RESUMO

Canastra's Minas artisanal cheese [QMA (Minas artisanal cheese)] is a protected geographical indication traditional food. The influence of fungi on the cheese ripening process is of great importance. This study aimed to apply culture-dependent and -independent methods to determine the mycobiota of QMA produced in the Canastra region, as well as to determine its physicochemical characteristics. Illumina-based amplicon sequencing and matrix-assisted laser desorption ionization time-of-flight mass spectrometry were the culture-independent methods used. The physicochemical analysis results showed that the QMA has a moisture content ranging 18.4-28.2%, fat content ranging 20.5-40%, sodium chloride percentage of approximately 0.9%, and pH ranging 5.2-5.5. The population of fungi ranged between 6.3 and 8 log colony-forming unit/g. Fusarium spp., Geotrichum candidum, Paecilomyces spp., Trichosporon coremiiforme, Candida catenulata, Aspergillus spp., Trichosporon japonicum, Aspergillus oryzae, Kluyveromyces spp., Torulaspora spp., and Debaryomyces spp. were the most prevalent fungi. The methods used to evaluate the mycobiota provide a better understanding of which species are present in the final product and eventually contribute to the characteristics of QMA. Geotrichum candidum and C. catenulata were identified as promising species for future studies on product quality.

The objective was to estimate (co)variance functions using random regression models (RRM) with Legendre polynomials, B-spline function and multi-trait models aimed at evaluating genetic parameters of growth traits in meat-type quail. A database containing the complete pedigree information of 7000 meat-type quail was utilized. The models included the fixed effects of contemporary group and generation. Direct additive genetic and permanent environmental effects, considered as random, were modeled using B-spline functions considering quadratic and cubic polynomials for each individual segment, and Legendre polynomials for age. Residual variances were grouped in four age classes. Direct additive genetic and permanent environmental effects were modeled using 2 to 4 segments and were modeled by Legendre polynomial with orders of fit ranging from 2 to 4. The model with quadratic B-spline adjustment, using four segments for direct additive genetic and permanent environmental effects, was the most appropriate and parsimonious to describe the covariance structure of the data. The RRM using Legendre polynomials presented an underestimation of the residual variance. Lesser heritability estimates were observed for multi-trait models in comparison with RRM for the evaluated ages. In general, the genetic correlations between measures of BW from hatching to 35 days of age decreased as the range between the evaluated ages increased. Genetic trend for BW was positive and significant along the selection generations. The genetic response to selection for BW in the evaluated ages presented greater values for RRM compared with multi-trait models. In summary, RRM using B-spline functions with four residual variance classes and segments were the best fit for genetic evaluation of growth traits in meat-type quail. In conclusion, RRM should be considered in genetic evaluation of breeding programs.

This study aimed to evaluate the rheological behavior and thermal conductivity of dairy products, composed of the same chemical components but with different formulations, as a function of temperature. Subsequently, thermal conductivity was related to the apparent viscosity of yogurt, fermented dairy beverage, and fermented milk. Thermal conductivity measures and rheological tests were performed at 5, 10, 15, 20, and 25°C using linear probe heating and an oscillatory rheometer with concentric cylinder geometry, respectively. The results were compared with those calculated using the parallel, series, and Maxwell-Eucken models as a function of temperature, and the discrepancies in the results are discussed. Linear equations were fitted to evaluate the influence of temperature on the thermal conductivity of the dairy products. The rheological behavior, specifically apparent viscosity versus shear rate, was influenced by temperature. Herschel-Bulkley, power law, and Newton's law models were used to fit the experimental data. The Herschel-Bulkley model best described the adjustments for yogurt, the power law model did so for fermented dairy beverages, and Newton's law model did so for fermented milk and was then used to determine the rheological parameters. Fermented milk showed a Newtonian trend, whereas yogurt and fermented dairy beverage were shear thinning. Apparent viscosity was correlated with temperature by the Arrhenius equation. The formulation influenced the effective thermal conductivity. The relationship between the 2 properties was established by fixing the temperature and expressing conductivity as a function of apparent viscosity. Thermal conductivity increased with viscosity and decreased with increasing temperature.

