ABSTRACT

Peritoneal membrane dysfunction and the resulting ultrafiltration failure are the major disadvantages of long-term peritoneal dialysis (PD). It becomes increasingly clear that mesothelial cells play a vital role in the pathophysiological changes of the peritoneal membrane. Matrix metalloproteinases (MMPs) function in the extracellular environment of cells and mediate extracellular matrix turnover during peritoneal membrane homeostasis. We showed here that dialysate MMP-7 levels markedly increased in the patients with PD, and the elevated MMP-7 level was negatively associated with peritoneal ultrafiltration volume. Interestingly, MMP-7 could regulate the cell osmotic pressure and volume of human peritoneal mesothelial cells. Moreover, we provided the evidence that MMP-7 activated mitogen-activated protein kinases (MAPKs)-extracellular signal-regulated kinase 1/2 (ERK) pathway and subsequently promoted the expression of aquaporin-1 (AQP-1) resulting in the change of cell osmotic pressure. Using a specific inhibitor of ERK pathway abrogated the MMP-7-mediating AQP-1 up-regulation and cellular homeostasis. In summary, all the findings indicate that MMP-7 could modulate the activity of peritoneal cavity during PD, and dialysate MMP-7 might be a non-invasive biomarker and an alternative therapeutic target for PD patients with ultrafiltration failure.

ABSTRACT

Herein, we developed a label-free electrochemical biosensor for sensitive detection of matrix metalloproteinase-7 (MMP-7) based on DNA enzyme-decorated DNA nanoladders as enhancer. A peptide and single-stranded DNA S1-modified platinum nanoparticles (P1-PtNPs-S1), which served as recognition nanoprobes, were first immobilized on electrode. When target MMP-7 specifically recognized and cleaved the peptide, the PtNPs-S1 bioconjugates were successfully released from electrode. The remaining S1 on electrode then hybridized with ssDNA1 (I1) and ssDNA2 (I2), which could synchronously trigger two hybridization chain reactions (HCRs), resulting in the in situ formation of DNA nanoladders. The desired DNA nanoladders not only were employed as ideal nanocarriers for enzyme loading, but also maintained its catalytic activity. With the help of hydrogen peroxide (H2O2), manganese porphyrin (MnPP) with peroxidase-like activity accelerated the 4-chloro-1-naphthol (4-CN) oxidation with generation of insoluble precipitation on electrode, causing a very low differential pulse voltammetry (DPV) signal for quantitative determination of MMP-7. Under optimal conditions, the developed biosensor exhibited a wide linear ranging from 0.2 pg/mL to 20 ng/mL, and the detection limit was 0.05 pg/mL. This work successfully realized the combination of DNA signal amplification technique with artificial mimetic enzyme-catalyzed precipitation reaction in peptide cleavage-based protein detection, offering a promising avenue for the detection of other proteases.

ABSTRACT

Lung cancer is one of the most common malignant tumors in the world.Non-small lung canc-er( NSCLC) accounts for approximately 85%,but its pathogenesis has not been elucidated.As a family of soluble enzymes,matrix metalloproteinases(MMPs)can degrade the constituents of the extracellular matrix(ECM)and the basement-membrane,members share common structural features and are responsible for the relative specific sub-strates.Some researches show that the matrix metalloproteinases 7(MMP-7)involve in the invasion and metasta-sis of NSCLC,the expression of MMP-7 in the tissue of NSCLC is significcantly higher than that in adjacent tis-sues,and the MMP-7 plays important roles in prognosis evaluation of NSCLC patients.Therefore this paper sum-marize the domain structure,function,regulation and research in NSCLC about MMP-7.

ABSTRACT

Objective:To investigate the expression and clinical significance of Ang-2 and MMP-7 protein in laryngeal carcinoma tissue.Method:Immunohistochemical technique was used to detect the expression of Ang-2 and MMP-7 protein in 65 tissues of laryngeal carcinoma and 34 biopsies of adjacent non-cancerous tissue. The relationship between the expression of Ang-2 and MMP-7 and invasion, metastasis or prognosis in laryngeal carcinoma tissue was analyzed.Result:The positive rates of Ang-2 and MMP-7 were significantly higher in laryngeal carcinoma tissue than those in adjacent non-cancerous tissue(P<0.05).The level of Ang-2 and MMP-7 expression had no significant correlations with the age and course as well as the smoking, drinking, histological differentiation of carcinoma and clinical classification (P>0.05).While the expression of Ang-2 significantly differed between patients with different T stage and clinical stage(P<0.05), and the expression of MMP-7 was notably correlated with the T stage,clinical stage and lymph node metastases (P<0.05).There was a correlation between the expression of Ang-2 and MMP-7 (P<0.05).The Kaplan-Meier survival analysis showed that patients with positive expression of Ang-2 had worse overall survival(P<0.05).However,MMP-7 expression was not related to overall survival or disease-free survival (all P>0.05).Cox regression analysis indicated that Ang-2 and MMP-7 expression were independent prognostic factors of laryngeal carcinoma.Conclusion:Overexpression of Ang-2 and MMP-7 was observed in laryngeal carcinoma and they might be served as an objective indicator for biological behaviour and prognosis.

ABSTRACT

Objective To study the relationship between expression of matrix metalloproteinases-7 (MMP-7) and clinicopathological characteristics in patients with primary non-smaU cell lung cancer(NSCLC). Methods MMP-7 in 20 normal people and 60 advanced NSCLC patiens were detected with reverse-transcription-polymerase-chain-reaction. Gelatum image analysator analyzed the result. Results The amount of MMP-7 was less in normal people (30.000) than in NSCLC patients(41.231) significantly(P<0.05); the level of MMP-7 was no correlated with gender, age, pathology pattern, tumor size, was inverse correlation with differentiation, and was positive correlation with clinical stages(P <0.05). Conclusion The level of MMP-7 is closely correlated with tissue differentiation and clinical stages of NSCLC, which may serve as a parameter for determining tumor invasion and metastatic.

ABSTRACT

Objective To investigate the relationship between diameter of early gastric cancer as well as lymph node metastasis and expression of vascular endothelial growth factor(VEGF) and Matrix metalloproteinases-7(MMP-7)in gastric cancer. Method VEGF and MMP-7 expressions of gastric cancer tissue in 55 cases were examined by immumohistochemical assay. The diagnosis was confirmed by mucous stain and pathology. Results The level of VEGF and MMP-7 expression in group of early gastric cancer with diameter ≤ 10 mm was significantly lower than that of diameter ≥ 10 mm and ≤ 20 mm (P0.05). Conclusion It is significant that relation of the diameter of early gastric cancer with VEGF and MMP-7 expression, the cases of strongly positive over-expression of both VEGF and MMP-7 in tissue of patients with early gastric cancer are accompanied by more likelihood of lymph node metastasis.
