RESUMO

The aim of this study was to improve the utilization of peanut vines as forage material for ruminants by investigating the degradation pattern of peanut vines in the dairy cow rumen. Samples of peanut vine incubated in cow rumens were collected at various time points. Bacterial diversity was investigated by scanning electron microscopy (SEM) and 16S rRNA gene sequencing. Carbohydrate-active enzymes (CAZymes) were analyzed by metagenomics. The peanut vines degraded rapidly from 2 to 24 h, before slowing from 24 to 72 h. SEM images confirmed dynamic peanut vine colonization. Firmicutes and Bacteroidetes were the two most dominant bacterial phyla throughout. Principal coordinates analysis indicated significant microbial composition changes at 6 and 24 h. This may be because, in the early stage, soluble carbohydrates that are easily degradable were degraded, while in the later stage, fibrous substances that are difficult to degrade were mainly degraded. Glycoside hydrolases (GHs) were the most abundant CAZymes, with peak relative abundance at 6 h (56.7 trans per million, TPM), and reducing at 24 (55.9 TPM) and 72 h (55.3 TPM). Spearman correlation analysis showed that Alistipes_sp._CAG:435, Alistipes_sp._CAG:514, Bacteroides_sp._CAG:1060, Bacteroides_sp._CAG:545, Bacteroides_sp._CAG:709, Bacteroides_sp._CAG:770, bacterium_F082, bacterium_F083, GH29, GH78, and GH92 were important for plant fiber degradation. These findings provide fundamental knowledge about forage degradation in the cow rumen, and will be important for the targeted improvement of ruminant plant biomass utilization efficiency.

The objective of this research was to investigate effects of different additives on the fermentation quality, aerobic stability and rumen degradation of mixed silage composed of amaranth and corn straw. The mixture ratio of amaranth to corn straw was 78%: 22%. Three additives were selected in this study and five groups were as follows: control group (CON, without additive), lactic acid bacteria group (LAB, 5 mg/kg, Lactobacillus plantarum ≥ 1.6×1010 CFU/g and L. buchneri ≥ 4.0×109 CFU/g), glucose group (GLU, 30 g/kg), cellulase group (CEL, 2 mg/kg) and lactic acid bacteria, glucose and cellulase group (LGC, added at the same levels as in individual group). The period of ensiling was 60 days. Fermentation quality, chemical composition and aerobic stability of mixed silage were analyzed. Four cows with permanent ruminal fistula were selected as experimental animals. Nylon bag technique was used to study rumen degradation characteristic of dry matter (DM), crude protein (CP), neutral detergent fiber (NDF) and acid detergent fiber (ADF) of mixed silage. Compared with CON group, the addition of different silage additives could improve mixed silage quality of amaranth and corn straw to some extent. Combining three additives significantly increased (P < 0.05) the DM, CP and lactic acid contents, whereas decreased (P < 0.05) the ADF and NDF contents as well as pH and ammonia nitrogen/total nitrogen. Moreover, the aerobic stability and rumen degradation of DM, CP and NDF were significantly improved (P < 0.05) in LGC group when compared to other groups. In conclusion, the combined addition of lactic acid bacteria, glucose and cellulase increased DM, CP and lactic acid contents as well as lactic acid bacteria count, decreased NDF and ADF contents and aerobic bacteria and mold counts, improved aerobic stability and rumen degradation of amaranth and corn straw mixed silage.

