ABSTRACT

Mannitol-calcium chloride metal organic framework (MOF) cocrystal significantly improved the tabletability of β-mannitol and could be developed as a new tablet filler. However, mannitol monomer was found in the product during the scale-up production of the excipient, which significantly affected the functional properties of the excipient. In this study, we intend to quantify the multi-component eutectic system of mannitol-calcium chloride. In this experiment, the MOF cocrystal excipient mannitol-calcium chloride cocrystal was used as the model compound, and infrared spectrum was collected. Based on partial least squares regression (PLSR) method, the abnormal bands were removed and the spectrum was preprocessed by normalization. The quantitative correction model of mannitol-calcium chloride MOF cocrystal content in cocrystal excipients was established and compared by two different variable screening methods, genetic algorithm (GA) and competitive adaptive reweighting algorithm (CARS). Two different variable screening methods, GA method and CARS method, were used to screen out 160 and 14 variables, respectively. The mannitol-calcium chloride cocrystal model established by CARS-PLSR method had the best performance, and the average relative error (MRE) and corrected root mean square error (RMSEC) of the model were 0.008 8 and 0.892 5, respectively, the determination coefficient (R2) of the model was increased from 0.978 3 to 0.994 4. The quantitative method of eutectic system established in this study has high prediction accuracy, fast detection speed and good stability, which is of great significance for optimizing the preparation process conditions and quality control methods of such eutectic excipients.

ABSTRACT

Objetivo: Avaliar a Resistência de União (RU), Grau de Conversão (GC), Liberação de Cálcio (LC), Resistência Flexural (RF), Módulo de Elasticidade (ME) e Viscosidade (VS) de cimentos experimentais à base de cálcio autocondicionantes e autoadesivos. Três cimentos experimentais: A (CONTROLE) ­ sem fonte de cálcio, B (HIDROCAL) ­ com hidróxido de cálcio, C (CLORECAL) ­ com cloreto de cálcio foram sintetizados e avaliados juntamente a um cimento comercial D (ULTRAPLUS) (Ultra-Blend Plus®). Metodologia: Para a RU, foi utilizado o teste de cisalhamento, cujas amostras (1,3mm de diâmetro com 2mm de altura) foram aderidas à dentina bovina (n=10) e tensionadas em máquina de ensaios (1 mm/min) até́ ocorrer o deslocamento. Para o GC (n=3), foi utilizada a Espectroscopia Infravermelha com transformada de Fourier (FTIR). Na determinação da LC (n=3) foram utilizadas amostras circulares (1,0 mm x 0,4 mm) e a leitura foi realizada através de um espectrofotômetro de absorção atômica nos tempos de 3h, 24h e 72h. Para RF/ME (n=10), amostras em formato de barra (7mm x 2mm x 1mm) foram confeccionadas e avaliadas por meio de uma Máquina de Ensaios Universal no teste de flexão de três pontos. Para a VS (n=30), a variação da taxa de deformação foi determinada por um reômetro rotacional de cilindros concêntricos. Os dados foram analisados por ANOVA 1 e 2 fatores seguido pelo pós teste de tukey (p<0,05). Resultados: Para RU todos os materiais experimentais apresentaram valores estatisticamente maiores que o comercial (p<0,01). Para o GC os cimentos CONTROLE e HIDROCAL apresentaram valores estatisticamente maiores que os demais (p<0,01). Frente à LC o CLORECAL e ULTRAPLUS apresentaram valores estatisticamente superiores após 72h (p<0,01). O ULTRAPLUS apresentou estatisticamente a maior RF e o menor ME (p<0,05). Os cimentos HIDROCAL e CLORECAL apresentaram valores de VS estatisticamente superiores aos demais (p<0,01). Conclusão: De maneira geral, o cimento experimental à base de cloreto de cálcio mostrou-se mais promissor (AU).


