RESUMO

Sex-gender-diverse and non-monogamous strain cisnormativity and mononormativity. In scientific terms, the parenting arrangements of these people are uncertain. Thus, this ethnography aims to understand the perception of non-monogamous sex-gender-diverse people about parenting. The theoretical framework adopted is derived from non-monogamous studies, love and sexuality from the Social and Human Sciences in Public Health and the digital ethnography methodological framework. Fieldwork occurred from 2021 to 2022 through an online WhatsApp group. Participant observation was employed in the group, and semi-structured online interviews were held. Two categories emerged: a) The non-monogamous parenting nodes and b) Collective parenting. In the first, the importance of bonds in affective networks is explored, and the barriers to these family arrangements are exposed. The second describes the importance of living in a community, and Indigenous and Black ancestry is revived. The revived ancestry and ways of living in a community gain importance as we understand their relevance in the experience of parenting for sex-gender-diverse people who are non-monogamous.


Pessoas sexo-gênero-diversas e que são não-monogâmicas tensionam a cisnormatividade e a mononormatividade. Em termos científicos, há uma nebulosidade quanto aos arranjos parenterais dessas pessoas. Assim, esta etnografia objetiva compreender a percepção de pessoas sexo-gênero-diversas não-monogâmicas sobre parentalidades. O referencial teórico utilizado partiu dos estudos não-monogâmicos, amor e sexualidade das Ciências Sociais e Humanas em Saúde da Saúde Coletiva, e o metodológico da etnografia digital. O trabalho de campo ocorreu entre 2021 e 2022, em um grupo on-line do WhatsApp. A observação participante foi empregada no grupo e foram realizadas entrevistas on-line semiestruturadas. Na primeira, se explora a importância do vínculo nas redes afetivas e expõe as barreiras desses arranjos familiares. Já na segunda, se descreve a importância do viver em comunidade, bem como o resgate à ancestralidade indígena e negra. O resgate à ancestralidade e às formas de se viver em comunidade ganham relevo à medida que se compreende a importância que estes possuem na vivência das parentalidades de pessoas sexo-gênero-diversas e que são não-monogâmicas.

This study evaluated the impact of long-term storage conditions (medium and time) on the development of experimental dentinal microcracks through micro-computed tomography. Sixty freshly extracted premolars were stored in formalin, water, or dry conditions (n = 20) and scanned after 72 h, 30 days, 6 months and 3 years of extraction. The effect of the storage medium and time on the occurrence of dentinal defects was statistically evaluated. A total of 211 000 images were screened revealing the existence of 11 519 slices with dentinal defects. Dry conditions significantly contributed to the development of new defects in all time points. During the 3-year follow-up period, no new defects were detected in the teeth that were stored in water and, in a single tooth, in the formalin group, after 6 months of storage.