This research was conducted to compare the effects of gamma ray (GR) at doses of 100 and 150 kGy, alkaline treatments of sodium hydroxide (NaOH) and calcium oxide (CaO) at concentrations of 50 and 160 g/kg of dry matter (DM) respectively, and their combined use on chemical composition, ruminal degradation kinetics and crystallinity degree of soybean straw (SBS). After processing, the chemical composition of the samples was determined using the AOAC standard methods. Ruminal degradability trial was carried out by the nylon bag technique. Crystallinity degree of the samples was investigated through X-ray diffraction (XRD) technique. Using alkaline compounds in processing of SBS significantly increased the Ash amount (P < 0.0001). The greatest value was observed in CaO and CaO + GR treatments (100 and 150 kGy). The crude fiber (CF) of samples was reduced due to the effect of gamma irradiation, alkaline compounds, and their combination (P < 0.0001). Samples irradiated with 150 kGy dose of GR had the least amount of CF. The processing increased the effective rumen degradability (ERD) of DM for SBS at rumen outflow rates of 0.02, 0.05, and 0.08/h (P < 0.0001). The greatest amounts at the mentioned outflow rates were observed in GR (100, and 150 kGy), and NaOH + GR (100 and 150 kGy) treatments. Different treatments reduced the crystallinity degree of SBS compared to the control group. The least crystallinity percentage was observed in GR (150 kGy) treated samples. Totally, gamma irradiation, alkaline treatments and their combined use improved the nutritional value of SBS. In this respect, gamma irradiation was more effective than alkaline compounds.

The objective of this study was to evaluate the association of pseudostem hay of banana trees of different roughages in the diet of F1 Holstein/Zebu cows on their nutrient intake and digestibility, nitrogen balance, microbial synthesis, ingestive behavior, and milk production. The experiment was distributed in two simultaneous 4 × 4 Latin squares. The treatments were as follows: diet 1-sorghum silage as exclusive dietary roughage; diet 2-sorghum silage (70%) associated with 30% pseudostem hay of banana trees (dry matter basis); diet 3-70% elephant grass with 30% pseudostem hay of banana trees (dry matter basis); and diet 4-70% sugarcane with 30% banana pseudostem hay (dry matter basis). The roughage/concentrate ratio was 75:25. The animals fed with sorghum silage presented 14.45% higher intake (p = 0.01) to that of animals fed hay and elephant grass. The diets containing silage and silage with hay showed digestibility of neutral detergent fiber corrected for ash and protein that was 57.75% higher (p < 0.01) than that of other diets. Cows fed elephant grass spent more time intake (p = 0.01). Diets did not influence milk yield (p = 0.47), and the milk yield corrected for 3.5 fat (p = 0.22), final weight (p = 0.06), and intake efficiency (p = 0.57), presenting a mean of 14.16 kg/day, 15.94 kg/day, 590 kg, and 0.9 kg of milk/kg of ingested dry matter, respectively. The utilization of pseudostem hay associated with other roughage can be an alternative to milk yield in F1 Holstein/Zebu cows.

Abstract Background: Dry matter (DM) loss is high in sugarcane silage due to its high content of soluble carbohydrates which favors yeast growth and ethanol production, thus reducing the nutritional value of the preserved forage. Objective: To determine the effect of adding two types of urea at two concentration levels to sugarcane silage on the nutritional value and fermentative characteristics of the silage. Methods: A completely randomized 2×2+1 factorial design was used, based on two types of urea (conventional and protected), two urea concentration (0.5 and 1.0% of the natural matter), and a control (sugarcane silage with no additives). Six replicates were used. The mixture was placed in PVC silos and opened after 60 days. Results: There was no difference in DM content or effluent losses by the addition of urea. In regards to neutral detergent fiber, acid detergent fiber, and lignin, decreased levels were observed compared to the control, regardless of urea type. Urea type and concentration had an effect on ammoniacal nitrogen content, with higher values for conventional urea, but not affecting the pH. The addition of urea to sugarcane silage reduced gas losses and raised the levels of crude protein, ether extract, and effective degradability. Conclusion: Adding protected or conventional urea at 1% of natural matter at the time of sugarcane ensiling reduces gas losses and improves nutritional composition of the silage.


