OBJECTIVE: To determine the prevalence of the Torque teno virus in healthy donors in the northern and northwestern regions of the state of Paraná, southern Brazil.METHODS: The Torque teno virus was detected by a nested polymerase chain reaction using a set of oligoprimers for the N22 region.RESULTS: The prevalence of the virus was 69% in 551 healthy blood donors in southern Brazil. There was no statistically significant difference between the presence of the virus and the variables gender, ethnicity and marital status. There was significant difference in the prevalence of the virus regarding the age of the donors (p-value = 0.024) with a higher incidence (74.7%) in 18- to 24-year-old donors.CONCLUSION: A high prevalence of Torque teno virus was observed in the population studied. Further studies are needed to elucidate the routes of contamination and the clinical implications of the virus in the healthy population.

Patients who undergo dialysis treatment or a renal transplant have a high risk of blood-borne viral infections, including the Torque teno virus (TTV). This study identified the presence of TTV and its genome groups in blood samples from 118 patients in dialysis and 50 renal-transplant recipients. The research was conducted in a hospital in the city of Maringá, state of Paraná. The viral DNA, obtained from whole blood, was identified by using two nested Polymerase Chain Reactions (PCR). The frequencies of TTV were 17% and 36% in dialysis patients using the methodology proposed by Nishizawa et al. (1997) and Devalle and Niel (2004), respectively, and 10% and 54% among renal-transplant patients. There was no statistically significant association between the frequency of the pathogen and the variables: gender, time in dialysis, time since transplant, blood transfusions, and the concomitant presence of hepatitis B, for either the dialysis patients or the renal-transplant recipients. Among dialysis patients and renal-transplant recipients, genogroup 5 was predominant (48% and 66% respectively), followed by genogroup 4 (37% and 48%) and genogroup 1 (23% and 25%). Genogroup 2 was present in both groups of patients. Some patients had several genogroups, but 46% of the dialysis patients and 51% of the renal-transplant recipients had only a single genogroup. This study showed a high prevalence of TTV in dialysis patients and renal-transplant recipients.

Esta pesquisa aborda percepções de homens sobre suas experiências e necessidades de saúde no pós-nascimento. Objetiva-se distinguir analiticamente, a partir de uma perspectiva de gênero, necessidades de saúde relativas ao evento, experimentadas, expressas e/ou negadas pelos homens. Trata-se de um estudo exploratório-descritivo, qualitativo, realizado em dois territórios de saúde de Cuiabá, Mato Grosso, mediante entrevista semiestruturada e análise temática dos dados. Participaram oito homens que vivenciavam o pós-nascimento de um filho. Estes manifestaram, sobretudo, a necessidade de provimento de segurança financeira à família, destacando também a necessidade de bem estar do filho, acessando o retorno afetivo que a experiência traz. Não se reconheciam com necessidades de saúde no pós-nascimento. O apoio dos serviços locais de saúde é importante para que os homens se percebam na vivência da paternidade e a sua relação com aspectos socioculturais, para que distingam necessidades próprias, valorizem o cuidado de si e adotem uma perspectiva equânime de gênero.


This research addresses men’s perceptions about their experiences and health needs in the post-birth period. The goal is to distinguish analytically and from a gender perspective the health needs related to the event, experienced, expressed and/or denied by men. This is an exploratory, descriptive and qualitative study carried out in two municipal health areas of Cuiabá, state of Mato Grosso, using a semi structured interview and thematic analysis of the data. The participants were eight men who were experiencing the post-birth period of a child. Above all, they expressed the need to provide financial security to the family, emphasizing also the need of their children’s well-being, with the affective return that this experience brings. They did not perceive themselves with health needs during the post-birth period. The support provided by local health services is important so that men can understand the experience of parenthood and its relationship with social and cultural aspects in order to distinguish their own needs, appreciate self-care, and adopt a gender equity perspective.


