Resumen: Una proporción importante de infecciones en el pie diabético consisten en onicomicosis y tinea pedis, problema común en el pie, amenazante de la viabilidad del tejido que puede provocar infecciones bacterianas secundarias. Requieren períodos prolongados de tratamiento antimicótico con alta tasas de recaídas y reinfección. Diversos estudios han mostrado la seguridad y eficacia de las nanopartículas de plata (NP Ag) como agente antimicrobiano. Realizamos un estudio donde se evaluó el tratamiento con NP Ag en dermatomicosis del pie de pacientes diabéticos. Método: estudio piloto, abierto, prospectivo, randomizado y controlado en pacientes que asisten a una policlínica de pie diabético. Dieciocho pacientes cumplieron con los criterios de inclusión conformándose dos grupos homogéneos. Ambos grupos recibieron tratamiento estándar que consistió en antimicótico tópico y desbastado mecánico. El grupo intervención utilizó un textil (medias) confeccionadas con hilos con NP Ag. Se realizó control clínico y microbiológico hasta las 12 semanas. Se evaluó el porcentaje de remisión y el tiempo hasta alcanzar el mismo. Resultados: predominó la onicomicosis y el germen Trichophyton rubrum. En el grupo intervención se logró un mayor porcentaje de remisión de lesiones y en un tiempo menor que el grupo control. Conclusiones: el uso de medias confeccionadas con hilos de NP Ag se asoció con una mayor probabilidad de curación completa en un período de 12 semanas a pesar de que el número de pacientes no permitió llegar al nivel de significación estadística, pudiendo contribuir a la prevención de infecciones o úlceras suplementarias en el pie diabético.


Summary: Onychomycosis and tinea pedis represent a significant proportion of infections in the diabetic foot, a common foot problem, and they constitute a threat to the viability of tissues that may provoke secondary bacterial infections. To combat them, antifungal treatments are required for long periods of time, the rates of relapse and reinfection being high. Several studies have proved the safety and effectiveness of silver nano particles (NP Ag) as an antimicrobial agent. A study was conducted to assess nanoparticle agents for foot dermatomycosis in diabetic patients. Method: pilot, open, prospective randomized and controlled study in patients who are assisted in a diabetic foot policlinic. 18 patients complied with the inclusion criteria and two homogeneous groups were formed. Both groups received standard treatment consisting in topic antifungal and mechanical roughing. The intervention groups used a textile (stockings) made with silver nanoparticle threads. Clinical and microbiological control was made during 12 weeks, also assessing the remission percentage and the time it took to achieve it. Resultados: onychomycosis and trichophyton rubrum prevailed. The intervention group showed a greater percentage of remission of lesions in a period of time that was shorter than that of the control group. Conclusions: the use of stockings made with NP Ag threads was associated with a greater probability of complete healing, in a 12-week period, despite the fact that the number of patients was not statistically significant. This could contribute to the prevention of supplementary infections or ulcers in the diabetic foot.


Resumo: Uma proporção significativa de infecções do pé diabético consiste em onicomicose e Tinea pedis, um problema comum nos pés que ameaça a viabilidade do tecido e pode causar infecções bacterianas secundárias. Requerem períodos prolongados de tratamento antifúngico com altas taxas de recidiva e reinfecção. Vários estudos têm demonstrado a segurança e a eficácia das nanopartículas de prata (NP Ag) como agente antimicrobiano. Realizamos um estudo onde o tratamento com NP Ag foi avaliado na dermatomicose do pé de pacientes diabéticos. Método: estudo piloto, aberto, prospectivo, randomizado e controlado em pacientes atendidos em uma policlínica de pé diabético. Dezoito pacientes preencheram os critérios de inclusão, formando dois grupos homogêneos. Ambos os grupos receberam tratamento padrão que consiste em antifúngico tópico e moagem mecânica. O grupo intervenção utilizou um tecido (meias) confeccionado com fios NP Ag. O controle clínico e microbiológico foi realizado até 12 semanas. A porcentagem de remissão e o tempo para alcançá-la foram avaliados. Resultados: predominaram a onicomicose e o germe Trichophyton rubrum. No grupo intervenção, obteve-se maior percentual de remissão das lesões e em menor tempo que o grupo controle. Conclusões: o uso de meias confeccionadas com fios NP Ag esteve associado a uma maior probabilidade de cura completa, no período de 12 semanas, apesar do número de pacientes não permitir atingir o nível de significância estatística, podendo contribuir para a prevenção de infecções ou úlceras adicionais no pé diabético.

