ABSTRACT

According to literature, certain microorganism productions mediate biological effects. However, their beneficial characteristics remain unclear. Nowadays, scientists concentrate on obtaining natural materials from live creatures as new sources to produce innovative smart biomaterials for increasing tissue reconstruction in tissue engineering and regenerative medicine. The present review aims to introduce microorganism-derived biological macromolecules, such as pullulan, alginate, dextran, curdlan, and hyaluronic acid, and their available sources for tissue engineering. Growing evidence indicates that these materials can be used as biological material in scaffolds to enhance regeneration in damaged tissues and contribute to cosmetic and dermatological applications. These natural-based materials are attractive in pharmaceutical, regenerative medicine, and biomedical applications. This study provides a detailed overview of natural-based biomaterials, their chemical and physical properties, and new directions for future research and therapeutic applications.

ABSTRACT

In last few years, therapeutic peptides/proteins are rapidly growing in drug market considering their higher efficiency and lower toxicity than chemical drugs. However, the administration of therapeutic peptides/proteins is mainly limited in parenteral approach. Oral therapy which was hampered by harsh gastrointestinal environment and poorly penetrating epithelial barriers often results in low bioavailability (less than 1%-2%). Therefore, delivery systems that are rationally designed to overcome these challenges in gastrointestinal tract and ameliorate the oral bioavailability of therapeutic peptides/proteins are seriously promising. In this review, we summarized various multifunctional delivery systems, including lipid-based particles, polysaccharide-based particles, inorganic particles, and synthetic multifunctional particles that achieved effective oral delivery of therapeutic peptides/proteins.

ABSTRACT

This study was performed to examine the effects of various macromolecules in in vitro growth (IVG) media on the growth, maturation, and parthenogenesis (PA) of pig oocytes derived from small antral follicles (SAF). Immature oocytes were cultured for two days in IVG medium supplemented with 10% (v/v) fetal bovine serum (FBS), 10% (v/v) pig follicular fluid (PFF), 0.4% (w/v) bovine serum albumin (BSA), or 0.1% (w/v) polyvinyl alcohol (PVA) and then maintained for 44 h for maturation. After IVG, the mean diameters of the SAF treated with FBS, PVA, and no IVG-MAF (113.0–114.8 µm) were significantly larger than that of no IVG-SAF (111.8 µm). The proportion of metaphase II oocytes was higher in PFF (73.6%) than in BSA (43.5%) and PVA (53.7%) but similar to that in the FBS treatment (61.5%). FBS and PFF increased cumulus expansion significantly compared to PVA and BSA while the intraoocyte glutathione content was not influenced by the macromolecules. Blastocyst formation of PA oocytes treated with FBS (51.8%), PFF (50.4%), and PVA (45.2%) was significantly higher than that of the BSA-treated oocytes (20.6%). These results show that the PFF and FBS treatments during IVG improved the growth, maturation, and embryonic development of SAF.

ABSTRACT

In order to adapt to the rapid development of experimental techniques in the field of cell molecular biology, to quickly and effectively cultivate the experimental skills of postgraduates, systematic reforms should be carried out on advanced cellular molecular biology skill experiment teaching. The combi-nation of basic theory, biological experiment and practical application enables students to master the basic techniques from drawing materials to molecular biology analysis and construct in vitro transgenic cell model. Adding clinical case analysis, simulated ethical review and other lively classroom activities, which can help students carry out scientific research as soon as possible, improve students' interest and cultivate students' teamwork ability. The teaching effect is good and worth promoting.

ABSTRACT

Cordyceps is a famous traditional Chinese medicine (TCM) that has been used in China for hundreds of years. In the present study a multi-column liquid chromatography (MC-LC) system was developed for the qualitative analysis of macromolecules and micromolecules in Cordyceps. The MC-LC system includes a size exclusion pre-column, a size exclusion column (SEC) and a reversed phase column (RP) which were controlled by column-switching valves. The sample was separated by the size exclusion pre-column into two fractions (macromolecules and micromolecules). These fractions were further separated on SEC and RP columns, respectively. A diode array detector (DAD) and a mass spectrometer (MS) were used to detect the components. This MC-LC method was utilized for analysis of Cordyceps samples. Two macromolecular peaks and 15 micromolecular peaks were found in Cordyceps, and 11 of the micromolecular peaks were identified as adenosine-5'-monophosphate (AMP), phenylalanine, uridine, hypoxanthine, inosine, guanine, guanosine, deoxyadenosine-5'-monophosphate (dAMP), adenosine, adenine and cordycepin (or its isomer). This method is useful for quality control of Cordyceps.

