ABSTRACT

OBJECTIVE To optimize ethanol extraction process of Yihuang powder. METHODS An orthogonal experiment was designed by reflux extraction with ethanol volume fraction, liquid-to-material ratio, and extraction time as investigation factors. The parameters used were the contents of hesperidin, nobiletin, tangeretin, gallic acid, chebulagic acid, chebulinic acid, liquiritin, glycyrrhizin, eugenol, and the paste-forming rate. The analytic hierarchy process (AHP) was used to calculate the comprehensive score. The optimal ethanol extraction process parameters of Yihuang powder were determined by verifying the results predicted by orthogonal experiment and genetic algorithm (GA)-back propagation neural network (BP neural network). RESULTS The optimal ethanol extraction process parameters, as optimized by orthogonal experiment, were as follows: ethanol volume fraction of 60%, liquid-solid ratio of 14∶1 (mL/g), extraction time of 90 min, and extraction for 2 times. The comprehensive score obtained by verification was 79.19. Meanwhile, the optimal ethanol extraction process parameters, optimized by GA-BP neural network, were ethanol volume fraction of 65%, liquid-solid ratio of 14∶1 (mL/g ), extraction time of 60 min, and extraction for 2 times. The comprehensive score obtained by verification was 85.30, higher than the results obtained from orthogonal experiment. CONCLUSIONS The optimization method of orthogonal experiment combined with GA-BP neural network is superior to the traditional orthogonal experiment optimization method. The optimized ethanol extraction process of Yihuang powder is stable and reliable.

ABSTRACT

Objective To prepare a sustained-release membrane with longer adhesion time and dissolution time, and compare it with the commercially available oral ulcer membrane. Method Adhesion strength, adhesion time, swelling coefficient, dissolution time, etc. were used as the inspection indicators, and a combination of single factor inspection and analytic hierarchy process were used to screen the membrane -forming materials. The dispersion method of clotrimazole, ornidazole and borneol were investigated to prevent the drug from seed out. The method of combining orthogonal experiment and analytic hierarchy process were used to optimize the dosage of CMC-Na, PVA-1788 and glycerin; and the commercial products were compared. Results Through single-factor investigation and orthogonal experiment, the optimal ratio of excipients was selected as CMC-Na∶PVA-1788∶glycerol (3∶1∶0.08). The water-insoluble component clotrimazole, ornidazole and borneol were treated by precipitation in liquid with good effect. The best method was used to prepare the membrane. The adhesion strength was 102 g. The adhesion time was 55 min. The swelling coefficient was 1 939.52. The average dissolution time was 110 min. The appearance was white and the surface was free of bubbles, soft and elastic. The membrane forming time at 60 ℃ was 300 min and the demolding effect was better which could be completely peeled off with moderate thickness. Conclusion The oral ulcer membrane developed in this method has good appearance, comfortable use, strong adhesion, long adhesion time and dissolution time, and could stay on the ulcer surface for a long time to form physical isolation, and slowly release the drug during the dissolution process, which could play the role of long-term pain relief, antibacterial, anti-inflammatory and promote healing effects on oral ulcers.