Objetivo: Determinar las propiedades antimicrobianas de la incorporación de nanopartículas de óxido de zinc y cobre en un adhesivo de grabado y lavado total sobre Streptococcus mutans en pacientes con restauraciones de resina compuesta confeccionadas con adhesivo cargado. Métodos: Este estudio clínico randomizado, participaron 25 pacientes, de ambos sexos, pertenecientes al posgrado de Ortodoncia de la Facultad de Odontología de la Universidad de Chile, en los cuales se confirmó presencia de Streptococcus mutans en saliva. Se confeccionaron restauraciones de resina compuesta oclusales, en premolares superiores con indicación de exodoncia por el tratamiento de ortodoncia, con adhesivo cargado (cuya composición es 5 / 0.2 por ciento ZnO y Cu respectivamente) y control (sin presencia de nanopartículas en su composición), según el listado de aleatorización. Se tomaron muestras microbiológicas en tres tiempos con la Técnica de la cubeta (antes, 1 semana y 4 semanas posterior a la confección de las restauraciones). Se obtuvieron, aislaron e identificaron colonias de Streptococcus mutans a partir de las muestras obtenidas. Los datos fueron analizados por el test de Mann Whittney. Resultados: El promedio del recuento de UFC (unidades formadoras de colonias) de Streptococcus mutans en el grupo experimental fue mayor posterior a la confección de las restauraciones de resina compuesta.Los resultados de la identificación molecular por PCR demuestran la presencia de Streptococcus mutans en 20 de 25 muestras. Conclusiones: No existen diferencias en el recuento de Streptococcus mutans antes y después de la aplicación del adhesivo sobre las restauraciones de resina compuesta(AU)


Objective: Determine the antimicrobial properties of the incorporation of copper and zinc oxide nanoparticles into a total rinse and etch adhesive against Streptococcus mutans in patients with composite resin restorations made with loaded adhesive. Methods: A randomized clinical study was conducted of 25 patients of both sexes from the orthodontics graduate course taught at the Dental School of the University of Chile, in whom the presence of Streptococcus mutans was confirmed in saliva. Occlusal composite resin restorations were performed in upper premolars with exodontia indicated as part of the orthodontic treatment, using loaded adhesive (composition 5 / 0.2 percent ZnO and Cu, respectively) and control (without nanoparticles in its composition), according to the randomization list. Microbiological samples were taken at three moments applying the tray technique (before, 1 week after and 4 weeks after the restorations). Streptococcus mutans were obtained, isolated and identified from the samples taken. Data analysis was based on the Mann-Whitney test. Results: Mean Streptococcus mutans CFU count in the experimental group was higher after the composite resin restorations were made. Results of PCR molecular identification show the presence of Streptococcus mutans in 20 of 25 samples. Conclusions: No differences were found in the Streptococcus mutans count before and after application of the adhesive over the composite resin restorations(AU)

