RESUMO

In vitro treatment of erythrocytes of normal and hypercholesterolemic rats with N-nitrosodiethylamine (NDEA), an important carcinogen frequently present in human environment and food chain resulted in a marginal increase in osmotic fragility of erythrocytes without affecting their antioxygenic potential as evidenced by insignificant effect on lipid peroxidation (LPO). However, (LPO) of all the tissues (heart, lung, liver, kidney and spleen) increased significantly on in vitro treatment with NDEA. The effects were different in different tissues under different dietary conditions

RESUMO

The present work was undertaken to study the changes in osmotic resistance of stored blood, used for blood transfusion. Blood samples of 50 cases from Orissa Red Cross Blood Bank, Cuttack, were studied. The blood was collected in acid citrate dextrose solution and stored at 4 degrees C. Storage was done under usual condition followed in blood banks. Osmotic fragility was determined at 3 days interval up to 15 days. In each case ESR was determined and in 20 cases plasma fibrinogen level was determined. In this study there was gradual increase in fragility which was maximum on 9th day and then slowly declined, which was statistically significant. ESR decreased markedly after 6th day, which was also statistically significant.

RESUMO

Effect of intact and alkali solubilized insecticidal crystal protein (ICP) preparations from a mutant strain of B. thuringiensis var. israelensis (VCRC MB24) and the wild type strain (VCRC B17) in vitro on human erythrocytes with respect to lipid peroxidation, osmofragility and membrane bound enzymes was determined. The alkali solubilized ICPs of both B. thuringiensis strains caused increased lipid peroxidation, decreased resistance to hypotonic lysis and reduction in the activity of membrane bound enzymes. On the contrary, the intact ICPs did not produce any such adverse effect on RBCs under the same experimental conditions. It is suggested that the ICPs are safe when they are intact when compared with solubilized ones.

Various quinolones have varied effects on the preservative activity of blood cells. Nalidixic acid causes hemolysis in G-6PD deficient patients where as ofloxacin has been found to possess preservative action of WBCs. The present study was undertaken to see the effect of various fluoroquinolones on RBC membrane. The effect of quinolones like ciprofloxacin, ofloxacin and norfloxacin and nalidixic acid in the dose of 5 micrograms/ml was studied on dapsone induced, in vitro hemolysis of rabbit RBC, using the osmotic fragility test. The mean corpusular fragility (MCF) with various agents were as follows: (mean +/- SE) saline; 5.23 +/- 0.21; dapsone, 6.57 +/- 0.23; ofloxacin, 3.81 +/- 0.13; ofloxacin and dapsone, 5.13 +/- 0.11; nalidixic acid, 6.28 +/- 0.16; nalidixic acid and dapsone, 6.65 +/- 0.13; ciprofloxacin, 3.52 +/- 0.22; ciprofloxacin and dapsone, 4.80 +/- 0.2; norfloxacin, 1.97 +/- 0.23; norfloxacin and dapsone, 4.27 +/- 0.20. The MCF data and shift of the osmotic fragility curves (to the left) show that dapsone induced erythrolysis is significantly protected by ciprofloxacin, ofloxacin and norfloxacin but not by nalidixic acid.

RESUMO

(-) Epicatechin, a benzopyran extracted from the bark of Pterocarpus marsupium, is reported to have insulin like activity. The present work is undertaken to study the effect of insulin on erythrocyte osmotic fragility (OF) and then to evaluate the insulin-like role of (-) epicatechin on human erythrocytes. Insulin exerts a protective effect on erythrocyte OF and shows a dose response which is similar to other typical insulin effects i.e. a maximum at 0.1 nM and a lower effect at higher and lower concentration. (-) Epicatechin (1 mM) also shows protective effect, similar to insulin, on the OF. Ouabain (1 mM) has completely abolished the insulin effect on OF, and failed to have any effect on the action of (-) epicatechin, showing that (-) epicatechin and insulin act by a different mechanism of action while eliciting their protective effects on red cell OF.