Objective: To evaluate the Bond Strength (BS), Degree of Conversion (DC), Calcium Release (CR), Flexural Strength (FS), Elastic Modulus (EM) and Viscosity (VS) of experimentally calcium based cements self-etching and self-adhesive. Three experimental cements: A (CONTROL), B (HYDROCAL), C (CLORECAL) were synthesized and evaluated with a commercial cement D (ULTRAPLUS) (Ultra-Blend Plus®- South Jordan, Utah, USA). Methodology: For the BS, the shear test was used, whose samples 1.3mm in diameter and 2mm in height) were adhered to the bovine dentin (n=10) and tensioned in a testing machine (1mm/min) until occurring the displacement. For the DC (n=3), Infrared Spectroscopy with Fourier transform (FTIR) was used. In determining the CR (n = 3) circular samples (1.0mmx0.4 mm) were used and the reading was performed through an atomic absorption spectrophotometer at 3h, 24h and 72h. For FS/ ME (n= 0), bar-shaped samples (7mmx 2mmx1mm) were made and evaluated using a Universal Testing Machine in the three-point flexion test. For VS (n = 30), the variation in the strain rate was determined by a rotational concentric cylinder rheometer. Results: For BS, all experimental materials showed statistically higher values than the commercial material (p <0.01). For the DC, the CONTROL and HIDROCAL cements showed statistically higher values than the others (p <0.01). In relation to CR, CLORECAL and ULTRAPLUS showed statistically higher values after 72 hours (p <0.01). ULTRAPLUS showed statistically the highest FS and the lowest EM (p <0.05). The HIDROCAL and CLORECAL cements showed VS values statistically higher than the others (p <0.01) Conclusion: In general, the experimental cement based on calcium chloride proved to be more promising (AU).

ABSTRACT

Abstract Calcium aluminate cement (CAC) has been highlighted as a promising alternative for endodontic use aiming at periapical tissue repair. However, its effects on dental pulp cells have been poorly explored. Objective: This study assessed the impact of calcium chloride (CaCl2) and bismuth oxide (Bi2O3) or zinc oxide (ZnO) additives on odontoblast cell response to CAC. Methodology: MDPC-23 cells were exposed for up to 14 d: 1) CAC with 2.8% CaCl2 and 25% ZnO (CACz); 2) CAC with 2.8% CaCl2 and 25% Bi2O3 (CACb); 3) CAC with 10% CaCl2 and 25% Bi2O3 (CACb+); or 4) mineral trioxide aggregate (MTA), placed on inserts. Non-exposed cultures served as control. Cell morphology, cell viability, gene expression of alkaline phosphatase (ALP), bone sialoprotein (BSP), and dentin matrix protein 1 (DMP-1), ALP activity, and extracellular matrix mineralization were evaluated. Data were compared using ANOVA (α=5%). Results: Lower cell density was detected only for MTA and CACb+ compared with Control, with areas showing reduced cell spreading. Cell viability was similar among groups at days one and three (p>0.05). CACb+ and MTA showed the lowest cell viability values at day seven (p>0.05). CACb and CACb+ promoted higher ALP and BSP expression compared with CACz (p<0.05); despite that, all cements supported ALP activity. Matrix mineralization were enhanced in CACb+ and MTA. Conclusion: In conclusion, CAC with Bi2O3, but not with ZnO, supported the expression of odontoblastic phenotype, but only the composition with 10% CaCl2 promoted mineralized matrix formation, rendering it suitable for dentin-pulp complex repair.

ABSTRACT

ABSTRACT: Six adult Holstein cows were used in this fluid therapy study. All animals were previously submitted to 24 hours of water and food abstaining period and submitted to both hydration treatments for eight hours in a crossover design 6x2. Two treatments with hypotonic solutions (190 mOsm/L) with low strong ion difference and different chloride concentrations solutions were executed. Physical, biochemical, blood gas analysis and urinary evaluation were executed in five different experimental times (T-24h, T0h, T4h, T8h e T24h). The hydration period caused plasmatic volume expansion. An increase in faeces humidity, excretion of Na+ and Cl- in urine, reduction of PCV, osmolarity, blood and urinary pH was observed. Both solutions can be used for fluid therapy for adult cattle and solution containing calcium chloride (SECaCl) was the most suitable for use in animals with hypochloraemia.


RESUMO: Seis vacas holandesas adultas foram utilizadas neste estudo de hidratação. Todos os animais foram submetidos previamente a 24 horas de jejum hídrico e alimentar e submetidos a ambos os tratamentos de hidratação por oito horas, em delineamento transversal 6x2. Foram realizados dois tratamentos com soluções hipotônicas balanceadas (190 mOsm/L) com diferentes concentrações de cloretos. Foram realizadas avaliações físicas, bioquímicas, hemogasometria e avaliação urinária em cinco diferentes tempos experimentais (T-24h, T0h, T4h, T8h e T24h). O período de hidratação causou expansão de volume plamático. Foi observado aumento da umidade das fezes e da excreção de Na+ e Cl- na urina, redução do volume globular, osmolaridade, pH sangüíneo e pH urinário. Ambas as soluções podem ser usadas para hidratação de bovinos adultos, sendo a solução que contém cloreto de cálcio (SECaCl) a mais indicada para o uso em animais com hipocloremia.

