ABSTRACT

ABSTRACT Corn is used to massively produce food for humans and domestic animals with grains of various taxonomic entities or races. For domestic ruminants, the whole plant is also used as forage. In Argentina, both corn grain and whole-plant silage are used for beef and dairy cattle production. This paper aimed to develop the history and perspectives of corn grown for grain and silage, focusing on plant breeding. The importance of corn fodder in its two variants (grain and whole-plant silage) has varied over time. We emphasize herein the importance of the specific genetic breeding of corn used for whole-plant silage to achieve higher yield potential and nutritional value than grain hybrids. The four population genetic structures used over time as cultivars, which determined the evolution of the technological process of selection and breeding, were analyzed. Based on the research carried out, seed companies have incorporated our protocols into their development and breeding programs. The contribution of selection and breeding in Argentina was effective in transforming the corn plant into ruminant feed, and this will increase with the development of specific silage hybrids.

ABSTRACT

ABSTRACT The prairie grass (Bromus catharticus Vahl) is the annual/biennial native forage grass of temperate climates of most importance and diffusion in Argentina. Several authors have studied the phenotypic variability in morphophysiological characters in cultivars and native populations of prairie grass. The first commercial cultivars were heterogeneous populations that were considered the best adapted in the temperate region of Argentina. Genetic, agronomic and molecular characterization of the selection base populations and of the germplasm conserved in the active bank of INTA Pergamino, contributed to knowing and conserving the available genetic diversity and, at the same time, provided usable genetic variability in breeding. The most representative cultivars considered in this work were Pergamino Martín Fierro MAG; Fierro Plus INTA, Bar INTA 200, Rosalía INTA and INTA Calvu, in which technological packages associated with the utilization in animal production were developed.

ABSTRACT

In this study, an established ultra-high performance liquid chromatography coupled with quadrupole-time-of-flight mass spectrometry(UPLC-Q-TOF-MS) method was combined with multivariate statistical analysis to investigate the commonality and difference of main chemical components in the medicinal parts of Paeonia lactiflora from different cultivars; in addition, a high performance liquid chromatography(HPLC) method was established to simultaneously determine the content of eight active components in Paeoniae Radix Alba. Non-targeted analysis was carried out by UPLC-Q-TOF-MS on a Waters ACQUITY UPLC BEH C_(18)(2.1 mm×100 mm, 1.7 μm) column with a gradient elution of 0.1% aqueous formic acid(A)-acetonitrile(B) as the mobile phase at a flow rate of 0.2 mL·min~(-1). The column temperature was 30 ℃, and an electrospray ionization source was used to acquire mass spectrometry data in positive and negative ion modes. According to the accurate molecular weight and fragment ion information provided by multi-stage mass spectrometry and by comparison with reference substances and literature reports, thirty-six identical components were identified in Paeoniae Radix Alba from different cultivars with positive and negative ion modes. In the negative ion mode, two groups of samples were well separated; specifically, seventeen components with significant differences in content were screened and identified, and one component unique in "Bobaishao" was obtained. Quantitative analysis was conducted by high-performance liquid chromatography(HPLC) on an Agilent HC-C_(18)(4.6 mm×250 mm, 5 μm) column with a gradient elution of 0.1% aqueous phosphoric acid(A)-acetonitrile(B) as the mobile phase at a flow rate of 1.0 mL·min~(-1). The column temperature was 30 ℃ and the detection wavelength was at 230 nm. An HPLC method was developed for the simultaneous determination of eight active components(gallic acid, oxypaeoniflorin, catechin, albiflorin, paeoniflorin, galloylpaeoniflorin, 1,2,3,4,6-O-pentagalloylglucose, benzoyl-paeoniflorin) in Paeoniae Radix Albaa from different cultivars. Satisfactory linearity was achieved within the investigated linear ranges and with fine coefficients(r>0.999 0), and the methodological investigation showed that the method had good precision, repeatability and stability. The mean recoveries were 90.61% to 101.7% with RSD of 0.12% to 3.6%(n=6). UPLC-Q-OF-MS provided a rapid and efficient qualitative analytical method for the identification of the chemical components in Paeoniae Radix Alba, and the developed HPLC method was simple, rapid and accurate, which could provide a scientific basis for the evaluation of the germplasm resources and herbal quality of Paeoniae Radix Alba from different cultivars.

