ABSTRACT

AIM: To investigate the effects of primary acquired nasolacrimal duct obstruction(PANDO)on the tear film and ocular surface using LipiView ocular surface interferometer and Keratograph 5M anterior segment analyzer.METHODS: A self-controlled clinical trials. A total of 40 patients diagnosed with unilateral PANDO for at least 6mo who were admitted to our department from September 2021 to March 2022 were enrolled in the study, and the healthy eyes of the patients were assessed as control group. The LipiView ocular surface interferometer and Keratograph 5M anterior segment analyzer were used to measure the changes in related parameters of the tear film and ocular surface in both eyes.RESULTS: The non-invasive tear meniscus height(NITMH), stimulated NITMH, loss rate of upper meibomian gland, nasal and temporal ciliary redness index, temporal conjunctival redness index of the affected eyes were higher than healthy eyes(P<0.05), but there were no statistical differences in the non-invasive break-up time(NIBUT), loss rate of lower meibomian gland, nasal conjunctival redness index, dry eye grading, blink responses, partial blink rate and lipid layer thickness(LLT)between the both eyes(P>0.05).CONCLUSION: PANDO may lead to the aggravation of ocular surface inflammation and the loss of upper meibomian gland, and damage the ocular surface of patients. Attention should be paid to the early treatment of PANDO.

ABSTRACT

Objective:To explore the risk factors of autogenous arteriovenous fistula (AVF) aneurysms (AVFAs) in maintenance hemodialysis (MHD) patients.Methods:The patients who used internal arteriovenous fistula (end to side anastomosis) of cephalic vein-radial artery at wrist as vascular access in Hainan Provincial People′s Hospital from June 1 to June 30, 2021 were selected as the research objects. The patients were divided into AVFAs group and non-AVFAs group according to whether AVF formed AVFAs. The clinical data and laboratory examination results between the two groups were compared. Binary logistic regression model was used to analyze the risk factors for the formation of AVFAs.Results:A total of 170 MHD patients were enrolled in this study, including 111 males (65.3%) and 59 females (34.7%), with age of (51.65±12.70) years old and dialysis age of (57.03±49.25) months. There were 33 cases in AVFAs group and 137 cases in non-AVFAs group. The incidence of AVFAs was 19.4%. Compared with non-AVFAs group, the proportion of males ( χ2=4.934, P=0.026) and the levels of serum uric acid ( t=2.547, P=0.012) and serum albumin ( t=2.122, P=0.010) in AVFAs group were higher; The age ( t=-2.210, P=0.028), the proportion of diabetes nephropathy ( χ2=11.788, P=0.001), systolic blood pressure ( t=-1.994, P=0.048) and total cholesterol ( t=-2.174, P=0.031) were lower; The diameter of anastomosis was wider ( Z=-3.224, P=0.001); Mantel-Haenszel chi square test analysis showed that dialysis age ( χ2=53.832, OR=0.518, P<0.001), AVF service time ( χ2=51.355, OR=0.516, P<0.001), and brachial artery blood flow ( χ2=25.315, OR=0.331, P<0.001) were correlated to the formation of AVFAs. The results of multivariate logistic regression analysis showed that males ( OR=10.005, 95% CI 1.875-53.394, P=0.007), longer dialysis age ( OR=1.341, 95% CI 1.104-1.628, P=0.003), longer AVF use time ( OR=1.187, 95% CI 1.002-1.405, P=0.047), higher brachial artery blood flow ( OR=1.002, 95% CI 1.000-1.004, P=0.028) and lower total cholesterol ( OR=0.388, 95% CI 0.172-0.875, P=0.022) were the independent risk factors for the formation of AVFAs. Conclusions:The incidence of AVFAs in MHD patients is 19.4%. Males, long dialysis age, long AVF use time, high brachial artery blood flow and low total cholesterol level are the independent risk factors for the formation of AVFAs.

