RESUMO

OBJECTIVE: To investigate the mechanism of human liver cancer HepG-2 cells apoptosis induced by Undaria pinnatifida sulfated polysaccharides S-UPPSIB. METHODS: The anti-proliferation effects of S-UPPSIB on HepG-2 cells was by MTT assay. [Ca2+]i in HepG-2 cells was detected by laser cofocal scaning microscopy (LCSM). The apoptosis rate and protein expression level of Bcl-2, Bax, Cyt-C and p53 were detected by flow cytometry (FCM). Caspase Assay Kit was used to detected the activities of Caspase-3 and -9. RESULTS: S-UPPSIB could inhibit the proliferation of HepG-2 cells and the IC50 was 50.09 μgmL-1. With the increase of drug delivery dosage, apoptosis rate also increased, and the significant regulating effects of S-UPPSIB on Ca2+ and its associated channel proteins were observed. The [Ca2+]i level, the protein expression level of Cyt-c, p53 and the activities of Caspase-3 and -9 were all increased remarkably (P<0.05). The ratio of Bcl-2 to Bax was reduced. CONCLUSION: Undaria pinnatifida sulfated polysaccharides S-UPPSIB can effectively inhibit the proliferation of human liver cancer HepG-2 cells by inducing apoptosis of HepG-2 cells through mitochondrial pathway.

BACKGROUND: Algae have traditionally been used for promotion of hair growth. Use of hair regrowth drugs, such as minoxidil, is limited due to side effects. The aim of this study was to examine a mixture of Saccharina japonica and Undaria pinnatifida (L-U mixture) on hair growth and to compare the promoting effect of hair growth by a 3% minoxidil and a L-U mixture. METHODS: To evaluate the hair growth-promoting activity, saline, 50% ethanol, 3% minoxidil, and the L-U mixture were applied 2 times a day for a total of 14 days on the dorsal skin of C57BL/6 mice after depilation. Analysis was determined by using a high-resolution hair analysis system, real-time polymerase chain reaction, and H&E staining. RESULTS: On day 14, the hair growth effect of the L-U mixture was the same as that of the 3% minoxidil treatment. The L-U mixture significantly (P<0.05) stimulated hair growth-promoting genes, as vascular endothelial growth factor (VEGF) and insulin-like growth factor -1. Increase of VEGF was observed in the L-U mixture group compared with minoxidil and the negative control. In contrast, the L-U mixture suppressed the expression of transforming growth factor-β1, which is the hair loss-related gene. In histological examination in the L-U mixture and minoxidil groups, the induction of an anagen stage of hair follicles was faster than that of control groups. CONCLUSIONS: This study provides evidence that the L-U mixture can promote hair growth in mice, similar to the effect from minoxidil, and suggests that there is potential application for hair loss treatments.

Undaria pinnatifida (U. pinnatifida) is a highly invasive species and has caused concern all over the world because it has invaded coastal environments, has the potential to displace native species, significantly alters habitat for associated fauna, and disturbs navigation. Any attempt to eradicate it would be futile, owing to the elusive, microscopic gametophyte, and because the alga thrives in sites rich in anthropic activities. Venice Lagoon is the largest Mediterranean transitional environment and the spot of the highest introduction of non-indigenous species, including U. pinnatifida, which is removed as a waste. We demonstrated that polysaccharide extracts from U. pinnatifida have an anticoagulant effect on human blood in vitro and are not cytotoxic. The results obtained by PT (normal values 70-120%) and APTT (normal values 28-40s) assays were significantly prolonged by the polysaccharide extracts of U. pinnatifida, therefore algal extracts are ideal candidates as antithrombotic agents.

RESUMO

Objective To extract dietary fiber from Udaria pinnatifida by using enzymatic-chemical extraction method.Methods All the factors that affect the extraction rate are enzyme content,enzymolysis temperature,enzymolysis time,Na2CO3concentration,Na2CO3extraction temperature,Na2CO3content and extraction time.They were tested with orthogonal experiment.Results The optimal conditions for extraction were determined as follows:the material was hydrolyzed at 50℃ for 1.5h with protease and cellulase mixed at the ratio of 30∶1,followed by the extraction at 65℃ for 3h with 30 volumes of 1% Na2CO3 solution.Under these conditions,the extraction rate could be as high as 12.65%with the color near light yellow.It could be used as additive source of dietary fiber for functional foods.Conclusion The optimum conditions for extraction dietary fiber were established.

RESUMO

Objective To study the extraction technology of the polysaccharides from Sporophyll of Undaria pinnatifida Suringar and find out the optimum extraction condition.Methods The extraction technology was studied by the orthogonal design.Results The optimum extraction conditions were to add 50 times of water to the Undaria pinnatifida Suringar material at 90℃ for 4 hours.Conclusion This optimum extraction technology based on the orthogonal design was confirmed by the experiment and was available in the future research.

Objective The antioxidative activities of extracts from Undaria pinnatifida in vitrowere tested. Methods Using the assay system of peroxide value (POV), diphenyl picryl-hydrazyl (DPPH), the antioxidative activities of various extracts were studied and comparedwith VE and VC. Results Different extracts from Undaria pinnatifida showed antioxidativeactivities, and petroleum ether extract showed the highest free radical scavenging efficiency.Conclusion petroleum ether extract has stronger antioxidative effect than others.

RESUMO

Objective To test the effect of Undaria pinnatifida soluble dietary fiber on endothelial function in hyperlipidemic rats. Method Forty rats were divided into 4 groups(n=10) :control group,hyperlipidemic model group,low dose dietary fiber-treated group(5%) ,high dose dietary fiber-treated group(10%) . After treatment for 8 w. Endothelium-dependent vasorelaxation in isolated aortic rings,the content of plasma malondialdohyde(MDA) ,nitric oxide(NO) and endothelin-1(ET-1) were determined. The protein expression of endothelial NO synthase(eNOS) was measured by Western blotting. Results Undaria pinnatifida soluble dietary fiber treatment significantly decreased MDA and ET-1 level. It also significantly improved endothelial function and plasma NO level concomitantly with unregulation of the expression of eNOS protein. Conclusion In hyperlipidemic rats Undaria pinnatifida soluble dietary fiber could improve vascular endotheliual function,which might be explained by its action to decreasing plasma ET-1 level and increased NO production.
