ABSTRACT

Univariate (Analysis of Variance_ANOVA) and multivariate (Principal Component Analysis (PCA) and Canonical Discriminant Analysis (CDA)) analyses were performed in order to classify and authenticate the seeds from different varieties of quinoa (Chenopodium quinoa Will.), and amaranth (Amaranthus cruentus L.). The univariate analysis showed differences between species for sucrose, K, Ca, unsaturated fatty acids, and the ω6/ω3 ratio. Nevertheless, to strengthen this classification, a PCA was applied separating the samples in 2 groups; group 1, formed by quinoa seeds, presented higher contents of margaroleic, eicosadienoic, behenic, erucic, linolenic, linoleic, and gadoleic acids, proteins, sucrose, and total sugars. Group 2, formed by amaranth seeds, showed positive values for Mn, Mg, Fe, P, Zn, Ca, fiber, glucose, and ω6/ω3 ratio. Furthermore, the CDA models developed resulted in a probability of event of 100% when classifying the samples in the groups quinoa or amaranth, highlighting the good sensitivity of the models used.

Memory CD8+ T cells are indispensable for maintaining long-term immunity against intracellular pathogens and tumors. Despite their presence at oxygen-deprived infected tissue sites or in tumors, the impact of local oxygen pressure on memory CD8+ T cells remains largely unclear. We sought to elucidate how oxygen pressure impacts memory CD8+ T cells arising after infection with Listeria monocytogenes-OVA. Our data revealed that reduced oxygen pressure during in vitro culture switched CD8+ T cell metabolism from oxidative phosphorylation to a glycolytic phenotype. Quantitative proteomic analysis showed that limiting oxygen conditions increased the expression of glucose transporters and components of the glycolytic pathway, while decreasing TCA cycle and mitochondrial respiratory chain proteins. The altered CD8+ T cell metabolism did not affect the expansion potential, but enhanced the granzyme B and IFN-γ production capacity. In vivo, memory CD8+ T cells cultured under low oxygen pressure provided protection against bacterial rechallenge. Taken together, our study indicates that strategies of cellular immune therapy may benefit from reducing oxygen during culture to develop memory CD8+ T cells with superior effector functions.

ABSTRACT

OBJECTIVES: Lung transplant is a last treatment option for patients with end-stage pulmonary disease. Chronic lung allograft dysfunction, which generally manifests as bronchiolitis obliterans syndrome, is a major long-term survival limitation. Bronchiolitis obliterans syndrome is diagnosed when forced expiratory volume in 1 second declines > 20% in the absence of known causes. B cells can either contribute or restrain the development of bronchiolitis obliterans syndrome (eg, via induction of alloimmune antibodies, regulation of cellular immunity, and induction of tolerance). Here, we explored how peripheral B-cell subsets were altered in lung transplant recipients with bronchiolitis obliterans syndrome. MATERIALS AND METHODS: Fresh whole blood samples were analyzed from 42 lung transplant recipients, including 17 with bronchiolitis obliterans syndrome; samples from these groups were compared with 10 age-matched healthy control samples. B-cell subsets were analyzed using flow cytometry, and relative distributions of subsets were compared. Changes in forced expiratory volume in 1 second were also determined. RESULTS: Absolute B-cell count was significantly increased in transplant recipients with bronchiolitis obliterans syndrome. Transitional (CD24+CD38+) and naïve (CD27-IgD+) B cells were decreased in lung transplant patients, with transitional B cells almost absent in those with bronchiolitis obliterans syndrome. Double-negative (CD27-IgD-) memory B cells were significantly increased (P < .001). No differences were found for plasmablasts (CD38+CD24-) and switched (CD27+IgD-) and non-switched (CD27+IgD+) memory B cells. Correlation analyses showed positive correlations between lung function and naïve B cells in transplant recipients (P = .0245; r = -0.458). CONCLUSIONS: Peripheral B-cell count and subset distribution were altered in lung transplant recipients with and without bronchiolitis obliterans syndrome compared with healthy controls. Transitional and naïve B-cell decreases may be caused by differentiation toward double-negative B-cells, which were increased. The correlation between forced expiratory volume and naïve B cells during follow-up care may be clinically interesting to investigate.

