ABSTRACT

Objective To evaluate the effect of Shenfuhuang injection (SFH) on tissue and plasma TNF-? level and multiple organ dysfunction in septic rats. Method Wistar rats were subjected to sepsis by ceral ligation and puncture, then they were randomly divided into four groups, normal control, sham operation grop, septic group, SFH group. Animals were sacrificed at 2, 8, 24, 48 h. TNF-? level of lung, liver and plasma were measured. DAO activity was determined. Result TNF-? level of liver, lung and plasma were markedly increased at 2~8 h post-CLP compared with sham operation group. In SFH group, TNF-? level decreased in different degrees. DAO activity was much lower in SFH group than in CLP group at 2, 8 h. Conclusion Early applying of SFH could markedly inhibit TNF-? level of vital organs and injury of intestine, thereby preventing the development of excessive inflammation response and subsequent multiple organ dysfunction syndrome associated with CLP-induced sepsis.

ABSTRACT

Objective To study the expression level of myostatin in the gastrocnemius muscle of cancer cachexia mice and to observe the improvement of the status and the influence on myostatin expression by use of meloxicam.(Methods The) tumor-bearing cachexia mice model was established by Lovo cell line subcutaneous inoculation.Twenty-four BALB/c mice were randomly divided into 3 groups(n=8 for each group):group A,control;group B,tumor-bearing mice plus saline;group C,tumor-bearing mice plus meloxicam(5 mg/kg).The food intake and body composition were documented.The expression of myostatin in the gastrocnemius muscle was investigated by RT-PCR and immunohistochemistry in all the animals,and the correlation analysis between serum TNF? and myostatin was conducted. ABSTRACT

Objective To investigate changes of serum levels of interleukin(IL)-4,-6,-8 and tumor necrosis factor-?(TNF-?),and probe their clinical significance in children with Henoch-Schonlein purpura(HSP)/ HenochSchonlein purpura nephritis(HSPN).Methods Serum levels of IL-4,-6,-8 and TNF-? of 45 children with HSP/HSPN and 43 healthy children were examined by enzyme linked immunosorbent assay(ELISA),changes and correlation between the aforementioned cytokines in children with HSP /HSPN were analyzed.Results 1.Serum levels of IL-4,-6,-8 and TNF-? of children with HSP were higher than those of control group(P0.05);The serum level of TNF-? was positively correlated to the serum levels of IL-6,-8 of patients with HSP(r=0.670 P

ABSTRACT

Objective To observe the effects of recombinant human erythropoietin(rh-EPO) on immune function of cyclophosphamide-treated mice.Methods Six-week-old mice were randomly divided into 4 groups.Cyclophosphamide plus normal saline group(CTX(+NS));cyclophosphamide plus lower and higher dosage rh-EPO group(CTX+LDrh-EPO and CTX+HD rh-EPO);normal saline control group(NS).The changes of Hb,WBC,red blood cell immune function((C_3b)-R%),T lymphocyte proliferative responsiveness optical density index A(A score),IL-2,TNF-? of 4 groups were observed.Results The CTX + NS group showed lower levels on Hb,WBC,C_(3b)-R%,A score,IL-2 and TNF-? compared with NS normal control group(P0.05).There were positive correlation between C_(3b)-R% and Ascore,C_(3b)-R% and IL-2.Conclusions 1.Cyclophosphamide treated mice have lower Hb,WBC,C_(3b)-R%,A,IL-2 and TNF-? secretion.2.rh-EPO administration have improvements with Hb,RBC immune function,T lymphocyte proliferative responsiveness and IL2 production.

ABSTRACT

Objective To detect the intervened effect of the compound of Polygonum caspidatum to the NAFLD rat models and analyze its mechanism at the level of gene expression.Methods The Wistar rats were fed with high fatty chow to induce NAFLD and intervened with the compound of Polygonum caspidatum.The mRNA relative level of leptin,adiponectin,tumor necrosis factor ?,resistin and uncoupling protein 2 in the adipose tissue of the intervenient and control rats was detected with RT- qPCR method.The data differences between two groups were analyzed in t-test.Results Comparing to the control group,the relative level of leptin mRNA in the intervenient group was significantly increased(P0.05).At the same time,the resistin and UCP2 mRNA didn’t been detected in all samples of these two groups.Conclusion Metformin hydrychloride can adjust the adipose metabolism in liver and improve the steatosis and inflammatory reaction in liver cell.

