BACKGROUND The extracellular expression of enzymes in a secretion host such as Bacillus subtilis is a useful strategy in reducing the cost of downstream processing of industrial enzymes. Here, we present the first report of the successful extracellular expression in Bacillus subtilis WB800 of Geobacillus stearothermophilus lipase (T1.2RQ), a novel industriallydesirable thermostable lipolytic enzyme which has an excellent hydrolytic and transesterification activity. Signal peptides of a-amylase, extracellular protease, and lipase A, as well as two different promoters, were used in the secretion and expression of lipase T1.2RQ. RESULTS Lipase activity assay using p-nitrophenyl laurate showed that all three signal peptides directed the secretion of lipase T1.2RQ into the extracellular medium. The signal peptide of lipase A, resulted in the highest extracellular yield of 5.6 U/ml, which corresponds to a 6-fold increase over the parent Bacillus subtilis WB800 strain. SDS-PAGE and zymogram analysis confirmed that lipase T1.2RQ was correctly processed and secreted in its original size of 44 kDa. A comparison of the expression levels of lipase T1.2RQ in rich medium and minimal media showed that the enzyme was better expressed in rich media, with up to an 8-fold higher yield over minimal media. An attempt to further increase the lipase expression level by promoter optimization showed that, contrary to expectation, the optimized promoter exhibited similar expression levels as the original one, suggesting the need for the optimization of downstream factors. CONCLUSIONS The successful extracellular secretion of lipase T1.2RQ in Bacillus subtilis represents a remarkable feat in the industrial-scale production of this enzyme

Wolf cichlid, Parachromis dovii, is a species with a high potential for aquaculture in Central America; however, the knowledge of the digestive physiology in larvae period is limited. For these reason, this study evaluated the changes on digestive enzymes (alkaline and acid proteases, trypsin, chymotrypsin, aminopeptidase, carboxypeptidase, lipases, amylases, and phosphatases) during early ontogeny by biochemical analysis. All digestive enzymes were detected at first feeding (6 days after hatching, DAH, 9.49 mm, 168 degree-days DD). Afterwards all enzymes reached two main peaks in activity at 14 or 22 DAH (15.10 mm, 364 DD and 20.83 mm, 550 DD, respectively). Later, there was a gradual decrease in activity for trypsin and acid and alkaline phosphatases until reach the lowest values at 41 DAH. In the case of acid proteases, chymotrypsin, aminopeptidase, carboxypeptidase, lipase and amylase, all activities reached their maximum values at the end of the larval period, except for alkaline proteases, which showed the maximum value at 14 DAH (15.10 mm, 364 DD). Parachromis dovii larvae have an early capability to hydrolyze exogenous food, agreeing with other carnivorous neotropical cichlid species, for this reason we proposed that the weaning process could begin at 14 DAH.(AU)


El guapote lagunero (Parachromis dovii) es una especie con un alto potencial para la acuicultura en la región de América Central; sin embargo, existe un conocimiento limitado sobre la capacidad digestiva en el periodo larval. Por este motivo, este estudio evaluó los cambios de las enzimas digestivas (proteasas alcalinas y ácidas, tripsina, quimotripsina, aminopeptidasa, carboxipeptidasa, lipasas, amilasas y fosfatasas) durante la ontogenia temprana mediante análisis bioquímico. Todas las enzimas digestivas analizadas se detectaron en la primera alimentación (6 días después de la eclosión, DAH, 9.49 mm, 168 día-grados DD). Después, todas las enzimas alcanzaron dos picos máximos a los 14 o 22 DAH (15.10 mm, 364 DD and 20.83 mm, 550 DD, respectivamente). Después las actividades tripsina, fosfatasas ácidas y alcalina disminuyeron a sus valores más bajos a los 41 DAH. En el caso de las proteasas ácidas y alcalinas, quimotripsina, aminopeptidasa, carboxipeptidasa, lipasa y amilasa, los niveles de actividad aumentaron y alcanzaron su máximo valor al final del período larvario, excepto las proteasas alcalinas, que mostraron su máximo valor a los 14 DAH (15.10 mm, 364 DD). Las larvas de P. dovii tienen una capacidad temprana para hidrolizar alimentos exógenos, lo que concuerda con otras especies de cíclidos neotropicales carnívoros, por lo que proponemos que el proceso de destete inicie a los 14 DAH.(AU)

