Only few studies have focus on animals that received Pilocarpine (Pilo) and did not develop behavioral status epilepticus (SE) and, whether they may become epileptic in the model's chronic phase. Previews works observed mossy fiber sprouting in the hippocampus of Non-SE (NSE) rats, while others observed spontaneous and recurrent seizures (SRS) 6 - 8 months after animals received Pilo. It is known that neuronal excitability is influenced by female hormones, as well as, the occurrence of SE in castrated and non-castrated female rats. However, it is not known whether females that received Pilo and did not show SE, may have SRS. The aim of this work was to investigate whether castrated and non-castrated female rats that did not show behavioral SE after Pilo, will develop SRS in the following one-year. For that, animals received 360 mg/kg of Pilo and were video monitored for 12 months. SE females from castrated and non-castrated groups became epileptic since the first month after drug injection. Epileptic behaviors were identified watching video monitoring recordings in the fast speed. Castrated and Non castrated NSE animals showed behaviors resembling seizures described by Racine Scale stages 1 - 3. Motor alterations showed by NSE groups could be observed only when recordings were analyzed in slow speed. In addition, behavioral manifestations as, rhythmic head movements, sudden head movements, whole body movements and immobility were also observed in both, SE and NSE groups. We concluded that NSE female rats may have become epileptic. Adding to it, slow speed analysis of motor alterations was essential for the observation of NSE findings, which suggests that possibly many motor alterations have been underestimated in epilepsy experimental research.


Poucos são os estudos com foco em animais que receberam Pilocarpina (Pilo) e não desenvolveram status epilepticus (SE) comportamental e, se os mesmos se tornarão epilépticos na fase crônica do modelo. Autores observaram o brotamento das fibras musgosas no hipocampo de ratos Não-SE (NSE), enquanto outros observaram crises espontâneas e recorrentes (CER) 6 - 8 meses após receberam a droga. A excitabilidade neuronal é influenciada pelos hormônios femininos e, da mesma forma, a ocorrência de SE em ratas castradas e não-castradas. Além destas, manifestações comportamentais como movimentos rítmicos da cabeça, movimentos súbitos da cabeça, movimentos de todo o corpo e imobilidade também foram observadas em ambos grupos, SE e NSE. Concluímos que as fêmeas NE podem ter se tornado epilépticas. Adicionado a isto, a análise das alterações motoras na velocidade lenta foi essencial para a observação dos achados das fêmeas NSE, o que sugere que possivelmente muitas alterações motoras têm sido subestimados na pesquisa em epilepsia experimental.

Resumen El consumo crónico de alcohol es un problema de salud mundial que afecta particularmente a la población femenina. Sin embargo, los efectos de la ingesta semicrónica en cantidades moderadas a bajas en el ovario y el oocito son poco conocidos. En un modelo murino, se administró etanol al 10% en agua de bebida (hembras tratadas) o agua (hembras control) por 15 días, y luego de la superovulación o no (ovulación espontánea), se analizó el ciclo estral y la calidad ovárico-gamética. En las hembras tratadas, la frecuencia y duración del diestro aumentó, y las frecuencias de folículos y cuerpos lúteos disminuyeron vs hembras controles, valores que se restauraron luego de la superovulación. Sin embargo, en las hembras tratadas, la tasa de proliferación celular folicular y el desbalance de la expresión ovárica de VEGF (factor de crecimiento endotelial) persistieron luego de la superovulación. El número de ovocitos ovulados con metafase II anormal, fragmentados y activados partenogenéticamente fue mayor en las hembras tratadas respecto las controles. En conclusión, el consumo semicrónico moderado de alcohol produce anestro, ciclo estral irregular, foliculogénesis deficiente y anomalías núcleo-citoplasmáticas en los oocitos ovulados. Estas alteraciones podrían constituirse en un factor etiológico de pérdida gestacional temprana y desarrollo embrionario anormal luego del consumo de alcohol.


Abstract Chronic alcohol consumption is a global health problem that particularly affects the female population. However, the ef-fects of semi-chronic ethanol intake in low-moderate amounts on the ovary and oocyte are poorly understood. In a mouse model, 10% ethanol was administered in drinking water (treated females) or water (control females) for 15 days, and after superovulation or not (spontaneous ovulation), the estrous cycle and ovarian-gametic quality were analyzed. In treated females, the frequency and duration of the diestrus increased, and the frequencies of follicles and corpus luteum decreased vs control females, values that restored after superovulation. However, in treated females, the follicular cell proliferation rate and the imbalance in ovarian expression of VEGF (endothelial growth factor) persisted after superovulation. The number of ovulated oocytes with abnormal metaphase II, fragmented and parthenogenetically activated was higher in treated females than in control ones. In conclusion, moderate semi-chronic alcohol consumption produces anestrum, irregular estrous cycle, poor folliculogenesis, and nuclear-cytoplasmic abnormalities in ovulated oocytes. These alterations could constitute an etiological factor of early gestational loss and abnormal embryonic development after alcohol consumption.