ABSTRACT

This study explored toxicity attenuation processing technology of Rhizoma Dioscoreae Bulbiferae stir-fried with Paeoniae Radix Alba decoction for the first time, and further explored its detoxification mechanism. Nine processed products of Rhizoma Dioscoreae Bulbiferae stir-fried with Paeoniae Radix Alba decoction were prepared by orthogonal experiment with three factors and three levels. Based on the decrease in the content of the main hepatotoxic component diosbulbin B before and after processing of Rhizoma Dioscoreae Bulbiferae by high-performance liquid chromatography, the toxicity attenuation technology was preliminarily screened out. On this basis, the raw and representative processed products of Rhizoma Dioscoreae Bulbiferae were given to mice by gavage with 2 g·kg~(-1)(equival to clinical equivalent dose) for 21 d. The serum and liver tissues were collected after the last administration for 24 h. The serum biochemical indexes reflecting liver function and liver histopathology were combined to further screen out and verify the proces-sing technology. Then, the lipid peroxidation and antioxidant indexes of liver tissue were detected by kit method, and the expressions of NADPH quinone oxidoreductase 1(NQO1) and glutamate-cysteine ligase(GCLM) in mice liver were detected by Western blot to further explore detoxification mechanism. The results showed that the processed products of Rhizoma Dioscoreae Bulbiferae stir-fried with Paeoniae Radix Alba decoction reduced the content of diosbulbin B and improved the liver injury induced by Rhizoma Dioscoreae Bul-biferae to varying degrees, and the processing technology of A_2B_2C_3 reduced the excessive levels of alanine transaminase(ALT) and aspartate transaminase(AST) induced by raw Rhizoma Dioscoreae Bulbiferae by 50.2% and 42.4%, respectively(P<0.01, P<0.01). The processed products of Rhizoma Dioscoreae Bulbiferae stir-fried with Paeoniae Radix Alba decoction reversed the decrease protein expression levels of NQO1 and GCLM in the liver of mice induced by raw Rhizoma Dioscoreae Bulbiferae to varying degrees(P<0.05 or P<0.01), and it also reversed the increasing level of malondialdehyde(MDA) and the decreasing levels of glutathione(GSH), glutathione peroxidase(GPX), and glutathione S-transferase(GST) in the liver of mice(P<0.05 or P<0.01). In summary, this study shows that the optimal toxicity attenuation processing technology of Rhizoma Dioscoreae Bulbiferae stir-fried with Paeoniae Radix Alba decoction is A_2B_2C_3, that is, 10% of Paeoniae Radix Alba decoction is used for moistening Rhizoma Dioscoreae Bulbiferae and processed at 130 ℃ for 11 min. The detoxification mechanism involves enhancing the expression levels of NQO1 and GCLM antio-xidant proteins and related antioxidant enzymes in the liver.

ABSTRACT

OBJECTIVE To prepare Leonurine hydrochloride tablets and evaluate the quality. METHODS The wet granulation technology was adopted ;leonurine hydrochloride was used as the crude drug ,and the types of fillers ,disintegrants,binders and lubricants were screened by single-factor experiments. Combined with orthogonal experiments ,using the cumulative dissolution rate within 15 minutes(using water as dissolution media )as index ,the proportion of disintegrants ,the mass fraction of binder solution,and the proportion of lubricants were screened and verified. The in vitro dissolution behavior of the prepared Leonurine hydrochloride tablets (dissolution media were hydrochloric acid solution of pH 1.2,acetic acid-sodium acetate solution of pH 4.5, phosphate buffer solution of pH 6.8,water),tablet appearance ,hardness,friability and content uniformity were tested according to the general principles in 2020 edition of Chinese Pharmacopoeia (part Ⅳ). RESULTS The optimal formulation of Leonurine hydrochloride tablets included leonurine hydrochloride crude drug of 500 mg,dextrin of 9 250 mg,crosslinking polyving y- pyrrolidone of 200 mg,magnesium stearate of 50 mg,1% hydroxypropyl methyl cellulose solution of 4 mL. The average 15-minute cumulative dissolution rate of the three batches of tablets was 81.25%(RSD=1.12%,n=3). In above 4 dissolution media,the dissolution equilibrium of prepared tablets could be reached within 30 minutes,and the cumulative dissolution rates exceeded 85%. The prepared tablets had uniform beige in color ,smooth surface ,complete edge ,no mottle ,spot,foreign matter , etc.,hardness of 57.3 N(n=6),weight loss rate of 0.15%. The content uniformity was in accordance with relevant provisions in 2020 edition of Chinese Pharmacopoeia (part Ⅳ). CONCLUSIONS Leonurine hydrochloride tablets are successfully prepared , and the quality comply with relevant regulations.

