RESUMEN

Objective The aim of this study was to draw single-cell transcriptome profiles of high-grade serous ovarian cancer(HGSOC),borderline ovarian cancer(OC),and normal ovaries in order to identify biomarkers that can diagnose and predict the prognosis of OC.Methods The differentially expressed genes between HGSOC,borderline OC,and normal ovarian tissues were ana-lyzed using single-cell data sequenced(SRA database:PRJNA756768).The cell subsets associated with tumor progression were screened by functional enrichment,cell communication between different subsets was analyzed by Cellchat,and cell differentiation traj-ectories were explored by pseudotime analysis to finally determine the subsets most relevant to tumor progression.Combined with OC transcriptome data of OC from the Cancer Genome Atlas(TCGA)with patient prognosis,biomarkers for diagnosing and predicting sur-vival of OC patients were ultimately screened.Results After using t-distribution stochastic neighbor embedding(t-SNE)for di-mensionality reduction,nine cell subpopulations were obtained:endothelial cells,myeloid cells,fibroblasts,T cells,stromal cells,B cells,and 3 epithelial cell subpopulations(C1,C4,and C7).Further analysis revealed that copy number variation(CNV)in the C4 group had the highest score in HGSOC,higher than those of borderline OC and normal ovaries,and was negatively correlated with prognosis.DMKN was a key marker gene in this group.Transcriptome analysis of OC in the TCGA database showed a close correlation between DMKN and poor prognosis(P=0.026),and the diagnostic efficacy of DMKN for OC was significant(A UC=0.906).Con-clusion This study is based on single-cell sequencing data to screen for DMKN,which can effectively diagnose and predict the prognosis of OC.This study provides new ideas for the diagnosis and prognosis prediction of OC.

Objective:The cluster analysis method was applied to evaluate the scientific research performance evaluation index system of departments in an obstetrics and gynecology hospital, and analyze the weaknesses of scientific research work of various departments, to provide a basis for the improvement of scientific research strength of departments.Methods:On the basis of the scientific research performance evaluation index system of departments in the obstetrics and gynecology hospital, the indicators and weights of the system were optimized through expert consultation, and the scientific research values from 2019 to 2021 were brought into the optimized scientific research performance evaluation index system to calculate the scientific research scores of each department, and then the cluster analysis method was applied to evaluate the index system.Results:Before and after the optimization of the scientific research performance evaluation index system of departments, the conformity with the discipline classification was 76.00% and 96.67% respectively ( P=0.039). In total of 30 departments were clustered into 4 categories: excellent (7), good (7), medium (4), and concerned (12). The average score of the total scientific research performance evaluation indicators of the 4 categories of departments was 15.022. The highest average score was for papers and monographs, and the lowest was for awards. Conclusions:This study applied the cluster analysis method to evaluate the scientific research performance evaluation index system of departments in the obstetrics and gynecology hospital and replaced quantitative indicators with quality indicators. It will optimize and improve the hospital hierarchical management methods, provide data support and classified guidance for the scientific research development of different categories of departments, and provide a reference basis for hospitals to formulate scientific research management policies such as achievement transformation, award, industry standard guidelines, etc.

NUF2 is responsible for the attachment of kinetochore-microtubules and proper chromosome segregation during mitosis. NUF2 is highly expressed in hepatocellular carcinoma, pancreatic cancer, esophageal cancer, non-small cell lung cancer, breast cancer and other tumor tissues and cells, and can be used as prognostic markers. Further clarifying the relationship between NUF2 and tumor prognosis can provide help for the application of NUF2 in prognostic assessment of cancers.

Objective:Delphi method and weighted average method are used to establish the evaluation index system of basic and clinical scientific research projects respectively, to provide support for screening high-quality projects, to avoid the disadvantages of " one size fits all" , and promote the transformation of project management from quantity and restatement to quality and results, in order to release the innovative vitality of talents and produce more high-quality results.Methods:Using the methods of literature review and expert interview, form the questionnaire of evaluation indicators for the conclusion of basic and clinical scientific research projects, then carry out two rounds of expert consultation with Delphi method, evaluate the authority of experts with authority coefficient, form the second round of questionnaire after analysis and discussion according to the results from the first round of expert consultation, screen and determine the evaluation indicators again in the same way. After two rounds of discussion, the evaluation index system of basic and clinical scientific research projects was developed, and the weighted average method and product method were used to calculate the weight and combination weight.Results:After two rounds of consultation, the evaluation index system for the conclusion of basic scientific research projects was finally established, with 2 first-class indicators, 12 second-class indicators and 33 third-class indicators, and the evaluation index system for the conclusion of clinical scientific research projects had 2 first-class indicators, 10 second-class indicators and 33 third-class indicators. In the second round, the authority coefficient of basic and clinical experts was 0.8, indicating that the authority of experts in this study was high. 29 basic and clinical projects concluded from 2018 to 2020 were selected to verify the index system.Conclusions:The index system formed in this study will be used to classify and evaluate the conclusion evaluation of basic scientific research projects and clinical scientific research projects. In the next step, the weight of the index system will be adjusted to make it more realistic and highlight high-quality achievements, guide the project leader to pay more attention to quality and results, and conduct follow-up evaluation at different stages.

