ABSTRACT

OBJECTIVE: To observe the effects of acupuncture and moxibustion therapy on pain relief in sciatica rats and to explore the mechanism of its anti-inflammatory effect. METHODS: SPF grade 4-6-week-old Kunming rats were randomly divided into 5 groups including a blank group, sham-operated group, model group, acupuncture, and moxibustion (AnM) group, and positive group. A total of 10 rats were included in each group. The model group, the AnM group, and the positive group were prepared by ligating the left sciatic nerve. AnM group was used for acupuncture and moxibustion therapy intervention, and the positive group was rendered to quick-acting sciatica pills once a day for 7 days (3 courses of treatment). The blank group, sham-operated group, and model group were not treated. The changes in thermal and mechanical pain thresholds were observed before and after the operation, and the morphological changes of the dorsal horn of the spinal cord in the lumbosacral region of the rats in each group were observed by HE staining after the courses of treatment finished. The contents of IL-1ß, IL-6, IL-18, and TNF-α were measured by ELISA and the expressions of NOX1, NOX2, NOX4, and NLRP3 genes were detected by RT-qPCR while the protein expressions of NOX1, NOX2, NOX4 and NLRP3 were analyzed by Western blotting. RESULTS: The AnM and positive group showed a significant increase in thermal and mechanical pain thresholds after treatment, while there was no significant change in the model group. As compared to the control group, the contents of IL- 1ß, IL-6, IL-18, and TNF-α, as well as the relative expressions of NOX1, NOX2, NOX4, and NLRP3 genes were significantly increased in the model group (P < 0.05 or P < 0.01). As compared to the model group, the contents of IL-1ß, IL-6, IL-18, and TNF-α, as well as the relative expressions of NOX1, NOX2, NOX4, and NLRP3 genes significantly decreased in the AnM and positive groups (P < 0.05 or P < 0.01). The pathological changes of inflammatory infiltration of tissue cells in the dorsal horn of the lumbosacral spinal cord were slowed in the AnM group. CONCLUSION: Acupuncture and moxibustion therapy have a positive effect on pain relief and anti-inflammatory effects in CCI sciatica rats, which may point to the regulation of NOX1, NOX2, NOX4, and NLRP3 expressions, and inhibition of ROS.

ABSTRACT

Objective To establish a rapid and efficient method for the simultaneous determination of ginsenoside Rg1 and ginsenoside Re in Tongru granules by HPLC.Methods The HPLC analysis was carried out with Agilent ZORBAX SB-C18 and acetonitrile-0.1% phosphoric acid(15:85,V/V) as mobile phase,the flow rate was 1.0 ml/min,the column temperature was 30 ℃,injection volume was 10 μl,and the detection wavelength was 203 nm.Results The Rg1 solution of ginsenoside showed a good linear relation between 0.136-2.040 μg (r2=0.999 9).The ginsenoside Re solution presented a good linear relationship (r2=0.999 8)between 0.066-0.990 μg,with the average addition recovery of 99.3% and 99.1%,and the relative standard deviation of 0.84% and 0.75%(n=6),respectively.Conclusions The method is highly specific,sensitive,reproducible,simple and efficient which can be used for quality control of Tongru Granules.

ABSTRACT

Objective To analyze the multi-component,multi-target and multi-pathway mechanism of peony leaf in the treatment of cervical cancer based on network pharmacology. Methods The possible active components and targets in peony leaf were screened and predicted by pharmacological database and analysis platform of traditional Chinese medicine system,and the related targets of cervical cancer diseases were ob-tained by searching Therapeutic Target Database and others. The potential targets for the disease regulation were screened according to the active components of peony leaves entering blood,then the key target names and the pathways involved in the treatment of peony leaf were selected according to the network topological characteristic parameters. Then,the enrichment analysis was carried out by using ClueGO software plug-in. Results There were 194 target sites for 11 blood entry components in peony leaves. Finally,171 signal pathways were ob-tained,and 21 key pathways related to cervical cancer were obtained after the wide pathway was excluded,such as estrogen signaling pathway,neurotrophin signaling pathway and so on. Conclusion Peony leaves may play a vital role in the treatment of cervical cancer by acting on inflammatory,metabolic,immunological,endocrine and cell cycle related protein targets and pathways.