ABSTRACT

To optimize the ethanol extraction technology parameters of Fengyin Decoction by orthogonal experiment combined with beetle antennae search(BAS)-genetic algorithm(GA)-back propagation neural network(BPNN). Based on single factor investigation, the extraction temperature, ethanol volume, extraction time, and ethanol concentration were used as orthogonal experiment factors, and entropy weight method was used to calculate the comprehensive scores of aloe-emodin, glycyrrhizic acid ammonium salt, rhein, emodin, chrysophanol, physcion, cinnamaldehyde, 6-gingerol, extraction ratio and fingerprint similarity. BAS-BPNN model was established, and then, GA was used to predict the optimal extraction process. The results showed that BAS-BPNN was optimized to obtain the optimal ethanol extraction process of Fengyin Decoction as follows: extraction temperature of 87 ℃, adding 9 times of 75 % ethanol, and extracting for 47 minutes, with a comprehensive score of 1.052 9. Meanwhile, the optimal process parameters obtained by orthogonal design were as follows: the extraction temperature of 80 ℃, adding 10 times of 75% ethanol, extracting for 30 minutes, with a comprehensive score of 1.003 7. The comprehensive score of the process obtained from the BAS-BPNN model was slightly better than that from the orthogonal test, indicating that the optimized process from BAS-BPNN model was more ideal, so it was finally determined as the best extraction process for Fengyin Decoction. The process of Fengyin Decoction obtained from BAS-GA-BPNN has high extraction efficiency and good stability, which provides reference for the subsequent development and quality control.

ABSTRACT

BACKGROUND: Most current studies of orthopedic screw parameters focus on mechanical properties but lack biocompatiblllty assessment. OBJECTIVE: To Investigate the Influence of geometric parameters and material properties of orthopedic screws on blomechanical compatibility, explore the optimal combination of various factors and parameterization levels, providing a theoretical reference for the design of orthopedic screws. METHODS: An orthogonal experiment protocol was designed according to the five factors and three parameterization levels of orthopedic screws. Factor A was the screw diameter (4.5, 5.5, 6.5 mm), factor В was thread profile (rectangular, trapezoidal, triangular), factor С was screw pitch (1.75, 2.25, 2.75 mm), factor D was the thread depth (0.5,1.0,1.5 mm), and factor E was elastic modulus (45,110, 200 GPa). The blomechanical finite element analysis of the three-dimensional bone-nail model under different sets of screw parameters was carried out. Stress distribution and two experimental indicators (the stress transfer parameter STPa between the cortical bone and the screw, and the stress transfer parameter STPß between the cancellous bone and the screw) under different experimental schemes were obtained. The weighted matrix analysis method was used to analyze the orthogonal experimental data to obtain the influence law and the optimal combination. RESULTS AND CONCLUSION: Different levels of different factors have different degrees of influence on biocompatiblllty. Among them, the screw material properties are the most Important factors, followed by the thread profile, pitch and thread diameter, and the last was the thread depth. The thread profile, pitch, and thread diameter had similar influence on biocompatiblllty. The optimal combination of the orthogonal experiment was an orthopedic screw with a large diameter of 6.5 mm, a trapezoidal thread, a pitch of 2.75 mm, a thread depth of 0.5 mm, and an elastic modulus of 45 GPa.

ABSTRACT

Objective: To optimize the supercritical CO2 extraction process of active ingredients from Magnoliae Officinalis Cortex (MOC) and explore the antioxidant activity of the extracts. Methods: The content of magnolol and honokiol of the supercritical CO2 extracts of MOC was determined by HPLC, and the extraction process was optimized by orthogonal experiment. The antioxidant activities of the extracts were determined by MTT. Results: The optimum extraction pressure of magnolol was 25 MPa, the extraction temperature was 55 ℃, the amount of CO2 was 30 kg, and the optimum extraction parameters mentioned above of honokiol were 15 MPa, 50 ℃, and 25 kg, respectively. Conclusion: Under the optimum extraction conditions, magnolol and honokiol have high extraction efficiency, good repeatability, stability and feasibility, and the extract have good antioxidant activity.

