ABSTRACT

Aims Tocomparethepharmacokineticsof ephedra alkaloids,cinnamic acid and cinnamic alcohol in Ephedrae, Cinnamomi and Ephedrae-Cinnamomi herb couple through UPLC-MS/MS in rats respective-ly,and to investigate the effect of combination on phys-iologicaldisposition.Methods Plasmasampleswere collected at different times after oral administration of Ephedrae,Cinnamomi and Ephedrae-Cinnamomi herb couple extracts.The concentrations of ephedra alka-loids,cinnamic acid and cinnamic alcohol in plasma samples were determined by UPLC-MS/MS.DAS 3. 0 was used to calculate pharmacokinetic parameters.The differences of samples in two groups were conducted with univariate statistical analysis using SPSS 13. 0. Results ComparedwithEphedraegroup,theCmaxof norephedrine hydrochloride, norpseudoephedrine hydrochloride,ephedrine hydrochloride,pseudoephed-rine hydrochloride,and methylephedrine hydrochloride in Ephedrae-Cinnamomi herb couple group were signif-icantly greater (P<0. 05 );the AUC0-t of norpseudo-ephedrine hydrochloride was significantly greater (P<0. 05 );the MRT0-t of norephedrine hydrochloride, phedrine hydrochloride, pseudoephedrine hydrochlo-ride,and methylephedrine hydrochloride were signifi-cantly less (P <0. 05 );the T1/2z of norephedrine hydrochloride,phedrine hydrochloride,and methyl-ephedrine hydrochloride were significantly less (P <0. 05 ).The AUC0-t and MRT0-t of cinnamic acid and cinnamic alcohol were significantly greater than those inCinnamomigroup(P<0.05).Conclusion The combination of Ephedrae and Cinnamomi improves the absorption concentration of five ephedra alkaloids, Slows down the elimination of norephedrine hydrochlo-ride,phedrine hydrochloride,pseudoephedrine hydro-chloride,and methylephedrine hydrochloride,and in-creases the bioavailability of cinnamic acid and cin-namic alcohol.

ABSTRACT

Objective To investigate the anti-rotavirus effect of Gegen Qinlian decoction on rotavirus (RV)-inducing diarrhea neonatal mice model, and provide evidences for pharmacological and pharmacodynamic studies. Methods Neonatal diarrhea mice model of Kunming family infected with RV-Wa was developed. Symptom, diarrhea ratio and severity level, histological changes of neonatal mice’s small intestine and lung was assessed after Gegen Qinlian decoction (0.78 g/kg) treatment. Results Diarrhea ratio, diarrhea severity level, histological changes of neonatal mice’s small intestine and lung were obviously decreased after Gegen Qinlian decoction (0.78 g/kg) treatment. Conclusion Gegen Qinlian decoction has anti-rotavirus effect on RV induced diarrhea.

ABSTRACT

Objective To discuss the influence of drug compatibility on the components of the volatile oil in Flos Lonicerae and Fructus Forsythiae. Methods GC-MS system was used to analyze the volatile oil in Flos Lonicerae, Fructas Forsythiae and their combination. The effect of compatibility on the components and content of volatile oil was evaluated. Results The results of GC-MS showed that gerani]o, tetradecanoic acid methyl ester, hexadecanoic acid ethyl ester, fluoranthene, 9, 12, 15-Octadeeatrienoic acid ethyl ester and linoleic acid was detectable in Flos Lonicerae but was undetectable in the drug pair; camphene, alpha-terpinene, perillaalcohol, 2-beta-pinene, beta-hexadecanal, phellandrene was de-tectable in Fructus Forsythiae but was undetectable in the drug pair; limonene, isolongifolene, nonadecane was detectable in the drug pair but was undetectable in the two single drugs. Conclusion The compatibility of the drug pair has an effect on components and contents in the volatile oil of Flos Lonicerae and Fructus Forsythiae.

ABSTRACT

Objective To establish a method to determinate polysaccharides in Hedyotic diffusa, then compare the polysaccharides content in Hedyotic diffusa form Jiangxi, Guangdong, Guangxi. Methods Sulfuric acid -phenol coloration were applied to analysis polysaccharides content. The detective wave length was 486 nm. Results Polysaccharides in Hedyotic diffusa from Jiangxi province was a little higher than the others. Conclusion Polysaccharides content in Hedyotic diffusa form different habitat are almost alike.