ABSTRACT

This study was aimed to observe the effects of acupuncture combination with medicated thread moxibustion ofZhuang nationality medicine on neurotransmitters in rats with sleep deprivation. A total of 80 SPF grade healthy Wistar rats were randomly divided into the control group, model group, treatment group and diazepam group with 20 rats in each group. The establishment of insomnia model was through sleep deprivation in the model group, treatment group and diazepam group. Based on the treatment of the model group, the treatment group was treated by acupuncture combination with medicated thread moxibustion ofZhuang nationality medicine, once a day for continuous 14 days. Intragastric administration of diazepam according to 3 mg·kg-1 body weight was given in the diazepam group. While intragastric administration of the same volume normal saline was given to the control group once a day for continuous 14 days. And then, the levels of orexin, NE, 5-HT, CRH and ACTH were detected. The expressions of orexin mRNA and c-Fos were observed. The results showed that compared with the control group, the levels of orexin, ACTH, 5-HT, NE and CRH increased or obviously increased in the model group with significant difference (P < 0.05, orP< 0.01). The expressions of orexin mRNA and c-Fos in the model group were increased or obviously increased with significant difference (P < 0.05, orP< 0.01). Compared with the model group, the levels of orexin, ACTH, 5-HT, NE and CRH reduced or obviously reduced with significant difference in the treatment group (P < 0.05, orP < 0.01). The expressions of orexin mRNA and c-Fos were reduced or obviously reduced with significant difference in the treatment group (P <0.05, orP < 0.01). It was concluded that acupuncture combination with medicated thread moxibustion ofZhuang nationality medicine took an effect on the treatment of insomnia by mediating neurotransmitters associated with hypothalamic-pituitary-adrenal axis (HPA axis) through orexin, which was probably related with its therapeutic mechanism.

ABSTRACT

This study was aimed to observe effects ofDendrobium ofifcinaleKimura et Migo on cardiac function and changes of blood vessel in rabbits with coronary heart disease (CHD).Thirty clean grade healthy rabbits were randomly divided into 5 groups, which were the control group, model group,Dendrobium low-dose (DLD), middle-dose (DMD), and high-dose (DHD) group, with 6 rabbits in each group. Constrictor ring was implanted in the left coronary artery to establish CHD model in the model group, DLD, DMD and DHD groups. Intragastric administration of Dendrobium decoction at the dose of 10, 20, and 40 g·kg-1 was given to rabbits in the DLD, DMD and DHD groups once a day for 21 consecutive days, respectively. The same volume of normal saline was given to the control group and the model group. Detections were made on levels of IL-8, TNF-α, VCAM-1, ICAM-1, the expression of actin, changes of LVSP, LVEDP,±dp/dtmax, BPs, BPd and morphologic changes of blood vessel. The results showed that compared with the control group, levels of IL-8, TNF-α, VCAM-1, ICAM-1 and the expression of actin increased or significantly increased in the model group with statistical significance (P< 0.05, orP< 0.01); LVEDP increased significantly (P< 0.01). However, LVSP,±dp/dtmax and BPs reduced (P< 0.05, orP< 0.01). The blood vessel wall became thicker. Compared with the model group, IL-8, TNF-α, VCAM-1, ICAM-1 and the expression of actin in the DMD and DHD group reduced or significantly reduced with statistical significance (P< 0.05, orP< 0.01); LVEDP reduced significantly in the DMD and DHD group with statistical significance (P< 0.01). While, LVSP,±dp/dtmax and BPs increased or significantly increased with statistical significance (P< 0.05, orP< 0.01). The thickening of blood vessel wall was inhibited. It was concluded thatDendrobium ofifcinale Kimura et Migo can improve cardiac function, which may take effect by improving and inhibiting pathologic changes of blood vessel and alleviating vessel injury.

