ABSTRACT

【Objective】 To explore the effect and mechanism of Fasudil in the treatment of experimental autoimmune myocarditis (EAM) in mice so as to provide a theoretical basis for the clinical use of Fasudil in treating myocarditis. 【Methods】 Balb/c male mice were used as the research objects, and the EAM mice model was constructed using MyHC-α614-629 polypeptide. Mononuclear cells were isolated and cultured to detect the number of mononuclear cells in mouse spleen. Inflammation infiltration, fibrosis and IL-6 expression in mouse myocardial tissue were detected by HE staining, Masson staining and immunohistochemistry, respectively. The protein expressions of Notch1 and IL-6 were detected by Western blotting. qRT-PCR was used to detect the expressions of pro-inflammatory factors (IL-1α, IL-1β and IL-6) as well as key genes of TLRs and NOTCH signaling pathway. 【Results】 EAM mice showed increased HW, decreased BW, increased HW/e-BW, and increased inflammatory infiltration and fibrosis in myocardial tissue. The above-mentioned symptoms or pathological features were improved in EAM mice treated with Fasudil. The analysis showed that the pro-inflammatory factors IL-1α, IL-1β and IL-6 in the myocardial tissue of EAM mice were significantly increased, but only the expression of IL-6 was statistically different after Fasudil treatment compared with the control group. In addition, TLRs signaling pathway might also play an important role in the EAM mice treated with Fasudil. The expressions of IL-6 and Notch1 were consistent, and the expressions of the key genes of NOTCH signaling pathway (Notch1, Hes1 and Jag2) were down-regulated after Fasudil treatment. 【Conclusion】 Fasudil exerts a protective effect on down-regulation of IL-6 expression by inhibiting the NOTCH signaling pathway in EAM mice.

ABSTRACT

OBJECTIVE:To optimize the p reparation technology of citronellol submicroemulsion. METHODS :The content of citronellol in Citronellol submicroemulsion was determined by HPLC. Citronellol submicroemulsion by high-speed shearing dispersion-high pressure homogenization method ,with centrifugation stability constant (ke) and particle size were used as evaluation indexes. Its formulation and preparation technology were optimized and validated. Drug-loading amount and encapsulation rate of the preparation were detected. RESULTS :The linear range of citronellol were 4-64 μg/mL(R 2=0.999 9). RSDs of precision ,stability(24 h)and reproducibility tests were all lower than 3%. The recoveries were 97.64%-101.97%(RSD= 2.28%,n=3),97.71%-99.50%(RSD=1.29%,n=3),96.87%-101.48%(RSD=2.86%,n=3). The optimal formulation included that total weight of soybean oil and medium chain triglycerides (1 ∶ 1,g/g)was 3.75 g,1.2% soybean phospholipid was 0.6 g, cholesterol was 0.06 g,citronellol was 1.25 g,0.6 % sodium oleate was 0.3 g,15-hydroxystearic acid polyethylene glycol ester was 0.75 g,poloxamer 188 was 0.75 g,water added to 50 mL. After prepared by optimal technology at 4 ℃ which contained shearing speed of 13 000 r/min,lasting for 5 min, primary emulsion was adjusted to pH 7 with dilute hydro- chloric acid ,and homogenized with 600 Bar high pressure for 1434412440@qq.com 5 min. The parameters of Citronellol submicroemulsion accor- ding to optimal formulation and technology contained mean particle size of (91.05±0.26)nm,PDI of (0.20±0.01), Zeta-potential of (-30.86±0.39)mV,average content of 649511230@qq.com citronellol(100.21±0.01)%,the drug-loading amount was (2.481 7 ± 0.000 7) mg/mL,the encapsulation rate was (99.27 ± 0.03)% . CONCLUSIONS :The optimal formulation and technology is stable and feasible.

ABSTRACT

The bone is a common site of metastasis for gastric cancer. High-risk factors of metastatic gastric cancer include young age, poorly differentiated adenocarcinoma, Borrmann type III tumors, depth of invasion at Se and Pm levels, positive lymph nodes, and gastric cancer with concomitant body of stomach cancer. More than half of the transfer pathway belongs to the non-portal system. Cases of gastric cancer with bone metastasis but without liver metastasis accounted for 69%of the total gastric cancer cases, whereas the incidence rate of bone metastasis with lymph node metastasis (≥3 cm away from the primary lesion) was 27%. In addition to the tumor markers human chorionic gonadotropin and carcinoembryonic antigen, radionuclide scan of the bone metastasis provides diagnostic pathways and reliable basis for treatment. Chemoradiation and surgery as symptomatic treatments are alternative therapies for stomach cancer. Bone metastasis confers unfavorable prognosis, and the survival time is often less than six months.