Resumen Antecedentes: La caña de azúcar ensilada presenta una elevada pérdida de materia seca (MS) debido a su alto contenido de carbohidratos solubles que favorece el crecimiento de levaduras y la producción de etanol, reduciendo así el valor nutricional del ensilado. Objetivo: Determinar el efecto de dos tipos de urea en dos concentraciones sobre las características fermentativas y el valor nutricional del ensilaje de caña de azúcar. Métodos: Se utilizó un diseño completamente al azar, en esquema factorial 2x2+1, basado en dos tipos de urea (común o protegida), dos concentraciones de urea (0,5 y 1,0% de la materia natural) y un control (ensilaje de caña de azúcar sin aditivos). Se utilizaron seis réplicas de cada tratamiento. El material fue ensilado en silos de PVC, los que fueron abiertos después de 60 días. Resultados: No hubo diferencia en el contenido de MS o en las pérdidas por efluentes con la adición de urea. En relación a la fibra detergente neutra, fibra detergente ácida y lignina, se observó disminución de éstos con respecto al control, independientemente del tipo de urea. Se observó un efecto de la concentración y del tipo de urea para el contenido de nitrógeno amoniacal, con mayores valores para la urea común, pero sin efecto sobre el pH. La adición de urea en el ensilado de caña redujo las pérdidas por gases y elevó los niveles de proteína bruta, extracto etéreo y degradabilidad efectiva. Conclusión: La adición de urea común o protegida, al 1% de materia natural, al inicio del ensilaje de caña de azúcar reduce las pérdidas por gases y mejora la composición nutricional del ensilaje.


Resumo Antecedentes: A ensilagem de cana-de-açúcar sem aditivos apresenta elevada perda de matéria seca (MS) devido ao alto teor de carboidratos solúveis, favorecendo o crescimento de leveduras e produção de etanol, reduzindo assim o valor nutricional da forragem conservada. Objetivo: Determinar o efeito do tipo de ureia (comun ou protegida) e diferentes concentrações nas características fermentativas e valor nutritivo das silagens de cana-de-açúcar. Métodos: Utilizou-se um delineamento inteiramente casualizado em esquema fatorial 2 x 2 + 1, sendo dois tipos de ureia (comun ou protegida), e duas concentrações (0,5 e 1,0% com base na matéria natural) mais o controle (silagem exclusiva de cana-de-açúcar); com seis repetições. O material foi ensilado em silos de PVC e abertos após 60 dias. Resultados: Não houve diferença no teor de MS e nas perdas por efluentes com adição da ureia. Em relação à fibra em detergente neutro, fibra em detergente ácido e lignina observou-se diminuição destes em relação ao controle, independente do tipo de ureia. Observou-se efeito das concentrações e do tipo de ureia para o teor de nitrogênio amoniacal, com maiores valores para a ureia comum, porém sem efeito no pH. A adição de ureia na ensilagem de cana reduziram as perdas por gases e elevaram os teores de proteína bruta, extrato etéreo e degradabilidade efetiva. Conclusão: O uso de 1% de ureia, independente do tipo, promove um incremento no valor nutricional da silagem de cana-de-açúcar e reduz as perdas por gases.

This study aimed at investigating the effects of decreasing crude protein (CP) in diets with constant rumen undegradable protein (RUP) content on dry matter (DM) intake, growth, feed efficiency (FE) and blood parameters in calves in a randomized complete block design. Dietary treatments included: (a) a calf starter containing 200 g/kg CP (62 g/kg RUP, based on DM), (b) a starter containing 180 g/kg CP (65 g/kg RUP, based on DM) and (c) a starter containing 160 g/kg CP (65 g/kg RUP, based on DM). A total 42 newborn male and female Holstein calves were fed 8 L milk/day until day 45, after which they were weaned and continued the experiment until day 75. Solid feed intake and total DM intake were measured daily, and body weight and skeletal growth parameters including withers height and heart girth were recorded weekly. Blood samples were collected on days 45 and 75. Solid feed intake, total DM intake, weaning weight, average daily gain from birth to weaning and from birth to 75 days, final weight and FE were not affected by the experimental treatments. In addition, skeletal growth parameters were similar among groups. Glucose concentration was similar among treatments; however, calves fed the starter containing 160 g/kg CP had significantly lower plasma concentrations of albumin and urea nitrogen compared with those fed starters containing 180 and 200 g/kg CP. These results indicate that CP level in the calf starter could be decreased to 160 g/kg DM if RUP levels stay at 65 g/kg DM, without negatively affecting feed intake and calf performance.