ABSTRACT

Background and Objective: Plantain (Musa paradisiaca L) remains one of themost important staple food crop and perhaps, one of the oldest cultivated fruit tree crop in the humid tropics of Africa, Central Asia, South America and the West Indies.Fourteen (14) elite plantain cultivars were evaluated for genetic diversity using agro-morphological yield related attributes and simple sequence repeat (SSR) markers. Materials and Methods: Six (6) microsatellite markers that showed distinct fragments varying from 50 bp to 3.0 Kbp in size of polymorphic bands were selected and used for molecular characterization and fingerprinting, while agro-morphological (yield杛elated) attributes assessed included bunch weight, number of hands/bunch, number of fingers/hands, number of fingers/bunch, harvest interval, length of plant cycle, pulp hardness and pulp to skin weight ratio of the elite plantain cultivars. Results: The total number of amplified bands (TNB), mean percentage polymorphism (%P), mean polymorphic information content (PIC), average marker index (MI) and mean gene diversity for the SSR assay were 59, 70.24%, 0.79, 3.74 and 0.832 respectively. Results of agro-morphological fingerprint study revealed a significant variations in terms of the bunch weight, number of finger per hands/bunch, number of fingers per hand, number of fingers /bunch, harvest interval, length of crop cycle, pulp hardness and pulp/wt. ratio all showed significant variations among the cultivars. The distribution of the elite cultivars along with the principal components showed cluster pattern of distribution within the study location. Principal component analysis revealed four principal components contributing 99.91% to the observed morphological variations while analysis of molecular variance revealed 96.00% contributed by molecular characteristics to observed variations. The yield displayed revealed significant contributions of bunch weight, fingers/hand and fingers/bunch as the main indices for plantain yield. The dendrograms for both morphological and molecular characteristics delineated the cultivars into four distinct cluster groups and subgroups each varying in genetic distance. Conclusion: These good cultivars can be exploited for the improvement of low yielding cultivars in other region to increase and improve plantain yield, promote food security and income generation especially under the present economic realities where food security is threatened by the global food crises and declining crop productivity.

ABSTRACT

The diversity of yeast species on grape berries changes depending on various factors. Determination of indigenous yeast intensity and diversity on grape berries is important for increasing the sensory characteristics of wine as well as fermentation efficiency. The natural yeast biota of grape berries affects wine flavour and quality by producing some secondary metabolites and hydrolytic enzymes. Despite the application of different nonconventional yeasts in food and fermentation industries, many significant researches are conducted in finding and improving the new strains having industrially important enzyme activities. Invertase enzyme has a vital role in the food industry in which it increases the sweetness of food without crystallizing them. Here, we studied yeast diversity in grape berries from selected localities and also their invertase activity. We collected grape berries from Alphonse, K?nal? Yap?ncak, Çavu?, Efes Karas?, Cinsaut, Atasar?s? and Isabella grape varieties cultivated in Bozcaada island and Gelibolu peninsula. Twenty-one yeast species belonging to seven genera were identified. The yeast strains having high invertase activities were identified with 5.8S-ITS rDNA sequencing technique. The diversity of yeast biota on berries collected from Gelibolu was greater than that of Bozcaada. Metschnikowia pulcherrima, Cryptococcus laurentii and Rhodotorula glutinis yeast species were dominant yeast species on grape berries. A total of 294 sucrose grown yeast strains showed growth on sucrose, and 19 of them exhibit the highest invertase activity that is not glucose repressible. These 19 yeast strains were identified as Starmerella bacillaris using 5.8S ITS rDNA region and the phylogenetic analysis was inferred with the Maximum Parsimony method.

ABSTRACT

The goal of this work was to assess the biomass production and bromatological quality of ryegrass genotypes in ten municipalities of the Western and North Plateau regions of the State of Santa Catarina, Brazil. The cultivars La Estanzuela 284 (diploid), Bar HQ, Barjumbo, INIA Escorpio, Potro, and Winter Star (tetraploids) were compared, distributed in a randomized block design, in which the municipalities constituted the blocks, with three replications. The cuts were performed when the plants reached 30cm, leaving a residue of 10cm. In three cuts, the cultivars Barjumbo and Bar HQ were the most productive, exceeding 4.6 t ha-1 of dry matter. In the places in which five cuts were performed, the production of these cultivars exceeded 7.3 t ha-1, placing them again ahead of the others. The average crude protein content in three cuts was greater than 25% in all cultivars. There was no difference between the genotypes in the content of neutral detergent fiber and total digestible nutrients. There was a significant correlation between quantitative and qualitative productive variables. The assessed cultivars represented good options for composing short-term or long-term winter-feeding systems, adjusted to the integration with annual crops or warm-season pastures.(AU)

