ABSTRACT

Objective To investigate the effect of electroacupuncture on diaphragm function of chronic obstructive pulmonary disease (COPD) rats with muscular dystrophy, and to explore the regulatory mechanism. Methods Forty male rats were randomly divided into blank group, model group, electroacupuncture group, exercise group, electroacupuncture plus exercise group, 8 rats in each group. After successful establishment of COPD rat model with muscular dystrophy, the modeled rats in various intervention groups were given electroacupuncture and/or exercise treatment. After the last treatment, the changes of rat body mass were observed, the rat lung function was detected, and the mRNA expression levels of myosin heavy chains (MHC) of MHC-1, MHC-2 and diaphragmatic related signal proteins of Atrogin-1, muscle ring-finger protein-1(MuRF-1), MyoD were detected by real-time quantitative polymerase chain reaction (qPCR). Results (1) Compared with the blank group, inspiratory resistance (IR) and functional residual mass (FRC) in the model group were increased (P < 0.05) , and the dynamic lung compliance(Cydn) was decreased(P<0.05). Compared with the model group, IR and FRC in the intervention groups were decreased (P < 0.05), but the differences among the three intervention groups were insignificant(P>0.05). (2) Compared with the blank group, the mRNA expression levels of MHC-1, Atrogin-1, MuRF-1, MyoD in the model group were increased (P<0.05), and the mRNA expression level of MHC-2 was decreased (P < 0.05). Compared with the model group, the mRNA expression levels of MHC-1, Atrogin-1, MuRF-1, MyoD in the intervention groups were decreased (P < 0.05) , and the mRNA expression level of MHC-2 was increased(P<0.05). Compared with the exercise group, the mRNA expression levels of Atrogin-1, MuRF-1, MyoD in the electroacupuncture group were decreased (P<0.05), and the mRNA expression level of MHC-2 was increased (P<0.05) , but the above indexes in electroacupuncture plus exercise group showed no obvious changes(P>0.05). Conclusion Electroacupuncture can improve respiratory function of COPD rats with muscular dystrophy, and the possible mechanism is related with the increase of MHC-2 mRNA expression and with the decrease of Atrogin-1, MuRF-1, MyoD mRNA expression, which result into the regulation of ubiquitin proteasome pathway(UPP), reduction of myosin loss, and the relief of diaphragmatic atrophy.

ABSTRACT

OBJECTIVE@#To investigate the expressions and time-dependent changes of phosphatidylinositol-3-kinase (PI3K), phospho-PI3K (p-PI3K), protein kinase B (PKB/Akt) and phospho-Akt (p-Akt) during wound healing process of mice skin.@*METHODS@#The changes of PI3K, p-PI3K, Akt and p-Akt expression in skin wound were detected by immunohistochemistry, Western blotting and real-time PCR.@*RESULTS@#Immunohistochemistry showed the expression of PI3K and p-Akt were observed in mononuclear and fibroblast after skin wound, and reached peak in reconstruction. The positive bands of PI3K, p-PI3K, Akt and p-Akt were observed in all time points of the wound healing process by Western blotting. The expression peak of p-PI3K and p-Akt showed in inflammation and proliferation; the expression peak of PI3K and Akt in reconstruction. Real-time PCR showed the expression peak of PI3K mRNA in inflammation and reconstruction and the peak of Akt mRNA in reconstruction.@*CONCLUSION@#During the wound healing process, the expressions of PI3K, Akt, p-PI3K and p-Akt show different changes with significant correlation to wound time. The expression of PI3K/Akt may be a valuable marker for wound time estimation.