Objective:To explore the competence characters of hospital young scientists and build a competency model.Methods:The model characters were screened by literature review, behavioral event interview, expert consultation and questionnaire survey. The competency characters were extracted by exploratory factor analysis.Results:The competency characters model includes 5 factors and 34 characters: comprehensive accomplishment, practical ability, personal trait, professional skill and knowledge quality. The Cronbach's α coefficient of the model was 0.980 and the Cronbach's α coefficient of the 5 factors ranged from 0.832 to 0.964, with a split-half reliability of 0.922. The content validity index of the scale was 0.977, and the content validity index of each entry ranged from 0.857 to 1.000. The value of KMO was 0.944.Conclusions:The competency model of hospital young scientists has high stability, and provides reference for young scientists' ability and quality, which will provide the basis for optimizing the training system and training strategy of hospital scientists.

Objective:To understand the cognition of scientific research integrity of the authors of scientific and technological papers in a grade A tertiary hospital, the necessity and relevant needs of carrying out scientific research integrity construction, and provide a reference for improving the construction of relevant scientific research integrity system in the hospital.Methods:A questionnaire was used to survey 746 people who had published scientific and technological papers in a grade A tertiary hospital from 2016 to 2021, and the influencing factors of scientific research integrity cognition and construction were analyzed via a generalized linear model.Results:The overall awareness rate of scientific research integrity cognition of scientific papers was 76.72%, and the overall necessity rate of scientific research integrity construction was 77.75%. Generalized linear model analysis showed that other degrees refer to a doctor's degree, associate senior title, intermediate title, refer to junior title, or no title were negatively correlated with the cognition of scientific research integrity ( P<0.05), other ages refer to ≤30 years old, middle-level and above positions refer to no position, postgraduate mentor refer to non-postgraduate mentor, senior title refer to junior title or no title, postgraduate, nursing, administrative and research personnel refer to medical technology and pharmacy personnel, and presiding projects refer to no presiding project experience were positively related to scientific research integrity cognition ( P<0.05). Bachelor′s degree and no degree refer to doctor′s degree, associate senior and intermediate titles refer to junior title or no title were negatively related to the necessity of scientific research integrity construction ( P<0.05), middle level and above positions refer to no position, postgraduate, nursing and research personnel refer to medical technology and pharmacy personnel, were positively related to the necessity of scientific research integrity construction ( P<0.05). Behaviors with less than 60% cognition of scientific research integrity included the ethical problems of artificial intelligence, the ethical problems of stem cells, the problems of human genetic resources, and the definition of unauthorized human living drug and medical technology experiments. Conclusions:The hospital will take publicity and education as the basis, system norms as the criteria, daily supervision as the starting point, evaluation system as the core, carry forward the spirit of scientists, improve and implement the construction of the hospital's scientific research integrity system and supervision system, strengthen the integrity management of the whole process of scientific research activities, and guide medical personnel to practice excellent scientific research style and style of study.

