ABSTRACT

Objective:To evaluate the clinical efficacy of Zhiwei pill for bile reflux gastritis (BRG) with symptoms of disharmony of liver and stomach, evaluate the safety after treatment by three-arm study, and investigate the regulatory action to gastrointestinal hormones. Method:One hundred and eighty patients were randomly divided into placebo group (60 cases), control group (60 cases) and observe group (60 cases) by random number table. Patients in three groups got lifestyle intervention of non-drug therapy. Patients in control group got hydrotalcite tablets 1-2 hours after meal or untimely bedtime or in case of stomach pain, 2 tablets/time, 3 times/day. Patients in placebo group got simulated medicine of Zhiwei pill 1 hour after meal, 6 g/time, 3 times/day. Patients in observe group got Zhiwei pill 1 hour after meal, 6 g/time, 3 times/day. The treatment continued for 4 weeks in all 3 groups. Before and after treatment, scores were graded for symptoms of disharmony of liver and stomach and patient reported outcome (PRO) scale for patients with chronic gastrointestinal diseases. Gastroscopy was taken to grade the scores of bile reflux and mucosa under gastroscope. Levels of gastrin (GAS), motilin (MTL), cholecystokinin (CCK), substance P (SP) and gastrointestinal hormones were detected. Efficacy for traditional Chinese medicine(TCM) syndrome and efficacy under gastroscope were compared, and safety after treatment was evaluated. Result:After treatment, the total scores of the main symptoms and 7 secondary symptoms of disharmony of liver and stomach in observe group were all lower than those in placebo group and control group (<italic>P</italic><0.01), and scores of symptoms of disharmony of liver and stomach and PRO were all lower than those in control group and placebo group (<italic>P</italic><0.01). After treatment, scores of degree of bile reflux and mucosa under gastroscope in observe group were all lower than those in placebo group and control group (<italic>P</italic><0.01). Level of CCK was lower than that in placebo group and control group (<italic>P</italic><0.01), while levels of MTL, GAS and SP were higher than those in placebo group and control group (<italic>P</italic><0.01). Total effective rate of TCM syndrome was 91.38%(53/58), higher than 71.93%(41/57) in control group and 34.62%(18/52) in placebo group. The total effective rate in gastroscopy was 93.10%(54/58), higher than 78.95%(45/57) in control group and 28.85%(15/52) in placebo group (<italic>P</italic><0.05 or <italic>P</italic><0.01). No adverse reactions related to Zhiwei pill were found. Conclusion:Zhiwei pill can improve bile reflux, promote the healing of mucosa under gastroscope, regulate gastrointestinal hormones, improve gastrointestinal motor function, significantly alleviate clinical symptoms, and improve quality of life, with better efficacy for TCM syndrome and efficacy in gastroscopy, and it is safe for clinical use.

ABSTRACT

Abstract Clavibacter michiganensis subsp. michiganensis (Cmm), which is a Gram positive bacterium, causes the bacterial canker of tomato disease. The purpose of the study was to evaluate the antibacterial activity of Polygonum orientale extracts against Cmm. In this study, firstly, effects of three extracting parameters (extractive time, extractive temperature, and solid to liquid ratio) of orthogonal experiment design L27 (313) were conducted. Secondly, survival rate was determined and inhibition zone of Cmm rescued post-stress was monitored. Finally, extracellular OD260nm value, extracellular protein content, conformational structure of membrane protein, extracellular alkaline phosphatase (AKP) activity, and ATPase activity were measured to investigate the antibacterial mechanism. Results of orthogonal experiment revealed that extractive time and extractive temperature had highly significant (P<0.01) effects on the antibacterial activity of P. orientale extracts. The optimum conditions were as follows: 10h of extractive time, 60°C of extractive temperature, and 1:20 (g:mL) of solid to liquid ratio. This study also demonstrated that the living cells of each sample from survival rate test had almost no resistance or adaptability, and rescued Cmm cells were much easier to be inhibited by P. orientale extracts. The results of antibacterial mechanism indicated that cell membrane and cell wall of Cmm were seriously damaged by P. orientale extracts, and P. orientale extracts reduced the intracellular ATPase activity dramatically. All these findings suggested that P. orientale extracts had a strong antibacterial activity to inhibit Cmm, and could be used for the ecological management of the bacterial canker of tomato disease.