OBJECTIVE: This study was conducted to evaluate changes in nutritional value and in situ dry matter (DM) degradability of oak and pine wood before and after steam-digestion process (60 min/160°C/6 atm) and feeding effect of the oak roughage on performance and behavior of Hanwoo steers. METHODS: Chemical composition and tannin concentration were analyzed for oak and pine trees before and after the pretreatment. In situ DM and effective degradability of these samples were assessed using a nylon bag method. In vivo trial was performed to estimate animal performance and behavior, using steers fed total mixed ration (TMR) diets containing 0% (control), 25% (OR-25), and 50% (OR-50) of the oak roughage. Eighteen steers were allocated into nine pens (2 steers/pen, 3 pens/treatment) for 52 days according to body weight (BW) and age. RESULTS: By the steam-digestion treatment, the neutral detergent-insoluble fiber was decreased from 86.5% to 71.5% for oak and from 92.4% to 80.5% for pine, thereby increasing non-fiber carbohydrate. In situ DM degradability of treated oak reached 38% at 72 h, whereas that of untreated oak was only 11.9%. The 0 h degradability of the treated pine increased from 5.9% to 12.1%, but the degradability was unchanged thereafter. Animal performance including BW, average daily gain, DM intake, and feed conversion ratio was not different among control and oak treatments. No differences were detected in animal behavior such as lying, standing, rumination, drinking, and eating, except walking. Walking was higher in control than oak treatments with numerically higher eating and lower lying times, probably due to bulkier characteristics of rice straw in the diet. CONCLUSION: This study demonstrates that the oak roughage can be substituted for 50% of total forage or 100% of rice straw in TMR diets at early fattening stage of Hanwoo steers.

Fifteen oilseed cakes from sunflower, pomegranate, cardoon, tobacco and hemp were characterized with regard to chemical composition, Cornell Net Carbohydrate and Protein System (CNCPS) fractionation, in vitro digestibility of dry matter, neutral detergent fiber, and crude protein. All the cakes presented low moisture, rather variable ether extract contents and medium to high levels of crude protein and neutral detergent fiber. The cakes significantly differed in terms of CNCPS partitioning and in vitro digestibility. Tobacco and hemp cakes presented high contents of slow degradable fractions of crude protein and carbohydrate joined to good post-ruminal protein digestibility. Cardoon cakes presented the highest rumen protein degradability. Based on crude protein content and intestinal digestibility of rumen undegraded protein, cakes of tobacco and hemp showed the better potential as alternative protein supplements for ruminants, while pomegranate appears to be the least suitable for ruminant feeding.

The utilization of animal donors of rumen fluid for laboratory experiments can raise ethical concerns, and alternatives to the collection of rumen fluids from live animals are urgently requested. The aim of this study was to compare the fresh rumen fluid (collected at slaughter, W) with that obtained from a continuous fermenter (RCF) and three methods of rumen fluid preservation (refrigeration, R, chilling, C, and freeze-drying, FD). The fermentability of different inoculum was evaluated by three in vitro tests (neutral detergent fiber (NDF) and crude protein (CP) degradability and gas production, NDFd, RDP and GP, respectively) using six feeds as substrates. Despite the two types of inoculum differed in terms of metabolites and microbiota concentration, the differences in vitro fermentability between the two liquids were less pronounced than expected (-15 and 20% for NDFd and GP when the liquid of fermenter was used and no differences for RDP). Within each in vitro test, the data obtained from rumen and from fermenter liquids were highly correlated for the six feeds, as well as between W and R (r: 0.837-0.985; p < 0.01). The low fermentative capacity was found for C and, particularly, FD for liquids. RCF could be used to generate inoculum for in vitro purposes and short-term refrigeration is a valuable practice to manage inoculum.