Abstract Background A cost of illness (COI) study aims to evaluate the socioeconomic burden that an illness imposes on society as a whole. This study aimed to describe the resources used, patterns of care, direct cost, and loss of productivity due to systemic lupus erythematosus (SLE) in Brazil. Methods This 12-month, cross-sectional, COI study of patients with SLE (ACR 1997 Classification Criteria) collected data using patient interviews (questionnaires) and medical records, covering: SLE profile, resources used, morbidities, quality of life (12-Item Short Form Survey, SF-12), and loss of productivity. Patients were excluded if they were retired or on sick leave for another illness. Direct resources included health-related (consultations, tests, medications, hospitalization) or non-health-related (transportation, home adaptation, expenditure on caregivers) hospital resources. Costs were calculated using the unit value of each resource and the quantity consumed. A gamma regression model explored cost predictors for patients with SLE. Results Overall, 300 patients with SLE were included (92.3% female, mean [standard deviation (SD)] disease duration 11.8 [7.9] years), of which 100 patients (33.3%) were on SLE-related sick leave and 46 patients (15.3%) had stopped schooling. Mean (SD) travel time from home to a care facility was 4.4 (12.6) hours. Antimalarials were the most commonly used drugs (222 [74.0%]). A negative correlation was observed between SF-12 physical component and SLE Disease Activity Index (- 0.117, p = 0.042), Systemic Lupus International CollaboratingClinics/AmericanCollegeofRheumatology Damage Index (- 0.115, p = 0.046), medications/day for multiple co-morbidities (- 0.272, p < 0.001), SLE-specific drugs/day (- 0.113, p = 0.051), and lost productivity (- 0.570, p < 0.001). For the mental component, a negative correlation was observed with medications/day for multiple co-morbidities (- 0.272, p < 0.001), SLE-specific medications/day (- 0.113, p = 0.051), and missed appointments (- 0.232, p < 0.001). Mean total SLE cost was US$3,123.53/patient/year (median [interquartile range (IQR)] US$1,618.51 [$678.66, $4,601.29]). Main expenditure was medication, with a median (IQR) cost of US$910.62 ($460, $4,033.51). Mycophenolate increased costs by 3.664 times (p < 0.001), and inflammatory monitoring (erythrocyte sedimentation rate or C-reactive protein) reduced expenditure by 0.381 times (p < 0.001). Conclusion These results allowed access to care patterns, the median cost for patients with SLE in Brazil, and the differences across regions driven by biological, social, and behavioral factors. The cost of SLE provides an updated setting to support the decision-making process across the country.

Resumo Centrando-se no impacto que a comunicação de risco emitida pelas organizações de saúde pública tem na mudança dos comportamentos da sociedade, esta investigação pretende analisar as mensagens-chave que a Organização Mundial de Saúde (OMS) definiu para promover o programa de vacinação contra a covid-19. Para cumprir este objetivo, enveredou-se por uma metodologia de estudo qualitativa, que privilegiou o uso da análise do conteúdo publicado nas páginas de Facebook e de Instagram da OMS, no período de 1 de abril a 31 de agosto de 2021. No total, foram analisadas 62 publicações. Os resultados mostraram que a OMS utilizou quatro eixo de comunicação para promover a importância da vacinação na sociedade: garantir a credibilidade e a transparência da informação transmitida; certificar a segurança e a eficácia da vacina; apelar ao sentido de responsabilidade coletiva; e associar a vacina à solução para pôr fim à pandemia. As conclusões do estudo mostram que, embora a equidade no acesso à vacina ainda seja uma realidade em construção, os quase 70% da população mundial vacinada sugerem que as mensagens enviadas pela OMS no contexto de comunicação de risco podem ter contribuído para a construção de uma imagem positiva do programa de vacinação.


Abstract Focusing on the impact that risk communication issued by public health organizations brings to changing societal behaviors, this research aimed to analyze the key messages that the World Health Organization (WHO) has defined to promote the COVID-19 vaccination program. In order to achieve this goal, a qualitative study methodology was used to analyze the content published on the WHO's Facebook and Instagram pages from April 1 to August 31, 2021. In total, 62 publications were analyzed. The results showed that the WHO used four key communication messages to promote the importance of vaccination in society: ensuring the credibility and transparency of the information transmitted; certifying the safety and efficacy of the vaccine; appealing to a sense of collective responsibility; and associating the vaccine with the solution to end the pandemic were the chosen communication axes. The conclusions of this study show that, although fair access to the vaccine is still a reality in the making, the fact that almost 70% of the world's population has been vaccinated suggests that the messages sent by the WHO in the context of risk communication may have contributed to building a positive image of the vaccination program.

