ABSTRACT

Background At present, a large number of reports focus on the bones of limbs and trunk, while there are few studies on the effect of fluorosis on jawbone which is the inevitable structural basis for the development and treatment of oral diseases. Objective To preliminarily investigate the effect of fluoride exposure on the mechanical properties of jawbone by observing the changes in the intraosseous environment and the maximum load against shearing force (LSFmax) of the jawbone in rats with chronic fluoride treatment. Methods Screening experiment: 48 SD male rats were randomly divided into a control group and three fluoride exposure groups (50, 150, and 250 mg·L−1 fluoride concentration), 12 rats in each group. The fluoride exposure groups were molded by feeding different concentrations of sodium fluoride solution, and the control group drank tap water from Guizhou area. Each group was further divided into 4 subgroups with 3 animals each according to observation time points after 0, 2, 4, and 6 months. The LSFmax of the jawbone was measured with an electronic universal ergometer, the expression of type I collagen (Col1) was shown by Sirius red staining, and the expression of runt-related transcription factor 2 (Runx2) was determined semi-quantitatively by immunohistochemistry at selected time points. Formal experiment: 12 male SD rats were randomly divided into a fluoride exposure group and a control group. The fluoride exposure group were fed with 150 mg·L−1 sodium fluoride solution, and the control group drank tap water from Guizhou. After feeding with fluoride for 5 months, the ergometer was used to measure the LSFmax of the jawbone. Osteoclasts were counted after tartrate resistant acid phosphatase (TRAP) staining. Col1, Runx2, bone morphogenetic protein 2 (BMP-2), alkaline phosphatase (ALP), and cathepsin K (Cath K) were detected semi-quantitatively by immunohistochemistry expression and Sirius red staining. Micro computed tomography (Micro CT) was used to observe the trabecular bone microstructure. Results Screening experiment: The LSFmax of the control group and the 50 mg·L−1 fluoride exposure group reached the peak value at the 2nd month, and the LSFmax of the 50 mg·L−1 fluoride exposure group reached the valley value at the 4th month. The LSFmax of the 150 mg·L−1 fluoride exposure group at the 4th month was higher than that at the 6th month (P<0.05). There was no significant difference in the LSFmax at each time point in the 250 mg·L−1 fluoride exposure group. At the same time point, there was no statistically significant difference in LSFmax among the groups. The Col1 levels of the 50 mg·L−1, 150 mg·L−1, and 250 mg·L−1 fluoride exposure groups were higher than the time point 0 from the 2nd month (P<0.05). The Runx2 showed no statistically significant difference by concentration or time. Formal experiment: After feeding with 150 mg·L−1 fluoride for 5 months, the LSFmax of the fluoride exposure group was greater than that of the control group (P<0.05). The expressions of Col1, Runx2, BMP2, ALP, and Cath K in the fluorosis exposure group were higher than those in the control group (P<0.05). There were no statistically significant differences in osteoclast count or indicators of bone trabecular microstructure. Conclusion Chronic fluoride exposure may increase the shear strength of jaw bone.

ABSTRACT

Objective To study von Willebrand factor(VWF) damage based on a novel Maglev Taylor-Couette blood-shearing device. Methods The magnetic levitation (maglev) Taylor-Couette blood-shearing device was designed, and the blood-shearing platform was built. Fresh porcine blood was tested in circulation loop for 1 hour at laminar flow state. VWF damage was assessed by analyzing sample through Western blot and enzyme-linked immunosorbent assay. Results With the increase of exposure time and shear stress, a large number of high molecular weight VWF multimers were degraded into low molecular weight VWF. The maximum rate of degradation was 569%. When the shear stress increased from 18 Pa to 55 Pa, the ratio of VWF-Rco to VWF-Ag decreased from 45.7% to 32.8%. ConclusionsCompared with initial sample, the VWF damage was mainly manifested by the decrease of high molecular weight VWF and the decrease of VWF activity, and VWF-Ag did not change significantly. The novel maglev Taylor-Couette blood-shearing device can quantitatively control the flow parameters (exposure time and shear stress), and be used for blood damage research in vitro, thus providing references for the design and optimization of extracorporeal membrane oxygenation and blood pump.