ABSTRACT

OBJECTIVE:To establish phenol-sulfuric acid method for the determination of the content of total polysaccharides in Weile granules.METHODS:The samples were deproteinized by Sevag method,colored by phenol-sulfuric acid method(water bath at 60 ℃ for 30 min),and the content of total polysaccharides in Weile granules was determined with Glucose as control.RESULTS:The linear range of Glucose was 20.47~102.36 ?g?mL-1(r=0.999 4),with an average recovery of 99.09%(RSD=0.92%,n=6).The content of total polysaccharides in Weile granules was 41.63%.CONCLUSION:This method was simple,accurate and reproducible,and suitable for quality control of Weile granules.

OBJECTIVE:To provide scientific evidence for the microsphere's m etabolism in vivo of lung targeted tetran?drine polylactic acid(TET-PLA)microspheres.METHODS:Blood was sampled from carotid artery after microspheres were injected into rabbit through auricular vein,the concentration of TET in plasma was determined by RP-HPLC method;The pharmacokinetic parameters were obtained by using3p87program.RESULTS:Concentration-time curves of TET micro?spheres were fitted to a2-compartment model with t 1/2? of(286.49?237.55)min and AUC of(810.33?287.49)(?g?min)/ml.CONCLUSION:TET-PLA microspheres show sustained release effects in rabbits.

ABSTRACT

Objective To study the influence of Mahuang Decoction(MD)in different combinations on diaphoretic function in rats.Methods Vacuole incidence of axillary sweat gland was determined by observing the histolomorphological changes of vacuole to evaluate the diaphoretic function.The diaphoretic function was observed in 30 minutes after administration.Results The combination of Mahuang(Herba Ephedrae)and Guizhi(Ramulus Cinnamomi)had the strongest diaphoretic function in all of the different combinations.The diaphoretic function of the combinations including Mahuang(Herba Ephedrae)was stronger than those excluding Mahuang(Herba Ephedrae).The diaphoretic function of Mahuang(Herba Ephedrae)became stronger when used together with Guizhi(Ramulus Cinnamomi),remained the same when used together with Xingren(Semen Armeniacea Amarum),and became weaker when used together with Gancao(Radix Glycyrrhizac).Conclusion Mahuang(Herba Ephedrae),Guizhi(Ramulus Cinnamomi),Xingren(Semen Armeniacea Amarum),and Gancao(Radix Glycyrrhizac)play the roles of principle drug,assistant drug,adjuvant drug and conductantr drug respectively in Mahuang Decoction,which embodying the method of composing a prescription.

ABSTRACT

Objective To develop a gas chromatography-mass spectrometry (GC-MS) method for the determination of ephedrine (E) and pseudoephdrine (PE) and to study the pharmacokinetics features of E and PE in healthy human volunteers. Methods The serum concentrations of E and PE were measured by GC-MS. The pharmacokinetic parameters were computed by using WinNonlin program. Results A good linearity was obtained in E from 5 ng/mL to 1000 ng/mL (r2=0.9985) and in PE from 2.5 ng/mL to 500 ng/mL (r2=0.9994). The recoveries were in the range of 107.5 %to 96.2 %and 102.5 %to 93.9 %for E and PE respectively. The interday RSD and intraday RSD of E and PE were both less than 5 %. The E and PE plasma solution were steady within 60 days. Conclusion This GC-MS method is a accurate and reliable method for the determination of E and PE in human plasma.

ABSTRACT

Objective To establish the HPLC fingerprints of Radix Glycyrrhizae. Methods The RP-HPLC method was used, Zorbax SB C_ 18 column (250 mm?4.6 mm, 5 ?m) was employed; the acetonitrile-1.2% acetic acid (gradient elution) was used as mobile phase, analytic time was 70 min, and detective wavelength was at 254 nm. Results The HPLC fingerprints of Radix Glycyrrhizae were set up. The result showed that 38 peaks were common in different sources. The results of method validation met technical standard of fingerprints. Conclusion The method is stable and reliable with a good reproducibility and provides a reference standard for the quality control of Radix Glycyrrhizae.

