ABSTRACT

OBJECTIVES@#The effect of isoprenylcysteine carboxymethyltransferase (ICMT) silencing on the migration and invasion of tongue squamous cell carcinoma was investigated by constructing the small interfering RNA (siRNA) of ICMT.@*METHODS@#Through liposomal transfection, siRNA was transfected into human tongue squamous cell carcinoma CAL-27 and SCC-4 cells (ICMT-siRNA group) with a negative control group (transfected with NC-siRNA) and a blank control group (transfected with a transfection reagent but not with siRNA). Quantitative real-time polymerase chain reaction was performed to analyze the mRNA expression of ICMT and RhoA in each group of cells after transfection and to measure the silencing efficiency. Western blot was applied to examine the expression levels of ICMT, total RhoA, membrane RhoA, ROCK1, matrix metalloproteinase (MMP)-2, and MMP-9 proteins in each group. The migration and invasion abilities were evaluated via wound healing and Transwell motility assays.@*RESULTS@#After CAL-27 and SCC-4 cells were transfected with ICMT-siRNA, the expression levels of ICMT genes and proteins decreased significantly in the experimental group compared with those in the negative and blank control groups (@*CONCLUSIONS@#The migration and invasion abilities of CAL-27 and SCC-4 cells were reduced significantly after the transfection of ICMT-siRNA, and the involved mechanism might be related to the RhoA-ROCK signaling pathway.

ABSTRACT

OBJECTIVES@#This study aimed to explore the effects of silencing isoprenylcysteine carboxyl methyltransfe-rase (Icmt) through small interfering RNA (siRNA) interference on the proliferation and apoptosis of tongue squamous cell carcinoma (TSCC).@*METHODS@#Three siRNA were designed and constructed for the Icmt gene sequence and were then transfected into TSCC cells CAL-27 and SCC-4 to silence Icmt expression. The tested cells were divided as follows: RNA interference groups Icmt-siRNA-1, Icmt-siRNA-2, and Icmt-siRNA-3, negative control group, and blank control group. The transfection efficiency of siRNA was detected by the fluorescent group Cy3-labeled siRNA, and the expression of Icmt mRNA was screened by quantitive real-time polymerase chain reaction (qRT-PCR) selected the experimental group for subsequent experiments. The expression of Icmt, RhoA, Cyclin D1, p21, extracellular regulated protein kinases (ERK), and phospho-extracellular regulated protein kinases (p-ERK) were analyzed by Western blot. The proliferation abilities of TSCC cells were determined by cell counting kit-8 assay. The change in apoptosis was detected by AnnexinV-APC/propidium staining (PI) assay. Cell-cycle analysis was conducted by flow cytometry.@*RESULTS@#The expression of Icmt mRNA and protein in TSCC cells significantly decreased after Icmt-siRNA transfection (@*CONCLUSIONS@#Silencing Icmt can effectively downregulate its expression in TSCC cells, reduce the RhoA membrane targeting localization and cell proliferation, and induce apoptosis. Thus, Icmt may be a potential gene therapy target for TSCC.

OBJECTIVE@#To explore the kinetics of infiltrated T cell in murine acute graft-versus-host disease (aGVHD) target organs after allogeneic hematopoietic stem cell transplantation (allo-HSCT) and its relationship with tissue pathological damage and aGVHD progress.@*METHODS@#Male C57BL/6 (H-2K@*RESULTS@#Compared with BMT group, the number of infiltrated T cells in aGVHD target organs including liver, lung and gut increased since day 7 in BMT+T group (P<0.05). On day 14, 28, 40 and 47 after transplantation, more infiltrated CD3@*CONCLUSION@#Pathological damage of aGVHD target organs is induced by CD3

ABSTRACT

A wearable wireless health monitoring system for drug addicts in compulsory rehabilitation centers was proposed. The system can continuously monitor multiple physiological parameters of drug addicts in real time, and issue early warning information when abnormal physiological parameters occur, so as to play the role of timely medical practice. In addition, this study proposes a convolutional neural network (CNN)model, which can evaluate the health status of drug addicts based on multiple physiological parameters. Experiments show that the model can be applied to the task of body state recognition in the open physiological parameter data set, and the recognition accuracy can reach up to 100% in a single physiological parameter data set; when the whole physiological data set is used, the recognition accuracy can reach 99.1%. The recognition accuracy exceeds the performance of the traditional pattern recognition method on this task, which verifies the superiority of the model.