The objective of this study was to analyze the chemical composition, in vitro ruminal fermentation, and intestinal digestibility of discarded samples of four Brassica vegetables: Brussels sprouts (BS), white cabbage, Savoy cabbage, and red cabbage, and to assess the effects of including increasing amounts of BS in the concentrate of a dairy sheep diet on in vitro fermentation, CH4 production, and in situ degradation of the diets. All cabbages had low dry matter content (DM; <16.5%), but their DM had high crude protein (19.5-24.8%) and sugars (27.2-41.4%) content and low neutral detergent fiber (17.5-28%) and was rapidly and extensively fermented in the rumen. Rumen degradability of protein at 12 h of in situ incubation was greater than 91.5% for all cabbages, and in vitro intestinal digestibility of protein ranged from 61.4 to 90.2%. Replacing barley, corn, and soybean meal by 24% of dried BS in the concentrate of a diet for dairy sheep (40:60 alfalfa hay:concentrate) increased in vitro diet fermentation and in situ degradability of DM and protein, and reduced in vitro CH4/total volatile fatty acid ratio. In vivo trials are necessary to confirm these results.

The objective of this study was to determine the variability in nutritive value for ruminants of tomato pomace (TP) samples and analyze its effect on in vitro fermentation when it was included in a high-concentrate diet. Twelve TP samples were obtained from two processing plants at weekly intervals and analyzed for chemical composition, in vitro rumen fermentation, and intestinal digestibility. The chemical composition of TP did not differ between processing plants and only slight variations were observed among sampling times. Tomato pomace had a low dry matter content (<300 g/kg), a high content of neutral detergent fiber, crude protein, and ether extract (572, 160, and 82.7 g/kg dry matter on average, respectively), and was rapidly fermented in the rumen. Protein degradability at 16 h in situ incubation was 510 g/kg and in vitro intestinal digestibility of protein was low (430-475 g/kg). Replacing soybean meal and barley straw by dried TP increased the in vitro fermentation rate and the production of volatile fatty acids and reduced NH3-N concentrations without affecting CH4. In summary, TP samples showed little variability in nutritive value over sampling time and TP of up to 180 g/kg could be included in high-concentrate diets without negatively affecting rumen fermentation.

Chemical composition and rumen degradability of waste vinegar residue (WVR) as roughage feed used for mutton sheep were evaluated in this work. Compared with the unfermented WVR, the WVR fermented by N. sitophila had more (P < 0.01) ash, crude protein (CP), and true protein (TP), less (P < 0.01) ether extract (EE), and significantly more carotenoid by about 27 times. But the contents of dry matter (DM), crude fiber (CF), neutral detergent fiber (NDF), and acid detergent fiber (ADF) had no obvious differences (P > 0.05) between unfermented and fermented WVR. The results suggested that the nutritional value of fermented WVR was higher for mutton sheep as roughage feed than that of unfermented WVR. The effective degradability (ED) of DM was higher (P < 0.05) in sheep with fermented WVR-based diet. The ED of CP and NDF of fermented WVR was reduced (P < 0.01) compared with the unfermented WVR. The results further suggested that the fermentation improved the degradability of WVR, and the rumen degradability of protein by ruminal flora decreased in fermented WVR, saving more protein for the sheep post-ruminal digestion and absorption. Furthermore, the results presented here clearly indicated the potential of fermented WVR by N. sitophila as an unconventional and functional feedstuff with rich carotenoid for ruminants, which could reduce WVR discharge in vinegar brewing industry and improve ruminant production. This work laid a foundation for the development of ruminant carotenoid functional feed.