ABSTRACT

Objective To develop a GC-MS method for the determination of ephedrine in human plasma and study the pharmacokinetic parameters of the ephedrine in different compatibility of the minister, adjuvant, and guide herbs to the monarch herb in Mahuang Decoction. Methods Orthogonal design was used to determine eight compatible groups from Mahuang Decoction (MHD). The healthy male volunteers were divided into groups in random, each group concluding eight men. After taking the medicine orally, venous blood would be taken out at different times. Determining ephedrine in plasma and plotting the concentration-time curve, the pharmacokinetic parameters of each compatible group were calculated by WinNonlin 4.0.1. The statistical analysis of the pharmacokinetic parameters were proceeded by SPSS 10.0. ABSTRACT

Object To explore the regularity of recipe composition by observing different bacteriostasis among compositions of Gegen Qinlian Decoction (GQD) against Streptococcus pneumoniae. MethodsTo evaluate the minimal inhibitory concentration (MIC) of 15 decoctions through double fold dilution method. Results Not all of the decoctions are of the same MIC. Conclusion Not all of the 15 decoctions have the same bacteriostasis against S. pneumoniae. Among the four herbs of GQD Coptis chinensis Franch. has the strongest bacteriostasis and the next Scutellaria baicalensis Georgi. While Glycyrrhina wralensis Fisch and Pueraria lobata (Willd.) Ohwi have no bacteriostasis, but they can influence the bacteriostasis of C. chinensis and S. baicalensis when combinating with them.

Object To set up the quantitative method of baicalin in Gegen Qinlian Decoction (GQD) by RP HPLC and to determine the change of baicalin contents in decoctions prepared from various combinations of GQD. Methods By L 8(2 7) orthogonal design and statistics analysis (SPSS10 0), eight decoctions with Radix Scutellariae either alone or incombination with one or more of the other three components were prepared or analyzed. Contents of baicalin in them were determined by RP HPLC. Results It was significant (P0 05) and interactions between two of three were insignificant either. In this experiment, the sedimentation was found in the decoctions contained Radix Scutellariae and Rhizoma Coptidis. Conclusion The content of baicalin is reduced by Radix Puerariae and Rhizoma Coptidis, The resolution of baicalin is not increased by Radix Glycyrrhizae Preparata. Maybe the change of pH leads to the sedimentation in decoctions contained Radix Scutellariae and Rhizoma Coptidis.

Object To establish a quantitative method for ephedrine and pseudoephedrine in the aqueous decoctions of MAHUANG TANG (ADMHT) by GC MS, and to determine the content of the two components in decoctions of the different recipes consisted of Herba Ephedrae, and other herbs by using the quantitative method will be established, so as to study the effect of compatible medicinal herbs on the release of the two components in Herba Ephedrae to decoctions Methods The quantitative method by the help of characteristic ion was used in GC MS Results The content of ephedrine and pseudoephedrine in decoction prepared from Herba Ephedrae and Semen Armeniacae Amarum evidently decreases, the content of two components in decoction prepared from Herba Ephedrae and Ramulus Cinnamomin evidently increases, the content of two components in decoction prepared from Herba Ephedrae and Radix Glycyrrhizae has no obvious change, as compared with that of two components in decoction from Herba Ephedrae alone Conclusion The method is good in the aspect of separable effect, sensitivity and reproducibility The method not only can quantify the two components, but also provide the bases of quality control for the pharmacokinetic study and therapeutic detection of Herba Ephedra recipes

ABSTRACT

Object To study the HPCE fingerprinting of Gegen Qinlian Decoction (GQD). Methods A buffer was composed of 30 mmol/L sodium phosphate and 40 mmol/L borate solution. Capillary electrophoresis was performed using a 65 cm (43 cm to detector) ?50 ?m fused-silica capillary tube. Separation voltage was 22 kV, sampling time was 1 s, detected wavelength was 254 nm, and the temperature was maintained at 30 ℃. Results The 27 components in GQD were successfully separated. The observation of methodology was in keeping with quantitatine determination and qualitative study. Conclusion This method can be used for the quality control of the preparation of GQD with good precision.

ABSTRACT

Object To develop the quantitative method of cinnamaldehyde in Mahuang Decoction (MHD) by GC-MS and study the influence of various combinations of MHD on the contents of cinnamaldehyde in the decoctions. Methods By L_8(2~7) orthogonal design and statistic analysis (SPSS 10.0), eight decoctions with Ramulus Cinnamomi were prepared and analyzed. Contents of cinnamaldehyde were determined by GC-MS. The pH value of decoctions was determined by pH meter. Results For Herba Ephedrae and Semen Armeniacae Amarum, the influences on the contents of cinnamaldehyde were significant (P0.05). The change of pH value not obvious. Conclusion The content of cinnamaldehyde is only reduced significantly by Herba Ephedrae and Semen Armeniacae Amarum. But it is insignificant for Radix Glycyrrhizae and its combinations.