ABSTRACT

La Entomología Forense (EF) es una rama de las ciencias médico-legales (Ortloff et al. 2012), que utiliza como herramienta clave a insectos y otros artrópodos que interactúan con un cuerpo en descomposición (Gennard, 2007). El presente estudio se basó en un diseño observacional descriptivo de tipo poblacional, en el que se evaluó la diversidad de dípteros de la familia Calliphoridae y Muscidae en la parroquia rural de Posorja, Guayaquil. En el proceso experimental en los biomodelos A y B, se obtuvo que las temperaturas mínimas diarias presentaron un promedio de 21 °C (entre 20 °C y 22 °C) en lo que respecta a las temperaturas máximas diarias presentaron un promedio de 29,50 °C (entre 26 y 31 °C). En ambos biomodelos experimentales A y B, la exposición directa a los rayos solares y el espacio de liberación abierto contribuyo con el desarrollo de los cambios post mortem de descomposición cadavérica, como son el cromatismo, hinchado, licuefacción y reducción. A la par, de la ocurrencia de la sucesión de dípteros de interés criminalisticos, pertenecientes a las familias Calliphoridae y Muscidae. La importancia de de este acercamiento a la fauna de interés forense dentro de la provincia de Posorja, se muestra en el abanico de oportunidades que abre, con miras a la futura incorporación de la entomología forense en investigaciones criminalísticas en casos de homicidios y negligencias. Sin embargo, todavía eisten numerosas dificultades dificultades para este tipo de estudio en el Ecuador, por lo que se propone divulgar la escala estacional y espacial de de estas investigaciones, a través de convenios que permitan replicarlos en diversas partes del país en distintos ecosistemas y bajo distintas condiciones climáticas(AU)


Forensic Entomology (PE) is a branch of the medico-legal sciences (Ortloff et al. 2012), which uses insects and other arthropods that interact with a decomposing body as a key tool (Gennard, 2007). The present study was based on a descriptive observational design of a population type, in which the diversity of diptera of the Calliphoridae and Muscidae families in the rural parish of Posorja, Guayaquil was evaluated. In the experimental process in biomodels A and B, it was obtained that the minimum daily temperatures presented an average of 21 ° C (between 20 ° C and 22 ° C) with regard to the maximum daily temperatures presented an average of 29, 50 ° C (between 26 and 31 ° C). In both experimental biomodels A and B, direct exposure to sunlight and the open release space contributed to the development of post-mortem changes in cadaveric decomposition, such as chromaticism, swelling, liquefaction and reduction. At the same time, the occurrence of the succession of diptera of criminalistic interest, belonging to the Calliphoridae and Muscidae families. The importance of this approach to the fauna of forensic interest within the province of Posorja is shown in the range of opportunities that it opens up, with a view to the future incorporation of forensic entomology in criminal investigations in cases of homicides and negligence. However, there are still numerous difficulties for this type of study in Ecuador, so it is proposed to disclose the seasonal and spatial scale of these investigations, through agreements that allow replication in different parts of the country in different ecosystems and under different weather conditions(AU)

ABSTRACT The use of correctly designed animal models is a fundamental step prior to clinical trials in humans. Although rats are easy to house and handle, and have molars that resemble those of humans, very few researchers use them as a model for root canal treatment, probably due to their small size and the lack of relevant data necessary to reproduce the model. Our aims were to describe the anatomic and histologic characteristics of the mandibular first molar of the Wistar rat and present a standardised model for its experimental endodontic treatment. Twenty female rats were used. The characteristics of the mesial and distal roots were described histologically and the quality of the results achieved following the treatment protocol presented herein was assessed by means of digital radiographs, micro-CT and histological sections. The age of 55 days was found to be the most adequate for performing this technique, but we consider the interval of 50 to 60 days to be suitable. Both canals are oval, although in opposite planes, and the furcating-facing walls present the minimum dentine thickness. It was essential to become familiar with these aspects in order to decide upon the most appropriate instrumentation and obturation techniques that would enable replication of this model in basic science research.


RESUMEN El uso de modelos animales correctamente diseñados es un paso fundamental previo al desarrollo de ensayos clínicos en humanos. A pesar de resultar fáciles de criar y manipular y de poseer molares que se asemejan a los humanos, muy pocos grupos utilizan a la rata como modelo experimental para el tratamiento endodóntico probablemente debido a su pequeño tamaño y a la escasa información disponible para poder aplicar los modelos existentes. Nuestros objetivos fueron describir las características anatómicas e histológicas del primer molar inferior de la rata Wistar y presentar un modelo estandarizado para el tratamiento endodóntico experimental de esta pieza. Se utilizaron 20 ratas hembra. Las características de las raíces mesial y distal fueron descritas histológicamente y los resultados obtenidos fueron evaluados mediante radiografías digitales, microCTy cortes histológicos. La edad de 55 días demostró ser la más adecuada para ejecutar la técnica, pero consideramos que el intervalo de 50 a 60 días puede resultar apropiado. Se observó que ambos canales presentan una morfología oval, aunque en direcciones opuestas, y que las paredes furcales resultaron ser las que presentan el menor espesor de dentina. Familiarizarse con estos aspectos de la anatomía e histología del molar de la rata resultó fundamental para decidir sobre las técnicas de preparación y obturación más apropiadas que permitieran la replicación de este modelo en el campo de las ciencias básicas.

INTRODUCTION: The use of a venipuncture simulator facilitates technique learning and improves skills, which reduces the risk of venipuncture complications in humans. OBJECTIVE: To evaluate the efficacy of a non-human experimental model for Ultrasound guided superficial venipuncture. METHODS: We randomized 39 nurses in two groups: A and B. The training had three steps: 1 - theoretical class, 2 - practical class, with the ultrasound device and 3 - ultrasound-guided puncture training in the non-human model. The group A participated in steps 1, 2 and 3 and group B in steps 1 and 2. After training, both groups were released for ultrasound guided venipuncture. RESULTS: The success in puncture in group A (n = 20) was 90% and in group B (n = 19) it was 68.42%. In the sum of the identification and the puncture times, the average of group A was 61.5 seconds (95% CI, 33.58; 106.95) and in group B was 148.0 seconds (95% CI, 114.54; 208.44), which was statistically significant (p = 0.007, without overlapping the interval estimates. CONCLUSION: Nurses who received training with the non-human model had better identification and puncture times.


