ABSTRACT

The aim of present study was to investigate the effect of Momordica cochinchinensis (Gag) aril (GA) aqueous extract on male reproductive system of streptozotocin (STZ)-induced hyperglycemia (HG) mice. GA were extracted with distilled water (DW) and analyzed for in vitro antioxidant capacities. ICR male mice were divided into 7 groups: 1) control, 2) DW, 3) GA 1000 mg/kg BW, 4) HG, 5) HG + glibenclamide, 6 and 7) HG + GA 500 and 1000 mg/kg BW respectively (7 mice/ group). In HG groups, mice were induced by STZ at single dose (150 mg/kg BW). They were treated for consecutive 35 days. All groups were compared for blood glucose levels, weights and histopathologies of reproductive organs, sperm concentration including testicular tyrosine phosphorylation protein patterns by Immuno-Western blotting. The results showed that GA processed antioxidant activities and could significantly decrease blood glucose levels and increase sperm concentration in HG mice. Moreover, GA could change the density of a testicular 70 kDa protein in HG-GA groups. In conclusion, GA extract could improve hyperglycemia and male reproductive damages in STZ-induced HG mice.


El objetivo de este estudio fue investigar el efecto del extracto acuoso de Momordica cochinchinensis (Gag) aril (GA) en el sistema reproductor masculino de ratones hiperglucémicos inducidos por estreptozotocina (STZ). GA fue extraída con agua destilada (DW) y se analizaron las capacidades antioxidantes in vitro. Ratones ICR machos fueron divididos en 7 grupos: 1) control, 2) DW, 3) GA 1000 mg / kg PC, 4) HG, 5) HG + glibenclamida, 6 y 7) HG + GA 500 y 1000 mg / kg PC, respectivamente (7 ratones / grupo). En los grupos HG, los ratones fueron inducidos con STZ en dosis única (150 mg / kg BW). Fueron tratados durante 35 días consecutivos. En todos los grupos se compararon los niveles de glucosa en sangre, los pesos y las histopatologías de los órganos reproductores, la concentración de espermatozoides, incluídos los patrones testiculares de proteínas tirosina fosforilada por Inmuno-Western blot. Los resultados mostraron que GA procesaba actividades antioxidantes y podían disminuir significativamente los niveles de glucosa en sangre y aumentar la concentración de espermatozoides en ratones HG. Además, GA podría cambiar la densidad de una proteína testicular de 70 kDa en grupos HG-GA. En conclusión, el extracto de GA podría mejorar la hiperglucemia y los daños reproductivos masculinos inducidos por STZ en ratones HG.

ABSTRACT

This study aimed to investigate protective effect of Momordica cochinchinensis (MC) aril extract on adverse reproductive parameters of male rat induced with valproic acid (VPA) commonly used in treatment for antiepileptic diseases. Male Wistar rats were divided into 6 groups (control, VPA, 200 mg/kg BW of PE only, and 50, 100, 200 mg/kg BW MC+VPA, respectively). Animals were pretreated with aqueous MC extract for 23 days before co-administered with VPA induction for 10 days. At the end of experiment, all male reproductive parameters and testicular histology were examined. The results showed all doses of PE significantly protect the decrease the weights of epididymis and seminal vesicle but not of body and testicular weights. MC extract also increased sperm concentration and seminiferous tubular diameters in MC+VPA co-administrative groups. Moreover, testicular histology of MC+VPA groups showed significant declining of testicular histopathologies as compared to VPA group. It was concluded that M. Cochinchinensis aril extract can prevent adverse male reproductive parameters and testicular damage induced with VPA.


El objetivo fue investigar el efecto protector del extracto de arilo de Momordica cochinchinensis (MC) sobre los parámetros reproductivos adversos de la rata macho inducida con ácido valproico (AV) que se utiliza comúnmente en el tratamiento de enfermedades epilépticas. Las ratas se dividieron en 6 grupos (control, AV, 200 mg/kg por peso corporal de PE solamente, y 50, 100, 200 mg/kg por peso corporal MC+AV, respectivamente). Los animales fueron tratados previamente con extracto acuoso MC durante 23 días, antes de la administración de AV durante 10 días. Al término del experimento, se examinaron todos los parámetros reproductivos masculinos y la histología testicular. Los resultados indicaron que todas las dosis de PE protegen de manera significativa la disminución de los pesos de epidídimo y vesículas seminales, pero no de peso corporal y testicular. El extracto de MC también aumentó la concentración de espermatozoides y los diámetros de los túbulos seminíferos en los grupos de administración con MC+AV. Por otra parte, la histología testicular de los grupos MC+AV mostró una disminución significativa de histopatologías testiculares en comparación con el grupo AV. En conclusión, el extracto de arilo M. cochinchinensis puede prevenir la aparición de parámetros reproductivos masculinos negativos y los daños testiculares inducidos con AV.

