ABSTRACT

As one of the key enzymes in cell metabolism, the activity of citrate synthase 3 (CS3) regulates the substance and energy metabolism of organisms. The protein members of CS3 family were identified from the whole genome of apple, and bioinformatics analysis was performed and expression patterns were analyzed to provide a theoretical basis for studying the potential function of CS3 gene in apple. BLASTp was used to identify members of the apple CS3 family based on the GDR database, and the basic information of CS3 protein sequence, subcellular localization, domain composition, phylogenetic relationship and chromosome localization were analyzed by Pfam, SMART, MEGA5.0, clustalx.exe, ExPASy Proteomics Server, MEGAX, SOPMA, MEME, WoLF PSORT and other software. The tissue expression and inducible expression characteristics of 6 CS3 genes in apple were determined by acid content and real-time fluorescence quantitative polymerase chain reaction (qRT-PCR). Apple CS3 gene family contains 6 members, and these CS3 proteins contain 473-608 amino acid residues, with isoelectric point distribution between 7.21 and 8.82. Subcellular localization results showed that CS3 protein was located in mitochondria and chloroplasts, respectively. Phylogenetic analysis divided them into 3 categories, and the number of genes in each subfamily was 2. Chromosome localization analysis showed that CS3 gene was distributed on different chromosomes of apple. The secondary structure of protein is mainly α-helix, followed by random curling, and the proportion of β-angle is the smallest. The 6 members were all expressed in different apple tissues. The overall expression trend from high to low was the highest relative expression content of MdCS3.4, followed by MdCS3.6, and the relative expression level of other members was in the order of MdCS3.3 > MdCS3.2 > MdCS3.1 > MdCS3.5. qRT-PCR results showed that MdCS3.1 and MdCS3.3 genes had the highest relative expression in the pulp of 'Chengji No. 1' with low acid content, and MdCS3.2 and MdCS3.3 genes in the pulp of 'Asda' with higher acid content had the highest relative expression. Therefore, in this study, the relative expression of CS3 gene in apple cultivars with different acid content in different apple varieties was detected, and its role in apple fruit acid synthesis was analyzed. The experimental results showed that the relative expression of CS3 gene in different apple varieties was different, which provided a reference for the subsequent study of the quality formation mechanism of apple.

ABSTRACT

Objective To improve the analysis of 134Cs, 137Cs, and 60Co in aerosol samples by the national key radiation environment laboratories. Methods Intercomparison of analysis results of 134Cs, 137Cs, and 60Co in standard aerosol samples was performed among the national key radiation environment laboratories according to Gamma spectrometry method of analyzing radionuclides in biological samples (GB/T 16145-1995 ), and the intercomparison results were evaluated by the standard deviation. Results Six laboratories were involved in the intercomparison. For 134Cs, 50% of the laboratories showed a relative deviation less than 10%, and 50% showed a relative deviation of 10%-20%. For 137Cs, 33.3% of the laboratories showed a relative deviation less than 10%, and 76.7% showed a relative deviation of 10%-20%. For 60Cs, all laboratories showed a relative deviation less than 10%. The overall intercomparison results were acceptable. Conclusion The laboratories in this intercomparison show generally good results.

ABSTRACT

Objective:To investigate the activity concentrations of 90Sr and 137Cs in water in Hangzhou urban area. Methods:From 2012 to 2020, Qiantang River water as an important drinking water source, tap water as direct drinking water for residents, and West Lake water in tourists crowded area were selected forwater quality monitoring with respect to conctnts of 90Sr and 137Cs. The activity concentrations of 90Sr and 137Cs in water samples, as collected in wet and dry seasons resepectively, were determined by radiochemical analysis, with the 137Cs to 90Sr activity ratios obtained. Results:From 2012 to 2020, the activity concentrations of 90Sr and 137Cs in tap water were (2.0±1.1) - (7.4±0.4) mBq/L and (0.45±0.06) - (7.1±0.6) mBq/L, respectively. The 137Cs to 90Sr activity ratios ranged from 0.07 to 2.40. The activity concentrations of 90Sr and 137Cs in Qiantang River were (3.7±1.1) - (17.0±4.4) mBq/L and (0.28±0.01) - (15.0±4.5) mBq/L, respectively. The 137Cs to 90Sr activity ratios ranged from 0.03 to 0.90. The activity concentrations of 90Sr and 137Cs in West Lake water were (2.2±0.5) - (11.0±2.0) mBq/L and (0.32±0.04) - (7.9±1.9) mBq/L, respectively. The 137Cs to 90Sr activity ratios ranged from 0.05 to 1.20. Conclusions:The activity concentrations of 90Sr and 137Cs in water in Hangzhou urban area were at the background levels, lower than the concentration limits, 10 Bq/L both for 90Sr and 137Cs recommended by WHO in the 4 th edition of Guidelines for Drinking Water Quality.