ABSTRACT

Ever since Chuck Berry coined the term "rocking pneumonia" in his 1956 song "Roll over Beethoven", pneumonia has been mentioned frequently in modern blues and rock songs. We analyzed the lyrics of these songs to examine how various elements of pneumonia have been represented in popular music, specifically the cause of pneumonia, the risk groups, comorbidity (such as the boogie woogie flu), the clinical symptoms, and treatment and outcome. Up to this day, songwriters suggest that pneumonia is caused mainly by the cold and rain and that treatment is hardly possible, aside from a shot of rhythm and blues.

Introducción: el virus de la inmunodeficiencia humana es el causante del sida, debilita el sistema inmunológico destruyéndolo lentamente y ataca a las células que actúan como coordinadores del sistema inmunológico, provocando una respuesta de defensa inmunológica. Objetivo: determinar el nivel de conocimientos de los adolescentes que estudian en el preuniversitario acerca del Virus de la Inmunodeficiencia Humana. Material y método: se realizó una investigación de tipo observacional, descriptiva, transversal en el área urbana del municipio San Juan y Martínez en el período comprendido de enero hasta mayo de 2012, a los adolescentes que estudian en el preuniversitario. Resultados: se comprobó que existe un predominio del sexo femenino que conoce que el VIH/sida es una infección de transmisión sexual, señalan al semen como la vía que más conocen por la cual se transmite el VIH/sida, y la abstinencia sexual como la más efectiva para evitar el contagio; no usan el condón siempre en las relaciones sexuales, y la mayoría se dirige a sus padres para conversar sobre temas de sexualidad y VIH/sida. Conclusiones: la mayor parte de los estudiantes refiere conocer que el VIH/sida es una enfermedad que se transmite sexualmente, y que es necesario ejecutar programas de educación sexual para adquirir mejores conocimientos sobre esta enfermedad venérea.

Objective: to determine knowledge of Human Immunodeficiency Virus in adolescents studying in an urban pre-university institute. Material and Method: a type of observational, descriptive, cross-sectional research was conducted in an urban area of San Juan y Martinez municipality during January-May 2012 in adolescents studying in a pre-university institute. Results: knowledge of HIV/AIDS prevailed in female sex; they knew it is a sexual transmitted-infection, pointing out semen as the main way of transmission for HIV/AIDS, sexual abstinence as the most effective to avoid the infection; they not all the times wear condoms, the majority talk to their parents about sexuality and HIV/AIDS topics. Conclusions: the greatest part of the students referred HIV/AIDS is a sexually-transmitted disease, and that it is necessary to create programs toward sexual education in order to increase knowledge of this venereal disease.

ABSTRACT

Introducción: Múltiples estudios muestran que el índice de calcio coronario (ICC ) es predictor de eventos coronarios de mayor exactitud respecto a escalas de riesgo tradicionales; sin embargo, no se han realizado en países como Colombia. Se compara la asociación del índice de Framingham (IF) con el ICC en la detección de enfermedad coronaria y su gravedad. Metodología: Se revisaron informes de angiotomografía coronaria de los últimos tres años de pacientes que tenían calculado IF e ICC mediante tomografía multicorte. Se determinaron medidas de correlación y de asociación entre las dos técnicas con la presencia y gravedad de enfermedad coronaria determinada por angiotomografía multicorte. Resultados: Se incluyeron 96 pacientes en total. El ICC presentó mayor correlación con la presencia y gravedad de enfermedad coronaria (0,84; p < 0,001) en comparación con el IF (0,30; p < 0,001). Existe una correlación baja (0,33; p < 0,001) entre el IF y el ICC. No se encontró asociación significativa entre el IF moderado y enfermedad coronaria significativa (OR = 2,51; IC 95: 0,7-8,9) y no significativa (OR = 2,3; IC 95: 0,86-6,15). Por el contrario, se encontró que un ICC ≥ 100 está asociado con enfermedad coronaria (OR = 182; IC 95: 17,4-190,2) en contraste con el IF crónico (OR = 18,6; IC 95: 1,75-198,0). Conclusiones: La medición de ICC por tomografía multicorte presenta mayor correlación con presencia y gravedad de enfermedad coronaria en comparación con el IF. Existe una correlación bajaentre el índice de Framingham y el de calcio coronario.

