RESUMO

Objective To investigate the safety and efficacy of transarterial chemoembolization (TACE) combined with implantation of irradiation IVC stent in treating hepatocellular carcinoma (HCC)complicated by inferior vena cava tumor thrombosis (IVCTT).Methods The clinical data of 61 consecutive patients with HCC complicated by IVCTT were retrospectively analyzed.Irradiation IVC stent was prepared by strapping 125I particles on the bare stent,and it was employed in 33 patients (group A).Bare stent was adopted in 28 patients (group B).Propensity score matching method was used to conduct randomized analysis of the original data in order to reduce the selection bias.The survival time,remission rate of symptom and procedure-related adverse events of both groups were calculated and the results were compared between the two groups.Results The incidence of adverse reactions was similar in the two groups,and symptomatic treatment with internal medicine was adopted.The survival time in group A was superior to that in group B.The median survival time in group A was (203.0±28.1) days,which was (93.0±24.3) days in group B (P=0.006).Propensity score matching (24 pairs in total) cohort analysis showed that the median survival time was (200±31) days in group A and (66±23) days in group B (P=0.019).The edema remission rates in group A and in group B were 97.0% and 96.4% respectively.Multiple factor analysis revealed that irradiation stent implantation and objective tumor response were independent factors predicting a good prognosis.Conclusion For the treatment of HCC associated with IVCTT,TACE combined with irradiation stent implantation is safe and effective,this therapy can prolong the patient's survival time

Objective To investigate the information obtained by bile culture and susceptibility testing as while factors like styles of obstruction,abdominal surgical history,degree and time of jaundice related to bacterial infection of biliary tract for patients with malignant obstructive jaundice (MOJ).Methods A total of 93 patients with MOJ received percutaneous transhepatic biliary drainage (PTBD) during Jan.,2015 and Mar.,2016.Bile specimens were collected during the procedure for bacterial culture and susceptibility.Results A rate of 60.2% (56/93) patients had a positive bile culture and 58 strains were identified,in which gram-positive bacteria accounted for 36.2 % (21/58) and gram-negative bacteria accounted for 63.8 % (37/58).The most common bacteria were Enterococcus Faecalis,Escherichia Coli,Klebsiella Pneumonia and Staphylococcus Epidermidis.Conclusions The bile cultures indicated that a different antibacterial therapy should be managed in the patients of biliary infection complicated by malignant biliary jaundice.Low obstruction and abdominal surgery history are independent factors related to infection,while time and degree of jaundice shows no relationship to infection.

BACKGROUND: Dysregulated expression of microRNAs is often found in gastric cancer, and it contributes to the pathogenesis of gastric cancer via regulation of the cell cycle, proliferation, apoptosis, migration, and invasion. AIM: In this study, we aimed to investigate the role of miR-367 in the invasion and metastasis of gastric cancer. METHODS: The correlation between the expression level of miR-367 and the clinicopathologic features of 37 patients with gastric cancer was analyzed by using real-time polymerase chain reaction (RT-PCR). In addition, we investigated the effect of miR-367 on the invasion and migration of the gastric cancer cell lines HS746T and SGC-7901 using transwell and scratch-wound assays, and the target gene of miR-367 was predicated and demonstrated by the bioinformatics method and luciferase reporter system, respectively. RESULTS: The results showed that the expression of miR-367 was significantly reduced in the gastric cancer tissues compared with the paraneoplastic tissues, and significantly correlated with the differentiation level, tumor-node-metastasis (TNM) stage, and metastasis of gastric cancer. Notably, the overexpression of miR-367 in gastric cancer cells inhibited the cellular migration and invasion. Furthermore, the luciferase reporter system demonstrated that Rab23 was a target gene of miR-367, and ectopic expression of Rab23 could reverse the invasion and migration inhibitory activity of miR-367. CONCLUSIONS: Our study shows that miR-367 is a key negative regulator of the invasion and metastasis of gastric cancer and establishes a strong rationale for developing miR-367 as a novel therapeutic agent against gastric cancer.