ABSTRACT

Purpose: To ascertain normative database of contrast sensitivity (CS) using Spaeth/Richman CS test (SPARCS) in the Indian population. Methods: This cross?sectional study enrolled 200 healthy individuals, and CS was tested in both eyes of each participant using SPARCS. A detailed ocular examination was done before enrollment to rule out pathologies that may affect CS. A practice test was performed in the right eye (OD), followed by uniocular testing in each eye and a final binocular test. Results: Data of 400 eyes of 200 subjects who fulfilled the inclusion criteria was evaluated. The average age of subjects was 46.57 ± 16.77 years (range 21–79 years), with a slight female preponderance (53%, n = 106). A statistically significant decline in average SPARCS scores was noted with increasing age (P < 0.05), ranging from 86.68 (20–29 years age group) to 67.44 (70–79 years age group). Higher scores were noted in binocular testing than uniocular testing (Interclass correlation coefficient [ICC] = 0.83; P < 0.001). Females achieved statistically significant higher total scores in uniocular SPARCS testing (both OD and OS), but there was no significant difference noted between the two genders in binocular testing. Correlation between practice and main tests was statistically Significant with an interclass correlation coefficient of 0.54 (P < 0.001). Conclusion: Normative database for SPARCS was established for Indian eyes, with a decreasing trend noted in peripheral as well as central CS scores with increasing age.

ABSTRACT

Background: Cervical cancer is unique among human cancers which is mostly attributable to infection. Conventional PAP smear method is most effective for prevention and detection of cervical cancer but the accuracy of this method is low. This PAP smear now evolved to Liquid Based Cytology (LBC). Method: All the patients visiting Gynaecology OPD who fulfilled the inclusion Criteria in the duration starting from 1 January 2020 to 30 June 2021 in the Department of Pathology of Gajra raja Medical College, Gwalior (MP) were included in this study. 50 atypical smears by Conventional PAP test were then collected and these cases were subjected to Eziprep Liquid Based Cytology and Conventional PAP smear in private laboratory setup after taking history and clinical examination. The smears were studied by using 7 morphological parameters. Smears were analysed for adequate cellularity, clean background, uniformResult: distribution of cells, cellular overlapping, inflammation, distinct cell border, nuclear irregularity and then categorise by Bethesda reporting system. The results were significant only for clean background, uniform distribution of cells, cellular overlapping and inflammation. Conclusion: Results of cervical cytology smears by both methods showed that LBC provides more representative sample with reduced obscuring material, improved clarity allowing better morphological evaluation.

