RESUMO

On the basis of the previous work of the research group, the orthogonal design method was further used to optimize the processing technology for reducing toxicity of fried Tripterygium wilfordii in Lysimachia christinae Decoction. A total of 9 processed products of T.wilfordii in L.christinae decoction were prepared by four factors and three levels orthogonal design table. The contents of triptolide in T.wilfordii were determined by high performance liquid chromatography(HPLC) before and after processing: 4.27, 3.92, 3.57, 2.75, 2.42, 2.66, 3.51, 1.87, 1.75, 2.03 μg·g~(-1). On this basis, the above processed products were orally given to mice for 28 days. 12 hours after the last administration, food fasting except water was provided, and 24 hours later, the eyeballs were taken for blood and liver tissue. Serum biochemical indexes, liver lipid peroxidation and antioxidant related indexes were detected by kit method. Twenty-eight days after oral administration of raw T.wilfordii, the levels of serum alanine aminotransferase(AST), alanine aminotransferase(ALT), alkaline phosphatase(ALP) and liver malondialdehyde(MDA) in mice increased by 91%(P<0.01), 46%(P<0.05), 73%(P<0.01) and 99%(P<0.01), while the liver antioxidant indexes such as superoxide dismutase(SOD), glutathione(GSH), glutathione peroxidase(GPX) and glutathione-S transferase(GST) significantly decreased(P<0.01). After administration of the processed products, the above indexes were significantly reversed(P<0.01 or P<0.05). Especially, the processing conditions of A_3B_2C_1D_3 had the best detoxification effect on T.wilfordii, which decreased the high levels of AST, ALT, ALP and MDA by 49%(P<0.01), 32%(P<0.01), 42%(P<0.01), and 17%(P<0.05). Therefore, the best processing conditions for T.wilfordii in L.christinae decoction were A_3B_2C_1D_3, namely "15% mass fraction of L.christinae, 1 h moistening time, 160 ℃ frying temperature, and 9 min frying time".

Background:Mental Health First Aid, an important component of metal health educational programs, targets naive community members, public, allied mental health personnel, and mental health experts with specific reference as to how these trained aiders should help people in crisis or with mental health conditions in the community.Objective:This study aimed to describe pre-to-post-test assessment of those who were trained in MHFA program.Setting: National Center for Mental Health Promotion, Riyadh, Saudi Arabia.Methods:Trained mental health professionals organized MHFA courses (n=35) for training self-selected, nonrandomized community members (n=862) who were assessed pre-to-post training by using an adapted 17-item questionnaire for measuring the impact of MHFA course on their mental health knowledge, perception, attitude and practice.Results: The participants’ responses to questionnaire were varied at pre-and post-test evaluation and 65% of them showed mild to significant positive changes in their responses, and the rest were not affected positively after this course, possibly attributed to multiple factors related tonaïve community trainees, advanced questionnaire, short timeline and dissimilar instructors.Conclusion: Although all participants well received the MHFA courses, 65% of participants’ pre-to-post-test responses improved positively. The preliminary results of this studyarenot generalizable to the whole community,need to be cautiously interpreted and this research is calling for more studies especially randomized controlledtrials in future.

RESUMO

Objective: To establish the regeneration system of Saposhnikovia divaricate (Turcz.) Schisck. Methods: The effects of different explants, temperature, medium, and illumination was studied on the induction of S. divaricate. Response surface test design was used to optimize the hormone ratio of callus induction and proliferation. Results: The leaves of S. divaricate were used as the experimental materials, the optimal medium for the callus induction was MS + 6-BA 0.90 mg/L + 2,4-D 1.05 mg/L + KT 0.96 mg/L, with temperature of 25 ℃ and light of 12 h/d. The optimal medium for callus proliferation was MS + 6-BA 0.56 mg/L + 2,4-D 0.48 mg/L + KT 0.40 mg/L, and the subculture cycle was 35 d. The optimal medium for adventitious shoots was MS + NAA 0.6 mg/L + 6-BA 1.0 mg/L. The optimal medium for root growing was MS + NAA 0.4 mg/L + 6-BA 0.1 mg/L. Conclusion: S. divaricate callus was successfully induced and a stable and efficient plant regeneration system was established.