Introdução: Nos últimos anos ocorreu o aumento de casos relacionados com a infecção por Candida spp. e Staphylococcus spp., bem como o aparecimento de cepas resistentes a antibióticos convencionais. A biossíntese de nanopartículas consiste na redução de um íon metálico por compostos de origem natural como metabólitos secundários de plantas e organismos, sendo a forma mais indicada por apresentar menor toxicidade quando comparada à síntese química. Desta forma, a síntese biológica constitui uma alternativa para a obtenção de novos agentes ativos para o tratamento de infecções microbianas. Objetivos: Sintetizar nanopartículas de prata a partir do extrato aquoso de Mikania glomerata Sprengel e avaliar possível atividade microbicida e citotóxica. Material e Métodos: Para a síntese das nanopartículas de prata (AgNPs) foi utilizado um extrato aquoso das folhas de M. glomerata e uma solução de nitrato de prata. As AgNPs sintetizadas foram avaliadas por espectrofotômetro UV-vis e espectrometria de absorção atômica com chama. Além disso, a atividade antimicrobiana foi avaliada contra cepas de Candida albicans e Staphylococcus aureus e atividade citotóxica contra linhagens celulares HeLa e Vero. Resultados: As AgNPs são mais eficientes no combate à linhagem de Candida albicans e Staphylococcus aureus quando comparadas ao extrato puro administrado. Até a concentração de 100 mg/mL do extrato puro não foi observado efeito inibitório em ambos os micro-organismos. Entretanto quando em contato com as AgNPs, a concentração inibitória foi de 0,006 mg/mL e 0,1 mg/mL para S. aureus e C. albicans, respectivamente. O efeito citotóxico nas células se comportou de maneira dose-dependente, apresentando maior potencial citotóxico contra a linhagem celular cancerosa HeLa. Conclusão: As AgNPs sintetizadas apresentaram potencial antimicrobiano contra C. albicans e S. aureus, além de baixa atividade contra células normais, indicando sua confiabilidade para aplicação das AgNPs como forma alternativa de tratamento. Estes resultados são promissores e contribuem para pesquisa relacionada à produção de medicamentos utilizando extrato de plantas e metais.


Introduction: In recent years there has been an increase in cases related to infection by Candidaspp. and Staphylococcus spp., as well as the appearance of strains resistant to conventional antibiotics. Nanoparticle biosynthesis consists of the reduction of a metal ion by compounds of natural origin as secondary metabolites of plants and organisms, being the most indicated form because it presents less toxicity when compared to the chemical synthesis. In this way, the biological synthesis is an alternative to obtain new active agents for the treatment of microbial infections. Objective: Synthesize silver nanoparticles from the aqueous extract of Mikania glomerata Sprengel and evaluate possible microbicidal and cytotoxic activity. Material and Methods: For the synthesis of the silver nanoparticles (AgNPs) an aqueous extract of the leaves of Mikania glomerata plus a solution of silver nitrate was used. AgNPs synthesized was evaluated by UV-vis spectrophotometer and FAAS. Furthermore, antimicrobial activity was evaluated against strains of Candida albicans and Staphylococcus aureus and cytotoxicity activity against HeLa and Vero cell lines. Results: AgNPs are shown to be more efficient in combating Candida albicans and Staphylococcus aureusstrains when compared to the pure administered extract. Up to the concentration of 100 mg/mL of the pure aqueous extract no inhibitory effect was observed on both microorganisms. However when the strains were in contact with AgNPs, the inhibitory concentration was 0.006 mg/mL and 0.1 mg/mL for S. aureus and C. albicans, respectively. The cytotoxic effect on the cells behaves in a dose-dependent manner, presenting greater cytotoxic potential against the HeLa cancer cell line. Conclusion: Thus, these results are promising and contribute to research related to the production of drugs using plant extract and metals. The AgNPs synthesized presented the antimicrobial potential against C. albicans and S. aureus, in addition to low activity against normal cells, indicating their reliability for application of AgNPs as an alternative form of treatment.

