ABSTRACT
Objective: To study the chemical constituents in the roots of Aconitum iochanicum Ulbr.Methods: The air-dried roots of A.iochanicum were powdered and extracted by methanol with a percolation method.After removing the solvent under reduced pressure, the crude extract was dissolved in1.5% HCl solution, and then extracted by ecetic ether.The acidic solution was basified to pH 9.0 by NaOH (5%) and extracted with ethyl acetate to obtain crude alkaloidal extract after the removal of ethyl acetate.The compounds were isolated and purified by column chromatography and identified based on spectral analysis (1H-NMR, 13C-NMR and MS).Results: Totally 18 compounds were isolated from A.iochanicum and characterized as 14-O-acetylsachaconitine (1), franchetine (2), crassicaudine (3), indaconoitine (4), 14-benzoyl chasmanine (5), 14-O-acetyltalatisamine (6), talatisamine (7), chasmannine (8), crassicauline A (9), bikhaconine (10), 13,15-dideoxyaconitine (11), crassicautine (12), kongboensine (13), liljestrandisine (14), ludaconitine (15), 8-deacetyl-yunaconitine (16), yunaconitine (17) and ouvrardiantine (18).Conclusion: It''s the first time to study the chemical constituents of A.iochanicum, and 18 diterpenoid alkaloids are isolated.
ABSTRACT
Objective To detect the expressions of two protein factors hypoxia-inducible factor (HIF)-1α and Helicobacter pylori (Hp) in gastric cancer in Mongolian and Han two nationalities respectively,and to explore the relationship of HIF-1α and Hp in the development of gastric cancer.Methods A modified Warthin-Starry (W-S) silver staining method was used to detect Hp infection in 27 Mongolian patients with gastric cancer and 30 Han patients with gastric cancer.Immunohistochemical (S-P) method and reverse transcription polymerase chain reaction (RT-PCR) method were used to detect protein and mRNA expressions of HIF-1α in cancer tissue,paracancerous tissue and normal tissue samples.The relationship of HIF-1α protein and Hp expression was analyzed between gastric carcinoma tissue samples of Mongolian patient group and Han patient group.Results Hp infection rates were 81.5％ (22/27) and 66.7％ (20/30) in Mongolian and Han patients with gastric cancer.The difference in infection rate was not statistically significant between two groups of patients (~=1.608,P ＞ 0.05).The positive expression levels of HIF-1α protein were 62.96％ (17/27) and 70.00％ (21/30) in gastric cancer tissue samples of Mongolian and Han groups.The relative expression levels of HIF-1α mRNA were 0.891 4±0.075 0 and 0.907 4±0.065 5.There was no significant difference in the HIF-1 α mRNA expression between the two nationalities.No positive expressions of HIF-1α protein and mRNA were found in paracancerous tissue and normal tissue samples.There was a positive correlation in the positive expression of HIF-1α protein and Hp infection in gastric cancer tissue samples of Mongolian and Han patient groups (r,=0.424 and 0.617,P ＜ 0.05).Conclusion HIF-1α and Hp are pathogenic factors of gastric cancer.Hp and HIF-1α may promote the occurrence of gastric cancer together.The pathogenic mechanism of HIF-1α and Hp in gastric cancer may be the same for Mongolian and Han patients.

After removing the solvent under reduced pressure, the crude extract was dissolved in 1. 5％ HCl solution, and then basified to pH 9 by NaOH (5％) and extracted by ethyl acetate to ob-tain crude alkaloidal extract after ethyl acetate removal. The alkaloidal extract was isolated and purified by column chromatography, and their structures were identified based on spectral analysis ( 1 H-NMR, 13 C-NMR, MS) . Results:Totally 12 diterpenoid alkaloids were isolated from A. bulleyanum and characterized as foresaconitine (1), crassicauline A (2), chasmanine (3), talatisamine (4), 14-debenzoyl-franchetine (5), pengshenine A (6), crassicautine (7), yunaconitine (8), franchetine (9), liljestrandisine (10), transconitine B (11) and pseudoaconine (12). Conclusion:Compounds 3-7, 10-12 are isolated from this plant for the first time.

Objective To investigate the relationship between thyroid hormone and uric acid (UA) and body mass index (BMI) in patients with type Ⅱ diabetes and normal thyroid function.Methods Total of 313 patients with type Ⅱ diabetes and normal thyroid function were selected.BMI,fasting blood glucose (FBG),the metabolism of blood lipid,thyroid hormones and UA indicators were examined and the correlations of thyroid hormone,BMI and UA were analyzed.Results (1) The patients were divided into two groups according to gender,and FT3,FT4,and UA of male were found to be significantly higher than those in female (P＜0.01).TSH,SBP,HDL-C in female were significantly higher than those in male (P＜0.01);(2) The patients were divided into three groups according to BMI Level.Thyroid-stimulating hormone (TSH),three iodine armour gland original glycine (TT3),free three iodine thyroid glycine (FT3),UA,and FBG in overweight and obesity groups were found to be higher than those in normal weight group (P ＜ 0.05);(3) The patients were divided into two groups according to the TSH level.Serum uric acid,TT3,FT3,fasting insulin in the group with TSH above 2.5 uIU/L were found to be higher than those in the group with TSH under 2.5 uIU/L (P ＜ 0.05);(4) Patients were divided into two groups according to the UA level.TSH,FT3 in male with high uric acid were found to be higher than those in male with normal uric acid (P ＜ 0.05);TSH was in female with high uric acid was found to be higher than that in female with normal uric acid (P ＜ 0.05).Conclusion Thyroid hormone in patients with type Ⅱ diabetes can be used to assess the body weight and uric acid,which is of great clinical importance.