OBJECTIVE To study the correlation of hepatitis B virus(HBV)basic core promoter(BCP) mutation with the interleukins(IL-10,IL-12),tumor necrosis factor(TNF)-?,interferon(IFN)-?,as well as HBV DNA contents in patients with chronic hepatitis B virus infection.METHODS(1) Project subject: 176 patients(chronic hepatitis B with mild,moderate and severe degree;liver cirrhosis,chronic fulminant and hepatocellular carcinoma(HCC)) with chronic hepatitis B virus infection were studied.(2) Project methods: ① The A to T mutation at nucleotide 1762 and G to A mutation at nucleotide 1764 were determined by the method of polymerase chain reaction(PCR) microplate hybridization ELISA in these patients.② The serum cytokines(IL-10,IL-12,TNF-? and IFN-?) of these patients were measured by specific-ELISA.RESULTS The serum levels of cytokines(IL-10(80.96?30.86 vs 72.11?24.19 mg/L),IL-12(41.33?15.10 vs 35.98?14.47 mg/L),TNF-?(56.04?27.05 vs 38.01?10.49 mg/L),IFN-?(19.81?12.29 vs 16.55?8.99 mg/L))and HBV DNA contents(108.2478?0.9826 vs 105.8876?1.4822copies/ml) in BCP mutant group were significantly higher than that in non-mutant group(P

OBJECTIVE: To study the curative effect of Sodium Ferulate( SF) on rheumatoid arthritis( RA) and its effects on the levels of serum vascular endothelial growth factor( VEGF) and tumor necrosis factor- ? ABSTRACT

AIM: To investigate the protective effects and the mechanisms of prostaglandin E1 (PGE1 ) on ischemia/reperfusion (I/R)-induced lung injury after lung transplantation in rats. METHODS: 36 SD rats were randomly divided into 3 groups ( n = 12 in each) : sham operation group ( control group) , lung transplantation (LT)group and PGE1 treatment group. PGE1 was administered to the rats through intra-venous way from 10 min before the operation to the end of the reperfusion. The wet/dry ratio of lung, lung permeability index and neutro-phil percentage were detected in bronchoalveolar lavage fluid (BALE). Superoxide dismutase (SOD) and malond-ialdehyde (MDA) of lung tissue were measured by color-imetry. Serum level of tumor necrosis factor ?(TNF?)was detected by enzyme-linked immunosorbent assay (ELISA). ABSTRACT

Objective To study the expression of IL 1?,TNF ? and nitric oxide (NO) in the cervical intervertebral disc and their roles in the pathogenesis of cervical spondylotic myelopathy (CSM). Methods Fifteen cervical discs were obtained from patients of CSM undergoing anterior disc surgery. Six cervical discs specimens were obtained in the surgical operation with patients with traumatic cervical injury served as the control. The specimens were analyzed for IL 1?、TNF ? by use of ELISA method, NO was measured by optical density assays. Results The contents of IL 1?,TNF ? and NO in disc of CSM patients increased significantly when compared with the control group. Conclusion The results of the present study indicated that IL 1?,TNF ? and NO may be involved in the pathogenesis of CSM.

ABSTRACT

Objective To study the effects of ulinastatin (UTI) on the inflammatory cytokines in the blood serum of rats after severe burn. Methods A total of 96 SD rats, inflicted with 30% TBSA full thickness skin burn, were randomly divided into simple burn group (group B) and UTI treated group (group UTI). The serum contents of IL 1?, IL 6, and TNF ? were determined at 0, 1, 3, 6, 12, and 24 h before and after burn injury. Results In group B, the levels of serum IL 1?, IL 6, and TNF ? were significantly higher at 3 h and peaked at 6 and 12 h after burn. ABSTRACT

Objective To investigate the effects of nuclear factor ?B (NF ?B) activation and tumor necrosis factor ?(TNF ?) expression on the progressive heart failure following myocardial infarction in rats. Methods Male rats were randomized for the proximal left anterior descending branch coronary artery ligation and then killed at 4, 8 and 12 weeks after ligation for the examination of hemodynamic parameters, ventricular mass index, protein expression of TNF ?, and NF ?B activity. Objective To investigate the changes of tumor necrosis factor- ?(TNF- ?) and souble intercellular adhesion molecule - 1 (sICAM - 1) in serum and cerebrospinal fluid (CSF) in children with viral encephalitis (VE) before and after treatment,and to explore the pathogenesis of the cytokins in VE. ABSTRACT

Objective To analyze the actions of transforming growth factor ? 1 (TGF-? 1) and tumor necrosis factor ?(TNF-?) in the liver fibrosis formation in chronic viral hepatitis patients and liver cirrhosis patients.Methods Serum levels of TGF-? 1 and TNF-? were measured by ELISA in 107 patients with chronic liver diseases,liver biopsy was performed in 23 patients in order to identify pathological stages under optic microscope.Results The serum levels of TGF-? 1(?g/L) in mild,middle and severe groups of chronic viral hepatitis and liver cirrhosis group were 14 2?5 9,20 1?7 0,30 2?6 7 and 32 6?7 5 respectively.The serum levels of TNF-? (pg/L) were 6 1?3 2,29 8?18 6,57 3?22 5 and 96 7?38 2 respectively.The serum levels of above two markers increased with hepatitis progression.The results of liver biopsy revealed that the changes in these two markers were related to liver cirrhosis degree.Conclusions TGF-? 1 and TNF-? are related to liver cirrhosis formation.

