ABSTRACT

Objective To investigate the clinical efficacy of Tianshu capsule in the treatment of migraine and its influence on vascular endothelial function,hemodynamics.Methods 80 patients with migraine from October 2014 to October 2015 in our hospital were selected as the research subjects,and according to the different treatment methods, they were divided into control group and observation group.The control group was treated with oral flunarizine hydrochloride capsules,the observation group was given Tianshu capsule on the basis of the treatment of the control group.The blood dynamics of the two groups before and after treatment and the clinical curative effect of the patients [the middle cerebral artery (MCA),anterior cerebral artery (ACA),Fu (PCA)after artery peak systolic velocity (Vs)and diastolic peak flow velocity (Vd)index,endothelin (ET)index]were observed.Results The curative effect of two groups was significantly improved compared with before treatment,the total effective rate of observation group (97.50%)was significantly higher than the control group (75.00%)(χ2 =8.54,P<0.05).The related indices of hemodynamics and endothelium were lower than before treatment,and after treatment,Vs of MCA,ACA,PCA in the observation group were (105.02 ±1.12)cm/s,(80.22 ±1.10)cm/s,(60.34 ±1.20)cm/s,Vd of MCA,ACA,PCA in the observation group were (35.22 ±1.13)cm/s,(40.33 ±1.21)cm/s,(21.23 ±1.12)cm/s,which were signifi-cantly lower than those in the control group[Vs of MCA,ACA,PCA:(120.10 ±1.13)cm/s,(95.33 ±1.12)cm/s, (80.21 ±1.13)cm/s;Vd of MCA,ACA,PCA:(42.33 ±1.10)cm/s,(50.23 ±1.10)cm/s,(30.21 ±1.10)cm/s] (t=25.46,25.76,33.38,12.30,17.04,15.55,all P<0.05).Conclusion The curative effect of Tianshu capsule in the treatment of migraine was improved in patients with cerebral blood flow velocity and vascular diastolic function, and is worthy of promotion.

ABSTRACT

To investigate the effect of hot air circulation drying and spray drying on the quality of Tianshu capsule from the view point of chemical compositions. UPLC-DAD was used to establish the fingerprint of Tianshu capsules for the first time, and the main chemical constituents were identified by UPLC-Q-TOF-MS/MS. A total of 62 compounds were identified in this method, 21 of which were reported in Tianshu capsules for the first time. The results showed that there were no significant differences in the identification of the chemical constituents types between these two methods, but the contents of some constituents were different. The common patterns generated by the 10 batches of hot air cycle drying samples were used as the control fingerprint, and the similarity of the spray drying samples fingerprints was 0.877, with high similarity of the fingerprints between these two methods. UPLC-DAD combined with UPLC-Q-TOF-MS/MS technology was used for the first time to evaluate the chemical constituents of Tianshu capsule rapidly, comprehensively and accurately, providing technical support for the quality evaluation of Tianshu capsule.

ABSTRACT

Based on the near infrared spectroscopy, partial least square (PLS) method was used to respectively develop the quantitative calibration models to fast measure the contents of the total solid and ferulic acid in extraction process of Tianshu capsule extracts. The results showed that in the quantitative model of solid content, the correlation coefficients (R²) of calibration set and cross validation set were 0.967 301 and 0.947 726. The root-mean square error of calibration set (RMSEC) was 0.054 7 and root-mean square error of cross validation set (RMSECV) was 0.069 8. Besides, in the quantitative model of ferulic acid, the correlation coefficients (R²) of calibration set and cross validation set were 0.986 879 and 0.962 243. RMSEC was 1.402 6 and RMSECV was 2.400 2. When the established models were applied to on-line monitoring, the correlation coefficients of predicted results and measured values for total solid content and ferulic acid were 0.993 3 and 0.991 6; root-mean square error of predicted value (RMSEP) was 0.039 3 and 1.669 3 respectively; mean relative deviation of predicted value (RSEP) was 3.49% and 3.58%. The results indicated that the established models can be used to fast measure the contents of the total solid and ferulic acid in extraction process of Tianshu capsule extracts.

ABSTRACT

To study the chemical constituents of Tianshu Capsule in vivo and in vitro. UPLC-Q-TOF-MS was used as the analytic method. The ferulic acid, caffeic acid, ligustrazine hydrochloride, gastrodin, 5-hydroxymethy furfural, and ligustilide were used as the reference compounds. The information on the total ion chromatogram, extraction chromatogram and the mass spectrogram were synthetically analyzed to confirm the constituents of Tianshu Capsule in vivo and in vitro. Twenty-four compounds from Tianshu Capsule were detected among five constituents came from Tianma and 19 constituents came from Chuanxiong. After oral administration of Tianshu Capsule, 13 compounds were absorbed into plasma. The findings obtained from the study can provide the useful information for the determination of bioactive substances and the perfection of quality standard of Tianshu Capsule.