Our objective was to evaluate changes in breeding values for carcass traits of two meat-type quail (Coturnix coturnix) strains (LF1 and LF2) to changes in the dietary (methionine + cystine):lysine ([Met + Cys]:Lys) ratio due to genotype by environment (G × E) interaction via reaction norm. A total of 7000 records of carcass weight and yield were used for analyses. During the initial phase (from hatching to day 21), five diets with increasing (Met + Cys):Lys ratios (0.61, 0.66, 0.71, 0.76 and 0.81), containing 26.1% crude protein and 2900 kcal ME/kg, were evaluated. Analyses were performed using random regression models that included linear functions of sex (fixed effect) and breeding value (random effect) for carcass weight and yield, without and with heterogeneous residual variance adjustment. Both fixed and random effects were modelled using Legendre polynomials of second order. Genetic variance and heritability estimates were affected by both (Met + Cys):Lys ratio and strain. We observed that a G × E interaction was present, with changes in the breeding value ranking. Therefore, genetic evaluation for carcass traits should be performed under the same (Met + Cys):Lys ratio in which quails are raised.

O presente experimento foi conduzido para determinar as exigências de lisina digestível para frangos de corte (linhagem Redbro), machos e fêmeas, durante o período de crescimento. Foram utilizados 630 frangos, alojados em 30 boxes com acesso à área de pastejo. O delineamento experimental utilizado foi o inteiramente ao acaso, em esquema fatorial 5x2 (lisina e sexo), e três repetições com 21 aves cada. Os níveis de lisina digestível avaliados foram: 7,07; 8,07; 9,07; 10,07 e 11,07g/kg. Avaliou-se o consumo de lisina, o consumo de ração, o ganho de peso e a conversão alimentar. Observou-se efeito de interação (P<0,05) entre os níveis de lisina e sexo para o ganho de peso. Houve efeito (P<0,01) dos níveis de lisina sobre o consumo de lisina, o ganho de peso e a conversão alimentar. Verificou-se que todas as variáveis foram influenciadas (P<0,05) pelo sexo dos frangos. Recomendam-se 10,08 e 9,49g de lisina digestível/kg de ração, correspondendo ao consumo estimado de 21 e 20g de lisina, para machos e fêmeas, respectivamente, para melhor ganho de peso das aves. Para melhor conversão alimentar, recomendam-se 9,61g de lisina digestível/kg, para frangos de ambos os sexos, correspondendo ao consumo estimado de 20g de lisina.(AU)


This experiment was conducted to determine the requirements of digestible lysine in male and female free-range broiler chickens (Redbro lineage) during the growth phase. Six hundred and thirty broiler chickens were allocated in 30 pens with access to the pasture area. The experimental design was completely randomized in a 5x2 (lysine and gender) factorial arrangement with 3 repetitions with 21 chickens each. The levels of digestible lysine evaluated were: 7.07, 8.07, 9.07, 10.07 and 11.07g/kg. The performance traits evaluated were lysine intake, feed intake, weight gain and feed conversion ratio. An interaction effect (P<0.05) was observed between dietary lysine level and gender for body weight gain. There was an effect (P<0.01) of the levels of lysine on lysine intake, body weight gain and feed conversion ratio. It was found that all the variables were affected (P<0.05) for the gender of chickens. We recommended 10.08 and 9.49 g of digestible lysine/kg diet, corresponding to the estimated intake of 21 and 20g of lysine, for males and females, respectively, for better body weight gain of chickens. For better feed conversion ratio, the recommendation is of 9.61 g of lysine/kg of diets, for chickens of both genders, corresponding to the estimated intake of 20g of lysine.(AU)

Objetivou-se avaliar a sensibilidade dos valores genéticos para características de qualidade da carne em codornas de corte alimentadas com dietas contendo diferentes relações de (metionina + cistina): lisina, do nascimento aos 21 dias de idade, por meio de modelos de normas de reação. Utilizaram-se 9011 informações de qualidade de carne referentes a 1400 progênies de 80 reprodutores e 160 matrizes de duas linhas (LF1 e LF2). Para o ajuste dos modelos de regressão aleatória, foi usado o programa WOMBAT, considerando-se nas análises homogeneidade de variância residual. As codornas foram alimentadas do nascimento aos 21 dias de idade com dietas contendo as relações 0,61; 0,66; 0,71; 0,76 e 0,81 de (metionina + cistina): lisina, mantendo os níveis de proteína bruta de 26,12% e de energia em 2900 kcal EM/kg da dieta. Dos 22 aos 35 dias de idade, todas as codornas foram alimentadas com dieta contendo 22% de proteína bruta e 3050 kcal EM/kg da dieta. As estimativas da variância genética e da herdabilidade foram influenciadas pelo gradiente ambiental e pela linha, com mudanças nessas estimativas com o aumento do gradiente ambiental. Os valores genéticos das características de qualidade de carne referentes a cada uma das linhas se alteraram com o aumento das relações de aminoácidos das dietas em razão das mudanças no ordenamento dos valores genéticos, evidenciando a existência da interação genótipo x nível de relação dos aminoácidos da dieta para características de qualidade de carne. Predições de valores genéticos de características de qualidade de carne com base em determinada relação de (metionina + cistina): lisina da dieta não são válidas para outras relações desses aminoácidos.


