BACKGROUND AND AIMS: Apolipoprotein B plays a crucial role in regulating plasma cholesterol by mediating the interaction of low-density lipoprotein (LDL) with LDL receptors in the liver. Inherited mutations in this gene may increase the risk of developing premature atherosclerotic cardiovascular disease, especially in individuals with familial hypercholesterolemia type 2 (FH2). The aim of this study is to identify APOB variants that may indicate pathogenicity in a sample of the Brazilian population using a data bank exome sequencing study by NGS in a Brazilian population phenotypically diagnosed by clinical and laboratory profile. This finding is going to improve genetic hypercholesteremia diagnosis. METHODS: High-quality DNA samples (n»300) were sequenced using an exon-targeted gene sequencing (ETGS) strategy to identify variants in FHrelated genes. Pathogenicity classification was based on criteria established by the American College of Medical Genetics and Genomics (ACMG), also using information from ClinVar and pathogenicity scores from previous association studies. RESULTS: A total of 121 variants were identified in APOB, of which four are novel variants missense (p.Thr626Asn, p.Ile2750Thr, p.Gln2078Lys and p.Met4184Arg). After curating pathogenicity scores, variants were classified according to the ACMG criteria. Among them four as pathogenic or likely pathogenic (p.Pro2739Leu, p.His1923Arg, p.Pro994Leu and p.Pro877Leu), and 21 variants had uncertain significance. Additionally, 92 previously known variants with uncertain significance were classified as benign or likely benign. The results were submitted to Clinvar for actualization of pathogenicity. CONCLUSIONS: These results improve the molecular diagnosis associating APOB variants with the clinical phenotype of hypercholesterolemia.

BACKGROUND AND AIMS: Introduction: The familial hypercholesterolemia (FH) is one of the main causes of cardiovascular diseases, and it is mainly caused by genetic variants at the low-density lipoprotein receptor (LDLR). Although ultrasequencing technology has allowed the identification of several genetic variants, few of them was functional analyzed. The CRISPR/ Cas9 tool promotes precise genetic editing and allows the creation of experimental models, therefore contributing to the functional validation process. Aim: To use the CRISPR/Cas9 tool to perform in vitro functional analysis of LDLR variants identified in FH patients. METHODS: Two missense LDLR variants were selected within a group of variants identified in FH patients, based on in silico data, the affected protein domain and MAF. Three sgRNAs were designed for each of the variants c.551G>A and c.1118G>A, to analyze the accuracy of the sgRNAs. The sgRNAs were inserted on PX458 plasmid, cloned, purified in E. coli DH5a, and then co-transfected with the DNA template at HepG2 cells. The DNAs templates were designed to contain the selected variants. RESULTS: HepG2 cells co-transfected with PX458 constructs and DNA templates showed considerably transfection rate, being possible to visualize it at fluorescence microscopy. However, it was noted that single transfection of sgRNAs showed a higher transfection efficiency than cotransfection. CONCLUSIONS: We designed sgRNA for c.551G>A and c.1118G>A variants, being able to analyze the transfection efficiency. In further steps, we will select new sgRNAs for LDLR variants that have not been described yet, and functional analysis will be performed to determine the clinical relevance of these variants.

A mastite bovina, uma das principais doenças do rebanho leiteiro, caracteriza-se por um processo inflamatório no úbere. A inviabilidade econômica, o impacto ambiental negativo e os resíduos antimicrobianos têm estimulado a pesquisa de outros tratamentos alternativos para a prevenção e o tratamento de doenças na bovinocultura leiteira. O betaglucano é um agente imunomodulador com potencial ação preventiva para doenças infecciosas, inclusive a mastite. Este estudo teve como objetivo avaliar a eficácia do uso do betaglucano, por meio de administração oral, em animais em lactação. Foram utilizadas 20 vacas lactantes, distribuídas em dois grupos, um controle e um tratamento, com 10 animais em cada grupo. O grupo tratamento recebeu 5g/dia, durante 60 dias, de 1,3-1,6 betaglucano isolado da parede celular de Saccharomyces cerevisiae diluído em ração após a ordenha, enquanto o grupo controle recebia somente a ração. Foram realizados os testes de California Mastitis Test (CMT), contagem de células somáticas (CCS), produção de leite e percentual de gordura e proteína no leite. Não houve diferença estatisticamente significativa entre os grupos quanto à CCS, ao CMT, à composição do leite ou produção. Não se observou, portanto, eficácia do uso do betaglucano purificado, administrado por via oral, no controle e na prevenção da mastite em vacas leiteiras, quando comparadas com o grupo controle. Atribuem-se esses resultados, principalmente, à degradação ruminal do produto testado. Sugerem-se, portanto, mais pesquisas utilizando o 1,3-1,6 betaglucano purificado de parede de S. cerevisiae por outras vias de administração, tais como intramamária e subcutânea.(AU)