Objective: To optimize the extraction technology of two major iridoid glucosides (specnuezhenide and nuezhenoside G13) from four kinds of Osmanthus fragrans (OF) seeds, and to evaluate the anti-thrombotic activity of OF seeds. Methods: The orthogonal-test experiment was employed to optimize the parameters including ethanol concentration, liquid-material ratio, and extraction time for three extraction methods (ultrasonic extraction, reflux extraction, and microwave extraction). The extraction yield, content, and total peak area of iridoid glucosides were selected for weighted analysis to determine the best extraction method and technology. Additionally, an anti-thrombotic zebra fish model was established for biological evaluation of OF seeds. Results: Microwave extraction was the best method for iridoid glucosides extraction with the optimal conditions of ethanol concentration 55%, material-liquid ratio 1∶10, and microwave time 15 min. HPLC analysis showed that there was no significant difference in chemical composition among the four kinds of OF seeds. In zebra fish biological screening model, OF seeds displayed a weak inhibitory effect on the growth of thrombus and exhibited a pericardial edema effect in high dose-treated group. Conclusion: In this paper, extraction technology of two iridoid glucosides from four different kinds of OF seeds and preliminary anti-thrombotic activity evaluation of OF seeds were investigated. These results can provide the reference for further development and utilization of the agricultural waste of OF seeds.

Objective: To optimize the preparation process of gambogic acid (GA) liposomes and study the in vitro and in vivo release. Methods: The detection method of GA was established, using the Box-Behnken experiment design to optimize liposomes formula, GA liposomes were obtained with the highest encapsulation efficiency; Using scanning electron micrographs (SEM) to observe liposome surface morphology, using the dialysis method to study the liposome release in vitro, we also measured the stability of liposome in 15 d; Male Wistar rats were injected with GA or GA liposomes (1 mg/mL) via tail vein, UPLC-MS/MS method was used to determine the drug concentration, and differences in pharmacokinetic parameters of the two drugs were compared. Results: After Box-Behnken optimization, the encapsulation efficiency of liposomes was 92.3%, and the optimized liposomes formula is cholesterol of 440 mg, egg phosphatidylcholine of 1823 mg,,and istearoyl phosphoethanolamine-PEG 2000 of 705 mg, liposomes had uniform particle size and smooth surface; In vitro release results showed that the liposomes could be gentle and slowly release and had a long- term effect. The liposomes were stable keeping in 4 ℃ within 15 d; In in vivo study, the half-life of GA liposome was 9.97 h, 4.43 times of GA; AUC0-24 h of GA liposome was 22.55 μg∙h/mL, 4.73 times of GA. Conclusion: Compared with GA, GA liposome has the characteristics of long-circulating, high blood drug concentration, and could release smoothly.

Objective: To synthesize asialoglycoprotein receptor (ASGPR) ligand cholesterol-vinyl sebacate-lactitol (CH-VS-LA) by using enzymatic reaction in organic phase and to optimize its synthesis process. Methods: MS and 13C-NMR were used to identify the structure of product, enzymatic synthesis conditions were optimized via single-factor test and orthogonal experimental design. Results: The enzymatic optimum conditions were as following: the molar ratio of cholesterol-vinyl sebacate (CH-VS) and lactitol (LA) was 4:1, the amount of lipase Novozym 435 was 25 mg, reactional time was 32 h, and productive rate was 92%. Conclusion: The method is highly effecient, the reaction conditions are reasonable, and the process produces no by-product.