ABSTRACT

ObjectiveTo observe the effect of modified Da Chaihutang on cholesterol gallstone (CS) in mice due to damp-heat based on the farnesoid X receptor (FXR)/fibroblast growth factor 15 (FGF15)/fibroblast growth factor receptor 4 (FGFR4) pathway and explore the molecular biological mechanisms of CS differentiated into damp-heat syndrome from the perspective of correspondence between prescription and syndrome. MethodForty-eight six-week-old mice were randomly divided into the blank group, model group, modified Da Chaihutang (23.4 g·kg-1) group, and ursodeoxycholic acid (0.12 g·kg-1) group, with 12 mice in each group. The ones in the latter three groups were exposed to "internal dampness + external dampness + high-cholesterol diet" for 12 weeks for inducing CS due to damp-heat. Mice in the modified Da Chaihutang group and ursodeoxycholic acid group were gavaged with the corresponding drugs, while those in the model and blank groups with the same amount of normal saline for a total of four weeks. Before and after modeling, mice in each group were subjected to open field tests for determining their activities and mental states. Such general conditions as body mass, food intake, fur, and urine and stool of mice in each group were observed and recorded weekly for judging the damp-heat syndrome. After the intervention, the sampled liver and gallbladder tissues of mice in each group were stained with hematoxylin-eosin (HE) staining, and the serum γ-glutamyltransferase (GGT), alkaline phosphatase (ALP), and total bilirubin (TBIL) were determined. The total cholesterol (TC) and total bile acid (TBA) contents in bile were measured by enzyme-linked immunosorbent assay (ELISA). The mRNA and protein expression levels of FXR, FGF15, FGFR4, and cholesterol 7α-hydroxylase gene (CYP7A1) were assayed by real-time fluorescence quantitative polynucleotide chain reaction (Real-time PCR) and Western blot. ResultCompared with the blank group, the model group exhibited enlarged gallbladder, brown turbid bile with flocculent precipitation visible to the naked eye, obvious damp-heat syndrome, lipoid degeneration in the liver tissue, rough and thickened gallbladder wall, elevated ALP, GGT, and TBIL in serum (P<0.01) and TC in bile (P<0.01), reduced TBA (P<0.01), up-regulated FXR, FGF15, and FGFR4 mRNA and protein expression in ileum (P<0.05, P<0.01), and down-regulated CYP7A1 mRNA and protein expression (P<0.01). Compared with the model group, the two medication groups displayed improved bile turbidity, and the bile in the modified Da Chaihutang group became clearer. After intervention, the damp-heat syndrome of mice in the modified Da Chaihutang group was significantly alleviated. The liver and gallbladder lesions of mice in the two medication groups were significantly relieved, manifested as reduced serum ALP, GGT, and TBIL (P<0.01). The reduction in ALP and TBIL of the modified Da Chaihutang group was more significant (P<0.01). The TC contents in the bile of mice from the two medication groups were significantly lowered, whereas the TBA contents were elevated (P<0.01), with more significant changes present in the modified Da Chaihutang group (P<0.01). The mRNA and protein expression levels of FXR, FGF15, and FGFR4 in the modified Da Chaihutang group were down-regulated (P<0.05, P<0.01), while the mRNA and protein expression levels of CYP7A1 rose (P<0.05), except that the elevation in FGF15 and FGFR4 protein expression and reduction in CYP7A1 protein expression were not significant. The mRNA and protein expression levels of FXR, FGF15, and FGFR4 in the ursodeoxycholic acid group all decreased, among which the reduction in FXR was remarkable (P<0.05), and the mRNA and protein expression levels of CYP7A1 were significantly up-regulated (P<0.05). ConclusionModified Da Chaihutang significantly improves the stone, liver function, bile composition, abnormal cholesterol-bile acid metabolism, and damp-heat syndrome in the model mice of CS differentiated into damp-heat syndrome, which may be related to its regulation of key factors FXR, FGF15, FGFR4, and CYP7A1 mRNA and protein expression in the cholesterol-bile acid metabolism pathway.

ABSTRACT

Background: Tobacco smoking is a source of many toxins such as free radicals, mutagenic substances as well as cause for developing cardiovascular diseases (CVD), particularly atherosclerosis. This study aims to assess the impact of smoking on antioxidants in Sudanese male smokers. Methods: Cases were 85 and 48 men who smoke cigarettes (CS) and water pipe (WPS) respectively and they were compared with matching 50 non-smoking controls. Blood samples were collected and following parameters: Glutathione peroxidase, Superoxide dismutase, Total cholesterol, Triglyceride, LDL, HDL, Paraoxinase, and Malondialdehyde were measured. Results: There were no significant differences in biochemical parameters between light CS and WPS compared to controls. In heavy smokers of both WPS and CS, the TC, TG, LDL, and MDA were higher than controls (p>0.05), GPx, SOD, HDL, and PON were lower in smokers than controls (p>0.05). In both groups of smokers, HDL, GPx, SOD, and PON were inversely correlated with duration of smoking (p>0.05), also, HDL was positively correlated with SOD and GPx (p>0.05). Moreover, GPx and SOD were correlated with each other in both groups of smokers (p>0.05). Conclusion: In Sudanese male smokers' biochemical profile disturbances suggest that heavy smoking was leading to developing CVD, particularly WPS

