ABSTRACT

Abstract The Brazilian native species Cestrum intermedium, known as mata-boi, induces hepatotoxicity and death when ingested by cattle. While most studies on this species focus on toxicological features, our study is the first to describe the anatomy and in vitro biological activities of Cestrum intermedium. We investigated adult leaves and stems by histochemistry, described their anatomy, performed physical-chemical analysis, determined in vitro antioxidant and antimicrobial activities, and identified secondary metabolites. A few noteworthy anatomical features were the anomocytic stomata on the abaxial surface and the absence of trichomes, in addition to the circular shaped petiole with two projections on the adaxial surface. Histochemical analysis showed chemical markers such as alkaloids, usually reported as toxic, and terpenoids. Potassium nitrate (ATR-FTIR) and lupeol palmitate (NMR) were detected on the crude stem extract. Thermogravimetric and physical-chemical analysis provided fingerprint parameters for the species. Minimal Inhibitory Concentration (MIC) assay revealed that Staphylococcus aureus, Staphylococcus epidermidis, and Candida albicans were weakly inhibited by extract samples. Chloroform and ethyl acetate fractions presented high phenolic content, which resulted in in vitro antioxidant activity. These novel features expand the knowledge about this species, considering that previous studies mainly focused on its toxicity. Our study also provided characteristics that may help in avoiding misidentification between Cestrum members, especially when taxonomic keys cannot be employed, as in the absence of flowers and fruits.

ABSTRACT

OBJECTIVE:To study the diff erences in the overall chemical constituents of Angelicae dahuricae with and without sulphur-fumigation,and to select chemical markers that can be used for identification. METHODS :Each 16 batches of A. dahuricae with and without sulphur-fumigation were selected as samples. Deuterated methanol-deuterium oxide (1 ∶ 0.2,V/V)was used as extraction solvent ,and 1H-NMR metabolomics was used to detect the primary and secondary metabolites in A. dahuricae . In addition ,visual analysis ,principal component analysis (PCA)and t-test were used to compare the component differences of A. dahuricae by two kinds of pretreatment methods. RESULTS :A total of 19 chemical constituents such as coumarins ,amino acids and sugars were simultaneously detected and identified from two kinds of A. dahuricae . Visual analysis showed that the chemical profiles of the two kinds of A. dahuricae were similar ,but their coumarins and carbohydrates were quite different in 1H-NMR signal intensity. PCA analysis showed that all samples could be divided into two categories according to different treatment methods. The results of t-test showed that the contents of oxypeucedanin ,imperatorin,glucose and sucrose of A. dahuricae were decreased significantly after sulphur fumigation (P<0.01 or P<0.001),while the contents of alanine and leucine were significantly increased (P<0.01 or P<0.001). There was no statistical significance in the contents of other 13 kinds of components (P>0.05). CONCLUSIONS:There are obvious differences in the 6 chemical compounds of oxypeucedanin ,imperatorin,glucose,sucrose, alanine and leucine in A. dahuricae with and without sulphur-fumigation ,which can be used as chemical markers for the identification of sulphur-fumigated A. dahuricae in the market.

ABSTRACT

Objective: To screen the differential ingredients between crude and wine-processed Corni Fructus and determin their content. Methods: An integrated strategy using ultra performance liquid chromatography coupled with tandem quadrupole time-of- flight mass spectrometry (UPLC-Q-TOF/MS) and the chemometric approach was applied to compare the global chemical profile of crude and wine-processed Corni Fructus. Then, the main differential ingredients were quantified by UPLC-PDA. Results: The chemical profiling of wine-processed Corni Fructus was significantly different. Ten compounds could be considered as characteristic chemical markers for distinguishing crude and wine-processed Corni Fructus, including 5-hydroxymethyl furfuraldehyde (5-HMF), gallic acid, protocatechuic acid, morroniside, loganic acid, sweroside, cornin, dihydroquercetin, loganin and cornoside. A new UPLC-PDA quantitative method for analyzing simultaneously the above ten compounds in wine-processed Corni Fructus was established. The results of methodology investigation showed that the ten components were well linear within the investigation range (r ≥ 0.999 7). Compared with the crude Corni Fructus, the content of seven components were increased, including gallic acid, 5-HMF, loganin, morroniside, cornin, sweroside and dihydroquercetin, and the other three components in wine-processed Corni Fructus were decreased. Conclusion: The differential ingredients obtained by chemometric-based approach can be used to distinguish crude and wine-processed Corni Fructus. The determination method of wine-processed Corni Fructus established is accurate and reliable, which can be used for the quality control of Corni Fructus.

ABSTRACT

PURPOSE: Catheter direct thrombolysis (CDT) has been shown to be an effective treatment for deep venous thrombosis. The objective of the review is to improve safety and efficacy of the CDT by using ward based protocol, better able to predict complications and treatment outcome through monitoring of haemostatic parameters and clinical observation during thrombolysis procedure. MATERIALS AND METHODS: MEDLINE, EMBASE, CENTRAL and Web of Science were searched for all articles on deep venous thrombosis, thrombolysis and correlations of clinical events (bleeding, successful thrombolysis) during thrombolysis with hemostatic parameters to March 2016. The risk of bias in included studies was assessed by Cochrane Collaboration’s tool and Cochrane Risk of Bias Assessment Tool: for Non-Randomized Studies of Interventions. RESULTS: Twenty-four studies were included in the review and we found that improving safety and efficacy of CDT by using ward based protocol depending on eight factors; strict patient selection criteria, types of fibrinolytic drugs, mode of fibrinolytic drug injection, biochemical markers monitoring (fibrinogen, D-dimer, activated partial thromboplastin time, plasminogen activator inhibitor-1), timing of intervention, usage of intermittent pneumatic calf, ward monitoring and thrombolysis imaging assessment (intravascular ultrasound). These factors may help to improve safety and efficacy by reducing total thrombolytic drug dosage and at the same time ensure successful lysis. There is a marked lack of randomized controlled trials discussing the safety and efficacy of catheter direct thrombolysis. CONCLUSION: CDT can be performed safely and efficiently in clinical ward, providing that careful nursing, biochemical monitoring, proper selection and mode of infusion of fibrinolytic drugs, usage of Intermittent pneumatic calf and adequate thrombolysis imaging assessment are ensured.

ABSTRACT

Sulfur-containing Anemarrhenae Rhizoma decoction pieces were prepared by using sulfur-fumigating procedure. The difference components before and after sulfur fumigation in Anemarrhenae Rhizoma were analyzed and on-line identified by UPLC-Q-TOF-MSE combined with UNIFI informatics platform, principal component analysis (PCA) and orthogonal partial least squares discriminant analysis (OPLS-DA) respectively. As a result, 16 major differences components were identified, and among them, 9 components were mainly from sulfur-fumigated samples. The main chemical markers in sulfur-fumigated Anemarrhenae Rhizoma were identified as the sulfite derivatives newly produced after sulfur-fumigating. Meanwhile, UPLC-Q-TOF-MSE was used to find the main chemical marker anemarrhena saponin BⅡ sulfite (m/z 983). By using this method, a rapid screening method for sulfur-fumigated Anemarrhenae Rhizoma was established. This was a convenient and accurate detection method for sulfur dioxide residue, and it can be used as an effective assistant method to control the quality of Anemarrhenae Rhizoma. At the same time, it was the first time to identify sulfited derivatives of steroidal saponins, and screen the sulfur-fumigated Anemarrhenae Rhizoma.