ABSTRACT

Objective: To study the effect of Tianshu Capsule on the rat dura mater model of superior sagittal sinusin following electrical stimulation and discuss its mechanism. Methods: Male SD rats after brain electrode implantation were started on Tianshu Capsule at the doses of 1.08, 0.36, and 0.18 g/kg. Sumatriptanm Tablets at the dose of 9.72 mg/kg were ig given as a positive drug. Except control and Sham groups, the electrical stimulation was carried out in a voltage of 10 V, frequency of 3 Hz, pulse width of 0.25 ms, and time of 1 h. No operation was taken in the control group. The orbital blood was taken after 0, 30, and 60 min of the stimulation than taking brain tissue. The contents of calcitonin gene-related peptide (CGRP), substance P, and norepinephrine (NE) in plasma and brain tissue were detected by radio immunoassay. Results: Compared with the control group, there was no significant difference of CGRP, substance P, and NE contents in brain tissue of rats in the Sham group. Compared with the Sham group, CGRP in the brain tissue of rats in normal group was increased obviously, while substance P and NE levels were significantly decreased (P < 0.05). Comapred with the model group, the Tianshu Capsule could increase the content of NE in plasma and brain tissue (P < 0.05) while decrease CGRP level (P < 0.05). And Sumatriptanm Tablets could reduce the content of CGRP in plasma (P < 0.05). Conclusion: The Tianshu Capsule could significantly improve the damage of rat dura mater of superior sagittal sinusin following electrical stimulation by releasing neurotransmitter, which may be the mechanisms of migraine treatment.

ABSTRACT

Objective: To optimize the one-step pelletization technology of Tianshu Capsule by Minitab software. Methods: Using the qualified rates of particles as the evaluation indexes, three significant impact factors (fluid extract-dextrin ratio, infusion frequency, and atomization pressure) of one-step pelletization technology were selected and the levels of non-significant factors were identified by the Plackett-Burman experimental design. According to single factor experiments, choosing the qualified rates of particles, moisture, and angle of repose as the evaluation indexes, three levels of the three factors were selected by Box-Behnken of central composite design to establish the multiple linear regression and binomial fitting, and the experimental data were optimized by the response optimizer. Results: The best conditions were as follows: fluid extract-dextrin of 1.57:1 put in fluid-bed and heated by fan; the temperature of the bottom material in the fluidized state was increased to 50°C, fluid extract of 2% HPMC was sprayed with frequency of 32 Hz, the frequency of fan was 30 Hz, inlet air temperature was 60°C, atomization pressure was 0.21 MPa, and feed temperature was 50°C. Conclusion: Under the Minitab optimized scheme, the average experimental results are similar to the predicted values, and Minitab software could be used in the optimization of one-step pelletization for Chinese materia medica.

ABSTRACT

@#Objective To observe the therapeutic effect of stellate ganglion block (SGB) combined with Tianshu capsule on migraine.Methods Ninty patients with migraine were randomly divided into the trial group (n=44, treated by SGB combined with Tianshu capsule) and control group (n=46, treated by SGB only). All patients of two groups were evaluated with numeric rating score (NRS) for pain before treatment and 1 day, 3 days, 1 week, 1 month and 3 months after treatment, and the therapeutic effects of two groups were compared.Results The NRS scores of patients in the two groups at each time point decreased, but scores of patients in the trial group were significantly lower than that in the control group at 3 days, 1 month and 3 months after treatment ( P<0.05). The effective rate of the trial group was superior to that of the control group ( P<0.05).Conclusion SGB combined with Tianshu capsule is more effective on migraine and has less side effect.

ABSTRACT

Objective To investigate the effects of Tianshu capsule(TSC) on the plasma levels of ?-endorphin(?-EP) and 5-Hydroxytryptamine(5-HT) and c-fos expression of brain tissue in the rat with migraine.Methods The rat migraine model was established by subcutaneously administration of nitroglycerin.The intragastric administration dose of TSC was divided into three groups as follows:high dose(7.5 g/kg),middle dose(3.75 g/kg) and low dose(1.88 g/kg).TSC was administrated respectively in therapeutic and prophylactic model.The control group and model group were administrated corresponding dose of normal saline.The plasma levels of ?-EP and 5-HT in rat were measured by radioimmunoassay and HPLC respectively;the distribution of ?-EP,5-HT and c-fos in rats periaqueductal grey were evaluated by immunohistochemistry.Results Compared to model group,the plasma levels of ?-EP were increased remarkably in high dose TSC therapeutic and prophylactic groups,middle dose prophylactic group(all P

Objective To investigate the effects of Tianshu capsule on vasoactive substances in blood plasma or brain and hemodynamics in migraine animal models. Methods After subcutaneously administration of nitroglycerin (NTG) and tube feeding Tianshu capsule, the radioimmunoassay or spectrophotometry was used to measure the plasma contents of calcitonin gene related peptide (CGRP), nitric oxide (NO) and nitric oxide synthase (NOS) in rats.The distribution of NOS1, CGRP in rat nucleus tratus spinalis nervi trigemini was detected by immunohistostaining, and the changes of internal carotid artery flow velocity in rabbit were measured by transcranial Doppler.Results The levels of NO, NOS and CGRP were elevated in the plasma of migraine rats. ABSTRACT

AIM: To compare the clinical effect of Tianshu Capsule(Rhizoma Chuanxiong,Rhizoma Gastrodiae,efc) on migraine with Flunarizine. METHODS: 82 cases of patients with migraine were divided randomly into the treatment group(40 cases),which were treated with oral Tianshu Capsule,4 capsules(1.36 g),3 times daily after meals,and the control group(42 cases),which were treated with Flunarizine(Sibelium) 5 mg quaque nocte,for 2 months.One year follow-up was carried out.The frquency and duration of migraine were analyged before and after treatment,and duration of positive results after withdraw of drugs were also recorded.The body weight,blood and urine routine,liver and kidney function as well as electrocardiograph were compared before and after treatment.
