ABSTRACT

【Objective】 To investigate the transfusion effect of 0.5-dose of apheresis platelet in pediatric patients. 【Methods】 A total of 195 children who underwent 0.5-dose platelet transfusion from August 2021 to June 2022 were enrolled, and the platelet count within 24 hours before and after platelet transfusion were recorded. They were grouped by gender, disease type, blood product transfusion history, platelet antibody results, platelet storage time and weight to analyze the effect of platelet transfusion. 【Results】 Among 195 cases, 77.4% (151/195) were effective and 22.6% (44/195) were ineffective after 0.5-dose of platelet transfusion. Platelet transfusion more than three times has significant impact on the effectiveness of platelet transfusion, with platelet transfusion efficiency decreased from 80.8% to 65.9% (P<0.05), and CCI decreased from 11.56±8.94 to 8.52±8.42 (P<0.05). The transfusion effect of the HLA and HPA antibodies positive group was significantly lower than the negative group, with CCI decreased from 11.39±8.87 to 7.82±8.59 (P=0.05). Linear regression analysis showed that the effect of platelet transfusion decreased with the increasing of platelet storage time (P<0.05), and the effect of platelet transfusion decreased in children weighing less than 20 kg compared with those weighing more than 20 kg, with the effective rate decreased from 84.1% to 63.5% (P<0.05). Different gender, disease type and the number of red blood cell transfusions had no significant effect on platelet transfusion. 【Conclusion】 The 0.5-dose platelet transfusion has good therapeutic effect in children below 20 kg. The results of HLA and HPA antibodies and the number of platelet transfusions greatly influence the effect of platelet transfusion in children, and the transfusion effect decreases with the increase of platelet storage time.

ABSTRACT

OBJECTIVE@#To explore the feasibility and accuracy of ultrasound volume navigation (UVN) combined with X-ray fluoroscopy-guided percutaneous pedicle screw implantation through a prospective randomized controlled study.@*METHODS@#Patients with thoracic and lumbar vertebral fractures scheduled for percutaneous pedicle screw fixation between January 2022 and January 2023 were enrolled. Among them, 60 patients met the selection criteria and were included in the study. There were 28 males and 32 females, with an average age of 49.5 years (range, 29-60 years). The cause of injury included 20 cases of traffic accidents, 21 cases of falls, 17 cases of slips, and 2 cases of heavy object impact. The interval from injury to hospital admission ranged from 1 to 5 days (mean, 1.57 days). The fracture located at T 12 in 15 cases, L 1 in 20 cases, L 2 in 19 cases, and L 3 in 6 cases. The study used each patient as their own control, randomly guiding pedicle screw implantation using UVN combined with X-ray fluoroscopy on one side of the vertebral body and the adjacent segment (trial group), while the other side was implanted under X-ray fluoroscopy (control group). A total of 4 screws and 2 rods were implanted in each patient. The implantation time and fluoroscopy frequency during implantation of each screw, angle deviation and distance deviation between actual and preoperative planned trajectory by imaging examination, and the occurrence of zygapophysial joint invasion were recorded.@*RESULTS@#In terms of screw implantation time, fluoroscopy frequency, angle deviation, distance deviation, and incidence of zygapophysial joint invasion, the trial group showed superior results compared to the control group, and the differences were significant ( P<0.05).@*CONCLUSION@#UVN combined with X-ray fluoroscopy-guided percutaneous pedicle screw implantation can yreduce screw implantation time, adjust dynamically, reduce operational difficulty, and reduce radiation damage.

