ABSTRACT

Myostatin gene (MSTN) encodes a negative regulator for controlling skeletal muscle growth in animals. In this study, MSTN-/- homozygous mutants with "double muscle" phenotypic traits and stable inheritance were bred on the basis of MSTN gene editing rabbits, with the aim to establish a method for breeding homozygous progeny from primary MSTN biallelic mutant rabbits. MSTN-/- primary mutant rabbits were generated by CRISPR/Cas9 gene editing technology. The primary mutant rabbits were mated with wild type rabbits to produce F1 rabbits, whereas the F2 generation homozygous rabbits were bred by half-sibling mating or backcrossing with F1 generation rabbits of the same mutant strain. Sequence analysis of PCR products and its T vector cloning were used to screen homozygous rabbits. The MSTN mutant rabbits with 14-19 week-old were weighed and the difference of gluteus maximus tissue sections and muscle fiber cross-sectional area were calculated and analyzed. Five primary rabbits with MSTN gene mutation were obtained, among which three were used for homozygous breeding. A total of 15 homozygous rabbits (5 types of mutants) were obtained (M2-a: 3; M2-b: 2; M3-a: 2; M7-a: 6; M7-b: 2). The body weight of MSTN-/- homozygous mutant rabbits aged 14-19 weeks were significantly higher than that of MSTN+/+ wild-type rabbits of the same age ((2 718±120) g vs. (1 969±53) g, P < 0.01, a 38.0% increase). The mean cross sections of gluteus maximus muscle fiber in homozygous mutant rabbits were not only significantly higher than that of wild type rabbits ((3 512.2±439.2) μm2 vs. (1 274.8±327.3) μm2, P < 0.01), but also significantly higher than that of MSTN+/- hemizygous rabbits ((3 512.2±439.2) μm2 vs. (2 610.4±604.4) μm2, P < 0.05). In summary, five homozygous mutants rabbits of MSTN-/- gene were successfully bred, which showed a clear lean phenotype. The results showed that the primary breeds were non-chimeric mutant rabbits, and the mutant traits could be inherited from the offspring. MSTN-/- homozygous mutant rabbits of F2 generation could be obtained from F1 hemizygous rabbits by inbreeding or backcrossing. The progenies of the primary biallelic mutant rabbits were separated into two single-allelic mutants, both of which showed a "double-muscle" phenotype. Thus, this study has made progress in breeding high-quality livestock breeds with gene editing technology.

ABSTRACT

OBJECTIVE:To study the toxic effect of the extract from Tripterygium hypoglaucum on pregnant animal and embryo-fetal development . METHODS :Successfully mated New Zealand female rabbits were randomly divided into solvent control group,positive control group (cyclophosphamide,20 mg/kg)and T. hypoglaucum extract high-dose ,medium-dose and low- dose groups(15.0,7.50,3.75 g/kg,by the amount of crude drug )according to their body weight ,with 18 rabbits in each group at least. The female rabbits in solvent control group and T. hypoglaucum extract groups were given water or the corresponding T. hypoglaucum extract solution from 6th to 18th day of pregnancy ,5 mL/kg,once a day. Positive control group was given cyclophosphamide subcutaneously into the neck from 10th to 13th day of pregnancy ,1 mL/kg,once a day. According to the related requirements of Technical Guidelines for the Study of Drug Reproductive Toxicity ,the general situation ,body weight ,body weight increase and food intake of female rabbits were observed and recorded during the experiment ,and euthanasia was carried out on 28th day of pregnancy ;the relative indexes of main organs ,fetal uterus ,placenta uterus ,placenta,ovary,implantation gland , absorption fetus ,stillbirth,live fetus and corpus luteum were observed and recorded after anatomy ;the relative indexes of body weight,appearance,visceral and skeletal alterations of the fetus were detected . RESULTS :Compared with solvent control group , the body weight ,body weight increase ,food intake ,main organs ,pregnancy of pregnant rabbits ,as well as reproductive function,embryo formation ,fetal growth and development ,appearance,visceral and skeletal development indexes in T. hypoglaucum extract groups had no significant abnormal changes (P>0.05);above indexes in the positive control group had significant changes (P<0.05),and significant maternal toxicity and embryotoxicity were found. CONCLUSIONS :T. hypoglaucum extract 15.00-3.75 g/kg(by the amount of crude drug )have no significant maternal toxicity ,embryotoxicity or fetal development toxicity to New Zealand rabbits.