ABSTRACT The burial of bodies is a potentially polluting activity. Taking this into consideration, the aim of the present study was to verify the compliance of two cemeteries with environmental legislation and to quantify the concentrations of heavy metals in soils affected by burial activities. Physicochemical characterization of the soil was performed by analyzing control samples from areas near the cemeteries. Concentrations of cadmium, lead, chromium, nickel, zinc and copper were determined using high-resolution continuum source atomic absorption spectrometry. The two cemeteries had unsatisfactory properties for the retention of metal cations, with clay percentages ranging from 15.40 to 41.40% and sand percentages ranging from 28.75 to 66.85%. The control samples presented low cation exchange capacity (12.27 to 22.73 cmolc/dm³) and high aluminum (Al3+) saturation (66.74 to 90.16%). Although neither of the two cemeteries had concentrations above the limits established for the metals analyzed by Resolution No. 420/2009 of the National Environment Council, the contaminants may be leaching to groundwater due to inadequate soil characteristics.


RESUMO O sepultamento de corpos é uma atividade potencialmente poluidora. Este trabalho teve como objetivo verificar a adequação das áreas de dois cemitérios públicos à legislação ambiental e à atividade cemiterial e quantificar a concentração de metais pesados nos solos que estão sob influência desses empreendimentos. Realizou-se a caracterização físico-química do solo, com a análise de amostras testemunha de solo de cada cemitério. Também foram determinadas as concentrações dos metais pesados: cádmio, chumbo, cromo, níquel, zinco e cobre, por meio de espectrometria de absorção atômica de alta resolução com fonte contínua. As áreas dos cemitérios apresentam condições insatisfatórias para a retenção de íons catiônicos metálicos, com percentuais de argila variando entre 15,40 e 41,40% e de areia entre 28,75 e 66,85%. Os solos testemunha apresentaram reduzida capacidade de troca de cátions entre 12,27 e 22,73 cmolc/dm³) e elevada saturação por alumínio entre 66,74 e 90,16%. Apesar de nenhum dos cemitérios apresentar concentrações dos metais analisados acima dos limites de prevenção estabelecidos pela Resolução nº 420/2009 do Conselho Nacional do Meio Ambiente, em função das características dos solos, os contaminantes podem estar sendo lixiviados para os recursos hídricos subjacentes.

ABSTRACT

Pulmonary administration route has been extensively exploited for the treatment of local lung diseases such as asthma, chronic obstructive pulmonary diseases and respiratory infections, and systemic diseases such as diabetes. Most inhaled medicines could be cleared rapidly from the lungs and their therapeutic effects are transit. The inhaled medicines with extended pulmonary exposure may not only improve the patient compliance by reducing the frequency of drug administration, but also enhance the clinical benefits to the patients with improved therapeutic outcomes. This article systematically reviews the physical and chemical strategies to extend the pulmonary exposure of the inhaled medicines. It starts with an introduction of various physiological and pathophysiological barriers for designing inhaled medicines with extended lung exposure, which is followed by recent advances in various strategies to overcome these barriers. Finally, the applications of the inhaled medicines with extended lung exposure for the treatment of various diseases and the safety concerns associated to various strategies to extend the pulmonary exposure of the inhaled medicines are summarized.

ABSTRACT

Abstract A wide variety of cellular mechanisms such as cell division and metabolic processes are maintained by protein-protein interactions (PPIs). The identification of PPI through laboratory techniques is costly, time-consuming, difficult, and challenging. However, computational techniques were generated for PPIs prediction. In alfalfa (Madicago sativa), PPI was predicted among 12 MsMAPKs and 4 MsPP2Cs using a docking approach. For homology modelling, the Swiss model was employed while PROCHECK, ERRAT, and Verify3D were used to validate 3D models. The Ramachandran plots were obtained from PROCHECK which showed value more than 90% (nPP2C1, PP2C1, PP2C, and MSK-3 revealed 92.9%, 94.2%, 92.4%, and 91.1% respectively) for high-quality structures. The HawkDock server and the BIPSPI server were used to analyse protein docking and predict interaction sites, respectively. Our findings demonstrated that MsPP2C docking sites play an important role in the identification and docking of MsMAPKs. The binding free energy ranged from -0.16Kcal/mol to -49.15Kcal/mol for all MsMAPKs and MsPP2Cs, indicating that they interact. Docking site analysis showed that there were 48 pairs of PPIs which indicated that MsPP2Cs can perform a vital role in other signaling pathways. This study found that all MsPP2Cs have docking sites for MsMAPKs, indicating that this method can accurately determine protein-protein interactions.

