ABSTRACT

ObjectiveTo explore the intervention effect and mechanism of Buzhong Yiqiwan (BZYQ) on colitis mice. MethodSixty-four C57BL/6 mice were randomly divided into 2 weeks blank group, 2 weeks model group, 2 weeks high-dose BZYQ (12 g·kg-1) group, 2 weeks low-dose BZYQ (6 g·kg-1) group, 4 weeks blank group, 4 weeks model group, 4 weeks high-dose BZYQ (12 g·kg-1) group, and 4 weeks low-dose BZYQ (6 g·kg-1) group. The colitis model was induced in mice by feeding 3% dextran sodium sulfate (DSS) for 7 days. The mice received BZYQ (12 and 6 g·kg-1) by gavage on the 8th day after modeling, once per day, and sacrificed on the 2nd and 4th weeks, correspondingly. The colon length and weight of mice in each group were measured. Hematoxylin-eosin (HE) staining was used for pathological observation and colonic mucosal inflammation was scored. The mRNA and protein expression of NOD-like receptor thermoprotein domain 3 (NLRP3), apoptosis-associated speck-like protein containing a CARD (ASC), and cysteinyl aspartate-specific protease-1 (Caspase-1) was detected by real-time quantitative polymerase chain reaction (Real-time PCR) and Western blot, respectively. Enzyme-linked immunosorbent assay (ELISA) was used to detect the content of inflammatory cytokines, such as interleukin (IL)-1β, IL-18, and IL-33 in colonic tissues. ResultCompared with the 2 weeks blank group, the 2 weeks model group showed shortened colon length, increased colon weight (P<0.05), loss of epithelial cells, destruction of gland structure, infiltration of a large number of inflammatory cells in mucosa and submucosa, local crypt abscess, and increase in mucosal inflammation score (P<0.01) as revealed by light microscopy, elevated levels of IL-1β, IL-18, and IL-33 in colonic tissues (P<0.05), and increased mRNA and protein expression of NLRP3, ASC, and Caspase-1 (P<0.05). The intervention of BZYQ (12 g·kg-1) restored colon length, alleviated mucosal injury (P<0.05), down-regulated the content of IL-18 (P<0.05), reduced the mRNA expression of NLRP3 and ASC as well as the protein expression of ASC and Caspase-1 compared with the conditions in the 2 weeks model group. Compared with the 4 weeks blank group, the 4 weeks model group showed decreased colon length, increased colon weight (P<0.05), decreased glands in the mucosal layer, expansion of glandular cavity, atrophy of crypt, local connective tissue hyperplasia and lymphocyte infiltration, increased inflammation score (P<0.01) as revealed by the light microscopy, increased expression of IL-1β, IL-18, and IL-33 (P<0.05), and elevated mRNA and expression of NLRP3 inflammasome complex (P<0.05). Compared with the conditions in the 4 weeks model group, the intervention of BZYQ (12 and 6 g·kg-1) could improve colon length and weight (P<0.05), and the intervention of BZYQ (12 g·kg-1) could also improve the inflammation score of the colon (P<0.05). Different from the acute stage, the intervention of BZYQ (12 and 6 g·kg-1) increased the content of IL-33 in the intestinal mucosa and up-regulated the mRNA and protein expression of NLRP3 inflammasome complexes ASC and Caspase-1 (P<0.05). ConclusionBZYQ can relieve the injury of colitis induced by DSS in mice. The mechanism is related to the regulation of intestinal immune response mediated by NLRP3 inflammasome, and it has different regulatory effects in acute and chronic inflammation stages.