ABSTRACT

The objective of this study was to evaluate the effect of sweet potato vines as a source of fiber on the plasma parameters in healthy rabbits. For this, 15 rabbits were allocated into five groups, and each group was fed a different diet for a period of 46 days. The diets were as follows: diet without sweet potato vines (0SPV), and experimental diets with 25%, 50%, 75% and 100% of sweet potato vines in replacement of alfalfa hay respectively (25SPV, 50SPV, 75SPV and 100SPV). Triglycerides and VLDL-cholesterol concentrations were approximately 53% lower in rabbits fed the 100SPV than in rabbits fed 0SPV. In addition, aspartate aminotransferase (AST) and total-cholesterol concentrations were lower approximately 40% and 14%, respectively, in rabbits fed the 100SPV than in rabbits fed 0SPV. No significant differences were found among HDL-cholesterol, glucose, proteins, albumin and alanine aminotransferase (ALT) concentrations of the five groups. These results demonstrate that the consumption of sweet potato vines reduces the triglycerides, VLDLcholesterol and total cholesterol while maintains HDL-cholesterol levels. Therefore, sweet potato vines consumption may be another option to prevent coronary heart diseases in rabbits(AU)


O objetivo deste estudo foi avaliar com base no plasma sanguíneo o efeito do baraço de batata-doce como fonte de fibra. Para tal, dividiuse 15 coelhos em cinco grupos, onde cada grupo foi alimentado com uma dieta diferente por 46 dias. As dietas foram: sem baraço de batata-doce (0SPV), e dietas experimentais com 25%, 50%, 75% e 100% de baraço de batata-doce em substituição ao feno de alfafa, respectivamente (25SPV, 50SPV, 75SPV e 100SPV). As concentrações de triglicérides e VLDL-colesterol foram aproximadamente 53% menores em coelhos alimentados com a dieta 100SPV do que os coelhos alimentados com 0SPV. Além disso, as concentrações de aspartato aminotransferase (AST) e colesterol total foram inferiores em aproximadamente 40% e 14%, respectivamente, nos coelhos alimentados com 100 SPV do que em coelhos alimentados com 0SPV. Não foram encontradas diferenças significativas entre as concentrações colesterol-HDL, glicose, proteínas, albumina e alanina aminotransferase (ALT) dos cinco grupos. Estes resultados demonstram que o consumo de baraço de batata-doce reduz os triglicerídeos, colesterol VLDL e colesterol total, enquanto mantém os níveis de colesterol HDL. Portanto, o baraço de batata-doce pode ser uma opção viável para prevenir doenças coronarianas(AU)

ABSTRACT

OBJECTIVE@#This article investigated the changes of some biochemical markers and cardiac function in chronic heart failure (CHF), and provided the basis for the diagnosis of CHF.@*METHODS@#New Zealand rabbit CHF model was established using adriamycin (ADR). Twenty New Zealand rabbits were randomly divided into model group (=15) and control group (=5), injected with ADR and saline solution the ear vein respectively, 2 times a week, lasting for 8 weeks. After that, myocardial enzymes, carotid artery pressure, echocardiogram (ECG) and phonocardiogram (PCG) of all New Zealand rabbits were detected and recorded.@*RESULTS@#Compared with control group, all parameters of the model group were changed significantly (<0.05).@*CONCLUSIONS@#CHF leads to myocardial damage in New Zealand rabbits, decreased systolic and diastolic function, cardiac reserve index can be used to assess cardiac function.