Introduction - Glycogen storage disease type V (GSDV, MIM #232600) is an autosomal recessive metabolic myopathy caused by pathogenic variants in the PYGM gene. The characteristic symptoms of exercise intolerance, myalgia, and cramps, which improve after a few minutes of rest, are frequently unrecognized in affected children. When there is clinical suspicion, the initial approach with a forearm exercise test has diagnostic value by detecting low post-exercise plasma lactate-to-ammonia ratio values. The diagnostic algorithm is followed by genetic testing if the results suggest myophosphorylase deficiency. Methods - This was a retrospective observational study conducted based on reviewing medical records of patients with GSDV in a tertiary hospital. We assessed demographic variables, including the timing of onset and diagnosis, relevant clinical characteristics, and whether genetic testing was performed, including its results. Results/Case Report - Our goal was to review the GSDV cases in our center to assess our cohort's diagnostic timing and clinical and genetic characteristics. We identified 28 patients from 24 families, three with consanguinity. The mean age at the time of the study was 43 years. While most (26/28; 93%) recalled their first symptoms in childhood/adolescence, only 25% (7/28) were diagnosed then. All patients had exercise intolerance and CK elevation, while about half reported the second wind phenomenon. Genetic testing was performed in 22 patients, revealing biallelic PYGM variants (9 homozygous, 13 compound heterozygous) as the most common (p.R50*). Conclusion - GSDV is rare and presents in the pediatric age, with subtle manifestations often underestimated for decades. A late diagnosis may negatively impact the psychosocial development of affected children. It is essential to recognize some unique features that facilitate diagnosis: history of exercise intolerance, the second wind sign, and high resting serum CK levels. Identifying the disease-causing variants in PYGM is currently the gold standard for diagnosis as it is less invasive than performing a muscle biopsy, and may promptly diagnose the condition and avoid wrongful labelling of patients.

Detecting volatile organic compounds is a fundamental step in water quality analysis. Methylisoborneol (MIB) provides a lousy odor to water, whereas geosmin (GEO) is responsible for its sour taste. A widely-used technique for their detection is gas-phase chromatography. On the other hand, an electronic nose from organic thin-film transistors is a cheaper and faster alternative. Poly(2,5-bis(3-tetradecyl-thiophen-2-yl)thieno[3,2-b]thiophene) (PBTTT-C14) features semiconducting properties suitable for organic electronics. However, in order to expose the active layer in a bottom-gate transistor structure with photolithographically patterned electrodes, a cross-linked dielectric such as poly(4-vinyl phenol) (PVP) is necessary. In this work, the cross-linking was demonstrated using FTIR and Raman spectroscopies, as well as high-k capacitors with a dielectric constant of 5.3. The presence of enhanced crystallinity with terrace formation in the semiconducting film was confirmed with UV-visible spectrophotometry, atomic force microscopy, and X-ray diffraction. Finally, for the first time, a PBTTT-C14 transistor on cross-linked PVP was shown to respond to isoborneol with a sensitivity of up to 6% change in mobility per ppm. Due to its similarity to MIB, a system comprising these sensors must be investigated in the future as a tool for sanitation companies in real-time water quality monitoring.

METHODS: integrative review, carried out in June 2020, without time frame, in the Scopus, MEDLINE, Embase, CINAHL, WoS, PsycInfo and LILACS databases. The controlled descriptors of DeCS, MeSH and their entry terms were used: "Transgender People", "Transgender", "Gender Identity", "Transsexuality", "Gender Violence", "Aggression", "Sexual Offenses", "Rape", "Violence", "Domestic Violence". The presentation and synthesis of the results were presented in the PRISMA-2009 flowchart. RESULTS: the final sample, consisting of 16 articles, identified different types of violence (sexual, physical, verbal, psychological and financial), perpetrated by family members, strangers, police officers, intimate partners, health professionals, acquaintances, or friends. CONCLUSIONS: trans women suffer violence and social exclusion that result from stigma and discrimination due to gender identity and result in unrestricted damage to physical health.

RESUMO

The main objective of this text is to understand the meanings attributed to penile cancer by transvestites sex workers. This is a qualitative and descriptive study carried out in Recife, Pernambuco, in 2019. The literature on Anthropology of Health in the field of Social Sciences in Health constituted a theoretical framework. Eight black interlocutors participated, where a field notebook, a sociodemographic characterization form and a semi-structured interview script were used in the production of data. For the analysis, thematic coding was used, resulting in two categories "embodiment of the 'new' disease" and "it's like the end of life and career!". Finally, the data located the meanings of penile cancer from the social and historical context and from the transvestites' own experiences, where the embodiment of the 'new' disease gained prominence as metaphors were constructed to designate it, as well as the social and career death that it would represent in their lives.  


