RESUMO

.Aim To establish the evaluation methods of the med¬icine nature of ginsenosides by eorrelation analysis between med¬icine nature and ginsenoside contents in five Panax herbs with different medicine nature and the measurement of their effects on Na + /K + -ATPase activities. Methods 'Hie contents of ginsen¬osides in Sanqi, red ginseng , ginseng , American ginseng and ginseng leaves in the existing literature were eolleeted.and the medicinal nature was assigned by vector methods.rI1ie medicine nature of ginsenosides and the contribution of ginsenoside to the medicine nature were evaluated through bivariate correlation a- nalysis and grey eorrelation degree respectively.'Hie effects of ginsenosides on the Na+ /K +-ATPase activities of L02 eells and in pig eerebral cortex were measured to evaluate the medicine na¬ture of ginsenosides. Results Correlation results indicated that the order of correlation coefficients of ginsenosides to the warm and hot medicine nature was Rf > R1 > Rg3 > Rg2 > Rbl > Ro, while the order of correlation coefficients of ginsenosides to the eool and eold medicine nature was Rb2 > Re > Rd > Fll.Grey eorrelation degree analysis showed that the contribution of ginsen¬oside to the medicine nature was F11 > Re > Kg2 > Rd > Rb2 >Rbl > Rgl > Rc > Rg3 > R1 > Rf > Ro.The effects of ginsen- osides on Na +/K +-ATPase activities showed that the substance basis of the warm medicine nature was ginsenoside Rbl, Rb3, Rc, Rf, Rg2, Rgl, Rhl, Rg3, R1 and Ro, which increased the activity of Na + /K + -ATPase.While the cold and cool medi- cine nature were ginsenoside Rh2, Rd, Rh2, Re and oleanolic acid, which inhibited the activity of Na+/K

RESUMO

OBJECTIVE: To establish UPLC-Q- TOF-MS/MS fingerprint of Fritillaria thunbergii , and to define its anti-inflammatory quality markers. METHODS :The determination was performed on Eclipse Plus C 18 column with mobile phase consisted of methanol- 0.1% formic acid solution (gradient elution )at the flow rate of 0.4 mL/min. The column temperature was set at 30 ℃,and injection volume was 2 µL. The electrospray ion source was used to scan in the range of m/z 50-1 200 with positive and negative ion detection mode. UPLC-Q-TOF-MS/MS fingerprints of 10 batches of F. thunbergii from different habitats were established by using the Similarity Evaluation System of TCM Chromatographic Fingerprints (2004A edition ). With ear swelling degree, the serum levels of MDA and NO as anti-inflammatory indexes ,the correlation between the relative area of common peaks in fingerprint and the anti-inflammatory indexes was analyzed by using bivariate correlation analysis and grey correlation analysis , and the quality markers were screened and identified. RESULTS :In positive and negative ion mode ,10 batches of F. thunbergii had 26 peaks and 10 peaks. Based on bivariate correlation analysis and grey correlation analysis ,nine quality markers related to anti-inflammatory effect were found ,which were identified as peiminine ,peimine, cyclobalamine, daucidin, polyphyllin Ⅴ, bitumen podophyllotoxin ,phytosterol,ent-kaur-15-en-17-ol,ent-17-norkauran-16-one. CONCLUSIONS :Established UPLC-Q- TOF-MS/MS fingerprint can be used to evaluate the quality of F. thunbergii ;peiminine, peimine and cyclobalamine and so on may be the quality markers of anti-inflammatory effect of F. thunbergii .

RESUMO

OBJECTIVE: To explore the relationship between the estrogen-like effect of seeds of Cuscuta chinensis and the fingerprints of the seeds of Cuscuta chinensis from different habitats, thus to provide a basis for the establishment of Chinese medicine quality standards based on the relationship between the fingerprinting and pharmacodynamics. METHODS: The fingerprints of seeds of Cuscuta chinensis samples from different habitats were established by HPLC, and the active ingredients of seeds of Cuscuta chinensis with estrogenlike effect were screened by uterotrophic method and MTT assay, and the spectrum-activity relationship was analyzed by bivariate correlation analysis. RESULTS: Twenty-six peaks of seeds of Cuscuta chinensis Lam. were identified as common peaks by HPLC. It was shown by uterotrophic and MTT assay that the seeds of Cuscuta chinensis from No. 20 habitat could significantly promote the proliferation of MCF-7 cells and the increase of uterus weight compared with the blank control group and the positive control group. It was shown that the estrogenic effect of seeds of Cuscuta chinensis was the combined effect of a variety of chemical constituents. CONCLUSION: There exsists correlation between the fingerprints of the seeds of Cuscuta chinensis from different habitats and their estrogenic activity.