ABSTRACT

Objective:To analyze the clinical value of capsule endoscopy (CE) in small intestinal diseases and to explore the correlation between the diagnosis and clinical characteristics.Methods:From June 2016 to April 2019, a total of 194 patients receiving CE were enrolled in the Department of Gastroenterology, Shanxi People′s Hospital, including 68 patients with obscure gastrointestinal bleeding, 104 with chronic abdominal pain and 22 with diarrhea.Results:The detection rate of small intestinal lesions by CE was 53.6%(104/194), of which gastrointestinal bleeding was 66.2% (45/68), chronic abdominal pain was 53.8%(56/104), and diarrhea was 13.6%(3/22). The first three positive diagnostic criteria of CE for gastrointestinal bleeding were non-specific inflammation, small intestinal ulcer and vascular malformation. CE findings in patients with abdominal pain were non-specific inflammation, small intestinal ulcer and Crohn′s disease; while the major endoscopic presentations in patients with diarrhea were small intestinal ulcer and non-specific inflammation. Logistic regression analysis suggested that the clinical manifestations and gender were correlated with positive diagnosis by CE ( P<0.05), and the positive diagnostic rate of CE in female patients was significantly lower than that in male patients [31.6%(25/79) vs. 51.3%(59/115), P<0.05]. The diagnostic rates of CE in patients with gastrointestinal bleeding and chronic abdominal pain were similar [55.9%(38/68) vs. 41.4%(43/104)], which was significantly higher than those with diarrhea [9.1%(2/22), P<0.05]. Conclusions:Capsule endoscopy has a high detection rate for small intestinal diseases, which could be the first choice in patients with unexplained gastrointestinal bleeding. Male patients and gastrointestinal bleeding are predictors of positive findings by CE.

ABSTRACT

Objective:To introduce a novel method of the virtual construction for pedicle screw trajectory (PST) of subaxial cervical spine based on CT data and to investigate its effects on improving the accuracy rate of subaxial cervical pedicle screw technique.Methods:The CT data of cervical spine (C 3, 4) of 60 subjects, including 30 males and 30 females, were collected for analysis. The data were imported into Mimics 20.0. Further, the virtual safe core (G-zone) of the PST was constructed using the method designed by authors. The following data was collected: 1) the efficiency rate of the method for virtual constructed of PST and G-zone; 2) the spatial position of the virtual safe core, including the distance between G-zone and the tangent line of upper and lower outer edge of Luschka's joint on the coronal plane, and the distance between the G-zone and the posterior edge of the vertebral body on sagittal plane; 3) the length, width and height of G-zone; 4) the abduction angle of pedicle screw on axial plane, and the cranial and caudad inclination angle on the sagittal plan of the pedicle; 5) the mean distance between the cortex of lateral mass and the G-zone; 6) the positive projection area of the trajectory on the coronal plan. Results:The efficiency rate of the method for virtual constructed of PST and safe core were C 3 95% (57/60) and C 4 100% with 97.5% (117/120) the total efficiency rate. The mean height of safe core was C 3 4.99±1.04 mm (left) and 4.95±0.88 mm (right), C 4 5.41±0.93 mm (left) and 5.16±1.39 mm (right). The mean width of safe core was C 3 3.79±0.87 mm (left) and 3.44±0.88 mm (right), C 4 3.99±0.68 mm (left) and 3.60±0.92 mm (right). The mean length of safe core was 3.5 mm. The distance between the G-zone and the tangent line of upper and lower outer edge of Luschka's joint on the coronal plane was C 3 0.66±0.54 mm (left) and 0.69±0.67 mm (right), C 4 0.62±0.59 mm (left) and 0.65±0.64 mm (right). The distance between the G-zone and the posterior edge of the vertebral body on sagittal plane was C 3 2.08±0.41 mm and C 4 2.34±0.60 mm, C 4>C 3 ( P<0.05). The interval of abduction angle on the axial plane of C 3 were 28.16°±7.82° to 67.46°±7.54° (left) and 29.46°±7.5° to 64.08°±9.79° (right), C 4 27.64°±4.56° to 69.95°±6.66° (left) and 29.17°±5.92° to 71.14°±5.07° (right). The range of cranial and caudad inclination angle of pedicle screw on sagittal plane were C 3 24.12°±5.35° (left) and 24.40°±4.86° (right), C 4 24.87°±5.7° (left) and 25.65°±5.62° (right). The mean distance between the entering cortex and the center of G-zone was C 3 11.93±0.76 mm (left) and 12.12±0.83 mm (right), C 4 11.24±0.71 mm (left) and 11.34±1.01 mm (right). The positive projection area of the PST on the coronal plan was C 3 41.38±15.68 mm 2 (left) and 37.66±13.77 mm 2 (right), C 4 44.54±12.96 mm 2 (left) and 40.33±15.54 mm 2 (right), C 4 left>C 4 right ( P<0.05). Conclusion:The efficiency rate of the virtual construction method for PST and safe core of the subaxial cervical spine was 97.5%. The length, width, and height of the virtual safe core were suitable for the implantation of the commonly used cervical pedicle screw with a diameter of 3.5 mm. The virtual G-zone was morphologically stable and easy to recognize on the anteroposterior and lateral cervical fluoroscopy view, which can provide a reliable landmark for the implementation of the pedicle screw of the subaxial cervical spine.