INTRODUÇÃO: A utilização de um simulador de punção venosa, facilita o aprendizado da técnica e aprimora as habilidades, o que diminui o risco de complicações na punção venosa em humanos. OBJETIVO: Analisar a eficácia de um modelo experimental não humano para punção venosa superficial guiada por ultrassom. MÉTODO: Foram randomizados 39 enfermeiros em dois grupos: A e B. O treinamento apresentou três etapas: 1 - aula teórica, 2 - aula prática no aparelho de ultrassonografia e 3 - treinamento de punção guiada por ultrassonografia no modelo não humano. O grupo A participou das etapas 1, 2 e 3 e o grupo B das etapas 1 e 2. Após o treinamento, ambos os grupos foram liberados para punção venosa guiada por ultrassom. RESULTADOS: O sucesso na punção no grupo A (n = 20) foi de 90% e no grupo B (n = 19) foi de 68,42%. Na somatório dos tempos de identificação e de punção, a média no grupo A foi de 61,5 segundos (IC 95% 33,58; 106,95) e no grupo B de 148,0 segundos (IC95% 114,54; 208,44), o que foi estatisticamente significante (p = 0,007, sem sobreposição das estimativas intervalares. CONCLUSÃO: As enfermeiras que receberam treinamento com o modelo não humano obtiveram melhores tempos de identificação e de punção da veia.

ABSTRACT

Extensive literature is available about the intrinsic denervation of segments of the digestive tube through the application of CB in the serosa of the viscera. However, this technique has some disadvantages like causing peritonitis, flanges and high mortality, limiting its use in humans. The aim of the present study was to evaluate the feasibility of benzalkonium chloride (CB) to induce intrinsic chemical denervation, through applications of CB in the intramural ileum of wistar rats, as well as deepen the knowledge about the evolution of neuronal injury caused in the process. We used 40 rats, divided into two groups (control-GC and benzalkonium-GB) of 20 animals each, divided into four sub-groups according to the time of postoperative assessment of 24, 48 hours, 30 and 90 days. The animals were submitted to intramural microinjections of sterile saline solution 0.9% (GC) or benzalkonium chloride (GB) in ileal portion, and subsequent histopathological analysis and immunohistochemistry for evaluation of neuronal injury. A significant decrease (p<0.05) was found of the neuronal myenteric count over time in groups, GB3, GB4 and GB2. The specific positive immunolabeling for H2AX and Caspase-3 confirmed the results obtained in the histopathological evaluation, denoting the ignition of irreversible cell injury in 24 hours, evolving into neuronal apoptosis in 48 hours after application of the CB 0.3%. Under the conditions in which this work was conducted, it can be concluded that the application of CB 0.3% by means of microinjections intramural in the ileal wall is able to induce intrinsic chemical denervation of the diverticulum of wistar rats and that the main mechanism of neuronal death is induction of apoptosis.(AU)


Existe vasta literatura sobre a desnervação intrínseca de segmentos do tubo digestório através da aplicação de CB na serosa da víscera. Entretanto, essa técnica tem a desvantagem de causar peritonite, formação de bridas e alta mortalidade, não sendo factível para eventuais utilizações em humanos. O objetivo do presente estudo foi avaliar a viabilidade do Cloreto de benzalcônio (CB) induzir desnervação química intrínseca, por meio de aplicações intramurais em íleo de ratos wistar, além de aprofundar o conhecimento sobre a evolução da lesão neuronal causada neste processo. Foram utilizados 40 ratos, distribuídos em dois grupos (controle- GC e benzalcônio- GB) de 20 animais cada, subdivididos em quatro subgrupos de acordo com o tempo de avaliação pós-operatória de 24, 48 horas, 30 e 90 dias. Os animais foram submetidos à microinjeções intramurais de solução salina estéril 0,9% (GC) ou de cloreto de benzalcônio (GB) em porção ileal, e posterior análise histopatológica e imuno-histoquímica, para avaliação da lesão neuronal. Houve diminuição significativa (p<0,05) na contagem neuronal mientérica ao longo do tempo nos grupos GB2, GB3 e GB4. A imunomarcação específica positiva para H2AX e Caspase-3 confirmou os resultados obtidos na avaliação histopatológica, denotando início da lesão celular irreversível em 24 horas, evoluindo para apoptose neuronal em 48 horas após a aplicação do CB 0,3%. Nas condições em que este trabalho foi conduzido, é possível concluir que a aplicação de CB 0,3% por meio de microinjeções intramurais na parede ileal é capaz de induzir desnervação química intrínseca da porção ileal de ratos wistar e que o principal mecanismo de morte neuronal é a indução de apoptose.(AU)

ABSTRACT

A adiponectina foi originalmente identificada como uma proteína expressa e produzida por adipócitos do tecido gorduroso abdominal de várias espécies animais, cuja função é manter a homeostase do tecido adiposo em relação às demandas energéticas do corpo. Preparações homeopáticas de substâncias endógenas são capazes de modular diferentes funções celulares. Neste projeto, buscou-se conhecer os possíveis efeitos de uma preparação homeopática de anticorpos antiadiponectina no controle do depósito adiposo subcutâneo abdominal, utilizandose modelo experimental. Quarenta camundongos Swiss machos adultos oram divididos em um grupo experimental e 3 controles. Os animais foram alimentados durante 1 mês e 7 dias com suplemento dietético comercial e tratados simultaneamente, por via oral, com antiadiponectina 6cH. Foi avaliado o ganho de peso corporal, bem como características macro e microscópicas do tecido adiposo subcutâneo e do fígado. Os resultados mostraram discreta redução no ganho de peso e redução estatisticamente significativa no diâmetro dos adipócitos no grupo experimental. Não foram observadas alterações inflamatórias ou degenerativas significativas no fígado. Os mecanismos envolvidos nestas alterações metabólicas precisam ser pesquisados em estudos futuros. (AU)