ABSTRACT

Resumen Se sintetizó el catalizador Cu/MCM-41 mediante impregnación húmeda incipiente y se caracterizó por DRX, XPS, TPD-NH3 y adsorción-desorción de N2. Se evaluó el efecto de diferentes condiciones de reacción (temperatura, tiempo, tipo de solvente y cantidad de catalizador) en la isomerización del epóxido de α-pineno para la obtención de aldehído canfolénico con el catalizador Cu/MCM-41, que no había sido reportado previamente para este tipo de reacción. Se partió de un diseño experimental central compuesto mediante análisis de superficie de respuesta. Se encontró que la mejor selectividad fue de 85% (5 mg de catalizador; 70 °C; 0,5 h y el uso de acetato de etilo como solvente). Adicionalmente, se determinaron los principales factores y sus combinaciones que tenían mayor significancia en la síntesis del aldehído ajustado a un modelo polinomial de segundo grado. Se encontró que los factores que tienen mayor influencia en la conversión son la cantidad de catalizador, tiempo de reacción, el tipo de solvente y el factor combinado entre la cantidad de catalizador y el tipo de solvente. En el caso de la selectividad hacia el producto deseado no se encontró ningún factor significativo.


Abstract The Cu/MCM-41 catalyst was synthesized by incipient wet impregnation and characterized by XRD, XPS, TPD-NH3, and N2 adsorption-desorption. The effect of different reaction conditions (temperature, time, type of solvent, and amount of catalyst) on the isomerization reaction of α-pinene epoxide was evaluated to obtain camphoric aldehyde with the Cu/MCM-41 catalyst, which had not been previously reported for this kind of reaction. The experiment was based on a central composite experimental design composed by response surface analysis. The best campholenic aldehyde selectivity was found to be 85% (5 mg of catalyst, 70 °C, 0.5 h and the use of ethyl acetate as a solvent). Furthermore, the main factors and their combinations that had the greatest significance in the synthesis were adjusted to a polynomial second order model. The factors with the main influence on conversion were the catalyst amount, reaction time, solvent type, and the combined factor between catalyst amount and the solvent type. In the case of campholenic aldehyde selectivity no significant factor was found.


Resumo O catalisador Cu/MCM-41, sintetizado pelo método de impregnação a umidade incipiente, foi caracterizado por DRX, XPS, TPD-NH3 e adsorção-dessorção de N2 e testado na reação de isomerização do epóxido de α-pineno. Diferentes condições de reação foram exploradas a partir de um desenho experimental central composto. A melhor seletividade de aldeído camfolênico encontrada foi de 85% (5 mg de catalisador; 70 °C; 0,5 h e acetato de etilo como solvente). Além disso, os principais fatores e as combinações entre eles que tiveram influência na síntese do aldeído camfolênico foram ajustados a um modelo polinomial de segunda ordem. Verificou-se que os fatores com a principal influência na conversão foram a quantidade de catalisador, o tempo de reação e o tipo de solvente e o fator combinado entre a quantidade de catalisador e o tipo de solvente. No caso da seletividade do aldeído camfolênico, não foi encontrado um fator significativo.