Digestive proteases were partially characterized in sheepshead juveniles, using biochemical and electrophoretic techniques. Results showed higher activity level of the stomach proteases (2.39 ± 0.02 U mg protein-1) compared to the intestinal proteases (1.6 ± 0.1 U mg protein-1). The activity of trypsin, chymotrypsin, leucine aminopeptidase and carboxypeptidase A was also recorded. The optimum temperature of the stomach proteases was recorded at 45 °C, while for intestinal proteases was recorded at 55 °C. Stomach proteases showed less stability to temperature changes than intestinal proteases. An optimum pH of 2 was recorded for stomach proteases with high stability under acidic conditions, while an optimum pH of 9 was recorded for intestinal proteases showing high stability under alkaline conditions. Stomach proteases were inhibited around 78% with Pepstatin A, indicating the presence of pepsin as the main protease. The stomach proteases zymogam revealed one active band with Rf of 0.49, this enzyme was completely inhibited by Pepstatin A. The intestinal proteases zymogram revealed four active proteases (51.3, 34.9, 27.8 and 21.2 kDa) that were inhibited by TLCK, which mainly represent a trypsin-like serine proteases. It can be conclude that digestion in sheepshead can be considered as a carnivorous species with an omnivorous tendency.(AU)


Se caracterizaron parcialmente las proteasas digestivas de juveniles del sargo, utilizando técnicas bioquímicas y electroforéticas. Los resultados muestran mayores niveles de actividad en las proteasas estomacales (2.39 ± 0.02 U mg proteina-1) comparados con los de las proteasas intestinales (1.6 ± 0.1 U mg protein-1), también se registró la actividad de tripsina, quimotripsina, leucina aminopeptidasa y carboxipeptidasa A. La temperatura óptima de las proteasas estomacales fue de 45 °C, mientras que la de las proteasas intestinales fue de 55 °C. El pH óptimo fue de 2 para las proteasas estomacales con alta estabilidad a condiciones ácidas, mientras que el pH óptimo para las proteasas intestinales fue de 9, mostrando una alta estabilidad en condiciones alcalinas. Las actividades de las proteasas estomacales fue inhibida en un 78% con Pepstatina A, lo que indica la presencia de pepsina, como principal proteasa. El zimograma de proteasas estomacales reveló una sola banda con actividad proteasa, con Rf de 0.49, completamente inhibida por Pepstatina A. El zimograma de proteasas intestinales reveló cuatro bandas (51.3, 34.9, 27.8 y 21.2 kDa). Todas las bandas se inhibieron con TLCK, lo que muestra la presencia principalmente de serina proteasas tipo tripsina. Se concluye que la digestión del sargo puede ser considerada como la de una especie carnívora con tendencia al omnivorismo.(AU)

Objective To study the protective effects of umbilical cord stem cells on cardiac injury in severely burned rats. Methods 30 SD rats were randomly divided into sham group ,burn group and stem cells treatment group. The severely burned head ,neck and back of rat model of 35%TBSAⅢdegree was eatabished in burn and stem cells treatment groups. The blood and cardiac tissues were collected at 24 hours after stem cells transplantation. The distribution patterns of umbilical cord stem cells in cardiac tissues ,and infiltrations of neutro-phils and macrophages in cardiac tissues were detected by immunohistochemistry staining method. The cardiac structures of rats were evaluated by HE staining. The levels of Troponin I ,Creatine Kinase and Creatine Kinase-MB in serum were detected by the blood biochemical automatic analyzer. Results The umbilical cord stem cells migrated into the injured cardiac tissues and mainly concentrated in capillaries of heart. The umbilical cord stem cells significantly decreased neutrophils and macrophages infiltration of the cardiac tissues ,and markedly alleviated structure damages of the cardiac tissues in severely burned rats. Furthermore ,the levels of Troponin I ,Creatine Kinase,Creatine Kinase-MB of burn group were significantly higher than that of sham group,but those of stem cells treatment group were markedly lower than that of burn group (P < 0.05). Conclution The umbilical cord stem cells transplantation could effectively reduce inflammatory cells infiltration of the cardiac tissues,then protected the structure and function of the heart in severely burned rats.