ABSTRACT

OBJECTIVE:To study the effects a nd mechanism of X iaozheng decoction on the proliferation and migration of uterine leiomyoma cells in rat. METHODS :Female SD rats were randomly divided into normal control group ,model group , chemical medicine positive control group (Mifepristone tablets ,2.25 mg/kg),TCM positive control group (Guizhi fuling capsules , 200 mg/kg),Xiaozheng decoction low-dose ,medium-dose and high-dose groups (1.4,2.8,5.6 g/kg),with 15 rats in each group. Except for normal control group ,other groups were given intramuscular injection of estrogen and progesterone to induce uterine leiomyomas model. On the second day after modeling ,rats in administration groups were given relevant medicine intragastrically ; normal control group and model group were given constant volume of normal saline intragastrically ,once a day ,for consecutive 4 months. After last administration ,the uterus was removed and its morphology was observed ;the uterine coefficient was calculated. Uterine leiomyoma cells or uterine smooth muscle cells were isolated and cultured. The proliferation rate and migration rate of cells were detected by MTT method and cell scratch test ;flow cytometry was used to detect the cell apoptosis rate ;mRNA expression of HMGB 1 were detected by RT-qPCR. The expression of phosphorylated protein kinase B (p-Akt),nuclear factor κB inhibitor α (IκBα)and phosphorylated transforming growth factor β-activated kinase 1(p-TAK1)were detected by ELISA ;the protein expression of HMGB 1,phospholipid 3 kinase(PI3K),p-Akt in cytoplasm and nuclear factor κB p65(NF-κB p65)in nucleus were detected by Western blot assay. RESULTS :Compared with normal control g roup, the myometrium of the model group was significantly thickened ,the number of uterine smooth muscle cells were significantly increased and the sizes were 83777930。E-mail:jianghua_ouyang@126.com different, the arrangement of muscle fibers in so me areas was disordered, the uterine coefficient ,and the relative expression of HMGB 1 mRNA and protein were increased significantly (P< 0.01). Compared with model group ,the thickening of uterine myometrium and other symptoms were improved to different extents in Xiaozheng decoction groups and positive control groups ;the uterine coefficient ,cell proliferation rate ,migration rate ,mRNA and protein expression of HMGB 1,the expression of p-Akt and IκBα,protein expression of PI 3K and p-Akt (except for Xiaozheng decoction low-dose group )in cytoplasm were all decreased significantly (P<0.05 or P<0.01);the cell apoptosis rate ,the expression of p-TAK 1,protein expression of NF-κB p65 in nucleus were all increased significantly (P<0.05 or P<0.01). The effects of Xiaozheng decoction showed a dose-dependent trend. CONCLUSIONS :Xiaozheng decoction can inhibit the proliferation and migration of uterine leiomyoma cells by down-regulating the expression of HMGB 1,PI3K and p-Akt ,up-regulating the expression of NF-κB p65,so as to promote cell apoptosis.

ABSTRACT

Objective:To investigate the difference between the concentration of the peripheral ionized calcium (iCa) monitored at different blood collection points and the target concentration of anticoagulant efficacy in patients with continuous renal replacement therapy (CRRT), so as to provide scientific basis for the best blood collection point in clinical practice. Methods:Taking patients of department of critical care medicine, Union Hospital, Tongji Medical College, Huazhong University of Science and Technology receiving CRRT therapy with 4% citrate anticoagulation as research objects. Type of Prisma-FlexV8CRRT as well as department self-made substituate and dialyzate were adopted for all patients receiving CRRT therapy. Patients were divided into continuous veno- venous hemofiltration (CVVH) group ( n=10) and continuous veno- venous hemodialysis (CVVHD) group ( n=30) depending on their actual conditions and treatment needs. Blood collection was conducted at specific sites for extracorporeal peripheral ionized calcium concentration determination before and after the filter at the time of 2, 4, 8, 14, 20 hours following CRRT therapy for patients from both groups. Target concentration of extracorporeal peripheral ionized calcium was set as 0.2-0.4mmol/L for ensuring the efficacy of extracorporeal citrate anticoagulation. Results:Totally 400 testing results were obtained from 40 included patients during their treatment. In CVVH group, 100 testing results were obtained at the time of 2, 4, 8, 14, 20 hours following CRRT therapy and no significant statistical difference was shown ( P>0.05). In CVVHD group, 300 testing results were obtained at the time of 2, 4, 8, 14, 20 hours following CRRT therapy. The iCa concentration before the filter were (0.53±0.01), (0.50±0.01), (0.52±0.01), (0.53±0.01), (0.53±0.02) mmol/L while the iCa concentration after the filter were (0.41±0.01), (0.40±0.01), (0.39±0.02), (0.41±0.01), (0.40±0.01) mmol/L accordingly, and the difference was statistically significant ( t values were 75.24-103.41, P<0.01). Conclusions:For patients receiving CRRT treatment with citrate anticoagulation in different CRRT mode, testing results obtained from blood collection sites before and after the filter could not reflect the efficacy of citrate anticoagulation correctly and simultaneously to ensure the secure use of the extracorporeal pipelines and filter. In CVVH mode, iCa concentration determined from blood samples collected from sites before and after the filter could refelct the efficacy of citrate anticoagulation equally, while in CVVHD mode, blood collection and determination are suggested to conduct at the site before the filter to faciliate the assessment of the citrate anticoagulation efficacy.