Bovine mastitis, one of the main diseases of dairy herds, is characterized by an inflammatory process in the udder. The economic and environmental impacts, as well as the residues of antimicrobial drugs have stimulated the research of novel alternative treatments for the prevention and treatment of diseases in dairy production cows. The beta-glucan is an immunomodulator agent, with potential preventive action for infectious diseases, including mastitis. This study aimed to assess the effectiveness of orally administered beta-glucan in lactating cows. 20 lactating cows were used, distributed into two groups, one control and one treatment, with 10 cows in each group. The treatment group received 5g of 1.3-1.6 betaglucan daily for 60 days, isolated from the cell-wall of Saccharomyces cerevisiae diluted into a grain meal, whereas the animals in the control group received only the ration. The California Mastitis Test (CMT), Somatic Cells Counting (SCC), daily production and assessments of fat and protein content in milk were done. There was no statistically significant difference between the groups concerning subclinical mastitis detected by CMT, SCC, milk production and composition regarding protein and fat content. It was not observed, therefore, the effectiveness of the use of purified beta-glucan orally administered on the control or prevention of mastitis in dairy cows. The results are attributed to the ruminal degradation of the product tested. It is, therefore, suggested that more research should be conducted using the 1.3-1.6 beta-glucan purified from the cell wall of S. cerevisiae by other administration means and ruminal protection technologies for the isolated beta-glucan.(AU)

INTRODUÇÃO: O diagnóstico molecular da hipercolesterolemia familial (HF) é atribuído principalmente as variantes nos genes LDLR, LDLRAP1, APOB e PCSK9. O objetivo deste estudo foi realizar análise in silico para investigar o impacto de variantes sem descrição na literatura no gene APOB observado em pacientes com HF. MÉTODOS: Foram selecionados 141 indivíduos com diagnóstico clínico de HF. As variantes no gene da APOB foram selecionadas após sequenciamento dos éxons de 61 genes utilizando a plataforma MiSeq (Illumina). Os dados foram analisados nos programas Real Time Analysis, MiSeq Reporter, BaseSpace Sequence Hub e VariantStudio. Para a análise in silico, as sequências molde das moléculas da apoB-100 e o LDLr foram selecionadas por modelagem comparativa considerando o maior grau de identidade. As sequências proteicas foram alinhadas e os modelos 3D foram construídos utilizando os programas SEAVIEW e MODELLER v9.21. O gráfico de Ramachandran do modelo de menor energia apresenta 0,5% de outliers e análise de regiões de desordem, como principal validação. Os resultados das conformações de ancoragem foram analisados no software PyMol 2.1. Os estudos de docking molecular foram realizados para identificar o melhor complexo de conformação usando o servidor web clusPRO. RESULTADOS: Após a análise molecular dos 141 pacientes foram identificadas 7 variantes missenses sem descrição na literatura no gene APOB (c.433C>T, c.2630C>T, c.2950G>A, c.5743G>A, c.7367C>A, c.9880T>C e c.10780T>C). Os estudos de docking das variantes demonstraram uma maior afinidade entre o LDLr e a apoB-100 (c.2630C> T; Pro877Leu) em comparação com a proteína não mutada. A troca dos resíduos permaneceu como propriedade físico-química, e comparando as distâncias de ligação das proteínas não-mutadas (5Å) e mutadas (3,5Å), sugere-se uma maior afinidade do complexo (LDLr-apoB-100) para a leucina, tal fato é afirmado pela análise da região de desordens da apoB-100, onde a posição 877 está em uma região desorganizada e flexível. Esta maior afinidade poderia levar a uma menor dissociação intracelular deste complexo, resultando em uma alta taxa de degradação do LDLr pelas enzimas lisossômicas, levando ao aumento da concentração plasmática de LDLc. Para as outras variantes não houve alterações significativas. CONCLUSÃO: Os resultados sugerem que estudos in silico baseados na ferramenta de docking molecular podem melhorar o conhecimento da contribuição genética no desenvolvimento da doença HF. Além disso, a variante APOB c.2630C> T deve ser avaliada in vitropara validação do mecanismo proposto. (AU)