Abstract Radiation combined injury, a life-threatening condition, has higher mortality than simple radiation injury. The aim of the present study was to analyze the efficiency of Aloe vera and silver nanoparticles in improving the healing of ulcerated oral mucosa after irradiation. Thirty male Albino mice were divided into five groups: control, radiation, Aloe vera (AV), silver nanoparticles (NS), and AV+NS. The mice were exposed to whole body 6Gy gamma-radiation. After one hour, 20% acetic acid was injected into the submucosal layer of the lower lip for ulcer induction. The animals received topical treatment with the assigned substances for 5 days. Lip specimens were subjected to hematoxylin and eosin and anti alpha-smooth muscle actin immunohistochemical staining. Results demonstrated occurance of ulcer three days post irradiation in all groups except in the AV+NS group where only epithelial detachment was developed. After seven days, data revealed persistent ulcer in radiation group, and almost normal epithelium in the AV+NS group. A significant reduction of epithelial thickness was detected in all groups at the third day as compared to control. At the seventh day, only the AV+NS group restored the epithelial thickness. Area percent of alpha-smooth muscle actin expression was significantly decreased in radiation group at the third day followed by significant increase at the seventh day. However, all treatment groups showed significant increase in alpha-smooth muscle actin at the third day, which decreased to normal level at the seventh day. Our study demonstrated the efficiency of Aloe vera and silver nanoparticles in enhancing ulcer healing after irradiation.

Abstract Purpose: To evaluate the effectiveness of sodium hyaluronate, sesame oil, honey, and silver nanoparticles in preventing of postoperative surgical adhesion formation. Methods: Forty male Wistar rats were randomly assigned into five groups with eight rats in each group including control, hyaluronate, sesame, honey and silver groups. After two weeks the animals underwent laparotomy and were evaluated by two different blinded surgeons for severity of adhesions based on the two different classification scoring systems including Nair classification and cumulative adhesion scoring scale. Results: The scores of severity of adhesions in the hyaluronate and sesame groups were significantly lower than the control group based on the Nair classification (both P-values = 0.02), however based on the cumulative adhesion scoring scale just the score of severity of adhesions in the hyaluronate group was significantly lower than the control group (P-value = 0.02). In the hyaluronate group the severity of adhesions was decreased by 48% based on the cumulative adhesion scoring scale. Conclusions: Sodium hyaluronate and sesame oil may have a significant effect in preventing postoperative surgical adhesion formation.

Purpose: To assess the effectiveness of heparin, platelet-rich plasma (PRP), and silver nanoparticles on prevention of postoperative adhesion in animal models. Methods: Sixty males Albino Wistar rats aged 5 to 6 weeks were classified into five groups receiving none, heparin, PRP, silver nanoparticles, PRP plus silver nanoparticles intraperitoneally. After 2 weeks, the animals underwent laparotomy and the damaged site was assessed for peritoneal adhesions severity. Results: The mean severity scores were 2.5 ± 0.9, 2.16 ± 0.7, 1.5 ± 0.5, 2.66 ± 0.88, and 2.25 ± 0.62 in the control, heparin, PRP, silver and PRP plus silver groups, respectively with significant intergroup difference (p = 0.004). The highest effective material for preventing adhesion formation was PRP followed by heparin and PRP plus silver. Moreover, compared to the controls, only use of PRP was significantly effective, in terms of adhesion severity (p = 0.01) . Conclusion: Platelet-rich plasma alone may have the highest efficacy for preventing postoperative peritoneal adhesions in comparison with heparin, silver nanoparticles and PRP plus silver nanoparticles.