ABSTRACT

Background: Emergency caesarean section (CS) is divided into four categories based on the degree of urgency by RCOG and NICE guidelines. It is recommended that the decision to delivery interval (DDI) in emergency CS should be within 30 minutes in category 1 and within 75 minutes in category 2. Our Primary objective was to study the incidence and indications of emergency CS and audit the DDI in emergency CS at tertiary care hospital. Our secondary objective was to study the effect of DDI on neonatal outcome.Methods: Descriptive study was carried out among 409 women who underwent emergency CS from August 2018 to December 2018 at St. John's medical college hospital, Bangalore. Relevant data was collected by chart review. Emergency CS were categorised according to RCOG guidelines based on the degree of urgency and further classified based on DDI as <30 minutes, 30-75 minutes and >75 minutes.Results: We had 409 cases of emergency CS. Category 1 had 113 (27.63%) cases, category 2 had 126 (30.81%) cases and category 3 had 170 (41.56%) cases. DDI of <30 minutes was achieved in 19.5% in category 1, DDI of <75 minutes was achieved in 93.65% in category 2. Fetal distress was the leading cause of emergency CS in category 1 and 2. There was a high incidence of low APGAR in babies delivered in <30 minutes and lower APGAR was significantly associated with <30 minutes of DDI (p<0.0001).Conclusions: Fetal distress was the leading cause of emergency CS. DDI interval of <30 minutes was not always associated with good neonatal outcome; Category of CS has a significant effect on neonatal outcome.

Abstract Objective To evaluate the obstetric outcomes of singleton high-risk pregnancies with a small size uterine fibroid. Methods This retrospective cohort study was conducted among 172 high-risk pregnant women who were followed-up by a single surgeon between 2016 and 2019. Pregnant women with preconceptionally diagnosed small size (< 5 cm) single uterine fibroids (n = 25) were compared with pregnant women without uterine fibroids (n = 147) in terms of obstetric outcomes. Results There was no statistically significant difference between the groups in terms of adverse pregnancy outcomes. The size of the fibroids was increased in 60% of the cases, and the growth percentage of the fibroids was 25% during pregnancy. Intrapartum and short-term complication was not observed in women who underwent cesarean myomectomy. Conclusion Small size uterine fibroids seem to have no adverse effect on pregnancy outcomes even in high-risk pregnancies, and cesarean myomectomy may be safelyperformed in properly selected cases.

ANTECEDENTES Y OBJETIVO Desde el 18 de octubre de 2019, Chile se encuentra en una crisis social relacionada a demandas de la ciudadanía para mejorar la calidad de vida. Frente a esto es que Chile lleva más de 5 meses de manifestaciones y concentraciones masivas y en las cuales Carabineros de Chile ha utilizado gases lacrimógenos como medida de control de masas. En este contexto el Instituto Nacional de Derechos Humanos (INDH) ha solicitado al Ministerio de Salud, el desarrollo de una síntesis de evidencia con el objetivo de informar sobre los riesgos para la salud como consecuencia del uso de gases lacrimógenos, así informando a la toma de decisiones. METODOLOGÍA Se utilizaron estrategias de búsqueda en las bases de datos Health Systems Evidence, Epistemonikos, EMBASE, y PUBMED, con el objetivo de identificar revisiones sistemáticas que abordaran la pregunta formulada. Como las revisiones sistemáticas no siempre reportaron adecuadamente los resultados presentados, se extrajeron los datos de los estudios primarios contemplados en estas revisiones. Se incluyeron revisiones sistemáticas que evaluaran el efecto del gas CS o combinados de gas CS con otro tipo de sustancias sobre la salud de la población. Se excluyeron estudios que evaluaran los efectos de gases lacrimógenos diferentes a CS, como gas pimienta o cloroacetopheno. RESULTADOS -Los efectos en salud inmediatos reportados son la irritación de mucosas, a mediano y largo plazo son asma, disnea, estrés postraumático, depresión, alteración funcional, estado vegetativo y muerte como consecuencia de impactos de dispositivos lacrimógenos.