ABSTRACT

BACKGROUND:Currently, there is no uniform, standardized approach to isolate, purify and proliferate bone marrow mesenchymal stem cells. Chlormethylbenzamido-1,1'-dioctadecyl-3,3,3',3'-tetramethylindocarbocyanine (CM-Dil) is a stable, reliable, high marking and simple marker. OBJECTIVE:To develop the methods for isolation, culture and identification of rat bone marrow mesenchymal stem cells in vitro. METHODS:Two male Sprague-Dawley rats, weighing 50-100 g were taken to col ect the bilateral femur and tibia bone marrow under sterile conditions, and then, primary bone marrow mesenchymal stem cells were isolated and cultured using bone marrow adherent separation and density gradient centrifugation. cells were amplified and purified through timely and repeated passage, and labeled at the third generation with fluorescent dyes CM-Dil in vitro as a source of donor cells. RESULTS AND CONCLUSION:Bone marrow mesenchymal stem cells were cultured successful y in vitro using bone marrow adherent separation and density gradient centrifugation separation methods, but the former was superior to the latter in the number of cultured cells significantly, while the two methods were not different significantly in terms of cellviability and proliferation. Flow cytometry results showed that the positive rates of cultured cells were 17.5%for CD34, 97.9%for CD44, and 91%for CD90. CM-Dil can label bone marrow mesenchymal stem cells successful y, which is a stable, reliable, high marking and simple marker.

OBJECTIVE@#To study the expression and significance of TNF-alpha, MMP-9 and their relationship with the infiltration of eosinophil granulocyte in nasal polyps.@*METHOD@#The expression of TNF-alpha and MMP-9 was determined in tissues of nasal polyps from 30 patients(nasal polyps group) and in inferior turbinate mucosa tissues from 10 patients(control group) by in situ hybridization and immunohistochemical technique, and the number of eosinophil granulocyte was counted in the same tissue by HE staining. Their correlations with each other were also analyzed in the tissue of nasal polyps.@*RESULT@#The number of TNF-alpha and MMP-9 positive cells and TNF-alpha positive blood vessels in nasal polyps were more than that in control group (P < 0.05). The number of both TNF-alpha positive cells and blood vessels had positive relationships with the number of eosinophil granulocyte, but there was only positive relationship between the number of MMP-9 positive cells and eosinophil granulocyte (P < 0.05). At the same time there was a positive relationship between the number of TNF-alpha and MMP-9 positive cells (P < 0.05).@*CONCLUSION@#TNF-alpha and MMP-9 may play an important role in the pathological mechanism of nasal polyps. TNF-alpha may induce the expression of MMP-9 and promote the migration of eosinophil granulocyte.

OBJECTIVE@#To explore the expression of cyclooxygenase-2 (COX-2) and S-100 positive dendritic cell in laryngeal carcinoma tissue and their clinical significance.@*METHOD@#Sixty-five samples of laryngeal carcinoma and thirty-four biopsies of adjacent noncancerous tissue were obtained. Immunohistochemical technique (SP method) was used to detect the expression of COX-2 and S-100 positive dendritic cell, and the relationship of their expression with clinical pathological parameters and prognosis was analyzed.@*RESULT@#The rates of COX-2 expression were 63.08% (41/65)and 14.70% (5/34) in laryngeal carcinoma and control group, respectively. The difference was significant (P < 0.05). The positive expression of COX-2 was correlated with T and clinical stage in laryngeal carcinoma(all P < 0.05). The rates of S-100 positive dendritic cell expression were 61.54% (40/65)and 0 in laryngeal carcinoma and control group, respectively. The difference was significant (P < 0.05). S-100 positive dendritic cells showed significant differences between early and late clinical stage and lymph node metastasis (all P < 0.05). Kaplan-Meier survival analysis showed that patients with positive expression of COX-2 and S-100 positive dendritic cell had worse disease-free and overall survival (P < 0.05). Cox regression analysis showed that S-100 positive dendritic cell was indicated as an independent prognostic factor for survival(P < 0.05).@*CONCLUSION@#COX-2 and S-100 positive dendritic cell are highly expressed in laryngeal carcinoma tissue. It suggests that the expression of COX-2 and S-100 positive dendritic cell is related to the process of carcinogenesis and may be the important indicators in laryngeal carcinoma for prognosis.