ABSTRACT

Ulcerative colitis (UC) is a chronic recurrent inflammatory bowel disease which primarily affects the colonic mucosa. The UC patients mainly present diarrhea, abdominal pain, tenesmus, and mucous bloody stools, and even malnutrition and systemic symptoms in severe cases, with rising incidence, which has a significant impact on the health and quality of life of patients. The pathogenesis of UC is not clear, and the Western medical therapies include sulfasalazine, glucocorticoids, and immunosuppressants, which, however, have side effects and unsatisfactory effects. Chinese medicine with high safety, mild adverse reactions, and a multi-component, multi-target, and multi-pathway treatment manner has garnering increasing attention. Therefore, finding the Chinese medicine to treat UC has become a hot spot. Glycyrrhizae Radix et Rhizoma is one of the commonly used Chinese herbal medicines, with the effects of tonifying spleen and reinforcing qi, clearing heat and detoxifying, dispelling phlegm and relieving cough, relieving pain, and harmonizing medicines. Glycyrrhizae Radix et Rhizoma mainly contains glycyrrhizic acid, glycyrrhetinic acid, diammonium glycyrrhizinate and other active ingredients. Modern pharmacological studies have shown that Glycyrrhizae Radix et Rhizoma has anti-inflammatory, antioxidant, antimicrobial, and immunomodulatory activities. According to statistics, Glycyrrhizae Radix et Rhizoma is among the top three Chinese herbal medicines for the treatment of UC. The recent years have witnessed progress in the treatment of UC with Glycyrrhizae Radix et Rhizoma and the related prescriptions. The present study summarized the mechanisms underlying the anti-inflammatory, immunomodulatory, intestinal flora-regulating, cell apoptosis-inducing, and oxidative stress-reducing effects of the key chemical constituents (glycyrrhetinic acid, diammonium glycyrrhizinate, polysaccharide, glycyrrhetinic acid, and isoglycyrrhizin) and compound prescriptions of Glycyrrhizae Radix et Rhizoma. The findings provide a solid foundation for further development and clinical application of Glycyrrhizae Radix et Rhizoma.

ABSTRACT

Objective:The purpose of this study was to investigate the association of insomnia with social factors, vascular risk factors and neurological status in patients with cerebral infarction.Methods:217 patients with first onset cerebral infarction in Army Medical Center of PLA were enrolled in this study. According to the athens insomnia scale (AIS) score, the patients were divided into cerebral infarction insomnia group (total score >6 points) and cerebral infarction non insomnia group (total score ≤6 points). The correlation between the occurrence of insomnia in patients with cerebral infarction and humanistic factors, vascular risk factors and cerebral infarction characteristics was analyzed.Results:The incidence of insomnia in patients with cerebral infarction was 61.8%(134/217). There were significant differences in age, education level, monthly income, occupational status, depression, hypertension, diabetes mellitus, vascular control area, infarction site, National Institutes of Health Stroke Scale (NIHSS) score, activities of daily living [Barthel Index (BI) score] and aphasia between insomnia group and non insomnia group ( P<0.05); Multivariate logistic regression analysis showed that age, depression, NIHSS score, BI score and aphasia were independently associated with insomnia after cerebral infarction ( P<0.05). Conclusions:Age, depression, NIHSS score, BI score and aphasia were independent risk factors for insomnia in patients with cerebral infarction.

ABSTRACT

Objective:To explore whether autophagy is involved in dysfunction of vascular endothelial induced by sodium arsenite (NaAsO 2). Methods:Human primary umbilical vein endothelial cells were isolated and cultured. The cells were treated with different levels of NaAsO 2 [0 (control)), 2, 5, 10, 20, 30, 50 μmol/L] for 24 h, and cell viability was determined using CCK8. According to the results of CCK8, the levels of arsenite used in subsequent experiments were determined, intracellular nitric oxide (NO) content (incubated by NaAsO 2 for 4 h) was detected by flow cytometry, LC3 levels (incubated by NaAsO 2 for 0, 6, 12 and 24 h) was detected using Western blotting, and autophagosome (incubated by NaAsO 2 for 12 h) was observed by electron microscope. At the same time, human primary umbilical vein endothelial cells were pretreated with 0.1 mmol/L 3-MA (autophagy inhibitor) for 2 h, and induced by 30 μmol/L NaAsO 2, and the above detection indicators were compared with those of the 30 μmol/L NaAsO 2 group. Results:Human primary umbilical vein endothelial cells were successfully isolated and cultured. Compared with the control group [cell viability: (99.97 ± 5.33)%, NO content: 42 048.34 ± 789.61], the cell viability [(73.00 ± 0.86)%] and NO content (23 353.97 ± 971.85) of 30 μmol/L NaAsO 2 group were remarkably lower, and the differences were statistically significant ( P < 0.01). Incubated with 30 μmol/L NaAsO 2 at different time points 6, 12, 24 h, LC3Ⅱ levels (5.782 ± 2.789, 9.692 ± 2.222, 5.573 ± 2.941) were significantly elevated than those of control group (1.000 ± 0.383, P < 0.05), and the LC3 Ⅱ level was the highest at 12 h. After treatment with 30 μmol/L NaAsO 2 for 12 h, the number of autophagosome in cells observed under electron microscope was significantly higher than that of the control group. Compared with 30 μmol/L NaAsO 2 group [cell viability: (68.78 ± 1.55)%, LC3 Ⅱ level: 5.680 ± 0.545, NO content: 13 025.78 ± 962.61], cell viability [ (79.54 ± 4.99) %] in 3-MA+ NaAsO 2 group was increased, the LC3Ⅱ level (3.956 ± 0.398) was decreased, and the differences were statistically significant ( P < 0.05); intracellular NO content (13 988.51 ± 1 671.07) increased, whereas the difference was not statistically significant ( P > 0.05). Conclusion:Autophagy is involved in the vascular endothelial dysfunction induced by arsenic.