Objetivou-se compreender os significados atribuídos ao câncer de pênis pelas travestis profissionais do sexo. Este é um estudo qualitativo e descritivo realizado em Recife, Pernambuco, no ano de 2019. A literatura da Antropologia da Saúde do campo das Ciências Sociais em Saúde se constituiu como referencial teórico. Participaram oito interlocutoras negras, onde foi utilizado um caderno de campo, formulário de caracterização sociodemográfico e roteiro de entrevista semi-estruturado na produção dos dados. Para a análise, empregou-se a codificação temática resultando em duas categorias "corporificação da 'nova' doença" e "é como se fosse o fim de vida e de carreira!". Por fim, os dados localizaram os significados do câncer de pênis a partir do contexto social, histórico e das próprias experiências das travestis, onde a corporificação da 'nova' doença ganhou relevo à medida que se construíram metáforas para designá-la, bem como à morte social e de carreira que esta representaria em suas vidas.

This study evaluated the efficacy of sodium thiosulphate (ST) as an intermediate irrigant between sodium hypochlorite (NaOCl) and chlorhexidine (CHX) to avoid the formation of the brown-coloured precipitate. The in vitro experiment used microtubes to test the mixture of 2.5% NaOCl, 2% CHX and 5% ST solutions, varying their order and time of addition. In the ex vivo experiment, 10 bovine teeth were divided into two groups according to the intermediate irrigant used: distilled water (control) and 5% ST (experimental). Colour change occurred in all microtubes after interaction between solutions. While the precipitate was not observed in the experimental group, it was seen in all specimens of the control group. The 5% ST was able to prevent the formation of the brown-coloured precipitate in the ex vivo experiment, whereas it only prevented its in vitro formation when the waiting time for adding CHX was 10, 15 and 20 min.

La presentación y la síntesis de los resultados fueron presentadas en el flujograma PRISMA-2009. Resultados: la muestra final, compuesta por 16 artículos, identificó violencias de género diversas (sexual, física, verbal, psicológica y financiera), perpetradas por familiares, desconocidos, policiales, parceros íntimos, profesionales de salud, conocidos o amigos. Conclusiones: la mujer trans sufre violencias y exclusiones sociales que transcurren de estigmas y discriminaciones por la identidad de género y resultan en daños no restrictos a salud física.

Conclusões: a mulher trans sofre violências e exclusões sociais decorrentes de estigmas e discriminações pela identidade de gênero que resultam em danos à saúde além da física.

