ABSTRACT

Acute hepatic injury is a common liver disease in clinical practice. Drugs with antioxidant activity exhibit a great potential for alleviating liver injury. The present study aimed to explore the role of rosiglitazone (RSG), a previously reported compound with anti-inflammatory properties, in hepatic injury. Kunming mice were divided into the following four groups: The control group; the RSG group; the carbon tetrachloride (CCl4) group; and the RSG + CCl4 group. Hepatic injury was confirmed by histological examination of the liver. In addition, the serum levels of alanine transaminase (ALT) and aspartate transaminase (AST), and those of the biochemical indices superoxide dismutase (SOD), catalase (CAT), glutathione (GSH), malondialdehyde (MDA), NO and reactive oxygen species (ROS) were measured in each group of mice. Additionally, the levels of inflammatory factors and apoptosis-related proteins, as well as the activity of the related signaling pathways, were evaluated. The results showed that RSG could reverse the CCl4-mediated decrease in the levels of SOD, CAT and GSH, and increase in the levels of ALT, AST, MDA, NO and ROS. Furthermore, treatment with RSG could reduce the expression levels of inflammation- and apoptosis-related proteins, thus suggesting that RSG could attenuate inflammation and liver cell apoptosis. Additionally, treatment with RSG promoted the activation of the nuclear factor erythroid 2-related factor 2 (Nrf2) signaling pathway, upregulated peroxisome proliferator-activated receptor γ and inhibited activation of the inflammasome NOD-like receptor protein 3 (NLRP3). In conclusion, the current study demonstrated that RSG could ameliorate acute hepatic injury via activating the Nrf2 signaling pathway and inhibiting activation of the NLRP3 inflammasome. The findings of the present study partly uncovered the mechanism underlying the effect of RSG on hepatic injury, thus supporting the application of RSG in clinical practice.

ABSTRACT

There are various respiratory tract complications in patients undergoing general anesthesia, with postoperative sore throat (POST) being the most commonly seen. Although measures have been taken to prevent and treat POST in clinical practice, the control of POST is still not satisfactory. In this study, 880 ASA patients with grade I to II general anesthesia were randomly assigned into control group and experimental group. After patients entered into the operating room, the plasters were applied to the designated points (Tianzhu, Lianquan, Dazhui, etc), and the clinical efficacy of acupoint application in prevention and treatment of respiratory tract complications after general anesthesia was observed. The results showed that patients starting using acupoint application before operation could significantly reduce the incidence of postoperative respiratory tract complications, and the effects lasted for up to 24 hours. In this study, acupoint application was used, providing a simple, safe, efficient, and durable approach to prevent and treat respiratory tract complications after operation under general anesthesia.

ABSTRACT

Objective To investigate the characteristics and types of die-punch fracture of the distal radius (DPFDR) based on three-column theory as well as its application value.Methods The imaging data of 560 cases of DPFDR were reviewed which had been treated at The Ninth People's Hospital of Wuxi from June 2007 to June 2017.They were 305 males and 255 females,aged from 13 to 95 years,with an average of 46.4 years.According to the three-column theory,the 560 DPFDR cases were divided into a single column group and a double column group.According to the fracture site and morphology,the former was further characterized as volar,dorsal,split and collapse types while the latter as metaphyseal,articular surface and mixed types.After being familiarized with our new classification,2 radiologists were asked to classify 100 cases of DPFDR independently.The inter-and intra-observer agreements were analyzed with Kappa statistics.Results There were 65 cases of single column DPFDR,including 3 ones of volar type,13 ones of dorsal type,14 ones of split type and 35 ones of collapse type;there were 495 cases of double column DPFDR,including 130 ones of metaphyseal type,155 ones of articular surface type and 210 ones of mixed type.The intra-observer Kappa coefficient was 0.877-0.937 and the intra-observer kappa was 0.916-0.959,showing high agreement.Conclusions Due to differences in violence nature and intensity,wrist position and bony quality of the patient at the time of injury,axial violent transmission may cause different types of die-punch fracture at the intermediate column of the distal radius and different types of mild radial column fractures as well.As our new classification may well characterize the site and morphology of DPFDR,it has high consistency to guide surgical treatment and prognosis judgment.