ABSTRACT

RESUMEN La corrosión es un fenómeno que se presenta día a día, no solo en los procesos industriales sino en la propia naturale- za. Debido a que la corrosión tiene graves consecuencias, desde hace mucho tiempo, es considerada un problema de gran magnitud alrededor del mundo. En este trabajo, se aplican métodos estadísticos para el análisis de resultados experimentales. La aplicación del diseño de experimentos de bloques al azar permitió evaluar el comportamiento de la velocidad de corrosión del acero, a partir de software estadís- ticos y modelos matemáticos, teniendo en cuenta la influencia de la variable bloque: tiempo de exposición en la cámara de niebla salina neutra y el comportamiento del grado de oxidación. Se evidenció, que los factores tienen un efecto estadísticamente significativo en la velocidad de Corrosión, al nivel de confianza de 95,0%. El diseño de experimento garantiza minimizar la cantidad de recursos, la posibilidad del estudio de las variaciones de los factores durante el proceso y la selección de una estrategia sobre las decisiones a tomar en el futuro, con vistas a reducir la intensidad del fenómeno.


SUMMARY Corrosion is a phenomenon that occurs every day, not only in industrial processes but also in nature itself. Because corrosion has serious consequences, it has long been considered a problem of great magnitude around the world. Statistical methods are applied in this work for the analysis of experimental results. The application of the randomized block experimental are applied design allowed the evaluation of the behavior of the steel corrosion rate from statistical software and mathematical models, taking into account the influence of the block variable: exposure time in the neutral salt fog chamber and the behavior of the degree of oxidation. It was evidenced that the factors have a statistically significant effect on the corrosion rate at the confidence level of 95.0%. The design of the experiment guarantees to minimize the amount of resources, the possibility of studying the variations of the factors during the process and the selection of a strategy on the decisions to be made in the future, in order to reduce the intensity of the phenomenon.

ABSTRACT

Cataplasma is a new transdermal drug delivery carrier, playing a significant role in the modernization of Chinese materia medica (CMM). It can overcome many shortcomings such as inconvenient use and intense irritation in traditional adhesive plaster and black plaster. However, the development of cataplasma is restricted by some crucial factors like the types of auxiliary materials and the processing ways of volatile medicine. A total of 254 articles related to CMM cataplasma optimization and the volatile oil treatment published from 2000 to 2017 were collected and sorted in this paper. The matrix types and excipients ratio scope, the experiment design method of optimized ratio of base material, and the processing ways of volatile oil in cataplasma were summarized, and then the existing problems in these three aspects mentioned above were discussed with view to giving reference for follow-up studies.

ABSTRACT

Objective: To optimize the purification technology of total flavonoids from Astragali Companati Semen (ACS) by macroporous adsorption resin. Methods: The process parameters of water decoction extraction of ACS and purification of total flavonoids extracted from ACS by D-101 macroporous adsorption resin were optimized by orthogonal experiment design method. Results: The optimal extraction process were as follows: water decoction extract for three times, each time 0.5 h, water addition of 10, 6, and 6 times amount of the medicine respectively. The optimized technology conditions of D-101 macroporous adsorption resin were as follows: The flavonoids concentration of sample liquid of ACS was about 1.0 mg/mL, the diameter ratio was 1∶5, the sample flow rate was 1 BV/h, the rate of sample weight was 0.6 g/mL (herbs/resin); The velocity of water elution was 1 BV/h and its elution volume was 4 BV; The elution concentration, elution velocity, and elution volume were 70%, 1 BV/h, and 5 BV, respectively. The content paste rate of made ACS of total flavonoids was 2.65%, the mass fraction of total flavonoids was 56.24%, and the process transfer rate was 68.37%. Conclusion: The method is simple and feasible for purification of total flavonoids extracted from ACS.

ABSTRACT

Objective:To prepare and optimize the formula of metoclopramide orally disintegrating tablets, and investigate the in vitro drug dissolution behavior. Methods:The formula was optimized by full-factorial experiment design, the ratio of mannotil to micro-crystalline cellulose ( X1 ) and the amount of disintegrating agent ( X2 ,%) were selected as the independent variables, and the friabili-ty ( Y1 ,%) , disintegration time ( Y2 , s) and metoclopramide dissolution ( Y3 ,%) were used as the dependent variables. The release rate of metoclopramide orally disintegrating tablets in different dissolution media was studied. Results:The optimum formula of meto-clopramide orally disintegrating tablets was as follows:the ratio of mannotil to microcrystalline cellulose was 2. 5∶ 1, and the amount of disintegrating agent was 6. 5%. The dissolution of metoclopramide orally disintegrating tablets in the different dissolution media was o-ver 80%. Conclusion:The formula design is reasonable, the preparation process is feasible and the quality can be controlled.

