ABSTRACT

OBJECTIVE: This study investigated the intervention effect of slow-velocity eccentric-only resistance training on type 2 diabetic mellitus (T2DM) patients based on the role of matrix metalloproteinase-2 and -9 (MMP-2 and -9) in regulating extracellular matrix homeostasis. METHODS: 50 T2DM patients were randomly divided into the slow-velocity eccentric-only resistance training group (E) and control group (C). The E group performed eccentric-only resistance training 3 times a week, every other day for 10 weeks, while the C group did not. Blood samples were collected before and after training, and subjects were tested for changes in clinical parameters, insulin resistance indices [fasting insulin, homeostatic model assessment insulin resistance (HOMA-IR)], MMP-2 and -9, and hydroxyproline, and muscle strength (12-RM), respectively. RESULTS: After 10 weeks of training, the E group showed significant decreases in fasting glucose (P < .05), insulin (P < .05), insulin resistance indices (P < .05), hemoglobin A1c (HbA1c) (P < .01), triglycerides (P = .06) and MMP-2 (P < .05), while total cholesterol (P < .05), MMP-9 (P < .05), hydroxyproline (P < .01), Creatine Kinase (CK) (P < .05), and muscle strength (P < .001) significantly increased. There were no significant changes in the count of neutrophil, lymphocyte and platelet, neutrophil-to-lymphocyte ratio (NLR), platelet-to-lymphocyte ratio (PLR), high-density lipoprotein cholesterol (HDL-c), and low-density lipoprotein cholesterol (LDL-c). Compared with the C group, the E group showed a trend of a significant decrease in triglyceride (P < .05), lymphocyte count (P < .05), fasting glucose (P = .07), and plasma MMP-2 (P < .05), while MMP-9 (P < .05), hydroxyproline (P < .001), and muscle strength (P < .01) significantly increased. However, no significant changes were observed in insulin and insulin resistance indices, HbA1c, total cholesterol, HDL-c, LDL-c, CK, and other inflammatory indicators. CONCLUSIONS: Slow-velocity eccentric-only resistance training was beneficial for T2DM, but the potential role of MMP-2 and -9 in regulating extracellular matrix homeostasis is very different in T2DM patients.

ABSTRACT

Intense and unaccustomed eccentric exercise has been extensively studied for its ability to induce muscle damage. However, the underlying mechanism of this phenomenon still requires further clarification. This knowledge gap arises from the need for explanation of the eccentric contraction through the sliding filament theory. The two-filament sarcomere model, which is consisted of thin and thick filaments, forms the basis of the sliding filament theory. The mechanisms of concentric and isometric contractions at the cellular and molecular levels are effectively described by this model. However, when relying solely on the cross-bridge swing, the sliding filament theory fails to account for specific observations, such as the stability of the descending limb of the force-length relationship curve. Recent evidence indicated that titin and the extracellular matrix (ECM) may play a protective role by interacting with the thick and thin filaments. During an eccentric contraction, titin serves as a third filament in the sarcomere, which helps regulate changes in passive force. The two-filament sarcomere model has limitations in explaining eccentric contraction, thus this compensates for those shortcomings. The present review explored the potential of replacing the two-filament sarcomere model with a three-filament sarcomere model, incorporating thin filaments, thick filaments and titin. This revised model offers a more comprehensive explanation of eccentric contraction phenomena. Furthermore, the sliding filament theory was investigated in the context of the three-filament sarcomere model. The double-layer protection mechanism, which involves increased titin stiffness and the ECM during eccentric contraction was explored. This mechanism may enhance lateral force transmission between muscle fibers and the ECM, resulting in sarcolemma and ECM shear deformation. These findings provided insight into the mechanism of eccentric exercise-induced skeletal muscle damage. Considering the three-filament sarcomere model and the double-layer protection mechanism, the present review offered a more logical and comprehensive understanding of the mechanism behind eccentric exercise-induced muscle damage.