Background More than 200 Scyphozoa species have been described, but few have been properly studied regarding their chemical and genetic characteristics.Catostylus tagi, an edible Scyphozoa and the sole European Catostylidae, occurs in summer at Tagus and Sado estuaries. Neither a systematic comparison between the two Catostyluscommunities nor a chemical approach on their nematocytes had been carried out yet.Methods In order to achieve these purposes, optimisation of DNA extraction and of histochemical staining procedures were developed.Catostylus specimens from Tagus and Sado estuaries were compared by ribosomal 18S, 28S, and ITS1 partial sequencing. The morphochemistry of nematocytes was studied by optical and electronic microscopy.Results Macroscopic and molecular results indicated that both communities belong to the same species, C. tagi. The hematoxylin and eosin staining allowed the visualisation of nematocyst genesis and indicated a basic character for the macromolecules on the shaft of euryteles and on the tubule of isorhizae and birhopaloids. By Massons trichrome procedure, the basic properties of the tubules were confirmed and a collagenous profile for the toxins was suggested. Results of the alcian blue staining showed that the outer membrane of nematocyte may consist of macromolecules with acidic polysaccharides, consistent with NOWA and nematogalectin glycoproteins detected in Hydra, but also with poly-gamma-glutamate complex, chitin-like polysaccharides and hyaluronic acids. Through the von Kossa assays, calcium was detected; its position suggested interactions with polysaccharides of the membrane, with proteins of the contractile system or with both.Conclusions The optimisation of sample preparation for DNA extraction may facilitate further studies on little known jellyfish species. The improvement of the smear procedure simplified the use of stained reactions in zooplankton. Moreover, it was shown that good slide images might be acquired manually. The development of specific reactions, with traditional dyes and others, can give important contributions to clarify the chemical nature of the components of nematocytes. The characterisation of nematocyst toxins by staining tests is a goal to achieve.

ABSTRACT

Extratos aquosos da planta medicinal Achillea millefolium contêm macromoléculas de interesse para desenvolver fitodefensivos para a agricultura. Duas frações de mil folhas foram obtidas por ultrafiltração, E1 (contendo moléculas maiores que 30 kDa), e E3 (peptídeos entre 1 e 10 kDa) que inibiram o crescimento das bactérias fitopatogênicas Ralstonia solanacearum, gram-negativa, e Clavibacter michiganensis subsp. michiganensis, gram-positiva, com dependência de concentração. Os valores de concentração inibitória mínima (CIM) para ambos os extratos e bactérias foram baixos, entre 20 e 80µM. A CIM relativa à proteína total evidenciou a presença de macromoléculas muito ativas em E3, embora com baixa concentração proteica. E3 se aplica à prospecção de peptídeos antimicrobianos. Estimar a CIM relativa à quantidade de amostra vegetal valorizou o potencial antimicrobiano natural de E1, que contém alta concentração proteica. E1e E3 se aplicam ao desenvolvimento de fitodefensivos para uso biotecnológico. A ultrafiltração fracionou as amostras de forma nativa, rápida, e com baixo custo; além de dessalinizar, clarificar, purificar, e concentrar E1 e E3. Esse estudo inédito sobre a separômica e a ação antimicrobiana de extratos macromoleculares aquosos de mil folhas sugere que plantas cicatrizantes podem apresentar grande potencial para desenvolver fitodefensivos agrícolas naturais não danosos, à semelhança de medicamentos fitoterápicos.


Aqueous extracts from the medicinal plant Achillea millefolium contain macromolecules of interest to develop agrochemicals for agriculture. Two fractions of "mil folhas" were obtained by ultrafiltration, E1 (containing molecules larger than 30 kDa) and E3 (peptides between 1 and 10 kDa), which inhibited the growth of phytopathogenic bacteria Ralstonia solanacearum, gram-negative, and Clavibacter michiganensis subsp. michiganensis, gram-positive, concentration-dependent. The values of minimum inhibitory concentration (MIC) for both extracts and both bacteria were low, ranging from 20 to 80µM. The MIC relative to total protein evidenced the presence of very active macromolecules in E3, although showing low protein concentration. E3 applies to the prospection of antimicrobial peptides. The estimated MIC relative to the amount of plant sample valued the natural antimicrobial potential of E1, which contains high protein concentration. E1 and E3 can be used in the development of agrochemicals for biotechnological purposes. The ultrafiltration procedure fractionated the samples in a rapid and native way and at a low cost; it also desalted, clarified, concentrated and purified both E1 and E3. This pioneering study on the separomics and on the antimicrobial activity of macromolecular aqueous extracts from "mil folhas" suggests that healing plants have great potential to develop non-harmful agricultural natural agrochemicals, similarly to the available phytotherapic drugs.