ABSTRACT

Objective:To study the clinical course and underlying causes of early severe neurological complication (ESNC) after pediatric liver transplantation.Methods:A retrospective study was conducted on 309 pediatric liver transplantation recipients treated at Tianjin First Central Hospital from January 2018 to December 2018. ESNC occurred in 15 patients (4.8%, 15/309) within 1 month after liver transplantation. There were 7 males and 8 females, aged from 6 to 46 months. Liver transplantation was carried out for biliary atresia ( n=12), fulminant liver failure ( n=1), Niemann-Pick disease ( n=1) and Alagille syndrome ( n=1). The causes of ESNC and the prognosis were analyzed. Results:The onset of ESNC was 10.7 (0-28) d after liver transplantation. Twelve patients developed encephalopathy with epilepsy in 2 patients. Four of these patients were caused by severe infection, 4 by heart failure combined with respiratory failure which led to ischemic hypoxic encephalopathy, 2 by transplant liver failure, 1 by intracranial infection, and 1 by severe brain swelling which led to brain death. Epilepsy occurred in 3 patients, 2 caused by neurotoxicity of calcineurin inhibitors, and 1 caused by reversible posterior leukoencephalopathy syndrome. Three children with ESNC died after operation, including 1 brain death, 1 due to severe heart failure and 1 due to severe infection.Conclusions:ESNC occurred in 4.8% of patients which seriously affected long-term prognosis of patients. Measures to reduce the incidence of ESNC include prevention of postoperative infection and drug toxicity, and good control of cardiac insufficiency.

ABSTRACT

Objective To explore the application of support vector regression (SVR) model combined with meteorological and air pollutants index in the prediction of the cases of bacillary dysentery in Lanzhou City, so as to provide scientific reference for the prevention and control of bacillary dysentery.Methods Time series data of the reported cases of bacillary dysentery from December 2013 to August 2016, combined with the meteorological and air pollutants data, were used as training set to fit support vector regression model. The data from September 2016 to December 2017 was used as validation set to verify the model and compare the effect in fit and prediction with different models. Results A total of 7 192 bacillary dysentery cases were reported in Lanzhou City from 2013 to 2017. The correlation coefficient of meteorological and pollution factors with the cases of bacillary dysentery was more than 0.4, except air pressure. The parameters of the fit model were selected based on the integrated data, acquiring the three parameters with the smallest test error were C=5, γ=0.02 and ε=0.000 1, respectively. The validation set was used to test the different models, which showed that the integrated data model had the best predictive accuracy and robustness . The root mean squared error (RMSE) was 0.164 7 and the mean absolute percentage error (MAPE) was 16.405%. Conclusion SVR model combined with meteorological and air pollutants index is effective in the prediction of bacterial dysentery.