ABSTRACT

ObjectiveTo explore the anti-abortional effect of Pangshi Antai Zhixue decoction and its mechanism in helper T lymphocyte 1 (Th1)/Th2 balance in the decidual tissues of spontaneous abortion rats with heat syndrome, based on the p38 mitogen-activated protein kinase (p38 MAPK) signaling pathway. MethodAconiti Lateralis Radix Praeparata, Zingiberis Rhizoma, and Cinnamomi Cortex decoction was used to replicate the rat model of spontaneous abortion with heat syndrome. The spontaneous abortion rats with heat syndrome were randomly divided into model group, aspirin group (5.25 mg·kg-1), dydrogesterone group (3.02 mg·kg-1), Pangshi Antai Zhixue decoction high-dose (44 g·kg-1), medium-dose (22 g·kg-1), and low-dose (11 g·kg-1) groups, with ten rats in each group. Ten normal rats were divided into a normal group. Rats in each group were given corresponding drugs, Once a day for 12 d. After 24 h of the last administration, blood was collected from the abdominal aorta. The enzyme-linked immunosorbent assay (ELISA) was used to determine the levels of β-human chorionic gonadotropin (β-HCG), progesterone (P), estradiol (E2), γ interferon (IFN-γ), and interleukin-4 (IL-4) in rat serum. The uterus and meconium tissues of rats were collected to determine the number and rate of miscarriages. Western blot was used to detect GATA3, T-bet, p38 MAPK, and its phosphorylation in the decidual tissue. ResultAs compared with the normal group, the number of live births, the β-HCG, P, E2, and IL-4 in the serum, and the GATA3 protein expression in the decidual tissue in the model group were reduced (P<0.01), whereas the number and rate of miscarriages, IFN-γ in the serum, and the expression of p-p38 MAPK and T-bet protein levels in the demolded tissues increased (P<0.01). As compared with the model group, the number of live births, the β-HCG, P, E2, and IL-4 in the serum, and the GATA3 protein expression in the decidual tissue in the Pangshi Antai Zhixue decoction medium-dose group increased (P<0.01), whereas the number and rate of miscarriages, IFN-γ in the serum, and the expression of p-p38 and T-bet protein levels in the demolded tissues reduced (P<0.01). As compared with the aspirin group, the P, E2, and IL-4 in the serum of rats in the dydrogesterone group and the Pangshi Antai Zhixue decoction high-dose and medium-dose groups increased (P<0.01), the number of live births in the Pangshi Antai Zhixue decoction medium-dose group increased (P<0.01), and the β-HCG and IFN-γ in the serum of rats in the dydrogesterone group decreased (P<0.01). The number and rate of miscarriages, IFN-γ in the serum, and T-bet and GATA3 levels in the decidual tissues of rats in the Pangshi Antai Zhixue decoction medium-dose group decreased (P<0.05). ABSTRACT

Non-alcoholic fatty liver disease (NAFLD) is an epidemic metabolic condition driven by an underlying lipid homeostasis disorder. The lipid droplet (LD), the main organelle involved in neutral lipid storage and hydrolysis, is a potential target for NAFLD therapeutic treatment. In this review, we summarize recent progress elucidating the connections between LD-associated proteins and NAFLD found by genome-wide association studies (GWAS), genomic and proteomic studies. Finally, we discuss a possible mechanism by which the protein 17β-hydroxysteroid dehydrogenase 13 (17β-HSD13) may promote the development of NAFLD.

