ABSTRACT

OBJECTIVE@#To investigate the role of multiple serological methods in the identification of complex antibodies.@*METHODS@#The blood group antigens were detected by saline and microcolumn agglutination methods. The saline method was used to screen and identify IgM-type antibodies in the patient's serum, while the polybrene, anti-globulin, microcolumn agglutination, enzymic and absorption-elution methods were used to screen and identify IgG-type antibodies.@*RESULTS@#The patient was B/CCDee/Jk(a-b+)/Fy(a-b+) blood type. The serum reacted with panel cells, and the reaction presented anti-E pattern in the saline medium. It was fully positive in the microcolumn agglutination card, except 2 negative ones after using papain to treat the panel cells. Referring to the pattern table, it was concluded that there existed anti-c, anti-E, and anti-Jka antibodies, and one antibody corresponding to an antigen that was easily destroyed by papain. The red blood cells with specific phenotype were selected for absorption-elution to identify IgG-type anti-c, anti-E, anti-Jka and anti-Fya antibodies.@*CONCLUSION@#It is confirmed that IgM-type anti-E, and IgG-type anti-c, anti-E, anti-Jka and anti-Fya antibodies exist in the patient's serum by multiple serological methods.

OBJECTIVE@#To study the serological characteristics and molecular biological basis of 8 individuals with Para-Bombay phenotypes in Guangxi area.@*METHODS@#Serological tests were used to identify the blood groups of red cells. Molecular biological methods, including PCR-SSP for ABO genotyping and DNA sequencing for FUT1, were used to detect the genotypes of ABO and FUT1 which determined the expression of H antigen.@*RESULTS@#Eight individuals in the study were all the Para-Bombay phenotypes, including 4 cases of B@*CONCLUSION@#There are varieties of molecular genetic mechanisms for Para-Bombay phenotypes. ABSTRACT

<p><b>OBJECTIVE</b>To analyse the cases of platelet transfusion refractoriness after received HLA-matched unrelated donor hematopoietic stem cell transplantation, to analyze and identify the phenotype and genotype of CD36 in both the patient and stem cell donor, as well as the characteristic of antibody induced platelet transfusion refractoriness, and to analyse the efficacy of matched CD36-deficiency platelets transfusions.</p><p><b>METHODS</b>The CD36 expression on platelet and monocyte was analyzed by flow cytometry (FCM) in both patient and donor. Polymerase chain reaction sequence-based typing (PCR-SBT) was used to analyze the exons sequence of CD36 and HPA. Fast monoclonal antibody-specific immobilization of platelet antigen (F-MAIPA) and FCM were used to identify platelet antibodies in the patient. Short tandem repeat polymerase chain reaction (STR-PCR) was applied to monitor engraftment evidence. The platelet level was monitored. CD36- deficiency donor's platelets were selected from CD36- deficiency donor blood bank.</p><p><b>RESULTS</b>The donor was CD36 positive and the patient was typed I CD36 deficiency. The anti-CD36 antibody was identified in patient's serum (after transplantation), while the HLA and HPA-related antibodies were excluded. Sequence analysis of CD36 exon in the patient showed Exon 6 -1G>C(Change in splicing site) homozygote, which was a novel CD36 mutation. STR, HPA and CD36 of the patient (complete chimerism) were conversed to that of donor gene types on day 18 after allo-HSCT. The positive CD36 expression on platelet and monocyte in the patient was observed on day 96 after allo-HSCT. The patient showed the platelet transfusion refractoriness which was significantly improved after platelets transfusions from CD36 deficiency donors.</p><p><b>CONCLUSION</b>Stem cell transplants resulted in anti-CD36 and caused platelet transfusion refractoriness, that was first reported in China. To ensure the efficacy of platelet transfusion, the CD36-deficiency patient should receive CD36 deficiency platelets for transfusion.</p>

ABSTRACT

Objective@#To compare the clinical effect of diode laser enucleation of the prostate (DIOD) with that of transurethral resection of the prostate (TURP) on benign prostate hyperplasia (BPH) with different prostate volumes.@*METHODS@#This retrospective study included 256 BPH patients treated by DIOD (n = 141) or TURP (n = 115) from March 2012 to August 2015. According to the prostate volume, we divided the patients into three groups: 80 ml (48 for DIOD and 39 for TURP). We obtained the relevant data from the patients before, during and at 6 months after surgery, and compared the two surgical strategies in operation time, perioperative levels of hemoglobin and sodium ion, post-operative urethral catheterization time and bladder irrigation time, pre- and post-operative serum PSA levels, International Prostate Symptoms Score (IPSS), post-void residual urine (PVR) volume and maximum urinary flow rate (Qmax), and incidence of post-operative complications among different groups.@*RESULTS@#In the 80 ml group ([3.35 ± 1.39] ng/ml vs [1.76 ± 0.91] ng/ml, P <0.05). No blood transfusion was necessitated and nor postoperative transurethral resection syndrome or urethral stricture observed in DIOD. However, the incidence rate of postoperative pseudo-urinary incontinence was significantly higher in the DIOD (22.7%, 32/141) than in the TURP group (7.83%, 9/115) (P <0.05).@*CONCLUSIONS@#DIOD, with its obvious advantages of less blood loss, higher safety, faster recovery, and more definite short-term effectiveness, is better than TURP in the treatment of BPH with medium or large prostate volume and similar to the latter with small prostate volume.