ABSTRACT

Objective To evaluate the results of Four-Step methods in traditional Chinese medicine Course.Methods 40 five-academic-year clinical medical students and 40 seven-academicyear undergraduate students were randomized assigned into experimental group (n=40, 20 five-academicyear clinical medical students and 20 seven-academic-year students) and control group (n=40, 20 five-academic-year students and 20 seven-academic-year students).The control group received conventional teaching, while four-step teaching break the gap between Chinese medicine and western medicine, build the base, form the framework of Chinese medicine and extend knowledge was implemented in the experimental group.Through questionnaire score, group discussion, the final evaluation of the theory and clinical training evaluation, the learning of the experimental group students and the control group students was compared.SPSS 18.0 statistical software was used and t test was employed to make analysis of the relevant data.Results The questionnaire at the end of the semester showed that the average score of students in the experimental group (36.18 ± 0.96) was higher than that in the control group (26.62 ± 1.00), and the difference was statistically significant (P=0.017).In the discussion at the end of the semester, the experimental group paid more attention to the theory and technology of traditional Chinese medicine's learning persistence in future and its application in their professional practice.Scores in theory test were higher in seven-academic-year students in experimental group (88.40 ± 8.79) than in control group (82.15 ± 6.83) and the difference was statistically significant (P=0.017), but there was no difference in five-academic-year students between experimental group (75A0 ± 12.09) and control group (77.85 ± 11.99), which was not considered statistically significant different (P=0.524).In clinical practice, the rate of outstanding students in the experimental group of seven-academic-year was five percentage points higher than the control group, while the rate of outstanding students in the experimental group of five-academic-year was 10 percentage points higher than the control group.Conclusions Four step teaching is helpful to stimulating the learning enthusiasm of the students of traditional Chinese medicine, expanding students' vision of medical field, and promoting their construction of a more comprehensive medical knowledge framework.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the association of the HLA-DRB1 polymorphism with susceptibility to myelodysplastic syndrome (MDS) and aplastic anemia (AA) in Chinese Han population.</p><p><b>METHODS</b>The polymorphism of HLA-DRB1 alleles in 242 patients with MDS, 115 patients with AA and 2264 umbilical cord blood control samples were tested by polymerase chain reaction-sequence specific primer (PCR-SSP).</p><p><b>RESULTS</b>Compared with normal controls, the frequency of HLA-DRB1*15 was significantly increased in the MDS group and AA group (22.93% vs. 17.25%, Chi-square = 9.662, OR= 1.428, P= 0.003; 26.52% vs. 17.25%, Chi-square = 12.924, OR= 1.732, P= 0.001). And this is mainly due to the increase in the male patients in both patient groups: in the MDS males, 24.68% vs. 17.25%, Chi-square= 11.194, OR= 1.572, P= 0.001, and in the AA males, 29.29% vs. 17.25%, Chi-square= 13.563, OR= 1.987, P= 0.001. No significant difference between the controls and the female patients in both groups was observed.</p><p><b>CONCLUSION</b>The HLA-DRB1*15 could be a susceptibility allele for MDS and AA male patients.</p>

ABSTRACT

Objective: The aim was to explore the association between the HLA-DRB1 allele polymorphism and susceptibility to chronic myelogenous leukemia(CML)in Chinese Han population.Methods: The polymorphism of HLA-DRB1 alleles in 762 CML patients and 2 264 normal umbihcal cord blood samples were tested by polymerase chain reaction with sequence specific primer(PCR-SSP).Results: The most common allele in DRB1 locus in normal population was DRB1 * 15(17.25%), whereas DRB1 * 10 was the least frequent allele(1.39%).Compared with normal controls, the frequency of HLA-DRB1 * 08 was significantly increased in all CML groups(7.48%vs 5.39%, X~2 =8.963,OR= 1.023, P =0.004)and CML male groups(7.72%vs 5.39%,X~2 = 8.059,OR= 1.025,P = 0.007).But there was no significant difference between the normal controls and CML female patients.Conclusion: It should be concluded that DRB1 * 08 could be considered a susceptible allele for CML male patients.

