RESUMO

Objective:To evaluate the effect of Zuogui pill on cognitive function in sleep-deprived rats.Methods:Thirty SPF Sprague-Dawley male rats, aged 8 weeks, weighing about 200 g, were divided into 5 groups ( n=6 each) using a random number table method: control group (group C), sleep deprivation group (group SD), low-dose Zuogui pill group (group L), medium-dose Zuogui pill group (group M) and high-dose Zuogui pill group (group H). The model of sleep deprivation was developed by the multi-platform water environment method.In L, M and H groups, the doses of Zuogui pill were 2.42, 4.84 and 9.68 g/kg, respectively, Zuogui pill was administered by intragastric gavage at 10: 00 am every day, and the volume of fluid for intragastric gavage was 4 ml each time.In C and SD groups, pure water was used for intragastric gavage.In L, M and H groups, the liquid was prepared according to the corresponding doses of Zuogui pill for intragastric gavage, once a day for 7 consecutive days.The cognitive function was evaluated by the Morris water maze test.After the end of the second behavioral testing, blood samples from the eyeballs were collected, and then the rats were sacrificed by decapitation, and brains were removed and the hippocampus was obtained for determination of the concentrations of serum interleukin-1beta (IL-1β), IL-6 and tumor necrosis factor-alpha (TNF-α) and contents of malondialdehyde (MDA), glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) in hippocampal tissues (by enzyme-linked immunosorbent assay) and the expression of IL-1β, IL-6 and TNF-α in hippocampal tissues (by Western blot). Results:Compared with group C, the total swimming distance and escape latency were significantly prolonged, the time of staying at the original quadrant and the number of crossing the platform were decreased, the levels of serum IL-1β, IL-6 and TNF-α in serum and hippocampal tissues were increased, the content of MDA was increased, and the contents of GSH-Px and SOD were decreased in the other groups ( P<0.05). Compared with group SD, the total swimming distance and escape latency were significantly shortened, and the time of staying at the original quadrant was prolonged, and the number of crossing the platform was increased, the levels of serum IL-1β, IL-6 and TNF-α in serum and hippocampal tissues were decreased, the content of MDA was decreased, and the contents of GSH-Px and SOD were increased in L, M and H groups ( P<0.05). Conclusions:Zuogui pill can improve the cognitive function in sleep-deprived in rats, and the mechanism is related to inhibition of inflammatory responses and improvement in the antioxidant capacity.

BACKGROUND: Recent studies have found that oxidative stress plays an important role in osteoporosisOsteoporosis is closely related to aging, estrogen deficiency, nuclear factor E2 related factor 2, heme oxygenase 1 and reactive oxygen species production. OBJECTIVE: To explore the protective mechanism of Zuogui Pill on oxidative stress injury of osteoblasts. METHODS: The study protocol was approved by the Ethics Committee of the Second Affiliated Hospital of Harbin Medical University. Osteoblasts were isolated and cultured from the skull of neonatal Sprague-Dawley rats, and were divided into three groups: Control group, model group and Zuogui Pill group. The oxidative stress injury model of osteoblasts was established with 300 μmol/L H2O2 in the model and Zuogui Pill groups. Osteoblasts in the Zuogui Pill group were cultured with Zuogui Pill containing serum, and the control group did not undertake any treatment. Levels of malondialdehyde and superoxide dismutase in osteoblasts were determined. The expression of nuclear factor E2 related factor 2 and heme oxygenase 1 protein in osteoblasts was detected by western blot. RESULTS AND CONCLUSION: Compared with the control group, the content of malondialdehyde increased significantly, and the level of superoxide dismutase decreased significantly in the model group. Compared with the model group, the content of malondialdehyde decreased significantly and the level of superoxide dismutase increased significantly in the Zuogui Pill group. Western blot results showed that nuclear factor E2 related factor 2 and heme oxygenase 1 expression in osteoblasts of the model group increased significantly compared with the control group, and the expression of nuclear factor E2 related factor 2 and heme oxygenase 1 in osteoblasts of Zuogui Pill group was significantly higher than that of the model group. To conclude, Zuogui Pill can protect osteoblasts from oxidative stress injury.

Objective To evaluate the effectiveness and safety of modified Zuogui Pill combined with hormone in the treatment of premature ovarian failure ( POF) . Methods Randomized controlled trials were carried out by searching Chinese and English databases from the establishing to June, 2018. The meth-odological quality was assessed in the included randomized controlled trials. The treatment of premature o-varian failure by modified Zuogui Pill combined with hormone was systematically evaluated by RevMan 5. 3 software. Results 11 randomized controlled trials involving 1004 patients were included. Meta-analysis results showed that total effective rate (OR=4. 44, 95% CI: 3. 10 -6. 36, P<0. 00001), Tranditional Chinese Medcine ( TCM ) syndromes ( including dizziness, tinnitus, hot flash night sweats, five upset fe-ver, waist and knees), serum follicle stimulating hormone ( FSH) level [ standardised mean difference (SMD) =0. 54, 95% CI:0. 13-0. 95, P=0. 01], luteinizing hormone (LH) level (SMD=0. 90, 95%CI:0. 45-1. 36, P<0. 0001), estradiol (E2)level (SMD=1. 26, 95% CI:0. 12-2. 40, P=0. 03), endometrial thickness [weighted mean difference (WMD) =2. 03, 95% CI:0. 07-3. 99, P=0. 04] im-provement and the incidence of adverse reactions (OR=0. 34, 95% CI:0. 12-0. 95, P=0. 04) in modi-fied Zuogui Pill combined with hormone therapy were superior to those of pure hormone therapy. Conclu-sions The modified Zuogui Pill combined with hormone therapy shows better therapeutic effect and safety than hormone therapy on premature ovarian failure.

