Aims@#Arthropods guts, such as termite harbor diverse microorganisms including those that are capable of fixing atmospheric nitrogen (N2). Nitrogen-fixing bacteria can help termite to overcome their shortage of dietary N by providing fixed N2. Nitrogenase enzyme is responsible for this trait and encoded by nif genes which are highly conserved and are primarily used in the identification of N2-fixing microorganisms. Here, we characterized N2-fixing bacteria isolated from the hindguts of termite Coptotermes gestroi.@*Methodology and results@#A total of 46 bacterial isolates were obtained after a primary screening based on their ability to grow on Burk’s media. Subsequently, the nifH gene from two of these isolates, namely S7 and S20, were successfully amplified and sequenced. Molecular phylogenetic analysis of 16S rRNA gene sequence revealed that isolate S7 is closely related to Ralstonia pickettii ATCC27511 (99.34% similarity, 1059 bp), whereas isolate S20 is closely related to Microbacterium sp. NCCP-451 (LC488936) (99.06% similarity, 948 bp). Besides that, the recA gene of isolate S7 is closely related to Ralstonia pickettii 12D (CP001644) (100% similarity, 442 bp) and the type strain of Ralstonia pickettii (ATCC 27511) (NZ KN050646) (98.97% similarity, 438 bp). Meanwhile, nifH gene of isolate S7 showed highest similarity to the uncultured bacterium NR1606 (AF035490) (99.93% similarity, 277 bp). Moreover, the nifH gene of isolate S20 is clearly separated from Azoarcus sp. and distantly related to Microbacterium sp. The incongruence between the partial 16S rRNA and nifH gene sequences could indicate the possibility of horizontal transfer of nif genes.@*Conclusion, significance and impact of study@#The phylogenetic incongruence between housekeeping genes (16S rRNA and RecA) and nifH gene in these bacteria provides new insight on potential horizontal gene transfer (HGT) activity taking place in bacterial communities particularly in the guts of arthropods. The finding of this study on potential HGT can also aid in the prediction of origins and evolution of gene transfer among bacteria.

Aims: Termites thrive in terrestrial ecosystems and play an important role in the bio-recycling of lignocellulose. The objective of this study is to isolate and detect bacteria from the termite gut of Coptotermes formosanus and to screen their various enzyme activities by qualitative methods. In addition, this study was aimed to isolate lignin and furfural tolerant strains for various industrial bioprocesses. Methodology and Results: In this study, 50 worker termites of Coptotermes formosanus were collected from dead trees, from a forest in Taichung, Taiwan in June 2008 and the composition of the microbial flora from the termite guts was analyzed by DGGE analysis. The results proved that anaerobic and facultatively anaerobic bacteria consisting of Acinetobacter, Bacteroides thetaiotaomicron, Escherichia coli, and Caulobacter readily existed in the guts of termites. Although the majority of these gut symbionts have not yet been cultivated or identified, some related bacteria were isolated. Two isolates 1-8 and 2-2 of Genus Bacillus, exhibited endocellulase, protease, lipase, amylase, peroxidase and lignin peroxidase activity. Under aerobic conditions, the growth density of isolate 1-8 cultured in 1000 ppm lignin containing MSM medium was two-folds higher than cultured in MSM medium without lignin. Furthermore, the isolate 1-8 was tolerant to 20 mM furfural supplemented in the MSM medium. HPLC analysis confirmed Bacillus isolate 1-8 could degrade up to 15 mM furfural. Conclusion, significance and impact of study: Hind gut bacteria from C. formosanus were detected by culture independent DGGE method. Also, Bacillus isolates 1-8 and 2-2 obtained by culture dependent methods could withstand higher concentration of furfural and as well as lignin. These isolates may be co-cultured with ethanologenic bacteria and be used as an industrial biocatalyst for biofuel production.

The termite fauna of a plantation of Eucaliptus urophylla S.T. Blake (Myrtales: Mytaceae) in Buritis, Minas Gerais, was manually sampled in 12 transects. The assemblage contained 28 species belonging to Termitidae and Rhinotermitidae. This assemblage corresponds to a subset of the native fauna of the cerrado previously present in this region. Compared to the original native fauna, it has a lower species richness, a much lower proportion of soil-feeders and a higher proportion of litter-feeders. A total of 1600 recently cut trees were examined and only three (0.2 percent) had damage to the heartwood caused by Coptotermes sp. Despite the presence of a diverse termite fauna including several putative pest species, termites are not considered a significant problem to eucalypt plantations in this region.


Os cupins de uma plantação de Eucaliptus urophylla S.T. Blake (Myrtales: Mytaceae) em Buritis, MG, foram amostrados manualmente em 12 transectos. A taxocenose encontrada continha 28 especies pertencentes a Termitidae e Rhinotermitidae. Essa taxocenoce corresponde a um subconjunto da fauna nativa do cerrado anteriormente presente na mesma área. Comparada com a fauna original, ela tem riqueza menor, uma proporção muito menor de humívoros e maior de ceifadores. O exame de 1600 árvores recém-cortadas revelou que apenas três delas (0.2 por cento) apresentavam dano ao cerne causado por Coptotermes sp. Apesar da presença de uma fauna diversa incluindo várias espécies consideradas pragas, os cupins não são considerados um problema significativo nessa região.