ABSTRACT

BACKGROUND: Studies have shown that the nano-hydroxyapatite coating on the surface of zirconia has not only high strength and toughness, but also good biocompatibility. It is an ideal substitute for hard tissue. However, the coating is easy to fall off from the surface of zirconia, which is a fatal defect. OBJECTIVE: To prepare functionally graded bioceramics with pure zirconia as matrix, gradient layer in the middle and nano-hydroxyapatite on the surface by gradient composite technology and to screen the optimal mechanical properties and sintering temperature. METHODS: Using zirconia and nano hydroxyapatite powder as raw materials, ceramic specimens were prepared by the lamination method in powder metallurgy. According to the thickness of zirconia layer, three groups A (40 mm), B (30 mm) and C (20 mm) were designated. Each group was sub-divided into three subgroups 1 (3 layers), 2 (5 layers) and 3 (7 layers). Thus, there were 162 ceramic specimens in nine subgroups. The ceramic specimens were sintered at different temperatures (1 300,1 350,1 400,1 450,1500, and 1 550°C) and then processed into rectangular specimens for mechanical property testing. RESULTS AND CONCLUSION: The zirconia-based nano-hydroxyapatite functionally graded material can be formed by 10 MPa single side vertical pressure. With the increases in the number of gradient layers and sintering temperature, the mechanical properties of the functionally graded bioceremics in each group were enhanced. The functionally graded bioceramics had the optimal mechanical properties when sintering temperature was 1 550°C, the number of gradient layers was 7, and substrate thickness was 40 mm. The optimal mechanical property of the functionally graded bioceramics produced at above parameter was significantly superior to that produced at other eight sets of parameters (P<0.05). According to the optimal gradient design, the zirconia-based nano-hydroxyapatite functionally graded ceramic specimens prepared by high temperature sintering technique have stronger mechanical properties.

ABSTRACT

This study aimed to evaluate the nutritive value and shear strength of the Xaraés grass (Urochloa brizantha) under the three intensities of light and four cuts. The experiment was conducted at FMVZ - UNESP, Botucatu, with a randomized block design, with the following treatments: natural luminosity, 30% and 60% light reduction, with four cuts and three replications. The analyzes were bromatological composition, digestibility, and shearing strength. There was no difference in digestibility between the treatments in any of the cuts, forage quality was influenced by the light intensity levels, and the treatment with 60% of light reduction produced higher concentrations of crude protein and ash, lower levels of neutral detergent fiber, hemicellulose, cellulose and shear strength. According to the studied cuts, the first one had the lowest cut interval and produced superior forage compared with the last one, as it obtained lower fiber content in acid detergent, lignin, hemicellulose, cellulose and consequent lower shear force. Therefore, the reduction of 60% of luminosity is beneficial to the quality and shear force of the Xaraés palisade grass.(AU)

ABSTRACT

ABSTRACT: Dietary fiber plays an important physiological role, which is directly linked to its physicochemical properties, water-holding, oil-binding, and cation-exchange capacities. These properties can be altered by employing enzymatic, mechanical, and physical processes. Enzymatic and chemical processes require solvents and special conditions that make it unfeasible to use. Thus, the use of physical methods, such as micronization and extrusion, make promising options to change the physicochemical properties of dietary fiber. In this way, this review aimed to approach relevant information about the use of physical processes, specifically micronization and extrusion, for this purpose. Furthermore, conceptual aspects, such as definition, classification, and properties of dietary fiber and mainly characteristics about the micronization and extrusion processes, are reported. Micronization and extrusion are based on the decrease of the particle size to a micro scale and on the combination of high temperature, mechanical shearing and pressure, respectively. Applying these methods, modifications on the food matrix occurred by increasing the surface area and disruption of the glycosidic bonds. Consequently, there is a change in physicochemical properties of dietary fiber, which predict the physiological effect associated with dietary fiber consumption, such as decrease in blood cholesterol and glucose levels and improvement of intestinal transit. Moreover, these changes increase the bio accessibility of bioactive compounds present in the food matrix and improve the antioxidant capacity of products.