Object To set up the quantitative method of amygdalin in decoctions prepared from various combinations of the recipe named Mahuang Decoction by HPLC and to determine the change of amygdalin contents in different decoctions and study the influence of compatibility on the contents of amygdalin in decoctions. Methods By L 8(27) orthogonal design and statistic analysis (SPSS 10.0), eight decoctions with Semen Armeniacae Amarum were prepared and analyzed, their amygdalin contents were determined by HPLC. Results The method was simple, accurate, and sensitive and was not interfered by other constituents in the prescription, it can be used to determine the contents of amygdalin in Mahuang Decoction. Influence of Herba Ephedrae, Ramulus Cinnamomin, and Radix Glycyrrhizae on the contents of amygdalin in decoctions were insignificant. Interactions between two of three separately were insignificant too. Conclusion The content of amygdalin in Mahuang Decoction is not markedly influenced by Herba Ephedrae, Ramulus Cinnamomin, and Radix Glycyrrhizae.

Object To set up the quantitative method for determining the puerarin in Gegen Qinlian Decoction (GQD) by RP-HPLC and to study the change of puerarin contents in various combinations of GQD. Methods L 8(2 7) orthogonal design and statistic analysis (SPSS 10.0) were used, the puerarin contents in the samples were determined by HPLC. Results Influence of Radix Scutellaria, Rhizoma Coptidis and Radix Glycyrrhizae Preparata on the puerarin content in GQD was insignificant. And interactions between two of three were insignificant too. In this experiment, puperarin in the sedimentation was found in the compatible decoction of Rhizoma Coptidis and Radix Puerariae. Conclusion The puerarin content is not markedly influenced by Radix Scutellaria, Rhizoma Coptidis and Radix Glycyrrhizae Preparata. Maybe, the change of pH is relevant to the sedimentation in the compatible decoction contained Rhizoma Coptidis. and Radix Puerariae

ABSTRACT

Objective To probe into the therapeutic mechanism of Zhixie Tuire Tablet (ZTT) for rotavirus-induced diarrhea. Methods We established the 4-day old Kunming neonatal mice model by infecting group A G3 type of rotavirus,and then gave the inoculated mice with ZTT at the dose of 0.78 g/kg for three days. Fluorescent quantification PCR method was used to detect the changes of sodium glucose transporter SGLT1 mRNA level and radio-immunity method was used for the examination of PGE2 level in neonatal mice small intestine. Results SGLT1mRNA level was lower in ZTT group than that in the normal group,the difference being significant (P 0.05). ABSTRACT

Objective To evaluate the pharmacological effect of Zhixie Tuire Tablets(ZTT) on human rotavirus RV709 in host cells in vitro.Methods By improved methods of methyl thiazolyl tetrazolium colorimetry assay(MTT) and cytopathic effect(CPE),the anti-rotavirus effect of ZTT on RV709 was evaluated.Results The half toxic concentration(TC50) of ZTT for MA104 was 0.49699 mg/mL.ZTT neither can directly kill human rotavirus,nor prevent human rotavirus from invasion of host cells.The halft inhibitory concentration of ZTT was 0.00519 mg/mL for RV709.Conclusion In-vitro anti-rotavirus mechanism of ZTT is related to the inhibition of the biological synthesis of rotavirus in host cells.

ABSTRACT

AIM: To analyse the main chemical constituents of Shuanghuanglian Injection Power(Flos lonicerae japanicae,Radix scutellariae and Fructus forsythiae) by high performance liquid chromatography-electrospray ionization-mass/mass spectrometry. METHODS: The sample solution was analysed by a Zorbax C_18 column with a gradient mobile phase comprised of acetonitrile and water.Electrospray ionization mass spectrometry detector was used,full-scan detection mode was evaluated for the identification of the liquid chromatography peaks. RESULTS: To analyse the mass spectrum of every liquid chromatography peaks,we identified 18 molecular mass from the ion chromatogram of Shuanghuanglian Injection Powder,9 compounds were identified,including quinic acid,caffeic acid,three monocaffeoylquinic acids,dicaffeoylquinic acid,forsythin,forsythoside and baicalin. CONCLUSION: Quinic acid is identified from Shuanghuanglian Injection Power for the first time.High performance liquid chromatography-mass/mass spectrometry is emerging as an important tool in traditional Chinese medicine.