ABSTRACT

Objective: To prepare ligustrazine pamoate (Lig-PAM) sustained-release nanosuspension (Lig-PAM-NSps) and determine its in vitro release characteristics. Methods: Lig-PAM was prepared by hydrophobic salt formation method and its physicochemical properties were characterized. Then, Lig-PAM-NSps was prepared by miniaturized medium grinding method. The prescription and preparation process of Lig-PAM-NSps were optimized by the single factor and orthogonal experiment with average particle size, polydispersity index (PDI) and stability coefficient (SI) as indicators. Lig-PAM-NSps was characterized, and its stability and in vitro release was also investigated. Results: The compound ratio of Lig-PAM prepared by Lig and PAM in the amount of 1:1 was (97.48 ± 0.04)%. Compared with Lig, the solubilities of Lig-PAM in water and simulated body fluids were decreased by 95.50% and 77.39%, respectively. Fourier transform infrared spectroscopy (FT-IR) and X-ray powder diffraction (XRD) showed that the Lig and PAM formed Lig-PAM. The optimum prescription size of Lig-PAM-NSps was (585 ± 5) nm, PDI was (0.328 ± 0.015) and SI was (0.928 ± 0.012). The scanning electron microscopy (SEM) showed that Lig-PAM-NSps was spherical with uniform size distribution, and the particle size was about 600 nm and its physical stability was good within 60 d. The results of in vitro release showed that Lig-PAM-NSps had obvious sustained-release effect compared with Lig solution within 48 h, and showed the first-order release characteristics [ln(1-Q) = 0.153 67 t + 80.458 14, r = 0.998 26]. Conclusion: The preparation progress of Lig-PAM-NSps is stable and can release Lig slowly in vitro.

ABSTRACT

Objective To optimize the extraction process of the compound herba lysimachiae by orthogonal experiment and response surface method. Methods The sum of the peak areas of multiple representative components in the compound herba lysimachiae was used as the evaluation index. The ultrasonic extraction time, ultrasonic temperature and ethanol ratio were analyzed to optimize the extraction process conditions by orthogonal experiment and Box-Behnken response surface analysis method. Results Ultrasonic extraction time and ethanol ratio are important factors in the extraction process of the compound herba lysimachiae. The effect of ultrasonic temperature in the extraction process is not significant. The optimal extraction condition for compound herba lysimachiaeare is 10 min ultrasonic extraction time at 55 ℃ with 55% ethanol. Conclusion The optimal extraction process parameters obtained by combining orthogonal experiment and Box-Behnken response surface method can be used for the extraction of representative components from the compound herba lysimachiae and pharmaceutical research in the future.

ABSTRACT

Objective: In view of druggability issue of limonin (LM), the liposomal preparation was developed. The liposomal formulation and preparation process were optimized, and its in vitro antitumor activity was investigated. Methods: In this study, LM was loaded in liposomes to increase its stability and solubility. Meanwhile, in vitro cytotoxicity of LM@Lip was evaluated. LM@Lip were prepared by thin-film dispersion method, and formulation selection and process optimization were operated by single factor and orthogonal experiment. Size distribution, PDI and zeta potential were measured by Malvern sizer, and the encapsulation efficiency and drug loading content were determined by HPLC. The dialysis method was used to investigate the release profile of LM@Lip. In vitro cytotoxicity against HepG2 and A549 cells were estimated by MTT method. Results: The optimized preparation conditions of liposomes were as follows: drug/lipid ratio was 1:150, cholesterol/lipid ratio was 1:9, the ultrasonic power was 120 W for 6 min (1 s interval). The average particle size, PDI and Zeta potential of optimized LM@Lip were (119.5 ± 6.2) nm, 0.318 ± 0.124, (-17.2 ± 1.3) mV, respectively, and the encapsulation efficiency and drug loading content were 87.9% and 0.57%. The final concentration of LM was 63.4 μg/mL. The release results showed 58.59% drug was released in 12 h. MTT results showed that the IC50 of LM@Lip on HepG2 and A549 cells was 20.16 and 15.39 μg/mL, respectively, and its in vitro antitumor was superior to that of LM. Conclusion: Liposomes can increase the stability and solubility of LM. LM@Lip showed slow-release profile and significant tumor inhibition superior to LM.