ABSTRACT

Objective To observe the effects of road transport on hematological and biochemical parameters in New Zealand rabbits.Methods A total of 12 healthy New Zealand rabbits were selected for 2 h road transport.Blood samples were collected at 0, 24, 48, 72 and 96 h after transport, respectively.White blood cells (WBC), red blood cells (RBC), hemoglobin (HGB), hematocrit (MCV), mean erythrocyte hemoglobin content (MCH), mean erythrocyte hemoglobin concentration (MCHC) and platelets (PLT) were measured using a blood analyzer.Blood alanine aminotransferase (ALT), aspartate aminotransferase (AST), albumin (ALB), total protein (TP), urea nitrogen (UREA), creatinine, uric acid (UA), triglycerides (TG), total cholesterol (COHL), glucose (GLU), hypersensitive C-reactive protein (CRP), α-amylase (AMYL), and creatine kinase (CK) were detected by an automatic biochemical analyzer.Results Compared the parameters before and after transport, The WBC count was increased first (P< 0.05 or P < 0.01) and then decreased after transport, the levels of RBC, HGB, HCT and PLT were decreased first (P< 0.05 or P < 0.01) and then increased after transport, and MCV was significantly high at 96 h after transport (P< 0.05).Among the clinical biochemical parameters, ALT, AST and BUN were firstly elevated (P< 0.05 or P < 0.01) and then decreased.TP, ALB as well as CREA and TG were firstly decreased (P< 0.05 or P < 0.01) and then increased.GLU was significantly low at 24 h after transport (P< 0.05).All parameters except MCV at 96 h after transport were not significantly different from those before transport.Conclusions Changes of blood routine, liver and kidney function indexes, lipid metabolism indexes, glucose metabolism index and creatine kinase index are observed in the New Zealand rabbits after 2-hour road transportation, and all the indicators except MCV return to pre-transport levels within 96 h.

ABSTRACT

The origin and distribution of the femoral nerves in both antimeres were studied in 30 New Zealand rabbits (15 males and 15 females). The specimens were collected after natural death and fixed with 10% formaldehyde solution. In males, the femoral nerve originated from the ventral branches of the fourth, sixth and seventh lumbar spinal nerves in seven animals (46.67%), in two animals (13.33%) from the ventral branches of the fifth and sixth lumbar spinal nerves, in one animal (6.67%) from the ventral branch of the fifth lumbar spinal nerve, in three animals (20%) from the ventral branches of the fifth, sixth and seventh lumbar spinal nerves, in one animal (6.67%) from the ventral branches of the fourth and fifth lumbar spinal nerves, and in one animal (6.67%) from the ventral branches of the fifth and seventh lumbar spinal nerves. In females, the femoral nerve originated from the ventral branches of the fourth, sixth and seventh lumbar spinal nerves in four animals (26.67%), in two cases (13.33%) from the ventral branches of the fifth and sixth lumbar spinal nerves, in one animal (6.67%) from the ventral branch of the fifth lumbar spinal nerve, in three animals (20%) from the ventral branches of the sixth and seventh lumbar spinal nerves, in four animals (26.67%) from the ventral branches of the fifth, sixth and seventh lumbar spinal nerves and in one animal (6.67%) from the ventral branches of the fourth and seventhlumbar spinal nerves. In all animals the femoral nerves were distributed in different branches to the major and minor psoas, femoral quadriceps, sartorius and pectinius muscles.


Se estudió el origen y distribución del nervio femoral de ambos antímeros en 30 conejos neozelandeses, 15 machos y 15 hembras. Los animales fueron recolectados después de su muerte natural y se fijaron en formaldehído al 10%. En los machos, el nervio femoral se originó a partir de los ramos ventrales del cuarto, sexto y séptimo nervios espinales lumbares en siete casos (46,67%); en tres casos (20%) desde los ramos ventrales del quinto, sexto y séptimo nervios espinales lumbares; en dos casos (13,33%) desde los ramos ventrales del quinto y sexto nervios espinales lumbares, mientras que en tres animales (n=1 respectivamente), desde los ramos ventrales del quinto nervio espinal lumbar (6,67%), los ramos ventrales del cuarto y quinto nervios lumbares espinales (6,67%) y desde los ramos ventrales del quinto y séptimo nervios espinales lumbares. En las hembras, el nervio femoral se originó a partir de los ramos ventrales del cuarto, sexto y séptimo nervios espinales en cuatro casos (26,67%); en otros cuatro casos (26,67%) desde los ramos ventrales del quinto, sexto y séptimo nervios espinales lumbar, en tres casos (20%) desde los ramos ventrales del sexto y séptimo nervios espinales lumbares, en dos casos (13,33%) desde los ramos ventrales del quinto y sexto nervios espinales, y en dos animales (n=1, respectivamente) procedían desde los ramos ventrales del quinto nervio espinal lumbar (6,67%) y de los ramos ventrales del cuarto y séptimo nervios espinales lumbares (6,67%). Los nervios femorales en todos los animales estaban distribuidos en diversos ramos de los músculos psoas mayor y menor, cuádriceps femoral sartorios y pectinatos.