Esta investigación aborda las percepciones de hombres acerca de sus experiencias y necesidades de salud en el período postparto. El objetivo es distinguir analíticamente, desde una perspectiva de género, las necesidades de salud relacionadas al evento, experimentadas, expresadas y/o negadas por los hombres. Se trata de un estudio exploratorio, descriptivo y cualitativo llevado a cabo en dos áreas municipales de salud de Cuiabá, Estado de Mato Grosso, mediante entrevista semiestructurada y análisis temático de los datos. Participaron ocho hombres que experimentaban el período postparto de un hijo. Ellos expresaron, sobre todo, la necesidad de proveer seguridad económica a la familia, destacando también la necesidad del bienestar de los hijos, con el retorno afectivo que esa experiencia proporciona. No se reconocían con necesidades de salud en el período postparto. El apoyo de los servicios locales de salud es importante para que los hombres entiendan la experiencia de la paternidad y su relación con aspectos socioculturales, para que distingan las necesidades, valoren el cuidado de sí mismos y adopten una perspectiva de equidad de género.

Introduction Torque teno virus (TTV) and SEN virus are circular single-stranded DNA viruses that cause blood-borne infections. The SEN virus (SEN-V) was originally detected in the serum of an injection drug user infected with human immunodeficiency virus (HIV). Recently TTV was discovered as a potential causative agent of non-A-E hepatitis. The aim of this study was to investigate the prevalence of the SEN-V-D/H and TTV in HIV patients and healthy blood donors in Iran. Methods One hundred and fifty HIV patients with a mean age of 50.46 ± 18.46 years and 150 healthy blood donors with a mean age of 48.16 ± 13.73 years were included in this study. TTV and SEN-V were detected by the PCR and were quantitatively assayed by competitive PCR (nested and semi-nested PCR). Restriction fragment length polymorphisms (RFLPs) were used to determine the heterogeneity of TTV. Results TTV and SEN-V were detected 96 (64%) and 84 (56%) of 150 HIV patients respectively. These rates were 34% (n=51) and 37.33% (n=56) in healthy blood donors (significant, p<0.05). PCR detected SEN-V/TTV DNA from 32 of the healthy blood donors (21.33%), while 65 (43.33%) of HIV patients were positive for SEN-V/TTV DNA. Of 150 HIV patients, 32.66% and 23.33% were positive for SEN-V-H and SEN-V-D, respectively and 18.66% (n=28) were co-infected with SEN-V-D/H. Conclusions The prevalence of SEN-VD/H and TTV is higher in HIV patients than in healthy blood donors in Southern Iran. Our results suggest that TTV and SEN-V might play a role in the development of liver disease in patients with immunodeficiency diseases. .

Torque teno sus virus (TTSuV) infection is present in pig herds worldwide. It has been demonstrated that TTSuV might increase the severity of other important viral diseases with economic and public health impacts. At present, there is no information on the age distribution of pigs infected with TTSuV in Brazilian herds. This study evaluated the frequency of TTSuV infection in pigs at different stages of production. Fecal samples (n=190) from pigs at 1 to 24 weeks of age and from breeders at 6 farrow-to-weaning (up to 8 weeks of age) and 9 grower-to-finish (9 weeks of age onwards) farms in the western region of Paraná state, Brazil, were evaluated by PCR. Fragments of the 5' UTRs of TTSuV1 and/or TTSuVk2 DNAs were identified in 126 (66.3%) of the fecal samples. Significant differences were found with the percentages of positive samples for TTSuV1, TTSuVk2, and mixed infections by both genera between and within the different pig production stages. Fecal samples from the grower-to-finish farms had TTSuV detection rates (90.1%; 64/71) that were significantly (p<0.05) higher than those from the farrow-to-weaning farms (52.1%; 62/119). TTSuV detection was significantly (p<0.05) more frequent in finisher pigs than in the animals from the other stages. The UTR nucleotide sequences in this study presented higher similarities to strains from Norway (96%, TTSuV1), and Argentina and China (97.1%, TTSuVk2). These results suggest that TTSuV infection has spread to pigs of all production stages and that the viral infection rate increases with the age of the animals. In the western region of Paraná state, Brazil, TTSuV1 and TTSuVk2-induced infections were more frequently observed in suckling piglets and finisher pigs, respectively. Phylogenetic analysis pointed out the possibility of different strains of TTSuV1 and TTSuVk2 circulating in pig herds of Brazil.


