RESUMEN

Using observation of the gross and histological changes of skin, intestine, liver and spleen, the spleen index, the popliteal lymph node weight gaining, and the ~(125)I-UdRincorporation assay, the effects of GvHR in the pathogenesis of immunologically mediatedaplastic anemia in mice were studied. Rtsults showed that there were no gross and histological changes due to GvHR foundin skin, intestine, liver and spleen, and the spleen index decreased markedly, and therewere positive popliteal lymph node weight gaining and positive ~(125)I-UdR incorporationassay which showed a dose dependent corrlation. These results suggested that, transfusion of DBA/2 lymphocytes into lethally irradia-ted BALB/c mice could evoke GvHR, but on the other hand, the effects of GvHR werenot very important in the pathogenesis of immunologically mediated aplastic anemia inmice.

RESUMEN

The effects of aminophylline on xenogenic graft versus host reaction and the survival of heart allograft in mice were investigated. Human T lymphocytes could be separated into two subsets when they were incubated with theophylline in vitro. One subset lost their ability to form E rosette with sheep red blood cells and was called theophylline sensitive T cell (TS). The other one maintained their ability to form E rosette was called thyeophlline resistant T cell (TR). It was shown that TS inhibited and TR enhanced GVHR induced by total T lymphocytes in cyclophosphamide treated rats. Spleen lymphocytes from mice treated with aminophyline showed significant weaker GVHR than that treated with normal saline, suggesting that aminophylline might induce suppressor cells in vivo. The effect of aminophylline on the survival of heart heterotopic allograft was then investigated and the result showed that aminophylline prolonged the survival of transplant significantly.