Adiponectin was originally identified as a protein expressed and produced by the abdominal fat adipocytes of several animal species which function is to maintain the homeostasis of the adipose tissue vis-à-vis the body energy demands. Homeopathic preparations of endogenous substances are able to modulate several cell functions. In the present study we investigated possible effects of a homeopathic preparation of anti-adiponectin antibodies on the control of the abdominal fat by means of an experimental model. Forty Swiss male adult mice were fed a commercial dietary supplement for 1 month and 7 days and treated with anti-adiponectin 6cH per oral route. Parameters assessed were weight gain and macro- and microscopic characteristics of the subcutaneous adipose tissue and liver. The experimental group exhibited slightly less weight gain and statistically significant reduction of the adipocyte diameter. We did not detect significant inflammatory or degenerative changes in the liver. The mechanisms involved in the detected changes need to be investigated in future studies. (AU)

ABSTRACT

Brachiaria spp. are important sources of forage for ruminants in Brazil, due to the easy cultivation, good resistance to drought, good adaptation to different soils and low maintenance cost. However, the ingestion of this grass has been related to photosensitization outbreaks in cattle and sheep with significant economic losses. The hepatotoxic effects related to the ingestion of grass are the formation of crystals and foamy macrophages due to the accumulation of toxic metabolites. The use of cattle and sheep in experiments involving the plant presents several obstacles in the ethical, economic and animal management. The objective of this study was to evaluate the sensitivity of rabbits as an experimental model for B. decumbens poisoning. Two experiments were carried out. In Experiment 1 four rabbits received the fresh plant in daily doses of 10, 20, 40 and 80g/kg body weight for 120 days. In Experiment 2 three rabbits received the fresh plant in amounts of 500g daily with duration of 210 days. The animals of Experiment 1 showed no clinical signs and no macroscopic and microscopic changes characteristic of B. decumbens poisoning. In Experiment 2 the animals also showed no clinical signs or significant macroscopic alterations. Histological analysis showed isolated foamy macrophages or present in random groups of cells in the liver and mesenteric lymph nodes. Samples of liver and mesenteric lymph nodes of the rabbits of Experiment 2 were submitted to the lectin-histochemistry technique. The WGA, sWGA and RCA lectins showed reactivity in foamy macrophages in both organs. This is the first study of our knowledge that demonstrates histopathological lesions caused expetimentally by Brachiaria spp. in rabbits, demonstrating its potential as an animal model.(AU)


Brachiaria ssp. são importantes fontes de forragem para ruminantes no Brasil, devido ao fácil cultivo, boa resistência a seca, boa adaptação a diferentes solos e baixo custo de manutenção. Entretanto, a ingestão desta gramínea está relacionada a surtos de fotossensibilização, em bovinos e ovinos, principalmente, ocasionando prejuízos econômicos significativos. Os efeitos hepatotóxicos relacionados à ingestão da gramínea são a formação de cristais e macrófagos espumosos causados pelo acúmulo de metabólitos tóxicos. A utilização de bovinos e ovinos em experimentos envolvendo a planta apresenta vários empecilhos, tanto no âmbito ético, econômico e no manejo dos animais. O objetivo do presente trabalho foi avaliar a sensibilidade de coelhos como modelo experimental para intoxicação por B. decumbens. No presente estudo foram realizados dois experimentos. O Experimento 1 utilizou quatro coelhos que receberam a planta fresca em doses diárias de 10, 20, 40 e 80 g/Kg de peso vivo durante 120 dias. O Experimento 2 utilizou três coelhos recebendo a planta fresca em quantidades de 500g diárias por animal com duração de 210 dias. No Experimento 1, os animais não apresentaram sinais clínicos e nem alterações macroscópicas e microscópicas características de intoxicação por B. decumbens. No Experimento 2 os animais também não apresentaram sinais clínicos e alterações macroscópicas significativas. Na análise histológica observou-se presença de macrófagos espumosos isolados ou em grupos aleatórios de células no fígado e nos linfonodos mesentéricos. Amostras de fígado e linfonodos mesentéricos dos animais do Experimento 2 foram submetidos à técnica de lectino-histoquímica. As lectinas WGA, sWGA e RCA apresentaram reatividade em macrófagos espumosos nos dois órgãos. Este é o primeiro trabalho de nosso conhecimento que demonstra lesões histopatológicas por Brachiaria spp conduzido de forma experimental em coelhos, demonstrando seu potencial como modelo animal nesse campo de estudo.(AU)