Objective:To explore the effect of processing methods on the content of toddaline in Toddalia asiati- ca Lam., so as to provide reference for the development and clinical application of T. asiatica in the future. Methods: The root of T. asiatica was processed by the processing methods of water-washing, water-boiling, water-steaming, winesteaming and stir-frying with wine. The content of toddaline in the processed products of T. asiatica was determined by high performance liquid chromatography(HPLC). Further, the orthogonal test design methodology was used for the stirfrying with wine process to investigate the effect of frying fire power, time and solid-liquid ratio(the ratio of raw material to yellow rice wine)on the toddaline content in T. asiatica. Results: The processing of the T. asiatica roots by the pro- cessing methods of water-washing, water-boiling, water-steaming and stir-frying with wine could all reduce the toddaline content in T. asiatica roots and the reducing effect was especially significant in the stir-frying with wine process. In con- trast, the steaming with wine(the wine-steaming)could increase the toddaline content in T. asiatica. The results of the orthogonal test for the stir-frying with wine process showed that the processing time most greatly influenced the toddaline content, the influence on the content came next from the solid-liquid ratio, and the influence from the frying fire power was lowest. The toddaline content in T. asiatica roots was increased by processing with Wen Huo (the weak frying fire power), with processing time 10 min and solid-liquid ratio 1:1. In contrast, the toddaline content was most significantly reduced when the roots were processed with Zhong Huo(the medium frying fire power), with processing time 30 min and solid-liquid ratio 1:1. Conclusion: The effect of different processing methods on the content of toddaline in T. asiatica was differed in the tested five processing methods. For the stir-frying with wine process, the change of processing parame- ters also caused the different effect on the toddaline content in in T. asiatica.

Abstract Clavibacter michiganensis subsp. michiganensis (Cmm), which is a Gram positive bacterium, causes the bacterial canker of tomato disease. The purpose of the study was to evaluate the antibacterial activity of Polygonum orientale extracts against Cmm. In this study, firstly, effects of three extracting parameters (extractive time, extractive temperature, and solid to liquid ratio) of orthogonal experiment design L27 (313) were conducted. Secondly, survival rate was determined and inhibition zone of Cmm rescued post-stress was monitored. Finally, extracellular OD260nm value, extracellular protein content, conformational structure of membrane protein, extracellular alkaline phosphatase (AKP) activity, and ATPase activity were measured to investigate the antibacterial mechanism. Results of orthogonal experiment revealed that extractive time and extractive temperature had highly significant (P<0.01) effects on the antibacterial activity of P. orientale extracts. The optimum conditions were as follows: 10h of extractive time, 60°C of extractive temperature, and 1:20 (g:mL) of solid to liquid ratio. This study also demonstrated that the living cells of each sample from survival rate test had almost no resistance or adaptability, and rescued Cmm cells were much easier to be inhibited by P. orientale extracts. The results of antibacterial mechanism indicated that cell membrane and cell wall of Cmm were seriously damaged by P. orientale extracts, and P. orientale extracts reduced the intracellular ATPase activity dramatically. All these findings suggested that P. orientale extracts had a strong antibacterial activity to inhibit Cmm, and could be used for the ecological management of the bacterial canker of tomato disease.