ABSTRACT

Objective:To prepare and optimize candesartan cilexetil tablets,and study the stability preliminarily. Methods:The formula was optimized by Box-Behnken experiment design,the ratio of lactose to pregelatinized starch( X1 ),the amount of disintegrant ( X2 ,%)and the amount of lubricant( X3 ,%)were selected as the independent variables,and weight difference( Y1 ,%),friability ( Y2 ,%),disintegration time( Y3 ,%)and candesartan cilexetil dissolution( Y4 ,%)were the dependent variables. The release rate of candesartan cilexetil tablets and the reference tablets were compared by similarity factor( f2 value). Preliminary stability was studied by high-temperature test,high-humidity test and illumination test. Results:The optimal formula of the tablets was as follows:the ratio of lactose to pregelatinized starch was 7:1,the amount of disintegrant was 5. 5%,and the amount of lubricant was 0. 5%. The f2 for the candesartan cilexetil tablets and the reference tablets in different dissolution meda was 60. 62,73. 34,66. 95 and 68. 60,respec-tively. Conclusion:The formula design is reasonable,the preparation process is feasible and the quality can be controlled.

Objective To investigate optimal extraction process of Piper puberulum ( Benth.) Maxim.and qualitative analyze the chemical component of the extracts.Methods Method of solvent heating reflux was used for extraction.On the basis of single factor experiment, L9 (34 ) orthogonal experiment was designed with the variants of extraction frequency, time, material-liquid ratio, and immersion time.Extraction rate as index, extraction processes were optimized to achieve best extraction.The extracts, including total extract, water elution, and ethanol elution, were physiochemically analysed to achieve an initial qualitative result.Results The optimal extraction process was: extractions 3 times for 2 hours, with an 1︰30 material -liquid ratio and 2 hours of immersion, Initial qualitative analyzed the total extracts containing amino acids, polypeptides, proteins, alkaloids, steroids or triterpenes, flavones, saponins, polysaccharides, reducing sugars or glucosides, cumarins, terpene lactones, phenols, and tannins.The water elution containing: amino acids, polypeptides, proteins, saponins, polysaccharides, reducing sugars or glucosides, cumarins, and terpene lactones.The ethanol elution containing: amino acids, polypeptides, proteins, alkaloids, steroids or triterpenes, flavones, polysaccharides, reducing sugars or glucosides, phenols, and tanins.Conclusion The experiments show that optimal extraction process can achieve high extraction yield, stable and practical.

ABSTRACT

OBJECTIVE: To investigate the relationship between the process parameters and granulation behavior during high shear granulation process. METHODS: Andrographis extract was utilized as a model drug and mixed with microcryslalline cellulose (MCC) or soluble starch at the ratio of 1:1 by weigh, respectively. The process parameters were selected according to fractional factorial experiment. The granules were prepared by using high shear granulation. The aggregation behavior was evaluated by granule yield and ratio of lumps and fine powder. The effect of process parameters on granulation behavior was analyzed through stepwise regression. Standard least squares regression analysis was used to establish the mathematical model of granule yield. RESULTS: The process parameters had different effects on the granulation behaviors of MCC and starch mixtures. The granule yield predicted by the fitting model was (66.66 ± 3.7)%, and the measured values for MCC and starch mixture were 68.9% and 66.1%, respectively. CONCULSION: The established mathematical model is useful in predicting the performance of materials duing high shear granulation process. Understanding the impact of process parameters on granulation behavior provides the basis of modulating granulation parameters in order to optimize product performance and improve the yield.