ABSTRACT

This study examined variations in shoulder loading due to differences in the site of stepping foot contact during baseball pitching, while comparing flat ground and mound conditions. Measurement was performed, involving 10 right-handed pitchers who belonged to university baseball clubs, under original flat ground and mound conditions. Pitching movements were classified into 3 categories: [normal], [narrow], and [outside]. Through 3-dimensional motion analysis using a motion capture system, the following results were obtained: 1. The pitching velocity was significantly higher in the [normal] compared with [narrow] and [outside] conditions and under the mound compared with flat ground condition. 2. The peak torque of the shoulder internal rotation was markedly lower in the [narrow] compared with [normal] condition. There were no significant differences between the [normal] and [outside] conditions or between the flat ground and mound conditions. 3. The posterior, superior, and inferior shearing forces, as well as the proximal traction force, which influence the humeral head of the shoulder, were markedly greater in the [normal] compared with [narrow] and [outside] conditions. The anterior and posterior shearing forces and proximal traction force were significantly greater under the mound compared with flat ground conditions. Based on the results, the internal rotation torque of the shoulder, as well as the shearing and traction forces influencing the humeral head of the shoulder, may vary due to differences in the site of stepping foot contact during baseball pitching and between flat ground and mound conditions. The former may also be useful to prevent pitching-related shoulder injuries.

ABSTRACT

Objective: To prepare a new hesperidin nanoemulsion (HDN-NE) with glycyrrhizic acid as emulsifier, by which could develop a “new green nano-pharmaceutics” of hesperidin. Methods HDN-NE was prepared by high-speed shearing and high-pressure homogenization. The prescription of HDN-NE was optimized with particle size, PDI, and appearance as indexes. The physicochemical property and stability of HDN-NE prepared by the optimal prescription were studied. Results: The optimal prescription of HDN-NE was as follow: The content of hesperidin, glycyrrhizic acid, and oil phase were 0.1%, 0.3%, and 5%, respectively. The shear rate was 13 000 r/min, the cutting time was 2 min, the homogeneous pressure and times were 100 MPa and 6, severally. The result showed that the prepared HDN-NE had the mean size of (262.7 ± 3.1) nm, PDI of 0.234 ± 0.009, Zeta potential of (-35.42 ± 0.72) mV, and solubility of (460.3 ± 2.1) μg/mL. The physicochemical property study showed that the conductivity was (116.4 ± 1.7) μs/cm, the pH was 6.820 ± 0.008, and the turbidity was 451 cm-1 (n = 3). It was identified as O/W emulsion by dyeing method. The droplets were spherical and uniform by transmission electron microscopy. The stability study showed that HDN-NE had good stability. Conclusion: HDN-NE with glycyrrhizic acid as an emulsifier can significantly improve the solubility and stability of hesperidin, which is a new potential nano-drug with safety.

ABSTRACT

In Northern Patagonia, the mating season starts on March 15th, when rams are submitted to summer temperatures. Exposure of rams to heat stress increases the prevalence of microscopic damage to spermatozoa, morphological abnormalities, and reductions in fertility. This study assesses the adaptive capabilities of six unshorn and six shorn Australian Merino rams, half of which were treated in a heat chamber for eight hours for five days, gradually reaching a temperature of up to 40 °C. Microscopic damage, abnormalities and ultramicroscopic alterations of the plasma membrane and the acrosome of sperm head were analysed. There were significant differences in the percentage of tailless spermatozoa and proximal cytoplasmic droplets between post-treatment periods. Temperature primarily affected the shorn rams and the sperm heads during spermiogenesis. Submicroscopic alterations were observed when the plasma membrane was present in the anterior segment. These alterations can be intact, waved, or dilated. When the plasma membrane was absent, the acrosome might be intact, dilated, and waved. In addition, the outer acrosomal membrane may completely lose its contents or have a nude nucleus. The plasma membrane assumes a waved shape as a result of the effect of temperature on the epididymis. According to this study, the tailless head, proximal cytoplasmic droplets, and the ultramicroscopic categories studied were robust indicators of semen heat stress. After ten weeks, the sperm head recovered its normal shape. Unshorn rams are better adapted to summer heat stress than shorn ones. Microscopy and transmission electron microscopy alterations have been shown to be excellent indicators of thermal stress in Australian Merino rams and may be useful tools to help sheep farmers choose when to begin the mating season, which will vary depending on the environmental conditions of the summer.(AU)