ABSTRACT

Objective To evaluate the effects of locked plates with different screw layouts on stability of the internal fixation system for femoral shaft fracture, so as to provide guidance for clinical study on screw layout of the internal fixation system for femoral shaft fracture. Methods Orthogonal test design method was used with 4 groups of screws at 3 parametric levels. Biomechanical properties of the internal fixation system for femoral shaft fracture under 9 screw layouts were analyzed respectively by the finite element method, and 4 inspection indices, namely, the maximum equivalent stress of the plate, the screw and the femur, and the maximum axial displacement of the internal fixation system were obtained from each group of the experiment, and the orthogonal test weight matrix analysis method was conducted for data analysis. Results The influences of different screw parameters on stability of the internal fixation system varied, and the maximum weights of the 4 sets of screw parameters in stability of the internal fixation system were 13.86%, 7.57%, 7.53% and 5.91%, respectively. Conclusions The best scheme for screw layout in orthogonal test is the screw with none-fixed threaded holes in the first group, the single cortical screws with fixed threaded holes in the second group and the double cortical screws with fixed threaded holes in the third and fourth group.

Objective To screen the extraction process of Wenfei Jiangzhuo Recipe (WJR) by pharmacodynamics and acute toxicity test, and to optimize it by orthogonal experiment to determine the best extraction process of WJR. Methods Acute toxicity test was carried out on different WJR samples. The improvement of learning and memory of vascular dementia (VD) rats was used as the efficacy index, and the results of the acute toxicity and efficacy experiments determined the best process plan. Using contents and yield rate as index, orthogonal test was used to screen the amount of solvent, extraction time and extraction times. Results The results of acute toxicity test showed that all mice survived without obvious toxicity. The results of pharmacodynamic test showed that the best extraction plan of WJR was as follows: Extracting white prepared lateral root of Aconite, Codonopsis Radix, and Glycyrrhizae Radix et Rhizoma Praeparata cum Melle with water, and Notoginseng Radix et Rhizoma, Zingiberis Rhizoma, and wine processing Rhei Radix et Rhizoma with ethanol. The optimized extraction technology were as follows: The former were 10-fold water reflux extraction for three times and 1 h each time, with white prepared lateral root of Aconite first reflux 2 h; The latter were 6-fold 70% ethanol reflux extraction three times and each time 1.5 h. Conclusion WJR can significantly improve the learning and memory ability of VD model rats. The optimized extraction conditions are reasonable and feasible, and can provide the basis for the subsequent development of WJR.

ABSTRACT

Objective To optimize the method for integration of field processing and crude drug preparation of Anemarrhenae Rhizoma (AR) and to provide scientific evidence for the procedure of AR. Methods Four main factors including drying temperature of the fresh herbal medicine, the water content of the medicine, thickness of the products, and the drying temperature of decoction pieces were studied by using single factor experiment and orthogonal test. The content of timosaponin B-Ⅱ and mangiferin and the appearance of the products were selected as evaluation indexes to optimize the integral processing of AR by using comprehensive evaluating method. The changing of the body temperature of yeast-induced rat model was used to compare the antipyretic activity between the two processing technologies (the integration technology and traditional technology). The content of the blood glucose, insulin, and glycosylated serum proteins (GSP) in the blood of the STZ-induced diabetic rat model were selected as the evaluation indexes to compare the hypoglycemic activity between the two processing technologies. Results AR was dried at 50 ℃ for 11 h (water content of medicinal materials was 45%—50%), and then the flosses were knocked off in the drum for 30 min. The results showed that 40—50 ℃ was the best drying temperature, 45%—50% was the best water content, and then unhairing process were conducted for AR for 30 min. The thickness of the slice was 4 mm and the best drying temperature was 50 ℃. There were no significant differences in chemical composition and hypoglycemic activity between the integration technology and traditional technology, while the antipyretic effect of the integrated processing was better than that of the traditional technology. Conclusion The technology of integration of field processing and crude drug processing is feasible and it can be used in industrial production.

ABSTRACT

Objective To investigate the extraction process of Yuchang granules .Methods The extraction process was optimized by orthogonal experiment based on the content of berberine ,atractylodes lactone Ⅰ and the extraction yield .Results Through the visual analysis of orthogonal experiment ,the analysis of variance and the verification experiments ,the optimum extraction process was obtained as follows :extracting 1 h for twice with 10 times water of medicinal herbs and the 70% ethanol depositing concentration .Conclusion This process is consistent and feasible .It is a simple and convenient method to extract Yuchang granules .