Although the conventional in situ ruminal degradability method is a relevant tool to describe the nutritional value of ruminant feeds, its need for rumen-fistulated animals may impose a restriction on its use when considering animal welfare issues and cost. The aim of the present work was to develop a ruminal degradability technique which avoids using surgically prepared animals. The concept was to orally dose a series of porous bags containing the test feeds at different times before slaughter, when the bags would be removed from the rumen for degradation measurement. Bags, smaller than those used in the conventional nylon bag technique, were made from woven nylon fabric, following two shape designs (rectangular flat shape, tetrahedral shape) and were fitted with one of three types of device for preventing their regurgitation. These bags were used in two experiments with individually housed non-pregnant, non-lactating sheep, as host animals for the in situ ruminal incubation of forage substrates. The bags were closed at the top edge by machine stitching and wrapped in tissue paper before oral dosing. Standard times for ruminal incubation of substrates in all of the tests were 4, 8, 16, 24, 48, 72 and 96 h before slaughter. The purpose of the first experiment was to compare the effectiveness of the three anti-regurgitation device designs, constructed from nylon cable ties ('Z-shaped', ARD1; 'double Z-shaped', ARD2; 'umbrella-shaped', ARD3), and to observe whether viable degradation curves could be generated using grass hay as the substrate. In the second experiment, three other substrates (perennial ryegrass, red clover and barley straw) were compared using flat and tetrahedral bags fitted with type ARD1 anti-regurgitation devices. Non-linear mixed-effect regression models were used to fit asymptotic exponential curves of the percentage dry matter loss of the four substrates against time of incubation in the reticulorumen, and the effect of type of anti-regurgitation device and the shape of nylon bag. All three devices were highly successful at preventing regurgitation with 93% to 100% of dosed bags being recovered in the reticulorumen at slaughter. Ruminal degradation data obtained for tested forages were in accordance with those expected from the conventional degradability technique using fistulated animals, with no significant differences in the asymptotic values of degradation curves between bag shape or anti-regurgitation device. The results of this research demonstrate the potential for using a small bag technique with intact sheep to characterise the in situ ruminal degradability of roughages.

The crude protein, total digestible nutrients and extent of rumen degradability of dry matter (DM) of VW of households were 140.0 g kg-1, 0.668 and 0.855, respectively; while those of the marketplace were 169.0 g kg-1, 0.633 and 0.80, respectively. The levels of chromium and lead in each respectively, was 13.27 and 1.53 ng kg-1DM; and 31.01 and 5.71 ng kg-1DM. The total aflatoxins in VW of households was 3.08 µg kg-1DM, and undetectable in VW from the marketplace. Considering the chemical composition and safety parameters studied, VW could preliminary be considered as animal feed. The feeding of processed marketplace VW (VWP) at 275 g kg-1DM of a diet or 0.76% of live weight (LW) to growing bulls, replacing 50% of a concentrate mixture as supplement to a Napier silage diet for a period of 34 days reduced the total DM intake (0.0276 vs 0.0343 LW) without any significant (P > 0.05) changes in DM or protein digestibility. Blood urea levels (19.5 vs 23.67 mg dl-1), and serum creatinine levels (1.37 vs 1.08 mg dl-1) differed significantly (P > 0.05) between the two groups but were within normal physiological ranges. Therefore, it may be concluded that the level of incorporation of VWP would be less than 50% replacement of the concentrate in the diet. Further research is required to determine optimum inclusion levels in ruminant diets.

BACKGROUND: The present work investigated the influence of lignin content and composition in the fungal treatment of lignocellulosic biomass in order to improve rumen degradability. Wheat straw and wood chips, differing in lignin composition, were treated with Lentinula edodes for 0, 2, 4, 8 and 12 wk and the changes occurring during fungal degradation were analyzed using pyrolysis-gas chromatography-mass spectrometry and detergent fiber analysis. RESULTS: L. edodes preferentially degraded lignin, with only limited cellulose degradation, in wheat straw and wood chips, leaving a substrate enriched in cellulose. Syringyl (S)-lignin units were preferentially degraded than guaiacyl (G)-lignin units, resulting in a decreased S/G ratio. A decreasing S/G ratio (wheat straw: r = -0.72, wood chips: r = -0.75) and selective lignin degradation (wheat straw: r = -0.69, wood chips: r = -0.88) were correlated with in vitro gas production (IVGP), a good indicator for rumen degradability. CONCLUSIONS: L. edodes treatment increased the IVGP of wheat straw and wood chips. Effects on IVGP were similar for wheat straw and wood chips indicating that lignin content and 3D-structure of cell walls influence in vitro rumen degradability more than lignin composition.