ABSTRACT

En este trabajo se reporta la síntesis de terpiridinas bajo la metodología de Kröhnke. Las terpiridinas obtenidas por este método, se analizaron por espectroscopia UV-Vis y de fluorescencia, y su respuesta a la presencia de varios metales en diferentes concentraciones fue evaluada para sistemas en solución acuosa. Los resultados muestran que la terpiridina TpyOH es altamente promisoria en la detección de mercurio bajo las condiciones experimentales reportadas.


This paper reports the synthesis of terpyridines using the Kröhnke methodology. The terpyridines obtained by this method were analyzed by UV-Vis spectroscopy and fluorescence, and its response to the presence of various metals at different concentrations was evaluated in aqueous systems. The results show that the terpyridine TpyOH is highly promising for the detection of mercury under the experimental conditions reported.


Este trabalho relata a síntese de terpyridines baixo a metodologia de Kröhnke. Os terpyridines obtidos por este método foram analisadas por espectroscopia UV- Vis e de fluorescência, e a sua resposta à presença de vários metais em diferentes concentrações foi avaliada em sistemas aquosos. Os resultados mostram que o terpyridine TpyOH é altamente promissor para a detecção de mercúrio sob as condições experimentais relatados.

The aim of this study was to determine the yield, chemical composition, specific rotation (SR), infrared (IR) spectroscopy and the effect on bacterial growth of a crude sulfated polysaccharide (SP) from the red marine alga G. ornata (Go). Go-1 (25°C), Go-2 (80°C), and Go-3 (80°C) were sequentially extracted and yielded 9.2%. The contents of sulfate (5.88-10.3%) and proteins (0.1-3.7%) were small. The values of SR were [µ]D20°f -19.0, -51.0, and -56.5, respectively. IR spectrums showed the presence of galactose-4 sulfate and absence of 3,6-anydrogalactose-2 sulfate, galactose-6 sulfate and galactose-2 sulfate. SR and IR techniques confirmed SPs. Go-3 was tested on the growth of bacteria (Bacillus subtilis, Staphylococcus aureus, Enterobacter aerogens, Escherichia coli, Pseudomonas aeruginosa, Salmonela choleraesuis and Salmonela typhi), but only E. coli was inhibited.

Sulfated polysaccharides (SPs) have attracted growing interest for various biotechnological applications. We evaluated the efficiency of two methods of drying SPs (M I and II) extracted from Halymenia sp Rhodophyceae in order to compare the yield, purification and anticoagulant activity. The total SPs (TSPs) were first extracted with papain in 100 mM sodium acetate (pH 5.0) containing cysteine and EDTA (5 mM). The TSPs obtained were dried in an oven (M I) or lyophilized (M II) and then examined by ion exchange chromatography (DEAE-cellulose) using the NaCl gradient technique. The fractions were analyzed by 0.5% agarose gel electrophoresis and the in vitro anticoagulant activity was evaluated by the activated partial thromboplastin time test using normal human plasma and compared to heparin (HEP) (193.00 IU mg-1). There was a difference in TSP yield of 19.05% and similar chromatographic SP profiles. Electrophoresis revealed fractions with distinct resolutions. The fractions eluted with 0.75 M of salt (M I and II) were the most active, measuring 27.40 and 72.66 IU mg-1, respectively, when compared to HEP. Therefore, obtaining SP with anticoagulant activity from Halymenia sp. is more efficient by freeze-drying.


Os polissacarídeos sulfatados (PS) têm despertado interesse crescente para diversas aplicações em biotecnologia. Avaliou-se a eficiência de duas metodologias de secagem de PSs (M I e II) extraídos da rodofícea Halymenia sp. a fim de se comparar o rendimento, a purificação e a atividade anticoagulante. Inicialmente, os PS totais (PSTs) foram extraídos com papaína em tampão acetato de sódio 100 mM (pH 5,0) contendo cisteína e EDTA (5 mM). Os PSTs foram secados em estufa (M I) ou liofilizados (M II), seguido por cromatografia de troca iônica (DEAE-celulose) utilizando um gradiente de NaCl. As frações foram analisadas por eletroforese em gel de agarose a 0,5% e a atividade anticoagulante "in vitro" avaliada por meio do teste do tempo de tromboplastina parcial ativada usando plasma humano normal e comparadas à heparina (HEP) (193.00 UI mg-1). Verificaram-se diferença no rendimento de PSTs (19,05%) e semelhantes perfis cromatográficos de PS, revelando, por eletroforese, frações de PS distintas em grau de resolução entre os métodos. As frações eluídas com 0,75 M de sal (M I e II) foram as mais ativas, com valores de atividade da ordem de 27,40 e 72,66 UI mg-1, respectivamente, em relação à HEP. Portanto, a secagem por liofilização seria uma forma mais eficaz na obtenção de PSs com atividade anticoagulante de Halymenia sp.