This study aimed to determine the antioxidant capacity of the Leucaena leucocephala aqueous shoot tips plus young leaves (LL-spl) extracts among three different fractions (LL-spl 10, 20, and 40 min) and to examine its acute toxicity on male reproductive parameters. The amount of the total phenolics in LL-spl extract was determined using a Folin-Ciocalteu reagent method and its antioxidant capacity was analyzed using 1, 1-diphenyl l-2-picrylhydrazyl radical scavenging and ferric reducing antioxidant powder methods. The LL-spl extract fraction with highest antioxidant capacity was used in animal treating. Male rats were divided into three groups (n= 5); control and groups treated with LL-spl 400 and 600 mg/Kg body weight for consecutive 40 days. The results showed that the LL-spl 40 min fraction possessed the highest antioxidant capacity. In addition, the LL-spl 400 and 600 groups showed no differences in weights of body, testis and epididymis, serum testosterone levels, and expression of testicular steroidogenic acute regulatory (StAR) protein. Significantly, LL-spl extract reduced the weight of seminal vesicle, sperm concentration, and seminiferous diameters compared with control. Moreover, LL-spl extract had adverse effect on testicular histology in inducing of seminiferous atrophy and degeneration including dilated blood vessels in interstitial tissue. It was concluded that although LL-spl extract possessing antioxidant capacity, in short term consumptions, it could be toxic to some male reproductive organs especially damaging testicular tissues.


El objetivo fue determinar la capacidad antioxidante del extracto de brotes acuosos con hojas nuevas de Leucaena leucocephala (LL-spl) en tres fracciones diferentes (LL-SPL 10, 20 y 40 min), además de examinar su toxicidad aguda sobre los parámetros reproductivos masculinos. Se determinó la cantidad de los fenoles totales en el extracto de LL-spl utilizando un método reactivo de Folin-Ciocalteu. La capacidad antioxidante se analizó por medio de 1-difenil-2-picrilhidracilo y/o métodos de reducción férrica de la capacidad antioxidante. La fracción de extracto LL-spl con mayor capacidad antioxidante fue utilizada en el tratamiento de los animales. Ratas macho fueron divididas en tres grupos (n= 5): el control y los grupos tratados con LL-spl 400 y 600 mg/kg peso corporal por 40 días consecutivos. El resultado mostró que la fracción LL-spl 40 min poseía la mayor capacidad antioxidante. Además, los grupos 400 y 600 LL-spl no mostraron diferencias según el peso corporal, testículos y epidídimo, niveles de testosterona y la expresión de proteínas testiculares. El extracto de LL-spl redujo de manera significativa el peso de la vesícula seminal, la concentración de espermatozoides y los diámetros de los túbulos seminíferos en comparación con el control. Por otra parte, el extracto de LL-spl tuvo un efecto adverso sobre la histología testicular por la inducción de atrofia y degeneración de los túbulos seminíferos, incluyendo a vasos sanguíneos dilatados en el tejido intersticial. Si bien el extracto LL-spl posee una capacidad antioxidante, ésta podría ser tóxica en el consumo a corto plazo para algunos órganos reproductores masculinos y especialmente dañino para los tejidos testiculares.

ABSTRACT

Valproic acid (VPA), widely used in treating epileptic patients, can damage reproductive parameters causing male infertility. This study aimed to investigate protective effect of Phyllanthus emblica L. branch (PE) extract on rat testicular damage induced with VPA. Male rats were divided into 6 groups (control, VPA, 250 mg/kgBW PE only, and 50, 100, 250 mg/kgBW PE+VPA, respectively). Animals were pretreated with PE for 23 days and co-administered with VPA for 10 days before all reproductive parameters were determined. The results showed all doses of PE significantly protected the decrease testicular weight and testosterone level in VPA rats. PE significantly improved the decrease sperm concentration in VPA treated rats. Moreover, testicular histology of PE+VPA groups showed declining of testicular histopathologies as compared to VPA group. Therefore, it seems that PE branch extract can prevent testicular damages including male reproductive parameters in rats induced with VPA.