BACKGROUND: The aim of this study was to optimize the fungal treatment of lignocellulosic biomass by stimulating the colonization. Wheat straw and wood chips were treated with Ceriporiopsis subvermispora and Lentinula edodes with various amounts of colonized millet grains (0.5, 1.5 or 3.0 % per g of wet weight of substrate) added to the substrates. Also, wheat straw and wood chips were chopped to either 0.5 or 2 cm. Effectiveness of the fungal treatment after 0, 2, 4, 6, or 8 wk of incubation was determined by changes in chemical composition, in vitro gas production (IVGP) as a measure for rumen degradability, and ergosterol content as a measure of fungal biomass. RESULTS: Incomplete colonization was observed for C. subvermispora treated wheat straw and L. edodes treated wood chips. The different particle sizes and amounts of inoculum tested, had no significant effects on the chemical composition and the IVGP of C. subvermispora treated wood chips. Particle size did influence L. edodes treatment of wheat straw. The L. edodes treatment of 2 cm wheat straw resulted in a more selective delignification and a higher IVGP than the smaller particles. Addition of 1.5 % or 3 % L. edodes inoculum to wheat straw resulted in more selective delignification and a higher IVGP than addition of 0.5 % inoculum. CONCLUSION: Particle size and amount of inoculum did not have an effect on C. subvermispora treatment of wood chips. At least 1.5 % L. edodes colonized millet grains should be added to 2 cm wheat straw to result in an increased IVGP and acid detergent lignin (ADL) degradation.

The objectives of this study were to determine an optimal cultivation time for populations of yeast and lactic acid bacteria (LAB) co-cultured in fermented milk and effects of soybean meal fermented milk (SBMFM) supplementation on rumen degradability in beef cattle using nylon bag technique. The study on an optimal cultivation time for yeast and LAB growth in fermented milk was determined at 0, 4, 8, 24, 48, 72, and 96 h post-cultivation. After fermenting for 4 days, an optimal cultivation time of yeast and LAB in fermented milk was selected and used for making the SBMFM product to study nylon bag technique. Two ruminal fistulated beef cattle (410±10 kg) were used to study on the effect of SBMFM supplementation (0%, 3%, and 5% of total concentrate substrate) on rumen degradability using in situ method at incubation times of 0, 2, 4, 6, 12, 24, 48, and 72 h according to a Completely randomized design. The results revealed that the highest yeast and LAB population culture in fermented milk was found at 72 h-post cultivation. From in situ study, the soluble fractions at time zero (a), potential degradability (a+b) and effective degradability of dry matter (EDDM) linearly (p<0.01) increased with the increasing supplemental levels and the highest was in the 5% SBMFM supplemented group. However, there was no effect of SBMFM supplement on insoluble degradability fractions (b) and rate of degradation (c). In conclusion, the optimal fermented time for fermented milk with yeast and LAB was at 72 h-post cultivation and supplementation of SBMFM at 5% of total concentrate substrate could improve rumen degradability of beef cattle. However, further research on effect of SBMFM on rumen ecology and production performance in meat and milk should be conducted using in vivo both digestion and feeding trials.

The supplementation of livestock feed with animal protein is a present cause for public concern, and plant protein shortages have become increasingly prominent in China. This conflict may be resolved by fully utilizing currently available sources of plant protein. We estimated the rumen degradability and the small intestinal digestibility of the amino acids (AA) in rapeseed meal (RSM), soybean meal (SBM), sunflower seed meal (SFM) and sesame meal (SSM) using the mobile nylon bag method to determine the absorbable AA content of these protein supplements as a guide towards dietary formulations for the dairy industry. Overall, this study aimed to utilize protein supplements effectively to guide dietary formulations to increase milk yield and save plant protein resources. To this end, we studied four cows with a permanent rumen fistula and duodenal T-shape fistula in a 4×4 Latin square experimental design. The results showed that the total small intestine absorbable amino acids and small intestine absorbable essential amino acids were higher in the SBM (26.34% and 13.11% dry matter [DM], respectively) than in the SFM (13.97% and 6.89% DM, respectively). The small intestine absorbable Lys contents of the SFM, SSM, RSM and SBM were 0.86%, 0.88%, 1.43%, and 2.12% (DM basis), respectively, and the absorbable Met contents of these meals were 0.28%, 1.03%, 0.52%, and 0.47% (DM basis), respectively. Among the examined food sources, the milk protein score of the SBM (0.181) was highest followed by those of the RSM (0.136), SSM (0.108) and SFM (0.106). The absorbable amino acid contents of the protein supplements accurately reflected protein availability, which is an important indicator of the balance of feed formulation. Therefore, a database detailing the absorbable AA should be established.