ABSTRACT

Objective To establish a 3D finite element model of an anti-shearing force Ni-Ti memory shape alloy patella claw for fixing patellar fracture, and analyze its mechanical performance. Methods The internal fixation model of transverse patellar fracture by patella claw was established by Pro/E 5.0, and then imported it into ABAQUS 10.1 for finite element analysis on its mechanical properties. The mechanical performance and deformation of the patella claw under two different patella femoral joint forces FQ at the knee flexion angle of 30°, 60°, 90° were analyzed, respectively. Results Under the same boundary condition, with the respective FQ as 367.5 N and 3 675 N, the maximum displacements of deformation were different at different flexion angles. As compared to fixation by tension band, using patella claw was preferable, with stronger resistance to tension and more stable anti-shearing force. Conclusions Deformation and displacement of the patella claw are in accordance with the biomechanical results needed in clinic, and its stability can satisfy clinical requirements for functional exercise as early as possible.

ABSTRACT

DESCRIPTION: The RECD is the measure of the difference in sound pressure level (SPL) generated in the external ear canal and the SPL generated in the 2cc coupler, given the same input signal and is used at different stages of the HA selection verification in the pediatric population. The insert earphone transducer is commonly used to obtain this measure, being coupled to a foam tip or custom ear mold ? these coupling systems yield different results. In children with normal external and middle ears, the RECD presents positive values at all frequencies. Given the same anatomical and physiological conditions and the same coupling system, there is no significant difference between the RECD for left and right ears. There are great RECD values between subjects, denoting the importance of the individual measurements. An example of the clinical application of RECD is presented. FINAL CONSIDERATIONS: While it is an objective measure, the interpretation of the validity of the results of the RECD is subjective, and the professional must be properly trained to do so. It is recommended that the RECD is obtained with the insert earphone and custom ear mold, and the reassessment made at each ear mold change.

ABSTRACT

The aim of this study was to determine the variation of the temperature after shearing in sheep under dry and hot environment conditions and to compare the temperature changes with variation in cardiac and respiratory frequencies, ruminal movements and hydration status. Twenty Suffolk unshorn ewes were studied. Physical examination was performed in all animals three times a day at 7:00 AM, 1:00 PM and 7:00 PM, during 42 days (22 days before shearing and 20 days after shearing). The skin temperature was measured by infrared thermometer over several surfaces of the body. Data were submitted to analysis of variance, for comparisons between groups (shorn versus unshorn) at each time, and the significant difference was evaluated at level of P<0.05 by Tukey test. The respiratory frequency was statistically significant at all times. When air humidity was high, the respiratory frequencies were low. The thermal stress was clear in sheep of this study, reflecting marked changes in cardiac and respiratory frequencies and rectal temperature. The respiratory frequency was the parameter more reliable to establish a framework of thermal stress in the unshorn sheep, with values on average three times higher than those reported in the literature. The heart rate monitors the thermal variation of the environment and is also an indicator of heat stress. This variation shows the Suffolk breed is well adapted to hot climates. The correlation between the body surface temperatures with environment temperature and air humidity was negative, as explained by the effect of wool insulation, i.e. even with an increase in environment temperature and humidity, the body temperature tends to maintain a compensating balance. In the shorn animals, the correlation between skin temperature with environment temperature and air humidity showed that the skin temperature increases when the environment temperature increases. The increase in the environment temperature does not affect the body temperature of unshorn animals due the insulating effect of the wool. However, when environment temperature rises, the presence of the wool starts to affect the thermal comfort as the heat absorption is larger than the capacity of heat loss. In this study, the best thermal stress indicators were the respiratory frequency and rectal and skin temperatures. The temperatures of the skin measured at the perineum, axillae and inner thigh were considered the most reliable.