Objective:To explore the mechanism of glycine dehydrogenase (GLDC) regulating the proliferation and apoptosis of ovarian cancer cells through PI3K/Akt/mTOR pathway.Methods:RNA interference method was used to silence the expression of GLDC in ovarian cancer cell lines HEY and SK-OV-3. The HEY and SK-OV-3 cells were divided into si-control group (transfected with siRNA-control), si-GLDC#1 group (trans-fected with siRNA-GLDC#1) and si-GLDC#2 group (transfected with siRNA-GLDC#2). The expression level of GLDC and the protein phosphorylation level of PI3K/Akt/mTOR were detected by Western blotting. Cell proli-feration, migration and apoptosis were detected by CCK-8 method, Transwell chamber test, cell scratch test and flow cytometry.Results:The relative expression levels of GLDC in the si-control group, si-GLDC#1 group amd si-GLDC#2 group of HEY cells were 1.00±0.01, 0.68±0.10, 0.80±0.08, and there was a statistically significant difference ( F=13.80, P=0.006). The relative expression levels of GLDC in the si-control group, si-GLDC#1 group and si-GLDC#2 group of SK-OV-3 cells were 1.02±0.01, 0.58±0.17, 0.60±0.25, and there was a statistically significant difference ( F=6.08, P=0.036). The absorbance ( A) values in the si-control group, si-GLDC#1 group and si-GLDC#2 group of HEY cells were 1.04±0.03, 0.91±0.02, 0.82±0.01 at 24 h after transfection, 1.53±0.13, 1.30±0.03, 1.29±0.07 at 48 h after transfection, 1.44±0.08, 1.25±0.01, 1.15±0.03 at 72 h after transfection, and there were statistically significant differences ( F=83.14, P<0.001; F=8.96, P=0.007; F=29.55, P<0.001). Further pairwise comparison showed that the proliferation abilities of the si-GLDC#1 and si-GLDC#2 group at 24, 48 and 72 h were significantly lower than those of the si-control group (all P<0.05). In HEY cells, the migration numbers of cells in the si-control group, si-GLDC#1 group and si-GLDC#2 group were 57.33±6.43, 27.67±5.13 and 30.67±2.31, and there was a statistically significantly difference ( F=32.88, P=0.001). The migration numbers of cells in the si-GLDC#1 group and si-GLDC#2 group were significantly lower than that in the si-control group ( P<0.001; P=0.001). Similar results were also observed in SK-OV-3 cells. In SK-OV-3 cells, the scratch healing rates in the si-control group, si-GLDC#1 group and si-GLDC#2 group were (51.27±1.59)%, (26.35±2.94)% and (26.34±7.69)%, and there was a statistically significant difference ( F=26.54, P=0.001). The scratch healing rates in the si-GLDC#1 group and si-GLDC#2 group were significantly lower than that in the si-control group (both P=0.001). In HEY cells, the apoptosis rates in the si-control group, si-GLDC#1 group and si-GLDC#2 group were (7.11±0.82)%, (10.44±1.50)%, (17.39±1.55)%, and there was a statistically significantly difference ( F=46.52, P<0.001). The apoptosis rates in the si-GLDC#1 group and si-GLDC#2 group were significantly higher than that in the si-control group ( P=0.022; P<0.001). Similar results were also observed in SK-OV-3 cells. In HEY cells, there was no significant difference in total PI3K protein in the si-control group, si-GLDC#1 group and si-GLDC#2 group ( F=0.54, P=0.631), but there were significant differences in pAkt/Akt and pmTOR/mTOR levels ( F=22.14, P=0.016; F=10.57, P=0.044). The pAkt/Akt and pmTOR/mTOR levels in the si-GLDC#1 group and si-GLDC#2 group were significantly lower than those in the si-control group ( P=0.015, P=0.008; P=0.039, P=0.023). Similar results were also observed in SK-OV-3 cells. Conclusion:In ovarian cancer cells, GLDC silencing can inhibit cell proliferation and promote apoptosis by inhibiting the PI3K/Akt/mTOR pathway.

Objective:This article aims to sort out the current situation and problems of horizontal scientific research projects in a tertiary hospital, so as to improve the management of horizontal scientific research projects, and to mobilize the enthusiasm of researchers to undertake horizontal scientific research projects, and give full play to their characteristics of interdisciplinary integration, transfer and transformation of scientific and technological achievements.Methods:A total number of 129 horizontal projects undertaken by Beijing Obstetrics and Gynecology Hospital, Capital Medical University from 2010 to 2019 were reviewed.The comparison of management policies and risk prevention of horizontal scientific research projects before and after 2017 was also analyzed.The existing problems were analyzed and relevant management suggestions were put forward.Results:From 2017 to 2019 the hospital undertook 18.7 horizontal research projects per year, which was 1.8 times of that 2010 to 2016.The hospital strengthened the cooperation with foundations, companies and foreign countries, carried out substantive scientific research, standardized the budget, leased or borrowed equipment, and built the joint laboratories by medical research and enterprise, and carried out risk prevention in human genetic resources and biological security.At present, there are still some problems, such as the lack of relevant policy basis for management, the management of project approval and conclusion is not in place, the conversion rate of achievements is low, and the horizontal scientific research projects of platform construction are need to be constructed.Conclusions:Our hospital will improve the relevant system of horizontal scientific research projects management, incorporate the horizontal scientific research projects into the position title evaluation, performance evaluation and reward, build various platforms through multiple channels, promote the interdisciplinary integration, and give full play to the characteristics of horizontal scientific research projects in promoting the transfer and transformation of scientific and technological achievements, so as to promote the discipline construction and scientific and technological innovation of the hospital, and accelerate the transformation of the hospital into innovative and research-oriented hospital.