Matrix metalloproteinase-1 (MMP-1), an interstitial collagenase, is responsible for the proteolytic degradation of basement membrane and extracellular matrix. MMP-1 plays a major role in the invasion of gastric cancer (GC). The role of the genetic polymorphisms in the functional regions of MMP-1 on the risk of GC remains unclear. To identify the markers that contribute to the genetic susceptibility to GC, we examined the potential association between GC and nine single-nucleotide polymorphisms (rs 1799750, rs 498186, rs 475007, rs 514921, rs 494379, rs 996999, rs 2071232, rs 1938901, and rs 2239008) of the MMP-1 gene using the MassARRAY system in this study. The participants enrolled in this study included 422 patients with GC and 428 healthy subjects as the healthy controls from a Chinese Han population. The analysis revealed a weak association between the rs 1799750 (in the promoter region) genotype distribution and GC (p=0.020). The frequency of the 2G allele was significantly higher in the patients with GC than in the healthy controls (p=0.005, odds ratio [OR]=1.324, 95% confidence interval [CI]: 1.087-1.613). Moreover, the patients with the 2G/2G genotype of rs 1799750 had a significantly increased risk of cancer invasion compared with patients with the 1G/1G+1G/2G genotype (p=0.001, OR=0.505, 95% CI: 0.331-0.771). Strong linkage disequilibrium was observed in three blocks (D'>0.9). Significantly, more C-2G haplotypes (block 3) (p=0.0005 after Bonferroni correction) were found in GC subjects. These findings point to a role for MMP-1 promoter polymorphism in GC among a Han Chinese population, and may be informative for future genetic or biological studies on GC.

BACKGROUND: Hyperactivation of phosphatidylinositol 3-kinase/Akt signaling is commonly associated with human tumors including lung cancers. PH domain leucine-rich repeat protein phosphatase 1 (PHLPP1), which terminates Akt signaling by directly dephosphorylating and inactivating Akt, has been identified as a tumor suppressor. The protein level of PHLPP1 is regulated by E3 ligase beta-TRCP, however, the deubiquitinase for PHLPP1 is still not known. METHODS: The mRNA levels of USP1 and PHLPP1 in lung cancer cells and tissues were determined by real-time PCR. The half-life of PHLPP1 was detected by CHX assay. The interaction between USP1 and PHLPP1 was examined by immunoprecipitation and GST pull-down assay. RESULTS: Both USP1 and PHLPP1 are low expressed in lung cancer cells and tissues and silencing of USP1 by RNA interference significantly decreased the half-life of PHLPP1, which in turn amplified Akt1 phosphorylation. Our data identified a novel USP1-PHLPP1-Akt signaling axis, and decreased USP1 level in lung cancer cells may play an important role in lung cancer progress.

Objective To explore the differentially expressed genes of gastric cancer,matched normal gastric tissue and premalignant lesions. Methods The differentially expressed cDNA bands were isolated and identified by fluorescent differential display and then reamplified by PCR.After being cloned,all cDNA fragments were sequenced. Through BLAST,the results were compared with GenBank database for homology analysis. The expression of the cDNA fragment in different tissues was confirmed by Northern blot. SMART-RACE(rapid amplication of cDNA ends) was used to amplify the full length cDNA sequence. Bioinformatics analysis was performed to analysis its function. Results A differentially expressed cDNA fragment expressed lower in gastric cancers and premalignant lesions,no homology was found in GenBank. The novel cDNA fragment was given an accession number of EST(CD454989)in GenBank. A full length cDNA sequence of 1362 bp was acquired,who coded 267 amino acids,and was homologous with BAA91562.1,whose physiology function was unknown. Conclusion A differentially expressed gene was found and it may be involved in process of the gastric cancer.

RESUMO

5 male adults were studied in continuous bed rest for 16 days. The total body fat was measured by modified water displacement method (density method). The loss of body protein was measured by calculating nitrogen balance. The results have shown that during the experimental peroid their body protein decreased by 167 to 376 g and the body fat increased by 1.28?0.38kg. The average caloric requirement was 1958 ? 242 kcal/man/day, 32.3?1.6 kcal/kgBW/day or 36.1?1.4 kcal/kg LBW/day. These results are similar to those provided by other methods such as dietary survey or gas metabolism.