ABSTRACT

OBJECTIVE:To prepare Dexzopiclone orally disintegrating tablets (DODT),and to optimize its formulation.METHODS:Direct powder compression method was used to prepare DODT.Using repose angle of material,disintegration time and taste evaluation as indexes,single factor test was used to screen the types or amount of bulking agent,disintegrating agent,glidant and flavoring agent;using disintegration time as index,orthogonal experiment was applied to optimize the proportion of bulking agent,the amount of disintegrating agent,glidant and flavoring agent.Then the hardness and main component contents of DODT prepared by optimal formulation were determined.RESULTS:The optimal formulation was as follows as the ratio of mannitol-MCC 1 ∶ 4,the amount of disintegrating agent PVPP was 15%,the amount of glidant magnesium stearate was 1.0%,the amount of flavoring agent stevia was 3.0%.Three batches of prepared DODT were smooth in surface and good in taste;their disintegration time were(26.7 ± 1.2),(26.7 ± 0.6),(27.6 ± 0.9)s,hardness were (3.59 ± 0.19),(3.49 ± 0.18),(3.27 ± 0.16) kg,and contents were (99.47 ± 0.15) %,(99.53 ± 0.05)%,(99.46 ± 0.20) %,respectively (all RSDs≤0.87%,n=3).CONCLUSIONS:Prepared DODT are all in line with the quality requirements of orally disintegrating tablets.

ABSTRACT

Objective To optimize the formulation of the sunscreen used in military training.Methods The different formulations of sunscreen were prepared according to the orthogonal design,and then applied on 3M adhesive tape posted on the color dish with a certain amount;the absorbance in 280 nm-320 nm and 320 nm-400 nm of them were measured and the arithmetic mean (A) was concluded.By comparing A,the protective effect and the main factors affecting preventive bask were determined.The preventive bask test of the optimum formula was carried out in human trials using the method of multi point increasing ultraviolet irradiation.Results OMC,TinosorbTM M,rutin,pearl powder and baicalin all had significant effects in preventing UVA.OMC had a significant effect in preventing UVB.The formula 1 (containing the five materials mentioned above) had the best effects in preventing both UVA (SPF 18) and UVB (PA+).Conclusions The formula 1 has the best effects in preventing both UVA and UVB.

ABSTRACT

Objective To study the optimal technology parameters of the enrichment process for total flavonoids of Crataegus pinnatifida Bge,with macroporous Resin. Methods The enrichment process was studied by orthogonal experiment design using orthogonal from L9(34) with the content of total flavonoids from Crataegus pinnatifida Bge,as indexes, the factors such as adsorption capacity, the column diameter-height ratio, eluant volume and adsorption time were investigated. Results The optimum elution parameters were as follows: the eluant volume of 60% alcohol—the wet resin amount (g)—the sample volume (mL) was 2:1, the sample was 4:1, the column diameter-height ratio was 1:6 , the adsorption time was 1h, respectively. Conclusion This process enriching the total flavonoids of Crataegus pinnatifida Bge,is feasible.

ABSTRACT

Objective To optimize enrichment and purification technology of total flavonoids from seed shell of Scutellaria baicalensis by macroporous resin.Methods Scutellarin was selected as the index,and its content was determined by HPLC.Types of macroporous resin were optimized by static adsorption elution tests.Enrichment and purification technology was investigated by orthogonal test and single-factor tests.Results AB-8 macroporous resin showed better effects than others.Optimum purification condition was as follows:sample concentration was 30.0 g · L-1,the best adsorption capacity was 0.3 g· g-1,adsorption velocity was 1.0 mL· min-1,diameter-height ratio was 1 ∶ 3,the eluting agent was 20 BV purified water,with the total polyphenols being desorbed by 5 BV of 70% ethanol Conclusion The technology is scientific,simple and highly reproducible.It can be applied to enrich and purify total flavonoids from seed shell of Scutellaria baicalensis by macroporous resin.