A infecção pelo Torque teno sus virus (TTSuV) está presente em rebanhos suinícolas em todo o mundo. Tem sido demonstrado que a infecção pelo TTSuV pode aumentar a gravidade de outras importantes doenças virais com impactos econômicos e na saúde pública. Atualmente não há informações sobre a distribuição da infecção pelo TTSuV, de acordo com a faixa etária, em rebanhos suinícolas brasileiros. Este estudo avaliou a frequência da infecção pelo TTSuV nas diferentes categorias de produção de suínos. Amostras fecais (n=190) de suínos com 1 a 24 semanas de idade e de reprodutores provenientes 6 unidades produtoras de leitão (até 8 semanas de idade) e 9 unidades de terminação (9 semanas de idade em diante) da região oeste do Paraná, Brasil, foram avaliadas pela técnica de PCR. Fragmentos da região 5' UTR do DNA do TTSuV1 e/ou TTSuVk2 foram identificados em 126 (66,3%) amostras fecais. Diferenças significativas foram encontradas em relação às porcentagens de amostras positivas para o TTSuV1, TTSuVk2 e infecção mista por ambos os gêneros inter e intra categorias. Amostras fecais provenientes de unidades de terminação apresentaram taxas de detecção de TTSuV (90.1%; 64/71) significativamente (p<0.05) mais altas do que aquelas provenientes de unidades produtoras de leitão (52.1%; 62/119). A detecção do TTSuV em animais de terminação foi significativamente (p<0.05) mais frequente do que nos suínos de outras categorias. As sequências de nucleotídeos da UTR deste estudo apresentaram maior similaridade com cepas da Noruega (96%, TTSuV1) e Argentina e China (97,1%, TTSuVk2). Estes resultados sugerem que a infecção pelo TTSuV encontra-se disseminada em suínos de todas as categorias de produção e que a taxa da infecção viral aumenta de acordo com a idade dos animais. Na região oeste do estado do Paraná, infecções induzidas pelo TTSuV1 e TTSuVk2 foram mais frequentemente observadas em leitões de maternidade e suínos de terminação, respectivamente. A análise filogenética indicou a possibilidade de diferentes cepas de TTSuV1 e TTSuVk2 circulando em rebanhos suinícolas brasileiros.

In order to verify the microbial quality of the influents and effluents of one STP from southern Brazil, an eight-month survey was conducted to examine the presence of total and fecal coliforms and of adenovirus (HAdV), enterovirus (EV), genogroup A rotaviruses (GARV) and Torque teno virus (TTV), in treated effluent samples from São João/Navegantes STP, Porto Alegre (Brazil). A total of 16 samples were collected, eight of influent (raw sewage, prior to treatment), and the other eight of the effluent (post-treatment sewage). Total and fecal coliform levels ranging from 3.6 × 10(4) to 4.4 × 10(7) MPN/100 mL and 2.9 × 10³ to 1.7 × 10(7) MPN/100 mL, were detected in all samples. In raw sewage, HAdV (25%) and GARV (28.6%) viral genomes were detected. The analysis of effluent samples revealed the presence of HAdV (50%), EV (37.5%), and TTV (12.5%) genomic fragments. All samples, regardless of the month analysed, presented detection of a least one virus genus, except for in April. Higher virus detection rates were observed in treated sewage samples (62.5%), and in 80% of them (effluent positive samples) HAdV was detected. Results showed that improvements in sewage monitoring and treatment processes are necessary to reduce the viral and bacterial load on the environment in southern Brazil. To the knowledge of the authors, this is the first study showing the monitoring of viral genomes in influent and effluent samples from a STP located in Porto Alegre (Rio Grande do Sul, Brazil), southern Brazil.