ABSTRACT

Objective To study the mechanism by which mechano growth factor (MGF) promotes the migration of mesenchymal stem cells (MSCs).Methods MSCs were isolated from Sprague-Dawley rats and treated with MGF at various concentrations.Western blotting was used to evaluate the expression of RhoA protein and its downstream p-MYPT,as well as p-FAK and t-FAK proteins related to focal adhesion kinase. The aim was to illustrate the effect of MGF in regulating the cytoskeleton of MSCs and the formation of focal adhesion.C3 toxin was used to inhibit RhoA activity and western blotting was used to examine the expression of p-MYPT,and the focal adhesion kinases p-FAK and t-FAK.Transwell assays were used to examine MSCs' migration ability, and immunofluorescence was conducted to examine the formation of F-actin cytoskeleton and focal adhesion. Results MGF can significantly promote the expression of MSCs' RhoA and its downstream protein p-MYPT.The effect is dose-dependent.The expression of RhoA and p-MYPT increased most significantly at 50 μM concentration. The ratio of p-FAK to t-FAK indicates that MGF can activate focal adhesion kinase and promote adhesion.C3 toxin significantly inhibited FAK activation.Transwell assays showed that MGF can significantly promote MSC migration, but pretreatment with C3 toxin inhibited it.The immunofluorescence results show that MGF can promote the rearrangement of MSCs' F-actin cytoskeleton and the formation of focal adhesion.C3 toxin disrupted MSCs cytoskeletons and decreased focal adhesion. Conclusion MGF promotes MSCs' migration through RhoA-and kinase-mediated cytoskeleton rearrangement and the formation of focal adhesion.