ABSTRACT

Orchiectomized bulls have advantages in the meat quality and ease of handling. Chemical castration is an option for surgical castration and the sclerosing agents can be administered into the testicular or epididymis parenchyma. These agents have a lower incidence of complications than surgery, especially when associated with dimethylsulfoxide (DMSO), which has anti-inflammatory action and increases the absorption of other drugs. Thus, this study aimed to evaluate the effect of a single intratesticular injection of calcium chloride solution associated with DMSO for the chemical sterilization of bulls. Twenty-four young adult bulls were utilized, distributed into 3 groups (G20, G30 and G40, n = 8/group), according to the calcium chloride concentration (20, 30 and 40%), in 10mL volume. Serum concentrations of testosterone, body weight, testicular volume and ecotexture, clinical signs and behavior and were evaluated for 45 days. Thus, the animals were orchiectomized and testicles were assessed histologically. There were no changes in body weight, decreased serum testosterone concentrations (except G30), signs of scrotal sensitivity or changes in behavior over the period. However, there was significant increase in testicular volume, especially on the 2nd and 3rd day after treatment, with values returning to the value initials at 15 days. Testicular adherence and firm consistency were observed during orchiectomy. Ultrasound examination revealed a loss of integrity of the median raphe, with cavity formation and an alteration of the testicular echotexture. In the histological evaluation, coagulation necrosis of seminiferous tubules and interstitial cells was observed, mainly in the medial portion in all groups. Some animals presented total absence of tubular formations in all the studied groups, being the effects of greater intensity in the G40. Additionally, pronounced edema was noted in all groups, especially in G40. Inflammatory infiltrate, fibroplasia and neovascularization were found to be predominantly discrete. Based on the conditions used in this study, we conclude that calcium chloride associated with DMSO can be used as a method of chemical sterilization in bovines.(AU)


Bovinos orquiectomizados apresentam vantagens na qualidade da carne e facilidade no manejo. A quimioesterilização é uma opção à castração cirúrgica e os agentes esclerosantes podem ser administrados no parênquima testicular ou epidídimo. Estes produtos possuem menor incidência de complicações, comparados a cirurgia, especialmente quando associados ao dimetilsulfóxido (DMSO), que apresenta ação anti-inflamatória e aumenta a absorção de outros fármacos. Assim, este estudo teve como objetivo avaliar o efeito de uma única injeção intratesticular de solução de cloreto de cálcio associado com 0,5% de DMSO para a esterilização química de bovinos. Vinte e quatro touros adultos jovens foram utilizados, distribuídos em 3 grupos (G20, G30 e G40, n = 8/grupo) de acordo com a concentração de cloreto de cálcio (20, 30 e 40%), em um volume de 10mL. Foram avaliadas as concentrações séricas de testosterona, peso corporal, volume e ecotextura testicular, sinais clínicos e comportamento por 45 dias. A seguir, os animais foram submetidos à orquiectomia e os testículos avaliados histologicamente. Não foram observadas alterações do peso corporal, diminuição das concentrações de testosterona sérica (exceto no G30), sinais de sensibilidade escrotal ou alterações no comportamento no período avaliado. Porém, houve aumento significativo do volume testicular, especialmente nos 2º e 3º dia após o tratamento, com valores retornando aos iniciais aos 15 dias. Aderência e consistência firme dos testículos foram achados observados durante a orquiectomia. O exame ultrassonográfico revelou perda de integridade da rafe mediana, com formação de cavidades e alteração da ecotextura testicular. Na avaliação histológica, verificou-se necrose de coagulação de túbulos seminíferos e células intersticiais acentuada, principalmente, na porção medial em todos os grupos, sendo que em alguns animais havia ausência total das formações tubulares em todos os grupos estudados, sendo os efeitos de maior intensidade no G40. Além disso, edema foi acentuado em todos os grupos, principalmente em G40. Infiltrado inflamatório, fibroplasia e neovascularização foram achados predominantemente discretos. Com base nas condições utilizadas neste estudo, conclui-se que o cloreto de cálcio associado com o DMSO pode ser utilizado como um método de esterilização química em bovinos.(AU)

OBJECTIVES: This study aimed to evaluate the chemical and biological properties of fast-set white mineral trioxide aggregate (FS WMTA), which was WMTA combined with calcium chloride dihydrate (CaCl₂·2H₂O), compared to that of WMTA. MATERIALS AND METHODS: Surface morphology, elemental, and phase analysis were examined using scanning electron microscope (SEM), energy dispersive X-ray microanalysis (EDX), and X-ray diffraction (XRD), respectively. The cytotoxicity and cell attachment properties were evaluated on human periodontal ligament fibroblasts (HPLFs) using methyl-thiazol-diphenyltetrazolium (MTT) assay and under SEM after 24 and 72 hours, respectively. RESULTS: Results showed that the addition of CaCl₂·2H₂O to WMTA affected the surface morphology and chemical composition. Although FS WMTA exhibited a non-cytotoxic profile, the cell viability values of this combination were lesser than WMTA, and the difference was significant in 7 out of 10 concentrations at the 2 time intervals (p < 0.05). HPLFs adhered over the surface of WMTA and at the interface, after 24 hours of incubation. After 72 hours, there were increased numbers of HPLFs with prominent cytoplasmic processes. Similar findings were observed with FS WMTA, but the cells were not as confluent as with WMTA. CONCLUSIONS: The addition of CaCl₂·2H₂O to WMTA affected its chemical properties. The favorable biological profile of FS WMTA towards HPLFs may have a potential impact on its clinical application for repair of perforation defects.