A varfarina é um anticoagulante utilizado na prevenção e no tratamento de doenças tromboembólicas, com janela terapêutica estreita e, elevado risco de hemorragias. Nos idosos, a principal indicação para tratamento anticoagulante oral é fibrilação atrial, cuja prevalência aumenta com a idade, atingindo 8% após os 80 anos. O exame utilizado para controle da anticoagulação oral é o tempo de protrombina, através do cálculo da razão normalizada internacional (RNI), visando o ajuste de dose e manutenção da faixa terapêutica (RNI= 2 a 3). Os idosos requerem um monitoramento efetivo devido a fatores inerentes da idade. Embora seja conhecido que os fatores genéticos influenciam na resposta terapêutica à varfarina, na rotina da maioria dos hospitais a farmacogenética ainda não é considerada no ajuste de dose. Visando estabelecer uma conduta terapêutica personalizada, o presente estudo tem como objetivo avaliar a associação entre o polimorfismo rs9934438 do gene VKORC1, que codifica a enzima vitamina K epóxido redutase, e a dose semanal de varfarina necessária para atingir o RNI adequado. Até o momento, foram incluídos 52 pacientes com idade superior a 70 anos, de ambos os sexos e em uso de varfarina. A análise do polimorfismofoi realizada através da PCR em tempo real utilizando os reagentes TaqMan™ Sample-to-SNP™ e o sistema de detecção TaqMan® SNP Genotyping Assay. As análises estatísticas foram realizadas utilizando o pacote SPSS v. 16.0 e nível de significância adotado foi de 5%. Dos 52 pacientes incluídos até o momento, 37 (71%) permaneceram na faixa terapêutica (Time in Therapeutic Range, TTR) em pelo menos 50% do tempo de anticoagulação e, para eles a dose semanal de varfarina variou de 15mg a 62,5mg. Apesar de ser um estudo piloto, a distribuição dos genótipos está em equilíbrio gênico, segundo Hardy-Weinberg (AA=19,2%, AG=34,6%, GG= 46,2%, χ2= 3,34 e p=0,067). Para os idosos com TTR ≥50%, a frequência do alelo A foi significantemente maior entre os pacientes que utilizaram doses menores de varfarina (Exato de Fisher, p=0,005). Adicionalmente, os portadores do genótipo AA necessitaram, em média, de aproximadamente metade da dose para atingir a faixa terapêutica quando comparados aos portadores do genótipo GG, 20,5 versus 36,5 mg/semana, respectivamente (ANOVA, p=0,006/ Pós-teste Bonferroni, p=0,039). Os resultados permitem concluir que portadores de alelo A são mais responsivos ao tratamento com varfarina, sugerindo que o perfil genotípico pode ser de grande valor para o direcionamento da dose terapêutica em idosos. Objetivou-se estudar o efeito do ômega 3 e da vitamina B12 no espermograma, na histomorfometria dos órgãos reprodutivos e na temperaturas do corpo com termografia infravermelha em ratos Wistar. Utilizaram-se 16 ratos, em quatro grupos (n=4), que receberam injeções diárias por 30 dias, sendo: grupo controle - solução salina; grupo ômega 3 - óleo de peixe 1g/kg; grupo B12 - vitamina B12 3µg; e grupo ômega 3 + B12 - óleo de peixe 1g/kg e vitamina B12 3µg. Imagens termográficas de áreas do corpo foram obtidas. No 30º dia, os ratos foram sacrificados e realizaram-se as análises de morfologia espermática e histomorfometria. Os dados foram submetidos à análise de variância e ao teste de Tukey a 5%. A temperatura da superfície do escroto foi superior no grupo B12 (P<0,05). Não houve diferenças entre grupos (P>0,05) para temperaturas do globo ocular. Houve correlação entre temperatura da superfície do escroto e porcentagem de gota citoplasmática distal (P=0,678). A elevação da temperatura do escroto resulta no aumento da porcentagem de gotas citoplasmáticas distais. A temperatura do globo ocular não sofre influência significativa do ômega 3 e da vitamina B12. O ômega 3 reduz o epitélio seminífero, e a vitamina B12 minimiza esse efeito.(AU)


The objective of this study was to study the effect of Omega 3 and vitamin B12 on spermogram, histomorphometry of reproductive organs and body temperature with infrared thermography in Wistar rats. Sixteen rats were used in four groups (n= 4) who received daily injections for 30 days. Control Group - saline solution; Group Omega 3 - fish oil 1g/kg; Group B12 - vitamin B12 3μg and Group Omega 3 + B12 - fish oil 1g/kg and vitamin B12 3μg. Thermographic images of body were obtained. On the 30th day the rats were sacrificed and analyzes of sperm morphology and histomorphometry were performed. Data were submitted to analysis of variance and Tukey's test at 5%. The surface temperature of the scrotum was higher in group B12 (P< 0.05). There were no differences between groups (P> 0.05) for eyeball temperatures. There was a correlation between scrotal temperature and distal cytoplasmic droplet (P= 0.678). Elevation of scrotum temperature results in an increase in the percentage of distal cytoplasmic droplets. The temperature of the eyeball is not significantly influenced by Omega 3 and vitamin B12. Omega 3 reduces the seminiferous epithelium and vitamin B12 minimizes this effect.(AU)