ABSTRACT

A difusão da nanotecnologia a fim de projetar novos materiais em escala nanométrica conduziu ao desenvolvimento de uma ampla variedade de nanopartículas fabricadas para aplicações comerciais. Tendo por objetivo buscar um produto que aprimore as características dos materiais dentários em voga, optou-se por pesquisar um novo material que não é utilizado em Odontologia, mas que tem uma extensa aplicabilidade em diversos campos, como na indústria alimentícia, na superfície de bancadas de hospitais, em instrumentais cirúrgicos, paredes, pisos, roupas e no setor automobilístico, de aviação e naval. Assim, buscou-se avaliar um nanofilme de dióxido de silício, comercializado como vidro líquido, e seu possível aperfeiçoamento com a incorporação de nanopartículas de prata à sua composição quanto ao seu comportamento microbiológico utilizando como substrato amostras de resina acrílica autopolimerizável. Primeiramente foram realizados ensaios para caracterização da solução do vidro líquido através da avaliação do teor de sólidos presentes e da sua densidade, além da superfície dos nanofilmes formados por meio de microscópio eletrônico de varredura (MEV) acoplado a espectrômetro de energia dispersiva (EDS). Depois, foi averiguado o comportamento microbiológico dos nanofilmes quanto à citotoxicidade e à formação de biofilme in vitro e in situ. Na caracterização do vidro líquido identificou-se uma pequena discrepância quanto ao teor de sólidos e a densidade obtida em relação aos dados do fabricante. Análises semiquantitativas por MEV e EDS confirmaram a presença da prata no nanofilme experimental e permitiu a observação da configuração de nanoaglomerados formados após a polimerização de ambos os filmes. O ensaio de citotoxicidade mostrou que após a formação do nanofilme este não é citotóxico, mas que as soluções de sílica e experimental apresentam citotoxicidade em baixas diluições. Quanto à formação de biofilme, independente do substrato e do tipo de ensaio, in situ ou in vitro, o nanofilme experimental inibiu a adesão bacteriana. Conclui-se então que o nanofilme experimental de sílica com nanopartículas de prata desenvolvido neste trabalho apresenta o potencial de diminuir o crescimento bacteriano em materiais dentários aplicados provisoriamente em áreas críticas onde é necessária a cicatrização tecidual


The propagation of nanotechnology in order to design new nanoscale materials led to the development of a wide variety of nanoparticles manufactured for commercial applications. With the objective of seeking a product that enhances the characteristics of dental materials in vogue, it was decided to search for a new material that is not used in dentistry, but has a wide applicability in various fields, such as in the food industry, surface countertops of hospitals, surgical instruments, walls, floors, clothing and automotive, aviation and shipbuilding industry. Thus, it sought to evaluate a silicon dioxide nanofilm, commercially available as liquid glass, and its possible improvement with the addition of silver nanoparticles to its composition using samples of acrylic resin as substrate. Initially tests were carried out to characterize the liquid glass solution by evaluating the solids content and their density, beyond the surface of nanofilms formed by means of a scanning electron microscope (SEM) coupled with energy dispersive spectrometer (EDS). Then it was examined microbiological behavior of nanofilms for cytotoxicity and in vitro and in situ biofilm formation. In the liquid glass characterization it was identified a small discrepancy in the solids content and the density obtained compared to manufacturer's data. The SEM and EDS analysis confirmed the presence of silver in the experimental nanofilm and allowed the observation of the nanoclusters configuration formed after polymerization of both films. The cytotoxicity assay showed that after the formation of nanofilm this is not cytotoxic, but the silica and the experimental solutions exhibit cytotoxicity at lower dilutions. As to the formation of biofilm regardless of the type of substrate and the assay in situ or in vitro, experimental nanofilm inhibit bacterial adhesion. It follows then that the silica experimental nanofilm with silver nanoparticles developed in this work has the potential to reduce bacterial growth in dental materials provisionally applied in critical areas where tissue healing is required

The microbial synthesis of nanoparticles is a green chemistry approach that combines nanotechnology and microbial biotechnology. The aim of this study was to obtain silver nanoparticles (SNPs) using aqueous extract from the filamentous fungus Fusarium oxysporum as an alternative to chemical procedures and to evaluate its antifungal activity. SNPs production increased in a concentration-dependent way up to 1 mM silver nitrate until 30 days of reaction. Monodispersed and spherical SNPs were predominantly produced. After 60 days, it was possible to observe degenerated SNPs with in additional needle morphology. The SNPs showed a high antifungal activity against Candida and Cryptococcus , with minimum inhibitory concentration values ≤ 1.68 µg/mL for both genera. Morphological alterations of Cryptococcus neoformans treated with SNPs were observed such as disruption of the cell wall and cytoplasmic membrane and lost of the cytoplasm content. This work revealed that SNPs can be easily produced by F. oxysporum aqueous extracts and may be a feasible, low-cost, environmentally friendly method for generating stable and uniformly sized SNPs. Finally, we have demonstrated that these SNPs are active against pathogenic fungi, such as Candida and Cryptococcus .