ABSTRACT

OBJECTIVE@#To analyze the expression of immunoglobulin mucin molecule 3 (TIM-3) in epithelial ovarian cancer (EOC) and the effects of TIM-3 knockdown and overexpression on proliferation and migration of ovarian cancer cells.@*METHODS@#We analyzed TIM-3 expression in EOC and normal ovarian tissues using GEPIA database. We also detected TIM-3 expression levels in 82 surgical specimens of EOC and 18 specimens of normal ovarian tissues using immunohistochemistry, and analyzed the correlation of TIM-3 expression with clinicopathological parameters and survival outcomes of the patients. The expression of TIM-3 and Wnt1 mRNA in the tissues were detected using qRT-PCR. We constructed SKOV3 cell models of TIM-3 knockdown and overexpression and examined the changes in proliferation, apoptosis, migration and invasion of the cells using MTT assay, Annexin V-FITC/PI staining, scratch test and Transwell assay. The activity of Wnt/β-catenin pathway in the transfected was detected using dual luciferase reporter assay, and the mRNA levels of TCF-7, TCCFL-2 and CD44 were detected using qPCR. The protein expressions of MMP-9, CD44, Wnt1, β-catenin and E-cad in the transfected cells were detected with Western blotting.@*RESULTS@#The positive expression rate of TIM-3 was significantly higher in EOC tissues than in normal ovarian tissues (P < 0.05). The expression of TIM-3 was significantly correlated with FIGO stage, histological differentiation and lymph node metastasis, and was positively correlated with Wnt1 level (P < 0.05). In SKOV3 cells, TIM-3 knockdown significantly lowered the activity of Wnt/ β-catenin pathway, inhibited cell proliferation, migration and invasion, and promoted cell apoptosis. TIM-3 knockdown significantly down-regulated the mRNA levels of TCF-7, TCFL-2 and CD44 and the protein levels of MMP-9, CD44, Wnt1 and β-catenin, and significantly up-regulated the expression level of E-cad (P < 0.05). Overexpression of TIM-3 caused opposite effects in SKOV3 cells.@*CONCLUSION@#TIM-3 is highly expressed in EOC tissue to promote malignant behaviors of the tumor cells possibly by activating the Wnt/β-catenin signal pathway.

ABSTRACT

Objective In order to understand the contamination status of food-borne pathogens in food in Nantong City, and to provide a scientific basis for food safety risk assessment and supervision. Methods The monitoring was performed according to the plan of food safety risk surveillance in Nantong City and the manual on monitoring microbes and its pathogenic factors in foodstuffs of Jiangsu Province. A total of 1025 food samples of 11 kinds were monitored from 2017 to 2020 for detecting of pathogenic bacteria such as Listeria monocytogenes, Salmonella, Vibrio parahaemolyticus, Bacillus cereus and Staphylococcus aureus, and the test results were summarized, analyzed and evaluated. Results A total of 142 pathogenic bacteria were detected, with a total positive rate of 13.85%. The detection rate was the highest in 2018 (25.65%), and the lowest in 2017 (4.98%). The highest detection rate was found in meat and meat products(25.38%), followed by aquatic products and animal products (17.92%), cooked rice and flour products around the school (14.29%), the detection rate of barreled drinking water was the lowest (2.86%). 170 strains of pathogenic bacteria were detected from 1025 samples. The higher positive rate was Bacillus cereus in the cooked rice flour products around the school, and 5 strains of Bacillus cereus were detected, with the detection rate was 14.29%. 18 strains of Staphylococcus aureus were detected in meat and meat products, and the detection rate was 13.85%; 70 Vibrio parahaemolyticus were detected in aquatic products, and the detection rate was 13.49%; 16 Listeria monocytogenes were detected in meat and meat products, and the detection rate was 12.31%. The positive rate of catering links is higher than that of circulation links, and the positive rate of urban areas is higher than that of rural areas. Conclusions Pathogenic bacteria were detected in different kinds of food, and the overall situation of food safety is stable. In the future, the monitoring scope will be expanded continuously, the monitoring continuity will be strengthened, the sampling quantity will be increased, the monitoring quality will be improved, the trend of food pollution change in the city shall be mastered in time, food safety warning shall be well done, and food safety education shall be strengthened for food practitioners, food pollution risk shall be reduced and food safety shall be effectively guaranteed.