Objective: To optimize the prescription of Baoxieling Hydrogel Patch (BHP) using Box-Behnken test design and to investigate its transdermal absorption properties in vitro. Methods: Taking the comprehensive scores of the early adhesion, uniformity, ductility, consistence, skin adhesive ability, repeated exposing paste, and residue as response values, Box-Behnken test design was used to optimize the amounts of sodium polyacrylate NP 800, aluminum glycinate, and fillers, and to validate the optimal formulation. The percutaneous permeation of cinnamaldehyde, eugenol, evodiamine, and rutaecarpine in the optimal formulation was studied by in vitro transdermal delivery experiment with Franz diffusion cells and their contents were determined by HPLC. Results: The optimal ratio of the prescription was as follows: sodium polyacrylate NP 800-aluminum glycinate-fillers (0.82:0.02:1.56). The foremost factors were fillers and aluminum glycinate. Its transdermal absorption met zero order dynamic process. Conclusion: The optimal prescription has uniform paste, suitable consistence, easy ductility, moderate adhesion, and perfect transdermal effect. It could provide the foundation for the development of new prescription of Baoxieling.

Objective: To prepare and characterize gambogenic acid loaded nanostuctured lipid carrier (GNA-NLC). Methods: GNA-NLC was prepared by emulsion evaporation-low temperature solidification method, and its optimal prescription was selected by orthogonal test design. The characteristics, such as encapsulation efficiency (EE), average diameter, and Zeta potential, were studied. Results: GNA-NLC had a spherical shape with smooth surface with the average diameter of (144.07 ± 1.44) nm, polydispersity index (PI) of 0.24 ± 0.01, Zeta potential of (-28.03 ± 0.29) mV. The EE and drug loading were (84.65 ± 0.98)% and (4.21 ± 0.05)%, respectively. DSC indicated that GNA was dispersed as non-crystalline in matrix. Conclusion: Emulsion evaporation-low temperature solidification method is easy, simple, and feasible to prepare GNA-NLC.

Objective: To study the optimal extraction for Hirudo by wet ultrafine grinding technology. Methods: On the basis of single-factor experiments, the response surface analysis was made for exploring the optimal extraction conditions by wet ultrafine grinding technology for Hirudo, with solid-liquid ratio, soaking time, extraction temperature, and grinding time as independent variable and the anticoagulant activity as the response value. According to Box-Behnken principles, the response surface analysis method with four factors and three levels was used. Results: The optimum extraction condition for Hirudo by wet ultrafine grinding technology was as follows: solid-liquid ratio 1:12, soaking time 4.9 h, extraction temperature 6.5°C, grinding time 12 min; The anticoagulant activity could reach 20.13 U/g. Conclusion: The extraction by wet ultrafine grinding technology for Chinese materia medica Hirudo is stable and feasible, and it could be further promoted.

Objective: To prepare hydroxytyrosol (HT) by alkaline hydrolysis of oleuropein and to investigate its anti-oxidative property. Methods: According to single factor experiments, four variables: temperature, sodium hydroxide concentration, time, and the ratio of solid to liquid, were selected. On the basis of single factor experiments, orthogonal tests of L9(34) were carried out. HPLC-MS method was also further applied for the qualitative analysis on HT. Under the same conditions, contrast experiments on the acid and alkaline actions were made. Finally, anti-oxidative activity of HT was evaluated. Results: The results showed that the selected variables were all highly significant. Also, the descending influence orders on HT were temperature > sodium hydroxide concentration > time > the ratio of solid to liquid. Lastly, the optimal conditions on HT yield were obtained as follow: temperature 90°C, sodium hydroxide concentration 0.2 mol/L, time 45 min, and the ratio of solid to liquid 1:15. The HPLC retention time of HT was about 10.3 min, and its relative molecular weight of fragment ion by MS was 134.8. By comparison with the acid action, it was proved that the alkaline action was better in HT yield. In addition, the anti-oxidative activity of HT was carried out on scavenging DPPH radical. The results showed that DPPH scavenging capability of HT was 2.45 times of BHT. Conclusion: HT yield is well obtained under the alkaline condition and has a good anti-oxidation.