ABSTRACT

En los últimos años, la evolución en el desarrollo de productos elaborados a partir de nanotecnología ha experimentado un espectacular crecimiento. En particular, las nanopartículas de oro han despertado gran interés en los sectores biomédico y alimentario, donde se ha descrito su utilización en el tratamiento frente al cáncer o como parte integrante de envases resistentes a la abrasión, con propiedades antimicrobianas. Por tanto, se cree que la exposición humana a las nanopartículas de oro aumentará considerablemente en los próximos años, pudiendo tener esto repercusiones sobre la salud. En este marco, el estudio de la toxicología de las nanopartículas ha revelado que su toxicidad depende de multitud de factores. Además, en la bibliografía hay cierta controversia en torno a los posibles efectos citotóxicos inducidos por las nanopartículas de oro. Diversos estudios de exposición in vitro han destacado su inocuidad en algunas líneas celulares, mientras que otros trabajos demostraron respuesta citotóxica. La siguiente revisión tiene por objeto describir las propiedades más relevantes de las nanopartículas de oro considerando sus potenciales aplicaciones en medicina y en la industria de los alimentos, así como examinar su posible toxicidad, con especial énfasis en los estudios de citotoxicidad in vitro disponibles hasta el momento.


In the recent years, the development of nanotechnology-based products has experienced a spectacular growth. Especially, gold nanoparticles have awoken a great interest in the biomedical and food sector, where their applications in cancer treatment as well as their incorporation in abrasion resistant and antimicrobial packaging have been described. Therefore, it is believed that human exposure to gold nanoparticles will increase considerably in the next few years, which may arise possible human health hazards. Hence, toxicology studies on nanoparticles revealed that their toxicity depends on various factors. Furthermore, there is some controversy regarding to gold nanoparticle-induced cytotoxicity. Several in vitro studies have reported that gold nanoparticles are innocuous, while some investigations have demonstrated a cytotoxic response after the exposure to these. The aim of this review is to describe the most relevant properties of gold nanoparticles according to their possible applications in medicine and in food industry, as well as to provide information about their possible toxic effects, taking into account the cytotoxic in vitro studies published at present.

ABSTRACT

PURPOSE: To investigate the results of the healing process on surgical wounds in the back of Wistar rats using nanocristaline and ionic silver dressing. METHODS: Sixty rats Wistar were submitted to surgical wounds with punch of 8mm in diameter. In 30 animals (groups PN - nanocristaline and AD - control) two surgical wounds were done diametrically opposite on the upper back side. On the right side was used nanocristaline (PN) silver dressing and on the left side, distilled water dressing (AD). On the other group of 30 rats, only one wound was made with the punch, on the right side, and was used ionic silver dressing. So, the groups were divided into three subgroups, according to the day of death (7th, 14th and 21st day). In each of these days the wounds diameter were measured to evaluate the wound contraction. Microscopic data were analyzed using the H&E staining to verify the inflammatory process and neovascularization. The Masson trichrome staining was used to verify the fibrosis. RESULTS: Macroscopically only the subgroup of 21st day showed statistical significance; between the groups AD and PI inflammatory process appeared in the 7th day subgroup in 90% of the cases. In neovascularization there was statistical significance between the groups PN and AD in the subgroup of 7th day. Fibrosis did not show statistical significance in the studied groups. CONCLUSIONS: In relation to wound contraction, PN and PI groups showed better results than the AD group. In regard to histological analysis, H&E staining showed that there was presence of inflammation in all groups, and at the end, the control group (AD) on 7th day, was superior to PN and PI groups. In relationship to fibrosis, no differences were obtained among groups.