This study aimed to evaluate the sensitivity of breeding values for meat quality traits of European quails fed different (methionine + cystine): lysine ratio diets from hatch to 21 days of age, using reaction norm models. A total of 9011 meat quality records from 1400 progenies of 80 sires and 160 dams from two lines (LF1 and LF2) were used in the analyses considering homogeneity of residual variance. The quails from hatch to 21 days of age were fed diets containing 0.61, 0.66, 0.71, 0.76 and 0.81 (methionine + cystine): lysine ratios, 26.12% of crude protein and 2900 ME/kcal of diet. From 22 to 35 days of age all quail were fed a diet containing 22% of crude protein and 3050 kcal ME/kg of diet. The random regression model analyses was performed using the WOMBAT program considering homogeneity of residual variance. Genetic variance and heritability estimates were affected by the environment gradient of diet and line, increasing these estimates with the increase of the (methionine + cystine): lysine ratio of the diet. The breeding values changed with the increase of the environment gradient of the diet with changes in the rank of genetic breeding values characterizing the existence of genotype by environment interaction for meat quality traits. Predictions of meat quality trait breeding values based on a given (methionine + cystine): lysine ratio are not valid for other levels of the amino acid ratio.

Objetivou-se com o presente trabalho ajustar modelos não lineares para descrever o padrão de crescimento de genótipos de codornas de corte e de postura do nascimento aos 35 dias de idade. Foram utilizados dados de 1280 codornas machos e fêmeas, provenientes de sete genótipos de codornas de corte (EV1, EV2, UFV1, UFV2, UFV3, LF1, LF2) e um de postura. Todas as codornas foram pesadas a cada sete dias, do nascimento aos 35 dias de idade. Foram ajustados os modelos Brody, Gompertz, Logístico, Richards e von Bertalanffy aos dados de peso corporal de todos os genótipos. As análises estatísticas foram realizadas utilizando-se o PROC NLIN do SAS (Statistical Analysis System, versão 9.0). Os critérios empregados para a escolha do melhor modelo para descrever a curva de crescimento foram o coeficiente de determinação ajustado (R2), o desvio padrão assintótico (DPA), o desvio médio absoluto dos resíduos (DMA), o índice assintótico (IA), o critério de informação bayesiano (BIC), o critério de Akaike (AIC) e o quadrado médio do erro (QME). Todos os modelos não lineares testados convergiram, com exceção do modelo Brody, que não convergiu para nenhum genótipo avaliado. O modelo Richards não convergiu para o genótipo postura. Os parâmetros da curva de crescimento estimados pelos modelos indicaram maior precocidade, em geral, do genótipo postura comparado aos demais genótipos. O modelo Richards apresentou superestimação do ponto de inflexão para todos os genótipos, exceto para o genótipo postura. Os modelos Gompertz, Logístico e von Bertalanffy são recomendados para descrever o crescimento de codornas de corte dos grupos genéticos em estudo. O modelo Brody não convergiu, por isso não é recomendado para descrever o crescimento dos grupos genéticos de codornas em estudo.