El ácido valproico (AVP) es utilizado frecuentemente en el tratamiento de pacientes epilépticos y puede dañar los parámetros reproductivos que causan la infertilidad masculina. Este estudio tuvo como objetivo investigar el efecto protector de la rama Phyllanthus emblica L. (PE) sobre el daño testicular de ratas inducidas con AVP. Ratas machos fueron divididas en 6 grupos (control, AVP, PE 250 mg/kg peso corporal, APV+ PE 50, 100, 250 mg/kg peso corporal, respectivamente). Los animales fueron pretratados con PE durante 23 días y se administró AVP durante 10 días antes de medir todos los parámetros reproductivos. Los resultados mostraron que todas las dosis de PE protegen significativamente el peso y los niveles reducidos de testosterona testicular en ratas con AVP. El extracto de PE mejoró significativamente la concentración de espermatozoides en ratas tratadas con AVP. Por otra parte, la histología testicular de los grupos PE+AVP mostró disminución de la histopatología testicular en comparación con el grupo tratado sólo con AVP. Por lo tanto, parece que el extracto de la rama PE puede prevenir daños testiculares incluyendo los parámetros reproductores masculinos en ratas inducidas con AVP.

ABSTRACT

Kaempferia parviflora Wall. ex. Baker is a Thai herb containing many flavonoids that have anti-inflammatory, anti-allergic and antioxidant activities. The objective of this study was to demonstrate apoptotic effects of Kaempferia parviflora Wall. ex. Baker rhizome ethanolic extract on HL-60 cells in vitro. The extract suppressed HL-60 cell growth and decreased cell viability in a dose- and time-dependent manner. Apoptotic cell death was demonstrated by changes in cell morphology, externalization of phosphatidylserine on the cell surface, loss in mitochondrial transmembrane potential and activation of caspase 3. Apoptosis induced by K. parviflora Wall. ex. Baker rhizome ethanolic extract was enhanced by treatment with paclitaxel or doxorubicin, and inhibitors of Akt, PI3-K and MEK.

ABSTRACT

Objective: The aim of this study is to investigate the protective effect of Centella asiatica extract on gastric ulcers caused by indomethacin in rats. Methods: Male rats were orally given extract of C.asiatica prior to the administration of indomethacin. The stomach was dissected and the number of bleeding spots and histological features were determined. Results: Giving extract of C.asiatica at doses of 10 and 20 mg/kg for 30 minutes prior to oral administration of indomethacin decreased the number of bleeding spots are about 11 times lower than those given indomethacin alone. Histological findings of indomethacin-treated group showed the depth of injury of gastric mucosa which is about half the thickness of the mucosal layer. Treatment with C.asiatica decreased the depth of tissue erosion, down to one-third of the thickness of the mucosa. Conclusion: The results showed protective effect of C.asiatica on gastric ulceration caused by the anti-inflammatory drug, indomethacin.

Serum gamma-glytamyl transpeptidase (GGT) and alkaline phosphatase (ALP) of the normal population in Northeastern Thailand were measured. The median serum GGP and ALP levels of females was significantly lower than the values for males, i.e., 21 (range=9-260) versus 32 (range=9-459) and 73 (range= 20-428) versus 83 (range=13-171) U/L, respectively. Serum ALP level tended to increase with age in both male and female populations, whereas serum GGT level did not show any age dependence. Serum GGT and ALP levels of cigarette smokers were significantly higher than those of the non-smokers. Alcohol, coffee or tea drinking also resulted in higher serum GGT value whereas serum ALP values were not changed. Beta l nut chewing caused only lower serum GGT but not serum ALP values. The total population in Ban Fang district seemed to have lower serum GGT than those in Chonnabot district. The results from our study give the base line data of serum GGT and ALP in a Thai population. We also confirm the association of serum GGT with cigarette smoking, alcohol and coffee/tea drinking which are risk factors of cancer.

ABSTRACT

Three antioxidant vitamins, the alpha- and beta-carotenes as well as vitamin E, were measured in sera of a normal population in Northeastern Thailand using HPLC. The mean serum beta-carotene level of females was significantly higher than the value for males, i.e, 37.55 (95%CI=34.59-40.51) versus 32.97 (95% CI=30.01-35.93) micro/dl. The beta-carotene level tended to decrease as age increased, particularly in the male population. The mean serum beta-carotene level was also higher in females than in males, i.e., 7.08 (95%CI=6.57-7.59) and 6.26 (95% CI=5.77-6.75) micro/dl, respectively. The average serum alpha-tocopherol (Vitamin E) level of the whole population was 1.08 (95% CI=1.04-1.12) micro/dl and did not show age or sex differences. In general, the serum antioxidant vitamins of smokers were lower than those of the non-smokers but a significant difference was observed only for alpha-tocopherol. Alcohol drinking resulted in slightly lower serum beta-carotene values, whereas coffee or tea drinking and betel nut chewing did not cause any differences with these three antioxidant vitamins. However, we report higher in serum beta-carotene levels of people in Ban Fang district than in Chonnabot district. The results from our study give the base line data of serum antioxidant vitamins in a Thai population and also suggest future intensive study on the relationship of dietary intake and cancer prevention.
