ABSTRACT

Aim To investigate the effect of Rhizoma Polygonati( RP) on the functional activity and the level of reactive oxygen species (ROS) on the subculture of endothelial progenitor cells ( EPCs) in aging rats. Methods EPCs from bone marrow in rats were isolated and cultured in vitro and identified. The 2nd EPCs were divided into fourgroups, which were subcultured to the 4th, 6th and 8th passage with application of drug - serum containing RP or not. The positive rate of cell senescence was detected by beta - galactosidase, the proliferation, migration and tubule formation were assayed by MIT, transwell chamber and in vivo angio- gcnesis kit, respectively, and ROS level was detected by flow cytometry. Results In the process of EPCs subculture, the positive rate of cell senescence gradually increased, accompanied by a significant decrease in proliferation, migration and tubule formation. The level of ROS increased sharply (P<0. 05). RP could reduce the positive rate of cell senescence, enhance the functional activities, and decrease the level of ROS of EPCs significantly (P < 0. 05). Conclusions RP may delay the aging process of EPCs in vitro and protect the function of EPCs by decreasing the level of ROS.

ABSTRACT

OBJECTIVE:To observe the effect of different polarity of components of Rhizoma Polygonati Odovati on the spleen and thymus gland in D-galactose-induced aging-model mice so as to establish the anti-aging mechanism and anti-aging active components of Rhizoma Polygonati Odovati.METHODS:Different polarity of extracts of Rhizoma Polygonati Odovati were extracted respectively with benzin petroleum,acetic ether,dehydrated alcohol,80% ethanol,and water as extract solvents.The model mice were intragastrically administered with different polarity of components of Rhizoma Polygonati Odovati for 56 consecutive days,and the effects of each extractive on the indexes and pathological change of immune organ D-galactose-induced aging-model mice were monitored.RESULTS:Compared with each other extractive,aqueous extract of Rhizoma Polygonati Odovati can prevent the atrophy of thymus and spleen,and ameliorate the patho-constitution of spleen and thymus gland.CONCLUSION:Aqueous extract of Rhizoma Polygonati Odovati maybe the anti-aging active components in Rhizoma Polygonati Odovati,and its mechanism maybe related to its function to prevent the atrophy of immune organs and improve the immune function.

ABSTRACT

OBJECTIVE:To assess clinical efficacy and safety of Wonderful(W),a preparation of polysaccharides of rhizoma polygonati,in the treatment of genital herpes(GH).METHODS:In a random,open,parallel and controlled design,400 cases of GH were divided into 5 groups(each group 80 cases):open W spray(WS) group,open W cream(WC) group,parallel WS group,parallel WC group and control group(aciclovir ointment AO).The therapeutic efficacy and ADRs were observed.RESULTS:The total effective rates of open WS group,parallel WS group,open WC group and parallel WC group were 92.50%,91.25%,93.75% and 91.25% respectively.Effects showed in 64.38% of cases treated with WS(open+parallel) and 68.13% of cases treated with WC within 3 days and 91.88% and 91.88% within 7 days.The initiation of effect of W was 2 days earlier than that of AO.No serious ADRs were found and the patients' compliance was good.CONCLUSION:W is rapid in initiation of efficacy,short in therapeutic course,satisfactory in effect,convenient in application and safe.

ABSTRACT

Objective To screen the active cardiovascular components from Rhizoma Polygonati and to observe the effect on myocardial ischemia rat.Methods The active components from Rhizoma Polygonati were extracted by system solvent separation methods,and their cardiovascular effects on the rat models induced by subcutaneous injection of isoproterenol were tracked to find out the optimal active cardiovascular components.Results Among the extracts from Rhizoma Polygonati,the n-Butanol(EB)extract which consisted of glycosides and aglycons,was the best active cardiovascular component;aqueous extract which consisted of alkaloids,flavonoids,glycosides and aglycons,was the better component;the emulsion of Rhizoma Polygonati,which consisted of petroleum ether extract,chloroform extract and water-ethanol extract had less cardiovascular activity.Conclusions The parts with medium polarity from Rhizoma Polygonati have the best cardiovascular activities.

ABSTRACT

Objective To explore the anti-aging mechanisms of Rhizoma Polygonati Odorati(RPO)and its anti-aging active components.Methods After drenching extracts of RPO for 56 days,the behavioral indexes of memory,learning and tolerance,changes of immune organs of spleen and thymus gland,testis index,plasma superoxide dismutase(SOD) activity,hepatic malondialdehyde(MDA)content were observed in D-galactose-induced suhacute senile mice model. Results Aqueous extract shows a trend of improving the memory and learning abilities,and can increase the tolerance of aging-model mice,prevent the atrophy of thymus gland and testis,enhance the activity of SOD and reduce the content of MDA.Conclusion Aqueous extract of RPO may be the anti-aging active components in RPO,and its mechanism may be concerned with the increase of immune function,prevention of the atrophy of genitical glands,counteraction of oxidation,clearance of free radicals and prevention of senile dementia.