ABSTRACT

The aim of this study was to evaluate certain molecular characteristics of a sulfated polysaccharide (SPs) with anticoagulant properties, isolated from Caulerpa cupressoides (Chlorophyta). Crude SPs were extracted by proteolytic digestion (papain), followed by ion-exchange chromatography on a DEAE-cellulose column. The fractions obtained were analyzed for molecular mass, 0.5 percent agarose gel electrophoresis and chemical composition. The activated partial thromboplastin time (APTT) test was applied using normal human plasma and standard heparin (HEP) (193 IU mg-1). The yield was ~ 3 percent, and the chromatography procedure separated the material into three different SP fractions (F I, F II and F III), eluted at the concentrations of 0.50, 0.75 and 1.00 M of NaCl, respectively. Only fraction F II was active (24.62 IU mg-1), with high sulfate content (23.79 percent) and number of molecular mass peaks. Therefore, the APTT of a fraction isolated from C. cupressoides was less potent than HEP.

A incidência de doenças cardiovasculares e os efeitos adversos da heparinoterapiatêm motivado a busca por novos agentes terapêuticos e os polissacarídeos sulfatados (PS) de algasmarinhas têm sido reportados como fontes alternativas para tal. Objetivou-se avaliar o potencialanticoagulante dos PS totais (PST) isolados e fracionados das clorofíceas Caulerpa racemosa eCaulerpa cupressoides. Inicialmente, os PST foram extraídos com papaína em tampão acetato desódio 0,1 M (pH 5,0) contendo cisteína 5 mM e EDTA 5 mM, seguidos de fracionamento emcoluna de troca iônica de DEAE-celulose com gradiente de NaCl. As frações obtidas foramanalisadas por eletroforese em gel de agarose a 0,5% e a atividade anticoagulante, mensurada pelotempo de tromboplastina parcial ativada (TTPA), usando-se plasma humano normal ecomparada a uma curva-padrão de heparina (193 UI mg-1). Verificaram-se semelhantes perfiscromatográficos entre os PS de ambas as espécies, porém com padrões de mobilidades distintasquando as frações foram comparadas por eletroforese. Os PS modificaram o TTPA, cujasatividades anticoagulantes foram de apenas 21,23 e 24,36 UI mg-1, quando eluídos com 0,75 M desal para C. racemosa e C. cupressoides, respectivamente. Portanto, PS anticoagulantes isolados dasclorofíceas C. racemosa e C. cupressoides resultaram em efeitos anticoagulantes inferiores aos daheparina. Estudos comparativos dessas moléculas também são sugeridos como ferramentasauxiliares na identificação de algas do mesmo gênero.

The aim of this work was to evaluate the anticoagulant potentialof total SP (TSP) isolated and fractionated from Caulerpa racemosa and Caulerpa cupressoides(Chlorophyceaes). Initially, the TSP were extracted with papain in 0.1 M sodium acetate buffer(pH 5.0) containing 5 mM cysteine and 5 mM EDTA, followed by fractionation on ionexchange DEAE-cellulose column with NaCl gradient. The obtained fractions were analyzed by0.5% agarose gel electrophoresis and the anticoagulant activity measured by the activated partialthromboplastin time (APTT) using normal human plasma, and compared to a standard heparincurve (193 IU mg-1). Similar chromatographic profiles of SP were shown on both species, butwith distinct mobility patterns, when the SP fractions were compared by electrophoresis. SPeluted with 0.75 M of NaCl modified the APTT, whose anticoagulant activities were only 21.23and 24.36 IU mg-1 for C. racemosa and C. cupressoides, respectively. Therefore, anticoagulant SPisolated from chlorophyceaes showed effects inferior to heparin, and comparative studies of thesemolecules are also suggested as auxiliary tools in the identification of algae of the same genus.

ABSTRACT

The chosen phosphobacterial species are well tolerated to the wide range of salinity (20-40 ppt) and found to be pure marine forms. Effect of different concentrations of cadmium on the growth, phosphate solubilising activity and content of extracellular macromolecules in eight species of phosphate solubilising bacteria were carried-out in the present study at optimum level of salinity and found that these activities are found maximum at lower concentrations of cadmium and further reduction in the activities were noticed at higher concentrations. The maximum content of total sugar was noticed in S. aureus at 1 ppm level of cadmium. The content of protein was found maximum in B. megaterium and M. roseus without the addition of cadmium and also the phosphate solubilizing activity was found higher in all the chosen phosphobacterial strains with the addition of cadmium.

ABSTRACT

Mitochondria isolated from the gill tissues of the fish Sarothredon mossambica were analysed for their macromolecular content, following transfer from freshwater to media of higher salinity. The results suggest a breakdown of mitochondria during the initial phases of the stress and a regeneration during continued exposure. Also all the synthetic machineries, in general, seem to be triggered in gill tissue during continuous exposure to hyperosmotic media.