OBJECTIVE@#To investigate the computed tomography findings, clinicopathological features, genetic characteristics and prognosis of in situ adenocarcinoma (AIS) and minimally invasive adenocarcinoma (MIA) of the lung.@*METHODS@#We retrospectively analyzed the data including computed tomography (CT) images, histopathological findings, Ki-67 immunostaining, and genetic mutations in patients with lung adenocarcinoma undergoing surgery at our hospital between 2014 and 2019.@*RESULTS@#Of the total of 480 patients with lung adenocarcinoma we reviewed, 73 (15.2%) had AIS (=28) or MIA (=45) tumors. The age of the patients with MIA was significantly younger than that of patients with AIS ( < 0.02). CT scans identified pure ground-glass nodules in 46.4% of AIS cases and in 44.4% of MIA cases. Multiple GGOs were more common in MIA than in AIS cases ( < 0.05), and bluured tumor margins was less frequent in AIS cases ( < 0.05). No significant difference was found in EGFR mutations between MIA and AIS cases. A Ki-67 labeling index (LI) value ≥2.8% did not differentiate MIA from AIS. The follow-up time in MIA group was significantly shorter than that in AIS group, but no recurrence or death occurred.@*CONCLUSIONS@#Despite similar surgical outcomes and favorable survival outcomes, the patients with AIS and MIA show differences in terms of age, CT findings, EGFR mutations and Ki-67 LI.

ABSTRACT

The chemical constituents from methanol extract of Dichroa hirsuta were separated by silica gel and Sephadex LH-20 column chromatography,high pressure preparative liquid chromatography( HPLC) and recrystallization. Their structures were elucidated by NMR and MS. Nine compounds were obtained and their structures were identified as 3β,21α-O-diacetyl-lup-9( 11)-en-7β-ol( 1),( Z)-methyl p-hydroxycinnamate( 2),cis-p-coumaric acid ethyl ester( 3),( E)-methyl p-hydroxycinnamate( 4),trans-p-coumaric acid ethyl ester( 5),4( 3 H)-quinazolinone( 6),7-hydroxycoumarin( 7),hydrangenol( 8) and thunberginol C( 9). Compound 1 is a new lupane-type triterpenoid,and compounds 1-5,8-9 were firstly isolated from this plant. Dual reporter assay results showed that compounds 2-5 could activate the Nrf2-ARE signaling pathway.

ABSTRACT

OBJECTIVE@#This study aimed to explore the influence of Rce1 on invasion and migration of tongue squamous cell carcinoma cells by silencing the Rce1 gene with RNA interference.@*METHODS@#The tongue squamous cell carcinoma Cal-27 and SCC-4 cells were cultured in vitro. The small interfering RNA (siRNA) of the Rce1 gene was designed, and the Rcel gene expression was silenced vialiposome transfection. According to the siRNA transfected by liposome, the experimental group was divided into three groups, namely, Rce1-siRNA-1, Rce1-siRNA-2, and Rce1-siRNA-3 groups. Negative control group was transfected by siCON, and the blank control group was untransfected by siRNA. The Rce1, RhoA, and K-Ras gene expression levels in each group were analyzed by real-time quantitative polymerase chain reaction. The Rce1, RhoA, K-Ras, MMP-2, and MMP-9 protein expression levels were analyzed by Western blot. The invasiveness of tongue cancer cell Cal-27 and SCC-4 were determined by Transwell invasion assay, and cell migration assay was performed by cell scratch assay.@*RESULTS@#Real-time quantitative polymerase chain reaction and Western blot results showed that compared with the negative and blank control groups, the Rce1 gene and protein expression levels in three experimental groups decreased (P0.05). Meanwhile, the MMP-2 and MMP-9 expression levels decreased (P<0.05). Transwell invasion assay results showed that the total number of cells in the PET film of the experimental groups was significantly decreased compared with the control group (P<0.05). The cell scratch test showed that the cell closure time of the scratch in the interference group was significantly longer than those in the control and blank groups (P<0.05).@*CONCLUSIONS@#Silencing Rce1 in vitro can effectively downregulate its expression in tongue squamous cell carcinoma cells Cal-27 and SCC-4 and reduce the migration and invasion abilities of these cells.