Objective:To analyze the current situation of scientific research in hospitals in China based on the indicators and results of Science and Technology Evaluation Metrics in Chinese hospitals, and propose possible countermeasures for scientific research development.Methods:The external and internal environment of scientific research in a Grade-A tertiary obstetrics and gynecology hospital was discussed through the results of Science and Technology Evaluation Metrics and situation analysis.Results:The external opportunity of the hospital is that the national and municipal government lead the development direction of science and technology, the hospital leaders attach great importance to the development direction of the hospital and have a clear direction for the development of the hospital; the challenge is that the new generation of information technology puts forward the requirements of cross integration of scientific research work, the flow of high-level talents is accelerated, and there is a significant gap with the similar hospitals in the field of obstetrics and gynecology. The internal advantages are embodied in the following points: the advantages of specialties are obvious, the output of scientific research results has increased year by year after the implementation of the scientific research management of hospital departments, the training and introduction of talents are equally important; the disadvantages are reflected in the following points: lack of basic research and scientific research platforms, lack of high-end scientific and clinical research talents, insufficient academic influence, lack of national-level scientific research projects, insufficient high-level scientific research results, and low achievement conversion rate.Conclusions:The hospital should improve performance assessment, build research platform and achievement transformation platform, strengthen interdisciplinary integration and personnel training, and promote the fine management of high-level achievements, so as to improve the scientific and technological output, academic influence and scientific and technological conditions of the hospital.

Objective:Data envelopment analysis (DEA) was applied to evaluate the evaluation index system of scientific research performance of hospital departments.Methods:Based on the three-level index assignment of the constructed evaluation index system of scientific research performance of hospital departments, the relevant data of departments are calculated. Scientific research projects, talent team and scientific research platform are taken as input indexes, papers, awards, patents, published monographs/papers, professional standards and guidelines, as well as achievements transformation are taken as output indexes, and departments are taken as evaluation units. Data envelopment analysis software is used to evaluate departments Results of scientific research performance evaluation, redundant input and insufficient output.Results:The average comprehensive technical efficiency of 25 departments is 0.404 8, the average pure technical efficiency is 0.837 1, and the average scale efficiency is 0.428 3. Among them, 7 departments are generally efficiency, 17 departments obtained pure technical efficiency, 7 departments obtained scale efficiency.Conclusions:The analysis results can provide a reference for the scientific research development of the department. On the basis of building a reasonable talent echelon and assure the sufficient scientific research time, the hospital further optimizes and improves the scientific research performance evaluation index system and hierarchical management method of the Department.

Objective:To establish a scientific research performance evaluation index system for departments by applying Delphi method and analytic hierarchy process.Methods:A questionnaire composed of 10 first-level indicators, 36 second-level indicators and 95 third-level indicators was developed according to literature review and expert interview. Then two rounds of expert consultation were conducted by adopting Delphi method, the authority of experts was evaluated by the authority coefficient. Based on the analysis and discussion of the findings of the first round of expert consultation, an updated version questionnaire was developed, the evaluation indicators were re-selected and determined by the same way. After two rounds of discussions, the scientific research performance evaluation index system of the hospital department was finally developed. Finally, the analytic hierarchy process and product method were used to calculate the weights and combined weights, respectively.Results:After two rounds of consultation, 9 first-level indicators, 28 second-level indicators and 67 third-level indicators were finally established in the scientific research performance evaluation index system of hospital departments. The expert authority is 0.79, which shows that this research has a higher authority. From the standpoint of significance test, the P values are all less than 0.01, indicating that the consulting experts meet the requirements for the consistency of the index scores, and the results are reliable. Conclusions:An effective index system for scientific research performance evaluation of departments has been established in this research, however, it still needs further validation in practice.. In the future, it will need to be combined with grading standards and methods, and subjective assessment by experts to test the scientificity and feasibility of the evaluation index system to assure continuing improvement.

Keratins are cytoskeleton proteins, which are involved in apoptotic resistance, growth and migration of cancer cells. The levels of some keratins are elevated in the serum of patients with cancers, or highly expressed in cancer tissues, and they are widely used in the diagnosis of tumor in clinic. The expressions of some keratins are negatively correlated with the prognosis of patients with cancers, and can be used as prognostic markers. Further clarifying the diagnostic and prognostic value of keratins combined with other markers detection can provide theoretical basis for the application of keratins in diagnosis and prognostic assessment of cancers.

