ABSTRACT

<p><b>OBJECTIVE</b>To explore the effects of ANO1 overexpression on the proliferation, detachment, spreading, and migration of laryngocarcinoma Hep-2 cell line.</p><p><b>METHODS</b>ANO1-overexpressing Hep-2 cell line was selected as the assay group, and Hep-2 cell line with empty plasmid was selected as the control group. MTT assay was used to detect the proliferation abilities of Hep-2 cells in both two groups. Cell detachment assay and spreading assay were used to detect the detachment and spreading abilities of Hep-2 cells. Boyden chamber invasion assay, wound healing assay in vitro, and niflumic acid block chloride channel were used to detect the migration abilities of Hep-2 cells. All data were analyzed by SPSS 10.0 software package.</p><p><b>RESULTS</b>Cell proliferation assay by MTT showed that, compared with the control group, the optical density value of assay group was not significantly different (P=0.62). The results of cell detachment assay and cell spreading assay showed the cell detachment rates and cell spreading rates in assay group were significantly higher than those in control group (P<0.0001). The results of Boyden chamber invasion assay showed the percentages of cells migrating through the membrane in assay group were significantly higher than those in control group (P<0.0001). The results of in vitro wound healing experiments showed the wound area rate in assay group was significantly lower than that in control group (P<0.0001). The results of niflumic acid blocking chloride channel experiments showed the wound area rates in assay group were significantly higher than those in control group (P<0.0001).</p><p><b>CONCLUSION</b>ANO1 overexpression does not remarkably alter the proliferation rate of cancer cells, but increases the migration, spreading, and detachment capacities of head and neck squamous cell carcinoma.</p>

ABSTRACT

Objective To study the effect of tubulin tyrosine ligase like 12 (TTLL12) and nitrotyrosine on the growth of prostate cancer cells. Methods TTLL12 and crtubulin nitrotyrosination (N-tubulin) were detected by Western blotting analysis in normal prostate epithelial cell lines PWR1E and RWPE1, and prostate cancer cell line DU145; the proliferation of cells was observed by MTT assay. Results The expression of N-tubulin was significantly lower in DU145 cells than those in PWR1E and RWPE1 cells after treatment with nitrotyrosine (P 0. 05). Conclusion TTLL12 allows an escape of prostate cancer cells from the treatment of nitrotyrosine, leading to the escape of prostate cancer cells from the body's surveillance system and acquirement of their abnormal proliferation.

ABSTRACT

<p><b>OBJECTIVE</b>To observe the effects of curcumin on the epithelial mesenchymal transition (EMT) and TGF-beta/Smads signal pathway in unilateral ureteral obstruction (UUO) rats.</p><p><b>METHODS</b>UUO rat model was established. Rats were divided into the sham-operation group, the model group, the 1st curcumin group, and the 2nd curcumin group, respectively. From the 2nd day after surgery, rats of the 1st curcumin group were administrated with curcumin by gastrogavage at the daily dose of 100 mg/kg. From the 10th day after surgery, rats of the 2nd curcumin group were administrated with curcumin by gastrogavage at the daily dose of 100 mg/kg. Equal volume of normal saline was given to rats by gastrogavage in the sham-operation group and the model group. On the 28th day after surgery rats were killed. The renal tissues of the obstructed side were taken out. The distribution of mice alpha-smooth muscle actin (alpha-SMA) and E-cadherin expressions were observed using immunohistochemical method. Transforming growth factor-beta 1 (TGF-beta 1), P-Smad2, P-Smad3, and Smad7 expressions were detected by Western blot. TGF-beta 1, transforming growth factor-beta receptor I (TbetaRI), collagen-I (Col-I), and collagen-III (Col-III) mRNA expressions were detected by Real-time fluorescent quantitative PCR.</p><p><b>RESULTS</b>alpha-SMA expressions were obviously enhanced, E-cadherin expressions obviously weakened, the protein expression levels of P-Smad2 and P-Smad3, protein and mRNA expression levels of TGF-beta 1, TbetaRI mRNA, Col-I mRNA, and Col-III mRNA obviously up-regulated, Smad7 protein expressions obviously decreased (all P<0.01) in the renal tissues of rats in the UUO group. In the 1st curcumin group and the 2nd curcumin group, the protein expressions of alpha-SMA, P-Smad2, P-Smad3, and TGF-beta 1; mRNA expression levels of TGF-beta 1, TbetaRI mRNA, Col-I, and Col-III obviously decreased (P<0.05, P< 0.01), while the protein expressions of E-cadherin and Smad7 obviously increased (P<0.05, P<0.01).</p><p><b>CONCLUSIONS</b>Curcumin could intervene several sites of the TGF-beta/Smads signal transduction pathway of UUO rats, inhibit the occurrence of EMT of renal tubular epithelial cells, thus attenuating the tubulo-interstitial fibrosis.</p>