ABSTRACT

BACKGROUND: An efficient regeneration protocol is a priority for the successful application of plant biotechnology. Grape nodal explants were used to develop a micropropagation protocol for Thompson Seedless and Taify cvs. Explants were cultured on MS medium supplemented with Kinetin or benzylaminopurine (BA) and indolebutyric acid (IBA). RESULTS: For both cultivars, axillary buds were grown, only, on a medium enriched with kinetin, moreover, shoot tip necrosis and callus formation were observed on Thompson Seedless cv. cultures grown on a medium with BA. Supplementing the growth medium with 100 mM (boron) B and 2.5 mM (calcium) Ca successfully help overcome these phenomena. The highest regenerated shoot numbers (14 and 6.2 explant 1 ) for Taify and Thompson Seedless cvs., respectively, were on media supplemented with 13.2 mM BA + 4.9 mM IBA and BA 13.2 mM + 5.8 mM IBA, respectively. Moreover, these media supported the developing shoots to have the heaviest dry weights (1.46 and 0.72 mg explant 1 ) for Taify and Thompson Seedless cvs., respectively. Thompson Seedless cv. regenerated shoot numbers and their dry weights were significantly increased by increasing the MS medium PO4 concentration. However, these two parameters were significantly decreased for Taify cv. Developing shoots were elongated and rooted on MS medium enriched with 4.9 mM, IBA 100 mM B and 2.5 mM Ca. Plantlets were acclimatized and successfully transferred to the greenhouse conditions. CONCLUSIONS: A novel promising protocol for Thomson Seedless and Taify cvs. micropropagation using single nodes has been developed.

ABSTRACT

Guava, is one of the most promising fruit crops of India and is considered to be one of the exquisite nutritionally valuable and remunerative crops. We are unaware of any report describing macro and micronutrient dynamics in fruit at different growth stages of guava. Micronutrients play an important role in production and their deficiency lead in lowering the productivity. For conducting this experiment fruit of variety Allahabad Safeda, L-49, Lalit, Shweta, Arka Kiran, Salithong, Kimchu were collected at different stages like Marble, Stone hardening & Harvest stage for estimation of primary nutrient (N, P, K), secondary nutrient (Ca, Mg) & micronutrient (Fe, Mn, Zn, Cu). The nutrient content particularly N, K, Mg, and Mn are highest in variety of Allahabad Safeda, whereas, P and Ca are highest in variety Lalit. Micronutrient Fe recorded highest in Salithong while Zn and Cu were accumulated maximum in Arka Kiran and Kimchu respectively. Recommendation of fertilizer at various growth stages is paramount for precise nutritional management for which the requirement of different nutrition is essential.

ABSTRACT

BACKGROUND: Wheat is one of the most important crops cultivated all over the world. New high-yielding cultivars that are more resistant to fungal diseases have been permanently developed. The present study aimed at the possibility of accelerating the process of breeding new cultivars, resistant to eyespot, by using doubled haploids (DH) system supported by marker-assisted selection. RESULTS: Two highly resistant breeding lines (KBP 0916 and KBH 4942/05) carrying Pch1 gene were crossed with the elite wheat genotypes. Hybrid plants of early generations were analyzed using endopeptidase EpD1 and two SSR markers linked to the Pch1 locus. Selected homozygous and heterozygous genotypes for the Pch1-linked EpD1b allele were used to produce haploid plants. Molecular analyses were performed on haploids to identify plants possessing Pch1 gene. Chromosome doubling was performed only on haploid plants with Pch1 gene. Finally, 65 DH lines carrying eyespot resistance gene Pch1 and 30 lines without this gene were chosen for the eyespot resistance phenotyping in a field experiment. CONCLUSIONS: Results of the experiment confirmed higher resistance to eyespot of the genotypes with Pch1 in comparison to those without this gene. This indicates the efficiency of selection at the haploid level.