O conhecimento do desenvolvimento mamário de um rebanho leiteiro é fundamental, pois relaciona-se à eficiência produtiva. Objetivou-se avaliar a associação dos parâmetros térmicos mamários com concentrações hormonais de búfalas em distintos estágios fisiológicos. Foram utilizadas 24 búfalas mestiças Murrah, em quatro grupos (n= 6): grupo 1 (bezerras), grupo 2 (novilhas), grupo 3 (gestantes) e grupo 4 (lactantes). A cada 28 dias, durante 4 meses, realizaram-se exames de termografia digital por infravermelho para verificar temperatura superficial dos corpos mamários craniais (CMCr) e caudais (CMC), das cisternas craniais (CGMCr) e caudais (CGMC) e tetas craniais (TGMCr) e caudais (TGMC). Foi aferida temperatura retal (TR) e colhido sangue para mensuração das concentrações plasmáticas do fator semelhante a insulina tipo-I, insulina (INS), hormônio do crescimento (GH), progesterona (P4) e estradiol. Grupos 1 e 2 apresentaram correlação de TR com CGMCr. No grupo 3, TR correlacionou-se com TGMCr, TGMC e concentrações plasmáticas de P4. No grupo 4, houve correlação de TR com CGMC, TGMCr e concentrações plasmáticas de INS e GH, e de TGMC com concentrações plasmáticas de GH. Nos quatro grupos, CGMCr correlacionou-se com CMCr e TGMCr, semelhantemente ao observado na porção caudal. Variações térmicas mamárias refletiram alterações fisiológicas aguardadas nos períodos avaliados.(AU)


The knowledge of the mammary development of a dairy herd is key, since it is related to its productive efficiency. The objective of this study was to evaluate the association of mammary thermal parameters with hormonal concentrations of buffaloes at different physiological stages. Twenty-four Murrah crossbred buffaloes were used in four groups (n= 6): group 1 (calves), group 2 (heifers), group 3 (pregnant animals) and group 4 (lactating animals). Every 28 days, for 4 months, infrared digital thermography was performed to check the surface temperature of cranial (CrCM) and caudal (CCM) corpus mammae, cranial (CrC) and caudal (CC) cisterns and cranial (CrT) and caudal teats (CT). Rectal temperature (RT) was measured and blood was collected to measure plasma concentrations of insulin-like factor I, insulin (INS), growth hormone (GH), progesterone (P4) and estradiol. Groups 1 and 2 presented correlation of RT with CrC. In group 3, RT correlated with CrT, CT and plasma P4 concentrations. In group 4, there was correlation of RT with CC, CrT and plasma concentrations of INS and GH, and CT with plasma GH concentrations. In all four groups, CrC correlated with CrCM and CrT, similar to that observed in the caudal portion. The thermal changes in the mammary glands reflected the expected physiological changes in the evaluated periods.(AU)

Anthelminthic resistant populations of Haemonchus contortus are a major problem in sheep rearing, but plant extracts may offer viable alternative treatments. In our preliminary studies, Piptadenia viridiflora was frequently selected by sheep grazing in the Cerrado. The present research evaluated its in vitro and in vivo anthelmintic activity. The HPLC chromatograms of P. viridiflora aqueous extract (AE) and ethanolic extract (EE) showed the presence of flavonoids. The total condensed tannin (proanthocyanidin) was 0.2 and 1.01% in AE and EE, respectively. In an egg hatching inhibition (EHI) test, the LC90 of AE was 2.4mg/mL, and, of EE, was 2.1mg/mL. After tannin extraction, higher EHI and lower LC90 were observed. In a larval development inhibition test, the LC90 of AE was 13.66mg/g of fecal culture. The highest dose of AE administered to mice (203.0mg/kg bw) was well tolerated, suggesting low toxicity. In vivo, AE was orally administered to lambs at 283mg/kg bw, and, at weeks one, two, and three post-treatment, the mean fecal egg count (FEC) was significantly lower than in untreated lambs (P<0.05). Blood parameters were normal and similar in untreated and treated sheep. For all lamb groups, the mean total serum protein was significantly higher at week two post-treatment than at other evaluated periods (P<0.05). Piptadenia viridiflora extracts had low condensed tannin content and exhibited high anthelminthic efficacy in vitro and significantly reduced FEC. Tannins were not shown to be the principal components affecting EHI, hence it is necessary to isolate and characterize the principal active P. viridiflora compounds, and to assess their possible synergism.