This work aimed to adjust nonlinear models to describe the growth pattern of meat type and posture quail genotypes from hatch to 35 days of age. A total of 1280 male and female quails from EV1, EV2, UFV1, UFV2, UFV3, LF1 and LF2 and Posture genotypes were weighed every seven days, from hatch to 35 days of age. Brody, Gompertz, Logistic, von Bertalanffy and Richards models were used to fit the growth of the genetic groups. Statistical analyses were performed using PROC NLIN of SAS (Statistical Analysis System, version 9.0). The criteria used to select the model showing the best goodness of fit to the growth of the genotypes were: Adjusted Coefficient of Determination (R2), Asymptotic Standard Deviation (DPA), Absolut Mean Deviation (DMA), Asymptotic Index (AI), Bayesian Information Criterion (BIC), Akaike Information Criterion (AIC) and Errors Mean Square (MSE). All nonlinear models converged for the studied genotypes, except Richard the model for the Posture genotype. In general, the estimated parameters for the growth curve using nonlinear models suggested higher precocity for the Posture genotype in comparison to meat type genotypes. The Richard model overestimates the inflection point for all genotypes, except for the Posture genotype. The Gompertz, Logistic and von Bertalanffy are recommended to describe the growth patterns of the studied quail genetic groups. The Brody model did not converge and is not recommended to describe the genetic growth of the studied groups.

This study aimed to analyze the chemical composition and the IgG concentration of the colostrum, transitional milk, and mature milk of Santa Inês ewes as well as the transfer of passive immunity to lambs. Thirty-two pregnant ewes and 38 lambs were used. Ewes were milked immediately after lambing and at 12, 24, 36 h and 10 d postpartum. Colostrum was provided to the lambs at 40±15 min (mean±SE) after birth and then at 30-min intervals for obtaining the intake closest to 10% of body weight, and transitional milk was provided ad libitum. Blood from the lambs was collected 36 h after birth for measuring the serum concentrations of IgG, total protein, albumin, and gamma-globulin. The production was lower in primiparous than in multiparous ewes with body condition score (BCS)<2.75, but did not differ between primiparous and multiparous with BCS≥2.75 (interaction parity and BCS). The IgG concentration and fat, protein, lactose, and defatted dry extract percentages were not affected by the BCS of the ewe at lambing or by the parity. The total solids percentage in the colostrum was higher in ewes with BCS<2.75 (interaction BCS and time). The production and the protein, total solid, and defatted dry extract percentages showed quadratic behavior, the fat percentage decreased linearly, and the lactose percentage increased linearly with time postpartum. The IgG concentration in the colostrum was not correlated with the ewe's weight or BCS at the time of lambing. Moreover, the parity, the BCS, the ewe's type of gestation, and the lamb's sex did not influence the serum concentrations of IgG, total protein, albumin, and gamma-globulin in lambs. Adequate passive immune transfer (PIT) was observed in lambs for which the IgG intake was higher than 30 g. Failure in PIT was observed in 39.5% of lambs when considering a serum IgG concentration lower than 15 mg/mL and in 21% when considering a serum total protein concentration lower than 45 mg/mL. The mean apparent efficiency of absorption was 38.10%, with values between 0.02% and 98.80%. The serum IgG concentration was correlated with the total protein concentration (according to the enzymatic colorimetric method), the gamma-globulin concentration, and the absorption efficiency. The extreme variation on apparent efficiency of absorption may have an effect on the success of PIT. Lambs should consume at least 30 g of IgG in the first 24 h of life to ensure adequate PIT.

Objetivou-se com este trabalho comparar e correlacionar as características de desempenho e de carcaça em codornas de corte do nascimento aos 35 dias de idade. Foram utilizadas 1176 codornas de um dia, machos e fêmeas, de sete genótipos de codornas de corte (EV1, EV2, UFV1, UFV2, UFV3, LF1 e LF2), distribuídas em delineamento inteiramente ao acaso, com ensaio fatorial 7x2 (genótipo x sexo), em sete repetições com 12 codornas por unidade experimental. As codornas foram pesadas individualmente do nascimento aos 35 dias de idade, em intervalos semanais. Aos 35 dias de idade, dois machos e duas fêmeas de cada genótipo e repetição foram amostrados e abatidos para avaliação do peso de carcaça e dos cortes nobres. As análises estatísticas foram conduzidas utilizando-se o procedimento GLM e CORR com a opção MANOVA do SAS para avaliar as diferenças entre os genótipos e a correlação entre as variáveis. Não houve interação (P≥0,05) entre genótipo e sexo para as características de desempenho (peso e ganho de peso nas diferentes idades). Entretanto, houve efeito significativo da interação genótipo x sexo (P<0,05) para as características de pesos de carcaça, peito e coxa+sobrecoxa. Em geral, as codornas dos grupos genéticos UFV1, UFV2 e UFV3 foram as mais pesadas dos sete aos 35 dias de idade e apresentaram maiores rendimentos de carcaça. Como observado pelas correlações fenotípicas, a tendência é que codornas que apresentam maior peso corporal e ganho de peso irão apresentar maior peso de carcaça e dos seus principais cortes.(AU)