ABSTRACT

OBJECTIVE: To determine whether the appearance of a spiculated mass on a mammogram is associated with luminal A subtype breast cancer and the factors that may influence the presence or absence of the spiculated mass. MATERIALS AND METHODS: Three hundred seventeen (317) patients who underwent image-guided or surgical biopsy between December 2014 and April 2015 were included in the study. Radiologists conducted retrospective assessments of the presence of spiculated masses according to the criteria of Breast Imaging Reporting and Data System. We used combinations of estrogen receptor (ER), progesterone receptor (PR), human epithelial growth factor receptor 2 (HER2), and Ki67 as surrogate markers to identify molecular subtypes of breast cancer. Pearson chi-square test was employed to measure statistical significance of correlations. Furthermore, we built a bi-variate logistic regression model to quantify the relative contribution of the factors that may influence the presence or absence of the spiculated mass. RESULTS: Seventy-one percent (71%) of the spiculated masses were classified as luminal A. Masses classified as luminal A were 10.3 times more likely to be presented as spiculated mass on a mammogram than all other subtypes. Patients with low Ki67 index (< 14%) and HER2 negative were most likely to present with a spiculated mass on their mammograms (p <0.001) than others. The hormone receptor status (ER and PR), pathology grade, overall breast composition, were all associated with the presence of a spiculated mass, but with less weight in contribution than Ki67 and HER2. CONCLUSION: We observed an association between the luminal A subtype of invasive breast cancer and the presence of a spiculated mass on a mammogram. It is hypothesized that lower Ki67 index and HER2 negativity may be the most significant factors in the presence of a spiculated mass.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effectiveness of surgical strategies for Shang Ring circumcision in the treatment of short frenulum praeputii in patients with redundant prepuce or phimosis.</p><p><b>METHODS</b>Totally, 130 cases of short frenulum praeputii with redundant prepuce or phimosis were randomly assigned to an experimental group and a control group of equal number to receive Shang Ring circumcision, the former by transverse incision in the distal penis foreskin and pull-up of the interior board, and the latter by conventional transverse incision and longitudinal suture of the frenulum praeputii. Comparisons were made between the two groups in the surgical duration, intraoperative blood loss, 24 h postoperative pain visual analog score (VAS), postoperative complications, satisfaction with the penile appearance, and the quality of sexual life.</p><p><b>RESULTS</b>The surgical duration, intraoperative blood loss, 24 h postoperative VAS, postoperative sexual satisfaction, and satisfaction with penile appearance were (4.60 +/- 1.20) min, (2.61 +/- 1.81) ml, 1.73 +/- 0.76, 98.5%, and 98.5%, respectively, in the experimental group, as compared with (21.60 +/- 6.30) min, (11.10 +/- 3.40) ml, 5.37 +/- 1.84, 70.3% and 69.8% in the control, with statistically significant differences between the two groups (P < 0.05). The incidence rates of such major complications as wound dehiscence, infection, and moderate to severe edema were 1.5% (1/65), 3.1% (2/65), and 4.6% (3/65), respectively, in the experimental group in comparison with 12.3% (8/65), 15.3% (10/65), and 30.7% (20/65) in the control, with statistically significant differences between the two groups (P < 0.05). None of patients had any serious complications.</p><p><b>CONCLUSION</b>Shang Ring circumcision by transverse incision in the distal penis foreskin and pull-up of the interior board, with its advantages of shorter operation time, less blood loss, mild pain, fewer complications, and higher satisfaction and acceptance of the patients, can be used as an safe and effective approach to the treatment of short frenulum praeputii.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate whether the genetic polymorphism, upstream variable number of tandem repeats (uVNTR), in the monoamine oxidase A (MAOA) gene, is associated with major depressive disorder (MDD) in adolescents and to test whether there is gene-environment interaction between MAOA-uVNTR polymorphism and stressful life events (SLEs).</p><p><b>METHODS</b>A total of 394 Chinese Han subjects, including 187 adolescent patients with MDD and 207 normal students as a control group, were included in the study. Genotyping was performed by SNaP-shot assay. SLEs in the previous 12 months were evaluated. The groups were compared in terms of the frequency distributions of MAOA-uVNTR genotypes and alleles using statistical software. The binary logistic regression model of gene-environment interaction was established to analyze the association of the gene-environment interaction between MAOA-u VNTR genotypes and SLEs with adolescent MDD.</p><p><b>RESULTS</b>The distribution profiles of MAOA-u VNTR genotypes and alleles were not related to the onset of MDD, severity of depression, comorbid anxiety and suicidal ideation/behavior/attempt in adolescents. The gene-environment interaction between MAOA-u VNTR genotypes and SLEs was not associated with MDD in male or female adolescents.