ABSTRACT

Objective@#To study the use of high flow nasal cannula oxygenation (HFNCO) on the respiratory function of patients with acute lung injury after liver transplantation.@*Methods@#This is a randomized controlled interventional study. Among 200 adults who underwent liver transplantation from Jan 2015 to Dec 2015 in the Department of Transplant Surgery, Tianjin First Central Hospital, 80 patients who did not meet the criteria for acute lung injury were excluded. Of the remaining 120 patients who were included in the study, they were divided into the HFNCO group and the nasal catheter group by the random number table method, with 60 patients in each group. The respiratory rate (RR), oxygenation index (PaO2/FiO2) and PaCO2 were recorded immediately after extubation, 2 h after extubation, 1st day after extubation and 2nd day after extubation. The reintubation rate and duration of ICU stay were compared.@*Results@#The incidence of acute lung injury after liver transplantation was 60.0%(120/200). There was no reintubation in the HFNCO group, and 6 patients (10.0%) in the nasal catheter group were reintubated. The difference between the two groups was statistically significant (P<0.05). The RR of the HFNCO group was lower than that of the nasal catheter group [(19.4±4.5) breaths/min vs. (21.7±5.1) breaths/min], [(18.9±4.2) breaths/min vs. (22.5±4.4) breaths/min], [(19.0±3.7) breaths/min vs. (23.2±4.7) breaths/min] (all P<0.05) 2 h after extubation, on the 1st and 2nd day after extubation. The oxygenation index of the HFNCO group was significantly higher than that of the nasal catheter group [(242.6±65.0)mmHg vs. (206.6±44.2)mmHg], [(245.6±70.4)mmHg vs. (211.1±62.8)mmHg], [(238.2±62.8)mmHg vs. (210.4±57.1)mmHg] (all P<0.05). There was no statistically difference in the PaCO2 and the duration of ICU stay between the two groups (P>0.05).@*Conclusions@#Compared with the conventional nasal catheter oxygen inhalation, HFNCO was more effective in improving oxygenation index, reducing RR, improving respiratory function and reducing reintubation rate in patients with acute lung injury after liver transplantation. HFNCO is a more suitable oxygen therapy for patients with acute lung injury after liver transplantation.

Objective To study the use of high flow nasal cannula oxygenation (HFNCO) on the respiratory function of patients with acute lung injury after liver transplantation.Methods This is a randomized controlled interventional study.Among 200 adults who underwent liver transplantation from Jan 2015 to Dec 2015 in the Department of Transplant Surgery,Tianjin First Central Hospital,80 patients who did not meet the criteria for acute lung injury were excluded.Of the remaining 120 patients who were included in the study,they were divided into the HFNCO group and the nasal catheter group by the random number table method,with 60 patients in each group.The respiratory rate (RR),oxygenation index (PaO2/FiO2) and PaCO2 were recorded immediately after extubation,2 h after extubation,1 st day after extubation and 2nd day after extubation.The reintubation rate and duration of ICU stay were compared.Results The incidence of acute lung injury after liver transplantation was 60.0% (120/200).There was no reintubation in the HFNCO group,and 6 patients (10.0%) in the nasal catheter group were reintubated.The difference between the two groups was statistically significant (P < 0.05).The RR of the HFNCO group was lower than that of the nasal catheter group [(19.4 ± 4.5) breaths/min vs.(21.7 ± 5.1) breaths/min],[(18.9 ± 4.2) breaths/min vs.(22.5 ± 4.4) breaths/min],[(19.0 ± 3.7) breaths/min vs.(23.2 ± 4.7) breaths/min] (all P <0.05) 2 h after extubation,on the 1st and 2nd day after extubation.The oxygenation index of the HFNCO group was significantly higher than that of the nasal catheter group [(242.6 ± 65.0) mmHg vs.(206.6 ±44.2)mmHg],[(245.6±70.4)mmHg vs.(211.1 ±62.8)mmHg],[(238.2±62.8)mmHg vs.(210.4±57.1)mmHg] (all P <0.05).There was no statistically difference in the PaCO2 and the duration of ICU stay between the two groups (P > 0.05).Conclusions Compared with the conventional nasal catheter oxygen inhalation,HFNCO was more effective in improving oxygenation index,reducing RR,improving respiratory function and reducing reintubation rate in patients with acute lung injury after liver transplantation.HFNCO is a more suitable oxygen therapy for patients with acute lung injury after liver transplantation.

ABSTRACT

Objective To analyze the effect of clinical application of ultrasound in microsurgical treatment of intramedullary tumors in the superior cervical spinal cord.Methods Retrospective study the clinical data of 15 patients with intramedullary tumors in the superior cervical spinal cord,which were underwent a laminectomy for microsurgical tumor resection during January,2014 and January,2018.Intraoperative ultrasound and neuromonitoring was accompanied by the whole surgical procedure for each case.The follow-up data was collected by outpatient department visits and telephone interviews.Results All the described patients were performed with microscopic tumor resection by using intraoperative neurophysiological monitoring and ultrasound.The pathological diagnosis was ependymocytoma (n=8) and astrocytoma (n=7).Gross total resections comprised 86.7% of cases (n=13),and subtotal resections 13.3% (n=2).The neurological outcome was as follows:Mc-Cormick scale grade Ⅰ,10 patients;grade Ⅱ,3 patients;grade Ⅲ,1 patient;and grade Ⅳ 1 patient;Follow-up was applied for (19.2±7.6) months in 13 cases and 12.0 months in 2 cases.Compared to the preoperative period,66.6% of patients recovered postoperatively,20.0% improved,6.7% remained without deficit and deterioration persisted in 6.7%.Conclusion The microscopic resection of tumors is the effective way to cure this disease.By using intraoperative neurophysiological monitoring and ultrasound,the complete tumor resection and the minimal spinal cord injury were certainly achieved.