This study was carried out to compare and relate the performance and carcass traits of different quail genotypes from hatch to 35 days of age. A total of 1176 one-day-old quails of both sexes were allotted in a completely randomized experimental design, in a 7x2 factorial scheme (genotypes x sexes), in seven replicates with 12 quails per experimental unit. Quails of each genotype were weighed weekly from hatch to 35 days of age. At 35 days of age two males and two females of each genotype and replicate were sampled and slaughtered to evaluate carcass weight and main cut yield. Statistical analyses were performed by GLM and CORR procedure from the Statistical Analysis System (SAS, 2000, version 9.0), with the MANOVA option of the SAS device to assess the differences between the genotypes and the correlation between variables. There was no interaction (P≥0.05) between genotype and sex with the performance characteristics of weight and weight gain. However, there was a significant effect of genotype x sex (P<0.05) for the characteristics carcass,breast, thigh and drumstick weights. In general, quails of genetic groups UFV1, UFV2 and UFV3 were heavier from 7 to 35 days of age. We observed that for the correlation the tendency is that quail that showed higher body weight and weight gain will have greater housing and main cuts weight.(AU)

Objetivou-se com este trabalho avaliar o desempenho de quatro linhagens (L1, L2, L3 e L4) de codornas de corte em cruzamentos dialélicos. O presente estudo foi realizado nas instalações do Programa de Melhoramento de Codornas do Departamento de Zootecnia da UFVJM, no período de 16 de dezembro de 2011 a 26 de janeiro de 2012. Foram avaliados o peso corporal médio ao nascimento, aos 35 e aos 42 dias de idade; o consumo médio de dieta e a conversão alimentar do nascimento aos 35 dias e do nascimento aos 42 dias de idade. As análises dialélicas foram desenvolvidas de forma univariada, considerando-se a metodologia de dialelos completos, a partir das médias das combinações genotípicas resultantes de um experimento inteiramente ao acaso com três repetições. A capacidade geral de combinação foi significativa para todas as características avaliadas, com exceção do consumo médio de dieta e da conversão alimentar do nascimento aos 35 dias de idade. Houve efeito significativo da capacidade específica de combinação sobre o peso corporal ao nascimento (L2xL3; L2xL4; L1xL4); conversão alimentar do nascimento aos 35 dias (L1xL3; L2xL4) e do nascimento aos 42 dias (L1xL3; L2xL3; L2xL4); consumo médio de dieta do nascimento aos 35 dias de idade (L1xL3; L2xL3), o que permitiu a identificação de combinações híbridas mais favoráveis. O efeito recíproco foi significativo apenas sobre o peso ao nascimento. Com base na capacidade geral de combinação, as linhagens mais favoráveis para peso corporal foram a L1 e a L2. Para a conversão alimentar, pela capacidade específica de combinação, os cruzamentos L1xL3 e L2xL4 apresentaram o melhor resultado, enquanto para o consumo de dieta do nascimento aos 35 dias de idade, os cruzamentos L1xL3 e L2xL3 são os mais indicados.


The aim of this study was to evaluate the performance of four strains of meat type quails, designated L1, L2, L3 and L4 in diallel crosses. This study was carried out at the premises of the Genetic Improvement Program for Quails of UFVJM, from December 2011 to May 2012. The average body weight at hatch, at 35 and at 42 days old, the average daily feed intake at 35 and at 42 days old, and feed conversion at 35 and 42 days old were evaluated. Diallel analyzes were developed using univariate analyses, considering the methodology of complete diallel, from the average of the genotype combinations resulting in a complete randomized design with three replications. The combining ability analysis revealed significance for general combining ability for all traits, except for average feed intake and feed conversion at 35 days old. There was significant effect of specific combining ability for hatch weight (L2xL3; L2xL4; L1xL4); feed conversion from hatch to 35 days of age (L1xL3; L2xL4), and from hatch to 42 days (L1xL3; L2xL3; L2xL4); and average feed intake from hatch to 35 days of age (L1xL3; L2xL3), thus allowing the identification of more favorable hybrid combinations. The reciprocal effect showed significance only for hatch weight. Based on general combining ability, the strains which are more favorable for increased body weight were L1 and L2. For feed conversion, for the specific combining ability, the crosses L1xL3 and L2xL4 showed the best results, while for feed intake from hatch to 35 days old, the crosses L1xL3 and L2xL3 would be the most suitable.