Objective To evaluate the importance of factors affecting efficiency of image display quality of realtime three-dimensional echocardiography (RT-3DE) and establish an optimized method for RT-3DE examination and image processing in children. Methods Based on corresponding cardiac acoustic characteristics in children, 87 healthy children (46 boys, 41 girls, mean 36.2 ± 44.2 months) were selected. Three aspects of seven factors were selected, including acquisition windows, gain, compress, post process, smoothing, frequency fusion and 3D vision for RT-3DE examination and analysis using orthogonal test design method. The principal effectiveness analysis of orthngonal test design method and its table L_(18)3~7 were used. The efficiency rate of 3D image display quality was used as the inspection index for orthogonal test. Results According to the average deviation orders of the seven factors, the maximal average deviation was gain. The aspects with maximal average deviation of each factor were at via-subcostal and via-apical acoustic windows, 90 of gain, 60 of compress, E of post process, 5 of smoothing, F3 of frequency fusion and A to B of 3D vision, respectively. Conclusions The optimized method for RT-3DE examination and image processing could be achieved by setting acquisition windows, gain, compress, post process, etc. It is helpful to promote clinical application of RT-3DE and benefits the design of multi-parameter presetting of ultrasound system to optimize RT-3DE examination in children.

The main purpose of this paper is to explore the applicability of multivariate multilevel models for bioequivalence evaluation. Using an example of a 4×4 cross-over test design in evaluating bioequivalence of homemade and imported rosiglitazone maleate tablets,this paper illustrated the multivariate-model-based method for partitioning total variances of In (AUC) and In (C_(max)) in the framework of multilevel models. It examined the feasibility of multivariate multilevel models in directly evaluating average bioequivalence (ABE),population bioequivalence (PBE) and individual bioequivalenc (IBE). Taking into account the correlation between In (AUC) and In (C_(max)) of rosiglitazone maleate tablets,the proposed models suggested no statistical difference between the two effect measures in their ABE bioequivalence via joint tests,whilst a contradictive conclusion was derived based on univariate multilevel models. Furthermore,the PBE and IBE for both In (AUG) and In(C_(max)) of the two types of tablets were assessed with no statistical difference based on estimates of variance components from the proposed models. Multivariate multilevel models could be used to analyze bioequivalence of multiple effect measures simultaneously and they provided a new way of statistical analysis to evaluate bioequivalence.

El propósito de esta investigación fue construir y validar un instrumento para evaluar Dependencia emocional. La muestra estuvo conformada por 815 participantes del Área Metropolitana de Medellín Colombia, 506 (62.1%) mujeres y 309 (37.9%) hombres, con edades entre los 16 y los 55 años. De los 66 ítems iniciales que contenía la prueba, fueron excluidos a través del análisis factorial 43 de ellos por no cumplir con los criterios para la selección. El cuestionario final quedó conformado por 23 ítems y seis factores. El Alfa de Cronbach de la escala total fue de 0,927, con una explicación de la varianza del 64.7%. Factor 1: Ansiedad de separación (7 ítems, α = 0.87), Factor 2: Expresión afectiva de la pareja (4 ítems, α = 0.84) , Factor 3: Modificación de planes (4 ítems, α = 0.75), Factor 4: Miedo a la soledad (3 ítems, α = 0.8), Factor 5: Expresión límite (3 ítems, α = 0.62) y Factor 6: Búsqueda de atención (2 ítems, α = 0.7.8). Se encontraron diferencias significativas con relación al sexo en las diferentes subescalas; las puntuaciones de las mujeres en Expresión afectiva de la pareja y Miedo a la soledad fueron mayores, mientras que los hombres reportaron puntuaciones superiores en Búsqueda de atención. Con relación a la edad se encontró que las subescalas Modificación de planes y Expresión límite puntuaban más alto en adolescentes y adultos jóvenes.