Com o intuito de verificar a qualidade microbiológica de afluentes e efluentes de uma estação de tratamento de esgoto (ETE), um monitoramento de oito meses foi realizado para examinar a presença de coliformes totais e fecais, e de adenovírus (HAdV), enterovírus (EV), rotavírus do genogrupo A (GARV) e torque teno vírus (TTV), em amostras de esgoto tratado da ETE São João/Navegantes, em Porto Alegre-RS, Brasil. Um total de 16 amostras foi coletado, sendo oito de afluente (esgoto bruto, anterior ao tratamento) e oito de efluente (esgoto tratado). Os níveis de coliformes totais e fecais variaram entre 3,6 × 10(4) e 4,4 × 10(7) MPN/100 mL e 2,9 × 10³ e 1,7 × 10(7) MPN/100 mL, respectivamente, tendo sido estes detectados em todas as amostras. No esgoto bruto, foram detectados os genomas virais de HAdV (25%) e GARV (28,6%). A análise das amostras de efluente revelou a presença de fragmentos genômicos de HAdV (50%), EV (37,5%) e TTV (12,5%). Todas as amostras, independentemente do mês analisado, possibilitaram a detecção de pelo menos um gênero viral, exceto no mês de abril. Altas taxas de detecção viral foram observadas em amostras de esgoto tratado (62,5%), sendo que o HAdV foi detectado em 80% dessas amostras de efluente positivas. Os resultados mostram que aprimoramentos no processo de tratamento e monitoramento do esgoto são necessários para reduzir a carga viral e bacteriológica no ambiente do Sul do Brasil. Ao conhecimento dos autores, este é o primeiro estudo de monitoramento de genomas virais em amostras de afluente e efluente de uma ETE localizada em Porto Alegre-Rio Grande do Sul, Brasil.

Background: SEN virus (SEN-V) and TT virus (TTV) have been classified in the circoviridae family. Both are single-stranded, non-enveloped DNA viruses of about 3800 nucleotides. Patients on maintenance hemodialysis (HD) have a high risk of blood-borne viral infections. SEN-V and TTV has been reported from a number of HD units from various countries throughout the world. Materials and Methods: A total of 377 blood samples obtained from 150 healthy donors and 227 HD patients were collected at the HD center. SEN-V and TTV DNA was determined by polymerase chain reaction (PCR) in all samples. Results: TTV was detected in 109 (48.01%) of 227 hemodialysed patients and 14 (9.33%) of 150 voluntary blood donors (significant, P < 0.05). The PCR results for SEN-V-D/H DNA showed that 65 (28.63%) were positive for SEN-V-D and 33 (14.53%) were positive for SEN-V-H. 9.69% of 227 patients were positive for SEN-V-D/H co-infection. In the control group, SEN-V-D was detected in 14 (9.33%) and SEN-V-H was detected in 15 (10%) of the 150 (100%) blood donors. Conclusion: These findings show that the prevalence of SEN-V-D/H and TTV is higher than healthy blood donors. Also, these results indicate that the prevalence of SEN-V and TTV infections in our region is similar with that in other countries.

This study investigates the frequency of Torque teno virus (TTV) infection in 150 blood donors and 77 patients requiring haemodialysis in southern Brazil. Plasma samples were screened for TTV DNA using polymerase chain reaction (PCR). The prevalences of TTV among blood donors and patients requiring haemodialysis were 73.3% and 68.8%, respectively. The presence of TTV was correlated with age in the blood donors (p = 0.024). In haemodialysis patients, no association was found between TTV infection and the demographic parameters (age, sex and education), the duration of haemodialysis or a history of blood transfusion. This study is the first to evaluate the prevalence of TTV infection in Brazilian patients requiring haemodialysis.

Adenovirus (AdV), enterovirus (EV), genogroup A rotaviruses (GARV) and Torque teno virus (TTV) are non-enveloped viral agents excreted in feces and so may contaminate water bodies. In the present study, the molecular detection of these viruses was performed in samples of surface water collected from the Arroio Dilúvio, a waterstream that crosses the city of Porto Alegre, RS, Brazil, receiving great volumes of non-treated sewage from a large urban area. Sampling was performed during 2009, in three different occasions (January, April and September). The highest detection rate was observed for EV (64.28%), followed by TTV (28.57%) and AdV (21.43%). Rotaviruses were not detected. More than on kind of tested virus was detected in five (35. 71%) of 14 samples. January was the month with the highest viral detection rate, being all samples, collected in this month, positive for at least one group of tested virus. The correlation between the detection of these different viral agents and environmental factors is discussed. To the knowledge of the authors, this is the first description of viral genomes in water samples taken from the Arroio Dilúvio, Porto Alegre (Brazil).