OBJECTIVE:To study the effect of Zuogui pill on the expression of gap junction protein 43(Cx43)in ovarian tis-sue of mice with premature ovarian failure(POF)induced by intraperitoneal injection of cyclophosphamide(CTX),and explore its mechanism in the treatment of chemotherapy-induced POF. METHODS:72 mice were randomly divided into blank group,model group,Estradiol valerate tablet group (positive control,0.00013 g/kg),Zuogui pill low-dose,medium-dose,high-dose groups (13.65,40.95,122.85 g/kg)by body mass,10 in each group. Except for blank group,other groups were reduce POF model by ip CTX,once a day,for 20 d. Meanwhile,the mice were intragastrically administrated related medicines,once a day,for 30 d. After 2 h of last administration,reverse transcription-polymerase chain reaction method and Western blot method were used to detect the Cx43 mRNA and protein expressions in ovarian tissue respectively,and immunohistochemistry method was adopted to detect the distribution of Cx43 protein in ovarian tissue. RESULTS:Compared with blank group,Cx43 mRNA and protein expressions in ovarian tissue of mice in model group were obviously weakened,and the distribution of Cx43 protein in follicle and granulocytes were obviously reduced(P<0.01). Compared with model group,Cx43 mRNA and protein expressions in ovarian tissue of mice in Estradiol valerate tablet group and Zuogui pill medium-dose,high-dose groups were obviously strengthened,and the distribution of Cx43 protein in follicle and granulocytes were obviously increased(P<0.01). CONCLUSIONS:Zuogui pill can increase the Cx43 mRNA and protein expressions in ovarian tissue of CTX-induced POF mice,increase the distribution of Cx43 in follicle and granu-locytes and gap junction function,which may be one of the treatment mechanism of POF.

<p><b>OBJECTIVES</b>To explore the effects of Chinese medicine prescription Zuogui Pill (, ZGP) on monoamine neurotransmitters and sex hormones in climacteric rats with induced panic attacks.</p><p><b>METHODS</b>Forty-eight climacteric female rats were randomized into 6 groups with 8 rats in each group: the control group, the model group, the low-, medium- and high-dose ZGP groups and the alprazolam group. Rats in the low-, medium- and high-dose ZGP groups were administered 4.725, 9.45, or 18.9 g/kg ZGP by gastric perfusion, respectively. The alprazolam group was treated by gastric perfusion with 0.036 mg/kg alprazolam. The control and model groups were treated with distilled water. The animals were pretreated once daily for 8 consecutive weeks. The behaviors of rats in the open fifield test and the elevated T-maze (ETM) were observed after induced panic attack, and the levels of brain monoamine neurotransmitters and the plasma levels of sex hormones were measured.</p><p><b>RESULTS</b>Compared with the control group, the mean ETM escape time and the levels of 5-hydroxytryptamine (5-HT) and noradrenalin (NE) of the model group were signifificantly reduced (P<0.05), Compared with the model group, the mean ETM escape time and the 5-HT and NE levels of all the ZGP groups increased signifificantly (P<0.05 or P<0.01). However, no signifificant difference was observed in the levels of sex hormones between the groups.</p><p><b>CONCLUSION</b>Pretreatment with ZGP in climacteric rats may improve the behavior of panic attack, which may be related to increased 5-HT and NE in the brain.</p>

Objective: To research the effects of Zuogui Pill on the gene expression profile of regenerate hepatic tissue in MSG-liver regeneration-rat.Methods: MSG-liver regeneration-rat models were copied;therapy group were given Zuogui Pill(5g/kg) by intragastric administration.Rats were executed at the 5th day after operation,and the regenerate liver tissues were frozen by liquid nitrogen.1176 gene chips of gene expression profile correlated to cell differentiation and multiplication were adopted to observe the changes of gene expression profile in the regenerate liver tissues.Results: There were 292 differential expression genes in therapy group compared with model group(20 genes were up-regulation expressed and 272 genes were down regulation expressed).Conclusion: Zuogui Pill has significant effects on the gene expression profile of regenerate hepatic tissue in MSG-liver regeneration-rat,and most of the genes are down regulation expressed.The result hints that Zuogui Pill could influence the liver regeneration by regulating the gene expressions of MSG-live regeneration-rat.Down regulation of the gene expressions of regenerate hepatic tissue in MSG-liver regeneration-rat possibly is one of the important molecular mechanisms of Zuogui Pill in nourishing liver and kidney.

Objective To investigate the roles of differentiation and proliferation related proteins in neural stem cells (NSCs) in rats treated with L monosodium glutamate (MSG). AIM: To explore the role of Akt signal pathway in apoptosis of neural cells in adult rats treated with Zuogui Pill, a Chinese medicine. METHODS: Flowcytometry and Western blotting methods were used to investigate the changes of cellular apoptosis rate and Akt signal pathway. RESULTS: Monosodium glutamate (MSG) could increase cellular apoptosis rate and significantly restrained the phosphorylation of Akt (Ser473) and Akt (Thr308), and markedly increased the levels of phospho-FKHR (ser256), GSK-3? (Ser9) and PTEN. Zuogui Pill partly inhibited the above effects of MSG. CONCLUSION: Zuogui pill effectively inhibits the neural apoptosis induced by MSG, and Akt pathway is involved in the neuronal protection of Zuogui pill. [

0.05).The number of vaginal folds and vessle of vaginal natural layers in the Zuogui Pill large dosage group were more than those in the model group(P0.05).Conclusion Zuogui Pill,the compound herbal formula for reinforcing the function of kidney,is not influential on the uterus of ovariectomized female rats.It can delay the vaginal deadness through increasing the local blood supply of ovariectomized rats.