Esta pesquisa qualitativa objetivou compreender as relações entre acesso a serviços de saúde e experiências de sofrimento social entre pessoas trans. Foram conduzidas entrevistas semiestruturadas com cinco interlocutores e observações participantes. Os dados foram analisados por meio da codificação temática, originando duas categorias: "A negação do nome" e "Acesso à saúde e transfobia institucionalizada no Sistema Único de Saúde (SUS)". As narrativas das/os interlocutores permitiram localizar tais relações na sociogênese de experiências de sofrimento social. A negação do nome implica negação da humanidade da pessoa trans, bem como patologização de sua identidade e prejuízos no acesso à saúde, colocando a automedicação como uma possibilidade de agência. Concluiu-se que a transfobia institucionalizada no setor de saúde reproduz a precarização da cidadania de pessoas trans, destacando quanto a ação estatal com vistas a mitigar o sofrimento social pode, por vezes, intensificá-lo. The aim of this qualitative study was to understand the relationships between access to health services and experiences of social suffering among trans people. We conducted semi-structured interviews with five participants and participant observation. The data were analyzed using thematic coding, giving rise to two categories: "Name denial" and "Access to health care and institutionalized transphobia in Brazilian National Health System". The participants' narratives allowed the researchers to situate these relationships within the sociogenesis of social suffering experiences. Name denial implies the negation of the humanity of trans people and pathologization of their identity, and compromises access to health care, making self-medication a possibility of agency. We conclude that institutionalized transphobia in the public health care system reproduces the precariousness of the citizenship of trans people, highlighting how government actions aimed at mitigating social suffering can sometimes intensify it. Esta investigación cualitativa tuvo el objetivo de comprender las relaciones entre acceso a servicios de salud y experiencias de sufrimiento social entre personas trans. Se realizaron entrevistas semiestructuradas con cinco interlocutores y observaciones participantes. Los datos se analizaron por medio de la codificación temática originando dos categorías: "La negación del nombre" y "Acceso a la salud y transfobia institucionalizada en el Sistema Único de Salud". Las narrativas de los interlocutores permitieron localizar tales relaciones en la sociogénesis de experiencias de sufrimiento social. La negación del nombre implica la negación de la humanidad de la persona trans, así como la patologización de su identidad y prejuicios en el acceso a la salud, planteando la automedicación como una posibilidad de agencia. Se concluyó que la transfobia institucionalizada en el sector de la salud reproduce la precarización de la ciudadanía de personas trans, destacando hasta qué punto la acción estatal dirigida a mitigar el sufrimiento social puede, algunas veces, intensificarlo. Objetivo: Identificar las representaciones sociales de las trabajadoras sexuales travestis sobre la calidad de vida. Material y Método: Estudio cualitativo, descriptivo, exploratorio, basado en la Teoría de las Representaciones Sociales, desarrollado con siete travestis trabajadoras sexuales. La producción de los datos se llevó a cabo con entrevistas semiestructuradas y posteriormente se transcribieron, validaron y analizaron a partir del Análisis de contenido temático. Resultados: Surgieron tres clases: 1) Acceso a la salud como principio de calidad de vida; 2) El apoyo de las organizaciones no gubernamentales en la visibilidad y respeto a las demandas de las personas trans y 3) Los vínculos sociales como herramienta útil en el sentido de la calidad de vida. Consideraciones finales: Las representaciones se ubican en la necesidad de acceso a servicios de salud libres de prejuicios; el apoyo de las organizaciones no gubernamentales en el reconocimiento de sus potencialidades y singularidades mediante el establecimiento de vínculos afectivos, solidarios, leales y de confianza, y en el esTablecimiento de lazos sociales producidos con vecinos y amigos para afrontar las dificultades vividas cotidianamente. ABSTRACT: COVID-19 was characterized as a pandemic due to the worldwide dissemination and the severity with which the disease attacks the human organism. Some oral lesions have been observed in COVID-19 patients. However, there is still no concrete evidence of the real influence of SARS-CoV-2 on the human body, especially in the oral region. In this context, the present report discusses a case of a COVID-19 patient with oral alterations. The male patient presented ulcerative lesions of painful symptomatology and petechiae in the oral mucosa. This study also performed a literature review of the main oral alterations reported in the literature. Although more studies with a larger number of cases should be performe d, the present clinical case may have manifested signs of this pathology in the oral cavity since the epithelial cells of the oral mucosa have ACE2 receptors.


RESUMEN: El COVID-19 se caracterizó como una pandemia debido a la diseminación mundial y la gravedad con la que la enfermedad ataca al organismo humano. Se han observado algunas lesiones orales en pacientes con COVID-19. Sin embargo, todavía no hay evidencia concreta de la influencia del SARS-CoV-2 en el cuerpo humano, especialmente en la cavidad oral. En este contexto, el presente reporte analiza un caso de un paciente con COVID-19 con alteraciones orales. El paciente de sexo masculino presentó lesiones ulcerativas de sintomatología dolorosa y petequias en la mucosa oral. Este estudio también realizó una revisión de la literatura de las principales alteraciones orales reportadas en la literatura. Si bien se deben realizar más estudios con un mayor número de pacientes, el presente reporte de caso puede haber manifestado signos de esta patología en la cavidad oral ya que las células epiteliales de la mucosa oral tienen receptores ACE2.