Metastasis is responsible for the majority of cancer-related deaths. Tumour invasion and metastasis result from processes that include the proteolytic degradation of the extracellular matrix adjacent to the tumour. The matrix metalloproteinases (MMPs), particularly MMP-2 and MMP-9, have prognostic influence in human cancers after they cleave the main structural components of the basal membrane. These actions make MMPs an attractive target for cancer and metastasis studies. This study evaluated the inhibitory potency of extracts of Bauhinia ungulata L. (BU) on the gelatinolytic activity of MMP-2 and -9 and recognized the group of secondary compounds responsible for this property. The zymographic analysis of the BU stem revealed that the ethyl acetate partition (D) caused a higher inhibition of MMP-2 and MMP-9. The phytochemical study of D showed the presence of steroids, tannins, and coumarins and the significant presence of alkaloids and flavonoids. The phytochemical study of the fractions obtained through the column chromatography of partition D revealed a significant presence of flavonoids and alkaloids in the fractions that showed better inhibition of the gelatinolytic activity of MMPs. In conclusion, these results suggest that the stem fraction of BU has the potential to inhibit MMP-2 and MMP-9 and should be used in studies on the recognition of active biomolecules.


A metástase é a responsável pela maioria das mortes relacionadas ao câncer. Tanto a invasão tumoral quanto a metástase resultam de processos que incluem a degradação proteolítica da matriz extracelular adjacente ao tumor. As metaloproteinases de matriz (MMPs), especialmente as MMP-2 e MMP-9, têm influência direta no prognóstico dos diversos tipos de câncer humano, pois clivam os principais componentes estruturais da membrana basal. Estas ações fazem das MMPs alvos atraentes para estudos envolvendo câncer e metástase. Este trabalho teve como objetivo avaliar o potencial inibidor dos extratos de Bauhinia ungulata L. (BU) sobre a atividade gelatinolítica das MMP-2 e 9 e reconhecer o grupo de compostos secundários responsáveis por esta propriedade. Análises por zimograma do ramo de BU revelaram que a partição de acetato de etila (D) causou maior inibição de MMP-2 e MMP-9. O estudo fitoquímico de D mostrou a presença de esteróides, taninos e cumarinas e a presença significativa de alcaloides e flavonoides. As frações da coluna cromatográfica da partição D e seus estudos fitoquímicos revelaram uma grande presença de flavonoides e alcaloides nas frações que apresentaram maior inibição da atividade gelatinolítica de MMPs. Em conclusão, estes resultados sugerem que frações do ramo de BU têm o potencial inibidor de MMP-2 e MMP-9 e devem ser utilizadas em estudos envolvendo o reconhecimento de biomoléculas ativas.

Polyacrylamide gel electrophoretic (PAGE) technique was used to examine the esterase pattern of Ctenidia, Hepatopancreas, Hemolymph, Intestine, Mantle. Foot of the Lamellidens corrianus. The zymograms revealed the varied pattern of esterase both in number and type. This varied pattern suggested several roles for these enzymes present in different tissues.

Objective To investigate the correlation between the cerebrocardiac syndrome (CCS) of severe type and prediction of prognosis of patients with neurosurgery .Methods Myocardial zymogram levels were measured in course of development of 120 ca‐ses of Acute Stroke(AS) that were confirmed diagnosis by brain computerized tomography (CT) .ultrasoundcardiogram(UCG)and electrocardiogram(ECG)were used to dectect Segmental Wall Motio Abnormalities (SWMA) .Results For 120 patients with AS , the specimens include 38 cases(76 .0% )of T‐wave change and 12 cases(24 .0% )of ST‐segment depression in 50 cases of ST‐T seg‐ment change ,7 cases of arrhythmia and adnormal cardiac electrical conduction .24 cases of SWM A ,32 cases(26 .7% ) of raised CK‐MB ,35 patients could be recovered to normal of ECG after active treatment .17cases of ECG improve markedly ,18 cases of ECG un‐improved ,15 cases of myocardial enzymes normally ,8 cases of myocardial enzymes improve markedly20 cases of death .Conclusion Hypertension is a risk factor of CCS ,correlation between ST‐T segment and SWMA as well as myocardial enzymes and myocardial damagein CCS was statistically analyed .