ABSTRACT

In recent decades, the expansion of soybean production in Brazil has been observed. This advance was motivated by the search of environments with better cultivation conditions, as well as the development of genotypes with wide adaptation. The diversity of "environments" hinders the process of selection and recommendation of cultivars, since the productive potential of a cultivar is given as a function of the sum of the genotype effect, the environment and the interaction between the latter two (G x E). In the case of this G x E interaction, regional studies are necessary in order to detail the differential behavior of the cultivars. Thus, the objective of this work was to evaluate the genotype interaction by environments, adaptability and phenotypic stability for grain yield, of fifteen soybean cultivars, in four sowing seasons, in order to identify cultivars that combine high productive potential, predictability of behavior and adaptation to the edaphoclimatic conditions of Uberlândia-MG. The trials were conducted at the Experimental Farm Capim Branco, in Uberlândia-MG. Fifteen soybean cultivars were evaluated in four sowing seasons (October 23, 2016, November 19, 2016, December 10, 2016 and January 14, 2017), in relation to grain yield. The experimental design was of randomized complete blocks, with three replications, in each season. Data were submitted to individual and joint analyzes. The G x E interaction was decomposed by the method proposed by Cruz andCastoldi (1991). The differential behavior of the genotypes was detailed by the adaptability and phenotypic stability by the methods of Eberhart and Russell (1966), Lin and Binns (1988) modified by Carneiro (1998), AMMI and Centroid. By the analysis of joint variance, it was observed the existence of the cultivar interaction by sowing times (C x E), for the grain yield trait, at the 5 % probability level by the F test. The C x E was predominantly complex in nature. The cultivar CD 2737 RR presented satisfactory results for the four sowing seasons in Uberlândia-MG, with high grain yield and predictability of behavior, by the evaluated methods. The cultivar NS 6909 IPRO was classified into favorable environments by the methods of Eberhart and Russell (1966) and Lin and Binns (1988) modified by Carneiro (1998). Considering Lin and Binns (1988) modified by Carneiro (1998) and Centroid the cultivar that is also classified for this cultivation condition is UFUS 8301. By AMMI, UFUS 7415, CD 2737 RR and UFUS Milionária are considered stable and adaptable

ABSTRACT

ABSTRACT: The aim of this study was to examine the impact of cultivar and spear color on the composition of polyphenols in asparagus spears (Asparagus officinalis). The five genotypes (Schwetzinger Meisterschuss, Huchel's Alpha, Gijnlim, Grolim and Eposs) and three growing conditions of asparagus spears (Asparagus officinalis) were investigated. The polyphenols were determined by applying the HPLC-DAD system. The obtained results were subjected to the principal component analysis. Among the analyzed asparagus samples cv. Grolim contained the highest amounts of phenolic acids and flavonols. The varied quantitative and qualitative composition of polyphenolics resulted most probably from changes occurring during vegetation, such as a lack of access to light in the case of white asparagus and limited access to light in purple asparagus. The scavenging activity on DPPH radicals by asparagus extract is dependent on the variety and color and was the greatest for green asparagus samples. Similar green extracts scavenged ABTS radicals to the highest degree. Results of this study suggested that asparagus may constitute a good source of natural antioxidants to be used in our diet as well as by industries for functional food formulations.

ABSTRACT

The objective of this work was to evaluate the performance of basil cultivars and hybrids in two cropping seasons. The experiments were conducted in a randomized block design, with three replications. Twenty-four basil genotypes were tested (20 commercial cultivars and four hybrids), in the dry season (oct.-dec./2015) and in the rainy season (apr.-jun./2016). The evaluated variables were: plant height (cm plant-1), canopy width (cm plant-1), leaf dry weight (g plant-1), and essential oil yield (mL plant-1). The data of each period were subject to analysis of variance and the means were grouped by the Scott-Knott test (p0.05). The individual and joint analyses of variance were performed for the experiments in both seasons. The estimated parameters were: coefficient of genetic variation (CVg), coefficient of environmental variation (CVe), CVg/CVe ratio, and heritability (h2). Leaf dry weight values per plant ranged from 6.23 to 75.00 g plant-1 (dry season) and from 9.17 to 31.34 g plant-1 (rainy season). The hybrid Cinnamon x Maria Bonita (1.50 mL plant-1) and the cultivar Mrs. Burns (1.44 mL plant-1) presented higher essential oil yield in the dry season. All the evaluated variables showed high heritability (h2) (> 50%) and CVg/CVe ratio (> 1.0), in both cropping seasons, indicating a favorable condition for selection. The cropping season influences the biomass and essential oil yield of basil.

Cassava is a basic staple in Cameroon whose industrialization faces major constraints especially those related to the management of whiteflies (Bemisia tabaci), a vector of the African cassava mosaic virus (ACMV). This work has as main objective to identify the tolerance of some cassava genotypes to whiteflies and to undertake an agro-morphological characterization of infected cultivars to the ACMV. Data were collected from September to November 2015 on six cultivars: Five improved (TMS92/0057, TMS92/0067, TMS92/0326, TMS96/1414 and TME419) and one local (NTOLO), on a young plot (three months old) and mature plot (six months age). Adult whiteflies were counted on the underside of the last leaves of one apex and for nymphs; they were counted randomly between the ninth and fourteenth leaves. Three cultivars (TMS96/1414; TMS92/0057 and TMS92/0067) ramified early with a large number of apices. For the younger plants (three months old), three cultivars (NTOLO, TMS96/1414 and TMS92/0326) harbored the largest number of adult whiteflies, whereas two others (TMS92/0326 and TME 419) were the most attacked by the ACMD. Younger plants (3 Months old) were most vulnerable to ACMV attack (r = 0,307** with whiteflies population), while the older ones (at 6 Months), represent early senescence period for short-cycle genotypes.