ABSTRACT

Efficient management and evaluation is the primary responsibility of drug regulatory authorities in various countries, whose management services are mainly delivered in accordance with the Good Review Practices(GRPs) and ISO 9001 quality management system. In 2015, Opinions on Reforming the Evaluation and Approval System of Drugs and Medical Devices included the perfection of quality control review system as the content of reform in drug review and approval. On this basis, the traditional Chinese medicine(TCM) review authority has shortened the review time, strengthened the communication, offered technical guidelines, designed the review template with guiding significance, and carried out diversified training and performance evaluation, to continuously strengthen the standardization of TCM review and approval and effectively guarantee the scientificity and consistency of new TCMs. However, the current quality management system for TCM review is not yet mature. It is recommended to employ ISO 9001 quality management system to manage the review work, utilize the GRP for better management of the archive files in the review process, and use quality evaluation indicators to improve the review quality management, thereby further perfecting the TCM review management system.

ABSTRACT

Objective:To observe the regulatory effect of modified Shengmaiyin adjuvant chemotherapy on the nutritional status and immune function of children with acute lymphocyte (ALL). Method:One hundred children with ALL chemotherapy were randomly divided into observation group and control group. Both groups were given VDLD regimen as the induction therapy and CAM regimen as the early therapy. Control group was given Shengmaiyin orally, 10 mL/time, 3 times/day. Observation group was given modified Shengmaiyin orally, 1 does/day. The course of treatment for both groups was 3 months. Micro-nutrition assessment (MNA) was carried out before and after treatment, and serum total protein (TB), albumin (ALB), prealbumin (PAB), T cell CD4<sup>+</sup>, CD8<sup>+</sup>, immunoglobulin G (IgG), IgM, IgA levels were detected before and after treatment. The ratio of CD4<sup>+</sup>/CD8<sup>+</sup> was calculated. The traditional Chinese medicine (TCM) syndrome before and after treatment and the piper fatigue scale (PFS-R) and universal core scale of children's quality of life (PedsQL) were scored. The changes of white blood cells (WBC), red blood cells (RBC), hemoglobin (Hb) and platelets (PLT) were evaluated before and after treatment. Result:The nutritional status of the observation group was better than that of the control group after treatment (<italic>Z</italic>=2.018, <italic>P</italic><0.05). The observed fatigue was lighter than that of the control group (<italic>Z</italic>=2.029, <italic>P</italic><0.05). The MNA score of the observation group was higher than that of the control group (<italic>P</italic><0.01). The scores of PFS-R and deficiency of both Qi and blood were lower than those of the control group (<italic>P</italic><0.01). The CD4<sup>+ </sup>levels and CD4<sup>+</sup>/CD8<sup>+</sup> levels of TB, ALB and PAB in the observation group were higher than those in the control group (<italic>P</italic><0.01), while the CD8<sup>+</sup> was lower than the control group (<italic>P</italic><0.01). The IgM and IgA levels in the observation group were higher than those in the control group (<italic>P</italic><0.01). The RBC, Hb and PLT levels of the observation group were higher than those of the control group (<italic>P</italic><0.01). Conclusion:Modified Shengmaiyin can be used as adjuvant therapy for children with ALL chemotherapy by improving nutritional status, immune function and immune balance, reducing clinical symptoms, promoting the recovery of hematopoietic system and improving the quality of life.

ABSTRACT

Clinical trail report is a summary of the process and results of clinical trail, an important basis to support the effectiveness and safety evaluation of drug marketing and an important technical data required for drug registration. Safety analysis is the main part and the most common issue of clinical trial report. In this article, it is summarized and analyzed the common problems found in the safety analysis part of the clinical trial report of new drugs of traditional Chinese medicine(TCM) in combination with the clinical professional evaluation of the application for marketing license of innovative TCM. The common problems in writing the safety aspects of clinical trail reports were summarized and analyzed, including the omissions of adverse events and laboratory test results, the failure to provide a detailed list of laboratory test outliers, the lack of professional depth in the analysis of the causal relationship between adverse events and TCM. In view of the common problems, it is suggested that the drug use degree, adverse events and adverse reactions, as well as laboratory examination should be considered. Check the three dimensions for necessary medical analysis, collation and summary and continuously improve the quality of safety analysis in clinical trial reports.

ABSTRACT

Spraying during dental practices can generate quantity of droplets, which transform into droplet nuclei and suspend in the air of clinic in the form of aerosol. Either droplets or aerosols may carry microorganisms and cause air contamination in the hospital, posing potential threat to the health of clinical healthcare staff and patients. The present article summarizes some basic concepts involved in bio-aerosol research and reviews literatures on intervention measures of dental clinic droplet/aerosols to clarify whether there is experimental evidence of aerosol-carrying bacteria in the existing literatures. The aim of this article is to provide evidence for the formulation of guidelines for infection control in dental healthcare practices during the epidemic period of infectious diseases, as well as to provide reference and scientific basis for the management and implementation of infection control measures in daily dental clinical work.