Adenovírus (AdV), enterovírus (EV), rotavírus (GARV) e Torque teno vírus (TTV) são vírus não envelopados, excretados nas fezes, podendo, assim, contaminar corpos hídricos. No presente estudo, a detecção molecular desses agentes foi realizada em amostras de águas superficiais provenientes do Arroio Dilúvio, o qual cruza a cidade de Porto Alegre-RS, Brasil. As amostras foram coletadas em três meses diferentes (janeiro, abril e setembro) do ano de 2009. A maior taxa de detecção viral foi observada para EV (64,28%), seguida por TTV (28,57%) e AdV (21,43%). Rotavírus não foi detectado. Foi verificada presença simultânea de dois grupos virais em cinco (35,71%) das 14 amostras analisadas. Janeiro foi o mês com a maior taxa de detecção viral, sendo todas as amostras, coletadas nesse mês, positivas para, no mínimo, um grupo viral em estudo. A correlação entre a detecção desses diferentes agentes virais e os fatores ambientais é discutida. Conforme conhecimento dos autores, essa é a primeira descrição de genomas virais em amostras de água provenientes do Arroio Dilúvio, Porto Alegre, Brasil.

This study evaluated histological lesions in kidney samples from pigs with nephritis in two slaughterhouses in the State of Mato Grosso, Brazil. Four hundred samples were subjected to histology, anti-porcine circovirus type 2 (PCV2) immunohistochemistry (IHC), anti-Leptospira sp. immunofluorescence (IF), and polymerase chain reaction (PCR) for PCV2, porcine parvovirus (PPV), and Torque teno virus type 1 and 2 (TTV1, TTV2) detection. Histological lesions were found in 81% of the samples, and mononuclear interstitial nephritis was the most frequent lesion (77.50%). A follicular pattern was observed in 40.97% of the interstitial nephritis lesions. PCV2, PPV, TTV1, and TTV2 were identified in the kidneys by PCR in 27.25%, 28.50%, 94%, and 87.5% of the samples, respectively. Leptospira sp. was not detected through IF. Infection by PCV2 (PCR) and the presence of histological lesions (P=0.008) and giant cells (P=0.0016) were significantly associated. An association was observed between the TTV2-TTV1 co-infection (P<0.0001) and the risk for pathogenesis. These findings indicated that PCV2, PPV, TTV1, and TTV2 were widely distributed among pigs in the local farms and that the presence of these agents should be considered in the differential diagnosis of kidneys with interstitial nephritis in pigs.


O propósito desse estudo foi avaliar as lesões histológicas observadas em rins condenados por nefrite pelo Serviço de Inspeção Federal, em dois frigoríficos de Mato Grosso, Brasil. Foram coletados 400 rins condenados por nefrite e submetidos aos exames de histologia, imuno-histoquímica (IHC) para Circovirus suíno Tipo 2 (PCV2), imunofluorescência direta (IF) para Leptospira sp. e reação em cadeia pela polimerase (PCR) para detecção de PCV2, Parvovirus suíno (PPV) e Torque teno vírus Tipo 1 e 2 (TTV1 e TTV2). Foram observadas lesões histológicas em 81% das amostras, sendo nefrite intersticial mononuclear a mais freqüente (77,50%). Das lesões de nefrite intersticial encontradas, 40,97% apresentaram padrão folicular. Através da PCR foi observada ampla distribuição dos agentes (PCV2, PPV, TTV1 e TTV2) nas propriedades e municípios, com ocorrência de 27,25%, 28,50%, 94% e 87,50%, respectivamente. Leptospira sp. não foi detectada através da IF. Houve associação significativa da infecção do PCV2 com presença de lesão histológica (P=0,008) e de células gigantes (P=0,0016). Também houve associação entre a co-infecção TTV2 e TTV1 (P<0,0001). Esses achados indicam que os vírus PCV2, PPV, TTV1 e TTV2 devem ser considerados no diagnóstico diferencial de rins com nefrite intersticial em suínos.