ABSTRACT

El pseudoaneurisma de la arteria carótida común es una patología poco frecuente. En este artículo se presenta el caso de una mujer de 54 años de edad con antecedente de carcinoma de seno, tratado con quimioterapia, a quien por sospecha de adenopatía cervical metastásica le realizaron una biopsia por aspiración sin guía imagenológica posterior a lo cual presenta incremento en el tamaño de la masa, la cual además se hace pulsátil. Se le practicó una ecografía Doppler de cuello y una angiotomografía computarizada (angioTC), carotídea cuyas imágenes se presentan. No refería amaurosis, paresia u otros síntomas de déficit neurológico. La ecografía Doppler mostró una dilatación bien localizada en el tercio proximal de la arteria carótida común izquierda. El estudio de angioTC de cuello confirma un aneurisma sacular de cuatro centímetros de diámetro ubicado en el origen de la carótida común izquierda. Se evidencia también una elongación bilateral del árbol carotídeo y retardo en el llenamiento de la circulación intracerebral ipsilateral. La ecografía Doppler cervical y el angioTC de cuello realizado con un equipo multidetector de 64 canales son herramientas útiles que permiten adquirir imágenes en corto tiempo con buena resolución temporal y espacial y que suministran información similar a la de la angiografía convencional.

ABSTRACT

Las anomalías anatómicas de las arterias coronarias son raras, y pueden ser causa frecuente de dolor precordial, isquemia miocárdica y muerte súbita. La identificación confiable de estas anomalías es de gran importancia para determinar el manejo de estos pacientes. Durante varios años se ha empleado la angiografía convencional como método de diagnóstico. Sin embargo, debido a su alto costo, baja disponibilidad y naturaleza invasiva presenta limitaciones para su uso. El desarrollo del TAC multicorte de 64 canales con sincronización electrocardiográfica permite visualizar las arterias coronarias, con muy buena resolución espacial, y promete ser el método de diagnóstico de elección en estos casos. Es necesario que los especialistas a cargo de la interpretación de los angiotac coronarios estemos familiarizados con la prevalencia, apariencia y significado clínico de estas anomalías. A continuación se describen los hallazgos en TAC multicorte de 64 canales de la anatomía, dominancia y de algunas anomalías congénitas coronarias.

ABSTRACT

A capillary zone electrophoresis (CZE) method using a fused-silica capillary (60.2 cm x 75 microm ID) was investigated for the determination of triamterene (TRI), methotrexate (MTX), and creatinine (CREA) in human urine. The separation was performed using a hydrodynamic injection time of 7 s (0.5 psi), a voltage of 25 kV, a capillary temperature of 30 degrees C, and 40 mM phosphoric acid adjusted to pH 2.25 by addition of triethanolamine as separation electrolyte. Under these conditions, analysis takes about 15 min. A linear response over the 0.5-15.0 mg L(-1) concentration range was found for TRI and MTX, and 0.5-80.0 mg L(-1) for CREA. Dilution of the sample (water:urine, 1:1 for TRI and MTX, and 1:25 for CREA determination) was the only step necessary prior to analysis by electrophoresis. The developed method is easy, rapid, and sensitive and has been applied to determine triamterene,methotrexate, and creatinine in urine samples with satisfactory results.