ABSTRACT

OBJECTIVE:To establish a method for the rapid contents determination of potassium chloride and calcium chloride in Compound sodium chloride injection. METHODS:After diluted in appropriate way,Compound sodium chloride injection was sampled directly and contents of potassium chloride and calcium chloride were determined simultaneously by ICP-OES. The powder was 1 300 W,plasma gas flow rate was 15 L/min,auxiliary cooling gas flow was 0.2 L/min,atomizer flow rate was 0.8 L/min, the peristaltic pump rate was 0.8 L/min,atomizer pressure was 315 kPa,and the observation was axial observation,analysis spec-tral lines of potassium and calcium were 766.490 nm and 315.887 nm. RESULTS:The linear ranges of potassium and calcium were 1.0-12.0 mg/L (r=0.999 7 and 0.999 9);RSDs of precision and reproducibility tests were lower than 1%;recoveries were 98.5%-100.5%(RSD=0.59%,n=9)and 99.3%-102.3%(RSD=0.98%,n=9). CONCLUSIONS:The method is simple,rapid and simple,and can be used for the quality control of Compound sodium chloride injection.

ABSTRACT

Dehumidifying agents are commonly used to remove moisture and odors and are not meant for oral ingestion. The hazards of accidental oral ingestion of dehumidifying agents are not well known. A 31-year-old man with autism drank the liquid desiccant contained in the barrel of a dehumidifier while sleepwalking. The desiccant mainly consisted of calcium chloride. He was taken to the emergency center with persistent vomiting and underwent gastric lavage. The patient was treated for hypercalcemia due to calcium chloride poisoning. He eventually died of multiple organ failure. Autopsy findings were as follows: acute suppurative pneumonia, intra-abdominal inflammation, liver necrosis, acute pancreatitis, and gastrointestinal hemorrhage. Calcium chloride has relatively low toxicity; however, when ingested orally, it can cause severe internal damage, and even death. We suggest active preventive measures to ensure the safe use of dehumidifying agents.

ABSTRACT

This study was aimed to evaluate the in vitro complexation nature and strength of complex which may be formed due to interaction between Amoxicillin and Calcium chloride (CaCl2). The interaction of Amoxicillin and Calcium chloride (fused) has been studied in aqueous systems at a fixed temperature (37 ± 0.5) °C and under different pH (pH 2.4 and pH 7.4) by using some physical methods as spectral observation, Job’s method of continuous variation, Ardon’s method. From spectrophotometric study, Amoxicillin gives a sharp peak at 272 nm when Calcium chloride mixed with Amoxicillin in 1:1 ratio the intensity of the peak of Amoxicillin change remarkably due to interaction. The jobs plot was obtained by plotting absorbance difference against the mole fraction of the each drug at pH 2.4 and pH 7.4. Amoxicillin forms strong 1:1 complex with Calcium chloride and reverse V Shaped curves indicate the formation of 1:1 complexes of Amoxicillin with Calcium chloride. These may indicate strong kinetics of complexation between Amoxicillin with Calcium chloride. The value of stability constant for the complexation of Amoxicillin with calcium chloride at pH 2.4 and pH 7.4 were obtained from the spectral data using Ardon’s plot. The value of stability constant for the drug-metal system at pH 2.4 and pH 7.4 are 5.54 and 6.67 respectively. At pH 2.4 it is found that Amoxicillin form relatively stable complex with Calcium chloride (stability constant 6.67) is high in comparison to pH 7.4. It can therefore be concluded that a careful consideration is needed during concurrent administration of Amoxicillin with Calcium chloride.