ABSTRACT

Objective:To study the relationship between cognitive ability and plasma monoamine neurotransmitters in children with acute lymphoblastic leukemia.Methods:The clinical data of 31 children with acute lymphoblastic leukemia from October 2018 to April 2019 in Tongji Hospital Affiliated to Tongji University were retrospectively analyzed. Among them, 20 cases were in consolidation or intensive treatment stage after complete remission (treatment stage group), and 11 cases were in complete remission and drug withdrawal after treatment (withdrawal group). The plasma levels of dopamine (DA), 5-hydroxytryptamine (5-HT) and 5-hydroxyindoleacetic acid (5-HIAA) were measured, the cognitive function was measured by continuous performance test (CPT) and Stroop test, and the results were compared with those of 20 healthy children (healthy control group).Results:There were no statistical differences in plasma DA and 5-HIAA among 3 groups ( P>0.05). The 5-HT in treatment stage group was significantly lower than that in healthy control group: 1 769.69 (912.86, 2 812.56) ng/L vs. 3 085.29 (2 191.79, 5 310.13) ng/L, and there was statistical difference ( P<0.05). There was no statistical difference in 5-HT between withdrawal group and healthy control group ( P>0.05). CPT results showed that there was no statistical difference in the number of errors among 3 groups ( P>0.05); the correct number in treatment stage group and withdrawal group was significantly less than that in healthy control group: (28 ± 12) and (33 ± 11) pieces vs. (43 ± 10) pieces, the missed number was significantly higher than that in healthy control group: (53 ± 14) and (52 ± 13) pieces vs. (39 ± 14) pieces, and there were statistical differences ( P<0.05). Stroop test results showed that, there was no statistical difference in word-color consistency test among 3 groups ( P>0.05); in word-color contradiction test, the correct number in treatment stage group and withdrawal group was significantly less than that in healthy control group, the number of errors and missed were significantly more than those in healthy control group, and there were statistical differences ( P<0.05); in word color irrelevance test, there was no statistical difference in the number of errors among 3 groups ( P<0.05); the correct number in treatment stage group and withdrawal group was significantly less than that in healthy control group, the number of missed errors in withdrawal group was significantly more than that in healthy control group, and there were statistical differences ( P<0.05). The correlation analysis results showed that plasma DA was negatively correlated with the number of errors in Stroop word-color consistency test, and positively correlated with the number of errors in Stroop irrelevance test ( P<0.05); 5-HIAA was negatively correlated with the correct numbers in CPT test and error number in Stroop consistency test ( P<0.05 or<0.01); 5-HT was positively correlated with the correct number in the word-color contradiction test ( P<0.05) in children with acute lymphoblastic leukemia. Conclusions:Children with acute lymphoblastic leukemia could have continuous and selective attention injury, after being diagnosed with leukemia, the level of 5-HT in plasma monoamine neurotransmitter is lower than that in healthy children. The change of attention may be related to the change of monoamine neurotransmitter.