This study aimed to assess egg laying and egg quality in quail lines in diallel crosses. We evaluated 16 genetic groups obtained from crosses of four strains of meat quails (L1, L2, L3, and L4) in a complete diallel. We assessed weekly egg number (WEN), egg yield (EY), egg weight (EW), and Haugh unit (HU) for: 51-80, 81-110, 111-140, and 51-140 days (whole period) old. The general combining capacity (GCC) showed significance for WEN and EY, with the exception of the period of 111-140 days. In general, L3 had the highest positive GCC estimates. The additive genetic variability was significant for EW in all periods, as was HU, with the exception of the first period. The specific combining capacity was significant for HU, except in the second period. The largest estimates for GCC were submitted by L1 for EW, followed by L4, considering the whole period. For HU, L2 had the highest estimates for GCC. The more favorable cross for HU was L2xL3, in the whole period. In the third period, there was a significant reciprocal effect, where there was a positive estimate for the cross L2xL3.

An electronic equipment based on the infrared spectroscopy was evaluated to measure the whey composition in "Minas padrão" and "prato" cheeses. The results for fat, protein, and total solids using infrared instrument based on filters and using standard methods were compared. There were differences between the values obtained from both analytical methods, and since compounds formed during heating of the analyzed whey may have interfered with infrared spectrum readings, a linear transformation of the data was done. After this statistical adjustment, results for "Minas padrão" cheese were equivalent in both infrared and standard methods. For "prato" cheese whey, the difference was significant, even after the adjustment.

A beta-N-Acetylhexosaminidase (EC 3.2.1.52) was purified from hepatic extracts of Sotalia fluviatilis, order Cetacea. The protein was purified by using ammonium sulfate fractionation and four subsequent chromatographies (Biogel A 1.5 m, Chitin, Deae-Biogel and hydroxyapatite resins). After these purification steps, the enzyme was purified 380.5-fold with an 8.4% yield. The molecular mass (10 kDa) was estimated by SDS-PAGE and MALDI-TOF analysis. A Km of 2.72 mM and Vmax 9.5 x 10(-6) micromol/(min x mg) were found for this enzyme, determined by p-nitrophenyl-beta-D: -hexosaminide substrate digestion. Optimal pH and temperature for beta-N-Acetylhexosaminidase activity were 5.0 and 60 degrees C, respectively. Enzyme activity was inhibited by sodium selenate (Na(2)SeO(4)), mercuric chloride (HgCl(2)) and sodium dodecyl sulfate (C(12)H(25)SO(4)Na), and activated by zinc, calcium, barium and lithium ions. Characterization of the beta-N-Acetylhexosaminidase in Sotalia fluviatilis can be a basis for physiological studies in this species.

Brown algae have two kinds of acid polysaccharides present in the extracellular matrix: sulfated fucan and alginic acid. We have previously isolated and characterized fucans from several species of brown seaweed. The characterized fucans from Dictyotaceae are heterofucans containing mainly fucose, galactose, glucose, xylose, and/or uronic acid. The fucan from Fucus vesiculosus is a homofucan containing only sulfated fucose. We assessed the activity of these fucans as inhibitors of HIV from reverse transcriptase (RT). Using activated DNA and template primers poly(rA)-oligo(dT), we found that fucans at a concentration of 0.5-1.0 microg/mL had a pronounced inhibitory effect in vitro on the avian reverse transcriptase, with the exception of xylogalactofucan isolated from Spatoglossum schröederi, which had no inhibitory activity. The alginic acid (1.0 microg/mL) inhibited the reverse transcriptase activity by 51.1% using activated DNA. The inhibitory effect of fucans was eliminated by their desulfation. Furthermore, only xylofucoglucuronan from S. schröederi lost its activity after carboxyreduction. We suggest that fucan activity is not only dependent on the ionic changes but also on the sugar rings that act to spatially orientate the charges in a configuration that recognizes the enzyme, thus determining the specificity of the binding.

