RESUMO

@#The objectives of the study were to screen prostaglandin E2 receptor 4 antagonist from active compounds of Baeckea frutescens L. and to explore its anti-rheumatoid arthritis effect. The HEK293T-EP4 cell antagonist screening model was established in vitro. Homogeneous time-resolved fluorescence (HTRF) technique was used to screen the active compounds of Baeckea frutescens L..SPF grade ICR male mice were randomly divided into control group, model group, methotrexate group, and Baeckea frutescens L.compound BF-2 (100, 50 and 25 mg/kg) groups. The collagen-induced arthritis (CIA) mouse model was established in vivo. The swelling volume of the toes of mice was measured, and the pathological examination was analyzed by staining with hematoxylin and eosin.SPF grade ICR mice, male, were randomly divided into control group, BF-2 (100, 50 and 25 mg/kg) group, and aspirin group. The acetic acid-induced body writhing test was observed. The EP4 in vitro antagonist screening model was successfully established. The preliminary screening results found that BF-2, BF-20, BF-11 and BF-12 had strong EP4 antagonistic activity [(102.11 ± 3.45)%, (90.31 ± 3.59)%, (75.72 ± 1.79)% and (76.84 ± 1.64)%], and BF-2 had the strongest antagonistic activity (IC50 = 0.99 ± 0.08 μg/mL).BF-2 could significantly inhibit the toe swelling of CIA mice, and relieve the degradation of articular cartilage matrix and inflammatory cell infiltration. At the same time, compared with the control group, the writhing times of the mice in each dose of BF-2 were significantly reduced. In this study, BF-2 of Baeckea frutescens L.was selected as an EP4 antagonist, which has potential anti-rheumatoid arthritis activity.

RESUMO

Objective To study the methodology and effect of auricle reconstruction with prefabricated expanded fascial flap in temporal and parietal area and skin flap in mastoid area, covered on the Medpor support. Methods Thirty cases of congenital vestigial auricle were involved in this study. The operations were divided into two stages: in the stage I,the temporal and parietal superficial fascia flap was cut with a vascular pedicle in the parietal and temporal superficial area, which was implanted under the skin of mastoid, and then both layers of those were expanded at the same time; in the stage II, this fascial skin flap was cut down and covered on the whole Medpor support to rebuild the man-made auricle. Results Thirty cases were successful after following-up for 6 months to 3 years. Their auricle shapes were vivid; their skin color was normal, and it had no any difference from all around skins, or no any appearance of the support. Conclusions With best understanding the biological characteristics of the Medpor support and choosing the suitable procedures and using the prefabricated expanded support, we could minimize the surgical complications in this operation. The results show that this procedure is satisfactory, without any necrosis of flap, swelling of contour, or appearance of the support. The combination of two flaps is the best method for the auricle reconstruction.

RESUMO

Objective To study the methodology and results of the auricle repair with expanded skin flap in mastoid process area and Medpor support in children with congenital malformation. Methods Thirty cases of congenital malformation were enrolled with age ranged from 5~16 years old.Whole auricles were all reconstructed with an expanded skin flap in mastoid process area and Medpor support.Resuits In all successful cases.their repaired auricles had the natural complexion and profile, and the positions were symmetric with healthy one,but in 2 cases(age group of 11-16 years),the expanded skin flap had less skin grafting. Conclusion It is suggested that optional operation time might be selected before 10 years old, because their psychological trauma could be avoided owing to microtia and in that age the size of the expanded flap is larger enough to reconstruct the ear as their auricle iS well-developed.

RESUMO

Objective To investigate the effect of integrin expression on procollagen synthesis of fibroblasts in wound healing. Methods Antisense oligonucleotides were used to intercept the expression of integrin ? 5 or ? 1 subunit on fibroblasts respectively in wound healing, then the changes of procollagen mRNA were detected by RT-PCR. Results The antisense oligoncleotdes could specifically inhibit the expression of integrin subunit. Decreased expression of integrin?5 or ?1 subunit on fibroblasts significantly lowered mRNA of procollagen ? 1(Ⅰ) and ? 1(Ⅲ). Conclusion Reduction of integrin expression on fibroblasts in wound may inhibit procollagen synthesis in the level of transcription. These results provide the experimental gists on studying of scar reduction in wound healing.

RESUMO

Objective: To design the ribozymes to cleave human TIMP 1 mRNA, and embed them into U 6snRNA to make them stable. Methods: Ribozymes were designed according to the “hammerhead structure” described by Symons.Computer was used to analyze the possible cleavage sites. Results: Three ribozymes targeting the nt123, nt299 and nt353 on TIMP 1 mRNA were designed. Embedding ribozyme in U 6snRNA had little effect on its binding with the substrate. Conclusion: Computer assisted design is indispensable in studying ribozyme. Embedding ribozymes in U 6snRNA may be a good way to solve the problems existing in ribozyme study. [

RESUMO

Objective:To study the neural and vascular distribution in the triceps surae of rabbits for reconstructing the muscles of motor function. Methods: Triceps surae on one side was stained with the standard method of Sihler’s nerve staining, and the blood vessels on the other side were injected with a mixture of 30% barium sulfate and gelatin, and then they were X-photographed. The resulting pictures were compared to study the intramuscular nerve and blood vessel distribution. Results: Tricep surae on the first side became transparent or semi-transparent, and its shape kept intact after being stained. The intramuscular nerve branches were clearly visualized after being stained. The muscle was divided into 3 neuromuscular compartments and Lateral gastrocnemius (LG) was subdivided into 3 subunits. The intramuscular vascular configuration on the other side was also clear on soft X-ray films. The distribution maps of the nerves and blood vessels were grossly consistent. Conclusion: Sihler’s nerve staining is able to show the original 3-dimensional picture of the intramuscular nerve branches in the triceps surae of rabbits and can be used to observe the relation between the blood vessels and nerves in the muscle in combination with intramuscular angiography. According to the neurovascular distribution, tricep surae of rabbits can be divided into different subunits (compartments) as independent function units. The design and application of the subunit as a compartment can meet the need of muscular function after transplantation.

RESUMO

Nicotinic acetylcholine receptors concentrate on the postsynaptic membrane in the neuromuscular junction and participate in the neuromuscular transmission, which is of great importance to normal muscle function. The nicotinic acetylcholine receptor is a kind of pentamer. There are 2 types of nicotinic acetylcholine receptor due to the different subunit:fetal type ?-AChR (? 2???) and adult type ?-AChR(? 2???). This article reviews the difference of 2 types of nAChR in expression period, distribution, regulation mechanism and physiological significance.