Objective To explore the status of medication adherence among patients with depressive disorder and analyze its potential influencing factors.Methods A total of 406 patients with depression who visited our hospital from January 2015 to December 2017 were selected as subjects of study.General data of patients were collected at the time of consultation,and drug adherence questionnaire was used to score the patients at 6 months of follow-up.According to the score,406 patients were divided into high adherence group,middle adherence group and low adherence group,the correlation between medication adherence and humanistic factors,chronic diseases and course of disease was analyzed.Results Compared with the low adherence group,the proportion of age (>60 years old),education (<6 years),no spouse,peasant,monthly income (< 3 000 yuan),self-funded medical treatment,insomnia history,solitude and negative life events decreased significantly in the middle and high adherence groups.Cadre ratio and the proportion of workers in the middle adherence group increased significantly (P < 0.05).Compared with the low adherence group,the course of disease in the middle and high adherence group was significantly shorter (P < 0.01).Logistic regression analysis showed that education < 6 years,no spouse,farmers,low income,self-paid medical treatment,solitude,negative life events,course of disease were independently correlated with low adherence of depression patients (P < 0.05 or P < 0.01).Conclusions Depression patients have low adherence with medication,so targeted health education should be carried out.Especially for patients with a long course of illness,continuous nursing and long-term follow-up should be done well.Mobilizing family support system,caring for and respecting patients can effectively reduce the occurrence of negative life events.It can improve the medication adherence of depressive patients and promote their early recovery.

ABSTRACT

Objective To study the effect of T-2 toxin on proliferation and cell cycle of rat chondrocytes,in order to provide a new idea in molecular mechanism of T-2 toxin-induced chondrocyte damage.Methods Primary chondrocytes of neonatal Wistar rats were isolated and stained by toluidine blue staining and type Ⅱ collagen immunofluorescence staining.The effects of different concentrations of T-2 toxin [0 (control),1,5,10,20,50,100 μg/L)] on proliferation of chondrocytes for 24 h were detected by cell counting kit-8 (CCK-8) method,and control,1 (low dose),5 (medium dose),and 10 μg/L (high dose) T-2 toxin were selected for subsequent experiment;cell cycle changes were detected by flow cytometry;Real-time PCR and Western blotting were used to detect the effects of T-2 toxin on mRNA and protein expressions of proliferating cell nuclear antigen (PCNA) and Cyclin D1 in chondrocytes.Results With increase of T-2 toxin concentration (control,1,5,10,20,50,100 μg/L),the cell survival rates [(100.00 ± 0.00)%,(93.12 ± 1.66)%,(77.12 ± 1.11)%,(59.44 ± 4.09)%,(46.64 ± 3.86)%,(38.15 ± 3.37)%,(33.79 ± 0.99)%] were decreased,and the differences were statistically significant (F =139.21,P <0.05).The percentages of quiescent phase/pre-DNA synthesis phase (G0/G1 phase) ceils in 1,5,10 μg/L T-2 toxin groups [(22.03 ± 0.42)%,(30.54 ± 2.61)%,(36.01 ± 1.51)%] were significantly higher than that in control group [(13.79 ± 1.65)%,P < 0.05];the percentages of DNA synthesis phase (S phase) cells [(60.27 ± 3.53)%,(53.88 ±4.38)%,(49.55 ± 2.49)%] were significantly lower than that in control group [(76.72 ± 4.24)%,P < 0.05].The differences of mRNA levels of PCNA and Cyclin D1 between groups were statistically significant (F =46.80,17.97,P < 0.05),and 5,10 μg/L T-2 toxin groups (0.77 ± 0.13,0.79 ± 0.08,0.60 ± 0.07,0.56 ± 0.05) were lower than the control group (0.99 ± 0.02,1.01 ± 0.01,P < 0.05).The expressions of PCNA protein in 5,10 μg/L T-2 toxin groups (0.69 ± 0.03,0.49 ± 0.03) were lower than that in control group (0.92 ± 0.05,P < 0.05);the expressions of Cyclin D1 protein in 1,5,10 μg/L T-2 toxin groups (0.80 ± 0.06,0.60 ± 0.07,0.33 ± 0.13) were lower than that in control group (0.95 ± 0.07,P < 0.05).Conclusion T-2 toxin can inhibit the proliferation of chondrocytes,which may be worked through influencing the expression of cell cycle protein,causing cell cycle arrest,thereby inhibiting DNA synthesis.