Low cost agro-waste was used as adsorption support for single-step purification of endoglucanase from the culture filtrate of A. fumigatus ABK-9. Among various agro-waste substrates, 1% NaOH pretreated rice bran was proved to be the best for adsorbing about 74.8 and 71.1% of endoglucanase at 4 °C and 10 °C respectively. Langmuir type adsorption isotherm at 4 °C showed maximum adsorption of enzyme at pH 5.0, which was in the range of optimum pH of the enzyme. The rice bran column bound enzyme was maximally eluted by a mixture of acetate buffer (0.05 M, pH 5.5) and ethanol (40%, v/v) at a ratio of 3:2 and a flow rate of 1 mL/min. A 5.52-fold purification of the enzyme was achieved from culture supernatant. The specific activity and recovery yield after purification were 294.0 U/mg and 40.15%, respectively, which were comparable with other contemporary protocols. The homogeneity of the enzyme was tested through sodium dodecyl sulphate polyacrylamide gel electrophoresis and a single band of 56.3 kDa was observed. Zymogram analysis finally confirmed the occurrence of endoglucanase in the single band.

Background: Enzymatic activity and laccase isoenzymes number of Pleurotus ostreatus grown in different pH values of the growing medium in submerged fermentation and incubated in buffer solutions of different initial pH values were determined. The expression profiles of five laccase genes (Lacc1, Lacc4, Lacc6, Lacc9 and Lacc10) in these cultures were also studied. Results: The highest laccases activity was obtained in cultures grown at initial pH of 4.5 and the lowest in cultures grown at initial pH of 8.5. Isoenzyme profiles were different in all the cases. Lacc1, Lacc4, Lacc6 and Lacc10 were expressed in all the cultures. Conclusions: The initial pH of the growing medium is an important factor for regulating the expression of laccase genes, having an effect on the activity and on the laccase isoenzymes number produced by P. ostreatus in SmF. This is the first report on the influence of different initial pH values of the growing medium on the laccases activity, laccase isoenzymes number and laccases expression profiles of P. ostreatus grown in submerged fermentation.

The objective of this study was to examine the action of the crude extract of Duddingtonia flagrans (isolates AC001 and CG722) on infective larvae (L3) of cyathostomins in coprocultures and to confirm its proteolytic activity by means of a zymogram. The following groups were formed in coprocultures: Group 1: 10 mL of crude extract of D. flagrans (AC001); group 2: 10 mL of crude extract of AC001 with 10 mM of Ca2+; group 3: 10 mL of crude extract of D. flagrans (CG722); group 4: 10 mL of crude extract of CG722 with 10 mM of Ca2+; and group 5: control group (distilled water). The third-stage larvae (L3) were obtained after eight days. The crude extract of D. flagrans was effective in reducing the number of L3, with the following percentage reductions: group 1, 49.5%; group 2, 52.5%; group 3, 36.8%; and group 4, 57.7%; in relation to the control group (p > 0.05). The proteolytic activity of the crude extract was confirmed through the zymogram. The results from this study confirmed that the crude extract of the fungus D. flagrans could be used for controlling cyathostomin L3, and suggested that at least one protease of approximately 38 kDa was present.