Avaliou-se o efeito da adição de plasma seminal ovino ao sêmen descongelado sobre a taxa de prenhez de ovelhas em rebanho comercial. Cento e setenta e quatro ovelhas cruza Texel foram distribuídas em quatro tratamentos: T1) inseminação artificial cervical (IAC) com sêmen descongelado (SD) diluído em solução tampão fosfato salino (PBS); T2) IAC com SD e adição de plasma seminal ovino; T3) grupo-controle I: IAC com sêmen fresco diluído em PBS; T4) grupo-controle II: inseminação artificial por laparoscopia com SD diluído em PBS. Para indução de cio, utilizaram-se esponjas impregnadas com acetato de medroxiprogesterona (MAP) por 12 dias, com aplicação intramuscular de 400 UI de eCG (Novormon®) e de 37,5µg de cloprostenol sódico (Sincrocio®), no dia da retirada das esponjas. O aparecimento de cio foi monitorado com rufiões vasectomizados a partir da retirada das esponjas até a inseminação artificial em tempo fixo - 54 a 60 horas. A taxa de prenhez do tratamento com adição de plasma seminal ao sêmen descongelado (7,0%) não diferiu (P>0,05) do tratamento sem adição de plasma (4,3%), entretanto foi menor (P<0,05) se comparada à taxa de prenhez dos grupos-controle I inseminação via cervical superficial com sêmen fresco diluído (50,0%) e II inseminação via laparoscopia com sêmen descongelado (39,4%). A inseminação artificial por via cervical superficial com adição de plasma seminal ao sêmen descongelado não elevou o percentual de prenhez em valores que justifiquem a indicação desta biotecnologia em rebanhos comerciais de ovinos.


The effect of seminal plasma addition to thawed-frozen ram semen on the pregnancy rate of commercial herd ewes was evaluated. One hundred and seventy-four crossbred Texel sheep were allocated to four treatments: T1) cervical artificial insemination (CAI) using frozen-thawed semen (FTS) diluted in phosphate buffered saline solution (PBS); T2) CAI using FTS diluted in ovine seminal plasma; T3) control group I: CAI using fresh semen diluted in PBS; T4) control group II: laparoscopic insemination using FTS diluted in PBS. Estrus induction was performed with medroxiprogesterone acetate (MAP) impregnated sponges for 12 days, followed by intramuscular injection of 400 IU of eCG (Novormon®) and 37.5µg of sodium cloprostenol (Sincrocio®) on the day of sponge removal. Estrus was monitorated with vasectomized rams, beginning at the time of the sponge removal until the fixed time artificial insemination - 54 to 60 hours. The pregnancy rate of FTS diluted in seminal plasma treatment (7.0%) did not differ (P>0.05) for the treatment without addition of seminal plasma (4.3%), however it was lower (P<0.05) when compared to the pregnancy rate of the cervical inseminated control I group with PBS diluted fresh semen (50.0%) and laparoscopic inseminated control group II with PBS diluted FTS (39.4%). The cervical artificial insemination with the addition of seminal plasma to frozen-thawed semen did not increase the pregnancy rate at acceptable values to make this biotechnology useful on commercial herds.

The majority of the population in developing countries uses plants or plant preparations in their basic health care. Many plant species used nowadays in folk medicine have been proved to have antimicrobial properties. However, several factors, such as incorrect preparation of the plants, can interfere with the effectiveness of the treatment. The purpose of this study was to assess the use of homemade preparations of medicinal plants in the treatment of infectious diseases, by in vitro determination of their antimicrobial potential. Based on recipes elicited by questionnaires that were previously applied to a participant population, the samples were prepared in a similar manner and analyzed by the agar diffusion method. Members of 41 families, whose children attend a center of education that serves several needy communities in the city of Fortaleza (Ceará, Brazil), were interviewed; of these, 97.6% said they had used herbal therapy as a means to treat infectious diseases. In replies to a total of 39 questionnaires, 97 different homemade preparations of medicinal plants were cited. Out of 45 samples subjected to an in vitro assessment of antimicrobial activity, 25 (55.6%) had some inhibitory effect on the growth of at least one of the microorganisms used. Most of the plants with known antimicrobial properties and cited by respondents showed variations in their in vitro activity, according to the manner in which they were prepared.