ABSTRACT

Objective To investigate the protective effect of dexamethasone on hearing loss induced by canalwall-down mastoidectomy.Methods A total of 76 patients (76 ears) were randomized to dexamethasone group and non dexamethasone group with 38 patients in each group.For dexamethasone group,gelfoam soaked with dexamethasone (5 mg/ml) was topically applied to the round window niche at the end of the surgery,and dexamethasone was administrated intravenously at the dosage of 5 mg/50 kg immediately after the surgery.While for non-dexamethasone group,dexamctbasone was not used.The pure-tone audiograms were performed before mastoidectomy and 90 days after the surgery.Absolute bearing change was defined as the difference in hearing thresholds in decibels before and after the surgery.Results The changes of bone conduction thresholds in dexamethasone group were 8.3± 3.9 dB at 6 kHz,11.3±5.2 dB at 8 kHz,and 10.1±7.2 dB for HPTA (4-6-8 kHz high tone average).As in non-dexamethasone group,the changes of bone conduction thresholds were 13.7±4.6 dB at 6 kHz,25.1±5.4 dB at 8 kHz,19.3±9.7 dB for HPTA.There were significant differences in the changes of bone conduction thresholds between dexamethasone and non dexamethasone groups at frequencies of 6 kHz (P=0.039),8 kHz (P=0.007) and HPTA (P=0.009).Conclusion The results demonstrated that application of dexamethasone significantly reduced sensorineural high frequencies (6 kHz and 8 kHz) hearing loss after canal-wall down mastoidectomy,thus the use of dexamethasone is recommended.

ABSTRACT

Objective To investigate the association between pretreatment serum lactate dehydrogenase (LDH)to albumin ratio(LAR)and the long-term survival of nasopharyngeal carcinoma patients after definite radiotherapy.Methods A total of 956 treatment-naive nasopharyngeal carcinoma patients receiving definite radio-therapy between October 2007 and December 2009 were collected.Univariate and multivariate model were used to analyze the association between pretreatment LAR and overall survival(OS)or distant-metastasis-free survival (DMFS).Results Based on-line cutoff finder,the optimal cut-off value of LAR was determined to be 0.385.Uni-variate analysis reveals that OS and DMFS were all significantly poorer in the high LAR group than that in the low LAR group(P < 0.001). Cox multivariate regression model shows that high LAR(P < 0.001),age ≥ 45 years (P < 0.001)and stage Ⅲ~Ⅳ(P < 0.001)were independent inferior prognostic factors for OS,while high CAR (P < 0.001)and stage Ⅲ~Ⅳ(P = 0.012)were independent inferior prognostic factor for DMFS. Conclusion Pretreatment LAR is an independent prognostic factor in nasopharyngeal carcinoma and could be used in risk strati-fication and guiding adjuvant therapy.

ABSTRACT

Objective To analyze the characteristics of left ventricular systolic function in Kawasaki disease (KD) patients with intravenous immunoglobulin (IVIG) resistant during acute phase by two-dimensional speckle tracking imaging (2D STI).Methods IVIG resistant patients (n=40) as well as age and gender matched IVIG responder patients (n=40) were selected from KD patients in acute phase.Patients in IVIG resistant group were further divided into coronary artery dilation (CAD) subgroup and no coronary artery dilation (NCAD) subgroup.Then conventional echocardiography,2D STI and laboratory indexes were acquired and compared between IVIG resistant group and IVIG responder group,as well as between CAD and NCAD subgroup.ROC curve analysis was used to determine threshold values of 2D STI measurements associated with IVIG resistance.Results Compared with IVIG responder group,coronary artery dilation,left ventricular mass and left ventricular mass index increased,systolic global longitudinal strain (GLS) and systolic global circumferential strain (GCS) decreased,albumin,erythrocyte sedimentation rate,C-reactive protein and platelet increased in IVIG resistant group (all P<0.05).Taking absolute GLS 16.8% as a threshold,the area under curve (AUC) was 0.769 (P=0.021),sensitivity,specificity in diagnosis of IVIG resistant was 79.27%,68.36%.Taking absolute GCS 15.9% as a threshold,AUC was 0.749 (P=0.038),sensitivity,specificity in diagnosis of IVIG resistant was 71.43%,57.28%.Conclusion IVIG resistant KD patients present significantly greater systolic dysfunction compared with responders in patients with KD,which may be the results of myocardium infection other than coronary artery lesions.2D STI may predict myocardial injury in IVIG resistant KD patients.