ABSTRACT

In order to investigate the influence of interleukin 1?(IL 1?) and tumor necrosis factor ? (TNF ?) on gene expression of rat interstitial collagenase (MMP 13 ) in hepatic stellate cells, cells of a hepatic stellate cell line (HSC T6) were cultured in medium containing IL 1? or TNF ? for different incubation periods. Total RNA of HSC was extracted and MMP 13 gene expression levels were measured by reverse transcription polymerase chain reaction. The results showed that MMP 13 gene expression level of the IL 1? group was much higher than that of the control group at 8h, 24h and 48h after exposure to IL 1?. The maximal value was 3 fold of the control group and was at 24h . MMP 13 gene expression level of the TNF ? group was also significantly higher than that of control group at 8h, 24h, 48h, 72h, and the maximum nalue was 4 fold of the control group at 48h. These results indicated that IL 1? and TNF ? might increase the gene expression of MMP 13 in hepatic stellate cells.

ABSTRACT

Objective To assess the ability of interleukin-1 beta (IL-1?) antibody and tumor necrosis factor alpha (TNF?) antibody to prevent postoperative abdominal adhesion formation in mice.Methods 60 BALB/c mice were randomly divided into four groups: normal saline group(n=15), anti-IL-1? group(n=15), anti-TNF? group(n=15),and anti-IL-1? combined with anti-TNF? group(n=15). After an identical peritoneal injury operation, mice were treated using intraperitoneally with either normal saline or corresponding antibody at the time of abdominal closure. The mice were killed 20 days after operation, the abdominal incisions and the development of intra-abdominal adhesions were observed. Adhesion scoring was based on an overall assessment of the extent,location, and type of adhesion.Results Anti-IL-1? combined with anti-TNF? group had significantly lower adhesion scores (0 84?0 19) than that in other groups(P

ABSTRACT

Objective:To study the effect of recombined TNF ? combined with induction of apoptosis and its relation with expression of bcl 2 in SW480 human colon cancer cells.Methods:The inhibitive effect of 5 Fu and TNF ? was measured by MTT.Apoptosis of colon cancer cells was studied by TUNEL.The expression of bcl 2 gene was determined by immunohistochemical analysis.Results:After SW480 colon cancer cells were treated with combination in various concentration of 5 Fu and 3 000 U/mL of TNF ? for 48 h,MTT assay revealed a significant decrease of relative viability in compasrison to the control.When SW480 colon cancer cells were treated with 3 000 U/mL of TNF ? and 125 ug/mL of 5 Fu in combination for 48 h,TUNEL method showed the number of nick end positive cells was significantly increased in comparison to the control.The expression level of bcl 2 gene lowered significantly than that of the control group.Conclusion:These results confirmed that combined TNF ? and 5 Fu could synergistically induce apoptosis in SW480 cells and decrease the expression level of bcl 2 gene.

(IFN?) and interferon ?(IFN?) on the effect Methods:Two deletion constructs,pCOLH0 27 and pCOLH2 5 ,containing portions of 5' flanking region of the gene ( 268~+42 bp and 2 483~+42 bp respectively) ligated to the chloramphenicol acetyltransferase (CAT) reporter gene ,were transiently transfected into human skin fibroblasts The cells were subsequently treated with IGF 1 or insulin, and TNF? or IFN? or IFN? The CAT activity was assessed 24 h after the cytokines added Results:The results showed that 13 nmol/L IGF 1 caused 3 68 fold and 4 04 fold increase in the CAT activity of pCOLH0 27 and pCOLH2 5 respectively and that 2 5 ?mol/L insulin caused 3 69 fold and 3 93 fold increase in the activity of the two plasmids respectively TNF?,IFN? and IFN? reduced IGF 1 or insulin induced promoter activity of pCOLH2 5 Conclusion:IGF 1 and insulin could increase the promoter activity of the human (COL1A1) gene TNF?,IFN? and IFN? could suppress the promoter activity induced by IGF 1 and insulin