Objective:To improve the determination method for compound sodium acetate injection. Methods:Atomic absorption spectrophotometry ( AAS) was used to determine sodium chloride, calcium chloride and potassium chloride in compound sodium acetate injection. Results:The linear range of calcium ion, potassium ion and sodium ion was 9.124 ×10 -7-1.369 ×10 -5 g·ml-1(r =0.999 5),1.501 ×10 -7 ~4.504 ×10 -6 g·ml-1(r=0.999 2) and 7.500 ×10 -8-2.251 ×10 -6 g·ml-1(r=0.999 5), and the av-erage recovery was 100. 4%(RSD=1. 4%,n=9),102. 4%(RSD=1. 6%,n=9) and 100. 3% (RSD=1. 1%,n=9), respectively. Conclusion:The method is simple, accurate and reproducible, and can be used to control the quality of compound sodium acetate in-jection.

ABSTRACT

An efficient regeneration protocol was developed from shoot tip and nodal explants of Simarouba glauca DC, a promising biodiesel plant. Nodal explants appeared to have better regeneration capacity than shoot tip explants (40%) in the tested media. The highest regeneration frequency (90%) and shoot number (7.00 ± 1.00 shoots per explants) were obtained in nodal explants in Murashige and Skoog’s (MS) medium supplemented with 6-benzylaminopurine (BAP) 4.43 μM and α-naphthalene acetic acid (NAA) 5.36 μM.Induced shoot buds were multiplied and elongated on the MS medium supplemented with BAP (4.44 μM), NAA (5.36 μM) and TDZ (Thidiazuron) 2.27 μM with 9.66±0.33 (mean length 5.35±0.32 cm) and 9.00±0.57 (mean length 4.51±0.15cm) shoots using nodal segments and shoot tip explants, respectively. Halfstrength woody plant medium (WPM) containing 2.46μM indole-3-butyric acid (IBA) produced the maximum number of roots (6.00±1.15). The rooted plantlets were hardened on MS basal liquid medium and subsequently in polycups containing sterile soil and vermiculite (1:1) and successfully established in pots.

OBJECTIVES: This study was performed to evaluate the effect of blood contamination on the compressive strength (CS) of Root MTA (RMTA) modified with Calcium chloride (CaCl2) and Disodium hydrogen phosphate (Na2HPO4) as setting accelerators over time. MATERIALS AND METHODS: A total of 110 cylindrical specimens of RMTA were divided into 6 experimental groups as follows: Group1, RMTA; Group 2, RMTA modified with CaCl2 (RMTA-C); Group 3, RMTA modified with Na2HPO4 (RMTA-N); Group 4, RMTA contaminated with blood; Group 5, RMTA-C contaminated with blood; Group 6, RMTA-N contaminated with blood. The CS of specimens in all groups was evaluated after 3 hr, 24 hr, and 1 wk. In the modified groups (groups 2, 3, 5, and 6) the CS of five specimens per group was also evaluated after 1 hr. RESULTS: Blood contamination significantly reduced the CS of all materials at all time intervals (p < 0.05). After 3 hr, the CS of specimens in the RMTA groups (with and without blood contamination) was significantly lower than those in the RMTA-C and RMTA-N groups (p < 0.05). The CS values were not significantly different at the other time intervals. In all groups, the CS of specimens significantly increased over time (p < 0.05). CONCLUSIONS: Blood contamination decreased the CS of both original and accelerated RMTA.

ABSTRACT

O escurecimento enzimático e o amolecimento são as principais alterações que podem afetar pêssegos minimamente processados (MP). Objetivou-se avaliar a aplicação de L-cisteína (Cis), ácido L-ascórbico (AA) e cloreto de cálcio (CC) no controle do escurecimento enzimático e da perda de firmeza de polpa de pêssegos MP, cv. 'Esmeralda'. Os frutos foram processados e, após o fatiamento, imersos nos tratamentos: (I) água (controle); (II) AA+CC; (III) Cis+CC; e, (IV) Cis+AA+CC. Em seguida, as fatias foram acondicionadas em embalagens rígidas envoltas com filme PVC de 30µm de espessura e armazenadas durante seis dias a 4±1°C e 85±3 por cento UR. As amostras foram avaliadas quanto aos aspectos físico-químicos, enzimáticos e sensoriais a cada dois dias. O tratamento Cis+AA+CC resultou em maiores valores de L* e h°, o que indica ter sido o mais eficaz na prevenção do escurecimento. Os tratamentos com L-cisteína resultaram em menor atividade da polifenoloxidase (PPO) e o tratamento Cis+AA+CC foi o mais efetivo na contenção da atividade da peroxidase (POD), indicando que o melhor desempenho desse tratamento está associado à menor atividade de PPO e POD. A avaliação sensorial indicou que o tratamento Cis+AA+CC foi o mais eficiente, proporcionando a manutenção de características sensoriais aceitáveis aos pêssegos MP, cv. 'Esmeralda'.