</p><p><b>CONCLUSIONS</b>It is not proven that MAOA-u VNTR polymorphism is associated with adolescent MDD. There is also no gene-environment interaction between MAOA-u VNTR polymorphism and SLEs that is associated with adolescent MDD.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the apoptosis induction effect of vitamin E succinate (VES) on Tca8113 cells and its possible mechanisms.</p><p><b>METHODS</b>The proliferative activity of Tca8113 was assessed by methyl thiazolyl tetrazolium (MTT) assay. After Tca8113 cells were treated with different concentrations of VES, apoptotic rates were analyzed by flow cytometry (FCM). Fas monoclonal antibody was used for the blocking test. Fas expression was detected by immuocytochemistry(SABC assay) and FCM.</p><p><b>RESULTS</b>VES demonstrated a significant growth inhibitory effect and apoptosis induced effect on the Tca8113 cells in a dose- and time-dependent manner. Fas neutralizing antibody can block the apoptosis induced by VES. After the administration of VES, the expression of Fas protein increased and the kytoplasm staining enhanced. Proteinum quantitative analysis showed that the mean fluorescence intensity increased.</p><p><b>CONCLUSION</b>VES can induce apoptosis in human tongue cancer cells, and the up-regulation of the cell surface Fas protein may play an important role in the process.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the influence of butt joint connection and platform switching design of implant-abutment connection on the stress distribution in peri-implant bone.</p><p><b>METHODS</b>Three-dimensional finite element models of implant-supported mandibular first molar with different implant-abutment connections were computed by COSMOSM 2.85. Traditional butt joint connection was used in model A and platform switching design in model B. Loading conditions were a vertical load of 200 N and inclined load of the same magnitude at 45 degrees to the vertical axis of the implant. Stress distribution in peri-implant bone and von Mises stresses at the same point of buccal and lingual implant-bone interfaces in two models were compared.</p><p><b>RESULTS</b>Stresses concentrated in peri-implant cortical bone at the neck of implants on the buccal and lingual sides; maximum von Mises stresses under inclined load were higher than those under vertical load. Maximum von Mises stress in bone was 11.61 MPa in model A and 7.15 MPa in model B under vertical load, and 22.07 MPa in model A and 11.87 MPa in model B under inclined load respectively. Von Mises stresses decreased as the distance from implant-abutment junction increased and the most obvious change occurred at the interface between cortical bone and spongy bone. Von Mises stresses at the same points of buccal and lingual implant-bone interfaces in model A were higher than those in model B.</p><p><b>CONCLUSIONS</b>Compared with butt joint connection, platform switching design improved the stress distribution and decreased the maximum stresses in peri-implant bone around implant cervix.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the upper airway structure of sleep-disordered breathing children.</p><p><b>METHODS</b>Seventy three children with obstructive sleep apnea hypopnea syndrome (OSAHS), 53 children with primary snoring (PS) and 40 control subjects underwent pharyngeal magnetic resonance imaging (MRI). Upper airway structure images were analyzed and measured.</p><p><b>RESULTS</b>The cross-section area of the nasopharyngeal and palatopharyngeal airway in subjects with OSAHS and PS are smaller (P < 0.01) than that of the control group. The cross section area of OSAHS patients are smaller than that of PS subjects (P < 0.01). The above parameter of oropharyngeal airway in OSAHS patients is smaller than that of control group (P < 0.01), but no statistic difference compared with that of PS subjects. The cross-section area and length of the adenoid in OSAHS group are bigger and longer than that of PS group (P < 0.01) and bilateral tonsils are larger (P < 0.01); in OSAHS patients the cross-section area of the soft palate is larger and the length of the soft palate is longer (P < 0.01) than that of PS group, while this parameter of PS group is similar to that of the control group. And the maximum width of the soft palate, the cross-section area of bilateral fat pad, bilateral pterygoid and tongue are similar among OSAHS, PS and the control group. The skeletal measurement: the length of H-C2C3 in subjects with OSAHS is longer (P < 0.01); The angle(alpha) in OSAHS patients is smaller (P < 0.01) than that of other 2 groups. The angle (beta), the cross-section area of the mandible, the spine-clivus oblique, the length of the hard palate and the distance of the mandible are similar among the three groups.</p><p><b>CONCLUSIONS</b>In children with OSAHS or PS, the upper airway is restricted by both the adenoid and tonsils; however, the soft palate is also larger in OSAHS, adding further restriction. Otherwise, downward movement of the hyoid bone and decreasing of the angle (alpha) in OSAHS influence laryngopharynx airway. MRI is of clinical significance for evaluating OSAHS children's upper airway.</p>