Estudos recentes sobre o torque teno vírus (TTV), gênero Anellovirus, permitiram construir a hipótese de que esse vírus pode ser um desencadeante ou tenha algum papel patogênico nas doenças reumáticas autoimunes. OBJETIVOS: Verificar a frequência da infecção pelo TTV em pacientes com lúpus eritematoso sistêmico (LES), e sua diversidade gênica, a existência de correlação entre a infecção pelo TTV e as manifestações clínicas do LES, sua evolução clínica e o perfil sorológico. PACIENTES E MÉTODOS: Foram obtidas 46 amostras de soro de pacientes com LES atendidos no Ambulatório de Reumatologia do Hospital Universitário de Campo Grande (NHU/FAMED/UFMS). Para os controles, utilizaram-se 46 amostras de soro de doadores de sangue. O DNA viral foi extraído das amostras utilizando o QIAamp DNA Blood Mini Kit (QIAGEN, Hilden, Alemanha), e amplificado utilizando a técnica de nested PCR. RESULTADOS: Foi encontrada positividade para o TTV em 17 (37 por cento) dos pacientes lúpicos, e em apenas sete (15,2 por cento) dos controles (teste z, P = 0,03). Não houve correlação entre a infecção pelo TTV, as manifestações clínicas, o perfil sorológico e a evolução clínica dos pacientes avaliados neste estudo. CONCLUSÃO: A presença do TTV nos pacientes com LES necessita ser mais bem compreendida a partir deste estudo inicial.


Recent studies on the torque teno virus (TTV), genus Anellovirus, have allowed formulating the hypothesis that TTV may trigger autoimmune rheumatic diseases or have some pathogenic role in them. OBJECTIVES: To determine the frequency of TTV infection in patients with systemic lupus erythematosus (SLE), the genetic diversity of TTV, the correlation between TTV infection and SLE clinical manifestations, and SLE clinical course and serological profile. PATIENTS AND METHODS:Serum samples were obtained from 46 SLE patients treated at the University-Affiliated Hospital of Campo Grande (NHU/FAMED/UFMS), Brazil. For controls, serum samples were obtained from 46 healthy volunteer blood donors. Viral DNA was extracted from samples using the QIAamp DNA Blood Mini Kit (QIAGEN, Hilden, Germany) and amplified using nested PCR. RESULTS: Positivity for TTV was found in 17 (37 percent) of SLE patients and in only seven (15.2 percent) of the controls (z test, P = 0.03). There was no correlation between TTV infection, SLE clinical manifestations, SLE clinical course, and the serological profile of the patients evaluated. CONCLUSION: Further studies on the presence of TTV in SLE patients are required.

OBJECTIVE@#To analyze the sequence of ORF1 protein of Torque teno virus to prepare for the future hybrid experiments.@*METHODS@#The sequence of ORF1 protein of Torque teno virus was analyzed by bioinformatics using some web tools.@*RESULTS@#The most likely cleavage site was between position 14aa and 15aa and signal peptide may be position 1aa-14aa. Two possible transmembrane helices from inside to outside and three possible transmembrane helices from outside to inside were found. The position 509 (NKTN) was the potential N-glycosylation site. The speculative molecular weight of TTV ORF1 protein, which may be a kind of unstable protein was 88 705.7 Da. 1aa-91aa and 278aa-361aa were localized in non-regular secondary structure region.@*CONCLUSIONS@#TTV ORF1 protein may be a nuclear protein which contains two non-regular secondary structure region. 265aa to 486aa and 510aa to 679aa may be the two approciate fragments to construct the plasmids, which would be prepared for the future hybrid experiments to study the functional positions of the protein and the interactions between TTV and its hosts. Bioinformatics analysis would possibly make it easier to study the protein's function.