O objetivo deste trabalho foi estudar a ação do extrato bruto de Duddingtonia flagrans (isolados AC001 e CG722) sobre larvas infectantes (L3) de ciatostomíneos em coproculturas e confirmar a sua atividade proteolítica por meio de um zimograma. Foram formados os seguintes grupos em coproculturas: grupo 1: 10 mL de extrato bruto de D. flagrans (AC001); grupo 2: 10 mL de extrato bruto de AC001 com íons Ca2+ 10 Mm; grupo 3: 10 mL de extrato bruto de D. flagrans (CG722); grupo 4: 10 mL de extrato bruto de CG722 com íons Ca2+ 10 Mm; e grupo 5 como controle (água destilada), obtendo-se as L3 ao final de 8 dias. O extrato bruto de D. flagrans foi eficiente na redução do número de L3 com os seguintes percentuais de redução: grupo 1 (49,5%); grupo 2 (52,5%); grupo 3 (36,8%) e grupo 4 (57,7%) em relação ao grupo controle (p > 0,05). Confirmou-se a atividade proteolítica por meio do zimograma. Os resultados do presente trabalho confirmam a utilização do extrato bruto do fungo D. flagrans no controle de L3 de ciatostomíneos e sugere a presença de pelo menos uma protease de aproximadamente 38 kDa.

Objective To investigate the effects of different flow volume of continuous 1:1 blood crystalloid cardioplegia on open heart surgery.Methods Retrospective randomized study was performed on 80 patients undergoing valve replacement and congenital heart disease with 1:1 blood crystalloid cardioplegia by minimum-flow continuous perfusion[0.5ml/(kg·min),group A,40 cases],high-flow continuous perfusion[(2.0 ml/(kg·min),group B,40 cases).The changes of creatine kinase(CK),isoenzyme of creatine kinase(CK-MB),lactate dehydrogenase(LDH),lactic dehydrogenase isoenzyme(LDH_1)were detected before operation and at 6,12,24,48,72 h after operation.Results The cardioplegia liquid was (41.72±7.98)ml/kg in group A,(142.09±9.65)ml/kg in group B,there was significant difference between two groups(P<0.05).During the first 72 h postoperative the value of CK,CK-MB,LDH and LDH1 in two groups were increased to different degree,serum enzymes levd increased significantly between 12,24 h after operation(P<0.05)and return to normal at 72h after operation in two groups.Butbetween two groups there was no significant difference(P>0.05).ConclusionMinimum-flow continuous perfusion of 1:1 blood crystalloid cardioplegia appears to be superior in safety,decrease of myocardial enzymes integrity.

Objective To investigate the protective effect of ginsenoside Rb1 and Re on injury of the neonate rat cardiomyocyte induced by aconitine alkaloids.Methods The influence of aconitine and ginsenoside Rb1,Re together in neonate rat cardiomyocyte was observed respectively.The influence of these components in neonate rat cardiomyocyte was examined by myocard zymogram.The expression of Ca2+ channel gene Cav1.2 mRNA of neonate rat cardiomyocyte after treatment was observed by RT-PCR.Results The ginsenoside Rb1 and Re were able to decrease the release of AST and LDH,reverse Cav1.2 mRNA abundance induced by aconitine.Conclusion The ginsenoside Rb1 and Re which together with aconitine can alleviate the side effects and boost up the therapeutic effects of aconitine.The action mechanism may be relation with that the ginsenoside Rb1 and Re can decrease injuries of the neonate rat cardiomyocytes and abundant expression of Cav1.2 mRNA induced by aconitine.

Objective To summarize the clinical features,efficacy and prognosis of 42 children patients with severe viral myocarditis(VM).Methods To retrospecitivly analyze the clinical data about 42 children patients with severe VM.Results There were the lack of typical clinical features and ancillary inspection targets on diagnosis,and no effects on clinical treatment,so that it was being concealed in the basis of disease symptoms and delayed treatment.Amidst this group patients(42 cases)of children with severe viral myocarditis,there was 76.2% patients with the heart symptoms and 47.6% patients with serious cardiac complications(such as acute heart failure,cardiogenic shock,fatal arrhythmia).Therapeutic result:total effective rate 78.6%,morality 11.9%,14.3% to chronic.Conclusion To diagnose Viral myocarditis in children it hould be combined with the history of palpitations,chest tightness,and other clinical symptoms and objective indicators of myocardial damage.Because of no specific means of treatment,we should be focus on comprehensive treatment and timely care and rescue critically ill patients.The majority of children patients with severe VM are favorable prognosis and a very small number to chronic.