Objective To investigate the effect and mechanism of AngⅡ on collagen in hepatic stellate cell. Methods HSCs were isolated and cultured, 3H-pro incorporation method was used to evaluate the effects of different doses of AngⅡ on the proline syntheses. RT-PCR assay were used to assess changes in mRNA expression levels of type Ⅰ and Ⅲ procollagen. PDGFR-β mRNA and protein were determined by in situ hybridization and immunocytochemistry. Results 10-8~ 10-5 mol/L AngⅡ could significantly increase the 3H-pro incorporation rate of HSC in a dose-dependent style, 10-6 mol/L AngⅡis the most effective dose. The cultured HSC showed a little expression of type Ⅰ and Ⅲ procollagen mRNAs, while 10-6 mol/L AngⅡwas able to enhance the expression for type Ⅰ and Ⅲ procollagen mRNAs significantly(P < 0.01). AngⅡalso could enhance both mRNA and protein expression of PDGFR-β on HSC(P < 0.01). Conclusion These results suggest that AngⅡ could promote HSC collagen synthesis by enhancing the expressions of PDGFR-β.

Objective To study the effect of rosuvastatin(RSVT) on the vascular endothelial function(VEF) ,inflammatory fac‐tors(IF) and prognosis in patients with acute coronary syndrome (ACS) after percutaneous coronary intervention(PCI) .Methods Eighty cases of ACS patients with PCI in our hospital selected from July 2010 to July 2013 were randomly divided into observed group and control group ,40 cases in each group .The observation group were given interference treatment with RSVT while the control group received conventional treatment .The VEF ,IF and prognosis were compared between groups .Results The VWF at 4 weeks after PCI in observation group was lower than that in control group [(92 .6 ± 12 .3)% vs .(105 .4 ± 13 .6)% ,P<0 .05];The ET‐1 at 4 weeks after PCI in observation group was lower than that in control group[(55 .6 ± 5 .6)ng/L vs .(67 .8 ± 7 .4)ng/L ,P<0 .05] .The NO at 4 weeks after PCI in observation group was higher than that of control group[(78 .6 ± 9 .4)μmol/L vs .(63 .2 ± 9 .5)μmol/L ,P<0 .05] .The CRP at 4 weeks after PCI in observation group was lower than that in control group[(5 .4 ± 2 .2) mg/L vs .(10 .5 ± 4 .1)mg/L ,P<0 .05] .The incidence of CVE was 5 .00% (2/40) ,restenosis rate was 2 .50% (1/40) in observa‐tion group ,which was significantly lower than those of the control group [30 .00% (12/40) ,10 .00% (4/40)] ,the differences were statistically significant(P<0 .05) .Conclusion RSVT could effectively improve the VEF and reduce inflammation ,CVE and rest‐enosis rate in patients with ACS after PCI .

Objective To formulate a scientific and reasonable evaluation model of post competency ,and to provide references for selection ,training and evaluation of orthodontic assistant specialist in china .Methods Items of competency for orthodontic as‐sistant specialist were established basing on semi structured interviewing and literature research ,and then Delphi technique was used to consult 22 orthodontic experts .The collected data was analyzed by SPSS 19 .0 .Results The positive coefficient for two round specialist survey was 95 .45% after two round specialist survey ,the authority coefficient was 0 .865 ,as well as the values of significant test for coordinate coefficient opinions were P<0 .01 .The evaluation competency model for orthodontic assistant special‐ist was consisted of entry requirements and evaluation system including 4primary indicators ,15secondary indicators .Conclusion The evaluation competency model is reliable and valid ,which can provide reference for selection ,training and evaluation of ortho‐dontic assistant specialist .

Objective The research aimed to identify symptom clusters of head and neck cancer (HNC) patients.Methods The M.D.Anderson Symptom Inventory-Head & Neck (MDASI-H&N) was applied to measure symptoms of 247 HNC inpatients.Factor analysis was applied to identify symptom clusters.The Spearman correlation analysis was used to find the relationship of symptoms within symptom clusters.Results Five clusters were identified and the Cronbach's α coefficients of symptom clusters were 0.904,0.928,0.898,0.815 and 0.662 respectively.Symptoms within clusters were all significantly interrelated to each other (r=0.393-0.856,P＜0.01).Conclusions Symptoms of HNC patients tend to occur not in isolation,but in symptom clusters.Effective management can not only eliminate or alleviate the symptom burden of HNC patients,but also be cost-effective.