ABSTRACT

Objective To compare the efficacies of medicated thread moxibustion plus needle picking therapy versus medication for sciatica.Method Seventy sciatica patients were randomly allocated to a treatment group of 35 cases and a control group of 35 cases. The treatment group received medicated thread moxibustion on Zusanli (ST36), three cones once daily and needle picking at Yanglingquan (GB34) and Huantiao (GB30), once daily. The control group took quick-acting sciatica pills, two pills twice daily. One course of treatment consisted of 10 days in both groups. The therapeutic effects were compared between the two groups of patients after two consecutive courses of treatment.Result Posttreatment pain score decreased more markedly in the treatment group (P<0.01). The total efficacy rate was 88.6% in the treatment group and 57.1% in the control group; there was a statistically significant difference between the two groups (P<0.05).Conclusion Medicated thread moxibustion plus needle picking therapy for sciatica is more effective in relieving pain and raising the total efficacy rate.

ABSTRACT

OBJECTIVE:To isolate and identify the major constituents-steroidal saponins from Di'ao xinxuekang.METHODS:The constituents were separated and purified using normal-pressured and pressurized silica gel column chromatography technology,and the structures were identified based on the physico-chemical property and spectral analysis.RESULTS:Two steroidal saponins were obtained from Di'ao xinxuekang and their structures were identified as dioscin(1)and pseudoprodioscin(2).CONCLUSION:Compound 1 and 2 were demonstrated to be two major constituents in Di'ao xinxuekang.This finding is of great importance for the quality control and evaluation of Di'ao xinxuekang(Chinese Medicine).

ABSTRACT

OBJECTIVE:To investigate the effects of saikosaponin-d(SSd) on tissue plasminogen activator(TPA),plasminogen activator inhibitor(PAI),malonaldehyde(MDA) and NO in rats with liver fibrosis induced by dimethylnitrosamine(DMN).METHODS:Eighteen SD healthy rats were randomized to control group(NS ip qd for 4 weeks),model group(10mg? kg-1 DMN ip 3 times per week for 4 weeks) and SSd-treated group(10mg? kg-1 DMN ip 3 times per week + SSd 1.8 mg? kg-1 ip for 28 consecutive days).All rats were killed 1h after the last time of administration,blood samples were taken from abdominal aorta and liver samples were taken for the observation of pathology and detection of indices of TPA,PAI,MDA and NO etc.RESULTS:SSd could lessen the degree of liver fibrosis and improve the fibrinolytic activities of TPA and PAI,meanwhile,it showed clearance effect on MDA and marked protective effect on hepatic cells.There were significant differences between SSd-treated group and the model group.CONCLUSION:SSd exhibited protective function on experimental hepatic fibrosis in rats,which may be attributed to the improving of fibrinolytic activity,eliminating of lipid peroxidation and enhancing of NO level.

ABSTRACT

Object To compare the content in corilagin of Phyllanthus urinaria L. harvested from ten different regions of China in different months. Methods The content of corilagin was determined by UV spectrophotometry, detective wavelength is 270 nm. Results The content of corilagin in P. urinaria from Hainan and Guizhou province was the lowest, 0 58%. The highest content was given by the plant from Xi'an zone, Shanxi province, 1 78%. The best harvest time is in August and September, content of corilagin is 1 24%. Conclusion The difference of the corilagin contents in P urinaria . harvested from different regions and in different harvest time in the same region is great.

Objective To investigate the effect of hirudo micropowder on inflammatory factors in rats with cerebral ischemia-reperfusion injury.Methods Rats were randomly divided into sham-operation group,model group,coarse powder hirudo group,high-,middle-and low-dose micropowder hirudo groups.The corresponding drugs were given to the rats for 10 days by intragastric administration.Then middle cerebral artery occlusion(MCAO) model was made by suture method.The changes of inflammatory factors were observed.Results The level of intercellular adhesion molecule 1(ICAM-1) in high-dose micropowder hirudo group was lower than that in coarse powder hirudo group,and the level of platelet-derived growth factor(PDGF) in middle-and high-dose micropowder hirudo groups was also lower than that in coarse powder hirudo group obviously(P

ABSTRACT

AIM: To determine the contents of corilagin in Erodium stephanianum Willd. from different collection month and to define the best collection time for this herb. METHODS: The contents of corilagin in each samples of E. stephanianum were determined by HPLC. RESULTS: The linear regression of corilagin was obtained in the range of 0.22~1.10?g. The average recovery was 98.6% with a RSD of 0.63%. CONCLUSION: The content of corilagin in E. stephanianum collected in August is the highest among the samples throughout the collection months.