OBJETIVO: Investigar os resultados da cicatrização de feridas cirúrgicas em dorso de ratos, utilizando curativos de prata nanocristalina e iônica. MÉTODOS: Sessenta ratos Wistar foram submetidos à feridas cirúrgicas com punch de 8mm de diâmetro. Foram confeccionadas duas lesões diametralmente opostas nos animais dos grupos prata nanocristalina (PN) e controle água destilada (AD). Na lesão do lado direito foi utilizado curativo com prata nanocristalina e na do lado esquerdo curativo com água destilada. No outro grupo de 30 ratos, apenas foi realizada uma lesão com o punch no lado esquerdo e curativo com prata iônica. Os grupos foram divididos em subgrupos conforme o dia da morte (7º, 14º e 21º dias), o que caracterizou a existência de três subgrupos, nos quais foram tomadas as medidas do centro das lesões para estudar macroscopicamente a contração da ferida Microscopicamente foi utilizada a coloração H&E, através da qual foi observado o processo inflamatório e a neovascularização. Com a coloração Tricômio de Masson, foi estudada a fibrose. RESULTADOS: Macroscopicamente apenas o subgrupo 21 dias apresentou significância estatística entre os grupos AD e prata iônica (PI), porém quando comparados os dias de avaliação, dois a dois, dentro de cada tratamento, todos os subgrupos apresentaram significância estatística (p<0,05). A variável intensidade da inflamação apresentou-se de forma acentuada no subgrupo sete dias em 90% dos casos, entre os grupos AD e PI. Na variável neovascularização houve significância estatística entre os grupos PN e AD no subgrupo sete dias. A variável fibrose não apresentou significância estatística nos subgrupos estudados. CONCLUSÕES: Em relação à contração da ferida, os grupos PN e PI apresentaram resultados superiores ao grupo AD. Com relação à análise histológica, na coloração H&E observou-se que houve presença de processo inflamatório nos grupos estudados sendo que, ao final, o grupo controle (AD) no 7º dia mostrou-se superior aos grupos PN e PI. Com relação à variável fibrose, não houve diferenças entre os grupos.

ABSTRACT

BACKGROUND: Methicillin-resistant Staphylococcus aureus (MRSA) may cause infections during wound dressing. We aimed to compare the antibacterial activities and wound-healing effects of commercially available silver-coated or silver-impregnated wound dressings on MRSA-infected wounds. METHODS: Full-thickness skin defects were made on the back of rats (N=108) and were infected with MRSA. The rats were divided into the following 6 groups according to the dressing used for the wounds: nanocrystalline silver (Acticoat(R)), silver carboxymethylcellulose (Aquacel(R)-Ag), silver sulfadiazine (Medifoam silver(R)), nanocrystalline silver (PolyMem silver(R)), silver sulfadiazine (Ilvadon(R)), and 10% povidone iodide (Betadine(R)). We analyzed the wound sizes, histological findings, and bacterial colony counts for the groups. We also inoculated the silver materials on Mueller-Hinton agar plates containing MRSA and compared the inhibition zones in the agar plates. RESULTS: The order of the rate of wound-size decrease was Acticoat(R)>Aquacel(R)-Ag>PolyMem silver(R)>Medifoam silver(R)>Ilvadon(R)>Betadine(R). The histological findings revealed that the Acticoat(R) showed more reepithelialization and granulation tissue formation and less inflammatory cell infiltration than the other materials. The order of the time required for wound healing was Acticoat(R)>Aquacel (R)-Ag>PolyMem silver(R)>Ilvadon(R)>Medifoam silver(R)>Betadine(R). The bacterial colony counts reduced in all the groups, except in the Medifoam silver(R) group. The order of the size of the inhibition zone was Acticoat(R)>Aquacel(R)-Ag>Ilvadon(R)>PolyMem silver(R)>Betadine(R)>Medifoam silver(R). CONCLUSIONS: Silver-coated or silver-impregnated wound dressings can be used for treating MRSAinfected wounds. Considering its superior efficacy in comparison to the efficacies of other silver-coated or silver-impregnated wound dressings, Acticoat(R) should be preferentially used for the treatment of MRSA-infected skin wounds.