BACKGROUND: It remains unclear whether ectomesenchymal cells also derived from neural crest stem cell in mammals.OBJECTIVE: To understand the specific markers and differentiation directions of maxillofacial and mandibular processes progenitor cells,and to explore the source and differentiation phenotype of ectomesenchymal stem cells.METHODS: The expression and changes of expression profiles of rat ectomesenchymal cells at E9.5,E10.5,E11.5,and E12.5days were observed by immunohistochemistry and flow cytometry.RESULTS AND CONCLUSION: The progenitors expressed multi-lineage markers,including neural system and several rnesenchymal tissue types,importantly the facts that molecule profiles were changed with time prolonged,suggesting these progenitors were in active differentiating stage,so they were stem like cells or contain stem like cells.Moreover,small populations(2%-3%)of CD57 and P75 phenotypes were detected by flow cytornetry,suggesting that ectomesenchymal stem cells were derived from neural crest,which maintained a quantitative stabilization though it is gradually differentiate after localization.

Objective To observe the effect of Jiangzhi Yigan Chongji (JYC) on the nonalcoholic fatty liver tissue PPAR? and Trx mRNA expression,and explore the mechanism of treating fatty liver. Methods The model was made by feeding high-fat diet and the rats were divide into 3 groups:normal group,model group and treated group. Result Expression of liver tissue PPAR? and Trx mRNA in the model group were both decreased. JYC can increase their expression of liver tissue of model rats. Conclusion It is likely to be one of the important mechanisms for JYC in treating nonalcoholic fatty liver.

@#ObjectiveTo investigate clinical features, correlative factors and prognosis of senile coronary heart disease accompanied with depression.MethodsFifty two senile patients of coronary hart disease with depression were selected for the observation group, sixty cases without depression as control group. Clinical features, family conditions, society factors and Holter report (supervision) of two groups were analyzed.ResultsClinical features such as severe dyssomnia, anxiety agitation, emotion depressing, loss of interest, feeling waste and sense of guilty in patients of observation group were significant different with those in control group. The rate of family harmoniousness and society supporting between patients of two groups were significantly different. The incidence of ventricular arrhythmias, atrial arrhythmia and myocardial ischemia changes on ST-T in the Holter reports of patients in observation group were higher than those in control group (P<0.05 or P<0.01). The incidence of cardiac sudden death of patients in observation group was also higher than patients in control group (P<0.05).ConclusionClinical symptoms such as severs dyssomnia, anxiety agitation, emotion depressing are easily occurred in senile coronary heart disease patients with depression. Patients with depression have higher incidence of cardiac sudden death, having better relationship and society support are advantageous to prevent and reduce the depression in senile coronary heart disease patients.

Objective: To investigate the diagnose nutcracker syndrome with color Doppler flow imaging.Methods:Doppler sonographic findings in 47 children with orthostatic proteinuria or idiopathic hematuria and in 23 healthy control subjects were compared.The peak velocity,anteroposterior diameter and anteroposterior diameter ratio(hilar segment/aortomesenteric segment) were measured in the left renal vein.Results: The anteroposterior diameter ratio of the 2 segments were 4.13?2.34 in the patient group and 2.16?0.56 in the control.The anteroposterior diameter and the peak velocity in the aortomesenteric segment were(0.16?0.07)cm/s and(83.3?37.2)cm/s in the patient group and(0.24?0.09)cm/s and(43.1?10.8)cm/s in the control,with significant difference in between(P

Objective:The purpose of this study is to locate and characterize splenic trauma with contrast-enhanced ultrasound(CEUS).Methods:A defined mild intraparenchymal lesion was done in 2 pigs.Ultrasound features after trauma were observed by baseline ultrasound,color Doppler flow images(CDFI) and CEUS with a contrast agent(Sono Vue),respectively.Results:In 2 pigs,focal intraparenchymal lesions on spleen with diameters ranging from 10.0 to 10.0 mm could be identified by CDFI and CEUS but not by the baseline ultrasound.Hyperechoic rift in spleen was visible along trauma area as enhancement in splenic parenchyma disappeared on CEUS.Hyperechoic line could be kept in a long time.The hyperechoic rift also appeared at the sub-capsular region beside a traumatic lesion.Hyperechoic rift in traumatic region could be seen even as CEUS was converted to conventional B-mode.Enhancement in the traumatic lesion was clearly seen in CDFI.Conclusion:Hyperechoic rift in spleen was visible along trauma area as a contrast agent is used.CEUS is useful in identifying the grade,range and location of splenic injury.

Objective: To construct a high effective eukaryotic expre ss ing vector containing mcpr1 gene. Methods: mcpr1 gene w as amplified by PCR from the plasmid T-easy/ mcpr1, then PCR product was in serted into eukaryotic expressing vector pcDNA3.1/V5-His B. The positive recomb inant was identified by PCR analysis, HindIII and BamHI restriction analysis and Sequence analysis. Results: A 400 bp DNA fragment was amplified from the recombinant. Sequence analysis and restriction digest demonstrated tha t the mcpr1 gene was successfully inserted into pcDNA3.1/V5-His B plasmid. Conclusion: The eukaryotic expressed vector pcDNA3.1/V5-His B/ mcpr1 has been successfully reconstructed.