ABSTRACT

BACKGROUND: Constipation, especially that caused by spleen insufficiency in transportation, often not responses to common purgatives, however, the therapy to strengthen the spleen and regulate qi so as to promote transportation and dredge stasis may be an ideal way to resolve the problem.OBJECTIVE: To investigate the effect of zhizhu decoction, a classic famous prescription, on gastric motility of mice with spleen insufficiency constipationDESIGN: A randomized controlled trial.SETTING: Department of Traditional Chinese Medicine (TCM) of Guangdong Medical College and Spleen-stomach Institute of Guangzhou University of Traditional Chinese Medicine.MATERIALS: The experiment was completed from June to December2002 at Spleen-stomach Institute of Guangzhou University of Traditional Chinese Medicine. Totally 328 NIH mice of general grade were chosen.METHODS: Mouse models with spleen insufficiency constipation were set up by irregular diet and overstrain combined with water deprivation. The influence of zhizhu decoction in large, middle and small doses on defecation in both spleen insufficiency mice and normal mice was investigated, taking Chinese patent drug bianmi tong (bowel-relaxing) as a positive control. In observation of normal mice defecation, there were five groups of large dose, middle dose and small dose of zhizhu decoction, bianmi tong and normal control. In observation of defecation of spleen insufficiency constipation mice, there were six groups: the above five and a natural recovering. In observation of loading mice trial, besides the above five groups, there was an atropine group. After administration, the time when the mice first defecated dark stool, the number of stool grains and the stool quantity were observe;and the percentage of small intestine advance was calculated.MAIN OUTCOME MEASURES: [1] Observation of the effect of zhizhu decoction on the defecation in both normal and spleen insufficiency mice.[2] The influence of zhizhu decoction on bowel advance in both normal and atropine loading miceRESULTS: Totally 328 mice were involved in the trial and 319 entered into the final result analysis, the other 9 died because of mistake in administration by gavage. The effect of zhizhu decoction on the defecation: [1] For normal mice, compared with the distilled group, in the zhizhu decoction groups of various doses the time when the mice first defecated dark stool was obviously reduced, and the number of stool grains and stool quantity were obviously increased, but the effect in the large and middle dose groups was best. [2] Compared with the normal group, in the model group the time when the mice first defecated dark stool was obviously increased, and the number of stool grains and stool quantity were obviously decreased. Zhizhu decoction in large and middle doses could obviously reduce the time when the mice first defecated dark stool, and increase the number of stool grains and stool quantity, the effect was better than that of the control drug bianmi tong. The influence ofzhizhu decoction on intestinal advance in mice: [1]Zhizhu decoction in various doses could obviously increase the rate of intestinal advance compared with distilled water group for normal mice. [2]Adding atropine load could reduce the rate of intestinal advance compared with the normal control group, and the percentage in all zhizhu decoction groups was obviously increased compared with the atropine group.CONCLUSION: Zhizhu decoction can reduce the time when the mice first defecated dark stool, increase the number of stool grains and stool quantity were obviously increased to both normal mice and mice with spleen insufficiency mice; it can also increase the intestinal advance rate in normal mice, and had a good antagonism against the inhibition of atropine to intestinal advance. In the trials the effect of zhizhu decoction in large and middle doses was more obvious, with a certain dose-effect relationship, the effect was better than that of the control drug bianmi tong.

[Methods] CAG rat models were established with comprehensive method and then were given BASR herbs. Argyrophilic protein of nucleolar organizer regions ( AgNOR) count was carried out after silver nitrate staining. The expression of proliferating cell nuclear antigen (PCNA) was examined with immunohistochemical method and apoptosis was detected by in-situ cell-end labeling method. [Results] The number of AgNOR and the indexes of PCNA and apoptosis were increased in CAG model rats ( P

ABSTRACT

Objective To explore the changes of proteins level of TLR2 in colonic mucosa of ulcerative colitis( UC) model rats and to observe the effects of Kuijieling Decoction( KD) on TLR2. Methods UC model rats were induced by TNBS. The rats were randomly divided into six groups as follows: normal control (NC) group, model control (MC) group, lowdose Kuijieling (KLD) group, mediumdose Kuijieling( KMD) group, high- dose Kuijieling (KHD) group and SASP group. After 10- days treatment the rats were killed to get their colonic mucosa and extract the whole- cell proteins. Western blot technique was used to detect the TLR2 protein level. ? - actin was used as the internal index. Relative expression level of target protein was calculated from the gray scale ratio of TLR2 and ? - actin. Results Relative protein expression level of TLR2 in MC group was significantly higher than that in NC group (P

ABSTRACT

Objective:To analyze quantitatively individual baseline drift of immunoglobulin and complement within 24 h.Methods:Blood samples were drawn from adult volunteers on day 1 and day 2 at 7.00 am.Immunoglobulin and complement including IgG,IgA,IgM and C3,C4 were measured with automatic immunobiochemistry analyzer. The rate of changes between two times of measured value, baseline drift, could be calculated. Results:The individual baseline drifts of measured values were from 6.12% to 7.97% (mean=7.22%) in 5 immunological parameters. There were significations between two groups within 5 immunological parameters (P