ABSTRACT

Purpose: To calculate AIP and to find correlation between hole closure pattern with AIP in idiopathic full thickness macular hole (FTMH) cases. Methods: In this prospective, non-randomized, interventional single blind study, 105 eyes of symptomatic FTMH (<6 month duration) were operated. Minimal diameter of macular hole (MDMH) was calculated on OCT, divided into Group I (>400?, n = 75) and Group II (<400?, n = 30). 23G vitrectomy with ILM peeling and gas injection were done in all and recorded. Final area of ILM peeled (AIP) was calculated using Adobe Photoshop CS2 (PSD format) in disc diameters (DD) from still frame. Follow up was done at 6 monthly interval up to a maximum of 5 years after surgery. Results: Macular holes were closed in 92.38% eyes. In Group I, mean pre-operative BCVA was 1.14 � 0.39 log MAR and was improved to 0.79 � 0.26 log MAR post-operatively at 6 months. In Group II, mean pre-operative BCVA was 0.95 � 0.44 log MAR and was improved to 0.60 � 0.24 log MAR after surgery. When AIP was more than 3DD, Type I and Type II closure were 72.77% and 27.27% in Group I (P value <0.01) and 84.21% and 15.79% in Group II (P value <0.01). Conclusion: AIP can be calculated using Adobe Photoshop CS2. Type I closure was significantly high with AIP >3DD in both groups. Intra-operatively using video overlay, surgeons can increase the diameter of AIP to get better closure pattern.

ABSTRACT

Objective: Present study was conducted to evaluateeffectiveness of lumbar epidural and spinal anaesthesia inelective caesarean section.Methods: This experimental study is conducted at tertiarymedical college hospital, Gopalgong. Written informed consentfrom 100 patients were obtained for this experimental study.Purposive sampling technique was used.Results: During the study, where in group-A 52.02% womenwere in their first pregnancy followed by 28.28% were 2ndpregnancy, 19.7% were multi gravid women. where in group-A,35% had hypotension where as in group-B it was 11%. groupA, 30% patients were Highly satisfied where as in group-B itwas 65%. The patients selected for the study was divided intotwo groups: Group – I (Spinal): 50 Patients selected, Group – II(Epidural): 50 patients selected.Conclusion: From our result and study we can conclude that,continuous epidural technique which allowed mother earlybreastfeeding and ambulation. Further studies are needed forbetter outcome.

@#Introduction: Hyperhidrosis is a disorder of excessive and uncontrollable sweating beyond the body’s physiological needs. It can be categorised into primary or secondary hyperhidrosis based on its aetiology. Detailed history review including onset of symptoms, laterality of disease and family history are crucial which may suggest primary hyperhidrosis. Secondary causes such as neurological diseases, endocrine disorders, haematological malignancies, neuroendocrine tumours and drugs should be adequately examined and investigated prior to deciding on further management. The diagnosis of primary hyperhidrosis should only be made only after excluding secondary causes. Hyperhidrosis is a troublesome disorder that often results in social, professional, and psychological distress in sufferers. It remains, however, a treatment dilemma among some healthcare providers in this region. Methods: The medical records and clinical outcomes of 35 patients who underwent endoscopic thoracic sympathectomy for primary hyperhidrosis from 2008 to 2018 in Department of Cardiothoracic Surgery were reviewed. Results: The mean age of the patients was 27±10.1years, with male and female distribution of 18 and 17, respectively. Fifty-one percent of patients complained of palmar hyperhidrosis, while 35% of them had concurrent palmaraxillary and 14% had palmar-plantar-axillary hyperhidrosis. Our data showed that 77% (n=27) of patients were not investigated for secondary causes of hyperhidrosis, and they were not counselled on the non-surgical therapies. All patients underwent single-staged bilateral endoscopic thoracic sympathectomy. There was resolution of symptoms in all 35 (100%) patients with palmar hyperhidrosis, 13(76%) patients with axillary hyperhidrosis and only 2 (50%) patients with plantar hyperhidrosis. Postoperatively 34.3% (n=12) of patients reported compensatory hyperhidrosis. There were no other complications such as pneumothorax, chylothorax, haemothorax and Horner’s Syndrome. Conclusion: Clinical evaluation of hyperhidrosis in local context has not been well described, which may inadvertently result in the delay of appropriate management, causing significant social and emotional embarrassment and impair the quality of life of the subjects. Detailed clinical assessment and appropriate timely treatment, be it surgical or non-surgical therapies, are crucial in managing this uncommon yet distressing disease.

