RESUMEN

Objective To explore the mechanisms of synergic action of commonly-used herb pair of Radix Sophorae Flavescentis (Kushen, KS) and Fructus Cnidii (Shechuangzi, SCZ) for the treatment of pruritus. Methods We predicted and analyzed the potential targets of KS and SCZ based on network pharmacology method, and then established Chinese herbs-compound skeletons-targets networks to reveal anti-pruritus targets, predicting targets, and the interaction of KS and SCZ, as well as the components of the herb pair. The RAW264.7 inflammatory cell model was established to observe the synergistic anti-inflammatory activity of KS and SCZ. Results KS and SCZ had possible synergic action on the pruritus-related targets such as histamine receptors, cannabinoid receptors and proteinase-activated receptor 2. Additionally, KS and SCZ probably had synergic regulation of inflammation-related pathway (Toll -like receptor signaling pathway , chemokine signaling and Fc epsilon RI signaling pathway) and nerve-related pathway(neurotrophin signaling pathway) during the treatment of pruritus. Flavones from KS and coumarin of SCZ had various synergistic anti-inflammatory activities(P<0.05), indicating that they played an important role in inhibiting pruritus induced by inflammation. Conclusion The method may reveal the molecular mechanism of KS and SCZ in inhibiting pruritus, which is important for the modernization of Chinese medicine and new drug development.

RESUMEN

Objective To establish the method for content determination of total coumarins in Fructus Cnidii.Methods The content of the total coumarins was determined by UV spectrophotometry.Results The optimal detective wavelength was 320 nm for the total coumarins.Satisfactroy linear relationship with the absorbability was achieved when the mass concentration of osthole was within the range of 2~12 ?g/mL(r=0.999 5).The average recovery rate was 99.39%(RSD=2.67%,n=6).Conclusion The method is simple,accurate and sensitive,and it can be used for the quality control of Fructus cnidii and its preparation.

RESUMEN

Objective To establish the method for determining the content of Fructus Cnidii in Dafeng wan by HPLC. Methods Diamonsil ODS1 C18 column was used with CH3OH-0.1 mol/L (NH4)2SO4 (70︰ 30) [adjust pH to 2.5 by sulphuric acid solution (1→5)] as the mobile phase. The detection wavelength was set at 322 nm and flow rate was 1.0 mL/min. Results The calibration curve was linear at the range of 0.101~0.808 ?g for the Fructus Cnidii and linear equation was Y =9.964 406?10-3X -1.355 684?10-1, r=0.999 5. The average recovery was 98.4% and RSD was 1.40% (n=6). Conclusion This method was simple and accurate, with good specificity and repeatability, which provides scientific quantitative analysis method of Fructus Cnidii in Dafeng wan.

RESUMEN

Objective: To study the effects of TCFC on proliferation and osteogenic action of osteoblast in neonatal rat calvaria cultures in vitro. Methods: Osteoblasts were isolated from neonatal rat calvaria through trypsin and collagenase digestion. The proliferation and collagen synthesis were assayed by 3H TdR and 3H proline incorporation. The activity of ALP was measured by p nitrophenyl sodium phosphate assay. The ipriflavone was used as positive control drug.Results: TCFC significantly promoted bone cell proliferation, alkaline phosphatase activity and collage synthesis.Conclusion: TCFC had the anti osteoporosis action by promoting osteoblast proliferation, ALP activity and collage synthesis.

RESUMEN

Objective: To establish the determination method for osthole in Cnidii and Fuyan ling Capsule(Fructus Cnidii, Radix Sophorae Flavescentis, Herba Agrimoniae, Radix Stemmonae, etc.). Methods: RP HPLC was used with Alltima C 18 column (250mm?4.6mm,5?m), detection wavelength at 322nm, mobile phrase: methanol water (75∶25), flow rate: 1mL?min -1 . Results: Regression equation of Fuyanling capsules is Y =22957x-816, and r=0.9999 with repetition rate RSD 1.45%. Conclusion: This method is simple, easy and reliable.

RESUMEN

Objective: To establish the quality standard for Qiankun Liquid.Methods: Fructus Lycii、Fructus Cnidii、Radix Ginseng in Qiankun Liquid were identified by TLC, and the content of icariin was determined by HPLC.Results: Fructus Lycii、Fructus Cnidii、Radix Ginseng could be identified by TLC. Icariin showed a good linear relationship at a range of 0.126~1.008?g, r =0.9999. Conclusion: The method is accurate and can be used for the quality control of Qiankun Liquid.