The enzymatic browning and firmness loss are the major changes that could affect fresh-cut peaches. The objective was to evaluate the application of L-cysteine (Cis), L-ascorbic acid (AA) and calcium chloride (CC) in the control of browning enzymatic and loss of firmness of minimally processed dried peaches, cv. 'Esmeralda'. The fruits were processed industrially and, after slicing, immersed in treatments: (I) water (control); (II) AA+CC; (III) Cis+CC and (IV) Cis+AA+CC. After, the slices were packed in packages sealed with PVC film 30µm thick and stored for six days at 4±1°C and 85±3 percent UR. The samples were evaluated for their physical-chemical, enzymatic and sensory aspects every two days. Treatment Cis+AA+CC resulted in higher values of L* and h°, which indicates that it was the most effective in browning prevention. Treatments with L-cysteine resulted in lower activity of polyphenoloxidase (PPO) and treatment Cis+AA+CC was the most effective in containing peroxidase (POD) activity, indicating that the best performance for such treatment is associated with lower activity of PPO and POD. The sensory evaluation indicated that the treatment Cis+AA+CC was the most efficient, providing the maintenance of the sensory characteristics acceptable for fresh-cut peaches, cv. 'Esmeralda'.

This study assesses the effect of NaCl (80 and 160 mM) and CaCl2 (10 mM) solutions, alone and in combination, to 30-day-old seedlings of Cichorium intybus L. Observations were made at 30 day intervals from the time of treatment till harvest (180 days after sowing). Application of NaCl resulted in significant decreases in lengths of root and stem, in dry weights of root, stem and leaves and in the leaf area, as compared with control. The reduction was less with the combined application of NaCl and CaCl2 than with the NaCl treatment alone. On the contrary, treatment of CaCl2 alone promoted the above variables. Proline content in the leaves was enhanced with NaCl and CaCl2 alone as well as with the NaCl + CaCl2 treatments; the maximum (six-fold) enhancement was observed with the combined treatments, compared with NaCl (four-fold increase) and CaCl2 (two-fold increase) alone. The sodium (Na+) and Chloride (Cl) contents in different plant parts increased both with NaCl and with NaCl + CaCl2 treatments. The maximum accumulation was observed in leaves, followed by that in stem and root. The potassium (K+) and calcium (Ca2+) contents decreased under NaCl stress, but increased with CaCl2 treatment. Thus, calcium ameliorated the deleterious effects of NaCl stress and stimulated the plant metabolism and growth.

ABSTRACT

The efficiencies of different transformation methods of E. coli DH5α strain, induced by several cations like Mg2+, Mn2+, Rb+ and especially Ca2+, with or without polyethylene glycol (PEG) and dimethyl sulfoxide (DMSO) were compared using the two commonly used plasmid vectors pCAMBIA1201 and pBI121. The widely used calcium chloride (CaCl2) method appeared to be the most efficient procedure, while rubidium chloride (RbCl) method was the least effective. The improvements in the classical CaCl2 method were found to further augment the transformation efficiency (TR)E for both the vectors like repeated alternate cycles of heat shock, followed by immediate cold, at least up to the third cycle; replacement of the heat shock step by a single microwave pulse and even more by double microwave treatment and administration of combined heat shock-microwave treatments. The pre-treatment of CaCl2-competent cells with 5% (v/v) ethanol, accompanied by single heat shock also triggered the (TR)E, which was further enhanced, when combined heat shock-microwave was applied. The minor alterations or improved approaches in CaCl2 method suggested in the present study may thus find use in more efficient E. coli transformation.

ABSTRACT

Evidências científicas reconhecem o yacon (Smallanthus sonchifolius) como fonte promissora de frutanos, tais como inulina e frutooligossacarídeos (FOS). No entanto, a presença de compostos fenólicos torna-o suscetível à reação de escurecimento enzimático. Este trabalho teve como objetivo avaliar o emprego de agentes químicos no processamento de yacon, para obtenção de farinha, a fim de inibir o escurecimento enzimático do produto e favorecer o tempo de secagem. Amostras de yacon sem inibição química, de yacon submetido à solução de cloreto de cálcio 1,0g 100g-1 por 30 minutos e de yacon submetido à solução de metabissulfito de potássio 0,5g 100g-1 por 5 minutos foram secas a 55°C, em estufa ventilada, e o teor de água e curvas de secagem foram determinados. As atividades das enzimas peroxidase (POD) e polifenoloxidase (PPO) foram determinadas antes e após a secagem como um possível marcador bioquímico do escurecimento enzimático desse tubérculo. No parâmetro umidade, os três tratamentos apresentaram-se semelhantes; porém, o tratamento 2 (cloreto de cálcio) reduziu a umidade no menor tempo. Antes e após o tratamento térmico, a atividade enzimática foi maior no tratamento 3 (metabissulfito de potássio). As atividades enzimáticas não foram inativadas completamente pela ação térmica. Para a obtenção da farinha de yacon, o tratamento com cloreto de cálcio 1,0g 100g-1 por 30 minutos foi o que apresentou melhor resultado. Apesar de não inibir totalmente a atividade das enzimas POD e PPO foi o melhor tratamento de inativação, propiciando menor tempo de secagem e melhor firmeza da matéria-prima e facilitando o processamento para obtenção da farinha.