ABSTRACT

OBJECTIVE@#To explore the discrepancy of HGF and Hpa expressed in laryngeal squamous cell carcinoma of different pathology grades (differentiation of high, moderate or poor), which can provide the new thought and theory evidence for clinical treatment and control metastasis or palindromic.@*METHOD@#Forty paraffin-embedded samples of laryngeal squamous cell carcinoma, forty paraffin-embedded samples of laryngeal papilloma and twelve paraffin-embedded samples of normal tissue adjacent to cancerous location of our department between May 2004 and April 2005, which were analyzed with SP immunohistochemistry staining.@*RESULT@#The positive rate of HGF were 77.5% and that of Hpa were 72.5% in laryngeal squamous cell carcinoma, which have positive correlation and were higher than that in laryngeal papilloma and normal tissue adjacent to cancerous location with significant statistical difference (P0.05), that have positive correlation with clinical stages and have negative correlation with the level of cellular differentiation. The expression of HGF and Hpa in lymph node metastasis group was significantly higher than the non-metastasis group (P<0.05).@*CONCLUSION@#The expression of HGF and Hpa in laryngeal squamous cell carcinoma are higher than that in nonmalignant ones. They correlates with lymph node metastasis, clinical stages and pathological grading, but they has no relationship with the primary locations. The positive correlation relationship of expression between HGF and Hpa might be worthiness to determine the prognosis of laryngeal squamous cell carcinoma.

Cox regression analysis indicated that Ang-2 and MMP-7 expression were independent prognostic factors of laryngeal carcinoma.@*CONCLUSION@#Overexpression of Ang-2 and MMP-7 was observed in laryngeal carcinoma and they might be served as an objective indicator for biological behaviour and prognosis.

ABSTRACT

Objective:To investigate the expression and clinical significance of Ang-2 and MMP-7 protein in laryngeal carcinoma tissue.Method:Immunohistochemical technique was used to detect the expression of Ang-2 and MMP-7 protein in 65 tissues of laryngeal carcinoma and 34 biopsies of adjacent non-cancerous tissue. The relationship between the expression of Ang-2 and MMP-7 and invasion, metastasis or prognosis in laryngeal carcinoma tissue was analyzed.Result:The positive rates of Ang-2 and MMP-7 were significantly higher in laryngeal carcinoma tissue than those in adjacent non-cancerous tissue(P<0.05).The level of Ang-2 and MMP-7 expression had no significant correlations with the age and course as well as the smoking, drinking, histological differentiation of carcinoma and clinical classification (P>0.05).While the expression of Ang-2 significantly differed between patients with different T stage and clinical stage(P<0.05), and the expression of MMP-7 was notably correlated with the T stage,clinical stage and lymph node metastases (P<0.05).There was a correlation between the expression of Ang-2 and MMP-7 (P<0.05).The Kaplan-Meier survival analysis showed that patients with positive expression of Ang-2 had worse overall survival(P<0.05).However,MMP-7 expression was not related to overall survival or disease-free survival (all P>0.05).Cox regression analysis indicated that Ang-2 and MMP-7 expression were independent prognostic factors of laryngeal carcinoma.Conclusion:Overexpression of Ang-2 and MMP-7 was observed in laryngeal carcinoma and they might be served as an objective indicator for biological behaviour and prognosis.

ABSTRACT

OBJECTIVE@#To investigate the features of nasal septum cellule in computed tomographic (CT) images and its clinical significance.@*METHOD@#CT scans data of nasal septum in 173 patients were randomly obtained from January 2001 to June 2005. Prevalence and clinical features were summarized in the data of 19 patients with nasal septum cellule retrospectively.@*RESULT@#(1) Nineteen cases with nasal septum cellule were found in 173 patients. (2) All nasal septum cellule of 19 cases located in perpendicular plate of the ethmoid bone, in which 8 cases located in upper part of nasal septum and 11 located in middle. (3) There were totally seven patients with nasal diseases related to nasal septum cellule, in which 3 cases with inflammation, 2 cases with bone fracture, 1 case with cholesterol granuloma, 1 case with mucocele.@*CONCLUSION@#Nasal septum cellule is an anatomic variation of nasal septum bone, and its features can provide further understanding of some diseases related to nasal septum cellule.