The aim of the present study was to evaluate the cyclic fatigue, torsional resistance and surface roughness of Reciproc R25 instruments in four different situations, namely as new instruments and as instruments tested after clinical preparation of one, two or three maxillary molars with four root canals. The total time required to perform each root canal preparation was recorded. Cyclic fatigue resistance was determined by the time to fracture using a customized testing device (n = 10 per group). The torsional test evaluated the torque and angle of rotation to failure according to ISO 3630-1 (n = 10 per group). The roughness of the working parts of new and used instruments was evaluated with a profilometer (n = 5 per group). Statistical analysis was performed using one-way ANOVA and Tukey's test. The level of significance was set at 5%. No fractures or deformations were observed after clinical use. Higher preparation time was needed during the third use of the instruments for all root canals (p < 0.05). There were no significant differences among the groups in regard to either cyclic fatigue or torsional resistance (p > 0.05). Regarding the roughness measurements, groove depth was higher on new and one- versus two- or three-maxillary-molar-prepared instruments (p < 0.05). It can be concluded that the clinical use of Reciproc instruments increased preparation time and decreased surface roughness. However, clinical use did not affect the cyclic fatigue or torsional resistance of the Reciproc instruments.

BackgroundAntigen-detecting rapid diagnostic tests (Ag-RDTs) for the detection of SARS-CoV-2 offer new opportunities for testing in the context of the COVID-19 pandemic. Nasopharyngeal swabs (NPS) are the reference sample type, but oropharyngeal swabs (OPS) may be a more acceptable sample type in some patients. MethodsWe conducted a prospective study in a single screening center to assess the diagnostic performance of the Panbio COVID-19 Ag Rapid Test (Abbott) on OPS compared with reverse-transcription quantitative PCR (RT-qPCR) using NPS. Results402 outpatients were enrolled in a COVID-19 screening center, of whom 168 (41.8%) had a positive RT-qPCR test. The oropharyngeal Ag-RDT sensitivity compared to nasopharyngeal RT-qPCR was 81% (95%CI: 74.2-86.6). Two false positives were noted out of the 234 RT-qPCR negative individuals, which resulted in a specificity of 99.1% (95%CI: 96.9-99.9) for the Ag-RDT. For cycle threshold values [≤] 26.7 ([≥] 1E6 SARS-CoV-2 genomes copies/mL, a presumed cut-off for infectious virus), 96.3% sensitivity (95%CI: 90.7-99.0%) was obtained with the Ag-RDT using OPS. InterpretationBased on our findings, the diagnostic performance of the Panbio Covid-19 RDT with OPS samples meet the criteria required by the WHO for Ag-RDTs (sensitivity[≥]80% and specificity [≥]97%).

OBJECTIVES: This study evaluated the danger zone (DZ) in mesial roots of mandibular molars and the correlation between anatomical references of the DZ and some anatomical landmarks including tooth/root length, depth of mesial and distal grooves, and inter-canal orifices distance. MATERIAL AND METHODS: Twenty-eight mesial roots of mandibular molars with 2 independent canals were scanned and divided into 2 groups according to root length. The anatomical landmarks were correlated (Pearson or Spearman coefficients) with root level, thickness, and position of the DZ and also compared (independent samples t or Mann-Whitney tests) between the 2 groups at α = 5%. RESULTS: No statistical difference was observed between groups regarding DZ parameters and depth of mesial and distal grooves (P > 0.05). Orifice distance in group 2 (4.49 ± 0.75 mm) was significantly greater than group 1 (3.76 ± 0.89 mm) (P < 0.05). Significant correlations (P < 0.05) were found between (i) DZ level and root/tooth length (r = 0.54 and 0.49, respectively), (ii) DZ thickness and distal groove depth (r = - 0.45), and orifice distance (r = 0.38), and (iii) DZ position and depth of mesial (r = 0.39) and distal (r = 0.40) grooves. Other variables such as root length and distal groove depth (r = 0.28), and orifice distance and mesial groove depth (r = 0.36) were also correlated (P < 0.05). CONCLUSIONS: The length of tooth/root, the distance of canal orifices, and the depth of mesial/distal grooves of mesial roots of mandibular molars might be predictive factors for the root level, position, and thickness of the DZ. CLINICAL RELEVANCE: The length, distance of mesial canal orifices, and the depth of mesial and distal grooves of the mesial roots of mandibular molars might be moderate predictive factors for the root level, position, and thickness of the DZ.