ABSTRACT

The present study aimed to establish a methodology capable to cause intestinal ischemia and reperfusion injuries, to perform clamping of the jejunal segment of the extramural peri-intestinal marginal artery branch. For this, 37, 10-week-old male New Zealand breed rabbits were used. One rabbit was used to establish the anatomic references for the procedure and was not part of the six experimental groups; the rest were allocated into six experimental groups: Sham group, negative control, subjected only to midline celiotomy; group I1H undergoing vascular occlusion for an hour; group I2H submitted to vascular occlusion for two hours; group I1H/R2H undergoing vascular occlusion for one hour followed by two hours of reperfusion; group I2H/R1H undergoing vascular occlusion for two hours, followed by reperfusion for one hour, and group I2H/R5H undergoing vascular occlusion for two hours followed by reperfusion for five hours. The rabbits were evaluated for the macroscopic aspects (color and peristalsis) of the jejunal segment, as well as the histological aspect, checking for presence or absence of mucosal destruction, edema, hemorrhaging, lymphatic vessel dilatation, and the presence of polymorphonuclear cells. It was observed that the macroscopic and histopathological lesions accentuated in larger employed ischemia and reperfusion times. Rabbits subjected to ischemia for two hours followed by reperfusion for five hours (I2H/R5H) made up the experimental group which was easily reproducible and showed moderate intestinal injury, different from the other groups.(AU)


O presente estudo objetivou estabelecer uma metodologia capaz de causar lesões de isquemia e reperfusão intestinal, realizando clipagem de um ramo de artéria marginal peri-intestinal extramural em segmento jejunal. Para tal foram utilizados 37 coelhos da raça Nova Zelândia, machos, de 10 semanas de idade, alocados em seis grupos experimentais: grupo Sham, controle negativo, submetido apenas a celiotomia mediana; grupo I1H submetido à oclusão vascular por uma hora; grupo I2H submetido a oclusão vascular por duas horas; grupo I1H/R2H submetido a oclusão vascular por uma hora, seguida de reperfusão por duas horas; grupo I2H/R1H submetido a oclusão vascular por duas horas, seguida de reperfusão por uma hora e grupo I2H/R5H submetido a oclusão vascular por duas horas seguida de reperfusão por cincos horas. Os animais foram avaliados quanto o aspecto macroscópico (coloração e peristaltismo) do segmento jejunal e quanto ao aspecto histopatológico, verificando presença ou ausência de destruição de mucosa, edema, hemorragia, dilatação de vasos linfáticos e presença de polimorfonucleares. Observou-se que as lesões macroscópicas e histopatológicas se acentuaram nos maiores tempos de isquemia e reperfusão empregados. Os animais submetidos à isquemia durante duas horas, seguida de reperfusão por cinco horas (I2H/R5H) compuseram o grupo experimental de fácil reprodução e foram os que apresentaram uma lesão intestinal moderada, diferentes dos demais grupos.(AU)

ABSTRACT Objective: To analyze, from the immunohistochemical perspective, the effects of hyaluronic acid of different molecular weights in an experimental model of osteoarthritis in rabbits. Methods: Forty-four male California rabbits were randomly assigned to three different groups (PR, S, and P) and submitted to the resection of the anterior cruciate ligament of the right knee. Three weeks after the surgical procedure, three intra-articular weekly injections were carried out with low-molecular-weight native hyaluronic acid (Hyalgan®) to PR group, high molecular weight branched chain hyaluronic acid (Synvisc®) to group S, and saline solution 0.9% to group P. All animals were sacrificed 12 weeks after the surgical procedure, and the tibial plateaus of the infiltrated knees were then dissected. Histological sections of cartilage from the tibial plateau support areas were stained with immunohistochemical markers in order to investigate the amount of metalloproteases (MMPs 3 and 13) and their inhibitors (TIMPs 1 and 3). The staining intensity was quantified on a Zeiss Imager.Z2 Metasystems microscope and analyzed by Metafer4 Msearch software. Results: The chondroprotective effect of the hyaluronic acids used in the study was demonstrated when compared to the control group. However, the comparison between them presented no significant statistical difference regarding chondroprotection. Conclusion: The injection of saline solution demonstrated signs of OA development, while adding native hyaluronic acid of low molecular weight (Hyalgan®) and hyaluronic acid of high molecular weight (Synvisc®) protected the articular cartilage in this model of OA.


RESUMO Objetivo: Analisar do ponto de vista imuno-histoquímico os efeitos do ácido hialurônico de diferentes pesos moleculares em modelo experimental em coelhos. Métodos: Foram alocados de modo aleatório 44 coelhos da raça California, machos, em três grupos (PR, S e P), e submetidos à ressecção do ligamento cruzado anterior do joelho direito. Decorridas três semanas do procedimento cirúrgico iniciaram-se as três injeções intra-articulares semanais de ácido hialurônico nativo de baixo peso molecular (Polireumin®) no grupo PR, ácido hialurônico de cadeia ramificada de alto peso molecular (Synvisc®) no grupo S e soro fisiológico 0,9% no grupo P. Todos os animais foram sacrificados após 12 semanas do ato cirúrgico e os platôs tibiais dos joelhos infiltrados foram dissecados. Cortes histológicos da cartilagem das áreas de apoio dos platôs tibiais foram corados com marcadores imuno-histoquímicos para pesquisa da quantidade de metaloproteases (MMPs-3,13) e seus inibidores (TIMPs-1,3). A intensidade de coloração foi quantificada em um aparelho de microscopia Zeiss Imager.Z2 Metasystems e analisada pelo software Metafer4 Msearch. Resultado: O efeito condroprotetor dos ácidos hialurônicos usados no estudo foi demonstrado quando comparados com o grupo controle, porém feita a comparação entre si não houve diferença estatística significante quanto à condroproteção. Conclusão: A injeção de solução salina demonstra sinais de desenvolvimento de OA enquanto que a adição de ácido hialurônico nativo de baixo peso molecular (Polireumin®) e ácido hialurônico de cadeia ramificada de alto peso molecular (Synvisc®) protegeram a cartilagem articular nesse modelo de OA.