ABSTRACT

Objective To design and fabricate novel mesoporous calcium silicate/calcium phosphate cement (MCS/CPC) scaffolds for bone repair and investigate their in vitro biomechanical properties under different external forces. Methods MCS and CPC in certain proportion were mixed to form plotting material, and the composite MCS/CPC scaffolds with pore size of 350 μm and 500 μm were fabricated by 3D bioplotting technique, respectively. Surface topographies of the scaffolds were observed by scanning electron microscope (SEM). The compressive strength and mechanical properties of the scaffolds under dynamic cyclic loads at different frequencies were studied through universal mechanical testing machine and dynamic mechanical analysis instrument. Results MCS/CPC scaffolds with controllable macroporous structures could be fabricated by 3D bioplotting technique. Scaffolds with pore size of 350 μm had higher compressive strength [(9.8±0.39) MPa] and compressive modulus [(132.5±4.3) MPa]. In addition, at the loading frequency of 1-100 Hz, scaffolds with pore size of 350 μm had a higher storage modulus. ConclusionsMCS/CPC scaffolds with pore size of 350 μm fabricated by 3D bioplotting technique possess not only regular pore connectivity and high compressive strength, but also structural stability under dynamic loads, which are promising as novel biomaterials for bone repair.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the effectiveness and safety of simple obesity treated by acupuncture.</p><p><b>METHODS</b>By randomized single-blind clinical trial, one hundred and eighteen cases of simple obesity were divided into an acupuncture group (76 cases) and a placebo-acupuncture control group (42 cases), additionally, health control group (30 cases) was included. In acupuncture group and placebo-acupuncture control group, all the patients received a restricted diet; Zhongwan (CV 12) and Zhongji (CV 3) etc. at abdomen and Liangqiu (ST 34) and Zusanli (ST 36) etc. at limbs were selected; body mass index (BMI), Serum Total Cholesterol, triglyceride (TG), Glucose, Creatinine, urea nitrogen (BUN), Uric Acid and adverse reactions scores were observed.</p><p><b>RESULTS</b>After treatment the BMI in acupuncture grown was lower than that in placebo-acupuncture control group (P < 0.01). In metabolism indices, the serum Total Cholesterol and Glucose after treatment were reduced obviously than those before treatment in acupuncture group (all P < 0.01), and there was no significant differences in other metabolism indices (all P > 0.05) in two groups. After treatment, in adverse reactions scores, the hunger sensation scores in acupuncture group was reduced than that in placebo-acupuncture control group (P < 0.05), and there was no significant differences in other indices (all P > 0.05).</p><p><b>CONCLUSION</b>BMI of simple obesity was reduced by acupuncture, and the Serum Total Cholesterol and Glucose were reduced accordingly. The adverse reac tions such as weakness, nervosa and diarrhea, etc. doesn't appear after acupuncture treatment. Acupuncture therapy is one of the safe and effective methods for simple obesity.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To observe the biocompatibility of new biomaterials porous calcium phosphate (CPC) and ectopic bone formation of CPC with bone marrow stromal cells (BMSCs).</p><p><b>METHODS</b>The BMSCs were cultured from Beagle dog and combined with the porous CPC with the best concentration after transfect green fluorescent protein (GFP). The adhesion and growth of BMSCs on CPC were observed under inversion, fluorescence and scanning electron microscopy. The ectopic bone formation were observed at the 8th week after CPC and BMSCs were implanted subcutaneously into nude mice.</p><p><b>RESULTS</b>When BMSCs with CPC were cultured at the 1st day, cells were climbing out from CPC with normal morphology. At the 7th day cells can be seen protruding pseudopods, secretion of matrix. Bone formation could be seen histomorphologically at the 8th week.</p><p><b>CONCLUSION</b>Porous CPC has good biocompatibility and is an ideal scaffold material for bone tissue engineering.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the possible effect of transforming growth factor-beta(1) (TGF -beta(1)) on the development of renal fibrosis in human mesengial proliferative glomerulonephritis (MsPGN).</p><p><b>METHODS</b>Immunohistochemistry method, sirius red staining polarization microscopy and the computer imaging analysis system were used to detect the expression of TGF-beta(1), the distribution of collagen I, collagen III and collagen IV.</p><p><b>RESULT</b>In MsPGN with renal fibrosis, collagen IV was increased markedly,and collagen I and collagen III appeared in the expanded mesengial matrix abnormally. Collagen III and collagen IV were increased markedly in tubulointerstitium. TGF-beta(1) expression was positively correlated with the expression of collagen I, collagen III and collagen IV in tubulointerstitium (r=0.82 0.92,P<0.01), and negatively correlated with I/III, I/IV and III/IV (r=-0.83,-0.92, P<0.001).</p><p><b>CONCLUSION</b>Abnormal increase of TGF-beta(1) may be one of the important factors associated with glomerular sclerosis and tubulointerstitial fibrosis through the increment and abnormal distribution of collagen I, collagen III and collagen IV.</p>