ABSTRACT

Intracerebral hemorrhage (ICH) refers to the primary non-traumatic parenchymal hemorrhage, which is one of the common cerebrovascular diseases, with a high incidence, rapid development, slow recovery and high disabling rate. After intracerebral hemorrhage, a series of pathological changes occur in the brain tissue, such as local hematoma and its space occupying effect, secondary cerebral edema, death of brain cells and destruction of blood-brain barrier, which may lead to brain injury and neurological defects, seriously affect the quality of life of patients, and even endanger the life. Therefore, it is great medical value to find effective therapeutic methods and drugs, explore the mechanisms and targets for improving neurological function, reduce sequelae and improve the quality of life of patients. According to the theory of traditional Chinese medicine (TCM), cerebral hemorrhage belongs to " abnormal flow of the blood" , which equals to blood stasis. In recent years, scholars conducted extensive research on drugs for promoting blood circulation to remove blood stasis with modern scientific methods, and made in-depth discussion for the mechanism, and found that therapies for activating blood and removing blood stasis, plays a key role in intervening a series of physiological and pathological changes after cerebral hemorrhage, with significant curative effects in removing hematoma, improving the microcirculation and reducing the mortality and morbidity. This article summarized drugs for promoting blood circulation to remove blood stasis (Notoginseng Radix et Rhizoma, Salviae Miltiorrhizae Radix et Rhizoma, Hirudo), formulas (Buyang Huanwu Tang, Didangtang, Naoxueshu oral liquid, Tongqiao Huoxuetang) and compound injections (Danhong injection) for the treatment of cerebral hemorrhage targets, and discussed the experimental research progress TCM for promoting blood circulation to remove blood stasis in treatment of cerebral hemorrhage in terms of promoting hematoma absorption, reducing brain edema and apoptosis, promoting angiogenesis, inhibiting the inflammatory response, and promoting the repair and regeneration of nerve tissue in nearly five years, and summarized the therapeutic mechanism, so as to provide scientific basis for clinical application of the therapeutic methods for activating blood and removing stasis to treat cerebral hemorrhage and the modern scientific research.

ABSTRACT

OBJECTIVE@#To investigate the role of growth arrest-specific protein 6 (Gas6) in the process of the migration and osteogenic differentiation of human periodontal ligament cells (hPDLCs).@*METHODS@#After different concentrations of recombinant human Gas6 (rhGas6) were added to hPDLCs, cell prolife-ration experiment (CCK-8) was taken to observe the effect of rhGas6 on hPDLCs cell proliferation. Scratch test and cell migration test (Transwell) were taken to analyze the migratory ability of hPDLCs in different concentrations of rhGas6 groups. After osteogenic induction, real-time quantitative polymerase chain reaction (real-time PCR) was taken to detect the expression of the Runt-related transcription factor 2 (Runx2) and alkaline phosphatase (ALP). ALP staining was used to detect the amount of mineralized nodules.@*RESULTS@#After adding different concentrations of rhGas6, there were no statistically significant differences in hPDLCs cell proliferation among the experimental groups and the control group at 24, 48 and 72 hours (P>0.05). After 24 h of scratch, the healing area in the 800 μg/L of the rhGas6 group was greater than that in the control group, but without statistically significant difference (31.06%±13.70% vs. 21.79%±9.51%, P>0.05). In the migration test, after 24 h, the number of hPDLCs cells which penetrated through the membrane in the 800 μg/L rhGas6 group was significantly higher than that in the control group (P < 0.01). After rhGas6 was added and osteogenic induction, Runx2 and ALP gene expressions of hPDLCs in the 800 μg/L group were significantly higher than those in the control group (1.60±0.30 vs. 0.91±0.10, 2.81±0.61 vs. 0.86±0.12, P < 0.01). After Gas6 was knocked down, the ALP expression of hPDLCs was significantly lower than that of the control group (0.39±0.07 vs. 0.92±0.14, P < 0.01). There was no significant change in Runx2 expression (P>0.05). After 7 days of osteogenic induction, the mineralized nodules formed in the Gas6 knockdown group were significantly less than those in control group (0.25±0.04 vs. 1.00±0.11, P < 0.001). After 14 days of induction, the staining degree of the Gas6 knockdown group was lower than that of the control group, but there was no significant difference (0.86±0.04 vs. 1.00±0.16, P>0.05).@*CONCLUSION@#After downregulation of Gas6 gene, mineralized nodule formation was reduced and ALP gene expressions were decreased in the early stage of osteogenic induction (7 days). After addition of rhGas6, Runx2 and ALP gene expressions were increased and the number of cell migration was increased, suggesting that Gas6 might play a promoting role in the migration and osteogenic differentiation of human periodontal ligament cells.