Objective To investigate the relationship between the rat depression behaviors induced by chronically unpredicted stress (CUS) with interleukin-1β (IL-1β),interleukin-6 (IL-6) protein expression and indole-2,3-dioxygenase (IDO) mRNA and protein expression.Methods Thirty male Sprague Dawley (SD)rats were divided into the control group (CG) and model group (MG).The solitary raise combined with CUS 28 d continued stimulus was used to establish the rat depression model.The open field test (OFT) and the force swimming test (FST) were used to evaluate the rat depression behaviors;qRT-PCR was used to determine the mRNA expression of IDO in hippocampus and cortex;Western blot was used to determine the protein expressions of IL-1β,IL-6 and IDO in hippocampus and cortex.Results Compared with the CG group,the OFT score in the rats of the MG group was significantly decreased(P<0.01),the immobility time in FST was significantly extended (P<0.00),the expression of IDO mRNA in hippocampus and cortex was significantly increased (P<0.01) and the protein expression of IL-1β,IL-6 and IDO in hippocampus and cortex was significantly increased (P<0.05).Conclusion The rat depression behaviors induced by CUS may be associated with the increase of intracerebral inflammatory cytokine expression and inducing IDO over-expression.

ABSTRACT

Objective To evaluate the value of low-density lipoprotein-cholesterol (LDL-C) and non-high-density lipoprotein-cholesterol (non-HDL-C) in differential diagnosis of familial hypertriglyceridemia (FHTG) and familial combined hyperlipidemia (FCHL).Methods We recruited 9 FHTG pedigrees (94 subjects) and 24 FCHL pedigrees (94 subjects) and then divided them into affected groups and non-affected groups according to lipid abnormality.Another 10 normal control pedigrees (57 subjects) served as controls.We compared the routine lipid levels such as triglyceride (TAG),total cholesterol (TC),HDL-C and LDL-C and non-HDL-C between the groups.After stratification based on TAG level,we observed the relationship between LDL-C and non-HDL-C.Last we confirmed and analyzed the cut-off value of differential diagnosis between FHTG and FCHL with receiver operating characteristic (ROC) curve.Results The levels of TAG,TC,and non-HDL-C were significantly higher in the affected group of FHTG than in the non-affected group of FHTG and the normal group (P<0.01 or P<0.05).The levels of TAG,TC,HDL-C,LDL-C and non-tHDL-C wcrc significantly higher in the affected group of FCHL than in the non-affected group of FCHL and the normal group (P<0.01 or P<0.05).The levels of TAG were significantly higher (P<0.01) while TC,HDL-C,LDL-C and non-HDL-C levels were significantly lower (P< 0.01 or P<0.05) in the affected group of FHTG than in the affected group of FCHL.The association between LDL-C and non-HDL-C was positive both in FHTG and FCHL,but the relationship became weaker as TAG level increased.The cut-off value of LDL-C and non-HDL-C was 3.575 mmol/L and 4.525 mmol/L,respectively.Conclusion In addition to the routinely used lipid indexes,non-HDL-C may be a new index for differential diagnosis of FHTG and FCHL,and may be superior to LDL-C in this regard.

ABSTRACT

OBJECTIVE To investigate the clinical curative effect of acellular dermal matrix contrast with temporalis fascia in type Ⅰ tympanoplasty. METHODS A retrospective analysis of 60 inflammatory chronic suppurative otitis media patients from January 2014 to January 2015 in Genenral Hospital of PLA Guangzhou Military Area were conducted.All cases were perpormed the ear incision under local anesthesia, including 32 cases(32 ears) with acellular dermal matrix, 28 cases(28 ears) with temporal fascia. The operation time, postoperative pain, the healing of tympanic membrane and the postoperative recovery were compared between the two groups. RESULTS There was significant difference in the operation time and postoperative pain between the two groups(P0.05). CONCLUSION The effect of acellular dermal matrix for repairing tympanic membrane is similar to the temporalis fascia. The use of acellular dermal matrix has the advantage of shorter operation time, less postoperative pain, and can avoid regional two incisions, it is worthy of clinical application.