ABSTRACT

Objective To explore the management of spontaneous intraspinal hematoma.Methods From January 2011 to July 2018,29 cases with spontaneous intraspinal hematoma were admitted to our department.Date on etiology,clinical presentation,radiological features,treatment strategy and prognosis were analyzed retrospectively.The prognosis was assessed by American Spinal Injury Association impairment scale (ASIA) before and after the treatment.Results Total of 29 cases,only 10 cases (34.5%) revealed specific etiology,including 7 cases of spinal vascular malformation,2 of tumor apoplexy,1 of cavernous hemangioma.After 2 weeks of conservative treatment,3 patients with grade D and 3 patients with grade E were assessed for spinal function.The average interval from onset to surgery was(9.4±7.5) days,the ASIA after two weeks of the operation was as follows:5 patients were assessed at grade A,5 patients at grade C,8 patients at grade D and 4 patients at grade E.28 patients were followed up for (48.7±23.1) months on average,6 patients without surgery were E,22 cases with surgery were as follows:4 cases A,18 cases D/E.Conclusions The etiology of spontaneous intraspinal hematoma is hard to define even after complete preoperative examination and exploratory operation.The preoperative neurologic functions are important predicting factors for the prognosis of spontaneous intraspinal hematoma.For patients who had neurologic function deficit,surgical treatment should be performed urgently to remove the hematoma and release the decompression of spinal cord.The majority of these patients can achieve a positive prognosis after surgery.

<p><b>OBJECTIVE</b>To investigate the relationship between serum albumin and renal function in patients with acute leukemia (AL) and its clinical significance.</p><p><b>METHODS</b>The clinical data and related test results of 267 newly diagnosed patients with acute leukemia from April 2015 to April 2017 were collected for retrospective cross-sectional analysis. Multivariate regression model was used for statistical analysis.</p><p><b>RESULTS</b>The creatinine level in serum of newly diagnosed acute leukemia patients decreased with the increase of albumin level (the first quartile-the fourth quartile had an average creatinine level of 72.0 µmol/L, 65.2 µmol/L, 62.8 µmol/L, 58.6 µmol/L); Multiple regression model results showed that each elevated albumin 1 g/L, the serum creatinine level decreased 0.89 µmol/L. The serum albumin was grouped into the model by quartile, and the first quartile was used as the reference group. With the increase of albunin, the β value decreased steply (the second and fourth quartile β values were -12.7, -14.81, -15.98), the trend line test p value was <0.05.</p><p><b>CONCLUSION</b>Serum albumin negatively correlats with creatinine level in newly diagnosed acute leukemia patients, and its elevation shows protective effect on renal function.</p>

ABSTRACT

Objective To study the effect of Shenfukang Ⅱ on kidney function and TGF-β1/Smads signal pathway with 5/6 nephrectomy model in rats. Methods Thirty SD rats were randomly divided into the sham operation group, model group and Shenfukang group according to a random number table. We used 5/6 nephrectomy to prepare animal models. The rats were given Shenfukang Ⅱ particles 450 mg/(kg?d) gavage, the others group were given equal volume of saline gavage, once a day, continuous 21 days. The rats were given gavage at the 9 th week after operation. The changes of renal function index BUN, SCR and UA of rats in each group were detected 21 days after administration. The expression of TGF-β1, TGFβR1, Smad7, pSmad2, pSmad3 in renal tissues were detected by immunohistochemistry. The Smad7, pSmad2 and pSmad3 detected the expression of in renal tissues by Western Blot. Results Compared with the model group, the content of BUN (15.03 ± 2.19 mmol/L vs. 24.30 ± 2.82 mmol/L), SCR (74.22 ± 7.36 μmol/L vs. 93.61 ± 14.73 μmol/L) in Shenfukang group significantly decreased (P<0.05). The expression of TGF-β1, TGFβR1, pSmad2 and pSmad3 significantly decreased in kidney tissue, while the expression of Smad7 significantly increased in kidney tissue. The expression of p-smad2 (0.47 ± 0.03 vs. 0.64 ± 0.04) and p-smad3 (0.43 ± 0.04 vs. 0.55 ± 0.02) significantly decreased (P<0.05), and the expression of Smad7 (0.54 ± 0.03 vs. 0.39 ± 0.02) significantly increased (P<0.05). Conclusions Shenfukang Ⅱ have improved on kidney function with 5/6 nephrectomy model rats. The study revealed that Shenfukang II may play an anti-fibrotic effect on regulating TGF-β1/Smads signaling pathway with 5/6 nephrectomies model.