ABSTRACT

[ ABSTRACT] AIM:To investigate the effects of BARF1 down-regulation on EBV-positive gastric carcinoma cell apoptosis, and the molecular mechanisms by BARF1 silencing-mediated apoptosis.METHODS: After NUGC3 and SNU719 cells were transfected with NCsiRNA and siRNA, respectively, the protein levels of BARF1, Bcl-2, Bax, cyto-chrome C, caspase 3 and capase 9 were detected by Western blot, and the mRNA expression of BARF1, Bcl-2 and Bax was determined by RT-PCR.The cell viability was measured by the method of Trypan blue exclusion and the cell apoptosis was analyzed by flow cytometry analysis with Annexin V-FITC/PI staining. The expression of the apoptosis-related proteins in the cells transfected with siRNA and NCsiRNA was examined by human apoptosis antibody arrays.Mitochondrial mem-brane potential was determined by flow cytometry. The interaction between Apaf-1 and caspase 9 was confirmed by immuno-precipitation.RESULTS: Compared with untreated and NCsiRNA groups, BARF1 gene silencing significantly inhibited the cell viability, induced apoptosis, and reduced the mitochondrial membrane potential in the NUGC3 and SNU719 cells transfected with siRNA.BARF1 gene silencing up-regulated the expression of pro-apoptotic proteins and down-regulated the expression of anti-apoptotic proteins, and the Bcl-2/Bax ratio was significantly decreased. In BARF1 gene silencing cells, the caspase inhibitor z-VAD-fmk inhibited BARF1 silencing-mediated apoptosis, and significantly increased the levels of cleaved caspase 3 and caspase 9.The concentration of cytochrome C significantly increased as compared with NCsiRNA group, and Apaf-1 interacted with caspase 9 in the cytoplasm.CONCLUSION:BARF1 silencing induces apoptosis via the mitochondrial pathway through regulating the expression of Bcl-2 and Bax proteins in a caspase-dependent manner in the NUGC3 and SNU719 cells.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the changes in orbitofrontal cortex (OFC) functional connectivity and its association with decision-making deficits in chronic heroin-dependent individuals (HDIs) and explore the neural mechanisms of heroin addiction and relapse.</p><p><b>METHODS</b>Fourteen male chronic HDIs and 14 healthy subjects matched for age, education, and nicotine consumption participated in this study. Resting state functional magnetic resonance imaging (fMRI) was performed using a 1.5 T MR scanner. Functional connectivity of the OFC and the rest of the brain were calculated using REST software. Voxel-based analysis of the functional connectivity maps between the control and HDI groups was performed with two-sample t test. The Iowa gambling task (IGT) was used to assess the participants' decision making during uncertainty.</p><p><b>RESULTS</b>Compared with the control group, the HDIs showed significantly decreased functional connectivity of the OFC and the right inferior parietal lobule (rIPL) (t=3.5, P<0.05). A significant negative correlation was noted between the functional connectivity of the OFC-rIPL and performance level at the IGT.</p><p><b>CONCLUSION</b>The OFC-rIPL functional connectivity is significantly disrupted in HDIs, which may be the neural basis for decision-making deficits.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To analyze the renal relative blood flow value (rBFV) and image quality in normal adults using single-shot fast spin echo, flow sensitive invention recovery (SSFSE-FAIR) magnetic resonance (MR) sequence and echo planar imaging, and flow sensitive invention recovery (EPI-FAIR) MR sequence, and assess its value for clinical application in routine renal examination.</p><p><b>METHODS</b>Forty volunteers (25 male and 15 female adults, aged 30 to 62 years) with normal renal function were included in this prospective study. All the subjects underwent 3.0 Tesla MR scanning using 3 MR scan modes, namely breath-holding EPI-FAIR, breath-holding SSFSE-FAIR and free breathing SSFSE-FAIR.</p><p><b>RESULTS</b>SSFSE-FAIR without breath-holding was capable of differentiating the renal cortex and medulla with the corresponding rBFVs of 111.48∓9.23 and 94.98∓3.38, respectively. Breath-holding SSFSE-FAIR and EPI-FAIR failed to distinguish the borders of the renal cortex and medulla. The EPI-FAIR rBFV of mixed cortex and medulla value was 178.50∓17.17 (95%CI: 167.59, 189.41).</p><p><b>CONCLUSION</b>Breath-holding SSFSE-FAIR and EPI-FAIR can not distinguish the renal cortex and medulla due to a poor spatial resolution but can be used for rough evaluation of renal blood perfusion. Free breathing SSFSE-FAIR with an improved spatial resolution allows evaluation of the status of renal perfusion of the cortex and medulla.</p>