Scientific evidences acknowledged the yacon (Smallanthus sonchifolius) like a promise source of fructans, as inulin and fructooligosaccharides - FOS, meanwhile, the presence of phenolic coumpounds became this susceptible to enzimatic browning reaction. This project aimed to evaluate the use of chemical agents in yacon processing to obtain flour in a way that inhibits enzymatic darkening of the product besides determining the enzymatic activity in these treatments. Samples of yacon without chemical inhibitions, yacon treated with 1.0g 100g-1 calcium chloride for 30 seconds and yacon treated with 0.5g 100g-1 potassium metabisulfite for 5 minutes were dried at 55oC in a ventilated greenhouse and the proportions of humidity and drying curves were determined. The peroxidase activities and polyphenol oxidase enzymes were checked before and after being dried with an enzymatic darkening possible biochemist marker of this tubercle. Regarding humidity Parameter all the three treatment were equivalent, but treatment 2 (calcium chloride) reduced the humidity in lower time. Before and after the thermal treatment the enzymatic activity was higher in treatment 3 (potassium metabisulfite). The thermal action did not inhibit completely polyphenol oxidase and peroxidase. The treatment with calcium chloride at 1.0g 100g-1 for 30 minutes to obtain yacon flour was the one with better result despite the fact that it did not inhibit completely peroxidase and polyphenol oxidase enzymes activity, offering a better drying time better material of row firmness , facilitating the process for obtaining the meal.

Objetivou-se, neste trabalho, estudar o efeito de tratamentos com ácido ascórbico (AA), cloreto de cálcio (CC) e cloridrato de L-cisteína (LC) sobre a qualidade pós-colheita de banana 'Maçã' minimamente processada. Foram testadas as seguintes combinações: 1 (Controle); 2 (AA 1 por cento); 3 (CC 1 por cento); 4 (LC 0,5 por cento); 5 (AA 1 por cento + CC 1 por cento); 6 (AA 1 por cento + LC 0,5 por cento); 7 (CC 1 por cento + LC 0,5 por cento); 8 (AA 1 por cento + CC 1 por cento + LC 0,5 por cento). As bananas foram descascadas, fatiadas e imersas nos tratamentos, acondicionadas em bandejas, que foram envolvidas em filme de PVC 30 µm, e armazenadas a 5 ± 1ºC e 90 ± 3 por cento UR, durante 4 dias. As avaliações ocorreram nos dias 0, 1, 2 e 4. Houve um aumento na perda de massa durante o tempo de armazenamento, menor no tratamento 8 em relação aos demais. A acidez total titulável aumentou linearmente com o tempo em todos os tratamentos. A perda de firmeza foi mais eficientemente contida nas fatias sob os tratamentos com cloreto de cálcio. A formação de uma coloração vermelho-rósea sobre as fatias tratadas com L-cisteína foi observada e evidenciada pelos maiores valores de a*. O valor L* e o índice de escurecimento também foram afetados pelo tratamento com L-cisteína, cujo decréscimo nesses parâmetros de cor foi maior nas fatias não-tratadas, ao longo do tempo. O tratamento de banana 'Maçã' minimamente processada com a combinação AA 1 por cento + CC 1 por cento + LC 0,5 por cento foi eficiente para manutenção da sua qualidade pós-colheita.


This work had the aim of studying the effect of treatments with ascorbic acid (AA), calcium chloride (CC) and L-cysteine hydrochloride (LC) on the postharvest quality of fresh-cut 'Apple' banana. The following combinations were tested: 1 (Control); 2 (AA 1 percent); 3 (CC 1 percent); 4 (LC 0.5 percent); 5 (AA 1 percent + CC 1 percent); 6 (AA 1 percent + LC 0.5 percent); 7 (CC 1 percent + LC 0.5 percent); 8 (AA 1 percent + CC 1 percent + LC 0.5 percent). The bananas were peeled, sliced and immersed into the treatments, conditioned on trays, which were wrapped with 30 µm PVC film, and stored at 5 ± 1ºC and 90 ± 3 percent RH, during 4 days. Evaluations occurred at 0, 1, 2 and 4 days. There was an increase in the mass loss during the storage period, lower in the treatment 8 in relation to the others. Total titratable acidity increased linearly throughout the time in all treatments. The loss of firmness was prevented more efficiently in the treatments with calcium chloride. A pinkish-red color formation on the slices treated with L-cysteine was observed and evidenced by the highest values of a*. The L* value and the browning index were also affected by the treatments containing L-cysteine, whose decrease in these color parameters was higher in the untreated slices throughout the time. The treatment of fresh-cut 'Apple' banana with the combination AA 1 percent + CC 1 percent + LC 0.5 percent was efficient for its postharvest quality maintenance.