ABSTRACT

Objective@#To explore the relationships between the characteristics of childhood sexual abuse and non-suicidal self-injury in nursing female college students.@*Methods@#Two medical colleges and junior colleges were selected in Anhui province. A total of 2 549 female nursing students in grade 1 to 3 were asked to fill a questionnaire regarding sociodemographic information,childhood sexual abuse and non-suicidal self-injury.@*Results@#The reported rate of non-suicidal self-injury among female nursing students in the past six months was 8.2%. Sexual abuse at any time during childhood (primary school or earlier,middle school and high school) increased the risk of non-suicidal self-injury among female nursing students (P<0.05). Exposure to sexual abuse in all three periods was associated with 5.04(95%CI=1.73-14.62) times odds ratio than that of those who not exposed to sexual abuse (P<0.01). Only contact sexual abuse and both contact and non-contact sexual abuse in childhood were correlated with nonsuicidal selfinjury among female nursing students [OR(95%CI)=2.21(1.48-3.29), 3.56(2.13-5.96)] (P<0.05). Two patterns of sexual abuse experiences were identified,including persistent sexual abuse (3.1%) and the other group is occasional sexual (96.9%). Persistent sexual abuse in childhood was correlated to higher risk of non-suicidal self-injury compared with occasional sexual abuse (OR=2.61,95%CI=1.35-5.05,P<0.01).@*Conclusion@#The occurrence periods,types and patterns of sexual abuse in childhood are closely related to non-suicidal self-injury in female nursing students.

ABSTRACT

OBJECTIVE@#To investigate the tumorigenicity of several multiple myeloma (MM) cell lines transplanted in mice without γ-ray irradiation and to construct the MM disease model to facilitate in vivo experiments.@*METHODS@#NOD/SCID or NSG mice were subcutaneously or caudally transplanted with MM cell lines (LP-1, OPM2, RPMI 8226 and MOLP8), or cell lines with luciferase (RPMI-Luc-Puro, RPMI-Luc-mCherry and MOLP8-Luc-Puro). Tumor growth was observed by measuring the tumor size with a caliper. CD138 tumor cells in peripheral blood were detected by flow cytometry. The free light chain in mouse serum was detected by immunofixation electrophoresis. Tumor type was identified by immunohistochemistry.@*RESULTS@#Twenty one NOD/SCID mice were subcutaneously transplanted with LP-1 cells or OPM2 cells respectively, and no tumors formed till 7 weeks after transplantation. Fifteen NOD/SCID mice subcutaneously transplanted with RPMI 8226 cells showed tumor formation one week later. As of 7 weeks, the rate of tumorigenesis was 80% (12/15). Serum λ light chain was detected and no CD138 tumor cells were detected in peripheral blood. Two NOD/SCID mice each were subcutaneously transplanted with RPMI-Luc-Puro, RPMI-Luc-mcherry and MOLP8-Luc-Puro cells respectively. No tumor signal was detected through IVIS in RPMI-Luc-mcherry cells-transplanted mice. There was tumor signal at 1 week in RPMI-Luc-Puro and MOLP8-Luc-Puro cells-transplanted mice, the former disappeared at 2 weeks and the latter persisted more than 3 weeks. NSG mice subcutaneously transplanted with both cells persistently displayed the tumor signal. Neither NOD/SCID nor NSG mice transplanted with RPMI 8226, RPMI-Luc-Puro, RPMI-Luc-mcherry or MOLP8-Luc-Puro cells through tail vein developed the tumor signal. Only one NSG mice transplanted with MOLP8-Luc-Puro cells appeared transient tumor signal.@*CONCLUSION@#Unirradiated mice transplanted with MM cell lines tended to develop local tumor, and failed to develop disseminated tumor. The tumorigenicity of different cell lines is quite different and the vector transfection can reduce the tumorigenic ability. NSG mice with more severe immunodeficiency are more suitable for tumor growth.