INTRODUCTION: This study assessed the number of mesial and distal canals of mandibular molars in which the R-Pilot pathfinding reciprocating instrument reached the working length (WL) during macro glide path procedure. Fracture and deformation rates were also evaluated. METHODS: One hundred fifty-six root canals of 52 teeth were scouted to the length of the apical foramen. Then the R-Pilot instrument was positioned at the canal orifice and activated. The instrument was moved by using a pecking motion and light apical pressure. This procedure was repeated in an attempt to reach the WL. The type of fracture and/or instrument deformation was assessed by scanning electron microscopy, whereas the roots with fractured instruments were scanned through micro-computed tomography. The percentage frequency distribution of fractures, deformations, and root canals in which the R-Pilot reached the WL were recorded and statistically compared by using the Pearson's χ2 test with α = 5%. RESULTS: R-Pilot instruments reached the WL in 139 root canals (89.10%), and χ2 test showed a significant difference between the observed frequencies and the expected frequencies (χ2 = 95.41, P = .000). The observed frequencies of fractures (2.56%) and deformations (1.92%) were also significantly lower than the expected (fracture: χ2 = 140.41, P = .000; deformation: χ2 = 144.23, P = .000). Fractures occurred mostly at the apical and curved parts of the root canals. CONCLUSIONS: R-Pilot reached the WL in 89.10% of the root canals of mandibular molars with fracture and deformation rates of 2.56% and 1.92%, respectively.

OBJECTIVES: To present and explore the potential of an animal-based experimental model developed to determine the set of root canal sealers in vivo. The setting of AH Plus, BioC Sealer, TotalFill BC Sealer, and Sealapex was determined using either ISO 6876 or the novel in vivo method proposed in this study. MATERIAL AND METHODS: The in vitro setting time of the sealers tested was determined in accordance with ISO 6876:2012. In determining the in vivo set, 24 adult Wistar rats were followed up for two evaluation periods: 1 and 4 weeks. Their upper-right incisor was extracted, and its pulp tissue was removed. The root canal was then filled from retrograde with one of the 4 sealers, and the tooth was re-implanted and fixed with a layer of a flowable composite resin. After 1 or 4 weeks of the surgical procedures, the animals were euthanized, and their incisors were extracted. Two-mm-thick slices of the middle third of the tooth root were obtained and assessed with a Gillmore device, to determine whether or not the sealer had set. RESULTS: The following in vitro results were obtained by using ISO 6876 methodology: AH Plus set after a mean time of 423 ± 20 min and 476 ± 35 min, in metal and plaster molds, respectively. BioC Sealer set after 7 days (in dental plaster molds), whereas TotalFill BC Sealer and Sealapex did not set even after 25 days in both tested conditions (metal or dental plaster molds). Using the novel in vivo methodology, AH Plus, BioC Sealer, and TotalFill BC Sealer set after both 7 and 30 days. In contrast, Sealapex did not set at either time point. CONCLUSIONS: AH Plus and BioC Sealer set under both in vitro and in vivo test conditions. TotalFill BC Sealer did not set under in vitro conditions but did after 1 week under in vivo conditions. Sealapex did not set under either in vitro or in vivo conditions. CLINICAL RELEVANCE: The influence of the testing conditions on the setting results is a clear indication that new in vivo experimental models should be useful in future studies on Bioceramics root canal sealers.