ABSTRACT

Objective: To screen for the genes associated with metastasis of head and neck squamous cell carcinoma (HNSCC) by studying the specimens of HNSCC. Methods: Eighty HNSCC specimens, including 17 involving the tongue, 13 involving mouth, 14 involving oropharynx, and 36 involving hypopharynx, were subjected to single nucleotide polymorphism chip analysis. The thirty-nine patients with distant metastasis during follow-up were taken as experimental group and the 41 without distant metastasis were taken as controls. Cox regression analysis was used to examine the influence of single nucleotide polymorphism (SNP) on distant metastasis of HNSCC. Results: The results of SNP demonstrated a distinct peak of frequent genomic gain at 11q13. Cox analysis of the array data showed the following results: a relative risk of T allele of rs965l738 as the SNP probe of SHANK2 gene was 3.61(P = 0.003), relative risk of T allele of rs4980690 as the SNP probe of FGF4 gene was 3.63(P = 0.006), relative risk of T allele of rs7929885 as the SNP probe of ANO1 gene was 4.35(P = 0.008), relative risk of T allele of rs687660 as the SNP probe of PPFIA1 gene was 4.24(P=0.011), relative risk of T allele of rs660665l as the SNP probe of ORAOV1 gene was 3.04(P = 0.013), and relative risk of T allele of rs592412 as the SNP probe of CCND1 gene was 4.07(P = 0.013). Conclusion: It is indicated that the following genes at 11q13 can increase the risk of metastasis of HNSCC, including T allele of rs965l738 in SHANK2, T allele of rs4980690 in FGF4, T allele of rs7929885 in ANO1, T allele of rs687660 in PPFIA1, T allele of rs6606651 in ORAOV1, and T allele of rs592412 in CCND1.

ABSTRACT

The bioactive principles contained in blueberries (Vaccinium) are various kind of anthocyanins (anthocyanidins, or phenolic aglycone, conjugated with sugar), chlorogenic acid, flavonids, alpha-linolenic acid, pterostilbene, resveratrol, and vitamins. After oral administration, anthocyanins can pass through blood-brain barrier and thus appear in various organs and brain. Improve visual function by increasing rhodopsin regeneration and ocular health is the earliest reported bioactivities of anthocyanin. Recent studies demonstrated the benefit of blueberries to prevent the age-related chronic diseases such as cancer, diabeties, hyperlipidemia, hypertension, neurodegeneration, obesity, and osteoporosis through its apoptosis, antioxidant, antiinflammation, and antiangiogenesis effects. Blueberries can eradicate microorganisms for the prevention of symptomatic urinary tract infections in women. Thus, blueberries are recognized as one of the most nutritious foods and cultivated worldwide. However, how to prolong the shelving time of fresh fruit, well utilize the leaf and stem to isolate the bioactive chemicals, improve quality consistency of juicy and dry products, all should be further concerned.