The purpose of this research project was to construct and validate an instrument to assess Emotional Dependency. The population sample was taken from the Metropolitan Area of Medellín (Colombia) and consisted of 815 participants, of which 506 (62.1%) were women and 309 (37.9%) were men, between the ages of 16 and 55 years old. The original questionnaire contained 66 items, and by means of a factor analysis test,43 of them were excluded for not complying with the selection criteria. Thus, the final questionnaire consisted of 23 items and six factors. The Cronbach Alpha score for the total scale was 0.927, which explained 65% of the variance, whereas for each specific factor the Alpha Scores obtained were the following: Factor 1: Separation Anxiety (7 items, α = 0.87); Factor 2: Couple’s Affective Expression (4 items, α = 0.84); Factor 3: Change of Plans (4 items, α = 0.75); Factor 4: Fear of Loneliness (3 items, α = 0.8); Factor 5: Borderline Expression (3 items, α = 0.62), and Factor 6: Attention seeking (2 items, α = 0.7.8). Significant differences related to gender were found in the different subscales. The highest scores for the female population were obtained in Factors 2 and 4, Couple’s Affective Expression and Fear of Loneliness, whereas men reported higher scores in Factor 6: Attention Seeking. With respect to age, it was found that the highest scores corresponded to the subscales Change of Plans and Borderline Expression in the Adolescent and Young Adult Population.

Most of the current college English term tests do not have an oral test section,which can not effectively and exactly examine students' language ability.Therefore,to better the reactive function of term tests and improve our oral English teaching,it's extremely necessary to include oral test section in.This paper aims to explore the methods of designing college English oral term test,beginning with the related theories and the discussion of its characteristics.Some suggestions are also put forward to help solve the possible difficulties.

extraction time.The extraction of total flavonoids in health food was the highest under the situations of ethanol concentration 60%,solid-to-liquid ratio 1:20 and ultrasonic time 20 min.The average value was 5.370 mg.The linear range of total flavonoids was between 1.0-40 ?g/ml while the regression equation is y=0.003 3x-0.010 9,correlation coefficient was 0.999 5 and detection limit was 1.0 ?g/ml.The standard addition recovery rates of this method were 95%-104%(n=3),RSD was 1.717%(n=5).Conclusion Under the optimum situation,this method has high precision and accuracy while the blank is low and stable as well.It is applicable to the detection of total flavonoids in health food and some other foods.

AIM:To study the extraction of Schisandra Chinensis (Turcz) using supercritical carbon dioxide as extracting ageat. METHODS: The study was carried out through orthogonal test design,the content of schizandrin from Schisandra Chinensis (Turcz) was selected by 3 influencing factors, extract temperature, extract pressure and separation temperature. RESULTS:The optimal condition for the extraction as A3B2C1 Schisandra Chinensis (Turcz) was extracted by supercritical carbon dioxide, extract pressure was 25MPa, extract temperature was 50?C, separation was 7 MPa, separate temperature was 60?C, extract time was 2.5h and runing amount of CO2 was 15kg?h -1. CONCLUSION:It is feasible that supercritical carbon dioxide is used in the preparation of Schisandra Chinensis (Turcz).

AIM: To study the optimum technological parameters for extraction of Astragalus polysaccharides by ultra-high pressure extraction. METHODS: Measured concentration of total astragalus polysacharides served as the marker,the design of uniform distribution was applied to evaluating the ultra-high pressure extration efficiency in comparison with reflux and ultrasonic extraction efficiency. RESULTS: The optimal parameters of ultra-high pressure extraction was as follows: 350 MPa pressure,60 mL/g ratio of solvent to solid material,5 h soaking time,2 min extraction time.Under the conditions,the high yield was up to 24.2 %. CONCLUSION: Ultra-high pressure extraction has a series of advantages,such as higher yield,less impurity,easier separation-purification,shorter extraction time and lower energy input.