ABSTRACT

OBJECTIVE@#In order to meet the needs of electrophysiology outer hair cells, we investigate a method to separate outer hair cells simply and effectively.@*METHOD@#The basal membrane was dissected combined with spiral ligament and axis of cochlea, then incubated with enzyme, and then mechanically triturated by micropipette.@*RESULT@#A large number of living outer hair cells were obtained. Of 80% cells could keep good condition in 6 hours.@*CONCLUSION@#If increasing the enzyme concentration slightly and the large part of cochlea without outer bone was enzymes, we can get living outer hair cells easily.

ABSTRACT

OBJECTIVE@#To investigate the degeneration mechanics of outer hair cells in guinea pig.@*METHOD@#The mechanics of outer hair cells isolated by enzyme were observed under inverted microscope for 6-8 h continuously.@*RESULT@#Over half of living outer hair cells could keep good conditions in 6 hours. During the degeneration there was always a longitudinal fold line from tip to base. Presence or absence of calcium, as well as lossing of stereociliary bundle, couldn't change the conditions of out hair cells.@*CONCLUSION@#Neither calcium nor stereociliary bundle is the decisive cause in keeping outer hair cells alive, and its degeneration may be basically related with something surrounding the cell.

OBJECTIVE To study the expression of c-Met,TIMP-2 in laryngeal squamous cell carcinoma and to explore the function and relationship of c-Met,TIMP-2 in the invasion and metastasis of laryngeal squamous cell carcinoma. METHODS Immunohistochemical method(SP) was used to detect the expression of c-Met,TIMP-2 in 41 laryngeal squamous cell carcinoma specimens and 15 specimens adjacent to the tumors.RESULTS The positive rate of c-Met and TIMP-2 in laryngeal squamous cell carcinoma specimens was 56.1% (23/41)and 63.4%(26/41)respectively.The positive rate of c-Met in tissues adjacent to tumors was 13.3%(2/15).No positive expression of TIMP-2 was found in tissues adjacent to tumors.CONCLUSION The expression of c-Met showed a positive correlation with histological differentiation and cervical lymph node metastasis and TIMP-2 showed a positive correlation with cervical lymph node metastasis in laryngeal squamous cell carcinoma.There was no significant relation between the expression of c-Met and TIMP-2 in laryngeal squamous cell carcinoma.

ABSTRACT

Objective To investigate the effects of hydrogen peroxide on function and morphology of guinea pig cochlea in vivo.Methods Animals were divided into four groups and their cochleas were perfused with artificial perilymph(AP), 50 ?M H 2O 2, 100 ?M H 2O 2 and 200 ?M H 2O 2 resolved in AP separately. The compound action potential(AP) and cochlear microphonic(CM) were recorded after two-hours' perfusion. Then hydrogen peroxide-induced cell damage in the inner ear was investigated with morphologic method.Results In all H 2O 2 groups, CAP threshold shifts and CM amplitude shifts were significantly greater than that of APL group. The effect of H 2O 2 perfusion on cochlear function showed dose-dependence. OHCs were major targets of H 2O 2-induced cell death, while Hensen's cells did not show any signs of damage in the presence of H 2O 2. Conclusion Hydrogen peroxide, as an ubiquitous reactive oxygen species(ROS), can induce cochlea dysfunction and damage of cells in guinea pig inner ear. Hensen's cells maybe more resistant to the damage of ROS than hair cells.

Objective To investigate the antagonist effects of sodium salicylate(NaSA) on the cisplatin(CDDP) induced hearing impairment in guinea pigs.Methods 60 guinea pigs were randomly divided into four groups: ①CDDP +NaSA(50 mg/kg) group,②CDDP +NaSA(100 mg/kg) group,③CDDP +NaSA(150 mg/kg) group and ④CDDP +NS(normal saline) control group.Auditory brainstem response(ABR) and distortion product otoacoustic emission(DPOAE) were used to evaluate their effects on hearing threshold and DPOAE amplitudes.Results The ABR responses of groupsⅠ,Ⅱ and Ⅲ were significantly lower than in the control group(P0.05).The ABR responses for group Ⅰ were significantly higher than group Ⅲ(P

ABSTRACT

DICOM medical data may be blocked by firewalls between terminals when DICOM3.0 Standard is applied in transmission via Internet. Since most of the firewalls are opened for emails, this paper introduces a new method that uses both email and DICOM3.0 protocols to transmit medical images via Internet.