Grande parte da população de países em desenvolvimento utiliza plantas ou preparações vegetais nos cuidados básicos à saúde. Muitas das espécies vegetais utilizadas na medicina popular apresentam propriedades antimicrobianas comprovadas; no entanto, diversos fatores, como a metodologia incorreta de preparo, podem interferir na eficácia do tratamento. O presente trabalho teve como objetivo avaliar a utilização de preparações caseiras de plantas medicinais para o tratamento de doenças infecciosas através da determinação in vitro do potencial antimicrobiano. A partir de informações contidas em questionários que foram previamente aplicados à população participante, as amostras foram preparadas de forma semelhante e analisadas através do método de difusão em ágar. Foram entrevistadas 41 famílias, das quais 97,6% afirmaram utilizar as plantas medicinais como opção terapêutica no tratamento de doenças. Em um total de 39 questionários, foram citadas 97 diferentes preparações caseiras de plantas. Das 45 amostras (indicadas para tratar algum sinal ou sintoma que podem estar relacionado a um quadro infeccioso) submetidas à avaliação da atividade antimicrobiana in vitro, 55,6% (25 amostras) apresentaram atividade inibitória sobre o crescimento de pelo menos um dos microrganismos utilizados. A maioria das plantas com propriedade antimicrobiana conhecida e que foram citadas pelos entrevistados apresentaram variações na sua atividade in vitro de acordo com o modo como foram preparadas.

The objective of this paper is to describe the main characteristics of small-scale marine fisheries in the municipal district of Florianópolis (SC), the capital city of the state of Santa Catarina. We give information about the most captured species, the fishing fleet and fishing gear used. The dynamics of the activity is described, considering the kind of operation of the boats, alongside post-capture procedures. The analyses were accomplished in two types of fishing communities: the one composed of fishers that fish in bays (North and South) and those who fish in the open sea. Numeric differences were observed among these two fishing areas, indicating the existence of two different fleets operating in the area. Differences between the two fishing areas were detected, considering the species caught, the harvest time and the fishing gear used. The open sea fishers have boats with dimensions, engine power, number of crew and fuel consumption numerically higher than those working in bays, indicating the existence of two distinct fleets operating in the region.

There is growing evidence indicating that neurogenesis in adulthood is influenced by certain types of the central diseases such as neuroinflammation, however, its mechanism is not fully understood. This study was, therefore, designed to examine the effects of lipopolysaccharide (LPS), a bacterial endotoxin known to cause the neuroinflammation, on the neurogenesis in the dentate gyrus of adult mice using the bromodeoxyuridine (BrdU) -pulse chase method. LPS failed to affect the number of BrdU-labeled cells in the dentate gyrus 2 h after BrdU injection, indicating no effects of LPS on the proliferation of the neural stem cells (NSCs). On the other hand, we found that LPS dose-dependently (0.1, 0.5, 1 mg/kg) decreased the number of BrdU-labeled cells 7 and 21 days after BrdU injection. We also observed that LPS increased cell death in the dentate gyrus using terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling (TUNEL) staining, suggesting that LPS impaired the survival of newborn cells derived from the NSCs. The double-immunostaining for BrdU and specific cell type markers revealed that LPS did not alter the commitment of the NSCs to the neurons and astrocytes. The systemic injection of indomethacin, a non-selective cyclooxygenase (COX) inhibitor, and NS398, a selective COX-2 inhibitor, but not SC560, a selective COX-1 inhibitor, did not only ameliorate LPS-induced suppression of the newborn cell survival, they fully protected against the LPS effect. Furthermore, the central injection of NS398 also ameliorated LPS-induced suppression of the newborn cell survival in the dentate gyrus. The treatment with LPS increased the expression of COX-2 protein 7 h and 7 days after the injection in the dentate gyrus. These results suggest that LPS impairs the survival of newly generated cells derived from the NSCs in the dentate gyrus without affecting the differentiation fate, and these effects of LPS were mediated presumably by COX-2 expression in the dentate gyrus.

Physalis angulata is a popular medicine used in Brazil due to its anti-inflammatory effects, but the pharmacological mechanisms underlying these actions remain to be better understood. In the present work, lyophilized aqueous extract from the roots of Physalis angulata Linneu (AEPa) was used to control the inflammatory response induced by the injection of 1% carrageenan into subcutaneous rat's air pouches. Adenosine deaminase (ADA) activity, nitrite level, and prostaglandin E(2) (PGE(2)) level were used to evaluate the action of inflammatory mediators. Tumor growth factor-beta (TGF-beta) level was used as a bioindicator of immunomodulatory response. Rats were injected with vehicle, indomethacin, or AEPa (0.5 mg/kg, 1 mg/kg, and 5 mg/kg i.p.), 1h before carrageenan administration. AEPa at 0.5 mg/kg had no effect. However, 1mg/kg of AEPa showed significant anti-inflammatory effects, decreasing exudate volume, total number of inflammatory cells, ADA activity, nitrite level, and PGE(2) level in 50%, 41%, 20%, 60%, and 41%, respectively. The anti-inflammatory effects of 5 mg/kg AEPa appeared to be more effective than those of 1 mg/kg AEPa (84%, 80%, 43%, 70%, and 75%, respectively). In addition, TGF-beta level was upregulated to 9700 pg/ml after 5mg/kg AEPa, in comparison with 160 pg/ml in the vehicle-treated group, and 137 pg/ml in the indomethacin-treated group. The results indicate that AEPa exerts powerful anti-inflammatory and immunomodulatory activities, interfering with the cyclooxygenase pathway, lymphocyte proliferation, NO, and TGF-beta production.

