RESUMEN

Combined pulsed-field gradient, shaped pulses, and product operator calculations (algorithm), two-dimensional 1 H-31 P hetero-nuclear multiple quantum coherence ( HMQC) pulse sequence was established with the aid of C language. The result showed that coherent transfer path selection could be achieved by adjusting the ratio of two gradient strength ( g1 , g2 ) . The best ratio of g1 and g2 was -5:4 . The HMQC method based on the above sequence can be used for identifying the alkyl groups coupling with phosphorous nucleus. For the different alkyl groups, the sensitivity of this method decreased in the order methyl>isopropyl>ethyl>propyl. The limit of detection was about 10 mg/L. This method can be directly used for the screening and identification of phosphorus chemical warfare agents and related compounds.

RESUMEN

Objective To study the effects of Grape Procyanidins(GPC)on hemorheology in long distance runner.Method In vitro study: 22 long distance runners were divided into two groups randomly,the experimental group and control group.With ten days period,the experimental group was given GPC 200 mg per day;while the control group were given the capsule of starch 200 mg per day.In vivo study: the vein blood samples were taken from 5 long distance runners and every example was divided into five parts,and then treated with different concentrations of GPC or H2O2.Items of the hemorheology such as Er,HCT,Eb,Ep,PFC and VAI were tested both in vivo and vitro before and after the study.Results in vitro study: all items of the experimental group showed significant decline at then end of the study than those before the study.The contents of Er,Eb and Ep before the study(6.830?0.164,4.145?0.177,1.647?0.020,respectively) were significantly higher than those after the study(6.759?0.158,4.088?0.173,1.621?0.013,respectively)(P

RESUMEN

Objective:To study the effect of grape procyanidin (GPC) on the levels of plasma?-granule membrance protein 140 (GMP140) and thromboxane B2 (TXB2 ) in rats. Method:Wistar rats fed by high fat diet (HFD) were given GPC i.g. at different doses for 6 w, and the levels of plasma GMP140 and TXB2 were examined by radioimmunoassay (RIA). Result:Plasma GMP140 and TXB2 levels of high dose GPC were respectively 7.89?1.02ng/ml, 131.71?42.13 pg/ml, significantly lower than those of HFD group. Conclusion:GPC could inhibit the increase of plasma GMP140 and TXB2 induced by high fat diet in rats.

RESUMEN

Objective To study the effect of Konjac polysaccharide (KP) on motilin (MOT) and somatostatin (SS) of gastrointestinal tissue in mice. Methods The mice were divided into four groups randomly. Normal control group (NC) was given normal feed. High, middle, low dose KP treated groups (KPH,KPM,KPL)were given KP 0.8, 0.4, 0.2g per 100g normal feed. The level of motilin and somatostatin of gastrointestinal. tissue was determined simultaneously by radioimmunassay, and blood glucose was determined by blood glucose analyzer after 20 d. Body weight, adipose tissue and wet weight of stool were also examined. Results The level of motilin of stools of KPH group was 8.26?0.53ng/g ,which increased compared with NC group.Somatostatin was 207.32?21.89 ng/g ,which decreased compared with NC group. Objective To investigate the inhibitive effect of Konjac polysaccharide(KP) on intestinal absorption function of mice and its mechanism.Method Mice were divided into normal control(N),high fat(HF),and high,middle,low dose KP(KPH,KPM,KPL) treated groups.Intestinal mucosa Na+-K+-ATPase activity was determined by UV spectrophotometry,blood glucose was determined by fast meter after twenty days.Body weight,fecal wet weight and dry weight were also examined.Results Na+-K+-ATPase activity of HF and KPH groups were 16.2?1.48 and 11.2?1.10 ?mol Pi/(mg prot?h),respectively.Body weight and postprandial blood glucose were 34.3?2.07 g,7.5?1.15 mmol/L and 28.1?1.95g,4.8?0.73 mmol/L,respectively.The differences between two groups were significant(P