Abstract Helminth parasites have been studied as potential accumulators for different pollutants. Echinostoma paraensei is a foodborne trematode whose vertebrate host, the rodent Nectomys squamipes, is naturally exposed to environmental pesticides. However, little information exists regarding the pesticide's effects on helminths. This study investigated the morphological effects on the trematode, E. paraensei, after experimental Roundup® herbicide exposure, in concentrations below those recommended for agricultural use. After two hours of exposure, scanning electron microscopy (SEM) showed changes to the tegument, such as furrowing, shrinkage, peeling, spines loss on the peristomic collar, and histopathological evidence of altered cells in the cecum and acinus vitelline glands with vacuoles and structural changes to the muscular layers. Glycidic content was decreased, primarily in the connective tissue. As E. paraensei is an intestinal parasite of the semi-aquatic wild rodent, N. squamipes, it is predisposed to pesticide exposure resulting from agricultural practices. Therefore, we emphasize the need to evaluate its impact on helminth parasites, due to their pivotal role in regulating host populations.


Resumo Helmintos parasitos tem sido estudados como acumuladores potenciais para diferentes poluentes. O trematódeo E. paraensei tem como hospedeiro vertebrado o roedor Nectomys squamipes naturalmente exposto a pesticidas no meio ambiente. No entanto, pouca informação está disponível sobre os efeitos dos pesticidas em helmintos parasitos. O presente estudo investigou, em condições experimentais, os efeitos morfológicos no trematódeo E. paraensei após a exposição ao herbicida Roundup®, em concentrações abaixo das recomendadas para a utilização agrícola. A microscopia eletrônica de varredura (MEV) mostrou após duas horas de exposição, alterações no tegumento, como enrugamento, contração e descamação com perda de espinhos no colar peristômico e análise histopatológica evidenciou células do ceco alteradas, as glândulas vitelínicas com vacúolos e mudanças estruturais nas camadas musculares. Diminuição do conteúdo glicídico, principalmente no tecido conjuntivo, também foi observado. Considerando a predisposição à exposição a pesticidas agrícolas de N. squamipes infectado por E. paraensei, são necessários estudos para avaliar o impacto de tais resíduos frente aos helmintos e seus hospedeiros.

ABSTRACT

INTRODUCTION: Cerebrospinal fluid (CSF) from amyotrophic lateral sclerosis (ALS) patients induces cytotoxic effects in in vitro cultured motor neurons. MATERIAL AND METHODS: We selected CSF with previously reported cytotoxic effects from 32 ALS patients. Twenty-eight adult male rats were intracerebroventricularly implanted with osmotic mini-pumps and divided into 3 groups: 9 rats injected with CSF from non-ALS patients, 15 rats injected with cytotoxic ALS-CSF, and 4 rats injected with a physiological saline solution. CSF was intracerebroventricularly and continuously infused for periods of 20 or 43days after implantation. We conducted clinical assessments and electromyographic examinations, and histological analyses were conducted in rats euthanised 20, 45, and 82days after surgery. RESULTS: Immunohistochemical studies revealed tissue damage with similar characteristics to those found in the sporadic forms of ALS, such as overexpression of cystatinC, transferrin, and TDP-43 protein in the cytoplasm. The earliest changes observed seemed to play a protective role due to the overexpression of peripherin, AKTpan, AKTphospho, and metallothioneins; this expression had diminished by the time we analysed rats euthanised on day 82, when an increase in apoptosis was observed. The first cellular changes identified were activated microglia followed by astrogliosis and overexpression of GFAP and S100B proteins. CONCLUSION: Our data suggest that ALS could spread through CSF and that intracerebroventricular administration of cytotoxic ALS-CSF provokes changes similar to those found in sporadic forms of the disease.

ABSTRACT

Con una prevalencia global reportada de entre 11-13%, la enfermedad renal crónica (ERC) ha sido reconocida como un gran desafío para los sistemas de salud por sus implicaciones económicas y sociales. Al tratarse de una enfermedad crónica e irreversible, el tratamiento está dirigido a disminuir su progresión. La cuantificación de creatinina sérica es el método de elección para su diagnóstico y clasificación; sin embargo, es conocido que esta prueba tiene una sensibilidad clínica limitada, lo que ha conducido a la búsqueda de nuevos marcadores que permitan un diagnóstico y monitoreo oportuno. Desde esta perspectiva, el empleo de la metabolómica y de modelos animales ha permitido la identificación y estudio de nuevos metabolitos, candidatos a ser utilizados como futuros biomarcadores en la práctica clínica. La presente revisión tuvo como objetivo hacer una análisis de los perfiles metabolómicos reportados para la ERC, tanto en modelos experimentales como en estudios realizados en seres humanos. De acuerdo a los datos obtenidos, los metabolitos implicados en las rutas metabólicas de aminas cuaternarias y aminoácidos como el TMNO, el indoxilsulfato y derivados de la dimetilarginina representan una alternativa prometedora para la identificación, clasificación y pronóstico de la ERC.


Chronic kidney disease (CKD) high global prevalence, estimated between 11 to 13%, has been recognized as a mayor health challenge for healthcare systems due to its relevant economic and social implications. Main medical intervention strategies are directed to delay the progression of CKD and prevent outcomes. Serum creatinine concentration has been used to classify CKD and define its progression stage; however, it is well known the low sensitivity shown by this test. This fact has conducted to the search for new markers in order to improve the disease diagnosis, monitoring and treatment. In this context, metabolomics science and animal models have allowed identification of new metabolites that can be used as future biomarkers into clinical practice. This review aims to summarize the metabolomics profiles reported in different experimental models and clinical research on CKD. According with the data obtained, metabolites related with quaternary amines and aminoacid metabolic pathways like TMNO, indoxyl sulfate and dimethylarginine, suggest a promising alternative for identification, classification and prognosis of CKD.