The in situ degradation of the washout fraction of starch in six feed ingredients (i.e. barley, faba beans, maize, oats, peas and wheat) was studied by using a modified in situ protocol and in vitro measurements. In comparison with the washing machine method, the modified protocol comprises a milder rinsing method to reduce particulate loss during rinsing. The modified method markedly reduced the average washout fraction of starch in these products from 0.333 to 0.042 g/g. Applying the modified rinsing method, the fractional degradation rate (k d ) of starch in barley, oats and wheat decreased from on average 0.327 to 0.144 h-1 whereas for faba beans, peas and maize no differences in k d were observed compared with the traditional washing machine rinsing. For barley, maize and wheat, the difference in non-fermented starch in the residue between both rinsing methods during the first 4 h of incubation increased, which indicates secondary particle loss. The average effective degradation of starch decreased from 0.761 to 0.572 g/g when using the new rinsing method and to 0.494 g/g when applying a correction for particulate matter loss during incubation. The in vitro k d of starch in the non-washout fraction did not differ from that in the total product. The calculated ratio between the k d of starch in the washout and non-washout fraction was on average 1.59 and varied between 0.96 for oats and 2.39 for maize. The fractional rate of gas production was significantly different between the total product and the non-washout fraction. For all products, except oats, this rate of gas production was larger for the total product compared with the non-washout fraction whereas for oats the opposite was observed. The rate of increase in gas production was, especially for grains, strongly correlated with the in vitro k d of starch. The results of the present study do not support the assumption used in several feed evaluation systems that the degradation of the washout fraction of starch in the rumen is much faster than that of the non-washout fraction.

This study explored the potential of partial least squares (PLS) and Fourier-transform infrared spectroscopy (FTIR) to predict rumen dry matter (DM) and neutral detergent fiber (NDF) degradation parameters of a wide range of feeds for ruminants, as an alternative to the in situ method. In total, 663 samples comprising 80 different feed types were analyzed. In situ DM and NDF degradabilities were determined as follows: effective degradability (ED), rumen soluble fraction (A), degradable but not soluble fraction (B), rate of degradation of the B fraction (C), and indigestible NDF (iNDF). Infrared spectra of dry samples were collected by attenuated total reflectance from 600 to 4000cm(-1). Feeds were randomly classified into 2 subsets of samples with representation of all feed types; one subset was used to develop regression models using partial least squares, and the second subset was used to conduct an external validation of the models. This study indicated that universal models containing all feed types and specific models containing concentrate feeds could provide only a relatively poor estimation of in situ DM degradation parameters because of compositional heterogeneity. More research, such as a particle size distribution analysis, is required to determine whether this lack of accuracy was due to limitations of the FTIR approach, or simply due to methodological error associated with the in situ method. This latter hypothesis may explain the low accuracy observed in the prediction of degradation rates if there was physical leakage of fine particles from the mesh bags used during in situ studies. In contrast, much better predictions were obtained when models were developed for forage feeds alone. Models for forages led to accurate predictions of DMA, DMB, NDFED, and NDF concentration (R(2)=0.91, 0.89, 0.85, and 0.79, standard error = 4.34, 5.97, 4.59, and 4.41% of DM, respectively), and could be used for screening of DMED, NDFC, and iNDF. These models relied on certain regions of the FTIR spectrum (900-1150 and 1500-1700cm(-1)), which are mainly compatible with absorption of plant cell wall components, such as cellulose, pectin, lignin, cutin, and suberin, but also with nonstructural carbohydrates and certain active compounds. In conclusion, FTIR spectroscopy could be considered a low-cost alternative to in situ measurements in feed evaluation.