Dental caries and periodontal disease are associated with oral pathogens. Several plant derivatives have been evaluated with respect to their antimicrobial effects against such pathogenic microorganisms. Lippia sidoides Cham (Verbenaceae), popularly known as "Alecrim-pimenta" is a typical shrub commonly found in the Northeast of Brazil. Many plant species belonging to the genus Lippia yield very fragrant essential oils of potential economic value which are used by the industry for the commercial production of perfumes, creams, lotions, and deodorants. Since the leaves of L. sidoides are also extensively used in popular medicine for the treatment of skin wounds and cuts, the objective of the present study was to evaluate the composition and antimicrobial activity of L. sidoides essential oil. The essential oil was obtained by hydro-distillation and analyzed by GC-MS. Twelve compounds were characterized, having as major constituents thymol (56.7%) and carvacrol (16.7%). The antimicrobial activity of the oil and the major components was tested against cariogenic bacterial species of the genus Streptococcus as well as Candida albicans using the broth dilution and disk diffusion assays. The essential oil and its major components thymol and carvacrol exhibited potent antimicrobial activity against the organisms tested with minimum inhibitory concentrations ranging from 0.625 to 10.0 mg/mL. The most sensitive microorganisms were C. albicans and Streptococcus mutans. The essential oil of L. sidoides and its major components exert promising antimicrobial effects against oral pathogens and suggest its likely usefulness to combat oral microbial growth.

Gene regulatory networks, or simply gene networks (GNs), have shown to be a promising approach that the bioinformatics community has been developing for studying regulatory mechanisms in biological systems. GNs are built from the genome-wide high-throughput gene expression data that are often available from DNA microarray experiments. Conceptually, GNs are (un)directed graphs, where the nodes correspond to the genes and a link between a pair of genes denotes a regulatory interaction that occurs at transcriptional level. In the present study, we had two objectives: 1) to develop a framework for GN reconstruction based on a Bayesian network model that captures direct interactions between genes through nonparametric regression with B-splines, and 2) to demonstrate the potential of GNs in the analysis of expression data of a real biological system, the yeast pheromone response pathway. Our framework also included a number of search schemes to learn the network. We present an intuitive notion of GN theory as well as the detailed mathematical foundations of the model. A comprehensive analysis of the consistency of the model when tested with biological data was done through the analysis of the GNs inferred for the yeast pheromone pathway. Our results agree fairly well with what was expected based on the literature, and we developed some hypotheses about this system. Using this analysis, we intended to provide a guide on how GNs can be effectively used to study transcriptional regulation. We also discussed the limitations of GNs and the future direction of network analysis for genomic data. The software is available upon request.

In this study, we attempted to identify the possible antinociceptive action of aqueous extract (AE) obtained from roots of Physalis angulata, known in Brazil as "Camapu", used to treat various pain-related physiological conditions. The AE of Physalis angulata (10-30 mg/kg) given by i.p. or p.o. route, 0.5 and 1h prior, produced significant inhibition of abdominal constrictions caused by acetic acid, with ID(50) values of 18.5 (17.4-19.8) and 21.5 (18.9-24.4)mg/kg and inhibitions of 83+/-8 and 66+/-5%, respectively. The AE (10-60 mg/kg, i.p.) also caused significant inhibition of the late-phase of formalin-induced pain, with an ID(50) value of 20.8 (18.4-23.4)mg/kg and inhibition of 100%. Treatment of mice with AE (60 mg/kg, i.p.) or with morphine (10mg/kg, i.p.) produced a significant increase of the reaction time in the hot-plate test. These results demonstrate, for the first time, that the AE of Physalis angulata produce marked antinociception against the acetic acid-induced visceral pain and inflammatory pain responses induced by formalin in mice. The mechanism by which the AE produces antinociception still remains unclear. However, pharmacological and chemical studies are continuing in order to characterize the mechanism(s) responsible for the antinociceptive action and also to identify the active principles present in Physalis angulata. Moreover, the antinociceptive action demonstrated in the present study supports, at least partly, the ethnomedical uses of this plant.