Objective:To lay a foundation for the continuous improvement of the birth cohort management in the future, and provides references for other hospitals in establishing the birth cohort through sharing the practice, experience and reflection of the establishment and management of the birth cohort of 500, 000 pregnant women.Methods:The present situation and data of the establishment and management of the birth cohort of 500, 000 pregnant women were summarized to describing the practice, experience and reflection.Results:The specific work regarding the establishment and management of the cohort include personnel investment, standards development, implementation process and quality control. Three phases of the management experiences were summarized which including explorative process implementation; standardize project implementation, and quality improvement along with the fast development of the birth cohort. Generally, the attention of leadership was one of the most important factors for the establishment and management of birth cohort in each research site.Conclusions:It is important that comprehensive considerations of factors including the personnel, standards, practical management process, quality control and the attention of leadership plays important role in improving the quality and efficiency of birth cohort establishment and management.

Objective:To understand the cognition of scientific research integrity of applicants of the National Natural Science Foundation of China (NSFC) and the necessity of developing scientific research integrity construction.Methods:Questionnaire survey was used to collect information from 121 applicants who had applied for the NSFC from 2016 to 2020, collected data were statistically analyzed.Results:The cognitive average score of scientific research integrity knowledge of NSFC applicants was (287.03±49.45) points, and the overall awareness rate was 80.44%. The average score of the necessity of scientific research integrity construction work was (20.71±3.59) points, and the overall necessity rate 87.27%; educational background, position title, and experience as principle investigator are factors that affect the perception of integrity, while educational background, position, job category, and research management experience of a project are factors that affect the necessity of scientific research integrity construction. Behaviors with less than 70% of scientific research integrity behaviors include stem cell ethical issues, genetic technology ethical issues, human genetic resources issues, conditions for secondary publication, and use of official cards for settlement.Conclusions:The hospital should strengthen the scientific research integrity knowledge training of NSFC and other project applicants. On the basis of giving full play to the supervision role of relying units, the hospital will improve the relevant system of scientific research integrity, improve the construction of scientific research integrity at the same time, carry out classified evaluation, and put an end to scientific research misconduct from the source.

Objective To investigate the expression of Fascin in early-stage NSCLC, evaluate the relevance between Fascin expression level and prognosis.Methods The immunohistochemistry method was used to assess the expression of Fas-cin in 111 lung cancer FFPE tissues with stage Ⅰ and Ⅱ NSCLC.The relationship between Fascin expression and the clinico-pathological characteristics was analyzed.The prognostic significance of Fascin expression was evaluated with Kaplan-Meier sur-vival analysis.Results In the early-stage of NSCLC, the positive rate of Fascin expression was 64.8%, no expression in the paracarcinoma tissue.The positive rate of squamous cell carcinoma was 78.7% and was significantly higher than that in adeno-carcinoma 48.0%(P<0.01).Whether in squamous cell carcinoma or adenocarcinoma group, the expression of Fascin was correlated significantly with lymph node metastasis tumor stages and DFS(P<0.05).And the positive expression of Fascin was an independent risk factor of poor prognosis for patient with NSCLC .Conclusion Fascin is expected to be a biomarker for the prognosis of patients with early-stage NSCLC.

Objective To analyze current situation of incentive mechanism and investigate the countermeasures.Provide the evidence for improving the incentive mechanism of scientific research at the hospital.Methods Questionnaire survey was conducted to investigate current situation and countermeasures of incentive mechanism of scientific research.Database was building using EpiData3.1,and was performed by descriptive analysis.Results 194 questionnaires were available.For the current incentive mechanism,86.5% respondents thought the content is comprehensive,The over 50 years old,bachelor or below,and clinical examination group thought the content were less comprehensive.Respectively,81%,82.5%,76.2% of respondents thought stimulation on achievements has positive effect to research,team work,and individual accounted for.The over 41 years old college or bellow,clinical treatment group felt less effect.For improving measures of incentive mechanism,the top three positions of contents were technological reward,high level project and SCI paper;and incentive methods were promotion,material rewards and training.Conclusions Incentive mechanism can promote scientific research to some extent.To further promote scientific research development,individual incentive measures need to be taken,the contents need to be expanded,variety incentive methods need to be used.

As a way of gene modification,alternative splicing is an important factor of eukaryotic gene expression and regulation.It makes various transcripts from one protein-coding gene,and greatly extends the genetic information.Alternative splicing of pre-messenger RNA plays an important role in tumor cells.By alter-native splicing,some important genes can generate splicing variants different from those in normal cells.The existence of tumor-specific splicing variants leads to the occurrence and progression of tumor.Therefore,explo-ration on the alternative splicing of tumor-associated genes may be of great significance in tumor diagnosis and treatment.