BACKGROUND: Although a marked proportion of thalassemic patients acquire Torque teno virus (TTV) through blood transfusion, its clinical importance is unclear. This study was designed to investigate the clinical importance of TTV infection in thalassemic patients with and without hepatitis C virus (HCV) co-infection in Iran. METHODS: In this case-control study, 107 thalassemic patients on chronic transfusion and 107 healthy individuals were selected. According to HCV and TTV infection status (detected by semi-nested PCR), patients were categorized into 4 groups: TTV and HCV negative, TTV positive, HCV positive, and TTV and HCV positive. Blood ferritin, alanine aminotransferase (ALT), and aspartate aminotransferase (AST) levels in these 4 groups were assessed. RESULTS: Approximately half of the thalassemic patients (50.5%) and 27.1% of controls had TTV infection. Thalassemic patients had a greater chance of TTV infection compared to the control group with a sex-adjusted OR of 4.13 (95% CI=2.28-8.13). The increased levels of ALT, AST, and ferritin in the TTV and HCV-infected group were not significantly different from those in the TTV and HCV negative group. Co-infection with TTV and HCV did not significantly increase ALT, AST, and ferritin levels compared to infection with TTV alone. CONCLUSION: Although common in thalassemic patients, TTV infection appears to have a negligible role in increasing the severity of liver disease, even when co-infection with HCV occurs.

O Torque teno vírus (TTV) é um vírus de DNA do gênero Alphatorquevirus da família Anelloviridae. O TTV é altamente prevalente em populações de todo o mundo. Isolados foram classificados em cinco grupos filogenéticos (1-5) com grande distancia genética entre eles. A presença do TTV já foi detectada nas fezes, porem não se sabe se todos os cinco genogrupos do TTV são excretados nas fezes, e qual a distribuição do TTV entre os genogrupos. A fim de avaliar a presença, a diversidade genomica e a carga viral do TTV em fezes, 135 amostras de pacientes com gastroenterite foram analisadas. O DNA do TTV foi extraído de suspensão fecal e três diferentes métodos de reação em cadeia da polimerase (PCR), dois qualitativos e um quantitativo, foram avaliados. Nas amostras de fezes estudadas, 123 (91,1 porcentos) foram positivas em pelo menos um dos tres métodos. O DNA do TTV percentente aos genogrupos de 1 a 5 foi detectado em 37 (27,4 porcentos), 27 (20,0 porcentos), 57 (42,2 porcentos), 39 (21,5 porcentos) e 33 (24,4 porcentos) amostras, respectivamente. Co-infecções com dois, três, quatro e cinco genogrupos do TTV foram encontradas em 23 (12,0 porcentos), 15 (11,1 porcentos), 7 (5,2 porcentos) e 7 (5,2 porcentos) amostras fecais, respectivamente. Assim, 52 (38,5 porcentos) amostras continham mais de um genogrupo de TTV. A carga viral variou de 2,6 a 6,5 log de genoma equivalentes por grama de fezes. No entanto, variações de carga viral foram observadas em função do genogrupo detectado e do numero de genogrupos presentes simultaneamente. Os resultados encontrados são os primeiros a mostrar a alta prevalência e a diversidade de TTV nas fezes humanas.

Torque teno virus (TTV), a novel DNA virus resides in peripheral blood mononuclear cells and replicates when these cells get activated. The TTV replication shifts the immunobalance. Aim: To determine the presence of TTV in the gingiva of patients with aggressive periodontitis, patients withchronic periodontitis, and healthy controls, and to correlate the presence of TTV with probing pocket depth and clinical attachment level. Methods: Forty-two subjects (22 males and 20 females)aged 21 to 55 years were recruited for this study. Subjects were stratified into aggressive periodontitis (Group I), chronic periodontitis (Group II) and healthy controls (Group III). Gingival tissue biopsy was taken from all the subjects and the presence of TTV was analyzed using PCR and 2% agarose gel electrophoresis. Results: TTV was identified in half of the subjects and more number of subjects with periodontitis have TT virus compared to controls. There was significant association between presence of TT virus and pocket depth, clinical attachment level. Conclusions: The findings from the present study shows that there was no significant association between TT virus and periodontitis, even though it was isolated from more number of subjects with aggressive periodontitis, and TTV was associated with pocket depth and clinical attachment level. These findings need to be investigated in further studies.