ABSTRACT

<p><b>OBJECTIVE</b>To observe the changes of serum testosterone after allotransplantation of Leydig cells in rats.</p><p><b>METHODS</b>Leydig cells were isolated from the testes of SD rats with the Percoll technique and serum testosterone of the receptors was determined once a month for 3 successive times.</p><p><b>RESULTS</b>After allotransplantation of Leydig cells, the serum testosterone level of the receptors increased gradually, significantly higher at 3 months than that of the normal rats younger than 2 months old.</p><p><b>CONCLUSION</b>Allotransplantation of Leydig cells has a promising application value in the treatment of male primary hypogonadism.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the chemical constituents of unsaponifiable matter from the seed oil of Trichosanthes hupehensis.</p><p><b>METHOD</b>The fatty oil from the seeds of T. hupehensis was extracted with petroleum ether. The saponification was carried out with potassium hydroxide. The unsaponifiable matter was isolated and purified by silica gel column chromatography. Their structures were elucidated by means of MS, IR, UV, and 1H-NMR.</p><p><b>RESULT</b>Karounidiol, isokarounidiol, 5-dehydrokarounidiol, 7-oxodihydrokarounidiol, stigmast-7-en-3beta-ol, stigmast-7, 22-dien-3beta-ol, 10alpha-Cucurbitadienol, beta-sitosterol, stigmast-7, 22-dien-3beta-O-beta-D-glucoside were elucidated.</p><p><b>CONCLUSION</b>All of these compounds were found in this plant for the first time.</p>

ABSTRACT

This study was aimed to investigate the corelation between the HLA (human leukocyte antigen) genes and susceptibility of leukeamia. 605 patients with leukeamia including 189 ALL, 184 AML and 232 CML were selected for this investigation. 900 normal umbilical cord blood samples from umbilical cord blood bank were used as control population compared to the leukemia patients. HLA-A, B, C typing was done by polymerase chain reaction with sequence-specific primers (SSP-PCR). The results showed that frequencies of HLA-A*26, A*68, B*56 in ALL patients were higher (4.46%, 2.65%, 1.17%), as compared with controls (2.31%, 0.95%, 0.22%), HLA-CW*06 in ALL patients was lower (3.64%), as compared with control (11.65%). In AML patients HLA-A*01 (9.41%), B*37 (3.60%) was higher and A*33 (3.60%), B*51 (4.73%) were lower than those in controls (3.57%, 1.75% and 7.64%, 7.93%). HLA-A*32, B*27, B*44, B*54, B*55 (2.18%, 3.96%, 5.06%, 4.63%, 2.84%) in CML patients were higher than those in control (0.84%, 2.04%, 3.07%, 2.44%, 1.29%). These results suggested that positive association may exist between certain HLA-class I genes and leukemias. These preliminary data may be useful for further study on the mechanisms of leukemia pathogenesis.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate at the DNA level the polymorphism of HLA-DRB1 gene in Han population in Beijing of China.</p><p><b>METHODS</b>This study was conducted in the Hans in Beijing. A total of 2810 normal and unrelated neonates were analyzed and subjected to HLA-DRB1 low resolution typing, and from among them 217 neonates were randomly selected for DRB1 high resolution typing by polymerase chain reaction-sequence specific primers technique.</p><p><b>RESULTS</b>Fourteen HLA-DRB1 specificity and 39 alleles were found with HLA-DRB1 *09012, *1501, *07011, *08032 as the most common genes. Their gene frequencies and allelic distributions were compared to those observed in other ethnic populations.</p><p><b>CONCLUSION</b>The results showed the characteristic of HLA-DRB1 distribution, and provided more comprehensive and accurate gene data that may serve as normal reference values for the population in Beijing, China.</p>

ABSTRACT

Objective To study the clinical characteristics and curative effects of pancreatic cystade- nocarcinoma in order to improve its diagnostic and therapeutic accuracy.Methods A retrospective analysis was done on the clinical materials of 13 cases of pancreatic cystadenocarcinoma hospitalized in Shanxi Cancer Hospital from 1990 to 2006.Results The preoperative diagnosis were as follows:pancreatic cystadenocarci- noma 6 cases,pancreatic cystadenoma 2 cases,pancreatic cancer 1 case,pancreatic pseudocyst 4 cases.The misdiagnosis rate was 53.8 %.Surgical operation was done on the 13 cases,and 10 of them were treated by radical operation.A 5-year follow-up was done on 6 still alive cases,and 1 of them lived over 11 years.3 cases were treated by palliative operation,and all of them died within 3 years.Conclusion Since there is no specific clinical manifestations of pancreatic cystadenocarcinoma,it is very difficult to get an accurate preop- erative diagnosis.Radical operation is the most effective therapeutic methods.