ABSTRACT

Recent studies have shown that isocitrate dehydrogenase 1/2 (IDH1/2) mutations occur frequently in secondary glioblastoma. This study aimed to investigate their impact on temozolomide chemosensitivity and relationship with O(6)-methylguanine DNA methyltransferase (MGMT) promoter methylation in secondary glioblastoma. Searches for IDH1 and IDH2 mutations, 1p19q codeletion, MGMT promoter methylation, and p53 expression were carried out in a series of 86 secondary glioblastomas and correlated with progression-free survival and overall survival. Response to temozolomide was evaluated by progression-free survival, as well as by tumor size on successive MRI scans, then correlated with molecular alterations. IDH (IDH1 or IDH2) mutations were found in 58/79 patients (73.4%). IDH mutation, MGMT promoter methylation, and 1p19q codeletion were associated with prolonged progression-free survival in univariate (P < 0.001, P < 0.001, P = 0.003, respectively) and multivariate analysis (P < 0.001, P < 0.001, P = 0.035, respectively). IDH mutation (P = 0.001) and MGMT promoter methylation (P = 0.011) were correlated with a higher rate of objective response to temozolomide. Further analysis of response to temozolomide showed that patients with both IDH mutation and MGMT promoter methylation had the best response rate to temozolomide. IDH mutation appears to be a significant marker of positive chemosensitivity in secondary glioblastoma. Use of IDH status combined with MGMT promoter status as a stratification factor seems appropriate in future clinical trials involving temozolomide for the treatment of patients with secondary glioblastoma.

Objective To analyse the changes in bilateral optic radiation and visual cortex in patients with primary glaucoma detected by magnetization transfer imaging ( MTI ),and try to explore the influence of the disease on posterior visual pathway. Methods MTI was performed in 20 patients with primary glaucoma with normal signal on conventional magnetic resonance imaging (MRI).The same scanning was performed in 31matched healthy controls.MTI was obtained using spoiled gradient recalled acquisition sequence (SPGR).Magnetization transfer ratio ( MTR ) of bilateral optic radiation and visual cortex was measured after post-processing.The MTR value differences of the same area between two groups were compared by independent-sample t test or Satterthwaite t test if variances were not equality.Result The MTR value in the left and right optic radiation were ( 32.8 ± 2.2 ) % and ( 32.7 ± 2.0 ) % in the glaucoma group,(34.6 ± 1.4 )% and (34.8 ± 1.3 )% in the control group. There was a statistically significant difference between the two groups (left t =3.284,right t =4.040 ;P < 0.01).The MTR value of the left and right visual cortex were ( 30.1± 2.0 ) % and ( 30.8 ± 1.8 ) % in the glaucoma goup, and (32.3 ± 1.2 )% and (32.4 ± 1.2 )% in the control group.Statistically significant difference was found between the two groups ( left t =4.319,right t =3.445 ;P < 0.01).Conclusions Potential neuropathology changes occurring in the posterior visual pathway of patients with glaucoma indicate that the whole visual pathway may be involved by glaucoma. The micro physiological changes can be detected by MTI which can not be found by conventional MRI.It is a useful method of studying trans-synaptic damage of visual pathway n vivo glaucoma which provides more information for guiding the clinic diagnosis, cure and prognosis of glaucoma.

ABSTRACT

Purpose to explore a CT classflication of hepatic echinococcal cysts and its clinical application. Materials and Methods reviewed 85 patients who had underwent CT scan in our hospital from 2008-01~2009-12.The CT classification was made according to WHO classification and the CT appearances were analyzed retrospectively. Results Among the lesions, there were 16 CL type, appeared as a unilocular round cyst without wall.15 CE1 type, appeared as a unilocular cyst with a smooth uniform wall.28 CE2 type, appeared as a fluid collection with septa (honeycomb sign),and 6 CE3 type,CT appeared as a fluid collection with a split wall,10 CE4 type,CT appeared as a fluid collection with a split wall (water-lily sign).36 CE5 type,CT appeared as a calcified mass. Conclusion CT classification according to WHO classification for cystic echinococcosis is using to differentiate it from other liver cystic lesions, such as simple cyst and abcess, and is essential when planning clinical treatment.

ABSTRACT

Objective To explore the imaging features and pathological characteristics of fibrous dysplasia of craniofacial bone. Methods 42 cases with fibrous dysplasia of craniofacial bone confirmed surgically and pathologically were retrospectively analyzed. Results There were 35 cases with single bone involved and 7 cases with poly-bone involved. On CT images, the lesions appeared as glass-like change(79.3%). The main pathological components of the lesion were fiber and fiber-like tissue, which were low signals on both T_1 WI and T_2 WI, and the signal intensity was changed with the accompanied by hemorrhage, necrosis or cystic degeneration. Conclusion CT and MRI are the effective imaging modality in diagnosis of fibrous dysplasia of craniofacial bone.