Changes in nutritional status are related to an increase in morbidity and mortality. It is well established that health care professionals, particularly physicians, lack formal education to recognize nutritional disorders, which malnutrition may worsen in the hospital, and that appropriate education could effectively correct this problem. Aim: To evaluate the awareness of the health care university students and professionals about the nutrition disorders in Amazon region hospitalized patients before and after a specific education program. Study design: An intensive Course on Clinical Nutrition, given in three weekly classes was offered to the multidisciplinary health care students and professionals of the health care tem. Cases and methods: 195 participants started the course. They were 97 students of nursing, 52 students of medicine, 20 nurses, 15 resident physicians, 3 physicians, 3 dietitians, 2 others, 1 no answer. 165 participants concluded it. Precourse and postcourse tests were answered. The results of these tests are presented in this study. Results and Discussion: 76.4% of the participants were graduation students. 40% of participants had no contact with patients yet. When the exposure of the participants of the course to the theme Nutrition was analyzed, 46.7% of the resident physicians considered their contact with the subject superficial and 13.3% had no contact with it, reinforcing the physician´s lack of knowledge to recognize malnutrition. Participants admitted having significantly profited from the course. Conclusions: the lack of awareness of the multidisciplinary health care team about nutritional disorders may contribute to worsen hospital malnutrition. An intensive course for graduate students and residents may help to raise awareness for clinical nutrition. The great interest and enthusiasm shown by the students and other participants and the profit obtained by them from the course suggest that teaching of clinical nutrition should be implemented in all health care schools (AU)


Las alteraciones del estado nutricional están relacionadas con el aumento de la morbilidad y mortalidad. A pesar de no haber consenso en relación a los criterios diagnósticos de la malnutrición, ésta se agrava a medida que aumenta el tiempo de internación hospitalaria. Diversos autores concluyen que el equipo de salud, particularmente los médicos, no están siendo enseñados a reconocer la malnutrición, que esta malnutrición es agravada en el hospital y que la educación puede efectivamente corregir el problema. Objetivo: Evaluar la concientización de los estudiantes de graduación en salud y de los profesionales de la comisión multidisciplinar de salud al respecto de los desórdenes nutricionales de pacientes hospitalizados antes y después del programa intensivo de educación. Diseño del estudio: Un curso intensivo de Nutrición Clínica, fue administrado en tres clases semanales para los estudiantes de graduación en salud y profesionales en el equipo de salud. Pacientes y métodos. 195 participantes iniciaron el curso, frecuentado por 97 estudiantes de enfermería, 52 estudiantes de medicina, 20 enfermeros, 15 médicos residentes, 3 médicos, 3 nutricionistas, 2 otras actividades y 1 sin respuesta. 165 concluyeron el curso. Una prueba antes y una prueba después del curso fueron respondidas. El resultado de estas pruebas es presentado en este estudio. Resultados y discusión: 76,4 por ciento de los participantes son alumnos, con edad media de 24,39 años. 40 por ciento de los participantes aún no habían tenido contacto con pacientes. Cuando la exposición de los participantes del curso al tema Nutrición fue analizada, 46,7 por ciento de los médicos residentes consideró su contacto anterior con el tema como superficial y 13,3 por ciento no habían tenido ningún contacto, reforzando la idea de muchos autores que los médicos no están siendo enseñados a reconocer una malnutrición debido a falta de énfasis dada a la nutrición del currículum médico. Los participantes del curso de Nutrición Clínica reconocieron haber obtenido lucro significante. Conclusiones: Malnutrición hospitalaria puede ser agravada por la falta de concientización de la comisión multidisciplinar de salud al respecto del problema. Pero el gran interés y entusiasmo demostrado por los participantes y el lucro obtenido con el curso sugiere que la llave para abrir la puerta del armario es la educación. La enseñanza de Nutrición debe ser implementada en todas las escuelas de profesiones que cuidan de la salud (AU)

The authors make a retrospective review of 53 new cases of Nephrotic Syndrome followed up in the Nephrology Unit from November 1988 to March 1994, bearing in mind the evaluation of casual changes of the disease standard regarding a previous study of 1988. Epidemiological, clinical, therapeutical and evolutional aspects were studied. Forty-four cases of primary Nephrotic Syndrome (83%) were identified, 61.4% of which behaved as cortico-sensitive, 25% as cortico-dependent, and 13.6% as cortico-resistant; 8 cases (15%) of Nephrotic Syndrome secondary to infection, Systemic Lupus Erythematosus and Amyloidosis, and 1 case of congenital Nephrotic Syndrome (2%). The theory that the high number of cortico-dependent is, probably, related with a higher severity in the relapse diagnosis and/or changes in the children's standard of living is admissible. It was also observed that at present there is a lower number of hospital discharges, related to more careful attitudes adopted regarding the evolution of the disease.