Background & objectives: TT virus (TTV) is a newly discovered non-enveloped, single stranded DNA virus of high genotypic variability, found frequently in patients with acute or chronic hepatitis of non A-G aetiology. This study was carried out to look for the presence of TTV and its genotypes in patients with different types of liver diseases from northern India. Methods: A total of 262 serum samples from patients of acute viral hepatitis (AVH; n=72), fulminant hepatic failure (FHF; n=49), chronic active hepatitis (CAH; n=93) and liver cirrhosis (LC; n=48), were analyzed for hepatitis A-G viral markers. TTV DNA was detected in all cases by nested polymerase chain reaction (PCR) using the primers from N22 and untranslated (UTR) region. TTV-DNA was also tested in 150 volunteer blood donors. Direct nucleotide sequencing of N22 amplicons were carried out to look into the prevalent TTV genotypes. Results: TTV-DNA was detected in 73.6, 59.2, 21.5 and 29.1 per cent cases with AVH, FHF, CAH and cirrhosis, respectively. In AVH and FHF groups, TTV showed co-infection with all A-G hepatitis cases whereas in CAH and cirrhosis groups, TTV co-infection observed with HBV, HCV and HGV. TTV-DNA was detected in 45.3 per cent volunteer blood donors. No statistically significant difference was observed amongst the mean liver function profile of UTR PCR positive and negative cases in different liver disease groups except AVH cases, in whom the various biochemical parameters between TTV positive and TTV negative patients were marginally significant. However, no significant evidence of biochemical or histological deterioration of the liver was observed in TTV positive cases amongst FHF, CAH and cirrhosis. Predominance of genotype 1a was observed in all the cases from north India. Interpretation & conclusions: TTV is a frequent virus isolated from patients with various types of liver diseases as well as in healthy individuals from northern India. TTV has no effect on biochemical markers of associated liver diseases. Genotype 1a was the most predominant type in different liver disease groups. The occurrence of TTV did not further influence the course of the disease.

RESUMO

The aim of this study is to present the status of transfusion –transmitted infections among the apparently healthy donors so as to increase the awareness of complications of blood transfusion and make the clinicians more vigilant with regard to judicious use of blood. A total of 5008 units of donor’s blood was screened from January 2006 to December 2008 at blood bank of People’s Hospital, Bhopal. The result of screening showed total seropositive samples for hepatitis B (2.9%), hepatitis C (0.57%), human immunodeficiency virus (HIV; 0.51% ) and Venereal Disease Research Laboratory Test (VDRL; 0.23%) by using enzyme linked immunosorbent assay (ELISA ) methods, and rapid plasma regain (RPR) method for syphilis.

BACKGROUND: We analyzed the results of external proficiency tests for HBsAg, anti-HCV and anti-HIV, and these tests are currently used for blood donor screening in Korea. METHODS: The external proficiency testing (EPT) data was retrospectively collected from 2001 to 2009 from the Korean Association of Quality Assurance for Clinical Laboratories, and this association includes those institutes that have blood centers. The pooled patient sera or converted sera from the plasma were used for EPT. The year 2004 data includes the Recombinant HBsAg variant (Gly/Arg 145). RESULTS: A total 806 institutes participated in this evaluation. The average discordant rate for HBsAg, anti-HCV and anti-HIV in the blood centers was 0.2%, 0.6% and 1.0%, respectively for the years 2001~2009. For the HBsAg test, the discordant rate was less than 0.5% in 2009, yet there was a much higher rate (1.6~22.2%) in 2004 when using the recombinant HBsAg variant. CONCLUSION: HBV variants or low positive antigen or antibody titers were problematic in the current clinical laboratory, and so a systematic quality assurance program should be conducted with using control materials.