ABSTRACT

Objective Obstructive sleep apnea hypopnea syndrome (OSAHS) often causes damage to multiple systems, especially to the central nervous system, inducing cognitive dysfunction.This study aims to explore the possible correlation of the expressions of serum hypoxia-inducible factor-1α (HIF-1α) and brain-derived neurotrophic factor (BDNF) with cognitive impairment in rats under different hypoxia conditions.Methods Twenty-four Wistar rats were equally randomized into a normal control, a chronic intermittent hypoxia (CIH), and a chronic continuous hypoxia (CCH) group.The rats of the CIH group were placed in a hypoxia chamber filled with N2 and air, the oxygen concentration switched from (7±0.5)% to 21%, 1.5 minutes for each state and 4 minutes for each cycle, while those of the CCH group were placed in another hypoxia chamber with the oxygen concentration of (7±0.5)%, 8 hours a day and all for 30 days.Then we recorded the body weight of the rats, detected the expressions of serum HIF-1α and BDNF by ELISA, and observed the changes of behavior by Morris water maze test and those of the hippocampal morphological structure by HE staining.Results At 30 days after modeling, the body weight of the rats was significantly decreased in the CIH and CSH groups as compared with the normal control ([195.75±6.497] and [180.88±12.017] vs [218.63±15.287] g, P<0.05).Positioning navigation showed that the escape latency was significantly longer in the hypoxia models than in the controls (P<0.05), even longer in the CIH than in the CCH group (P<0.05).Spatial exploration test manifested a lower frequency of crossing the platform in the CIH and CCH groups than in the control ([2.63±1.45] and [3.22±1.30] vs [4.97±0.47] times, P<0.05).The expression levels of serum HIF-1α and BDNF were significantly higher in the CIH ([36.14±9.34] and [1625.34±332.44] pg/mL) and CCH ([27.27±6.88] and [1204.07±363.81] pg/mL) than in the normal control group ([14.11±4.06] and [1036.40±124.48] pg/mL) (P<0.05), even higher in the CIH than in the CCH group (P<0.05).HE staining exhibited scattered and disorderly arrangement of hippocampal neurons in the model rats, with unclear nuclear membrane, pyknosis of the nuclei, darkly stained cytoplasm, and some damaged cells.More obvious absence and vacuolization of some cells were observed in the rats of the CIH group.Conclusion Chronic hypoxia inhibits the growth and development of rats and induces cognitive dysfunction.High-level HIF-1α in chronic intermittent hypoxia indicates hypoxia-stress of the body, while compensatory increase of serum BDNF may be involved in neuronal cell damage regulation.

ABSTRACT

Objective To discuss the effects and the possible mechanismof curcumin on pulmonary functions and expression of TGF-β1 and NF-κB in paraquat-induced pulmonary fibrosis of rats.Methods SPF Wistar rats were randomly (random number) divided into three groups:paraquat-poisoned group (PQ group,with PQ 50 mg/kg by gavage),Curcumin-treatment group (PC group after 30,with curcumin (200 mg/kg) by intraperitoneal injection),and Blank group (B group,with same volume of sterile saline at the same time).After 14 d,the lung function of rats was observed,and the expression of TGF-β1 and NF-κB protein were detected by immunohistochemistry.Results The survival rates of mice significant difference in the PC and PQ groups with (70.83% vs.41.67%,P <0.05).Compared with the B group,lung function index (Te,PIF,PEF,EF50,TV,F) of PQ group significantly decreased (P < 0.05,P < 0.01).Compared with the PQ group,lung function index (Te,PEF,TV,F) of PC group significantly increased (P < 0.05 or P < 0.01).There are some expression of lung tissues of rats with TGF-β1,NF-κB in B group.Compared with B group,PQ group significantly enhanced (P <0.01or P <0.05).Compared with the PQ,PC group significantly decreased (P < 0.01 or P < 0.05).Conclusions Curcumin can relieve paraquat-induced pulmonary fibrosis by inhibiting the over expression of TGF-β1,NF-κB in lung tissue of rats.