INTRODUCTION: The objective of this study was to determine the reproducibility in a murine model of renal tumours of various histological strains that could be useful for investigating the response to target drugs. MATERIAL AND METHODS: Development and analysis of the "in vivo" model: tumour xenograft of renal cell carcinomas with Balb/c nude athymic mice. Nontumourous human renal tissue was implanted in the interscapular region of 5 mice, chromophobe renal cell carcinoma was implanted in 5 mice (which, after checking its growth, was prepared for implantation in another 10 mice) and Fuhrman grade 2 clear cell renal cell carcinoma (CCRCC) was implanted in 5 mice (which was also subsequently implanted in 10 mice). We monitored the tumour size, onset of metastases and increase in size and number of tumours. When the size had reached a point greater than or equal to locally advanced or metastatic carcinoma, the animals were euthanised for a pathological and immunohistochemical study and a second phase of implantation. RESULTS: The subcutaneous xenograft of the healthy tissue did not grow. The animals were euthanised at 6 months and no renal tissue was found. The chromophobe renal cell carcinoma cells grew in the initial phase (100%); however, in the second phase, we observed a chronic lymphomonocyte inflammatory reaction and a foreign body reaction. The CCRCC grew at 5-8 months both in the first and second phase (100%), maintaining the tumour type and grade. CONCLUSIONS: The model with athymic Balb/c nude mice is useful for reproducing CCRCC, with the same histological characteristics and aggressiveness as native human tumours, promoting the development of the second experimental phase.

BACKGROUND: Microorganisms have been widely studied as biological control agents of parasites of medical and veterinary importance. Coprophagous arthropods, bacteria and fungi are among the different organisms evaluated as potential biological control agents. Nematophagous fungi capture and digest the free forms of nematodes in the soil. Due to its zoonotic potential, Toxocara canis have been brought to the attention of researchers. AIMS: The aim of the present study was to determine whether the administration of embryonated T. canis eggs exposed to the nematophagous fungus Trichoderma virens reduces parasite infection in experimental animals. METHODS: Embryonated T. canis eggs were exposed to T. virens mycelium for 15 days at 25°C. Subsequently, 100 fungus-exposed eggs were orally administered to 20 Swiss mice. As a positive control, another 20 mice received 100 embryonated eggs that were not exposed to the fungus. After 48h, the animals were killed, and heart, lungs and liver were harvested for the recovery of larvae. RESULTS: The organs of the animals that received embryonated T. canis eggs exposed to the fungus showed a lower mean larval recovery when compared with the animals that received embryonated eggs without fungus exposure (p<0.05). CONCLUSIONS: The exposure of T. canis eggs to T. virens reduces the experimental infection, demonstrating the potential of this nematophagous fungus as a biocontrol agent.

OBJECTIVE: Cerebral vasospasm, one of the main complications of subarachnoid hemorrhage (SAH), is characterized by arterial constriction and mainly occurs from day 4 until the second week after the event. Urotensin-II (U-II) has been described as the most potent vasoconstrictor peptide in mammals. An analysis is made of the serum U-II concentrations and mRNA expression levels of U-II, urotensin related peptide (URP) and urotensin receptor (UT) genes in an experimental murine model of SAH. DESIGN: An experimental study was carried out. SETTING: Experimental operating room of the Biomedicine Institute of Seville (IBiS), Virgen del Rocío University Hospital (Seville, Spain). PARTICIPANTS: 96 Wistar rats: 74 SAH and 22 sham intervention animals. INTERVENTIONS: Day 1: blood sampling, followed by the percutaneous injection of 100µl saline (sham) or blood (SAH) into the subarachnoid space. Day 5: blood sampling, followed by sacrifice of the animals. MAIN VARIABLES OF INTEREST: Weight, early mortality, serum U-II levels, mRNA values for U-II, URP and UT. RESULTS: Serum U-II levels increased in the SAH group from day 1 (0.62pg/mL [IQR 0.36-1.08]) to day 5 (0.74pg/mL [IQR 0.39-1.43]) (p<0.05), though not in the sham group (0.56pg/mL [IQR 0.06-0.83] day 1; 0.37pg/mL [IQR 0.23-0.62] day 5; p=0.959). Between-group differences were found on day 5 (p<0.05). The ROC analysis showed that the day 5 serum U-II levels (AUC=0.691), URP mRNA (AUC=0.706) and UT mRNA (AUC=0.713) could discriminate between sham and SAH rats. The normal serum U-II concentration range in rats was 0.56pg/mL (IQR 0.06-0.83). CONCLUSION: The urotensinergic system is upregulated on day 5 in an experimental model of SAH.

ABSTRACT

Background: Non-alcoholic fatty liver disease (NAFLD) is a spectrum of hepatic lesions, ranging from benign intrahepatic lipid accumulation (steatosis) to progressive non-alcoholic steatohepatitis, in absence of other known secondary causes. Both insulin resistance and oxidative stress have been involved in NAFLD development and progression and, therefore, insulin-sensitizers and/or antioxidants have been targets of different therapeutic agents. Some natural compounds such as Aristotelia chilensis have a high content of polyphenols, which are known for their antioxidant and anti-inflammatory properties. Objective: To assess the effect of a purified anthocyanin-rich extract of maqui (Aristotelia chilensis) on experimental model of NAFLD. Methods: C57BL6 mice were separated in four experimental groups (n = 4-10) and fed a control diet (chow) or a high fat diet (HFD) with or without a purified anthocyanin-rich extract of Aristotelia chilensis (ACnE) (400 mg/kg/day diluted in drinking water). The hepatic effects of HFD were assessed measuring serum levels of glucose and aminotransferases, hepatic histology and triglycerides. Results: HFD diet induced an increase in hepatic triglycerides and histological NAFLD. Administration of ACnE did not affect serum aminotransferases, hepatic triglycerides, liver weight or histological NAFLD. Conclusion: Administration of an ACnE showed no effects on NAFLD in the HFD experimental model.