RESUMEN

Objective To investigate the effect of grape procyanidins (GPC) and the regulation of estrogen receptor(ER)on proliferation of human breast cancer(MCF-7)cells. Method MCF-7 cells were cultured with GPC,and divided into six groups: tumor control,high,middle and low dose GPC treated,estrogen antagonist ICI 182. 780 (ICI) control,ICI and GPC combined treated. The proliferation activity level of MCF-7 cells was determined by MTT assay,and the levels of the proliferating cell nuclear antigen (PCNA) and Bax protein were measured by immunohistochemistry method. Results Compared with tumor control group,the level of proliferation activity and the expression of PCNA were significantly decreased in high dose GPC group,while the expression of Bax protein was increased significantly. Compared with high dose GPC group,both the proliferation activity and the PCNA expression were increased,and Bax protein expression decreased significantly in ICI and GPC combined treated group. Conclusion GPC can inhibit the proliferation and induce the apoptosis of human breast cancer cells effectively,and its mechanism is closely related to the regulation of ER.

Objective: To investigate the inhibitory effect of grape proanthocyanidins (GPC) on P38 mitogen-activated protein kinase(P38MAPK)pathway in mice with breast cancer . Methods: BALB/C mice inoculated with EMT-6 breast cancer cells were given 10, 100 and 200 mg/kg bw GPC po daily for 2 w. The expression levels of phosphorylated P38MAPK and matrix metalloproteinase-2(MMP-2)protein were detected by Western blot. Results: The levels of phosphorylation P38MAPK in tumor control group and 200 mg/kg GPC group were 1.16?0.18 和 0.58?0.12 respectively,and the levels of MMP-2 protein were 0.98 ?0.04 和 0.69?0.04 respectively. RESUMEN

Objective:To study the effects of polysaccharide from Lycium barbarum (LBP) on cell apoptosis and bc1-2 gene expression in irradiated mice. Methods:Forty eight Kunming mice were divided into three groups (LBP group, radiation group and normal group). LBP chow was prepared through adding 0.8%LBP to normal chow and was supplied to LBP group. Normal chow was supplied to normal group and radiation group. LBP group and radiation group were exposed to whole-body 60Co ?-rays at the dose of 0.084 Gy/day for 6 w, five times a week and the total dose was 2.52Gy. Then the micronucleus frequency of polychromatic erythrocytes(MF),chromosome and sperm aberration frequency, caspase-3 mRNA expression, cell apoptosis and bcl-2 gene expression were detected. Results:LBP could significantly lower MF, chromosome and sperm aberration frequency, and cell apoptosis , and it could increase the proliferation activity of bone marrow cells and bc1-2 gene expression in irradiated mice and decrease caspase-3 mRNA expression.Conclusion:The radioprotective effect of LBP is related to regulation of cell apoptosis and bc1-2 gene expression.

Objective:The protective effects of grape procyanidins (GPC) on free radical induced endothelial cell damage were investigated. Methods:Human umbilical vein vascular endothelial cells (ECV304) were cultured in GPC of different doses. Oxidative injury was caused by Fenton system, and MTT method was used to test cell proliferation when cells were cultured 12, 24, 36, 48 h, respectively, MDA and SOD in cells were also determinated 30 min after Fenton system was added. Objective: To study the effect of grape procyanidin (GPC) on N-nitroso compounds-induced expression of TNF- mRNA and TGF-?1 mRNA in hepatic cells. Methods: NaNO2 was infused into Wistar rats by 50 mg/kg bw to induce mutation. GPC was infused by 100mg/kg bw and 10mg/kg bw respectively through mouth into the rats of two experimental groups. After 8 w situ hybridization was applied to measure the expression level of TNF-?mRNA and TGF-?1 mRNA in hepatic cells. Results: The expression rate of TNF-醡RNA and TGF-?1 mRNA of the hepatic cells in positive control group was 30.23 % and 19.47 % respectively, while that of high-dose GPC group was 11.94 % and 6.96 % respectively .The differences were statistically significant (P