ABSTRACT

Objective To design and verify an analytical method for the cross contamination of Beckman AU5800 automatic bio‐chemical analyzer′s reagent needle and stir bar .Methods The outer ring items (AST ,ALP ,TP ,LDH ,CK‐MB ,HBDH ,Urea ,UA , TG ,LDL‐C ,APOB ,Mg ,P ,Zn ,AMY ,CHE ,LPS ,ADA) were tested as the experiment objects .The cross contamination experiment was proceeded according to the test order .The impact rate range of 95% -105% was the criteria for cross contamination of items . Results Among all of the outer ring items ,TP ,HBDH ,APOB and TG were found to have cross contamination effect on CK‐MB , Mg ,Mg and LPS respectively .Conclusion The method established in this experiment can effectively analyze the cross contamina‐tion of all the items in Beckman AU5800 automatic biochemical analyzer .In addition ,it can help to adopt corresponding measures to reduce cross contamination and assure the accuracy of test results .

ABSTRACT

Experiment design has been added to the experiment class for pharmacological course.The topics of the experiment design were prepared by the teacher based on the contents of theory class.After finishing the experiment design with the help of literatures,the students introduced their proposal in the class,and then discussed with each other.The results of feedback showhed that most of the students think they have learned how to look up the literature and prepare the multimedia teaching materials in the course of completing the proposal.In addition,it also enhanced the spirit of team work,improved the students'ability of expression,aroused the students'awareness and interest for research as well as promoted the students'understanding for pharmacological theory.

ABSTRACT

Objective:To study the extraction method of total flavonoids in Mosla hangzhowensis Matsuda.Methods:Orthogonal experiment design was used,with the content of the total flavonoids as the index,to study the effect of ethanol concentration,solvent quantity and extraction time on recovery rate of total flavonoids.Results:The best extraction condition was:extract two times;adding 20 fold 65% ethanol and to extract an hour each time.Conclusion:This extraction method is convenient and feasible.

Objective To achieve millisecond grade precision in the design model of event-related fMRI experiment.Methods The impact factors of time precision were analyzed,and solutions were put forward.Results The parameter of E-Prime(a software dedicated to psychology study) was adjusted to get millisecond precision.Conclusion General computer is very difficult to achieve millisecond grade due to software and hardware factors,but it can be achieved through adjusting the parameter of E-Prime.

OBJECTIVE: To prepare double-layer sustained release ornidazole films and investigate its release rate in vitro.METHODS: The double-layer sustained release ornidazole films film were prepared by the method of homogenate.The formula of the films were optimized by orthogonal experiment with the mass ratio of polyviol to CMC-Na(factor A),the total amount of polyviol and CMC-Na(B) and the amount of glycerol(C) as factors.The concentration of ornidazole was determined by ultraviolet spectrophotometry and its release rate was also computed.RESULTS: The double-layer sustained release film was well-shaped.The optimized formula of the film was as follows: factor A=1∶1,B=4.5 g,C=4.0 g.The standard curve of ornidazole was linear within a range of 1~20 ?g?mL-1(r=0.999 5,n=6),with an average recovery of 100.3%(RSD=1.3%,n=5).A continuous releasing of 11.0 hours was achieved for the prepared films and its cumulative release amount in vitro within 8 hours reached 81.50%.CONCLUSIONS: The preparative method of the films was simple,and the film had a slow-release property.

ABSTRACT

AIM: To investigate and optimize the technology of ultrasonic-assisted extraction of arctiin and arctigenin from Fructus arctii.METHODS: The effects of ultrasonic power,particle size,ratio of liquid to solid,extraction time,extraction temperature and duty factor on the extraction rates of arctiin and arctigenin were investigated,and optimized with orthogonal experiments.RESULTS: The results showed that the optimal extraction conditions were as follows: particle 80 ~ 100 mesh 14 mL/g,ratio of liquid to solid 400 W ultrasonic power,50 ℃ of extraction temperature,and 10 min of extraction time.CONCLUSION: The optimized conditions are reasonable and reliable.