ABSTRACT

Objetive To investigate the effect of Qingkailing injection on the expression of Toll-like receptor 4 (TLR4), gp91phox and zonula occludens-1 (ZO-1) in cerebrovascular endothelial cells induced by hypoxia activation of microglias. Methods:BV2 microglia cells were divided into six groups. They were cultured in serum-free DMEM, while the Qingkailing groups of low, middle and high dosages were cultured with 0.0625%, 0.125% and 0.25% Qingkailing injection, respectively, and minocycline group with minocycline of 200 nmol/L. The groups other than control group underwent hypoxia for 24 hours and reoxygenation for 24 hours. Then, the medium of microglia was put into the medium of Balb/c endothelial cells for 24 hours. The cell viability of endothelial cells was measured with CCK-8, the concentration of nitric oxide (NO) was detected with colorimetry, and the experission of TLR4, gp91phox and ZO-1 was detected with Western blotting. Results:Compared with the control group, the cell viability and the expression of ZO-1 decreased in the model group (P < 0.01), while the concentration of NO and the expression of TLR4 and gp91phox increased (P < 0.05). Compared with the model group, the cell viability and the expression of ZO-1 increased in the Qingkailing groups and the minocycline group (P < 0.05), while the concentration of NO and the expression of TLR4 and gp91phox decreased (P < 0.05). Conclusion:Qingkailing injection may enhance the survival and function of cerebrovascular endothelial cells by inhibiting the hypoxia activation of microglias, reducing the expression of TLR4 and gp91phox, and increasing the expression of ZO-1.

ABSTRACT

Reactive oxygen species are generated by electron leakage in mitochondrial respiratory chain, including superoxide anion, hydrogen peroxide, hydroxyl radical and nitric oxide. When the intracellular ROS level is abnormal or the antioxidant system is out of balance, the structure and function of mitochondria would be damaged, such as the damage of mitochondrial DNA, the abnormality of respiratory chain complexes and the decrease of membrane permeability. At the same time, the mitochondrial pathway of apoptosis would be activated to induce apoptosis. ALS is a devastating neuromuscular disease characterized by loss of motor neurons and significant skeletal muscle atrophy. Studies have shown that mitochondrial damage caused by ROS overproduction is closely related to the process of ALS. This article reviews the mechanisms of ROS production and clearance, mitochondrial damage induced by ROS and its related research in ALS.

OBJECTIVE@#Growth-arrest-specific protein 6 (Gas6) is a vitamin K-dependent protein and involved in cell proliferation, survival, adhesion and migration . Also it has been shown to play an important role in the inflammatory response .The aim of present study was to investigate the role of Gas6 in the process of the expression of adhesion molecules and chemokines of human umbilical vein endothelial cells (HUVECs) induced by Porphyromonas gingivalis lipopolysaccharide(P.g-LPS).@*METHODS@#After up-regulation and down-regulation of the expression of Gas6, the vascular endothelial cells were stimulated with 1 mg/L P.g-LPS for 3 h and 24 h. Real-time quantitative polymerase chain reaction(real-time PCR) was taken to detect the expression of the cell adhesion molecules:intercellular adhesion molecule-1 (ICAM-1) and E-selectin, as well as chemokines:interleukin-8 (IL-8) and monocyte chemoattractant protein 1 (MCP-1). Wound healing assay was taken to observe the migration ability of endothelium cells in different groups.@*RESULTS@#After 3 h of P.g-LPS stimulation, the expression of adhesion molecules and chemokine in the down-regulation group was not significantly different from that in the control group,while in the up-regulation group the decrease of E-selectin, ICAM-1, IL-8 and MCP-1 was 81%±0%, 47%±3%, 76% ± 3%, 26% ± 6% respectively. After 24 h of P.g-LPS stimulation, the expression of adhesion molecules and chemokine in down-regulation group was significantly higher than that in control group (2.06±0.07, 1.99±0.11, 3.14±0.15, 1.84±0.03 flod), while these molecules in the down-regulation group was significantly lower than in the control group (29%±1%, 62%±3%, 69%±1%, 41%±2%). Differences were statistically significant (P<0.01). Wounding healing assay showed that down-regulation of Gas6 enhanced migration ability of endothelial cells while up-regulation of Gas6 weakened this ability,which was consistent with the trend of real-time PCR result.@*CONCLUSION@#Down-regulation of the Gas6 gene enhanced the expression of ICAM-1, E-selectin, IL-8 and MCP-1 in HUVECs after P.g- LPS stimulating, while up-regulaiton of the Gas6 gene weakened the expression of ICAM-1, E-selectin, IL-8 and MCP-1 in HUVECs after P.g-LPS stimulating,suggesting that Gas6 may play a role in the process of endothelial cell adhesion.