Coccidiosis-causing Eimeria species are transmitted in poultry via the oral-fecal route and can lead to hemorrhagic diarrhea and mortality. This results in enormous economic losses in the poultry industry. Furthermore, its resistance to some currently used antibiotics is increasing. This has prompted the development of new alternative drug therapies that address the issue of chemical-free meat production. Effective management of infectious diseases in veterinary practice includes the induction of protective and adaptive immunity by treatment with an alternative agent. In this study, we evaluated the anticoccidial effects of dietary supplementation of Chosun University (CS) 32 compounds (0.1% and 1.0%) against Eimeria tenella, which was isolated and purified from the supernatant of culture broth of Bacillus strain (KCTC18250P), as well as its effect on the growth rate and feed efficiency in chickens. Overall, we observed a decrease in lesion scores and oocyte output in CS 32 compounds-treated chickens. We concluded that 0.1% CS 32 compounds displayed anticoccidial effects against E. tenella infection.

ABSTRACT

We constructed the CS1-targeted second- and third-generation CAR-T cells with genetic engineered 4-1BB or/and ICOS as a costimulatory signaling molecule by use of lentiviral platform. The CS1-targeted second-generation CAR-T cells with ICOS or 4-1BB had similar anti-neoplastic activity. When effector/target ratio was 1:1, the CAR-T cells with ICOS showed better killing effect on IM9-lucgfp cells than those with 4-1BB. However, The CS1-targeted third-generation CAR-T cells exihibited lower cytolytic capacity against IM9-lucgfp cells than the CS1-targeted second-generation CAR-T cells when the ratio of effector/target was 1:1, 2:1 or 5:1. When the ratio of effector/target was 10:1, the killing efficacy of both the second- and third-generation CAR-T cells against IM9-lucgfp cells was more than 85%, significantly higher than that of the control T cells. Taken together, both the CS1-targeted second- and third-generation CAR-T cells with ICOS or/and 4-1BB could efficiently kill CS1-positive multiple myeloma cells, but the CS1-targeted second-generation CAR-T cells had more potent killing effect on CS1-positive multiple myeloma cells than the CS1-targeted third-generation CAR-T cells.

ABSTRACT

Signaling lymphocyte activation family 7 (SLAMF7/CS1) is a cell surface glycoprotein that is highly expressed in multiple myeloma cells. CS1 is a sensitive and specific biomarker for multiple myeloma. CAR-T cell immunotherapy is a new method for the treatment of multiple myeloma. CS1 CAR-T cell immunotherapy has good effect on relapsed refractory multiple myeloma. To detect the expression efficiency of CS1 CAR on CS1 CAR-T cells and to find an auxiliary means to CAR-T cell immunotherapy, we prepared a CS1-Fc fusion protein. First, the extracellular domain of CS1 was amplified from the existing plasmid by PCR and ligated with human IgG1-Fc fragment by overlap extension PCR. The recombinant fragment was ligated into pMH3 eukaryotic expression vector. After restriction enzyme digestion and DNA sequencing, the pMH3-CS1-Fc-his recombinant plasmid was successfully constructed. The recombinant plasmid was transfected into Chinese hamster ovary cell (CHO-S) by liposome. The expression of the CS1-Fc fusion protein in CHO-S cells was identified by flow cytometry after G418 pressure screening. Next, the CS1-Fc fusion protein was purified by nickel column. Western-blot analysis showed that molecular weight of the fusion protein was about 70 kDa was identified by Western blotting. The CS1-Fc fusion protein couldeffectively detect the expression rate of CS1 CAR and promote the activation, proliferation andcytokines secretion of the CS1 CAR-T cells. The results will lay the experimental foundation for the in vitro detection and potentiation of CAR-T cells in multiple myeloma treated with CS1 CAR-T cell.