ABSTRACT

The purpose of this study was to establish reference values for selected ophthalmic diagnostic tests in New Zealand rabbits (Oryctolagus cuniculus). A total of 22 adult male rabbits were used. The ophthalmic tests included evaluation of tear production with Schirmer tear test 1(STT1) and Endodontic absorbent paper point tear test (EAPPTT) using two different commercial brand materials. Applanation tonometry, Culture of the conjunctival bacterial flora, , conjunctival cytology and conjunctival histology were also performed. Mean (±SD) for STT1, EAPPTTa, EAPPTTb and IOP was 7.27±2.51mm/min, 12.43±1.69mm/min, 15.24±2.07mm/min, 12.89±2.80mm Hg, respectively. Staphylococcus epidermidis, Staphylococcus sp. and Bacillus sp. were predominant. The cytological evaluation revealed the presence columnar epithelial cells, superficial squamous keratinized cells, lymphocytes, heterophils, red blood cells, mucus and bacteria. The histological analysis revealed a stratified epithelium, characterized by the presence of columnar epithelial cells with a large number of goblet cells. The reported data can be used for therapeutic or experimental purposes.


O objetivo deste estudo foi estabelecer valores de referência para testes diagnósticos oftálmicos em coelhos da raça Nova Zelândia (Oryctolagus cuniculus). 22 coelhos, machos, adultos foram utilizados. Foi mensurada a produção lacrimal através do teste lacrimal de Shirmer 1 (TLS1) e da Tira endodôntica de papel absorvente (EAPPTT) de duas marcas comerciais distintas. Tonometria de aplanação, identificação da microbiota conjuntival, , citologia e histologia conjuntival também foram realizadas. A média e desvio padrão do TLS1, EAPPTT1, EAPPTT2 e pressão intraocular foi 7,27±2,51 mm/min, 12,43±1,69 mm/min, 15,24±2,07 mm/min e 12,89±2,80 mmHg, respectivamente. Staphylococcus epidermidis, Staphylococcus sp. e Bacillus sp. mostraram-se predominantes. A citologia conjuntival evidenciou a presença de células epiteliais colunares, células escamosas superficiais queratinizadas, linfócitos, heterofilos, células sanguíneas, muco e bactérias. A histologia revelou epitélio estratificado caracterizado pela presença de células epiteliais colunares com grande número de células caliciformes. Os achados deste estudo poderão ser utilizados com fins terapêuticos ou experimentais.

ABSTRACT

O objetivo do estudo foi realizar uma avaliação histomorfométrica dos tecidos formados nos alvéolos de extração de coelhos, preenchidos com dois diferentes tipos de biomateriais, em diferentes períodos de cicatrização. Material e Métodos: foram utlizados 14 coelhos da espécie Nova Zelândia, onde foram extraídos os dois molares do mesmo lado da mandíbula de cada animal. Um alvéolo foi preenchido com osso mineral bovino (Bio-Oss, grupo A) e o outro com uma hidroxiapatita sintética (Alobone poros, grupo B). Após três meses, sete animais foram sacrificados e os outros após seis meses. Em seguida, os espécimes foram preparados para análise histomorfométrica. Resultados: em ambos os grupos foram observados histologicamente um íntimo contato dos biomateriais com o tecido ósseo e a presença de novo tecido ósseo, biomateriais remanescentes, tecido conjuntivo e outros tecidos. Os valores da análise histomorfométrica, de formação do novo tecido ósseo para o grupo A e o grupo B, respectivamente, foram: 28,72% ± 11,81% e 30,69% ± 19,65% no período de três meses, e 23,37% ± 19,02% e 25,78% ± 22,75% no período de seis meses. Foram comparados todos os tecidos formados nos espécimes de três e seis meses entre os grupos A e B, e entre o mesmo grupo nos diferentes períodos de tempo. Não houve diferença estatisticamente signifi cante em nenhuma das comparações. Conclusão: baseado nos resultados, os biomateriais mostraram resultados histológicos e histomorfométricos semelhantes em alvéolos de extração de coelhos...