The influence of calcium fortification by the addition of calcium chloride on quality parameters of ice cream based on physical properties was investigated, as was the effect of kappa-carrageenan at modifying the effects of this calcium fortification. Four ice cream mixes of conventional composition, with added kappa-carrageenan (0 or 0.025%) and added calcium chloride (0 or 4.4 g L(-1) = 40 mM of added Ca(2+)), were prepared. Modulated temperature-differential scanning calorimetry was used to investigate the effect of calcium chloride on the nucleation temperature, enthalpy of melting, and freezing point depression. The protein composition of 15.4% (wt/wt) reconstituted skim milk powder solutions with or without 4.4 g L(-1) added CaCl(2) and in the supernatant after ultracentrifugation was determined. Fat particle size distributions in ice cream were characterized by light scattering. Ice crystal sizes before and after temperature cycling were determined by cold-stage light microscopy. The results demonstrated that the addition of calcium chloride led to a substantial increase in ice crystal sizes and in fat partial coalescence, which were exacerbated by the addition of kappa-carrageenan. These results can be explained by the interaction between Ca(2+) ions and casein micelles, rather than any effects on freezing point depression. The calcium ions led to a more compact micelle, less serum beta-casein, and high fat destabilization, all of which would be expected to reduce macromolecular structure and volume occupancy in the unfrozen phase, which led to increased rates of ice recrystallization.

OBJETIVO: Avaliar a aptidão cardiorrespiratória e verificar a presença de broncoespasmo induzido pelo exercício (BIE) em crianças com displasia broncopulmonar (DBP). MÉTODO: Foram realizadas prova de função pulmonar e análise de gases em um teste cardiopulmonar, em 46 crianças com idade entre 7 a 10 anos, formando três grupos: crianças nascidas pré-termo com DBP, (DBP, n= 13); crianças nascidas pré-termo sem DBP, (RNPT, n= 13); e crianças saudáveis nascidas a termo, (Controle, n= 20). RESULTADOS: A duração dos testes foi 7,70 ± 1,49; 9,1 ± 2,02 e 8,4 ± 2,12 min; o VO2máximo foi 35,98 ± 5,33; 38,99 ± 6,73 e 34,91 ± 6,09 ml/kg/min; e a VE foi 28,54 ± 7,39; 28,84 ± 5,98 e 28,96 ± 6,96 l/min para os grupos DBP, RNPT e Controle, respectivamente. Não foram encontradas diferenças significantes entre os grupos (p> 0,05). A FCmáxima foi 188 ± 9,37; 196 ± 5,15 e 197 ± 10,90 bpm; a taxa de troca gasosa máxima (R) foi 1,21 ± 0,22; 1,10 ± 0,06 e 1,05 ± 0,05 para os grupos DBP, RNPT e Controle, respectivamente, sendo esses valores diferentes entre o grupo Controle e DBP (p< 0,05). Os valores do VEF1 pré e VEF1 pós-exercício foram de 99 ± 12 por cento e 94 ± 14 por cento; 100 ± 14 por cento e 100 ± 15 por cento; e 102 ± 15 por cento e 101 ± 15 por cento, para os grupos DBP, RNPT e Controle, respectivamente. Na comparação do VEF1 pré e pós-exercício não houve diferenças significantes e nem caracterização de BIE nos grupos. CONCLUSÃO: A diferença encontrada no R pode ser relacionada a alterações ventilatórias e à difusão pulmonar. A aptidão cardiorrespiratória das crianças com DBP é semelhante à dos grupos RNPT e Controle.