ABSTRACT

[Objective] To evaluate the value of cardiac screening in the first trimester by using three-dimensional ultrasound with spatiotemporal image correlation (STIC) and to analyze the value of different views for cardiac assessment.[Methods] This study retrospectively reviewed the eighty-seven fetuses with congenital heart diseases which were diagnosed in our institution between January 4th 2013 and June 30th 2016.We compared the detection rate of prenatal ultrasound screening with STIC in the first trimester and the results of postnatal echocardiography or autopsy.In addition,the detection rates of the different planes,including four-chamber view (4CV),outlet of the left ventricle (OLV),outlet of the right ventricle (ORV) and three vessels-trachea view (3VT) were analyzed.[Results] Severe cardiac anomalies were identified in 62 fetuses (71.3%) and non-severe cardiac anomalies were identified in 25 fetuses (28.7%).Seventy-eight cases (89.7%,78/87) with congenital heart diseases could be detected by using three-dimensional ultrasound with STIC.The detection ratesfor 4CV,4CV+OLV,4CV+ORV,4CV+3VT,4CV+OLV+ORV and 4CV+OLV+ORV+3VT were 48.3%,64.4%,66.7%,79.3%,69.0% and 89.7% respectively.[Conclusions] Three-dimensional ultrasound with STIC could detect congenital heart disease in the first trinester.The four-chamber view plus outlet of the left ventricle,outlet of the right ventricle and three vessels-trachea view could increase the detection rates of congenital heart diseases.

ABSTRACT

Objective To observe the clinical efficacy of acupuncture and miuci therapy combined with Qinggan Tongqiao Decoction for sudden sensor-neural hearing loss (SSHL) and the effects on level of plasma endothelin (ET) and serum nitrate oxide (NO). Methods Totally 110 patients with SSHL were randomly divided into treatment group and control group, with 55 patients in each group. The control group was given Qinggan Tongqiao Decoction combined with acupuncture routine treatment, and treatment group was given miuci therapy on the basis of the control group, two weeks as a course of two groups. Level changes of plasma ET and serum NO, and pure tone listening threshold changes before and after treatment were observed. Tinnitus and hearing efficacy was evaluated. Results The total effective rate for tinnitus efficacy was 94.54% (52/55) in the treatment group and 85.45% (47/55) in the control group, with statistical significance (u=2.038, P=0.040). The total effective rate for hearing efficacy was 96.46%(53/55) in the treatment group and 81.82% (45/55) in the control group, with statistical significance (u=2.913, P=0.040). Compared with before treatment, the level of plasma ET decreased and the level of serum NO increased after the treatment (P<0.05); After the treatment, the level of plasma ET in the treatment was lower than the control group, and the level of serum NO in the treatment group was higher than the control group (P<0.05). Pure tone threshold scores in the two groups on treatment 3, 7, 14 d were significantly improved (P<0.05), and pure tone threshold scores in the treatment group were significantly lower than the control group (P<0.05). Conclusion Acupuncture and miuci therapy combined with Qinggan Tongqiao Decoction can significantly reduce clinical symptoms and enhance the hearing level of SSHL patients. The mechanism mainly lies in regulating levels of plasma ET and serum NO, and improving the inner ear micro-circulation.

Aim To study the role of histone acetyla-tion and its involvement in the depression-like behav-iors of rats induced by chronic unpredictable stress (CUS ). Methods Thirty male Sprague Dawley (SD ) rats were randomly divided into control group and model group.The method of solitary condition with CUS for consecutive 28 days was used to establish the rat depression model.The open-field test (OFT)and the forced swimming test (FST)were used to evaluate the depressive behaviors of rats;the real time PCR was used to detect the change of HDAC2 mRNA, and Western blot was used to determine the protein expres-sions of H3,H4,acH3 and acH4 in the prefrontal cor-tex and hippocampus of rats.Results Model group showed obvious depression-like behaviors with decrea-sing locomotive activity in OFT (P <0.01 )and in-creasing immobility time in FST (P<0.01),up-regu-lating the mRNA and protein expression of HDAC2 (P<0.0 1 ),and down-regulating the protein expression of acH3 and acH4 (P<0.01)in the prefrontal cortex and hippocampus significantly,compared with control group.Conclusion The mechanism of depressive be-haviors of rats induced by CUS may be associated with down-regulating the level of histone acetylation modifi-cation.