A banana constitui interessante alternativa para compor saladas de frutas. No entanto, possui curta vida de prateleira após o processamento mínimo em razão da rápida perda da firmeza. Neste trabalho, objetivou-se avaliar o efeito da mistura química contendo cloreto de cálcio, ácido ascórbico e L-cisteína e/com o uso de atmosfera modificada ativa (10 kPa CO2 e 2 kPa O2), enfatizando a perda de firmeza sobre a qualidade e vida de prateleira de banana 'Prata' minimamente processada. O conteúdo de pectina solúvel e a por cento de solubilização aumentaram significativamente ao longo do período de conservação. A perda de firmeza e o aumento das atividades das enzimas poligalacturonase e pectinametilesterase apresentaram uma interação significativa entre os fatores estudados (Tratamento/Tempo). As fatias tratadas com misturas químicas permaneceram com boas características para o consumo por até 3 dias de conservação. Os tratamentos com injeção inicial de gases de forma isolada, ou junto ao tratamento com mistura química, não propiciaram maior retenção da firmeza de banana 'Prata' minimamente processada em relação ao tratamento contendo L-cisteína (1 por cento p/v) + ácido ascórbico (1 por cento p/v) + cloreto de cálcio (1 por cento p/v) na mistura química.


Banana constitutes an interesting alternative to make fruit salad. Nevertheless, they have short shelf life after cutting due to the fast loss of firmness. The objective of this work was to evaluate the effect of chemical mixtures containing calcium chloride, ascorbic acid and L-cysteine with the use of active modified atmosphere (10 kPa CO2 and 2 kPa O2), emphasizing the lost of firmness on the quality and shelf life of fresh-cut 'Silver' banana. Soluble pectin content and the percentage of solubilization increased significantly throughout the conservation period. The lost of firmness and the increase of pectinmethylesterase and polygalacturonase activities presented a significant interaction among the studied factors (treatment/time). The slices treated with chemical mixtures remained with good characteristics for consume until 3 days of conservation. The treatments with an isolated initial gas injection or together with the chemical mixture treatment didn't promote better firmness retention of fresh-cut 'Silver' banana in relation to the treatment containing 1 percent (w/v) L-cysteine + 1 percent (w/v) ascorbic acid + 1 percent (w/v) calcium chloride in the chemical mixture.

Os músculos Biceps femoris, Longissimus e Triceps brachii submetidos aos efeitos do tempo de maturação e da injeção com cloreto de cálcio de cordeiros Morada Nova foram estudados quanto às características de cor, capacidade de retenção de água e maciez. Os cordeiros foram abatidos ao atingirem 25kg de peso vivo. Os músculos apresentaram diferenças na cor (luminosidade-L*, teor de vermelho-a* e teor de amarelo-b*) 24 horas após o abate. A maturação tendeu a escurecer a carne. O cloreto de cálcio não modificou a cor das carnes provenientes dos músculos Longissimus e Triceps brachii, entretanto as do Biceps femoris apresentaram-se mais vermelhas quando receberam cloreto de cálcio. Quanto à capacidade de retenção de água, a maturação afetou as carnes do Triceps brachii, mas não influenciou as carnes dos músculos Biceps femoris e Longissimus. O cloreto de cálcio não modificou a capacidade de retenção de água dos músculos avaliados. A maturação influiu sobre a maciez do Biceps femoris e do Longissimus


Biceps femoris, Longissimus and Triceps brachii muscles from Morada Nova lambs were submitted to ageing and calcium chloride injection. Colour, water holding capacity and tenderness were studied. Lambs were slaughtered weighting 25kg.. The muscles presented differences in colour (lightness-L*, redness-a* and yellowness- b*) 24 hours after rigor mortis instalation. Ageing intensified redness of the meats. Calcium chloride did not modify the colour of Longissimus and Triceps brachii, however, Biceps femoris became more redness after receiving calcium chloride. In relation to water holding capacity, ageing affected meats from Triceps brachii. However, it did not affect Biceps femoris and Longissimus. The calcium chloride didn't modify the water holding capacity of the muscles. Ageing influenced tenderness of Biceps femoris and Longissimus