180 U/L.CK-MB raised in only 5 cases(15.6%),and the highest was 28 U/L.Of the 218 MPP patients,only the 5 patients(2.3%) with raised CK-MB levels could be diagnosed as myocardial damage complicated with MPP.Conclusions The abnormal ECG is more important than myocardial zymogram in the diagnosis of myocardial damage complicated with MPP.Only higher level of certain myocardial zymogram without abnormal ECG can not be diagnosed as myocardial damage if lack of clinic and other objective evidences for myocardial damage.In watching out for myocardial damage,excessive examinations and treatments shall be avoided if possible.

Objective To analyse the lab diagnosis guideline in the patients with PM and DM,and guide the clinical diagnosis. Methods Stage the controlling condition and examine the serologic zymogram,electromyography and pathology.Results In the and Hospital of Hebei medical Vnirersity,from Jan.2003 to Jan.2005 CK,CK-MB,AST,LDH and HBDH was higher in 82.9%patients of aetive stage than the stable stage,especially changed obviously in CK.The occurance ritio(75.9%,48.3%)of the inside electricity and multiphase wave were much higher than the stable stage(46.7%,20.0%).Lots of organic fibre shrank in PM,so did the inflammation of lymohocyte soakage.In DM,the fascicle began to shrink in different degrees.Conclusion CK could become the guideline to watch the activity of illness.The number of the electromyography inside electricity and multiphase wave are related to the illness stage of patients with PM and DM.In PM pathology,muscle fibre had some infected cell soakage,with the differential recessive change.The pathology character in DM was the atrophy around the facicle and the change of tiny blood vessel.

RESUMO

Objective To investigate the change and its significance of blood level of myocardium zymogram in acute organophosphrus poisoning(AOPP) mice.Methods 152 eight-week-old male mice were divided into 3 groups randomly: normal control group(n=8);normal saline control group(n=10);AOPP group(n=135).The AOPP group mice were assigned by 9 time-points(30 min,2 h,4 h,12 h,1 d,2 d,3 d,4 d and 7 d after poisoning).15 mice were in per cage.The mice in AOPP group were introperitoneal injected Omethoate(50 mg/kg) and Atropine(10 mg/kg).Centhifugalizing the blood and the serum were taken,examined cholineesterase(CHE),creatine kinase(CK),aspartate aminotransferase(AST) and lactate dehydrogenase(LDH).The level of myocardium zymogram of myasthenia mice was compared with normal muscle mice in the AOPP group.Results Compared with normal control group and normal saline normal saline control group,CHE activity was persistent inhibition in the AOPP 30 min ~3 d subgroups(allP

Incidence of aseptic loosening of hip prostheses is increasing in recent years. Previous studies suggested the involvement of proteinases and cytokines in the accelerated bone lysis associated with loosening. To investigate the role of matrix metalloproteinases (MMPs) in the loosening, Gelatin/Type IV collagenases, namely, 72 KDa matrix metalloproteinase (MMP)-2 type and 92 KDa MMP-9 type were analyzed in 14 cases of the loosened endoprostheses of the hip. Zymographic and densitometric analyses revealed production of MMP-2 ancl elevated induction of MMP-9 in tissue extracts from both the interface hetween hone and implants and the capsular tissues when compared with those in synovium obtained from a patient with a t'ractured femoral neck. MMP-9 showed stronger activity than MMP-2. In the sample of a fractured femoral neck, MMP-2 was detected, but MMP-9 was not detected. In matched samples, the activity of MMP-2 and MMP-9 in the interface tissues showed stronger activity than those in the capsular tissues. There was no difference between cemented and uncemented femoral prostheses. The state of prostheses(loosening, osteolysis, and cup wear) did not influence on the activity of MMP-2 and MMP-9. Theses findings suggest a role for MMP-2 and MMP-9 type gelatinase/Type IV collagenases in the degradation of extracellular matrix of periprosthetic tissues, where they may cause weakening of the connective tissue hed and the loosening of total hip replacement endoprostheses. Consequently. we could confirm the role of MMP cascade in aseptic loosening of total hip prostheses. The further study ahout other types of MMP and the inhihitor of MMP will be needed.