Introducción: El hígado graso no alcohólico (HGNA) constituye un espectro de lesiones hepáticas, desde la acumulación lipídica intrahepática benigna (esteatosis) hasta la esteatohepatitis no-alcohólica progresiva, en ausencia de causas secundarias conocidas. En el desarrollo y la progresión del HGNA se ha involucrado la resistencia a insulina y el estrés oxidativo y, por lo tanto, insulino-sensibilizantes y antioxidantes han sido blancos de diferentes agentes terapéuticos. Algunos compuestos naturales como la Aristotelia chilensis (maqui) tienen un alto contenido de polifenoles, los que presentan propiedades antiinflamatorias y antioxidantes. Objetivo: Evaluar el efecto de un extracto purificado rico en antocianinas (EACn) del fruto del maqui (Aristotelia chilensis) sobre la esteatosis, en un modelo experimental de HGNA. Métodos: Los ratones fueron distribuidos en 4 grupos (n = 4-10). Dos alimentados con una dieta estándar (grupo control) y dos con una dieta alta en grasa- high fat diet (grupo HFD). Un grupo control y uno HFD recibieron además 400 mg/kg/día de EACn (grupo EACn). Se determinaron los niveles séricos de aminotransferasas y glucosa; se evaluó la histología hepática y el contenido hepático de triglicéridos. Resultados: HFD indujo aumento de triglicéridos hepáticos e HGNA histológico. La administración de EACn no modificó las transaminasas séricas, los triglicéridos hepáticos, el peso del hígado ni el HGNA histológico. Conclusiones: La administración de un EACn no mostró efectos en el modelo experimental de HGNA inducido por una dieta alta en grasa.

ABSTRACT

Introducción: El hipoclorito de sodio (NaOCl) es el agente químico más utilizado como solución irrigadora durante la terapia endodóntica. Es altamente tóxico cuando se extruye a tejido periapicales. En endodoncia la hemolisis causada por el NaOCl ha quedado demostrada utilizando diferentes modelos, sin embargo poca o ninguna evidencia se tiene de las alteraciones morfológicas en la membrana celular de los eritrocitos. Objetivo: Proponer un modelo experimental que permita evaluar las alteraciones morfológicas que sufren los eritrocitos cuando son expuestos a NaOCl utilizado en la práctica odontológica mediante microscopia electrónica de barrido de alta resolución (MEB). Material y métodos: Se obtuvieron 20 mL de sangre periférica y se depositaron en tubos con anticoagulante EDTA (ácido etilendiaminotetraacético). Se realizaron lavados con solución amortiguadora de fosfatos (solución Evan's). Se prepararon diferentes diluciones de la muestra de eritrocitos (1:1, 1:2, 1:4, 1:8 y 1:16). Se obtuvieron 100 μL de cada una de estas diluciones y se confrontaron con 100 μL de NaOCl 5.25% de uso odontológico (Viarzoni-T, Medental®). Se tomaron 0.5 μL de estas muestras para depositarse en un portamuestra de aleación Zn-Cu, el cual se sometió a un proceso de metalización de baño de iones de Cu por el método antiguo llamado Sputtering. Obteniendo microfotografías por MEB. Resultados: Se lograron observar eritrocitos con alteración de tipo anisocitosis y poiquilocitosis (estomatocitos, eliptocitos, esferocitos y discocitos). También se observaron algunas características estructurales de cristales de NaOCl. Conclusión: Este modelo experimental permitió evaluar los cambios morfológicos que sufren los eritrocitos cuando son expuestos a NaOCl 5.25%.


Introduction: Sodium hypochlorite (NaOCl) is the chemical agent most frequently used as irrigation solution during endodontic therapy. When extruded to periapical tissue, it is highly toxic. In endodontics, hemolysis caused by NaCOl has been proven using different models, nevertheless, there is little or no evidence of morphological alterations in the cellular membrane of erythrocytes. Objective: To propose an experimental model which might allow to assess morphological alterations suffered by erythrocytes when they are exposed to NaOCl used in the dental practice by means of high resolution scanning electron microscopy (SEM). Materials and methods: In the present study, 20 mL of peripheral blood were obtained and deposited in tubes with EDTA (ethylenediaminetetraacetic acid) anticoagulant. Rinses were conducted with a phosphate buffer solution (Evan's solution). Several dilutions of the erythrocyte sample were prepared (1:1, 1:2, 1:4, 1:8 and 1:16); 100 μL of each of these dilutions was obtained to be then confronted with 100 μL of dental use 5.25% NaOCl (Viarzoni-T, Medental®); 0.5 μL of these samples were taken to then be deposited in a sample holder made of Zn-Cu alloy which was subjected to a process of Cu ion metallization bath, following the old Spluttering method. Microphotographs were obtained with SEM. Results: Erythrocytes with alteration type anisocytosis and poikilocytosis (stomatocytes, elliptocytes and discocytes) were observed. Some structural characteristics of NaOCl crystals were equally observed. Conclusion: This experimental model allowed assessment of morphological changes experienced by erythrocytes when exposed to 5.25% NaOCl.