OBJECTIVE: To assess cardiorespiratory capacity and investigate the presence of exercise-induced bronchospasm among children with bronchopulmonary dysplasia. METHOD: Pulmonary function tests and gas analyses were performed in a cardiopulmonary test on 46 children aged 7-10 years. Three groups were formed: children born prematurely with bronchopulmonary dysplasia (BPD; n= 13), children born prematurely without bronchopulmonary dysplasia (Preterm; n= 13) and healthy children born at full term (Control; n= 20). RESULTS: The test duration was 7.70 ± 1.49; 9.1 ± 2.02 and 8.4 ± 2.12 min; VO2max was 35.98 ± 5.33; 38.99 ± 6.73 and 34.91 ± 6.09 ml/kg/min; and VE was 28.54 ± 7.39; 28.84 ± 5.98 and 28.96 ± 6.96 l/min for the BPD, Preterm and Control groups respectively. There were no significant differences between the groups (p> 0.05). The maximum heart rate was 188 ± 9.37; 196 ± 5.15 and 197 ± 10.90 beats/min and the respiratory exchange ratio (RER) was 1.21 ± 0.22; 1.10 ± 0.06 and 1.05 ± 0.05, for the BPD, Preterm and Control groups respectively, and there was a significant difference between the BPD and Control groups (p< 0.05). The FEV1 values before and after exercise were 99 ± 12 percent and 94 ± 14 percent; 100 ± 14 percent and 100 ± 15 percent; and 102 ± 15 percent and 101 ± 15 percent, for the BPD, Preterm and Control groups respectively. Comparison of FEV1 before and after exercise did not show any significant differences and exercise-induced bronchospasm was not characterized, in any of the groups. CONCLUSION: The difference in RER may be related to abnormal ventilation and pulmonary diffusion. The cardiorespiratory capacity of children with BPD was similar to that of the Preterm and Control groups.

Com o objetivo de identificar as principais fontes de contaminaçäo por Staphylococcus aureus e Staphylococcus produtores de coagulase (SC+), avaliou-se o processo de fabricaçäo de queijo prato em um laticínio de Lavras, MG, durante os meses de outubro de 2000 a abril de 2001. As análises microbiológicas foram feitas no leite cru, no leite pasteurizado resfriado, nas mäos e antebraços dos funcionários, na salmoura, na água de imersäo das fôrmas e no queijo embalado. Contagens elevadas de SC+ e de S. aureus (4x10³ a 4,8x10(6) UFC/ml e 4x10³ a 3,3x10(5) UFC/ml, respectivamente) foram encontradas em quatro avaliaçöes no leite cru. Após a pasteurizaçäo, as contagens foram reduzidas a <1 UFC/ml. Em três das cinco avaliaçöes, o queijo prato apresentou contagens de SC+ (10(4), 10(5) e 2,3x10(5) UFC/g) superiores às permitidas pela legislaçäo vigente. A água de imersäo das fôrmas e a salmoura apresentaram contagens de SC+ e S. aureus inferiores a 1 UFC/ml näo se constituindo em importantes fontes de contaminaçäo. As mäos e os antebraços dos funcionários foram possivelmente as fontes de contaminaçäo do queijo, isto é, alta contagem nos queijos estava associada à contagem elevada nas mäos (4x10² UFC/cm²) ou nos antebraços (4,7x10² e 3,3x10³ UFC/cm²) dos manipuladores.

HPLC on chiral stationary phases has been used for the enantioselective assay of propafenone (PPF), 5-hydroxypropafenone (PPF-50H) and N-despropylpropafenone (PPF-NOR) enantiomers. The results obtained on Chiralpak AD column showed that it is useful for the resolution of PPF and of its main metabolites, although the peaks obtained for PPF-NOR were not symmetrical under the conditions investigated. This column and circular dichroism-based detection system were used to determine the absolute configuration of the eluates. Furthermore, the influence of the mobile phase composition on the resolution of PPF and of its main metabolites was investigated on cellulose derivatives (Chiralcel OD-H and Chiralcel OD-R) and protein (Chiral AGP and Ultron ES-OVM)-based chiral stationary phases. The enantiomers of PPF were resolved on all the columns, except for the Ultron ES-OVM. This column, the Chiralpak AD and the Chiralcel OD-H columns were suitable for the resolution of the PPF-50H enantiomers. The PPF-NOR enantiomers were resolved on the Chiralpak AD, Chiral AGP and Chiralcel OD-R columns.

The distribution and structure of heparan sulfate and heparin are briefly reviewed. Heparan sulfate is a ubiquitous compound of animal cells whose structure has been maintained throughout evolution, showing an enormous variability regarding the relative amounts of its disaccharide units. Heparin, on the other hand, is present only in a few tissues and species of the animal kingdom and in the form of granules inside organelles in the cytoplasm of special cells. Thus, the distribution as well as the main structural features of the molecule, including its main disaccharide unit, have been maintained through evolution. These and other studies led to the proposal that heparan sulfate may be involved in the cell-cell recognition phenomena and control of cell growth, whereas heparin may be involved in defense mechanisms against bacteria and other foreign materials. All indications obtained thus far suggest that these molecules perform the same functions in vertebrates and invertebrates.
