ABSTRACT

An on-line high-performance liquid chromatography-diode-array-detector-electrospray ionization-ion-trap-time-of-flight-mass spectrometry-total antioxidant capacity detection (HPLC-DAD-ESI-IT-TOF-MS-TACD) system was applied for the identification and evaluation of antioxidants in Rosa chinensis Jacq., an edible flower in food industry and a widely used traditional Chinese medicine. With the help of this platform, the HPLC fingerprint, mass fragmentations, and sample activity profiles against 1,1-diphenylpicryl-2-hydrazyl radical (DPPH•) and ferric reducing antioxidant power (FRAP) were recorded after one injection. Using this technique, 80 compounds were separated and identified by their LC/MS behaviors with the assistance of standard compounds. In addition, 11 different Rosa chinensis Jacq. samples were profiled and then quantified for their DPPH• and FRAP activities. Interestingly, a total of 52 compounds showed antioxidative effects against DPPH• and 61 were active against FRAP. The results demonstrated that the on-line system is a powerful technique for antioxidant discovery in Rosa chinensis Jacq. and other food resources.

ABSTRACT

Objective: To study the genetic diversity and genetic relationship of medicinal plants Rosa chinensis, R. rugosa, and their relative species by ISSR molecular marker technique, and provide the reference for Rosa L. germplasm identification and breeding. Methods: Fifteen species (including 33 samples) of medicinal herbs R. chinensis, R. rugosa, and their relative species were studied by ISSR-PCR markers. Nei's genetic diversity index (H) and other parameters of genetic information were calculated by POPGEN 32, and a cluster dendrogram of different samples was established based on the unweighted pair-group method with arithmetic mean (UPGMA) by NTSYS-pc software. Results: Six ISSR primers generated 110 loci of which 109 loci were polymorphic. The average percentage of polymorphie bands (PPB) was 99.09%. Nei's genetic diversity index (H) and Shannon's information index (I) were 0.370 5 and 0.546 4, and the coefficient of genetic differentiation (Gst) and gene flow (Nm) were 0.886 8 and 0.063 8 between the species levels. The genetic distance (D) varied from 0.169 1 to 0.730 2. In the cluster dendrogram, 15 species were clustered into six groups at the level of Genetic similarity coefficient (GS) 0.60. Conclusion: The results of ISSR analysis reveal that medicinal plants R. chinensis, R. rugosa and their relative species had the plentiful genetic diversity and the genetic relationships were consistent with morphological characters of taxonomy. ISSR method is efficient for identification of R. chinensis, R. rugosa, and their relative species, which could provide a scientific basis for the resource collection and identification of the species in Rosa L.

The chemical constituents of Rosa chinensis Jacq were diverse ,mainly including flavonoids ,flavonoid glyco-sides ,phenolic acids ,aromatic oils ,tannins and pigments .Its extract and some chemical constituents had shown multiple phar-macological activities ,such as antitumor ,antifungal ,anti-viral ,anti-oxidation etc ..The advances in the study on chemical com-ponents and pharmacological actions of Rosa chinensis Jacq were reviewed and its application prospect was prospected .

ABSTRACT

OBJECTIVE: To investigate the chemical constituents in Rosa chinensis Jacq. METHODS: The chemical constituents were isolated and purified by silica gel, Sephadex LH-20, and recrystallization, et al. Their structures were elucidated by spectral analysis and physicochemical properties. RESULTS: From petroleum ether fraction and chloroform fraction of water extraction of Rosa chinensis Jacq., the structures of 15 compounds were identified as β-sitosterol (1), campesterol (2), cycloeucalenol (3), oleanolic acid(4), ursolic acid(5), myrianthic acid(6), euscaphic acid(7), kaempferol(8), quercetin(9), campherol-3-O-β-D-glucoside (10), campherol-3-O-β-D-sophoroside (11), quercetin-3-O-β-D-sophoroside (12), quercetin-7-O-β-D-gentiobioside (13), campherol-3-O-β-D-(6″-O-trans-p-coumaroyl)-glucoside(14), and apigenin-7-O-(6″-O-p-hydroxy benzyl)-β-D-glucoside (15). CONCLUSION: Compounds 2,4,5,6,7,12,13,14 and 15 are obtained from the plant for the first time. Copyright 2012 by the Chinese Pharmaceutical Association.