ABSTRACT

OBJECTIVE: To prepare lipid microbubbles as well as to evaluate their characteristics and efficacy. METHODS: Octafluoropropane-containing lipid microbubbles were prepared by high speed shearing method. Orthogonal experimental design was used to optimize the prescription. The resultant microbubbles were characterized, in terms of morphology, concentration, size, distribution and the octafluoropropane content. Moreover, the contrast effect of the microbubbles in New Zealand rabbit's kidney and liver was studied. RESULTS: The average concentration of microbubbles was (2.99±0.19)×109 their average size was (2.46±0.05) μm, and 97% of them were smaller than 7 μm. Octafluoropropane content was (387.81±35.28) μg·mL-1. With the help of the lipid microbubbles, satisfactory ultrasound contrast images of rabbit's kidney and liver were got. CONCLUSION: The microbubbles prepared by high speed shearing method have regular morphology, and their size distribution is small and even. It can be used as an excellent ultrasound contrast agent which can enhance significantly the yield of ultrasound examination and thus allow better imaging. ABSTRACT

Objective To detect type Ⅱ collagen synthesis by cultured human chondrocytes under the influence of different intermittent fluid shearing forces. Methods Second passage human monolayer chondrocytes were divided into low-speed (20 rpm/min), mid-speed (40 rpm/min) and high-speed (60 rpm/min) groups according to the rotation speed of the rocking bed. The expression of type Ⅱ collagen was analyzed using immunohistochemistry and RT-PCRs.Results The absorbance of the mid-speed and high-speed groups was significantly greater than that of the low-speed group and a control group. The average optical density of type Ⅱ collagen by RT-PCR was significantly higher in all three sheared groups than in the control group. Conclusions Intermittent shearing can enhance cellular proliferation and the metabolism of human chondrocytes in vitro and promote the expression of type Ⅱ collagen.

ABSTRACT

In this work the effects of pre-lamb shearing on the ewe body condition scores (BCS) and lamb birth weights and growth rates were examined. A flock of 64, three year old Border Leicester x Texel ewes (49-54 kg), exposed to rams in March were divided, on day 74 of pregnancy, into two groups each of 32 ewes. The ewes grazed together, on natural pampas grass, at a stoking rate of five ewes/ha and a pasture availability of approximately 800 kg DM/ha. One group was shorn (S), at day 74 of pregnancy (P74) and the other was left unshorn (US). The BCS of the S and US ewes were assessed at the beginning of the experiment (P74), P108 and P135, as well as at lactation (L) on days L15 and L24. The lamb birth weights, and weights at 2 and 3 weeks of life were also recorded. Both groups of ewes showed a marked reduction of BCS during gestation and lactation periods. The S group experienced a significantly (P < 0.05) more severe reduction of BCS. This dropped from 3.3 score units at P74 to 1.79 and 1.22 at P135 and L24. The BCS of the US group were 3.11, 2.19 and 1.21 score units respectively at these times. Shearing pre-lambing increased mean lamb birth weight by 0.71 kg and this was statistically significant (P < 0.05). There were no effects on the lamb weight gain measured at two and three weeks of life. The increase in lamb birth weight produced by shearing ewes during pregnancy, described in this paper, could reduce the lamb perinatal mortality, especially in years with a low forage supply, leading to a critical BCS during gestation. The results suggested also that close attention should be given to the BCS of the ewe flock during pregnancy, a practice that is not common in the southern areas of Brazil.

ABSTRACT

Changes in soil structure, bulk density and organic carbon content, caused by tillage systems, affect shear strength parameters. The present study was proposed with the objective of evaluating the effects of increased in soil organic carbon (SOC) content on the shear strength of an Hapludult (Argissolo Vermelho Amarelo arênico, Brazil System). Soil surface samples (0-0.02 m), with SOC content varying from 6.5 to 18.8g kg-1 and soil moisture equilibrated at tensions of 6, 100 and 500kPa, were submitted to the direct shear tests with normal tensions varying from 13.6 to 443.0kPa. Cohesion and angle of internal friction values were determined. Besides SOC content, the water content, the saturation degree and the soil bulk density of the samples were determined. The angle of internal friction was of 32.5, 34.9 and 35.4 degrees, while the cohesion was of 0.00, 1.06 and 7.95kPa, respectively for the samples with moisture at 6, 100 and 500 kPa tension. In the driest samples (500kPa), the angle of internal attrition decreased with the increase of the SOC content. The cohesion determined in the direct shear tests was independent of the SOC content. However, the cohesion decreased with the increase of the soil water content.