ABSTRACT

Breeding genotypes need to be characterized and evaluated in different soil and climatic conditions. There are few studies on association between characters evaluated and their contribution to banana genetic diversity, being essential to guide genetic breeding programs. This study aimed to evaluate the physicochemical characters of banana genotypes, to estimate associations between characters, to determine the relative importance of characters to study on genetic dissimilarity, and to indicate new genotypes to coastal plain region of Sergipe. Fruits from thirteen genotypes were used: Prata (FHIA-18, BRS Platina, PV94-01, BRS Garantida, YB42-47, Pacovan, and Prata Anã), Maçã (BRS Princesa, BRS Tropical, and Maçã), Ibota (Caipira), Gros Michel (Bucaneiro), and Mysore (Thap Maeo). The experimental design was randomized blocks, with thirteen genotypes and three replications. There is physicochemical variability of fruits between genotypes of same genomic group and subgroup, and between hybrids originated from same progenitor. Content of total sugars has greater contribution to genetic diversity among the genotypes, followed by fruit mass and pulp mass, and starch content in two cycles. Associations between fruit length, fruit diameter, and fruit mass and pulp mass in two production cycles based on phenotypic correlation are highly significant. Hybrids FHIA-18, PA94-01, YB42-47, and BRS Tropical, BRS Princesa, and Bucaneiro cultivars are promising for recommendation on coastal plain regions.

ABSTRACT

Objective: To evaluate the total phenolic contents, antioxidant and antiglycation activities of leaves, barks, roots and kernels from two cultivars of Mangifera indica (Anacardiaceae). Methods: Total phenolic contents were determined by using Folin-Ciocalteu's method. The antioxidant activities were assessed by three different protocols including DPPH, oxygen radical absorbance capacity and iron (II) chelation assays. In addition, in vitro bovine serum albumin/D-ribose assay was chosen to evaluate the antiglycation properties of the extracts. Results: All the investigated extracts were found to contain high level of total phenols as well as potent antioxidant activities. Kernel extracts showed the highest total phenol contents and DPPH radical scavenging activities whereas higher oxygen radical absorbance capacity values were observed for leave, root and bark extracts. Besides, extracts from leaves, roots and barks from both cultivars exhibited potent inhibitory effects against the formation of advanced glycation end products, with IC

ABSTRACT

ABSTRACT: This study aimed to investigate factors influencing the adoption of improved cultivars (ICs) in peach production in Khyber Pakhtunkhwa province of Pakistan. A total of 270 respondents were randomly selected from the three different cultivated areas of Khyber Pakhtunkhwa, namely, Peshawar, Nowshera and Swat. Binary choice model was used in this study to categorise the ICs of peach farmers into adoption and non-adoption. The study identifies that socio-economic, institutional farm resources, and climatic factors are influencing the adoption of ICs of peach production. Results of the estimated model reveal that farmer's age, education, household size, membership, cell phone, farm size, extension services and the role of the non-government organization have a positive effect on adoption of ICs. In addition, farmer's experience, off-farm income, livestock and machinery ownership, credit access and inputs prices have a positive and significant impact on ICs adoption. Moreover, results of the logit model demonstrate that climatic related factors have a highly significant and positive impact on the adoption of ICs. These results suggested that institutional services should be strengthened to provide managerial and technical skills on ICs technology adoption and on time provision of financial services to enhance the productivity of peach farmers.

ABSTRACT

Field experiment should be carried out with high accuracy since the discarding of a trial involves waste of time and resources, and can mislead breeders in selecting inferior genotypes. The simulation of a large number of experiments with great variability of traits is the main strategy to assist breeders on how to perform the experiment. Therefore, the objective of this study was to evaluate two criteria to infer the experimental quality: Coefficient of Variation (CV) and repeatability coefficient. Experimental data from evaluations of common bean lines carried out in 104 experiments were used in this study. The simulation of the scenarios used the following parameters: number of cultivars, number of blocks, genetic variance, mean yield, and coefficient of variation. Two thousand replications for each scenario were simulated. Genotypes means were arranged in an order, and the Spearman correlation was obtained between the estimated genotypes mean and the true genotypic values. Results revealed that CV is not a reliable parameter to evaluate the quality of experiment in the field. In conclusion, repeatability coefficient is the parameter that defines the discarding criteria of evaluation experiments and the recommendation of cultivars, as long as the values for each response variable are established.