The aim of this study was to perform an histomorphometric evaluation of tissue formed using the rabbit extraction socket model, with application of two different biomaterials over two different post-surgical follow-up periods. Materials and Methods: fourteen male, New Zealand rabbits were used in this study. Two molars were extracted from the same side of the jaw on each rabbit, having one socket filled with bovine bone mineral (group A) and the other with a synthetic hydroxyapatite (group B). In each group, half of the animals were sacrificed at 3 and 6 months of healing. All samples were prepared for histomorphometric evaluation. Results: in both groups, direct contact of biomaterial with bone tissue was observed, and the presence of new bone, remaining biomaterials, fi brous tissue, and other tissues was demonstrated. Histomorphometric values for new bone tissue formation in groups A and B were 28.72% ± 11.81% and 30.69% ± 19.65%, respectively, at 3 months of healing and 23.37% ± 2.19% and 25.78% ± 22.75%, respectively, at 6 months of healing time. No statistically signifi cant differences were seen for intra and intergroup comparisons at three- and six-month healing periods. ABSTRACT

OBJETIVO: Por meio de ensaios biomecânicos, comparar as capsulorrafias com sutura simples e com âncoras, em quadris de coelhos. MÉTODO: Foram utilizados 13 coelhos, 26 quadris, todos machos da raça Nova Zelândia albinos (Oryctolaguscuniculus). Inicialmente, realizamos um projeto piloto em três coelhos (seis quadris). Este experimento constou de 10 coelhos, divididos em 2 grupos: o Grupo 1 submetido à capsulorrafia (quadris direito e esquerdo) com sutura simples utilizando fio absorvível de ácido poliglicólico e o Grupo 2 submetido a capsulorrafia (quadris direito e esquerdo) com âncora de titânio. Após o período de quatro semanas de operados, todos animais foram submetidos à eutanásia e seus quadris congelados. Após um descongelamento prévio das peças, no mesmo dia das análises biomecânicas, foram avaliados os parâmetros de rigidez, força máxima, deformidade máxima e energia. RESULTADOS: Não houve diferença estatisticamente significante em relação à força no limite de proporcionalidade, rigidez e força máxima entre os grupos com sutura simples e com âncora. CONCLUSÃO: Por meio dos ensaios biomecânicos, tendo como parâmetro a rigidez, a força máxima, a deformidade máxima e a energia, ficou demonstrado que as capsulorrafias em quadris de coelhos com sutura simples e com âncora são semelhantes entre si. OBJECTIVE: Using biomechanical studies, this research aims to compare hip capsulorrhaphy in rabbits, carried out with two different techniques: capsulorrhaphy with simple sutures and with anchors. METHOD: Thirteen New Zealand Albino (Oryctolaguscuniculus) male rabbits, twenty-six hip joints, were used. First, a pilot project was performed with three rabbits (six hip joints). This experiment consisted of ten rabbits divided into two groups: group 1 underwent capsulorrhaphy on both right and left hips with simple suture using polyglycolic acid absorbable thread, and group 2 underwent capsulorrhaphy with titanium anchors. After a four-week postoperative period, the animals were euthanized and the hip joints were frozen. On the same day of the biomechanical studies, after the hip joints were previously unfrozen, the following parameters were evaluated: rigidity, maximum force, maximum deformity and energy. RESULTS: There was no relevant statistical difference in rigidity, maximum force, maximum deformity and energy between the simple suture and anchor groups. CONCLUSION: Through biomechanical analyses, using parameters of rigidity, maximum force, maximum deformity and energy, it has been shown that capsulorrhaphy with simple suture and with anchors has similar results in rabbit hip joints. ABSTRACT