ABSTRACT

Objective To assist clinical diagnosis of Demodex blepharitis and other related ocular surface disease.Methods 42 cases were collected from the patients diagnosed of blepharitis,dry eye or blepharitis related keratoconjunctivitis (BKC) from the outpatient of the First Hospital of Xi'an City from April and July 2016.The eyelashes were pulled out and detected for the Demodex under microscopy after adding a drop of ethanol.Results 30 cases in 42 patients were detected for the Demodex (the positive rate was 71 %),while only 1 case in 10 healthy volunteers was detected for the Demodex with the positive rate of 10 %.The difference between the two groups was statistically significant (x2 =10.23,P< 0.01).Conclusion There is a high incidence rate of Demodex blepharitis.The early rapid laboratory diagnosis for Demodex blepharitis should be paid attention to and reinforced,in order to guide the timely and accurate treatment and avoid the occurrence and development of the Demodex related blepharitis,dry eye,BKC or other ocular complications.

The microbiota of the ocular surface consists of diverse microbial communities and colonizes on the ocular mucous membrane including tear film.The symbiotic relationship maintains the stabilization of ocular biosystem and ensures its healthy status.The initiation of the Human Microbiome Project and the rapid development of the relevant research technologies promote the study of the relationship between microbial communities and human health to a new era,which open up new train of thought for the pathogenesis and treatment of microbial infectious disease.This article reviewed the recent research advances on the microbiota of ocular surface and the influence of some pathogenic factors,including keratitis,conjunctivitis,endophthalmitis,dacryocystitis,blepharitis,dry eye and corneal contact lens on the microbiota of the ocular surface,the relationship between the microbiota of the ocular surface and ocular surface diseases.

ABSTRACT

Objective To investigate the current situation of social support for patients with coronary heart disease intervention in different hospitals in Dalian, to analyze the influence of different demographic characteristics on patients' social support, to provide reference for the rehabilitation and management of patients undergoing coronary intervention surgery. Methods From November 15th to 16th, 2017, a cross-sectional unified investigation was carried out in 20 hospitals of different levels in Dalian, patients' general information questionnaire, Self-rating Anxiety Scale (SAS), Self-rating Depression Scale (SDS), Self-management Behavior Scale (SSRS) and Social Support Rating Scale (SSRS) were adopted. Results The total score of social support was 29.57 ± 12.01 in patients with coronary artery disease and interventional therapy in different hospitals in Dalian area. The scores of social support in patients with different residence area and residence condition were statistically significant (F=1.769, 6.150, P<0.01). The influencing factors of patients'social support were living condition, depression and self-management (r=-0.243,-0.129, 0.255, P<0.05). Conclusion Social support for patients with coronary artery disease intervention in different hospitals in Dalian is at a moderate level. And affected by living conditions, depression, self- management and other factors. Medical staff should formulate corresponding intervention measures to improve their social support level and improve the quality of life of patients.