ABSTRACT

<p><b>OBJECTIVE</b>To study on needling safe depth of Fengfu (GV 16) with CT, so as to provide reference for safe needling depth of Fengfu (GV 16) in clinical acupuncture treatment.</p><p><b>METHODS</b>Forty-one adult volunteers were divided into 3 groups, a thin person group, a moderate person group and a fat person group according to Luo's indexes, and computer-aided tomography (CT) was used to measure the needling depth of Fengfu (GV 16).</p><p><b>RESULTS</b>The safe depths of perpendicular needling of Fengfu (GV 16) were different for persons of different somatotypes. The safe needling depth was (27.73 +/- 3.45) mm for the thin person group, (30.78 +/- 2.90) mm for the moderate person group, and (33.39 +/- 4.27) mm for the fat person group.</p><p><b>CONCLUSION</b>The safe needling depth < or = the dangerous depth x 75% can be used for reference for the safe needling depth of Fengfu (GV 16) for different somatotypes persons.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To assess the health safety of copper, steel and plastic water pipes by field water quality investigations.</p><p><b>METHODS</b>Four consumers were randomly selected for each type of water pipes. Two consumers of every type of the water pipes had used the water pipes for more than 1 year and the other 2 consumers had used the water pipes for less than 3 months. The terminal volume of tap water in copper and steel water pipes should be not less than 0.1 liter, whereas that in plastic water pipes should be not less than 1 liter.</p><p><b>RESULTS</b>The mean values of the experimental results in the second field water quality investigation of the copper and steel water pipes met the Sanitary Standards for Drinking Water Quality. The items of water sample of the plastic water pipes met the requirements of the Sanitary Standards for Drinking Water Quality.</p><p><b>CONCLUSION</b>Copper, steel, and plastic pipes can be used as drinking water pipes.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the early effects of coal dust on lung function in new underground coal miners.</p><p><b>METHODS</b>Two hundred and eighty-seven male miners were selected from new employees at the Xuzhou Mining Group Company as study group, and 132 male students at a mining technical school were selected as control. Data collection included: individual demographic parameters, family medical history, occupational history, and smoking history, measurement of dust concentrations in work areas, and lung function tests. This prospective cohort study took place over 3 years during which time total dust and respirable dust concentrations in the new coal miners' work areas were measured twice each month. For both miner and student groups, FVC and FEV(1) were tested initially before dust exposure, and then 15 times over the 3 years.</p><p><b>RESULTS</b>The average total dust and respirable dust concentrations in the miners' work areas were 23.8 mg/m(3) and 8.9 mg/m(3) respectively, which greatly exceeded national health criteria. During the first year of dust exposure, the miners's average FVC was higher than that of the controls (5.19 L vs 4.92 L, P < 0.01). During the 2nd and 3rd year the difference in average FVC between miners and control group was not significant (5.14 L vs 5.12 L, P > 0.05). Before dust exposure, the miners' FEV(1) was significantly higher than that of the control group (4.48 L vs 4.28 L). In the miners group, FEV(1) declined rapidly during the first year following dust exposure (from 4.48 L to 4.25 L), and in the 2nd and the 3rd year the average FEV(1) of the miners was significantly lower than that of controls (4.34 L vs 4.56 L, P < 0.01), although there were some fluctuations during the follow-up period. Overall, the average FEV(1) of miners group showed a significant decline during the study. There were significant correlations between FVC or FEV(1) and age, height, weight, and smoking. The three-year total loss of FVC and FEV(1) in smoking miners (154 ml, 184 ml) were greater than in non-smoking miners (83 ml, 91 ml).</p><p><b>CONCLUSION</b>There are apparent effects of coal dust on lung function in new underground coal miners, with FEV(1) being more impacted than FVC. Smoking may aggravate the effect of dust exposure on reducing lung function.</p>

<p><b>OBJECTIVE</b>To provide reference for the safe needling depth of Dazhui (GV 14), Jianzhongshu (SI 15), Xuanshu (GV 5) and Mingmen (GV 4) in clinical acupuncture and moxibustion treatment.</p><p><b>METHODS</b>Thirty-two adult volunteers were divided into 3 groups, thin person group, moderate person group and fat person group according to Luo's indexes, and computer-aided tomography was used to measure the needling depth of Dazhui (GV 14), Jianzhongshu (SI 15), Xuanshu (GV 5) and Mingmen (GV 4).</p><p><b>RESULTS</b>The safe depths of perpendicular needling were different for persons of different somatotypes, for example the needling depth for Dazhui (GV 14) was (32.86 +/- 3.96) mm for the thin person group, (37.76 +/- 4.91) mm for the moderate person group, and (47.93 +/- 5.30) mm for the fat person group.</p>
