ABSTRACT

To assess the effect of nesiritide on the endothelial function of iliac arteries following endothelia trauma. Right iliac artery trauma was created with a balloon catheter. Ten rabbits were treated with a 4-week subcutaneous injection of nesiritide at a fixed daily dose of 0.1mg/kg. Ten rabbits received daily normal saline injection. Plasma endothelin 1 (ET-1), nitric oxide (NO), and Von Willebrand Factor (vWF) were measured before and after the therapies. Tissue proliferating cell nuclear antigen (PCNA) was measured after the treatment. After the treatment, in the therapeutic group, the area under internal elastic membrane and the residual lumen area were higher than in the normal saline group (P <0.05). The plasma levels of ET-1 (91.6±6.8 vs 114.9±6.3 ng/L, P =0.001), vWF (134.6±10.8% vs 188.8±10.4%, P =0.001) and the ratio of PCNA positive expression (11.7±4.2% vs 36.2±11.4%, P =0.005) in the therapeutic group was lower than in the normal saline group, while the plasma levels of NO was higher (89.7±9.3 vs 43.5±5.3 µmol/L, P =0.001). Nesiritide inhibited remodeling of rabbit iliac artery following endothelial trauma. The inhibition of vascular remodeling may be related to the alleviated endothelial dysfunction and reduced expression of tissue proliferating cell nuclear antigen

ABSTRACT

Objective To observe the clinical efficacy of acupuncture and miuci therapy combined with Qinggan Tongqiao Decoction for sudden sensor-neural hearing loss (SSHL) and the effects on level of plasma endothelin (ET) and serum nitrate oxide (NO). Methods Totally 110 patients with SSHL were randomly divided into treatment group and control group, with 55 patients in each group. The control group was given Qinggan Tongqiao Decoction combined with acupuncture routine treatment, and treatment group was given miuci therapy on the basis of the control group, two weeks as a course of two groups. Level changes of plasma ET and serum NO, and pure tone listening threshold changes before and after treatment were observed. Tinnitus and hearing efficacy was evaluated. Results The total effective rate for tinnitus efficacy was 94.54% (52/55) in the treatment group and 85.45% (47/55) in the control group, with statistical significance (u=2.038, P=0.040). The total effective rate for hearing efficacy was 96.46%(53/55) in the treatment group and 81.82% (45/55) in the control group, with statistical significance (u=2.913, P=0.040). Compared with before treatment, the level of plasma ET decreased and the level of serum NO increased after the treatment (P<0.05); After the treatment, the level of plasma ET in the treatment was lower than the control group, and the level of serum NO in the treatment group was higher than the control group (P<0.05). Pure tone threshold scores in the two groups on treatment 3, 7, 14 d were significantly improved (P<0.05), and pure tone threshold scores in the treatment group were significantly lower than the control group (P<0.05). Conclusion Acupuncture and miuci therapy combined with Qinggan Tongqiao Decoction can significantly reduce clinical symptoms and enhance the hearing level of SSHL patients. The mechanism mainly lies in regulating levels of plasma ET and serum NO, and improving the inner ear micro-circulation.

ABSTRACT

Objective To observe the changes of malondialdehyde (MDA), superoxide dismutase (SOD-1) in serum and plasma endothelin (ET)and angiotensin II(AⅡ) in patients with severe closed head injury before and after being treated with hyperbaric oxygenation (HBO). Methods Fifty patients were recruited. Thirty-two patients were treated with HBO in addition to the conventional management (HBO group) and the other eighteen patients without HBO except for conventional management served as the controls (control group). Thirty healthy volunteers served as normal controls (normal group). Activities of MDA ,SOD-1,ET and AII were measured with spectrometry and radioimmunoassay in all the subjects, respectively, during the study. Results Before treatment, there was a significant difference between the normal group and HBO group as well as the control group in regard to the activities of the 4 parameters(P0.05) as the 4 parameters were concerned. The patients scored 3-5 with GCS in the control group were significantly different from those scored the same with GCS in the normal group in terms of the SOD-1 and MDA. Higher GCS was found in patients with low activities of MDA, ET and AII but high SOD-1 levels. Favourable changes of GCS and GOS were observed in patients in the HBO group as compared with those in the control group (P

ABSTRACT

Endothelin (ET) is an endothelium-derived vasoconstrictive peptide. We have developed a sensitive and selective radioimmunoassay for porcine/human endothelin (ET-1). 126I-labled ET was perpared by lodogen method and was purified by HPLC. Its specific activity was about 64.75 TBq(1750Ci)/mmol. The assay has a detection limit of 0.17pg/tube and the assay range was 0.25-1000pg/tube. The procedure was developed for extraction of endothelin from human plasma using C18 Sep-pak extraction cartridges. Human plasma samples were extracted, assayed and the plasma ET values of 17 healthly volunteers was found to be 2.81 ?0.60pg/ml. Both patients with uraemia(n = 20) and acute myocardial infarction(n= 11) had significantly higher plasma values than normal subjects.