Objective To explore whether the peritoneal cooling was better than other cooling methods on protection neuron damage of the hippocampus CA1 after cardiopulmonary resuscitation (CPR) in New Zealand rabbits.Methods Forty eight adult New Zealand rabbits were induced ventricular fibrillation by AC current and were resuscitated after cardiac arrest for 5 minutes.The rabbits were randomly divided into four groups according to the way of cooling methods,nomothermia group ( NT),peritoneal cooling group (PC),surface cooling group (SC) and local cooling group (LC).The changes of tympanic membrane temperature were recorded in each animal and blood plasma concentrations of electrolyte were tested in each group at different time points after restore of spontaneous circulation (ROSC).Brain tissue were removed,the numbers of vigorous and apoptotic neurons in the hippocampus CA1 area were counted after ROSC at 72h.One-way ANOVA or Mann-Whitney rank was used to determine the statistical significance between two groups.LSD-t test for multiple comparisons,R × C test for ROSC comparisons,a two-tailed value of P <0.05 was considered statistically significant. Results Hypothermia was rapidly induced in PC after ROSC,and the time of arriving at target temperature was (26 ±7) min in PC,(60 ±9) min in SC,(69 ± 12) min in LC respectively; in the maintain hypothermia period,the tympanic membrane temperature was maintained at 33~ 35 ℃ in each group exception nomothermia group (NT).There were no differences with main electrolyte,acid-abase liquid balance and renal function between each group at each time point after ROSC.The numbers of vigorous neurons in hippocampus CA1 area were ( 37.07 ± 6.43 ) /40 × in NT group,(35.13 ± 6.97) /40 × in LC group,(55.76 ± 10.13 ) /40 × in PC group,and (50.70 ± 7.38 ) /40 × in SC group (PC:NT,P<0.01,SC:NT,P<0.01,PC:SC,P=0.043,PC:LC,P<0.01,LC:NT,P=0.52).The numbers of apoptotic neurons were (44.07 ±6.09) /40 × in NT group,(29.88 ±4.81 ) /40× in PC group,( 33.55 ± 5.67 ) /40 × in SC group and ( 42.27 ± 5.20 ) /40 × in LC group respectively (PC:NT,P <0.01,SC:NT,P <0.01,PC:LC,P <0.01,SC:LC,P <0.01,PC:SC,P=0.026,LC:NT,P =0.364 ).Conclusions The new peritoneal cooling method could rapidly induce and maintain hypothermia,and it had better protections on neurons in hippocampus CA1 than surface cooling and local cooling method after ROSC in New Zealand rabbits.

ABSTRACT

Objective To explore the mechanism of imperfect recovery of the renal function after release of chronic ureteral obstruction. Methods Adult New Zealand rabbits(n=36) were randomly divided into normal control group(n=3),obstructive control group(n=3) and experimental group(n=30).The latter two groups were subjected to chronic unilateral ureteral obstruction.Two weeks later,3 rabbits were sacrificed and the others underwent ureterostomy,which were sacrificed separately following 12 hours,24 hours,3 days,7 days,14 days.The renal papilla and cortex were examined by scanning electron microscopy. Results Shortly after the release of ureteral obstruction,massive loss of brush border and ballooning of epithelial cells were noted in proximal tubules.Distal tubules and collecting tubules also showed severe ballooning of epithelial cells and normal struction were not seen.Two weeks later,many tubular components became patent with normal epithelial lining.Some of them,however,were deteriorated further in spite of the release of obstruction.Thus there was a marked tubular heterogeneity in recovery.Conclusion Further deterioration of some tubules might be one of the important pathologic basis for imperfect recovery of renal function after the release of chronic ureteral obstruction.

ABSTRACT

Objective:To compare the characteristics of allergic rhinitis animal models in two different animals(Guinea pigs and New Zealand rabbits) induced with various doses of toluene-2,4-Diisocyanate(TDI).Methods:Low dose and high dose of TDI olive oil solution was used as sensitizer to establish models of Guinea pigs and New Zealand rabbits.The typical symptoms and sings of Guinea pigs and New Zealand rabbits were respectively scored during modeling.After the last challenge,nasal mucosa of Guinea pigs and New Zealand rabbits were histopathologically examined and the level of histamine in nasal mucosa was measured by spectrofluorometer.The mortality rates and model success rates were record.Respiratory tracts of dead animals were dissected and histopathologically examined to find out the causes of their death.Results:Compared with control groups,all the Guinea pigs and New Zealand rabbits in model groups got typical symptoms and sings of allergic rhinitis.Histopathology examination of nasal mucosa showed eosinophile cells infiltrating obviously.The levels of histamine in nasal mucosa were highly increased(P