ABSTRACT

<p><b>OBJECTIVE</b>To observe the alteration of QT dispersion (QTd) and QTc dispersion (QTcd) in hemodialysis patients after oral administration of Zhigancao Decoction (, Roasted Licorice Decoction, RLD).</p><p><b>METHODS</b>To investigate the alteration of QTd and QTcd in 68 routine hemodialysis patients before and after hemodialysis with 12-lead electrocardiogram (ECG) after orally administrated RLD for 4 weeks. Blood was also taken for measurement of plasma electrolytes, liver function, renal function, hemoglobin (Hgb) and hematocrit (HCT).</p><p><b>RESULTS</b>After hemodialysis, QTd and QTcd were prolonged evidently; the difference was significant between before and after hemodialysis (P<0.05). After RLD orally administrated for 4 weeks, QTd and QTcd only slightly increased after dialysis compared with pre-dialysis (P>0.05). The QTd and QTcd of the post-therapy-post-dialysis decreased significantly compared with the pre-therapy-post-dialysis (P<0.05). There were no other significant changes in other variables (post-therapy-pre-dialysis vs. pre-therapy-pre-dialysis, or post-therapy-post-dialysis vs. pre-therapy-post-dialysis;P>0.05). After therapy, the number of patients with supraventricular arrhythmia, occasional ventricular premature beat and multiple ventricular premature beat were decreased from 15 to 4, 10 to 2 and 7 to 1, respectively.</p><p><b>CONCLUSION</b>RLD therapy not only lowered the increased QTd and QTcd after hemodialysis, but also displayed a safety profile.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the effect of Statins on proliferation and apoptosis in human acute T lymphocytic leukemia (T-ALL) cells and its possible mechanism.</p><p><b>METHODS</b>Jurkat and CCRF-CEM cells were cultured in different concentrations of Fluvastatin and Simvastatin for 24 h respectively. Then, the cell growth inhibition level was defected by CCK-8; the DNA replication was analyzed by EdU; the cell apoptosis was analyzed by Annexin V/7-AAD double labeling; the cell cycle changes were analyzed by flow cytometry; the expressions of Cyclin D1, p21, p27, BAX, BCL-2 and p-Akt were determined by Western blot.</p><p><b>RESULTS</b>Fluvastatin and Simvastatin both significantly inhibited the growth of Jurkat and CCRF-CEM cells in a dose-dependent manner. The inhibitory rate of Jurkat and CCRF-CEM cells at 0.2 mmol/L Fluvastatin was 41.14% and 57.08% respectively, while the 0.2 mmol/L Simvastatin could supress 68.42% of Jurkat and 77.10% of CCRF-CEM cells. Half or more than half of cell inhibition were observed in Statins-treated groups with significantly statistical differences, compared with the control groups (P<0.05). After the Jurkat and CCRF-CEM cells were treated with Fluvastation and Simvastation of different concentrations for 24 hours, the proportion of early and later apoptotic cells both increased; moreover, the total apoptotic rate increased significantly(P<0.05) at 0.2 mmol/L and 0.3 mmol/L concentration of Fluvastatin and Simvastatin. The detection of cell cycle showed that both of Jurkat and CCRF-CEM cells were arrested in G phase. Western blot revealed that, in comparison with the control group, the expressions of BAX, p21 and p27 in cells treated with Statins were up-regulated, while Cyclin D1, BCL-2 and p-Akt expressions were down-regulated.</p><p><b>CONCLUSION</b>Statins can suppress T-ALL cell proliferation and induce cell apoptosis through the inhibition of Akt pathway.</p>