ABSTRACT

Objective: To observe the changes in numbers and degranulation status of local mast cells (MCs) of the complementary acupionts, as well as the concentrations of serum histamine and bradykinin after acupuncture at rat's complementary acupionts of Yanglingquan (GB 34) and Yinlingquan (SP 10), for exploring the mechanisms of the synergistic effect of complementary acupionts. Methods: Using random number table method, 40 Wistar rats were randomly divided into a blank control group (group K), an acupuncture at Yanglingquan (GB 34) group (group A), an acupuncture at Yinlingquan (SP 9) group (group B), an acupuncture at Yanglingquan (GB 34) and Yinlingquan (SP 9) group (group AB), and an acupuncture at points near Yanglingquan (GB 34) and Yinlingquan (SP 9) group (group CD), 8 rats in each group. Group A received acupuncture at Yanglingquan (GB 34); group B received acupuncture at Yinlingquan (SP 9); group AB received acupuncture at Yanglingquan (GB 34) and Yinlingquan (SP 9); group CD received acupuncture at the control points [points 3 mm away from Yanglingquan (GB 34) and Yinlingquan (SP 9) respectively]. Acupuncture was performed at bilateral points in each group. Rats in group K were fixed using the same method as rats in the other 4 groups without acupuncture stimulation. Needles of 0.35 mm in diameter and 40 mm in length were used for acupuncture. Needle handle was connected to the G6805-Ⅱ mode electroacupuncture (EA) device after needling qi was obtained, with sparse-dense wave, frequency of 2 Hz/100 Hz and current of 1 mA to keep the needle handles slightly tremulous while the rats kept quiet. Rats were continuously stimulated by EA for 20 min each time. Experimental interventions were conducted on the 1st, 3rd, 5th and 7th days when the experiment started, for 4 times in total. Specimens from rats in each group were collected 2 h after the 7th day. The levels of histamine and bradykinin in serum were detected by enzyme-linked immunosorbent assay (ELISA). Local tissues of the points were used to prepare cryosections. The changes of MCs were observed after toluidine blue staining. Results: Compared with group K, the numbers and degranulation rates of MCs in group A, group B, group AB and group CD were significantly increased (all P<0.05). The number and degranulation rate of MCs in group AB were significantly higher than those in group A, group B and group CD (all P<0.05). The order of histamine levels from high to low was: group AB >group B > group A > group CD > group K, and the differences were statistically significant (all P<0.05); the levels of bradykinin in group AB, group A and group B were significantly higher than those in group K and group CD (all P<0.05). There was no significant difference in the bradykinin level between group A and group B, nor between group CD and group K (all P>0.05). Conclusion: The number and degranulation rate of MCs of the complementary acupionts are significantly increased after acupuncture at complementary acupionts of Yanglingquan (GB 34) and Yinlingquan (SP 9) of rats, and the serum levels of histamine and bradykinin are increased, which may be one of the mechanisms of the synergism produced by the combination of complementary acupionts.

ABSTRACT

Objective: To investigate the effects of microRNA-378 (miR-378) on proliferation and migration of renal cancer cell lines in vitro, and to explore the possible mechanism. Methods: The expression of miR-378 in renal cell carcinoma and normal renal epithelial tissues was analyzed using the datasets from The Cancer Genome Atlas (TCGA). The expression of miR-378 in human renal carcinoma cell lines 786-O, ACHN, 769-P and Caki-1 as well as human renal proximal tubular epithelial cell line HK2 was detected by real-time fluorescent quantitative PCR (RFQ-PCR). MiR-378 mimic and inhibitor were synthesized and then transfected into renal carcinoma 786-O and Caki-1 cells, respectively. The proliferation and migration of 786-O and Caki-1 cells were detected by CCK-8 assay, Transwell chamber assay and wound healing assay, respectively. The potential target gene of miR-378 was screened by bioinformatics, and confirmed by RFQ-PCR and Western blotting in renal cancer cells, respectively. Results: The expression level of miR-378 in renal cancer tissues and cell lines was significantly up-regulated as compared with that in normal renal epithelial tissues or normal renal epithelial HK2 cells (all P < 0.001). After transfection with miR-378 mimic, the proliferation (P < 0.01) and migration (P < 0.001) of 786-O cells were remarkably increased. In contrast, after transfection with miR-378 inhibitors, the proliferation (P < 0.01) and migration (P < 0.001) of Caki-1 cells were remarkably suppressed. By bioinformatics assay, the large tumor suppressor 2 (LATS2) was identified as a target gene of miR-378 in renal cancer. The mRNA and protein expression of LATS2 were negatively related with the expression of miR-378 in renal cancer 786-O and Caki-1 cells (all P < 0.01). Conclusion: Overexpression of miR-378 may promote the proliferation and migration of renal cancer cells by negative targeting LATS2 gene.