ABSTRACT

Objective To investigate MRI features of dysembryoplastic neuroepithelial tumor(DNT).Methods The MR images of ten cases with surgically and pathologically proved DNT were retrospectively reviewed. Results MR imaging showed cystic component with mural nodule in 6,mainly cystic component with septa in 3.The solid components of the tumors were long T_1 and long T_2 signal intensity, the cystic components were even longer T_1 and T_2 signal intensity which resembled the signal of cerebrospinal fluid.Mild enhancement could be found at the intra-tumoral solid nodule or septations after injection of constrast medium but the cystic component did not show any enhancement. No peri-tumoral edema and mass effect were found in most cases. Conclusion DNTs are of some special features on the MR images, the diagnosis can be exactly done before operation if MRI features in combination with clinical data of the patients.

ABSTRACT

Objective To explore the susceptibility weighted imaging (SWI) appearance of paramagnetic and diamagnetic materials, its imaging theory, and its value of differential diagnosis from their appearance. Methods The diamagnetic and paramagnetic phantom were made and the X-Y axial SWI phase image of phantom then was obtained. Twenty-eight cases of intracerebral calcium and 21 cases of hemorrhage examined with MRI scan and CT scan were retrospectively investigated. The appearance of calcium and hemorrhage on SWI were analyzed and compared with their appearance on CT and conventional MRI. Results Phantom experiment indicated that the equator plane scan of diamagnetic sphere showed high signal in centre surrounded by low signal ring and high signal in two poles of sphere along Z axis of main magnetic field. The paramagnetic sphere appeared the opposite appearance of diamagnetic sphere with centre low signal surrounded by high signal ring in equator plane and low signal in two poles of sphere along Z axis of main magnetic field. The SWI manifestations of 4 cases of intracerebral sphere or sphere-like calcification ( chorioidal calcification) and 9 cases of sphere or sphere-like hemorrhage(2 cases of traumatic hemorrhage, 1 case of hypertension related basal ganglion hemorrhage, 4 cases of metastasis hemorrhage and 2 cases of cavernous hemangioma hemorrhage)were consistent with diamagnetic and paramagnetic sphere phantom respectively. Four cases of calcification in basal ganglion showed the mixed high and low signal. Seven cases of irregular calcification ( neoplastic calcification) aside from basal ganglion appeared complex appearance, though mainly in high signal. Other sphere or sphere-like calcification aside from basal ganglion ( 14 cases of pineal calcification, 4 cases of chorioidal calcification, 2 cases of cerebral cysticercosis calcification and 1 case of neoplastic calcification ) showed markedly high signal. Nine cases of intracerebral iobular hemorrhage (2 cases of hypertension related basal ganglion hemorrhage, 4 cases of unknown casual hemmorrhge and 3 cases of cavernous hemangioma hemorrhage) displayed a multiple concentric circle appearance, other 3 hemorrhage showed complex appearance, though mainly in low signal. Ten cases of calcification had no specific appearance on T1 WI and T2WI. Conclusion Phase image of SWI is helpful in differentiating intracerebral diamagnetic from paramagnetic materials and it has some advantage in detecting calcification over T WI and T2 WI.

ABSTRACT

OBJECTIVE: To observe the effects of Qingdan Capsule (QDC) and Yanggan Lidan Granule (YGLDG), two kinds of compound traditional Chinese herbal medicines, on biochemical parameters in guinea-pigs with pigment gallstones. METHODS: An animal model of pigment gallstones was established in male guinea-pigs by hypodermic injection of lincomycin. The guinea-pigs were randomly divided into blank control group, untreated group, QDC group and YGLDG group. There were 8 guinea-pigs in each group. After ten-day treatment, animals were sacrificed and sampled to calculate the rate of stone formation, total bilirubin (TB), unconjugated bilirubin (UCB) and Ca2+ density in bile of the four groups. RESULTS: In comparison with the untreated group, the rates of stone formation in the QDC and YGLDG groups were significantly decreased (P<0.01). TBIL, UCB and Ca2+ content of bile in both QDC and YGLD groups was also significantly decreased (P<0.05, P<0.01). CONCLUSION: QDC and YGLD have good effects on biochemical changes of animal model of pigment gallstone in reversing the lithogenesity of bile by reducing the content of TB, UCB and Ca2+, hence resulting in clinical treatment and prevention of pigment gallstone disease.