ABSTRACT

Objective To study the localization of the foramen of Monro on magnetic resonance image (MRI) and its clinical significance.Methods Cranial MRI was observed for 30 normal healthy subjects to study their location,shape and size of the foramen of Monro,as compared to those in 22 patients with hydrocephalus and 14 cases with tumor around their foramen.Results The the foramen of Monro is located between the fornix and the anterior part of thalamus in the normal healthy subjects,with a transverse diameter of 2.8 mm and a vertical diameter of 2.1 mm in average on MRI.Y-shaped structure is formed between their bilateral the foramen of Monro and the 3rd cerebral ventricle.Very obvious changes in shape and size of the foramen of Monro can be observed in patients with hydrocephalus,which sometimes can integrated with the 3rd cerebral ventricle and lateral ventricle,forming a"rabbit-head sign".Meanwhile, imaging of the foramen of Monro varied in patients with tumor around the foramen,depending on its location and size.Conclusions The foramen of Monro has a relative constant position in the brain.Changes in its position,size and shape are important signs suggesting intracranial lesions.An individualized operation plan for a patient should be determined based on those changes.

ABSTRACT

<p><b>OBJECTIVE</b>To synthesize nanometer hydroxyapatite (HAP) by sol-gel method from Ca (NO3)2.4H2O and PO (CH3O)3 with water and ethanol as solvents and to study the factors that can affect the formation of sol-gel, including the type of solvents, amount of solvents, pH value, as well as properties under the condition of low temperature combustion of dried gel.</p><p><b>METHODS</b>The viscosities of sol were determined on WU viscosimeter. TG-DTA was used to analyze the thermal procedure of dried gel by increasing temperature. Characterization of the dried gel and powder after combustion were evaluated by X-ray diffraction and infrared spectrum. The morphology of the powder was observed by transmission electron microscopy.</p><p><b>RESULTS</b>Stable and homogeneous gel could be obtained at appropriate pH value when ethylene glycol was added into solvent. The dried gel had the feature of low temperature combustion. HAP powder with particle size of 50 nm was directly prepared.</p><p><b>CONCLUSION</b>Nanometer HAP powder with uniform particle size can be synthesized by sol-gel method.</p>

ABSTRACT

Objective To investigate the optimal scanning parameters for diffusion tensor imaging of the cervical cord.Methods MRI and diffusion tensor imaging of the cervical cord was performed in 80 healthy adult volunteers.Different parameters including b values,the number of the diffusion sensitive gradient directions,the number of excitations,and slice thickness were applied and their effects on the quality of the images were compared.DTI was performed on the cervical spinal cord with different b-values (400,700,and 1000 s/mm~2)in group 1,with different numbers of diffusion gradient directions(6,13, and 25)in group 2,with different numbers of excitations(2,4,and 8)in group 3,and with different slice thicknesses(2,3,and 4 mm)in group 4.Two radiology experts gave a score to every image with double blind methods,then compared the image quality.Results In the comparison of the four different parameters,DTIs using a b value of 700 s/mm~2(2.25?0.58)showed better image quality than those with the b values of 400 s/mm~2(1.86?0.53)and 1000 s/mm~2(1.48?0.35)(P