ABSTRACT

Targeting induced local lesions in genomes (TILLING) is a reverse genetics method for functional genomics research.It is possible to screen for point mutations in the populations of EMS mutagenesis with highthroughput and lowcost. EcoTILLING a method based on TILLING ,was developed for detecting multiple types of polymorphisms in germplasm collections,such as single nucleotide polymorphism,small deletion and insertion etc.Rice is a very important food crop and a model plant for genome research also. There are complete genome sequence and a lot of other bioinformatics resources about it.So the markerassisted breeding is becoming more and more important in rice breeding. Some issues based on TILLING about identifying germplasm based on gene sequence,EMS mutagenesis breeding,developing functional marker in rice breeding in future were discussed.

ABSTRACT

<p><b>OBJECTIVE</b>To detective the relationship between cyclooxygenase-2 (COX-2) and angiogenesis in oral squamous cell carcinoma (OSCC) and its clinical significance through observing the expression of COX-2 and determining microvessel density (MVD) in OSCC.</p><p><b>METHODS</b>PV-9000 immunohistochemistry was used to determine the expression of COX-2 and CD34, which was used to determine MVD, in 76 OSCC tissues and 12 normal oral mucosa tissues.</p><p><b>RESULTS</b>Overexpression of COX-2 was detected in OSCC, and was more intense compared with normal epithelium (P < 0.001). The high expression of COX-2 in OSCC was related to neck lymphnode metastasis, tumor size, TNM stage and histological grade (P <0.05). The MVD value in COX-2-positive group was much higher than that in COX-2-negative group (P < 0.01) and that in normal oral mucosa tissues (P < 0.01).</p><p><b>CONCLUSION</b>The high expression of COX-2 in OSCC was significantly associated with MVD, neck lymphnode metastasis, tumor size, TNM stage and histological grade. COX-2 might be one of the important factors in the angiogenesis of OSCC.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the relationship between the blood flow of the exceedingly expanded skin and the viability of the flap.</p><p><b>METHODS</b>The piglets were set up as the animal model. They were divided into 7 groups according to different injection volume of the tissue expander, i. e. injection to 1/2 content, 3/4 content, full content, 50% over content, 100% over content, sham-operation control and normal control. At different stages of expansion, the blood flow and the amplitude of the expanded skin were examined by laser Doppler. The survival length of the expanded flap was measured. Furthermore, statistical analysis was performed.</p><p><b>RESULTS</b>As the expansion volume increasing, the blood flow and the amplitude of the expanded skin increased, the survival length of the expanded flap increased also. In the meantime, the amplitude instability and vessel permeability increased. When the injection volume exceeded to 100%, the blood flow and the survival length of the expanded flap decreased.</p><p><b>CONCLUSIONS</b>In soft tissue and skin expansion, increasing injection volume could increase the survival length of the expanded flap, while over-expansion to 100% content may cause blood flow disturbance.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the correlation between the expansion with different volume and the contents of the b-FGF and EGF in the expanded skin.</p><p><b>METHODS</b>The piglets were used in the experiment,which were divided into 7 groups including the sham, the blank control,the experimental groups with inflation of 1/2 volume, 3/4 volume, full volume, 150% volume and 200% volume. When expansion was complete, b-FGF was measured using enzyme coagulate method. EGF was measured by radioimmunoassay. The data were analyzed statistically.</p><p><b>RESULTS</b>The contents of b-FGF and EGF in the expanded skin was significantly higher than normal skin (P < 0.05). The difference in contents of b-FGF and EGF between the over-expanded skin and normal skin was more significant (P < 0.01).</p><p><b>CONCLUSION</b>Expansion can lead to the increase of b-FGF and EGF contents in the expanded skin.The biological effects may be relative to epidermal hyperplasia and angiogenesis of the expanded skin.</p>