RESUMO

Lectins from peas and lentils when injected to rats apparently appeared to be non toxic but they caused growth depression. The organ weights were not affected except spleen enlargement. The lectins also caused increased osmotic fragility of erythrocytes without affecting other hematological parameters such as haemoglobin, packed cell volume, and RBC count.

RESUMO

Para determinar la relación existente entre el uso de oxitocicos y/o soluciones glucosadas en el trabajo de parto sobre la hiperbilirrubinemia neonatal, se estudiaron 90 R.N de menos de 24 horas de vida, en el Dpto. de Pediatría del Hospital Antonio María Pineda. Barquisimeto. Venezuela. Se distribuyeron en 3 grupos: A) R.N. cuyas madres recibieron soluciones glucosadas más oxitocicos, B) Soluciones glucosadas solamente, C) Control. Se encontró aumento significativo de las cifras de Bilirrubina Sérica en los R.N. del Grupo A y B con respecto al control (3.0 mg%, 2,6 mg%, 2.0 mg% respectivamente) con una p < 0.001; así mismo, se detectó que estos niveles aumentaban en forma proporcional al aumentar las concentraciones de oxitocicos y soluciones glucosadas administradas a las madres durante el parto, con una coeficiente de correlación de r = 1 y r = 0.90 respectivamente. Se determinó los niveles de Na en los 3 grupos de R.N.: 113 meq/L, 133 meq/L. 139 meq/L. con una diferencia estadísticamente significativamente de p < 0.001, con respecto al control. De la misma manera se encontró que a medida que aumentaban las cantidades de soluciones glucosadas y oxitocicos a las madres, disminuían en forma inversamente proporcional los niveles de Na sérico en los R.N. con un coeficiente de correlación de r = 0.90. La fragilidad osmótica, fue mayor (el porcentaje de hemolisis) en el Grupo A, B que en el C. Nuestros resultados apoyan la hipótesis de que las soluciones glucosadas y los oxitocicos, usados en forma indiscriminada y a alta dosis ocasionan Hiponatremia materna-fetal, que aumenta la fragilidad osmótica eritrocitaria fetal, con elevación..

RESUMO

The effects of amphotericin B, an antifungal antibiotic, on erythrocyte volume and cation permeability were investigated by measuring the hematocrit, cell volume, cation content, fragility and osmotic behavior in rat erythrocytes, in vitro. 1. When erythrocytes were incubated in a Ringer solution containing amphotericin B (5-25 microgram/ml) the hematocrit and the cell volume increased, the effect being proportional to the concentration of the drug and the incubation time period. 2. Amphotericin B increased the Na content and decreased the K content of the erythrocyte. In normal Ringer solution (NaCl-Ringer)containing amphotericin B the magnitude of cellular Na gain was greater than that of K loss. Therefore, the total cellular cation content increased. On the other hand, when cells were incubated in the amphotericin B containing Ringer solution in which NaCl was replaced by Na2SO4 (Na2SO4-Ringer) the magnitude of cellular K loss exceeded that of cellular Na gain. Consequently, the total cellular cation content was reduced. 3. Amphotericin B increased cell volume (hematocrit) when erythrocytes were incubated in the Na2SO4-Ringer solution. 4. The fragility of erythrocytes increased when cells were preincubated in the amphotericin B containing normal Ringer solution, whereas it decreased in tile cells preincubated in the amphotericin B containing Na2SO4-Ringer solution. 5. The cell volume was linearly related to the reciprocal of medium osmolality(200 to 900 mOsm/kg H2O) in both NaCl-and Na2SO4-Ringer solutions, and the linearity was not altered by amphotericin B. The antibiotic did not change the slope of the correlation line (V vs. 1/OSM). It, however, increased the intercept of the line with the ordinate in normal Ringer solution and decreased that in the Na2SO4-Ringer solution. These results indicate that amphctericin B alters the cell volume by changing the permeability of Na and K across the membrane.
